SCE频率分析是研究环境诱变剂和致癌剂引起人类遗传物质损伤的有效手段。以人类外周血林巴细胞姊妹染色单体交换(SCE)为指标,评价了香烟烟雾水溶液的遗传毒性。首先,定量测定了香烟烟雾水溶液中含有H2O2、O2-等活性氧,从而间接证明了其潜在的遗传毒性。然后,将一定量的香烟烟雾水溶液加入人外周血淋巴细胞培养液中,结果表明实验组SCE频率明显高于空白对照组,因此直接证明了香烟烟雾水溶液的遗传毒性。  相似文献

应用IdU-毛玉米油体内SCE技术,以不同剂量的典型诱变剂MMC和CP对70尾黄鳝的脾、肾、血淋巴细胞进行了体内诱发SCE敏感性测试。结果:三种细胞的染色体SCE自发频率均较低,不同剂量MMC和CP诱发黄鳝三种细胞SCE频率均较对照组显著增加。诱变剂剂量与诱发SCE频率呈线性关系。三种细胞染色体SCE对MMC和CP的敏感性次序为肾>脾>血淋巴细胞。与几种鱼和其它动物比较,黄鳝三种细胞的SCE自发频率均较低,对MMC和CP诱发SCE的敏感性均较高,因此认为黄鳝可作为较理想的体内SCE检测系统。  相似文献

草鱼体内肾细胞姐妹染色单体分化及交换的初步研究   总被引:2,自引:0,他引:2  
本文确立了一个以草鱼体内肾细胞姐妹染色单体交换频率为指标的检测环境诱变或致癌物质的短期试验系统。采用硫堇-UV-Giemsa染色法,分析了草鱼体内肾细胞的SCD-2(注射BrdU后第二个细胞周期的中期分裂相的SCD)频率和SCE频率。用500微克/克体重BrdU体内标记5天,草鱼肾细胞SCD-2频率为8.58±0.22%;SCE频率为3.05±2.523 SCE_5/细胞。以丝裂霉素C(Mitomycin C,MMC)作为阳性对照,分析了化合物亚硝基胍(N-methyl-N~1-nitro-N-nitrosoguanidine,MNNG)和农药叶蝉散(Mipc)诱发SCE的能力。这项工作在评价水质污染方面将起一定的作用。  相似文献

姐妹染色单体交换(sister-chromatid exchange,SCE)是当前生物学研究的热点之一。SCE检测技术已广泛地应用于环境科学、医学、生物学等研究领域,具有重要的意义。环境胁迫会造成细胞内DNA不同程度的损伤,并可能进一步导致染色体畸变,甚至引发癌变。SCE作为一种灵敏而有效的指标,能够反映DNA损伤程度和遗传不稳定性。本文主要介绍了有关SCE的由来,色差显示原理、分子机理、SCE与染色体畸变的关系以及SCE的诱导因子。文章最后还对今后有关SCE的研究及其应用提出一些新的看法。  相似文献

目的观察不同胎牛血清对百日咳毒素在中华仓鼠卵巢细胞(Chinese hamster ovary cell,CHO)细胞簇聚试验中的影响。方法分别用两个厂家共6批次的牛血清培养CHO细胞,连续传3代后进行细胞簇聚试验,观察添加PT阳性对照、纯化PT、脱毒PT后其细胞的簇聚效果。结果 1、2号牛血清培养的CHO细胞生长缓慢,3~6号牛血清培养的CHO细胞生长正常。质量浓度16 ng/m L的PT阳性对照和纯化PT均未引起1~2号CHO细胞簇聚;3~6号牛血清培养的CHO细胞PT阳性对照判定终点分别为2、1、16、4 ng/m L,其纯化PT判定终点分别为1、0.5、8、2 ng/m L;脱毒PT质量浓度为40μg/m L时,1、2、5号牛血清培养的CHO不簇聚,而3、4、6号牛血清培养的CHO细胞脱毒PT判定终点分别为10、5、20μg/m L。结论 6种牛血清培养的CHO细胞簇聚程度存在差异,其中以4号牛血清培养的CHO细胞对PT阳性对照、纯化PT和脱毒PT最为敏感。需筛选对簇聚试验敏感度高的牛血清用于百日咳毒素CHO细胞簇集试验。  相似文献

目的:研究石油作业环境对女性习惯性流产的遗传毒性。方法:随机选择习惯性流产的石油作业女性38人和正常的育龄女性20人,检测其外周血淋巴细胞姐妹染色单体互换,记数SCE发生率。结果:观察组的外周血淋巴细胞SCE发生率为8.81±0.35,明显高于对照组(P0.05)。结论:SCE的发生可作为石油作业习惯性流产女性染色体结构稳定性的检测指标。石油作业环境对女性生育具有遗传毒性。  相似文献

本文通过对男性、女性成年人和儿童的外周血淋巴细胞的染色体畸变率、染色体畸变类型性的染色体畸变分析,微核,姐妹染色单体交换,及细胞有丝分裂指数、细胞周期比率、细胞增值指数的细胞动力学研究等8项参数、综合分析年龄和性别对其遗传安全性检测指标的影响。结果8项参数在成年人中均显示男性高于女性的趋势;除细胞畸变率外,其他7项参数在男性和女性中均显示成年人高于儿童的趋势。由本试验初步认为,年龄和性别对遗传安全性检测指标存在一定影响。  相似文献

目的探讨野菊花总黄酮(total fl avonoids of Chrysanthemu m,TFC)清除β淀粉样蛋白的作用和分子机制。方法采用CCK-8法观察不同浓度TFC对SH-SY5Y神经细胞株增殖的影响,ELISA法检测TFC对转染过表达淀粉样前体蛋白(APP)和淀粉蛋白前β位分解酶1(BACE1)的CHO细胞培养上清液中Aβ水平的影响,MDC染色检测TFC处理后的神经元内自噬小体的形成,Western blot检测自噬蛋白LC3表达水平。结果 CCK-8法分析显示,TFC对SH-SY5Y神经元活力无明显影响;ELISA检测显示,TFC处理使过表达APP和BACE1的CHO细胞培养上清液中Aβ水平呈浓度依赖性降低;荧光倒置显微镜下观察和Western blot检测发现,TFC处理后SH-SY5Y细胞自噬增加,LC3/LC3自噬标志物蛋白表达增强。结论 TFC可能通过增强细胞自噬而促进Aβ的清除,减少Aβ细胞毒性。  相似文献

用BrdU标记,改良的FPG法染色,建立了一种植物愈伤组织细胞姐妹染色单体分染的方法。并对培养基的不同附加成分对SCE的影响进行了研究。所有培养基上愈伤组织细胞的SCE率都显著高于正常根尖分生组织细胞。6-BA,AgNO_3,高浓度的2,4-D,蔗糖均可诱发SCE。  相似文献

用限制性内切酶PstⅠ,SalⅠ,PvuⅡ和BamHⅠ处理CHO细胞后,发现其SCE率升高,与对照相比,前三种酶具有显著性差异。但这些酶诱导SCE的效应与其致染色体畸变效应相比则较弱,提示引起DNA双链断裂的限制性内切酶不是SCE的强刺激物。实验结果表明,BrdU取代胸苷不能消除限制酶对底物DNA的识别及裂解。  相似文献

A new permanent staining procedure for sister chromatid differentiation (SCD) in cultured Chinese hamster ovary cells has been established by combining the three-way differentiation in third mitosis (M3) chromosomes and the immunoperoxidase reaction developed with 3,3'-diaminobenzidine using a monoclonal anti-bromodeoxyuridine (BrdU) antibody. This procedure allows SCD at very low BrdU concentrations, and the evaluation of the sister chromatid exchange frequencies on a per cell-cycle basis.  相似文献

For staining in toto, planarians are fixed in a mixture of 10 ml of commercial formalin, 45 ml of 95% ethanol and 2 ml of glacial acetic acid. After treatment with 70% ethanol 3-10 days, they are washed in distilled water and immersed in 10% CuSO4. 5H2O for 3 hr at 50° C, transferred without washing to 1% AgNO3 for 1.0-1.5 hr at 50° C; and then developed in: 10 ml of 1% pyrogallol, 100 ml of 56% ethanol and 1 ml of 0.2% nitric acid. Gold toning, 5% Na2S2O3 and dehydration follow as usual. For staining sections, material is fixed in the same fixative, embedded in paraffin and sectioned at 10 μ. After bringing sections to water, they are immersed in 20% CuSO4. 5H2O for 48 hr at 37° C; then rinsed briefly in distilled water and placed in 7% AgNO3 for 24 hr at 37° C. They are washed briefly in distilled water and reduced in: hydroquincne, 1 gm; Na2SO3, 5 gm and distilled water 100 ml. Gold toning, followed by 5% Na2S2O3 and dehydration completes the process. Sister chromatid exchange (SCE) is a sensitive indicator of genotoxicity. In this study we investigated the effects of alcohol consumption and cigarette smoking on the frequency of SCE in cultures of peripheral lymphocytes. The rate was higher in alcoholics who smoked (10.89+/-2.46) and in smokers (positive controls) (7.64+/-1.01) than in healthy non-smokers (negative controls) (6.96+/-2.18). Statistical analysis suggested that the increases were related to alcohol consumption and cigarette smoking (p<0.05).  相似文献

Sodium selenite (Na2Se03) was tested for its sister-chromatid exchange (SCE)-inducing ability in human whole blood cultures and for the effect of its co-exposure with methyl methanesulfonate (MMS) or N-hydroxy-2-acetyl aminofluorene (N-OH-AAF) on SCE frequency. Long exposure times (77 h and 96 h) to 3.95 × 10-6 M Na2SeO3 resulted in cell death as measured by mitotic indices, but mitotic figures were present after exposure to higher concentrations for a shorter time (19 h). High Na2SeO3 concentrations (7.90 × 10?6 and 1.19 × 10?5 M) resulted in a three-fold increase in the SCE frequency above background level (6–7 SCEs/cell). Exposure of lymphocytes to 1 × 10?4 M MMS for the last 19 h of culture yielded an average SCE frequency of 30.17 ± 0.75 while a similar exposure to 2.7 × 10?5 M N-OH-AAF resulted in 13.61 ± 0.43 SCEs/cell. Simultaneous addition of the high Na2Se03 concentrations and MMS or N-OH-AAF to the cultures resulted in SCE frequencies that were 25–30% and 11–17%, respectively, below the sum of the SCE frequencies produced by the individual compounds.  相似文献

Imidacloprid and metalaxyl are two pesticides that are widely used in agriculture, either separately, or in combination. These agents were studied for their possible genotoxic effects with respect to the following cytogenetic end-points: (1) in vitro micronucleus (MN) formation and sister-chromatid exchange (SCE) induction in human lymphocytes and (2) in vivo micronucleus induction in polychromatic erythrocytes (PCEs) of the rat bone-marrow. The results of the MN analysis indicate that MN frequencies after treatment with both pesticides, separately or as a mixture, do not significantly differ from those in the controls except after treatment with metalaxyl alone at 50 μg/ml (p < 0.05). The results of the SCE analysis show that SCE frequencies after treatment with imidacloprid do not differ significantly from those in the controls. A statistically significant increase (p < 0.05) in SCE frequency resulted from treatments with metalaxyl at 5, 10 and 100 μg/ml and with the combination of imidacloprid and metalaxyl at 100 and 200 μg/ml. Finally, the in vivo micronucleus assay with rat bone-marrow polychromatic erythrocytes showed a statistically significant effect upon separate treatments with imidacloprid and metalaxyl at doses of 300 mg/kg body weight (b.w.) (p < 0.01) or upon combined treatment with 200 mg/Kg b.w. (p < 0.001) and 400 mg/kg b.w. (p < 0.05).  相似文献
