Abstract The reversibility of adhesion of 3 representative strains of oral streptococci from a phosphate-buffered suspension onto 5 different solid substrata was studied.
Streptococcus mitis T9 (surface free energy γb= 39 mJ · m−2). Streptococcus sanguis CH3 (γb= 95 mJ · m−2) and Streptococcus mutans NS (γb= 117 mJ · m−2) were selected on basis of their surface free energy. Solid substrata were employed with a surface free energy γs ranging from 20 mJ · m−2 for polytetrafluorethylene to 109 mJ · m−2 for glass. Bacterial suspensions containing 2.5 × 109 cells per ml were incubated with 2 samples of each substratum. After 1 h the number of adhering bacteria was evaluated on one sample, while the second sample was kept for another hour at a 10-fold lower bacterial concentration. Bacteria with a low surface free energy desorbed only from substrata with a high surface free energy, while bacteria with a high surface free energy desorbed from substrata with a low surface free energy. Thus low energy bacterial strains adhered reversibly to high energy substrata and vice versa. Similar observations were made with polystyrene particles. Calculation of the interfacial free energy of adhesion (Δ F adh) for each bacterial strain as well as for the polystyrene particles showed that a reversible adhesion was associated with a positive Δ F adh, denoting unfavourable adhesion conditions upon a thermodynamic basis.  相似文献

Raw milk from 27 farms was sampled over 6 months for listerias, salmonellas, Yersinia enterocolitica and campylobacters. Total bacterial counts and somatic cell counts were measured. Lactococci, lactobacilli, dextran-producing leuconostocs, Brevibacterium linens , yeasts and moulds, Staphylococcus aureus and other Micrococcaceae, Pseudomonas , coliforms, Escherichia coli , enterococci, Clostridium perfringens and spores of anaerobic lactate-fermenting bacteria were also counted. Pseudomonas (2000 cfu ml−1), lactococci (760 cfu ml−1) and Micrococcaceae (720 cfu ml−1) were the most numerous groups. Lactic acid bacteria were detected in all samples. Coliforms were present in most samples, but 84% of samples had counts <100 cfu ml−1. Staphylococcus aureus was detected in 62% of milks, the average count was 410 cfu ml−1. About 80% of supplies had ≤10 E. coli cfu ml−1 and all samples had 1 Cl. perfringens cfu ml−1. Two of the tested milks were positive for salmonellas (2·9%), four were positive for Listeria monocytogenes (5·8%), 25 for Yersinia enterocolitica (36%) and one for campylobacters (1·4%).  相似文献

The effect of chlorine on β- D- galactosidase activity of sewage bacteria and Escherichia coli was studied. β- D- galactosidase activity of sewage was more resistant to chlorine than faecal coliform cultivability. At low initial dosage (0·05 mg Cl2 l−1) neither cultivability (colony-forming units (cfu)), nor enzyme activity of E. coli suspensions were severely impaired. When initial chlorine concentration was increased to 0·1 mg Cl2 l−1, the cfu number decreased whereas enzyme activity remained high, i.e. the enzyme activity calculated cfu−1 increased. At higher chlorine doses both cfu and enzyme activity were reduced, but non-cultivable cells retained assayable activity after chlorination. Mean values of the enzyme activity calculated cfu−1 decreased when the chlorine dosage was increased from 0·1 to 0·5 mg Cl2 l−1, but were not significantly different ( P > 0·05) for dosages of 0·2–0·7 mg Cl2 l−1. After chlorination, β- D- galactosidase activity of E. coli was less reduced than cfu and direct viable count numbers, but more reduced than 5-cyano-2-3, ditolyl tetrazolium chloride and total cell counts, and the enzyme activity represented an alternative activity parameter of chlorinated samples.  相似文献

Total viable counts, proteolytic, ureolytic, cellulolytic and NH+4 utilizing anaerobic bacteria in caecal contents, soft and hard faeces of rabbits were studied. No significant differences between the bacterial counts from the caecum and soft faeces were observed ( P < 0·05) but there were significant differences between the bacterial counts of the soft and hard faeces ( P < 0·01).  相似文献

The rate of degradation of n -alkanes C12-C18, in petrol (Slovene diesel) in an aqueous system, by free and immobilized Pseudomonas fluorescens in shaking flasks was investigated. Cells were immobilized to a biosupport, Biofix, and a biosorbant, Drizit. Analysis of cellular growth of the free and immobilized bacteria over 8 d of incubation with diesel as the sole carbon source, showed a reduction in the lag phase in the immobilized cultures in comparison to the free system. The free system degraded 52·3% of C12 and 11·6% of C13, but C14-C18 were not degraded. In comparison to the free system and diesel which had not been exposed to experimental conditions (unexposed), the immobilized systems degraded significantly more of C13-C18. Biofix-immobilized cells degraded 14·8% of C12 and an average of 53·5% of C13-C18. Drizit-immobilized cells degraded 24·5% of C12, 52·4% of C13 and an average of 91·2% of C14-C18. This study shows the successful use of immobilized bacteria technology to enhance the degradation of diesel in an aqueous system.  相似文献

Aims:  To evaluate the effectiveness of organic acids and supercritical carbon dioxide (SC-CO2) treatments as well as their combined effect for the reduction of nonpathogenic Escherichia coli and three pathogenic bacteria in fresh pork.
Methods and Results:  The different treatment conditions were as follows: (i) treatment with acetic (1%, 2% or 3%) or lactic acid (1%, 2% or 3%) only, (ii) treatment with SC-CO2 at 12 MPa and 35°C for 30 min only and (iii) treatment with 3% acetic or lactic acid followed by treatment with SC-CO2. Within the same organic acid concentration, the lactic and acetic acid treatments had similar reductions. For the combined treatment of lactic acid and SC-CO2, micro-organism levels were maximally reduced, ranging from 2·10 to 2·60 log CFU cm−2 ( E. coli , 2·58 log CFU cm−2; Listeria monocytogenes , 2·60 log CFU cm−2; Salmonella typhimurium , 2·33 log CFU cm−2; E. coli O157:H7, 2·10 log CFU cm−2).
Conclusions:  The results of this study indicate that the combined treatments of SC-CO2 and organic acids were more effective at destroying foodborne pathogens than the treatments of SC-CO2 or organic acids alone.
Significance and Impact of the Study:  The combination treatment of SC-CO2 and organic acids may be useful in the meat industry to help increase microbial safety.  相似文献

This paper describes a method for testing the effect of various concentrations of SO2 on lactic acid bacteria from ciders. The media and methods were devised to minimize loss of SO2 due to oxidation or binding with carbonyl compounds. Exposure of laboratory or freshly isolated strains to various concentrations of free SO2 at pH 4·0 did not readily kill them even at high concentrations of free SO2 ( c. 150 p/m or 0·97 p/m molecular SO2) yet they were suppressed at low concentrations ( c. 5 p/m or 0·032 p/m molecular SO2). Reducing the pH to 3·4 reaffirmed how much more effective SO2 is against lactic acid bacteria at lower pH levels because more is present as molecular SO2. As a result of this the idea of quoting SO2 values as p/m molecular SO2 is advocated. Addition of hydrogen peroxide or acetaldehyde to a test system containing 142 p/m free SO2 showed that they had a similar effect in nullifying its antimicrobial properties and allowing the test bacteria to grow. There was no indication that acetaldehyde bisulphite was toxic to the test bacteria.  相似文献

Streptococci and actinomyces that initiate colonization of the tooth surface frequently coaggregate with each other as well as with other oral bacteria. These observations have led to the hypothesis that interbacterial adhesion influences spatiotemporal development of plaque. To assess the role of such interactions in oral biofilm formation in vivo, antibodies directed against bacterial surface components that mediate coaggregation interactions were used as direct immunofluorescent probes in conjunction with laser confocal microscopy to determine the distribution and spatial arrangement of bacteria within intact human plaque formed on retrievable enamel chips. In intrageneric coaggregation, streptococci such as Streptococcus gordonii DL1 recognize receptor polysaccharides (RPS) borne on other streptococci such as Streptococcus oralis 34. To define potentially interactive subsets of streptococci in the developing plaque, an antibody against RPS (anti-RPS) was used together with an antibody against S. gordonii DL1 (anti-DL1). These antibodies reacted primarily with single cells in 4-h-old plaque and with mixed-species microcolonies in 8-h-old plaque. Anti-RPS-reactive bacteria frequently formed microcolonies with anti-DL1-reactive bacteria and with other bacteria distinguished by general nucleic acid stains. In intergeneric coaggregation between streptococci and actinomyces, type 2 fimbriae of actinomyces recognize RPS on the streptococci. Cells reactive with antibody against type 2 fimbriae of Actinomyces naeslundii T14V (anti-type-2) were much less frequent than either subset of streptococci. However, bacteria reactive with anti-type-2 were seen in intimate association with anti-RPS-reactive cells. These results are the first direct demonstration of coaggregation-mediated interactions during initial plaque accumulation in vivo. Further, these results demonstrate the spatiotemporal development and prevalence of mixed-species communities in early dental plaque.  相似文献

S ummary : The incidence of the main types of bacteria among 2,856 isolated on Yeastrel-milk agar incubated at 30° from 121 rinses of dairy equipment cleansed by chemical methods, showed that micrococci were dominant in low colony count (<104/ft2) rinses of equipment treated by immersion cleaning in caustic soda or cleansed in detergent-hypochlorite solutions, whereas nonpigmented Gram-negative rods were dominant in low count rinses of equipment cleansed with detergent-sterilizers containing quaternary ammonium compounds. Gram-negative rods were dominant and streptococci frequent in high count (<2·5 × 105/ft2) rinses irrespective of the chemical used. A high proportion of the streptococci and coli-aerogenes organisms gave acid reactions in litmus milk after 72 h at 22°, many of the anaerogenic Gram-negative rods and several of the aerobic sporeformers gave proteolytic reactions, whereas the majority of the micrococci, corynebacteria and other asporogenous Gram-positive rods showed no change. Bacteria isolated from high count rinses were more active in producing milk spoilage reactions at 22° than those isolated from low count rinses.  相似文献

The degradation and fermentation of microcrystalline cellulose were studied in monoculture of the polycentric anaerobic fungus Orpinomyces joyonii and in co-cultures with the rumen bacteria Megasphaera elsdenii and Eubacterium limosum. More than 25% of cellulose hydrolysis products (glucose and cellodextrins) were released by the fungus into the medium after 8 d of cultivation. These products were metabolized by bacteria in mixed cultures. In co-culture with the fungus M. elsdenii and E. limosum . increased the extent of microcrystalline cellulose degradation by 10·12% and 7·96%, respectively. Biomass yield in co-cultures was increased by 89·9% and 59·4% for M. elsdenii and E. limosum . Ycellulose for fungus alone was 52·29 g dry matter mol-1 glucose. These values were 64·93 and 55·92 g mol-1 glucose unit in co-culture with M. elsdenii and E. limosum , respectively.  相似文献

The sludge from hospital waste treatment facilities is a potential source of infectious organisms. The average numbers of micro-organisms in the sludge of hospital wastewater in Taiwan were as follows: total count 8·1 × 107 cfu g−1 (dry weight of sludge), and 1·4 × 106, 3·6 × 105, 1·6 × 105, 2·2 × 105 and 5·5 × 104 cfu g−1 (dry weight of sludge) for total coliforms, faecal coliforms, faecal streptococci, Pseudomonas aeruginosa and Salmonella spp., respectively . Salmonella spp. were detected in 37% (10 of 27) of the sludges from hospital wastewaters. Therefore, the treatment of such sludge to reduce pathogenic micro-organisms should be considered.  相似文献

Survival of Salmonella typhi was investigated in an anaerobic digester for cattle dung with volatile fatty acid (VFA) levels of 5000 mg l−1 and pH 6·0. The organism was added to the digester only once in the first experiment and daily in the other. Survival was monitored on alternate days. In the single dose experiment, the counts of Salm. typhi declined rapidly and the pathogen was completely eliminated within 12 d in the experimental digester (VFA ca 5000 mg l−1 and pH 6·0), whereas 26 d were required in the control digester (VFA ca 100 mg l−1 and pH 6·8). T 90 values for the experimental and control digesters were 2·44 d and 4·80 d, respectively. In the daily dose experiment, a four log reduction in the pathogen count was observed in the experimental digester, but only a two log reduction in the control digester at the end of the experimental period. The mean T 90 values for the experimental and the control digester were 4·22 d and 18·63 d, respectively. In both the experiments, statistical analysis of the data showed significant differences in the survival pattern of Salm. typhi in the two digesters.  相似文献

Slurry storage tanks on Lancashire farms were sampled in May, June, October and November. There was a seasonal pattern in the number of thermophilic campylobacters. The average log10 MPN per gram fresh weight (log10 MPN gfw−1) in stored samples in May and June was 0·78 ( S.D. 0·71) compared to 2·07 ( S.D. 0·70) in November and December. Campylobacters were readily detected in samples of mature slurry and composted bedding that farmers were about to put to land. The survival of campylobacters in slurry sprayed on land was studied in two seasons. In June, on farm A, stored slurry was mechanically aerated for 48 h and very low numbers of campylobacters (0·9 log10 MPN gfw−1) remained in the slurry before it was sprayed onto land. They became undetectable within 24 h once sprayed on land. In contrast, campylobacters in matured but unaerated slurry that was sprayed onto land on farm B in February were still detectable in samples taken 5 d after application to land, dropping from 2·11 log10 MPN gfw−1–1·37, a decline of only 0·74 log10 MPN gfw−1 in the first 5 d after application.  相似文献

When the Adriatic sturgeon Acipenser naccarii was exposed to progressive hypoxia under static conditions, it exhibited a linear decline in O2 uptake, behaving as an 'oxyconformer'. When, however, it was allowed to swim at a low sustained speed, it could regulate O2 uptake down to a mean ± s . e . critical ( P crit) of 4·9 ± 0·5 kPa ( n = 6). At moderate levels of hypoxia, static fish exhibited significant reductions in arterial blood O2 content, and increases in plasma lactate, which were not observed in swimming animals.  相似文献

Aims:  The aim of this study was to enrich, characterize and identify strict anaerobic extreme thermophilic hydrogen (H2) producers from digested household solid wastes.
Methods and Results:  A strict anaerobic extreme thermophilic H2 producing bacterial culture was enriched from a lab-scale digester treating household wastes at 70°C. The enriched mixed culture consisted of two rod-shaped bacterial members growing at an optimal temperature of 80°C and an optimal pH 8·1. The culture was able to utilize glucose, galactose, mannose, xylose, arabinose, maltose, sucrose, pyruvate and glycerol as carbon sources. Growth on glucose produced acetate, H2 and carbon dioxide. Maximal H2 production rate on glucose was 1·1 mmol l−1 h−1 with a maximum H2 yield of 1·9 mole H2 per mole glucose. 16S ribosomal DNA clone library analyses showed that the culture members were phylogenetically affiliated to the genera Bacillus and Clostridium. Relative abundance of the culture members, assessed by fluorescence in situ hybridization, were 87 ± 5% and 13 ± 5% for Bacillus and Clostridium , respectively.
Conclusions:  An extreme thermophilic, strict anaerobic, mixed microbial culture with H2-producing potential was enriched from digested household wastes.
Significance and Impact of the Study:  This study provided a culture with a potential to be applied in reactor systems for extreme thermophilic H2 production from complex organic wastes.  相似文献

Abstract Actinomyces viscosus is a predominant microorganism in dental plaque. It is, just as the oral Streptococcus spp., a saccharolytic and aero-tolerant organism. We have investigated the effects of oxygen on the growth and metabolism of A. viscosus . To this end A. viscosus Ut 2 was grown in a glucose limited chemostat culture on a chemically defined medium ( D = 0.2 h−1) with exposure to variable amounts of oxygen. The Yglucose increased from 62.5 g · mol−1 under anaerobic conditions to 149 g · mol−1 under aerobic conditions, while, concomitantly, the carbon recovery from acidic fermentation products decreased from 75% to 7%. Addition of [14C]glucose to the chemostat showed that the glucose, which was not converted to acidic fermentation products, was instead converted to carbon dioxide or used for the production of biomass. Under aerobic and anaerobic conditions identical cytochrome spectra, containing only two cytochrome b -type absorption bands, were found. It was concluded that electron transport phosphorylation probably occurs both under aerobic and anaerobic conditions. Anaerobically, fumarate served as the electron acceptor, while the high growth yields observed under aerobic conditions are likely to be explained by citric acid cycle activity coupled to electron transport phosphorylation.  相似文献

Control of seed development in Arabidopsis thaliana by atmospheric oxygen   总被引:2,自引:1,他引:1  
Seed development is known to be inhibited completely when plants are grown in oxygen concentrations below 5·1 kPa, but apart from reports of decreased seed weight little is known about embryogenesis at subambient oxygen concentrations above this critical level. Arabidopsis thaliana (L.) Heynh. plants were grown full term under continuous light in premixed atmospheres with oxygen partial pressures of 2·5, 5·1, 10·1, 16·2 and 21·3 kPa O2, 0·035 kPa CO2 and the balance nitrogen. Seeds were harvested for germination tests and microscopy when siliques had yellowed. Seed germination was depressed in O2 treatments below 16·2 kPa, and seeds from plants grown in 2·5 kPa O2 did not germinate at all. Fewer than 25% of the seeds from plants grown in 5·1 kPa oxygen germinated and most of the seedlings appeared abnormal. Light and scanning electron microscopic observation of non-germinated seeds showed that these embryos had stopped growing at different developmental stages depending upon the prevailing oxygen level. Embryos stopped growing at the heart-shaped to linear cotyledon stage in 5·1 kPa O2, at around the curled cotyledon stage in 10·1 kPa O2, and at the premature stage in 16·2 kPa O2. Globular and heart-shaped embryos were observed in sectioned seeds from plants grown in 2·5 kPa O2. Tissue degeneration caused by cell autolysis and changes in cell structure were observed in cotyledons and radicles. Transmission electron microscopy of mature seeds showed that storage substances, such as protein bodies, were reduced in subambient oxygen treatments. The results demonstrate control of embryo development by oxygen in Arabidopsis .  相似文献

The control of a thermophilic continuous anaerobic digestion system when subjected to potential inhibitory shock loadings was achieved through the regulation of dissolved H2, monitored using membrane inlet mass spectrometry, by the controlled addition of carbon source. At a feed pump switching threshold equivalent to 1 μmol/1 H2 a steady state rate of methanogenesis of approximately 40 μmol/1/min was obtained. Higher H2 thresholds resulted in an inhibition of methanogenesis, but precise control of H2 concentration was demonstrated with an oscillatory response of period 2·5–5·0 min.  相似文献

ABSTRACT. Strombidium purpureum Kahl, is an anaerobic oligotrichous ciliate with endosymbiotic phototrophic bacteria. Like other anaerobic ciliates, S. purpureum reacts to O2-pressure. In the light, the ciliates avoid even traces of O2 (< 1% atmospheric saturation). In the dark, however, the ciliates accumulate in water with a pO2 of 1–4% atmospheric saturation. Experiments show that ciliates which have accumulated in the dark under microaerophilic conditions react when the light is turned on, and their ability to escape is enhanced by steep O2-gradients. The ciliates orient themselves in O2-gradients by a series of phobic responses. They tumble whenever they swim towards a higher O2-tension and thus they eventually all swim in the direction of lower pO2. The ecological implications of such behaviour are discussed.  相似文献

The genus Pectinatus has been often reported in beer spoilage with off-flavours. The bacteria are strictly anaerobic, Gram-negative rods. Propionate and acetate are the main fermentation products from glucose in the two species belonging to the genus, P. cerevisiiphilus and P. frisingensis. Amino acids routinely present at a high level in beer were not growth substrates for both species, and a significant accumulation of succinate was observed with lactate as growth substrate. Both Pectinatus ssp. showed almost identical fermentation balances on glucose. Growth kinetics of both glucose-grown species were unchanged under a N2, H2 or 20% CO2-containing atmosphere. Combinations of culture medium pH values from pH 3·9 to pH 7·2, of glucose levels between 5 and 55 mmol l-1, and of lactate concentrations varied from 4 to 40 mmol l-1 demonstrated that biomass and volatile fatty acids production were proportional to glucose concentration for both Pectinatus species. A significant increase of volatile fatty acid production was measured for both species at the lowest pH values with a lactate or a glucose concentration increase. The maximum biomass production was observed at pH 6·2 for P. cerevisiiphilus , and between pH 4·5 and pH 4·9 for P. frisingensis. Glucose and lactate or pH value were dependent with regard to propionate and acetate production in P. frisingensis. On the other hand, the variations of these three parameters were independent with regard to biomass production for both strains, and to volatile fatty acids production for P. cerevisiiphilus. Addition of ethanol to glucose-grown cultures completely inhibited growth at 1·3 mol l-1 ethanol for P. cerevisiiphilus , and at 1·8 mol l-1 for P. frisingensis.  相似文献
