The defense characteristics of allelopathic rice accession PI312777 and its counterpart Lemont induced by exogenous salicylic acid (SA) to suppress troublesome weed barnyardgrass (BYG) were investigated using the methods of suppression subtractive hybridization (SSH) and real-time fluorescence quantitative PCR (qRT-PCR). The results showed that exogenous SA could induce the allelopathic effect of rice on BYG and this inducible defense was SA dose-respondent and treatment time-dependent. PI312777 exhibited higher inhibitory effect than Lemont on BYG after treated with different concentrations of SA. The activities of cell protective enzymes including SOD, POD and CAT in the BYG plants co-cultured with PI312777 treated by SA were highly depressed compared with the control (co-cultured with rice without SA-treatment). Similar but lower depression on these enzymes except for CAT was also observed in the BYG plants when co-cultured with Lemont treated by SA. It is therefore suggested that allelopathic rice should be more sensitive than non-allelopathic rice to exogenous SA. Seventeen genes induced by SA were obtained by SSH analysis from PI312777. These genes encode receptor-kinase proteins, ubiquitin carrier proteins, proteins related to phenylpropanoid metabolism, antioxidant related proteins and some growth-mediating proteins. The differential expressions of these genes were validated in part by qRT-PCR in the two rice accessions. Our work elucidated that allelopathic rice possesses an active chemical defense and auto-detoxifying enzyme system such as the up-regulated enzymes involved in de novo biosynthesis of phenolic allelochemicals and the glutathione-S-transferase (GST) associated with xenobiotic detoxification.  相似文献

This study aimed to determine the effects of exogenous application of salicylic acid (SA) on the toxic effects of salt in relation to ethylene and polyamine synthesis, and to correlate these traits with the expression of genes involved in ethylene and polyamine metabolism in two tomato species differing in their sensitivity to salt stress, Solanum lycopersicum cv Ailsa Craig and its wild salt‐resistant relative Solanum chilense. In S. chilense, treatment with 125 mM NaCl improved plant growth, increased production of ethylene, endogenous salicylic acid and spermine. The production was related to a modification of expression of genes involved in ethylene and polyamine metabolism. In contrast, salinity decreased plant growth in S. lycopersicum without affecting endogenous ethylene, salicylic or polyamine concentrations. Exogenous application of salicylic acid at 0.01 mM enhanced shoot growth in both species and affected ethylene and polyamine production in S. chilense. Concomitant application of NaCl and salicylic acid improved osmotic adjustment, thus suggesting that salt and SA may act in synergy on osmolyte synthesis. However, the beneficial impact of exogenous application of salicylic acid was mitigated by salt stress since NaCl impaired endogenous SA accumulation in the shoot and salicylic acid did not improve plant growth in salt‐treated plants. Our results thus revealed that both species respond differently to salinity and that salicylic acid, ethylene and polyamine metabolisms are involved in salt resistance in S. chilense.  相似文献

Soil salinity is one of the key factors adversely affecting the growth, yield, and quality of crops. A pot study was conducted to find out whether exogenous application of salicylic acid could ameliorate the adverse effect of salinity in lemongrass (Cymbopogon flexuosus Steud. Wats.). Two Cymbopogon varieties, Krishna and Neema, were used in the study. Three salinity levels, viz, 50, 100, and 150 mM of NaCl, were applied to 30-day-old plants. Salicylic acid (SA) was applied as foliar spray at 10?5 M concentration. Totally, six SA-sprays were carried out at 10-day intervals, following the first spray at 30 days after sowing. The growth parameters were progressively reduced with the increase in salinity level; however, growth inhibition was significantly reduced by the foliar application of SA. With the increase in salt stress, a gradual decrease in the activities of carbonic anhydrase and nitrate reductase was observed in both the varieties. SA-treatment not only ameliorated the adverse effects of NaCl but also showed a significant improvement in the activities of these enzymes compared with the untreated stressed-plants. The plants supplemented with NaCl exhibited a significant increase in electrolyte leakage, proline content, and phosphoenol pyruvate carboxylase activity. Content and yield of essential oil was also significantly decreased in plants that received salinity levels; however, SA overcame the unfavorable effects of salinity stress to a considerable extent. Lemongrass variety Krishna was found to be more adapted to salt stress than Neema, as indicated by the overall performance of the two varieties under salt conditions.  相似文献

Application of endogenous plant hormone salicylic acid (SA) or essential oil component carvacrol (CA) in elms enhances tree resistance to the Dutch elm disease pathogen, although the effect of these compounds on tree metabolism is unknown. The chemical changes induced by SA or CA treatments in Ulmus minor were studied through gas chromatography–mass spectrometry (GC–MS) analysis of xylem tissues. Treatments consisted of fortnightly irrigating seedlings with water, SA or CA at 600 mg L−1. The chemical composition of the xylem tissues sampled from treated trees was significantly altered depending on the treatment type. SA treatment induced an accumulation of the sinapyl alcohol, a precursor of lignin and other phenylpropanoid-derived products. CA treatment induced an accumulation of the methyl esters of palmitic, linoleic and stearic acids. Both treatments resulted in early bud burst and SA significantly reduced sapwood radial growth, possibly as a consequence of a trade-off between tree growth and tree defence. The enhanced resistance provided by these treatments is discussed.  相似文献

为探索宁夏枸杞对枸杞瘿螨致瘿过程诱导的激素响应及外源水杨酸对枸杞瘿螨的影响,利用LC-MS/MS技术对枸杞瘿螨为害后宁夏枸杞叶片中水杨酸(SA)、茉莉酸(JA)、生长素(Auxin)和脱落酸(ABA)4种内源激素进行测定,并通过浸叶法研究外源SA对枸杞瘿螨侵染过程、虫瘿生长和种群增长的影响。结果表明: 枸杞瘿螨的致瘿行为可引发枸杞叶片中多种内源激素产生变化,其中SA和JA的变化最显著,分别是对照组含量的4.0和13.0倍,而Auxin和ABA的变化不显著;外源SA对枸杞瘿螨虫瘿生长有显著抑制作用,且随时间的延长,抑制作用增强,同时虫瘿内瘿螨种群数量受到显著抑制。枸杞瘿螨能够同时诱导枸杞产生SA和JA抗性,外源SA对枸杞瘿螨虫瘿生长和瘿螨种群数量均具有显著抑制作用,利用外源SA防治枸杞瘿螨具有一定的应用前景。  相似文献

The temporal gradations of the investigated phenolics in Norway spruce bark after bark beetle (Ips typographus) attack followed the general eco-physiological concept. Treatment with salicylic acid inhibits bark beetle colonisation, alleviates the phenolic responses and activates the synthesis of condensed tannins on later sampling dates.

Abstract

Conifer bark is the target of numerous organisms due to its assimilated transport and nutrient storage functions. In the presented study, 100 mM salicylic acid (SA) was applied onto Norway spruce stems prior to being infested with bark beetles (Ips typographus L.), to study the temporal gradation of changes in condensed tannins (CT) and total phenolics (tPH) and their significance for mediating stress-tolerance. A significant accumulation of CT was monitored in untreated trees in response to progressive bark beetle infestation occurring from May onwards. In SA-treated infested trees, the CT values remained at control levels until May, but after the re-treatment of infested trees in June, the concentrations of CT rose significantly in comparison to the controls. The tPH values dropped 16 days after SA-treatment, independent of infestation, and later on remained at control level until July. In contrast, tPH contents accumulated in untreated infested trees in May, eased in June and increased again in July, when the trees were affected by the second generation of bark beetles. To sum up, in May and July when the highest beetle-flight activity was monitored the metabolic shift of phenolics within untreated infested trees differed significantly from the response of SA-treated trees. In addition, on SA-treated trees less entrance holes were monitored over the whole period of sampling when compared to untreated infested trees. These results provide evidence that SA-treatment alleviates the phenolic responses, activates the synthesis of condensed tannins and inhibits bark beetle colonisation.

Nitric oxide (NO), salicylic acid (SA), and reactive oxygen species (ROS) are important signal molecules that mediate plant resistance reactions and play important roles in secondary metabolism. To research the signal transduction pathway of the endophytic fungal elicitor from Fusarium sp. E5 promoting secondary metabolism in Euphorbia pekinensis suspension cells, the changes in NO, SA, ROS, and isoeuphpekinensin contents in the cells were investigated after elicitor addition to the cell suspension culture. The elicitor did not change H2O2 or O2 ? contents notably, whereas NO and SA contents were enhanced. Both the NO donator sodium nitroprusside (SNP) and SA enhanced isoeuphpekinensin content in the absence of the fungal elicitor, whereas the NO scavenger cPTIO and SA biosynthesis inhibitor cinnamic acid (CA) inhibited isoeuphpekinensin accumulation in the presence of the elicitor. In addition, cPTIO inhibited SA production induced by the fungal elicitor. CA did not inhibit NO production, but it significantly inhibited isoeuphpekinensin accumulation. The results demonstrated that in Euphorbia pekinensis suspension cells the endophytic fungal elicitor induced increased NO content and SA production, which promoted isoeuphpekinensin accumulation. ROS are clearly not involved in the endophytic fungus–host interaction signaling pathway.  相似文献

Salinity has a great influence on plant growth and distribution. A few existing reports on Artemisia annua L. response to salinity are concentrated on plant growth and artemisinin content; the physiological response and salt damage mitigation are yet to be understood. In this study, the physiological response of varying salt stresses (50, 100, 200, 300, or 400 mM NaCl) on A. annua L. and the effect of exogenous salicylic acid (0.05 or 0.1 mM) at 300-mM salt stress were investigated. Plant growth, antioxidant enzyme activity, proline, and mineral element level were determined. In general, increasing salt concentration significantly reduced plant growth. Superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were stimulated by salt treatment to a higher enzyme activity in treated plants than those in untreated plants. Content of proline had a visible range of increment in the salt-treated plants. Distribution of mineral elements was in inconformity: Na+ and Ca2+ were mainly accumulated in the roots; K+ and Mg2+ were concentrated in leaves and stems, respectively. Alleviation of growth arrest was observed with exogenous applications of salicylic acid (SA) under salt stress conditions. The activity of SOD and POD was notably enhanced by SA, but the CAT action was suppressed. While exogenous SA had no discernible effect on proline content, it effectively inhibited excessive Na+ absorption and promoted Mg2+ absorption. Ca2+ and K+ contents showed a slight reduction when supplemented with SA. Overall, the positive effect of SA towards resistance to the salinity of A. annua will provide some practical basis for A. annua cultivation.  相似文献

The constitutive and induced resistance were studied in two varieties (Khanyari and Kawdari) of kale, Brassica oleracea var. acephala in response to cabbage butterfly, Pieris brassicae infestation and exogenous application of jasmonic acid (JA) and salicylic acid (SA). Phenols, condensed tannins, flavonoids and proteins were measured at six days after JA (1?mM) and SA (1?mM) application and/or insect infestation. Plant damage and larval weights were also recorded. Khanyari variety showed highest amounts of phenols (208.23?μg/g FW), condensed tannin (347.76?μg/g FW), flavonoids (175.61?μg/g FW) and proteins (0.71?mg/g FW) in plants pre-treated with JA and infested with insects. The PAL activity was high in response to JA application followed by insect infestation. Insects reared on Khanyari and Kawdari plants pre-treated with JA prior showed significantly reduced larval weights (97.88 and 102.46?mg, respectively). Damage was low in plants pre-treated with JA in Khanyari at 3, 6 and 9?days after treatment (10.52%, 8.52%, and 5.30%, respectively). Thus, JA can play an important role in plant defense in kale against P. brassicae.  相似文献

There are two major modes for plant recognition of biotrophic microbial pathogens. In one mode, plant pattern recognition receptors (PRRs) recognize microbe associated molecular patterns (MAMPs, also called PAMPs), which are molecules such as flg22, a fragment of bacterial flagellin. In the other mode, the products of plant resistance (R) genes recognize pathogen effectors or host proteins modified by effectors. Salicylic acid (SA) -mediated defense responses are an important part of R gene-mediated resistance. It was not clear how these two signaling mechanisms interact with each other. Recently, we reported that treatment with flg22 triggered SA accumulation in Arabidopsis leaves. Disruptions of SA signaling components strongly affected MAMP-triggered gene expression responses. Flg22-triggered resistance to Pseudomonas syringae pv. tomato DC3000 (Pst DC3000) was partly dependent on SA signaling. Our results demonstrated the importance of SA signaling in flg22-triggered resistance and, at the same time, the importance of some other signaling mechanism(s) in this resistance. Heptanoyl salicylic acid (HSA) is a salicylic acid (SA) derivative obtained by esterification of 2-OH benzoic acid with heptanoic acid. In wheat, the protection levels obtained against Blumeria graminis f. sp. tritici (Bgt) increased from 50% with SA to 95% with HSA. Using molecular, biochemical and cytological approaches, we investigated here how wheat lipid metabolism is differentially activated by SA and HSA in both infectious and non-infectious conditions, and how Bgt infectious process is altered by both inducers. First, in the absence of Bgt, continuous lipoxygenase (LOX)-encoding gene expression and corresponding activity were specifically induced by HSA. Moreover, compared to SA, HSA treatment resulted in earlier up-regulations of the phospholipase C2-encoding gene expression and it specifically affected the expression of a lipid transfer protein-encoding gene. In infectious context, both HSA and SA sprayings impaired penetration events and therefore haustorium formation, leading to less frequent fungal colonies. While this alteration only slowed down the evolution of Bgt infectious process in SA-sprayed leaves, it completely impaired the establishment of successful infectious events in HSA-sprayed leaves. In addition, HSA induced continuous increases of a LOX-encoding gene expression and of the corresponding LOX activity when compared to SA-sprayed leaves. Lipid metabolism is therefore overall highly responsive to HSA spraying and could represent effective defence mechanism triggered during the induction of resistance in wheat toward Bgt. The concepts of priming and energy costs of the defences induced by SA and HSA are also discussed.  相似文献

The effect of biochemical responses in wheat seedlings cultivar Falat against lesion nematode (Pratylenchus thornei) was investigated by two factors inducer resistance salicylic acid (SA) and methyl jasmonate (MeJA). Foliar sprays with 1?ml SA 1?mM and 1?ml MeJA 100?μM was conducted on 20-day-old wheat (cv. Falat) seedlings. About 24?h after foliar sprays, plants were inoculated by root lesion nematode. Phenylalanine ammonia-lyase (PAL) activity was increased significantly in all treatments two days after inoculation over control. However, PAL activity was decreased in all treatments five days after inoculation in compared to control. What is more, in both time points, total phenol increased in all treatments over control. The comparison of both inducers SA and MeJA on total phenol in healthy and infected wheat seedlings in both days two and five after inoculation of nematode indicated that total phenol has decreased significantly in all treatments in the fifth day.  相似文献
