The electron transport rates and coupling factor activity in the chloroplasts; adenylate contents, rates of photosynthesis and respiration in the leaves as well as activity of isolated mitochondria were investigated in Pisum sativum L. leaves of plants grown under low or high light intensity and exposed after detachment to 5 mM Pb(NO(3))(2). The presence of Pb(2+) reduced rate of photosynthesis in the leaves from plants grown under the high light (HL) and low light (LL) conditions, whereas the respiration was enhanced in the leaves from HL plants. Mitochondria from Pb(2+) treated HL-leaves oxidized glycine at a higher rate than those isolated from LL leaves. ATP content in the Pb-treated leaves increased to a greater extend in the HL than LL grown plants. Similarly ATP synthase activity increased markedly when chloroplasts isolated from control and Pb-treated leaves of HL and LL grown plants were subjected to high intensity light. The presence of Pb ions was found inhibit ATP synthase activity only in chloroplasts from LL grown plants or those illuminated with low intensity light. Low light intensity during growth also lowered PSI electron transport rates and the Pb(2+) induced changes in photochemical activity of this photosystem were visible only in the chloroplasts isolated from LL grown plants. The activity of PSII was influenced by Pb ions on similar manner in both light conditions. This study demonstrates that leaves from plants grown under HL conditions were more resistant to lead toxicity than those obtained from the LL grown plants. The data indicate that light conditions during growth might play a role in regulation of photosynthetic and respiratory energy conservation in heavy metal stressed plants by increasing the flexibility of the stoichiometry of ATP to ADP production.  相似文献

Background

The marine cyanobacterium Prochlorococcus marinus, having multiple ecotypes of distinct genotypic/phenotypic traits and being the first documented example of genome shrinkage in free-living organisms, offers an ideal system for studying niche-driven molecular micro-diversity in closely related microbes. The present study, through an extensive comparative analysis of various genomic/proteomic features of 6 high light (HL) and 6 low light (LL) adapted strains, makes an attempt to identify molecular determinants associated with their vertical niche partitioning.

Results

Pronounced strand-specific asymmetry in synonymous codon usage is observed exclusively in LL strains. Distinct dinucleotide abundance profiles are exhibited by 2 LL strains with larger genomes and G+C-content ≈ 50% (group LLa), 4 LL strains having reduced genomes and G+C-content ≈ 35-37% (group LLb), and 6 HL strains. Taking into account the emergence of LLa, LLb and HL strains (based on 16S rRNA phylogeny), a gradual increase in average aromaticity, pI values and beta- & coil-forming propensities and a decrease in mean hydrophobicity, instability indices and helix-forming propensities of core proteins are observed. Greater variations in orthologous gene repertoire are found between LLa and LLb strains, while higher number of positively selected genes exist between LL and HL strains.

Conclusion

Strains of different Prochlorococcus groups are characterized by distinct compositional, physicochemical and structural traits that are not mere remnants of a continuous genetic drift, but are potential outcomes of a grand scheme of niche-oriented stepwise diversification, that might have driven them chronologically towards greater stability/fidelity and invoked upon them a special ability to inhabit diverse oceanic environments.  相似文献

Chlamydomonas reinhardtii was grown at photon flux densities (PFDs) ranging from 47 to 400 μE.m-2 s-1. The total cellular content of chlorophyll (Chl) was twice as high in the low light (LL) versus high light (HL) grown cells. On an equal Chl basis, photosystem II (PSII) and cytochrome f (Cyt f) content was higher in HL cells, but photosystem I (PSI) concentration displayed little variation with the light intensity during cell growth. Consequently, there was a shift in the ratio of PSII / PSI and Cyt / PSI from near unity in LL cells to greater than two in HL cells. The functional Chl antenna size of PSII and PSI ranged from 460 and 170 Chl (a + b)in HL-grown cells to 620 and 370 Chl (a+ b)in LL-grown cells, respectively. The initial slope of the Chl-specific photosyn-thesis-irradiance (P-I) curve was similar in LL- and HL-grown cells, but the light saturated rate of photosynthesis was lower under LL. The response to low light was beneficial at the cellular level, since there was an enhancement of photosynthesis in LL. The PFD for the onset of light saturation, 1 was a factor of 2 lower in LL- relative to HL-grown photosythetic membranes. Since growth PFD varied by a factor of ten, photosynthesis shifted from being light-limited in the LL regime to light-saturated in the HL regime. The requirement for balanced absorption of light by the two photosystems constrains the PSII / PSI ratio to near unity when growth is light-limited, but such a constraint does not apply in HL conditions. Instead the concentration of individual electron transport complexes way be related to the pool size necessary for maximum rates of steady-state electron transport. Thus the stoichiometry of electron transport complexes changes in response to growth PFD and this change is correlated with the response flexlbility of algal photosynthesis in diverse light environments.  相似文献

Elemental stoichiometry and organic composition were investigated in an Adriatic strain of Skeletonema marinoi, cultured at 25 [low light (LL)] and 250 [high light (HL)]µmol photon m?2 s?1. Inorganic carbon acquisition, fixation and allocation, and silicic acid and orthophosphate uptake were also studied. The C : P ratio was below the Redfield ratio, especially at LL. In HL cells, N quota was halved, C quota was similar, silica quota was lower, growth rate and long‐term net primary productivity were almost doubled, relative to LL cells. The HL : LL cell quota ratios were 6 for lipid, 0.5 for protein and 0.4 for carbohydrate. Phosphoenolpyruvate carboxylase (PEPc) and glutamine synthetase (GS) activities were unaffected by the growth irradiance; phosphoenolpyruvate carboxykinase (PEPck) was 2.5‐fold more active in LL cells. This suggests that in S. marinoi, C4 photosynthesis is unlikely, PEPc is anaplerotic and PEPck may be involved in the conversion of lipid C to carbohydrates, especially in LL cells. Because about 50% of the cost for the production of an HL cell is caused by lipid biosynthesis, we propose that the preferential allocation of C to lipid at HL takes advantage of the relatively high volume‐based energy content of lipids, in an organism that reduces its size at each vegetative cell division.  相似文献

In order to ensure the cooperative function with the photosynthetic system, the mitochondrial respiratory chain needs to flexibly acclimate to a fluctuating light environment. The non-phosphorylating alternative oxidase (AOX) is a notable respiratory component that may support a cellular redox homeostasis under high-light (HL) conditions. Here we report the distinct acclimatory manner of the respiratory chain to long- and short-term HL conditions and the crucial function of AOX in Arabidopsis thaliana leaves. Plants grown under HL conditions (HL plants) possessed a larger ubiquinone (UQ) pool and a higher amount of cytochrome c oxidase than plants grown under low light conditions (LL plants). These responses in HL plants may be functional for efficient ATP production and sustain the fast plant growth. When LL plants were exposed to short-term HL stress (sHL), the UQ reduction level was transiently elevated. In the wild-type plant, the UQ pool was re-oxidized concomitantly with an up-regulation of AOX. On the other hand, the UQ reduction level of the AOX-deficient aox1a mutant remained high. Furthermore, the plastoquinone pool was also more reduced in the aox1a mutant under such conditions. These results suggest that AOX plays an important role in rapid acclimation of the respiratory chain to sHL, which may support efficient photosynthetic performance.  相似文献

This study examined the relationship between preexercise muscle glycogen content and glycogen utilization in two physiological pools, pro- (PG) and macroglycogen (MG). Male subjects (n = 6) completed an exercise and dietary protocol before the experiment that resulted in one leg with high glycogen (HL) and one with low glycogen (LL). Preexercise PG levels were 312 +/- 29 and 208 +/- 31 glucosyl units/kg dry wt (dw) (P < or = 0.05) in the HL and LL, respectively, and the corresponding values for MG were 125 +/- 37 and 89 +/- 43 mmol glucosyl units/kg dw (P < or = 0.05). Subjects then performed two 90-s exercise bouts at 130% maximal oxygen uptake separated by a 10-min rest period. Biopsies were obtained at rest and after each exercise bout. Preexercise glycogen concentration was correlated to net glycogenolysis for both PG and MG for bout 1 and bouts 1 and 2 (r < or = 0.60). In bout 1, there was no difference in the rate of PG or MG catabolism between HL and LL despite a 26% increase (P < or = 0.05) in glycogen phosphorylase transformation (phos a %) in the HL. In the second bout, more PG was catabolized in the HL vs. LL (38 +/- 9 vs. 9 +/- 6 mmol glucosyl units. kg dw(-1). min(-1)) (P < or = 0.05) with no difference between legs in phos a %. phos a % was increased in HL vs. LL but does not necessarily increase glycogenolysis in either PG or MG. Despite both legs performing the same exercise and having identical metabolic demands, the HL catabolized 2.3 (P < or = 0.05) times more PG and 1.5 (P < or = 0.05) times more MG vs. LL in bouts 1 and 2, indicating that preexercise glycogen concentration is a regulator of glycogenolysis.  相似文献

The diatom Cyclotella meneghiniana Kütz. (SAG 1020‐a) was cultured under high‐light (HL) and low‐light (LL) conditions with either high (12 μM) or low (1 μM) iron in the media. Changes in cell morphology, especially cell volume and chloroplast size, were observed in cells grown under low iron. In contrast, HL had a much stronger influence on the photosynthetic apparatus. PSII function was unimpaired under lowered iron supply, but its quantum efficiency and reoxidation rate were reduced under HL conditions. As reported before, HL induced changes in antenna polypeptide composition. Especially the amount of Fcp6, an antenna protein related to LI818 and known to be involved in photoprotection, was increased under HL but was significantly reduced under lowered iron. The diatoxanthin content correlated with the amount of Fcp6 in isolated FCPa antenna complexes and was thus increased under HL and reduced under low iron as well. While the diatoxanthin (Dt) content of whole cells was enhanced under HL, no decrease was observed under lowered iron supply, ruling out the possibility that the decreased amounts in FCPa were due to a hampered diadinoxanthin de‐epoxidase activity under these conditions. Thus, diatoxanthin not bound to FCPa has to be responsible for protection under the slight reduction in iron supply used here.  相似文献

The response of microalgae to photooxidative stress resulting from high light exposure is a well-studied phenomenon. However, direct analyses of photosystem II (PSII) D1 protein (the main target of photoinhibition) in diatoms are scarce. In this study, the response of the diatom model species Phaeodactylum tricornutum to short-term exposure to high light was examined and the levels of D1 protein determined immunochemically. Low light (LL) acclimated cells (40 μmol photons m(-2) s(-1)) subjected to high light (HL, 1,250 μmol photons m(-2) s(-1)) showed rapid induction of non-photochemical quenching (NPQ) and ca. 20-fold increase in diatoxanthin (DT) concentration. This resulted from the conversion of diadinoxanthin (DD) to DT through the activation of the DD-cycle. D1 protein levels under LL decreased about 30% after 1 h of the addition of lincomycin (LINC), a chloroplast protein synthesis inhibitor, showing significant D1 degradation and repair under low irradiance. Exposure to HL lead to a 3.2-fold increase in D1 degradation rate, whereas average D1 repair rate was 1.3-x higher under HL than LL, leading to decreased levels of D1 protein under HL. There were significant effects of both HL and LINC on P. tricornutum maximum quantum yield of PSII (F(v)/F(m)), showing a reduction of active PSII reaction centres. Partial recovery of F(v)/F(m) in the dark demonstrates the photosynthetic resilience of this diatom to changes in the light regime. P. tricornutum showed high allocation of total protein to D1 and an active D1-repair cycle to limit photoinhibition.  相似文献

Cells of the red alga Porphyridium cruentum (ATCC 50161) exposed to increasing growth irradiance exhibited up to a three-fold reduction in photosystems I and II (PSI and PSII) and phycobilisomes but little change in the relative numbers of these components. Batch cultures of P. cruentum were grown under four photon flux densities of continuous white light; 6 (low light, LL), 35 (medium light, ML), 180 (high light, HL), and 280 (very high light, VHL) microeinsteins per square meter per second and sampled in the exponential phase of growth. Ratios of PSII to PSI ranged between 0.43 and 0.54. About three PSII centers per phycobilisome were found, regardless of growth irradiance. The phycoerythrin content of phycobilisomes decreased by about 25% for HL and VHL compared to LL and ML cultures. The unit sizes of PSI (chlorophyll/P700) and PSII (chlorophyll/QA) decreased by about 20% with increase in photon flux density from 6 to 280 microeinsteins per square meter per second. A threefold reduction in cell content of chlorophyll at the higher photon flux densities was accompanied by a twofold reduction in β-carotene, and a drastic reduction in thylakoid membrane area. Cell content of zeaxanthin, the major carotenoid in P. cruentum, did not vary with growth irradiance, suggesting a role other than light-harvesting. HL cultures had a growth rate twice that of ML, eight times that of LL, and slightly greater than that of VHL cultures. Cell volume increased threefold from LL to VHL, but volume of the single chloroplast did not change. From this study it is evident that a relatively fixed stoichiometry of PSI, PSII, and phycobilisomes is maintained in the photosynthetic apparatus of this red alga over a wide range of growth irradiance.  相似文献

Two procedures are described for the fractionation of chromatin containing unsubstituted (LL) DNA and DNA unifilarly substituted with bromodeoxyuridine (HL). The two procedures rely upon the sensitivity of bromodeoxyuridine-containing DNA to UV light to induce either strand breakage or protein crosslinking. When a mixture of LL and HL chromatin is irradiated with UV light, the HL DNA fragments into molecules of smaller molecular weight than the LL DNA and crosslinks more chromosomal protein than the LL DNA. LL and HL chromatin can be fractionated on the basis of size by centrifuging through a neutral sucrose gradient. The HL DNA-protein adducts that are generated by the UV light have a unique buoyant density and may be isolated by isopycnic centrifugation in CS2SO4. The ability to fractionate LL and HL chromatin permits certain studies on the structure of replicating chromatin.  相似文献

Berteroa incana (B. incana), a spring ephemeral species of Brassicaceae, possesses very high photosynthetic capacities at high irradiances. Exploring the mechanism of the high light use efficiency of B. incana under strong light conditions may help to explore mechanisms of plants' survival strategies. Therefore, the photosynthetic characteristics of B. incana grown under three different light intensities (field conditions (field): 200-1500μmolphotonsm(-2)s(-1); greenhouse high light (HL) conditons: 600μmolphotonsm(-2)s(-1); and greenhouse low light (LL) conditions: 100μmolphotonsm(-2)s(-1)) were investigated and compared with those of the model plant Arabidopsis thaliana (A. thaliana). Our results revealed that B. incana behaved differently in adjusting its photosynthetic activities under both HL and LL conditions compared with what A. thaliana did under the same conditions, suggesting that the potential of photosynthetic capacity of B. incana might be enhanced under strong light conditions. Under LL conditions, B. incana reached its maximum photosynthetic activity at a much higher light intensity than A. thaliana did, although their maximum photochemical efficiency of photosystem II (PSII) (F(v)/F(m)) was almost the same. When grown under HL conditions, B. incana showed much higher photosynthetic capacity than A. thaliana. A detailed analysis of the OJIP transient kinetics of B. incana under HL and LL conditions revealed that HL-grown B. incana possessed not only a high ability in regulating photosystem stoichiometry that ensured high linear electron transport, but also an enhanced availability of oxidized plastoquinone (PQ) pool which reduced non-photochemical quenching (NPQ), especially its slow components qT and qI, and increased the photochemical efficiency, which in turn, increased the electron transport. We suggest that the high ability in regulating photosystem stoichiometry and the high level of the availability of oxidized PQ pool in B. incana under strong light conditions play important roles in its ability to retain higher photosynthetic capacity under extreme environmental conditions.  相似文献

The thylakoid proteome of chloroplasts contains multiple proteins involved in antioxidative defense, protein folding, and repair. To understand this functional protein network, we analyzed the quantitative response of the thylakoid-associated proteome of Arabidopsis (Arabidopsis thaliana) wild type and the ascorbate-deficient mutant vtc2-2 after transition to high light (HL; 1,000 micromol photons m(-2) s(-1)). The soluble thylakoid proteomes of wild type and vtc2-2 were compared after 0, 1, 3, and 5 d of HL using two-dimensional gels with three independent experiments, followed by a multivariant statistical analysis and tandem mass spectrometry. After 5 d of HL, both wild-type and vtc2-2 plants accumulated anthocyanins, increased their total ascorbate content, and lost 10% of photosystem II efficiency, but showed no bleaching. Anthocyanin and total ascorbate concentrations in vtc2-2 were respectively 34% and 20% of wild type, potentially leading to enhanced oxidative stress in vtc2-2. Forty-five protein spots significantly changed as a consequence of genotype, light treatment, or both. Independent confirmation was obtained from western blots. The most significant response was the up-regulation of thylakoid YCF37 likely involved in photosystem I assembly, and specific fibrillins, a flavin reductase-like protein, and an aldolase, each located in thylakoid-associated plastoglobules. Fe-superoxide dismutase was down-regulated in vtc2-2, while Cu,Zn-superoxide dismutase was up-regulated. vtc2-2 also showed a systematic up-regulation of a steroid dehydrogenase-like protein. A number of other stress-related proteins, several thylakoid proteases, and lumenal isomerases did not change, while PsbS increased in wild type upon light stress. These findings are discussed in terms of plastid metabolism and oxidative stress defense, and emphasize that understanding of the chloroplast stress-response network must include the enzymatic role of plastoglobules.  相似文献
