One hundred sixty one soil samples from the Parco Nazionale d'Abruzzo were analyzed. The keratinophilic fungi present in the soil samples were isolated and classified with the hair-bait method. The most common was T. ajelloi (28.6% of the total samples) followed by Chrysosporium keratinophilum (14.3%), C. sp. (11.2%), C. asperatum and Trichophyton sp. (6.8%), T. terrestre (5.6%), C. pannorum and C. state of Ctenomyces serratus (3.7%), C. tropicum (3.1%), M. cookei (2.5%) and the C. state of Arthroderma tuberculatum (1.9%). Other species, not belonging to the dermatophytes, were also isolated. The absence of dermatophytes was correlated with low human and animal population density.Altitude and vegetation did not seem to influence their distribution.  相似文献

Natural mycoflora associated with fumonisins were analyzed in 150 samples of freshly harvested corn from Central-Southern, Central-Western and Northern regions of the State of Paraná, Brazil and correlated to climatic conditions. The corn samples were frequently contaminated with Fusarium sp.(98.7 to 100%) and Penicillium sp. (93 to 100%), when compared to Aspergillus sp. (not detected to 27.7%). The highest contamination with potentially mycotoxigenic fungi occurred in corn harvested in the Central-Western region, where total mould and yeast counts ranged from 5.5 × 103 to 5.2 × 106 CFU/g, with 98.7% contaminated byFusarium sp. and 93% by Penicillium sp. In this region F. moniliforme (F. verticillioides) was the predominant Fusariumsp., and was isolated in 85.9% of the samples. Aspergillus sp. was isolated from 27.7% samples. FB1 was detected in 100% of the samples (mean of 2.39 g/g) and FB2 in 97.7% (mean of 1.09 g/g). Fumonisins were also detected in all samples from Northern region, with mean of 4.56 g/g (FB1) and 2.20 g/g (FB2).Considering 1.0 g/g as the threshold, 72% of the corn samples from the Central-West and 92% from the North were contaminated with concentrations above this value, in contrast to a 18.5% contamination rate from Central-Southern samples. Between corn planting to harvesting season, the average maximum temperature and relative humidity were 26 °C and 77.1%(Central-Southern), 27 °C and 69% (Northern)and 29.9 °C and 89.1% (Central-Western).Therefore, the higher fumonisins contamination of corn from Northern region when compared to the Central-South were due to the differences in rainfall levels (92.8 mm in Central-Southern, 202 mm in Northern) during the month preceding harvest.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

The fungal microflora of orchard (cock's-foot) grass seed (Dactylis glomerata L.) was determined for 140 samples obtained from thirteen countries. The microflora was assessed by visual examination of dry seed (20–50 x), by microscopic examination of centrifuged seed washings and by incubating seed on 2 % malt extract agar (MA) under a 10-day regime of diurnal cycling of temperature (23-9oC/i8-3oC) and near-ultraviolet radiation (12 h NUV/ 12 h dark). More than twenty-five species of potentially pathogenic fungi and forty species of saprophytes were detected. The pathogens included an Ascochyta sp., Botrytis cinerea, nine species of Drechslera including D. dactylitis (infested 23% of seed lots) and D. Useptatum (10% of seed lots), five species oiFusarium including F. avenaceum (5 % of seed lots), Lep-tosphaeria sp., Phleospora sp., Phoma sp., Phomopsis sp., Pyrenochaeta sp., Rhizoctonia sp., Rhynchophoma sp., Septoria sp. and Stagonospora sp. Many more fungi were identified from seed plated on MA and exposed to NUV than by examination of seed washings or visual examination of dry seed. A cross-sectional study was carried out to identify species and determine the prevalence of Cryptosporidium sp. shedding in pre-weaned and post-weaned dairy calves and to identify management factors that may be contributing to disease. A total of 240 calf faecal samples were collected from 16 farms in two districts in Johor, Malaysia, and screened by PCR. The overall Cryptosporidium prevalence was 27.1%. The prevalence of Cryptosporidium species in pre-weaned calves was 32.4% for C. parvum, 26.5% for C. bovis, followed by C. andersoni (20.6%), C. ryanae (11.8%) and mixed sp. (8.8%). The prevalence of Cryptosporidium species in post-weaned calves was 35% for C. bovis followed by C. andersoni and C. ryanae (30% each) and mixed sp. (5%). Subtyping analysis of 8 of the 11 C. parvum isolates at the gp60 locus identified five isolates as IIdA15G1, one as IIa18A3R1 and two isolates as IIa17G2R1. Management factors that increased the risk of Cryptosporidium infection included having other cattle farms close by, feeding calves with saleable milk, keeping pre-weaned calves in pens with slatted floors and keeping post-weaned calves in pens with a sand floor.  相似文献

Microbial diversity of soil and water samples collected from pyrochemicals exposed areas of Virdhunagar district (Tamil Nadu, India) was studied. Soil and water samples from cultivable area, waste land and city area of the same region were also studied for a comparative acount. There is a remarkable reduction in total heterotrophic bacterial population (THB) in pyrochemicals exposed soil and water samples (42 × 104 CFU/g and 5.6 × 104 CFU/ml respectively), compared to the THB of cultivable area soil and water samples (98 × 107 CFU/g and 38.6 × 107 CFU/ml). The generic composition the THB of the pyrochemicals exposed samples too exhibited considerable change compared to other samples. Pseudomonas sp. was the predominant one (41.6%) followed by Achromobacter sp. (25%) in pyrochemical exposed soil and Pseudomonas sp. was the predominant one (25%) in pyrochemical exposed water samples followed by Bacillus sp. (25%) and Micrococcus sp. (16.6%). It was observed that Cornybacterium sp. and Micrococcus sp. were absent completely in pyrochemical exposed soil and Achromobacter sp. was missing in the pyrochemical exposed water samples, which were present in the other samples. The outcome of this study clearly demonstrates that pollutants such as chemicals used in pyrotechniques affect the microbial biodiversity and suitable measures have to be taken to control the pollution level and to save biodiversity.  相似文献

Twenty five surface samples/moss cushions were collected for palynological analysis from open areas of Reasi District, Jammu and Kashmir (India). These samples were used to investigate the relationships between extant vegetation and modern pollen spectra, which serve as modern analogue for the reliable ecological interpretation of fossil pollen records. The present vegetation in the region comprises tropical dry deciduous forests and subtropical pine forests with scattered stands of oak. The pollen analysis reveals that Pinus sp. (average 69% in the pollen assemblages), amongst the conifers, dominates the pollen rain, which can be attributed to its high pollen productivity and exceptional pollen dispersal efficiency. Cedrus sp. and Podocarpus sp. pollen contribute with an average of 16 and 5% to the total pollen rain. Other conifers such as Picea sp., Abies sp., Juniperus sp. and Tsuga sp., as well as broad-leaved taxa such as Quercus sp., Alnus sp., Betula sp., Carpinus sp., Corylus sp., Juglans sp., Ulmus sp., Salix sp., Elaeocarpus sp., Mallotus sp. and Aesculus sp., have lower averages of 1 to 4.5% in the total pollen rain which could be either due to their poor pollen dispersal efficiency or to the poor preservation in the samples. Tubuliflorae (average 25%), Poaceae (average 6.26%), Cerealia and other crop plants (average 7.68%) are other prominent taxa in the pollen rain. The nearly complete absence of members of tropical dry deciduous forests in the pollen spectra likely is due to the fact that most species in this vegetation type are not wind pollinated.  相似文献

Samples of traditionally smoke-dried Clarias gariepinus (Burchell), Chrysichthys nigrodigitatus (Lacepede), Sarotherodon galilaeus (Trewavas), Heterotis niloticus (Cuvier) and Heterobranchus bidorsalis (Geoffroy) were obtained from Oja Oba Market, Ago-Iwoye, Nigeria and examined for fungal infestation. The fish samples incubated on potato dextrose agar (PDA) for 7 days showed fungal infestation. Fungi isolated and identified included Mucor sp., Aspergillus sp., Rhizopus sp. and Fusarium sp. Six fungal species were isolated from C. nigrodigitatus, five each on C. gariepinus and H. niloticus. Intramural aeromycological survey was performed at the Central Milk Dairy, Calcutta, covering eight locations within the Dairyusing Burkard personal volumetric air sampler. The locations were butter cold storage (−2 °C), cold store (8 °C), packaging section (23 °C), milk processing section (24 °C), reconstituent of skimmed milk (24 °C), quality control lab (25 °C), raw milk reception (28 °C) and loading dock (26 °C). A number of fungal spores, conidia and mycelia were recorded in different rooms: the highest spore quantity was recorded in the packaging section (23 °C) and the minimum at the butter cold store (−2 °C). The dominant spores consisted of Aspergillus niger, A flavus,Cladosporium sp., Fusarium sp., Curvularia sp.,Alternaria sp., Torula sp., Myrotheciumsp., Helminthosporium sp., Periconia sp.,Nigrospora sp. and Pithomyces sp. Human activities have a tremendous impact not only on the macroscopic world, but also on the micro-organisms. Here, Amplified Ribosomal DNA Restriction Analysis (ARDRA) was used for assessing the effect of the industrial sewage on the microbial community in sludge of Dongting Lake, the second-largest freshwater lake in China. The sludge samples from the outfall of the representative nitrogenous fertilizer plant near the lake were collected in March, 2010, and the sludge samples from the surrounding waters were treated as the control. The multi-element analysis results showed that the content of nitrogen, phosphorus in Sample SY were 1.9 and 1.47 times of the control group respectively. Based on restriction patterns derived from ARDRA, 26 representative clones (15 clones in the SY group and 11 clones in the DZ group) were sequenced. The sequence data and phylogenetic analysis of 16S rRNA gene presented that microorganism diversity of two sludge samples were abundant. Bacterial diversity presented among the outfall samples was dominated by Aeromonas sp. (5.8%), Acidimicrobidae sp. (5.8%) and Gemmatimonas sp. (5.0%). In contrast, bacterial diversity presented among the control group was dominated by Xanthomonas sp. (8.0%), Lautropia sp. (5.8%) and Duganella sp. (5.1%). The results indicated that due to the excessive of nitrogen and phosphorus discharged by the nitrogen fertilizer plant, the eutrophication in Dongting Lake has great influence on the microbial community structure.  相似文献

The molecular phylogeny of 12 species of Camponotus ants in Korea was examined using random amplified polymorphic DNA (RAPD) markers as inputs for an analysis of molecular variance (AMOVA) and cluster analysis to describe the relationships between species. For comparison, morphometric data (based on 10 morphological characters) were also gathered for phylogenetic analysis. Assessments of similarity between species were made, and the results of these assessments are compared for the molecular and morphological data sets. In the morphometric analysis, the following groups were identified: (i) C. atrox, C. kiusuensis, C. japonicus and C. concavus (93% similarity); (ii) C. sp. 1 (be diverging at 80% similarity); (iii) C. jejuensis, C. sp. 3, C. itoi, C. nawai and C. tokioensis (94.5% similarity); and (iv) C. nipponensis and C. quadrinotatus (94.5% similarity). Formica fusca was 73.5% similar to the 12 Camponotus species studied here. The group comprising C. nawai and C. tokioensis had the highest similarity index (97.36%), followed by the group comprising C. jejuensis and C. sp. 3 (95%), then C. atrox and C. kiusuensis (94.5%), and then C. nipponensis and C. quadrinotatus (93.5%). In the molecular analysis the following groups were identified: (i) C. atrox and C. jejuensis (30% similarity); (ii) C. concavus, C. japonicus and C. itoi (25% similarity); (iii) C. kiusuensis, C. nawai, C. sp. 3, C. nipponensis, C. quadrinotatus and C. sp. 1 (24% similarity); and (iv) C. tokioensis (be diverging at 23% similarity). The most closely related group in the molecular analysis was C. nawai and C. sp. 3 (75% similarity), followed by C. concavus and C. japonicus (50.5% similarity), then C. atrox and C. jejuensis (30%), and then C. quadrinotatus and C. sp. 1. Camponotus tokioensis was the least closely related to other species among the 12 species studied. Although C. jejuensis and C. tokioensis were found to be 93.6% similar on the basis of morphometric data, molecular data indicated only 23% similarity, the lowest similarity index between any two species considered here. Camponotus jejuensis has marked morphological similarities to C. tokioensis, but on the basis of molecular data gathered in the present study, we refute the proposal that they are synonyms or sister species.  相似文献

The isolation of endophytic actinomycetes from surface-sterilized tissues of 36 plant species was made using humic acid–vitamin (HV) agar as a selection medium. Of the 330 isolates recovered, 212 were from roots, 97 from leaves and 21 isolates from stems with a prevalence of 3.9, 1.7 and 0.3%, respectively. Identification of endophytic actinomycetes was based on their morphology and the amino acid composition of the whole-cell extract. Most isolates were classified as Streptomyces sp. (n = 277); with the remainder belonging to Microbispora sp. (n = 14), Nocardia sp. (n = 8) and Micromonospora sp. (n = 4). Four isolates were unclassified and 23 were lost during subculture. The most prevalent group of isolates were the Streptomyces sp. occurring in 6.4% of the tissue samples of Zingiber officinale. Scanning electron microscopy investigation of this plant revealed that 7.5% of the root and 5% of the leaf samples contained endophytes. Three of the Streptomyces sp. isolates strongly inhibited Colletotrichum musae, five were very active against Fusarium oxysporum and two strongly inhibited growth of both test fungi.  相似文献

 As an addendum to a recent publication on the terrestrial arthropod fauna of the Byers Peninsula Site of Special Scientific Interest (SSSI), which included preliminary identification of four Collembola, we now present detailed information on Collembola occurring within the SSSI. Five species were recorded [Friesea grisea (Schaffer), Tullbergia mixta Wahlgren, Isotoma (Folsomotoma) octooculata (Willem), Cyptopygus antarcticus Willem, and Cryptopygus sp. nov.]. Species occurrence and abundance differed between samples collected from poorly vegetated stony ground and vegetation cores. F. grisea was both the commonest (58% of individuals) and the most widely distributed (18/19 samples) springtail in the former habitat. Cryptopygus sp. nov. was recorded only in the vegetation core samples. All five species were more evenly distributed in vegetation samples, although C. antarcticus and Cryptopygus sp. nov. were again the least abundant. Fungal endophytes were isolated from leaves of Centella asiatica (Apiaceae) collected at Mangoro (middle eastern region of Madagascar, 200 km from Antananarivo). Forty- five different taxa were recovered. The overall foliar colonization rate was 78%. The most common endophytes were the non-sporulating species 1 (isolation frequency IF 19.2%) followed by Colletotrichum sp.1 (IF 13.2%), Guignardia sp. (IF 8.5%), Glomerella sp. (IF 7.7%), an unidentified ascomycete (IF 7.2%), the non-sporulating species 2 (IF 3.7%) and Phialophora sp. (IF 3.5%). Using sequences of the ribosomal DNA internal transcribed spacer (ITS) regions, major endophytes (IF > 7%) were identified as xylariaceous taxa or as Colletotrichum higginsianum, Guignardia mangiferae and Glomerella cingulata. Results from in vitro fungal disk experiments showed a strong inhibitory activity of the xylariaceous non-sporulating species 1 against G. mangiferae and C. higginsianum and of C. higginsianum against G. mangiferae. The pollen content of 31 honey samples from 19 different apiaries of El Hierro (Canary Islands) were subject to qualitative and quantitative melissopalynological analysis. The quantitative analysis demonstrated that 13% of the honey belonged to Maurizio Class I (<2?000?grains), 68% to Class II (2?000–10?000?grains) and 19% to Class III (10?000–50?000?grains). The pollen density ranges from 1?042?grains/g of honey to 24?478?grains/g with an average of 7?471?grains/g. According to the qualitative analysis, six honeys were typified as unifloral and 25 as multifloral. The unifloral honey samples were broken down as follows: two of heather (Erica arborea L.), two of Chamaecytisus proliferus (L. f.) Link‐type (“tagasaste”), one of Fabaceae (Genisteae sp.) and one of Lamiaceae Origanum vulgare L. ssp. virens (Hoffmanns. &; Link) Ietsw.‐type (thyme: Micromeria hyssopifolia Webb &; Berthel.). Honeydew elements were practically absent. Sixty‐nine pollen types were identified belonging to 42 families. The number of pollen types range between 18 and 39 (mean of 27.42). Foeniculum vulgare Mill.‐type pollen is present in all the samples. Galactites tomentosa Moench‐type, Echium plantagineum L., Echium L. sp., Bituminaria bituminosa (L.) C. H. Stirt., Chamaecytisus proliferus ‐type and Origanum vulgare ssp. virens ‐type pollen were found in 96.8% of the samples. The sensorial analysis indicated that honey types are generally of good quality, because 62% were evaluated as very high (16%), high (23%), and good (23%).  相似文献

From March 1997 to June 1999, samples of lemonleaves infested with citrus leafminer (CLM),Phyllocnistis citrella Stainton(Lepidoptera: Gracillariidae), were collectedfrom the Al-Masalha Citrus Orchard in theCentral Jordan Valley in order to rear CLMparasitoids and to study their populationtrends. Nine species of eulophid parasitoidswere reared from CLM larvae; these were Cirrospilus ingenuus Gahan, C. pictusNees, Pnigalio agraules, Pnigalio sp. B,Pnigalio sp. C, Citrostichusphyllocnistoides Narayanan, Ratzeburgiolaincompleata Boucek, Semielacherpetiolatus Girault, and Zagrammosoma sp.Cirrospilus ingenuus was the most commonparasitoid of CLM from March 1997 to June 1999and could be a good potential biologicalcontrol agent after rearing and releasing it incitrus orchards in the Jordan Valley. Onehyperparasitoid species was reared fromimmatures of parasitoids of CLM larvae, andidentified as Tyndrichus sp. An increasein the activity of parasitoids was observed inautumn 1999 when the temperature was between 15–20 °C and the relative humidity was between 55–65%.  相似文献

The microbiota of the lower female genital tract was evaluated from vaginal swabs obtained from 623 healthy pregnant women at gestation periods of 35–40 weeks. Isolated and identified microorganisms were expressed as percentages of total samples. As expected, lactobacilli made up the dominant vaginal microbiota (70%). Enterobacteriaceae, mainly Escherichia coli, Klebsiella spp. and Proteus, were present in 38% of the samples, which might reflect the possible contamination of vaginal tract with rectal microorganisms. Candida albicans was present in 10% of healthy pregnant woman assayed. Streptoccocci (Streptococcus sp. and Enterococcus faecalis with 3% and 4%, respectively) and other gram-positive cocci (Staphylococcus sp., 5%), along with other microorgansisms such as Gardnerella vaginalis (5%) and Pseudomonas aeruginosa (2%) may represent a potential infection risk. Streptococcus agalactiae (group B streptococci β-hemolytic, GBS) was detected in 7% of the samples. GBS infection is a leading cause of neonatal morbidity and mortality in the developed world. Furthermore, GBS was often co-isolated with C. albicans (54.5%) in the samples. A complete and detailed evaluation of the vaginal biota swab, with particular attention to the presence of potential pathogens such as GBS, is a preventive strategy that can provide useful information to obstetricians and gynecologist in managing the last days of pregnancy and delivery. Bacterial communities in crude oil and oil field production water samples from an oil gathering and transferring system in Changqing Oil field in China were investigated by 16S rRNA denaturing gradient gel electrophoresis (DGGE) analysis followed by gene cloning and sequencing. DGGE profiles showed that bacterial communities are far more rich in the water samples than that in the crude oil samples, and that bacteria related to Ochrobactrum sp. and Stenotrophomonas sp. were detected in all crude oil and oil field water samples. Bacteria related to Burkholderia sp., Brevundimonas sp., and Propionibacterium sp. were detected in the crude oil samples but not in water samples. Bacteria related to Hippea sp., Acidovorax sp., Arcobacter sp., Pseudomonas sp., Thiomicrospira sp., Brevibacterium sp., Tissierella sp. and Peptostreptococcus sp. were detected in the water samples but not in crude oil samples. Using an archaea-specific primer set, methanogens related to Methanomicrobials and Methanosarcinales were found in water samples but not in crude oil samples. The comparability of the microbial communities in the water and crude oil phase during the period of oil gathering and transferring process was 83.3% and 88.2%, respectively, indicating a stable structure of the microbial communities.  相似文献

The aim of the present study was to determine the percentage of agents, which can give rise to superficial fungal infections in Istanbul, Turkey. Between 2000 and 2007, the clinical samples collected from 8,200 patients attending the outpatient Dermatology Clinic at Mihrimahsultan Medical Center were examined by direct microscopy and culture. Pathogen fungi were detected in 5,722 of the patients. Of the isolates were 4,218 (74%) dermatophytes, 1,196 (21%) Candida sp., 170 (3%) Malassezia furfur, and 138 (2%) Trichosporon sp. Among the dermatophytes, Trichophyton sp. was the most common isolate followed by Epidermophyton floccosum (243) and Microsporum sp. Among the Candida species, C. albicans (549) was also frequently found. Onychomycosis was the most prevalent type of infection, followed by tinea pedis, tinea cruris, tinea corporis, and tinea capitis. In conclusion, our study showed that the most common isolated agents from superficial infections were T. rubrum being Candida sp. the second most prevalent.  相似文献

The experiment was carried out to evaluate the antifungal potential of aqueous and ethanolic extracts of four spices (Allium sativum, Zingiber officinale, Cinnamomum zeylanicum and Capsicum annuum) against the important post-harvest spoilage fungi isolated from diseased fruits. In total, 276 isolates of post-harvest spoilage fungi were isolated from four different fruit types (Persea americana, Musa acuminata, Citrus sinensis and Lycopersicon esculentum), out of which 183 isolates were identified while 93 isolates were remain unidentified. The most dominant post-harvest spoilage fungus was Rhizopus sp. (26.45%), followed by Penicillium sp. (19.93%), Aspergillus sp. (10.86%) and Fusarium sp. (9.06%). Results of disc diffusion assay showed that ethanolic extract of C. zeylanicum was found to be most effective against Penicillium sp., followed by aqueous extracts of A. sativum. The ethanolic extract of C. zeylanicum in agar amended assay and minimal inhibitory concentrations were found to be very efficient (100% inhibition) against all the tested fungi. Results of in vivo study showed that pre-inoculated C. zeylanicum ethanolic extract and A. sativum aqueous extract were found effective in reducing the disease severity (6.24–13.67%) and (7.91–13.15%) against the Penicillium, Rhizopus, Aspergillus and Fusarium sp.  相似文献

Groundnut samples from 21 selected markets in the 10 regions of Ghana yielded high levels of the aflatoxigenic fungus Aspergillus flavus on half-strength potato dextrose agar. The fungus was associated with 31.7 and 12.8%, respectively, of all damaged and undamaged kernels assayed. Only 0.24% of total kernels assayed yielded A. parasiticus. Other fungi detected from total kernels assayed were A. niger (34%), A. candidus (1.45%), A. tamarii (3.93%), A. ochraceous (5.26%), Fusarium spp. (1.7%) Penicillium spp. (5.19%), a Mucor sp. (2.3%), a Trichoderma sp. (0.2%), Rhizopus stolonifer (12%) and certain unidentifiable fungi (11.72%). Total aflatoxin levels ranging from 5.7 to 22, 168 ppb were identified with damaged kernel samples. The mycotoxin was not detected in 50% of undamaged kernel samples tested and very low levels mostly ranging from 0.1 to 12.2 ppb were associated with the undamaged samples that tested positive for aflatoxins. In a novel in vitro microbial assay to determine the effectiveness of certain plant extracts against aflatoxin synthesis, extracts from Xylopia aethiopica, Monodera myristica, Cinnamomum verum and Piper nigrum permitted fungal growth in 1.5% potato-dextrose broth while completely suppressing NOR formation. These extracts, however, could not suppress NOR formation in a yeast extract sucrose medium.  相似文献
