东北红豆杉细胞两液相培养中紫杉醇释放行为研究   总被引:4,自引:0,他引:4  
在东北红豆杉细胞悬浮培养中,分别研究了稀土化合物(硫酸铈铵)、有机溶剂(油酸和邻苯二甲酸二丁脂)和稀土化合物与有机溶剂的协同作用对紫杉醇释放的影响。在此基础上深入研究了在东北红豆杉细胞两液相培养中,紫杉醇释放率随不同的有机溶剂(烷烃、有机酸、醇和脂)、有机溶剂的体积分数、有机溶剂的加入时间和有机溶剂相毒性的变化规律。结果表明分别加入稀土化合物和有机溶剂都明显促进紫杉醇的释放,特别是有机溶剂更显著促进紫杉醇的释放。但在东北红豆杉细胞两液相培养中,稀土化合物加入不能进一步促进紫杉醇的释放。因此两液相培养中有机溶剂本身就是很好的产物释放剂。紫杉醇的释放率由对照组的40%提高到75%以上。  相似文献

Summary Conditions have been established for the induction and maintenance of callus cultures of Taxus brevifolia (Pacific yew) from bark, stem, and needle tissues. Cultures were established on a modified Gamborg's B5 medium, 1% sucrose, 0.2% casamino acids and 1 mg/L 2,4-D. There was no apparent inhibition of callus induction as a result of taxol concentration in the explant material. Cell lines derived from explants of individual trees were used to investigate growth characteristics. Although none of the cell lines contained taxol, some contained low levels of related taxanes. Variability was observed with each cell line in response to light, and auxin type and concentration. Growth index was most affected by cell line, followed by auxin type and concentration. 研究了诱导子、前体及抑制剂的协调作用对东北红平杉生产紫杉醇的影响。结果表明,向培养基中加入80mg/L水到、80mg/L茉莉酸甲酯、0.5mmol/L乙酸钠、2mmol/L苯丙氨酸、0.5mmol/L丝氨酸、0.1mmol/L甘氨酸、10mg/L肉桂酸、0.5mmol/L苯甲酸钠、5mmol/L丙酮酸钠、10mg/L氯化氯胆碱、和1mg/L赤霉酸可使紫杉醇含量提高368.65%。并且证明交互作用对紫杉醇合成有显著作用。  相似文献

Cell culture of Taxus cuspidata may represent an alternative to extraction of bark as a source of taxol and related taxanes. Cell suspensions of a cell line of T. cuspidata were grown for 44 days in shake flasks containing B5C2 medium. Throughout the growth cycle, fresh and dry weight accumulation, taxol yield on a dry weight basis, taxol accumulation in the medium, pH and pigmentation variation in the medium, as well as the uptake of sucrose, glucose, fructose, nitrate, and inorganic phosphate from the culture medium were examined. The results showed that the growth was relatively slow (doubling times of 17 and 20 days for fresh and dry weight, respectively), and taxol accumulation in the cells was non-growth related (higher in the stationary phase) and at relatively low levels (up to 4 mug/g of the extracted dry weight). Taxol concentration in the medium had two peaks: one during the early (0.4mug/mL) and another during the late (0.1-mug/mL) parts of the growth cycle. On a volumetric basis, the average total amount of taxol produced during the stationary phase (day 38) was 0.15 mug/mL, of which approximately 66% was in the medium and 34% was in the cells. Total carbohydrate uptake was closely associated with the increase in dry biomass. Sucrose was apparently extracellularly hydrolyzed after the first 6 days of culture; glucose was used before fructose. Nitrate was assimilated throughout the growth cycle, but phosphate was absorbed within the first week of culture. The pH variation showed an initial drop followed by a trend toward alkalinization for most of the growth period. 研究了果糖和几种前体物质对东北红豆杉生产紫杉醇的影响,结果表明,在第12d加入6g/L果糖可以使紫杉醇产量增加63.89%,在糖协同的作用下,加入前体(0.05mmol/L乙酸钠,0.05mmol/L苯丙氨酸,0.1mmol/L苯甲酸钠)可显著提高紫杉醇的合成,同对照相比,含量分别增加49.36%、13.18%和64.26%,在第15d向培养基中加入0.05 mmol/L乙酸钠、0.1mmol/L苯甲酸钠、1mmol/L苯丙氨酸和6g/L果糖则使紫杉醇含量提高181.89%。  相似文献

研究证明, 多酚氟化酶(PPO)与东北红豆杉细胞生长存在密切关系, 利用Vc、NaCl、苯甲酸、亚硫酸钠、柠檬酸、草酸等可有效抑制PPO 的活性, 降低褐化程度, 促进东北红豆杉细胞培养物的生长。而利用硫酸铜、水解酪蛋白、椰乳、乙烯吡咯烷酮等可有效抑制PPO 活性·加重褐化、抑制东北红豆杉细胞培养物的生长。  相似文献

Methyl jasmonate (MeJA), a methyl ester of jasmonic acid (JA), is a well-established signal molecule in plant defense responses and an effective inducer of secondary metabolite accumulation in plant cell cultures such as the valuable anticancer diterpenoid taxol (paclitaxel) in Taxus spp. This work examines the involvement of nitric oxide (NO) in MeJA-induced plant defense responses and secondary metabolism in Taxus chinensis cell cultures. Exogenously supplied MeJA at 100 microM induced rapid production of NO in the Taxus cell cultures, reaching a maximum within 6 h of MeJA supply. Several other responses occurred concomitantly, including the production of hydrogen peroxide (H2O2), and the increases in intracellular malondialdehyde (MDA) content, lipoxygenase (LOX) and phenylalanine ammonium-lyase (PAL) activities. The MeJA-induced H2O2 production was suppressed by an NO donor, sodium nitroprusside (SNP), but enhanced by NO inhibitors, N (omega)-nitro-L-arginine (L-NNA) and 2-phenyl-4,4,5,5-tetramethyl-imidazoline-1-oxyl-3-oxide (PTIO). In contrast, the MeJA-induced MDA, LOX and PAL were all enhanced by the NO donor but suppressed by the NO inhibitors. The NO inhibitors also suppressed MeJA-induced taxol accumulation. These results are suggestive of a role for NO as a signal element for activating the MeJA-induced defense responses and secondary metabolism activities of plant cells.  相似文献

The effects of concentration of amino acids, nitrate, and ammonium on the growth and taxol production in cultures of cell line TY-21 of Taxus yunnanensis were investigated. Addition of 20 different amino acids each at 15–20 mg l–1 to B5 medium significantly improved callus growth but inhibited taxol formation in the cultures. The optimum nitrate concentration was 20–30 mM for both growth and taxol production. Ammonium greatly suppressed growth but strongly promoted taxol formation in the cells when it was the sole inorganic nitrogen in the medium. Culturing the suspension cells in nitrate-containing medium for 15 days and then in a medium in which ammonium was the sole inorganic nitrogen for 7 days increased taxol yield by 104%, reaching up to 28.1 mg l–1.  相似文献

Methyl jasmonate (MeJA) and cerium (Ce(4+)) elicitation share common features of increasing taxol accumulation of Taxus cuspidata cells. Interestingly, Ce(4+) induces programmed cell death (PCD), but this phenomenon is not observed with MeJA elicitation. Here, using a lipidomic approach to measure more than 100 membrane glycerophospholipids of T. cuspidata cells quantitatively, we discovered that lysophosphatidylcholine (LysoPC), phosphatidic acid (PA) and phosphatidylcholine were three potential lipid markers that were responsible for the differences between Ce(4+)-induced cells and MeJA-induced cells. Compared with MeJA elicitation, marked increase of phospholipase D (PLD) activity was observed following Ce(4+) elicitation, suggesting that the PLD activation and high concentrations of PA production might mediate the PCD. Rapid increase of phospholipase A(2) (PLA(2)) activity caused the release of fatty acids and LysoPC following Ce(4+) elicitation, which enhanced endogenous jasmonic acid (JA) accumulation. In contrast, PLA(2) activity was poorly induced following MeJA elicitation. PLA(2) inhibitor suppressed not only JA accumulation but also taxol production, suggesting that the PLA(2) activation mediated Ce(4+)-induced taxol production partially through a JA-dependent signaling pathway. These results demonstrate that differential alternation of glycerolphospholipids caused by phospholipases constitutes an important step in cell death response to Ce(4+) and increasing taxol production.  相似文献

美丽镰刀(Fusarium mairei)是一分离自南方红豆杉(Taxus chinensis var.mairei)的产紫杉醇内生真菌,用B5培养基培养6 d,去菌尘幺后制得美丽镰刀菌培养液,并从中提取其胞外多糖.研究了4%(V/V)美丽镰刀菌培养液及4%(W/V)胞外多糖两种处理,对东北红豆杉(T.cuspidata)悬浮细胞防御反应及紫杉醇合成的影响.结果表明:两种处理均能诱导东北红豆杉细胞的防御反应.但美丽镰刀菌培养液的影响明显大于胞外多特(P<0.05).另外,两种处理均可促进东北红豆杉细胞紫杉醇的合成与释放.美丽镰刀菌培养液处理得到的紫杉醇与其释放率分别是对照的2.5倍8.8倍,而胞外多糖处理得到的紫杉醇与其释放率则分别是对照的1.5倍与3倍.  相似文献

研究表明外加紫杉醇能够诱导悬浮培养的东北红豆杉(Taxus cuspidata)细胞总DNA发生梯带化降解。利用mRNA差异显示技术比较了紫杉醇诱导凋亡与不诱导凋亡的东北红豆杉细胞基因表达的差异,得到了8个特异表达的cDNA克隆,经Northern杂交证实其中3个在不发生凋亡的细胞中表达,5个在凋亡的细胞中表达。对这8个cDNA克隆单向序列测定后,与GenBank/EMBL/DDBJ中同源序列进行了比较,结果表明:1个cDNA片段与拟南芥中ABA应答蛋白基因的保守区有86%的同源性;2个cDNA片段与番茄内切壳聚糖酶前体基因的保守区有50%的同源性;其他5个cDNA片段无明显的同源基因,可能是新基因。  相似文献

Single cell growth and division was measured via flow cytometry in order to characterize the metabolic variability of Taxus cuspidata suspension cultures, which produce the valuable secondary metabolite Taxol. Good agreement was observed between the cell cycle distribution and biomass accumulation over the batch culture period. Specific growth rates of 0.13 days(-1) by fresh weight and 0.15 days(-1) by dry weight were measured. Elicitation with methyl jasmonate (MJ) significantly decreased both cell cycle progression and biomass accumulation, as the specific growth rate decreased to 0.027 days(-1) by fresh and dry weight. Despite the decrease in biomass accumulation for MJ elicited cultures, sucrose utilization was not significantly different from control cultures. MJ elicitation also increased the accumulation of paclitaxel and other taxanes. The accumulation of upstream taxanes (baccatin III and 10-deactylbaccatin III) increased during exponential growth, reached a maximum around day 12, and then declined throughout the stationary phase. The paclitaxel concentration increased during both exponential growth and stationary phase, reaching a maximum around days 20-25. Throughout the culture period, greater than 70% of the cells were in G(0)/G(1) phase of the cell cycle. Studies using bromodeoxyuridine (BrdU) incorporation showed that approximately 65% of the Taxus cells are noncycling, even during exponential growth. Although the role of these cells is currently unknown, the presence of a large, noncycling subpopulation can have a significant impact on the utilization of plant cell culture technology for the large-scale production of paclitaxel. These results demonstrate that there is a high degree of metabolic heterogeneity in Taxus cuspidata suspension cultures. Understanding this heterogeneity is important for the optimization of plant cell cultures, particularly the reduction of production variability.  相似文献

东北红豆杉(TaxuscuspidataSIEBETZUCC)是天然抗癌药物资源,其树皮中含有一种抗癌药物(TAXAL紫杉酚)引起各国的注意。由于红豆杉生长缓馒,往往遭到砍伐,远远不能满足抗癌药物研究和医疗药用的需要;加之资源又贫乏,因此确定此项研究。开展此项研究结果表明:黑龙江省、辽宁省及哈尔滨地区采样诱导率哈尔滨居首位,采样时间以五月份愈伤组织诱导率高达56.5%;叶片不同部位以叶前部和后部为好;叶脉方向对诱导率无明显差导。诱导愈伤组织中注意及时转移、继代、防止在培养中的褐变。克服褐变需对愈伤组织选择出组织紧密、褐变速度慢、同时在培养基中加入一些抗氧化剂,其效果明显。目前愈伤组织三颗粒状,已见绿芽点,现转入分化试验。  相似文献

The influences of salicylic acid (SA) on taxol production and isopentenyl pyrophosphate (IPP) biosynthesis pathways in suspension cultures of Taxus chinensis var. mairei were investigated by adding SA and mevastatin (MVS), a highly specific inhibitor of 3-hydroxy-3-methylglutaryl-CoA reductase in the mevalonate pathway for IPP biosynthesis, into the culture systems. The cell death and taxol production were induced upon the introduction of SA, and 20mg/l was proved to be the optimal SA concentration in terms of the less damage to Taxus cells and marked activation of phenylalanine ammonia lyase (PAL). In the coexistence of SA (20mg/l) and MVS (100 nmol/l), the taxol content (1.626 mg/g dry wt) was higher than that (0.252 mg/g dry wt) of the MVS-treated system but almost equal to that (1.581 mg/g dry wt) of the SA-treated system. It is thus inferred that the activated non-mevalonate pathway should be responsible for the formation of IPP in taxol biosynthesis in the presence of SA.  相似文献

The kinetics of biomass accumulation, nutrient uptake and taxol production of Taxus baccata cell suspensions were examined in three bioreactor configurations, viz. 250-mL Erienmeyerflasks, 1-L working volume pneumatically mixed (PMB), and stirred tank (STB) bioreactors. Qualitatively similar kinetics were observed in all three bioreactor types. Biomass accumulation and specific nutrient uptake rates exhibited biphasic characteristics. Carbohydrate uptake and biomass accumulation substantially ceased when phosphate was depleted from the medium. Phosphate was identified as a possible growth-limiting nutrient. Taxol accumulated exclusively in the second phase of growth. A maximum taxol concentration of 1.5 mg/L was obtained in the PMB which was fivefold greater than that obtained in the Erienmeyer flasks and the STB, but the relative kinetics of taxol production was the same in all three reactor types. Biomass yields were calculated from the kinetic data and a stoichiometry for biomass formation was evaluated. A cell suspension culture of Taxus media was established from a stable callus line of this species. The growth rate and production of paclitaxel and baccatin III of this cell suspension were significantly increased during the shake flask culture in its respective optimum media for cell growth and product formation, which were selected after assaying 24 different culture media. The highest yields of paclitaxel (2.09 mg L(-1)) and baccatin III (2.56 mg L(-1)) in the production medium rose (factors of 7.0 and 3.0, respectively) in the presence of methyljasmonate (220 microg g(-1) FW). When the elicitor was added together with mevalonate (0.38 mM) and N-benzoylglycine (0.2 mM), the increase in the yields of paclitaxel and baccatin III was even higher (factors of 8.3 and 4.0, respectively). Thereafter, a two-stage culture for cell suspension was carried out using a 5-l stirred bioreactor running for 36 days, the first stage being in the cell growth medium until cells entered their stationary growth phase (12 days) and the second stage being in the production medium supplemented with the elicitor and two putative precursors in the concentrations indicated above. Under these conditions, 21.12 mg L(-1) of paclitaxel and 56.03 mg L(-1) of baccatin III were obtained after 8 days of culture in the production medium.  相似文献
