以1年生紫斑牡丹幼苗为试验材料,采用不同浓度(0、100、300、500 mg/L)赤霉素(GA_3)喷施叶片处理,通过透射电镜、扫描电镜、光学显微镜观察幼苗叶片解剖结构,光合仪测定幼苗光合参数并以酶联免疫吸附法测叶片内源激素含量,探究外源GA_3对紫斑牡丹幼苗叶片解剖结构、光合特性和内源激素水平的影响。结果表明:(1)低浓度GA_3处理的紫斑牡丹叶肉细胞增大,栅栏组织外层细胞中叶绿体数量增加,高浓度GA_3处理则与之相反;GA_3处理叶片的栅栏组织/海绵组织比值(P/S)、组织结构紧密度(CTR)均下降,而其组织结构疏松度(SR)增加;GA_3处理的幼苗叶片的叶肉细胞内各叶绿体大小显著大于对照,随着GA_3处理浓度增加,紫斑牡丹叶肉细胞内叶绿体的体积趋于增大,类囊体垛叠凝聚逐渐松散,叶绿体上淀粉颗粒在300 mg/L GA_3处理中较明显;叶片气孔长度、宽度、气孔器大小、气孔开度和气孔密度随着GA_3浓度升高先升高后下降,同时叶片上表皮角质层厚度随GA_3浓度的升高而增加。(2)紫斑牡丹叶片净光合速率(P_n)、气孔导度(Cond)、蒸腾速率(T_r)、水分利用率(WUE)在100和300 mg/L GA_3处理下大都显著高于对照,且300 mg/L GA_3处理显著高于其余处理,而其在500 mg/L GA_3处理下显著低于对照。(3)紫斑牡丹叶片脱落酸(ABA)和吲哚乙酸(IAA)含量均在500 mg/L GA_3下显著高于对照,而在其余浓度处理下不同程度低于对照,叶片内源玉米素核苷(ZR)和GA_3含量均在300 mg/L GA_3处理下显著高于其余处理和对照,而其余处理相比对照均无显著变化;叶片的ZR/ABA、ZR/IAA、ZR/GA_3和(IAA+GA_3+ZR)/ABA比值都在300 mg/L GA_3处理下显著高于其他处理,叶片的IAA/ABA和ABA/GA_3比值均在500 mg/L GA_3处理下显著高于其他处理。研究发现,适宜浓度外源GA_3处理,能显著提高紫斑牡丹幼苗叶片光合速率、水分利用效率及蒸腾速率,调节植物体内源激素的含量及平衡,从而使叶片能合成较多有机物,促进幼苗生长。  相似文献

The role of endogenous gibberellin (GA) in the flowering of the short-day plant, Pharbitis nil, was investigated by using uniconazole, which is a specific inhibitor of GA biosynthesis. Both the endogenous GA level and flowering response decreased with increasing concentration of uniconazole applied via the roots. The strongest inhibition of flowering was observed when uniconazole was applied one day before a 15-h dark treatment. The inhibition by uniconazole was overcome by an application of GAs to the plumules, the order of effectiveness of the endogenous GAs in P. nil being GA1 ≧GA20>GA19≧GA44>GA53»GAH. This is the first report of the correlation between the endogenous GA level and flowering response in P. nil. It was found that endogenous GAs were required for the flowering of P. nil during or just after the dark period.  相似文献

Strategies based on the application of biotechnologies to crop improvement programmes generally require regeneration of whole plants from cells or tissues cultivated in vitro. In Cucurbitaceae, regeneration can occur either through a caulogenic or an embryogenic developmental pathway. Reports of somatic embryogenesis have dealt with the main cultivated crops, i.e. cucumber, melon, squash and watermelon. Somatic embryogenesis and plant recovery are obtained from numerous sources including protoplasts, but the best results are observed with explants coming from seedlings, especially cotyledons and hypocotyls. The genetic constitution of mother plants also seems to play a key role in the success of embryogenesis, but few systematic studies on genotype effect have been published. Somatic embryos can exhibit developmental abnormalities, particularly when they arise from protoplast-derived cultures. Generally, data concerning embryo yield, rate of germination and plant development and characteristics of regenerated plants and their progeny, has not been provided in previous reports. Environmental factors often affect plant growth bymodifying the levels of endogenous gibberellins (GAs).In this study, the involvement of GAs in theregulation of enhanced shoot growth in tomato (Lycopersicon esculentum Mill.) plants grown in soiltreated by solarization (a soil disinfestation method)was investigated. Seedlings at the cotyledonary stagewere transplanted into either solarized or untreatedcontrol soil. Plants in both soils grew free of anydisease symptoms. As soon as four days after planting,seedlings in solarized soil had a higher dry weightcompared to the control. Throughout most of theexperimental period of 18 days, leaf weight ratio washigher in the solarized vs. the control soil. Treatingshoot tips of control plants with 0.1 mg.L-1GA3 resulted in enhanced leaf and stem growth,thus reaching values similar to those of plants grownin solarized soil. The opposite effect was obtained bytreating plants grown in solarized soil with1 mg.L-1 uniconazole, a GA biosynthesisinhibitor. Quantitative GC-MS analyses revealed thatGA1 content in one and two-weeks old transplantsgrown in various solarized soils was up to 1.8 fold,and that GA3 content in two-weeks old plants wasup to five fold the values in the control. Theseincreases were linearly correlated with the increasein leaf dry weight. The growth and flowering response of a cold-requiring cauliflower (Brassica oleracea var. botrytis cv. 60 day) to a range of temperatures under 10 h photoperiod and to growth regulator application were investigated. Endogenous gibberellin A1(GA1) concentrations were also assessed under these treatments. Flowering and growth of the inflorescence stalk were correlated with plant developmental stage at the time of a vernalizing cold treatment. Temperature and its duration also affected flowering and inflorescence development. The most effective temperature for inflorescence induction was 10 °C. Flowering did not occur in non-vernalized plants (25 °C) even though they had been treated with GA3. Application of GA3 promoted inflorescence stalk elongation greatly in vernalized plants (10 °C), but less so in partially vernalized plants (15 °C or 20 °C). Paclobutrazol (PP333) sprayed at the 8–9 leaf stage significantly suppressed inflorescence stalk length and slightly delayed flower bud formation and anthesis. Vernalization at 10 °C increased endogenous GA1 content in both leaves and the inflorescence stalk irrespective of GA3 or PP333 treatment. Application of GA3 tended to increase GA1 levels, while PP333 significantly reduce GA1, both irrespective of vernalization. Vernalization is an important factor for flowering, but not curd formation in this cauliflower cv. 60 day and GA1 is likely a causal factor in inflorescence stalk elongation.  相似文献

为探讨GA_3和Spd对杜鹃(Rhododendron simsii)开花花期和开花品质的影响,研究了外源GA_3和Spd对杜鹃开花期光合特性和抗氧化系统的变化。结果表明,外源GA_3对花期有显著的提前作用,Spd对花期有明显的延迟作用,但两者均使花期延长、花径增大且成花率提高。GA_3和Spd处理提高了花期叶片的光合色素含量和净光合速率(Pn)、气孔导度(Gs)和胞间CO_2浓度(Ci);GA_3处理提高了叶片的蒸腾速率(Tr),而Spd使叶片的Tr下降,两者均有效缓解了末花期叶绿素含量的下降。GA_3和Spd处理显著降低了花瓣MDA含量,提高了抗氧化酶SOD、POD和CAT活性,并减缓了末花期SOD的下降,有效延缓了衰老进程,延长花期。以1 600 mg L~(–1) GA_3和0.10 mmol L~(-1) Spd处理效果较好,能有效提高杜鹃花的观赏品质。  相似文献

Earlier, we reported that mutation in the Male Sterile33 (MS33) locus in Arabidopsis thaliana causes inhibition of stamen filament growth and a defect in the maturation of pollen grains [Fei and Sawhney (1999) Physiol Plant 105:165–170; Fei and Sawhney (2001) Can J Bot 79:118–129]. Here we report that the ms33 mutant has other pleiotropic effects, including aberrant growth of all floral organs and a delay in seed germination and in flowering time. These defects could be partially or completely restored by low temperature or by exogenous gibberellin A4 (GA4), which in all cases was more effective than GA3 Analysis of endogenous GAs showed that in wild type (WT) mature flowers GA4 was the major GA, and that relative to WT the ms33 flowers had low levels of the growth active GAs, GA1 and GA4, and very reduced levels of GA9, GA24 and GA15, precursors of GA4. This suggests that mutation in the MS33 gene may suppress the GA biosynthetic pathway that leads to GA4 via GA9 and the early 13-H C20 GAs. WT flowers also possessed a much higher level of indole-3-acetic acid (IAA), and a lower level of abscisic acid (ABA), relative to ms33 flowers. Low temperature induced partial restoration of male fertility in the ms33 flowers and this was associated with partial increase in GA4. In contrast, in WT flowers GA1 and GA4 were very much reduced by low temperature. Low temperature also had little effect on IAA or ABA levels of ms33 flowers, but did reduce (>2-fold) IAA levels in WT flowers. The double mutants, ms33 aba1-1 (an ABA-deficient mutant), and ms33 spy-3 (a GA signal transduction mutant) had flower phenotypes similar to ms33. Summary Embryogenic masses were obtained from immature leaves of peanut (Arachis hypogaea L.) cultured on a medium containing 20 mg/l 2,4-D. Somatic embryos developed from these masses following transfer to a medium containing 3 mg/l 2,4-D. The embryo morphology was quite variable. Following transfer to hormone-free medium, these embryos germinated. Shoot elongation was obtained in 25% of the embryos following transfer to a medium supplemented with 0.5 mg/l each of BAP and Kn. Primary dormancy in A. retroflexus seeds wascompletely broken by dry storage or ethylene treatment and partially removedwith GA3. Norbornadiene counteracted the dormancy breaking action ofethylene and GA3. The GA3 effect was lowered bycobaltous ions. ABA increased the ethylene requirement in primary dormant seeds.Dormant seeds had a similar or different ability to produce ethylene and ACCoxidase in vivo activity than did non-dormant seeds,depending on the period of incubation. Dormant seeds contained less endogenousACC than non-dormant seeds. Thus, ethylene seems to play an essential role inthe release of primary dormancy in A. retroflexus seeds.Ethylene also participates in the release of dormancy achieved by GA3treatment. In this study, 5 μmol·L−1 abscisic acid (ABA) and gibberellic acid (GA3) were used to study the effect of both growth regulators on the morphological parameters and pigment composition of Andrographis paniculata. The growth regulators were applied by means of foliar spray during morning hours. ABA treatment inhibited the growth of the stem and internodal length when compared with control, whereas GA3 treatment increased the plant height and internodal length. The total number of leaves per plant decreased in the ABA-treated plants, but GA3 treatment increased the total number of leaves when compared with the control. Both growth regulators (ABA and GA3) showed increased leaf area. ABA and GA3 treatments slightly decreased the total root growth at all the stages of growth. The growth regulator treatments increased the whole plant fresh and dry weight at all stages of growth. ABA enhanced the fresh and dry weight to a larger extent when compared with GA3. An increase in the total chlorophyll content was recorded in ABA and GA3 treatments. The chlorophyll-a, chlorophyll-b, and carotenoids were increased by ABA and GA3 treatments when compared with the control plants. The xanthophylls and anthocyanin content were increased with ABA and GA3 treatments in A. paniculata plants.  相似文献

以紫斑牡丹种子为试验材料,研究不同浓度的赤霉素(GA_3)处理对种子生根以及生根过程中营养物质、酶活性和内源激素水平变化的影响,为探讨紫斑牡丹种子萌发机制提供依据。结果表明:(1)GA_3处理能够促进种子生根,并以300 mg/L GA_3处理对种子生根效果最好,与对照相比可提前14.67 d生根,生根率可达71.00%。(2)与对照相比,GA_3处理可以在0~15 d时促进种子淀粉水解和可溶性糖的积累,并加速可溶性蛋白的消耗,在0~30 d促进过氧化物酶(POD)活性的提高,从而促进种子萌发生根。(3)在种子沙藏生根过程中,种子脱落酸(ABA)含量呈下降趋势,赤霉素(GA)、玉米素核苷(ZR)和吲哚乙酸(IAA)含量均表现出先上升后下降的趋势,与对照相比,GA_3处理可使种子GA、ZR和IAA的含量在沙藏前期明显上升,以解除种子休眠。研究发现,外源GA_3处理可以调控紫斑牡丹种子内源激素含量和POD活性的变化,促进营养物质转化,从而提前解除种子休眠使其萌发。  相似文献

Gibberellin A1 (GA1), GA3 and GA4 inhibited the sprouting of nondormant bulbils of Chinese yam, Dioscorea opposita, where the effectiveness of the GAs was as follows: GA4>GA1+GA3. Uniconazole and prohexadione, plant growth retardants, promoted the sprouting of half-dormant bulbils. By contrast, these retardants inhibited the sprouting of nondormant bulbils. Gibberellin A3 (GA3) and A4 (GA4) which were applied to the stems of the sprouted bulbils, promoted stem elongation, but GAs applied to the bulbous parts inhibited this process. The effectiveness of the GAs on stem elongation was as follows: GA3+GA4 for the promotion and GA4 > GA3 for the inhibition. Uniconazole applied to the stem inhibited the stem elongation of the sprouted bulbils. These results suggest the possible involvement of endogenous GAs in the induction and maintenance of bulbil dormancy of D. opposita, as well as in the bulbil sprouting and subsequent stem elongation.  相似文献

Changes in gibberellins (GAs), indole-3-acetic acid (IAA), and cytokinins associated with the transition from vegetative growth to reproductive growth in Humulus lupulus L. buds and leaves harvested at fortnight intervals were studied. During vegetative growth, GA1 increased gradually and the lowest content was observed during flower development. Both GA3 and GA4 showed a dramatic increase in the samples taken from the apical part of axillary branches from plants 4–5 m high, which corresponds to the maximum vegetative development prior to macroscopically visible inflorescences. Notable increases in the cytokinins trans-zeatin (t-Z), isopentenyladenine (iP), and the riboside and ribotide forms of iP were also obtained. The auxin, indole-3-acetic acid, was the most abundant plant hormone, and its content was highest during vegetative growth. These results show for the first time a relationship between endogenous hormone profiles and both vegetative and reproductive development in hop plants, which may be relevant for future research on the control of the flowering by exogenous hormone applications.  相似文献

该研究采用ISSR分子标记,对黄枝油杉7个自然种群的遗传多样性进行了分析。结果表明:用12条ISSR引物对218个黄枝油杉个体进行扩增,共扩增出125个位点。在物种水平上,多态性位点百分数( PPL)为100.00%,Shannon信息多样性指数( I)为0.4177,Nei’ s基因多样性指数( H)为0.2666;在种群水平上,多态性位点百分数(PPL)在71.20%~92.00%之间,平均值为80.69%,Shannon信息多样性指数(I)在0.3273~0.3886之间,平均值为0.3548,Nei’ s基因多样性指数( H)在0.2139~0.2478之间,平均值为0.2291。这说明黄枝油杉在物种水平和种群水平上均显示出较高的遗传多样性。 Nei’ s遗传多样性分析( Gst=0.1433)和AMOVA分析(Φst=17.91%)表明,黄枝油杉的遗传变异主要存在于种群内,种群间的遗传分化程度较低,种群间保持一定的基因交流( Nm=2.9890>1)。 Mantel分析显示,黄枝油杉种群间的遗传距离和地理距离之间不存在显著的相关关系( r=0.4567, P=0.0610>0.05)。  相似文献

In vitro culture of Chenopodium murale L. (ecotype 197) green and herbicide SAN 9789 - treated "white" plants was established and the effects of benzylaminopurine (BAP), indole-3-acetic acid (IAA) and gibberellic acid (GA3) on growth and flowering were tested. Green plants did not flower on glucose free media, while 17 % of plants flowered on 5 % glucose-containing medium. SAN 9789 (10–5 M) inhibited growth and flowering. BAP and IAA (0.1 – 5 mg dm–3) also inhibited growth and flowering of green and "white" plants. GA3 (10 mg dm–3) stimulated leaf development in green plants, but had no significant effect on "white" plants, and stimulated flowering of green (41 %) and "white" (33 %) plants.  相似文献

The involvement of gibberellins in the control of flowering of sunflower was studied by direct application of GA3 to the apex of the plants, analysis of the endogenous levels of gibberellin-like substances at different plant ages, and indirectly by the application of paclobutrazol, an inhibitor of gibberellin synthesis. GA3 speeded-up flower initiation and floral apex development. The time of GA3 application was more critical than the amount of GA3 applied. The endogenous levels of gibberellin-like compounds increased significantly by day 15 after sowing. The application of paclobutrazol markedly delayed floral initiation and this effect was also depedent on plant age. Both GA3 and paclobutrazol had their greatest effects between 10 and 20 days after sowing suggesting that an increase in gibberellins in that time period plays a role in floral initiation.  相似文献

A very sensitive and specific bioassay using prohexadione calcium [BX-112, which blocks 2- and 3-hydroxylation of gibberellins (GAs)] with uniconazole (which blocks oxidation of ent-kaurene, ent-kaurenol and ent-kaurenal) in a microdrop assay was developed for several rice (Oryza sativa L.) varieties, including cv. Waito-C, which is already specific to 3-hydroxylated GAs. The sensitivity and specificity of cvs. Waito-C, Tan-ginbozu and Koshihikari to 3-hydroxylated GAs was greatly enhanced by treatment of the seeds with a combination of 40 mM prohexadione calcium and 80 M uniconazole. The minimum detectable doses of 3-hydroxylated GAs (GA1, GA3, GA4 and GA7) in the three cultivars treated with both chemicals were 1 to 10 fmol (i.e. ca. 350 fg to 3.5 pg) per plant. This is equal to 30-fold more sensitive than Waito-C treated with uniconazole alone, and 30 to 1000-fold more sensitive than Waito-C with no growth retardant soak. Minimum detectable doses of 3-nonhydroxylated GAs (GA9, GA19 GA20) and GAs with very low biological activity (GA8 and GA17) were equal to or more than 1000 fmol per plant. This is about equal to the activity in Waito-C treated with uniconazole alone. Application of this assay to an extract from Raphanus sativus was compared with the data by gas chromatography/mass spectrometry (GC/MS), confirming the conclusions reached using authentic test GAs, namely that use of uniconazole plus BX-112 appreciably enhanced the detection sensitivity to fractions shown by GC/MS to contain GA1 and GA4, both 3-hydroxylated GAs.Abbreviations GA gibberellin - BX-112 prohexadione calcium  相似文献

Picrorhiza kurrooa Royle ex Benth., a high value medicinal herb of alpine Himalaya and a source of hepatoprotective picrosides, is listed as ‘endangered’ due to heavy collection from its natural habitat. The present report deals with successful propagation of this species using both conventional and in vitro techniques. Vegetative propagation was achieved by rooting runner cuttings with indole-3-butyric acid (IBA) or α-naphtheleneacetic acid (NAA) treatment before planting. Nearly 87% rooting success was achieved by treatment of cuttings with 50.0 μM IBA. Seeds were given a presoaking treatment with gibberellic acid (GA3), 6-benzylaminopurine (BAP) or a combination of both to influence germination. More than 11-fold improvement in germination was recorded in seeds treated with 250.0 μM GA3. In vitro shoot multiplication was achieved through sprouting of axillary buds using nodal segment. Multiple shoots were formed following culture for 3 weeks on Murashige and Skoog (MS; 1962. Physiologia Plantarum 15: 473–497) medium containing 1.0 μM BAP. Cent percent rooting success, without basal callus formation, was observed when individual microshoots were placed in MS medium supplemented with IBA. The plantlets raised using conventional as well as tissue culture methods were hardened and successfully established in the experimental field located at 2450 m elevation. In addition, strategies have been discussed to encourage cultivation and in situ conservation of this highly valued medicinal herb so as to reduce pressure on its natural populations.  相似文献

Experiments with Grand Rapids lettuce seeds (Lactuca sativa L.) maintained in darkness or irradiated with red light have shown that the inhibition of germination induced by low concentrations of ABA (2, 4, 6 μM) could be overcome by gibberellins (GA3 or GA4). The same results were obtained, although to a lesser extent, under the influence of two out of the four cytokinins tested (K and BAP) for seeds maintained in darkness. To suppress the block induced by higher concentrations of ABA (for example 8 μM), it was necessary to apply a cytokinin (K, BAP, Z or 2iP) and a gibberellin (GA4 or GA3) simultaneously, or a cytokinin following a red light treatment. Experiments conducted in darkness in which ABA (8 μM) was applied together with a cytokinin (BAP) and a gibberellin (GA4) showed that the gibberellin and the cytokinin played similar roles towards each other and towards ABA.  相似文献

Growth of carrot and radish seedlings in nutrient culture was inhibited by pretreatment with three calmodulin inhibitors. There was little selective effect on specific organs, shoots, tap root and fibrous roots over a range of concentrations. Although pretreatment with CaCl2 (0.5 mM) did not affect growth of untreated seedlings, it partially reduced the inhibitory effects of trifluoperazine over the concentration range 0.01–0.05 mM.
