Pollination greatly accelerates petal senescence. The first observed event after pollination is an increase in the flower's sensitivity to ethylene, followed by an increase in ethylene biosynthesis. Our objectives were to study the mode of action of the increase in ethylene sensitivity and the possible involvement of short-chain saturated fatty acids (SCSFAs) in this process. Application of SCSFAs, ranging in chain length from 7 to 10 carbons onto stigmas of Phalaenopsis (Phalaenopsis hybrid, cv. Herbert Hager) flowers increased their sensitivity to ethylene in the same way as pollination. Following pollination, there was a significant increase in the endogenous content of these fatty acids in the flower's column and perianth, with octanoic acid (C8) being the main SCSFA observed. The increase in SCSFA content was observed as early as 6 h after pollination and began to decline 6 h later. Incorporation of octanoic acid into liposomes or microsomal membranes isolated from Phalaenopsis petals resulted in a decrease in lipid order that was detected by fluorescence polarization of dansyl pyrrolidine (DNSP) but not of 1,6-diphenyl-1,3,5-hexatriene (DPH). At peak ethylene sensitivity, 10 h after pollination, there was a significant decrease in the lipid order of microsomal membranes isolated from Phalaenopsis columns and perianths, again as detected by DNSP but not by DPH. Stigmatic application of octanoic acid mimicked the effect of pollination on membrane lipid order. We suggest that SCSFAs may be the ethylene 'sensitivity factors' produced following pollination, and that their mode of action involves a decrease in the order of specific regions in the membrane lipid bilayer, consequently altering ethylene action.  相似文献

The effect of short-chain saturated fatty acids (C5–C10) on the biosynthesis of ethylene in embryonic axes of chick-pea ( Cicer arietinum L.) seeds was investigated. The emergence of radicle and fresh weight of embryonic axes diminished with increasing number of carbons. The inhibition of germination caused by lower concentrations (1 m M ) of fatty acids (C5–C10) was partially reversed by exogenous 1-aminocyclopropane-1-carboxylic acid (ACC), whereas exogenous ethylene was able to overcome the inhibitory effect provoked by all concentrations (1–5 m M ) of applied fatty acids (C5–C10). Ethylene production rates, and enzyme activities of ACC synthase and ACC oxidase decreased concomitantly with the molecular mass and increasing concentration of fatty acids. The inhibitory effect of these acids on ethylene production seems to result not only from a decreased ACC synthesis, but also from an enhancement of 1-malonylamino)cyclopropane-1-carboxylic acid (MACC) synthesis.  相似文献

Normal and pollination-induced senescence of Petunia hybrida L cv. Pink Cascade flowers is accompanied by an increase in the sensitivity of the corolla to ethylene as indicated by an acceleration in the rate of corolla bluing after exposure to exogenous ethylene. Pollination resulted in the production of short-chain saturated fatty acids ranging in chain length from C6 to C10. Following pollination, these acids are synthesized in the stylar tissue via the acetate pathway within the first 12 hours. The fatty acids are transported rapidly to the corolla where they induce an increase in ethylene sensitivity. In unpollinated flowers, these acids are produced in the corolla during the early stages of senescence. Although the levels of these fatty acids decrease rapidly during the final stages of senescence, a significant increase in ethylene sensitivity could be detected prior to the decrease. It appears that the increase in ethylene sensitivity caused by the synthesis of short-chain saturated fatty acids occurs concurrently, but independent from ethylene synthesis.  相似文献

Abstract In this study the fatty acid composition of Flavobacterium halmephilum CCM2831 grown at different temperatures and salt concentrations is reported. At elevated growth temperatures the amount of cellular saturated long-chain acids (C18:0 and C20:0) and the branched-chain acid br-C17:0 increased, and the concentrations of both cyclic acids and the branched chain acid br-C15:0 decreased. Increasing the salt concentration in the medium resulted in a gradual increase in cellular cyclopropanoic acids and a concomitant decrease in the monounsaturated fatty acid content.  相似文献

Pollination induced an immediate increase in ethylene production in Dianthus caryophyllus and Petunia hybrida. In Cymbidium, a lag of several hours was observed. In all three species, pollination induced premature flower senescence. Treatment of the stigmatic surface with aminoethoxyvinylglycine prior to pollination effectively blocked the increase in ethylene production and alleviated the detrimental effect of pollination on flower life.In all three tested species, octanoic and decanoic acids, when applied to the stigmatic surface, had no effect on ethylene production and flower life. In isolated Cymbidium lips placed with their cut base in solutions containing these fatty acids, no effects on red colouration, ethylene production, and ethylene forming enzyme activity were observed. In addition, ethylene sensitivity of isolated lips was not affected. The putative regulatory role of short-chain saturated fatty acids in (pollination-induced) flower senescence is discussed.  相似文献

The inhibitory effects of the cyclic olefin 2,5-norbornadiene (NBD) on ethylene action were tested in carnation (Dianthus caryophyllus L. cv White Sim) flowers. Treatment of flowers at anthesis with ethylene in the presence of 500 microliters per liter NBD increased the concentration of ethylene required to elicit a response (petal senescence), indicating that NBD behaves as a competitive inhibitor of ethylene action. Transfer of flowers producing autocatalytic ethylene and exhibiting evidence of senescence (petal in-rolling) to an atmosphere of NBD resulted in a rapid reduction in ethylene production, petal 1-aminocyclopropane-1-carboxylic acid synthase activity, 1-aminocyclopropane-1-carboxylic acid content, and ethylene forming enzyme activity. Removal of NBD resulted in recovery of ethylene biosynthesis. These results support the autocatalytic regulation of ethylene production during the climacteric stage of petal senescence and suggest that continued perception of ethylene is required for maintenance of ethylene biosynthesis. The inhibition of ethylene action by NBD after the flowers had reached the climacteric peak was associated with interruption of petal senescence as evidenced by reversal of senescence symptoms. This result is in contrast to the widely held belief that the rate of petal senescence is fixed and irreversible once petals enter into the ethylene climacteric.  相似文献

The fatty acid compositions for 40 strains of moderately halophilic Gram-negative rods were determined by gas-liquid chromatography. The strains studied were included in the genera: Vibrio, Deleya, Alteromonas, Chromobacterium, Flavobac-terium and Pseudomonas. Although there were quantitative differences all strains showed more or less similar spectra of fatty acids ranging from C12 to C20 chain. The major fatty acid species were C16:0, C16:1 and C18:1. Most striking was the predominance of the C18:1 component, the major fatty acid in extracts of 29 of the 40 strains.  相似文献

The fatty acid composition of the total lipids from two Conidiobolus species was studied by gas—liquid chromatography. The major fatty acids of C. lamprauges were palmitic acid (C16:0), oleic acid (C18:1), linolenic acid (C18:3), and arachidonic acid (C20:4). For C. eurymitus , myristic acid (C14:0), C16:0, and linoleic acid (C18:2) were the most abundant acids. The fatty acid composition of C. eurymitus was quite different from that of the Conidiobolus species as mentioned in other reports. The lipid composition of the total lipids of C. lamprauges and C. eurymitus was also studied by thinchrography on quartz rods. Triglycerides and phospholipids were the major components in the two Conidiobolus species.  相似文献

Application of linoleic and linolenic acids to Phalaenopsis and Dendrobium flowers enhanced their senescence and promoted ethylene production. This effect was specific to unsaturated fatty acids which serve as substrates for lipoxygenase action, and did not occur following similar treatments with saturated fatty acids. Several major lipoxygenase pathway metabolites including jasmonic acid methyl ester, traumatic acid, trans -2-hexenal and cis -3-hexenol also enhanced flower senescence. Jasmonic acid methyl ester promoted ethylene production by Phalaenopsis flowers. In contrast, treating flowers with the lipoxygenase inhibitors salicylhydroxamic acid and n -propyl gallate. which inhibite(d) lipoxygenase activity in vitro, had no effect on pollination-induced senescence of the flowers. Furthermore, during the 50-h period following pollination, there was no increase in lipoxygenase activity in Phalaenopsis flowers. During the 10-h period from pollination of Dendrobium flowers until the initiation of ethylene production, there was no effect of pollination on jasmonate levels in either the perianth or the columns. These results suggest that lipoxygenase activity and jasmonates are not directly involved in pollination-induced Phalaenopsis and Dendrobium flower senescence.  相似文献

Factors accelerating flower senescence of Portulaca hybrid were investigated. Self‐and cross‐pollination, emasculation and removing of pistil significantly accelerated senescence. However, wounding of filaments was much more effective in accelerating flower senescence. Senescence was further accelerated by an increase in the number of wounded filaments, and ethylene production was also accelerated by wounding of filaments. Treatment with 0.1 µl 1−1 ethylene for 1 h significantly accelerated flower senescence, and the senescence of both the intact and filament‐wounded flowers was markedly delayed by exposure to norbornadiene (NBD), an inhibitor of ethylene action. The sensitivity to ethylene increased significantly within 1 h after wounding of filaments, but ethylene production did not. These results suggest that acceleration of flower senescence by wounding of filaments is caused by an increase in the sensitivity to ethylene and the subsequent production of ethylene.  相似文献

Abstract: We have devised an analytical method for the determination of fatty acid composition of erythrocyte membrane sphingomyelin by chemical ionization mass spectrometry combined with capillary column gas-liquid chromatography. Fatty acid composition of erythrocyte membrane sphingomyelin from 8 patients with adrenoleukodystrophy (ALD) and 16 healthy controls were examined by this method. The ratio of hexacosanoic acid (C26.0) to docosanoic acid (C22:0) in erythrocyte membrane sphingomyelin from ALD patients was 2.6-fold higher than that of the controls. This result suggests that biochemical diagnosis of ALD is possible by the analysis of fatty acid composition of erythrocyte membrane sphingomyelin. Furthermore, it demonstrates that biochemical abnormality in ALD is the generalized abnormal metabolism of very long-chain saturated fatty acids.  相似文献

The fatty acid composition from mycelia of Streptomyces hygroscopicus strains was studied. A significant proportion of C18 : 2 was found in cultures. High levels of C16 : 0, iso-C16 : 0 and C18 : 1 were also detected in all S. hygroscopicus strains. The different representatives of S. hygroscopicus had almost the same proportion of unsaturated fatty acids. Certain shifts in the amount of iso, anteiso and straight-chain fatty acids in some cultures were revealed. This might be explained by the adaptation capability of strains belonging to one species to form a variety of available fatty acids determined by particular cell membrane composition favouring certain antibiotic biosynthesis.  相似文献

Changes in water status, membrane permeability, ethylene production and levels of abscisic acid (ABA) were measured during senescence of cut carnation flowers ( Dianthus caryophyllus L. cv. White Sim) in order to clarify the temporal sequence of physiological events during this post-harvest period. Ethylene production and ABA content of the petal tissue rose essentially in parallel during natural senescence and after treatment of young flowers with exogenous ethylene, indicating that their syntheses are not widely separated in time. However, solute leakage, reflecting membrane deterioration, was apparent well before the natural rise in ethylene and ABA had begun. In addition, there were marked changes in water status of the tissue, including losses in water potential (ψw), and turgor (ψp), that preceded the rise in ABA and ethylene. As senescence progressed, ψw continued to decline, but ψp returned to normal levels. These temporal relationships were less well resolved when senescence of young flowers was induced by treatment with ethylene, presumably because the time-scale had been shortened. Thus changes in membrane permeability and an associated water stress in petal tissue appear to be earlier symptoms of flower senescence than the rises in ABA or ethylene. These observations support the contention that the climacteric-like rise in ethylene production is not the initial or primary event of senescence and that the rise in ABA titre may simply be a response to changes in water status.  相似文献

Abstract— The fatty acid composition of cerebrosides, sulphatides and ceramides has been determined at 20 days postpartum in the brains of Quaking mutant mice and of littermate controls. There was a significant deficit in the proportion of long-chain fatty acids (C22-C24) affecting both normal and a-hydroxy fatty acids of the cerebrosides. The proportion of normal but not the a-hydroxy long-chain fatty acids of the sulphatides was also decreased. Striking and disproportionate deficits of the C24:1 and C24 h:1 fatty acids of cerebrosides, sulphatides and ceramides characterized the brain of the Quaking mutant, and an increased proportion of C23 h:O fatty acid was found in the cerebrosides and sulphatides of the brain of this mutant. We compared these data with findings on the Jimpy mutant which has been examined by the same techniques. The deficiency of long-chain fatty acids which was found in the cerebrosides and sulphatides of both mutants was less extensive but more selective in the Quaking mutant.  相似文献

Temperature-dependent compositional changes of phospholipids and their fatty acids were analysed in Yersinia enterocolitica grown at 5°, 25° and 37°C. The relative amounts of the four phospholipids, phosphatidylethanolamine (75–78%), phosphatidylglycerol (10–11%), cardiolipin (<7%) and lysophosphatidylethanolamine (<5%), were essentially the same at all growth temperatures. The degree of fatty acid unsaturation of the four phospholipids increased with decrease in growth temperature, mainly due to an increase of C16:1 and C18:1 and a corresponding decrease of C16;0, C18:0 and cyclo C17:0. An electron spin resonance spectroscopic study of the membrane lipids showed that membrane lipid fluidity was enhanced by decreasing the growth temperatures. The changes in fatty acid composition of phospholipids in response to varied temperatures were consistent with the temperature-dependent changes in the membrane lipid fluidity of Y. enterocolitica , and were similar to those reported for other bacteria.  相似文献

Arthrobacter nicotianae KCC B35 isolated from blue-green mats densely covering oil sediments along the Arabian Gulf coast grew well on C10 to C40 n -alkanes as sole sources of carbon and energy. Growth on C20 to C40 alkanes was even better than on C10 to C18 alkanes. Biomass samples incubated for 6 h with n -octacosane (C28) or n -nonacosane (C29) accumulated these compounds as the predominant constituent alkanes of the cell hydrocarbon fractions. The even chain hexadecane C16 and the odd chain pentadecane C15 were the second dominant constituent alkanes in C28 and C29 incubated cells, respectively. n -Hexadecane-incubated cells accumulated in their lipids higher proportions of C16-fatty acids than control cells not incubated with hydrocarbons. On the other hand, C28 and C29-incubated cells did not contain any fatty acids with the equivalent chain lengths, but the fatty acid patterns of the cell lipids suggest that there should have been mid-chain oxidation of these very long chain alkanes. This activity qualifies A. nicotianae KCC B35 to be used in cocktails for bioremediating environments polluted with heavy oil sediments.  相似文献

PELIZAEUS-MERZBACHER DISEASE: BRAIN LIPID AND FATTY ACID COMPOSITION   总被引:2,自引:2,他引:0  
Abstract— Biochemical analysis of the leukodystrophy brain from a case of Pelizaeus-Merzbacher disease, classical type, was performed. A decrease in the amount of solid material present was found. The lyophilized brain weight was reduced to 76% of normal with a slightly greater decrease in the amount of extractable lipid. Total myelin was diminished to 7% of normal. Among specific lipids plasmalogens were present in slightly lowered amounts. Cerebrosides and sulphatides were drastically reduced to 8% of normal, whereas sphingomyelin was less severely affected. Fatty acids from phospholipids were close to normal, only enols being slightly diminished. Analysis of pure cerebrosides and sulphatides revealed that the a-hydroxylated compounds as well as very long chain fatty acids (over C18, especially C23 to C26) were greatly reduced. For chain lengths over C18, the ratio of leukodystrophy fatty acid to normal fatty acid was close to 10%. The defect in very long chain fatty acids is estimated at 99.2% in total brain.
Thus, we have found a marked decrease in the amount of very long chain fatty acids and a less marked decrease in sphingolipids. The reduced amount of these acids appears to be partially offset by an increase in the amount of medium-chain fatty acids in sphingolipids. We conclude that one aspect of Pelizaeus-Merzbacher disease may be a defect in the synthesis of myelin very long chain fatty acids (as these acids are far much reduced than any other myelin molecule).  相似文献

Abstract Type strains of Rhodocyclus purpureus, R. gelatinosus and R. tenuis along with three local isolates of R. gelatinosus were tested for growth in the light on n -fatty acids ranging in chain length from C5 (valerate) to C22 (docosanoate).
R. purpureus , the type species of the genus, was anomalous in its limited ability to grow on n -fatty acids; no fatty acids of chain length greater than C9 (nonanoate) were utilized. R. gelatinosus and R. tenuis , on the other hand, utilized all fatty acids in the range C5 to C18 inclusive. R. gelatinosus showed some restricted ability to use C20 (eicosanoate) and C22 (docosanoate) fatty acids.  相似文献

Plant cuticle is an extracellular lipid-based matrix of cutin and waxes, which covers aerial organs and protects them from many forms of environmental stress. We report here the characterization of CER8 / LACS1 , one of nine Arabidopsis long-chain acyl-CoA synthetases thought to activate acyl chains. Mutations in LACS1 reduced the amount of wax in all chemical classes on the stem and leaf, except in the very long-chain fatty acid (VLCFA) class wherein acids longer than 24 carbons (C24) were elevated more than 155%. The C16 cutin monomers on lacs1 were reduced by 37% and 22%, whereas the C18 monomers were increased by 28% and 20% on stem and leaf, respectively. Amounts of wax and cutin on a lacs1-1 lacs2-3 double mutant were much lower than on either parent, and lacs1-1 lacs2-3 had much higher cuticular permeability than either parent. These additive effects indicate that LACS1 and LACS2 have overlapping functions in both wax and cutin synthesis. We demonstrated that LACS1 has synthetase activity for VLCFAs C20–C30, with highest activity for C30 acids. LACS1 thus appears to function as a very long-chain acyl-CoA synthetase in wax metabolism. Since C16 but not C18 cutin monomers are reduced in lacs1 , and C16 acids are the next most preferred acid (behind C30) by LACS1 in our assays, LACS1 also appears to be important for the incorporation of C16 monomers into cutin polyester. As such, LACS1 defines a functionally novel acyl-CoA synthetase that preferentially modifies both VLCFAs for wax synthesis and long-chain (C16) fatty acids for cutin synthesis.  相似文献

Abstract: The free fatty acid (FFA) composition of peripheral nerve resembles that of erythrocytes but the composition of both is different from that of brain and other tissues. Approximately 75% of FFAs of nerve and erythrocytes are saturated and <5% are polyunsaturated whereas in brain and other tissues, 30-45% of FFAs are saturated and 25-50% are polyunsaturated. Approximately 10-15% of the total FFA of nerve have very long chain lengths [C24, C26, C28, and C30]. The presence of these very long-chain FFAs in endoneurium cannot be accounted for by the retention of erythrocytes or by lipid degradation. During Wallerian degeneration a significant increase of 18:1, associated with a decrease of saturated FFAs, was found in rat sciatic endoneurium, but normal values were approached when fiber regeneration was well under way. The FFA composition with chain length ≥C26 were not, however, significantly altered with degeneration or repair of nerves. The metabolic significance of this striking difference between nerve and brain FFA composition is unknown but may reflect different functional properties.  相似文献
