When retinoic acid (RA) is applied to the regenerating limb the positional information of blastemal cells is respecified and extra limb segments develop. We are trying to elucidate the molecular basis of the action of RA and report here experiments focused on the role that fibronectin (FN) might play in the process. The FN distribution in stump tissues, regeneration blastemas and RA-treated blastemas was investigated by immunocytochemistry. Two effects of RA were observed. Firstly, excessive dedifferentiation of the severed cartilage at the amputation plane, resulting in lumps of FN-positive matrix being released into the blastema; secondly, blastemal cells tend to aggregate together into FN-positive accumulations. Excessive dedifferentiation of the cartilage plays no role in the RA-induced respecification of pattern, because we show that extra segments are still produced in RA-treated limbs from which all the cartilage has been removed. The effect on blastemal cell FN distribution was investigated in several ways. Axolotl plasma FN and cellular FN were characterised on immunoblots, and no obvious change was observed after RA treatment; neither were there changes in amounts of FN detected by ELISA. Levels of FN synthesis were measured by [35S]-methionine labelling and again no change observed after RA treatment. We conclude that the change in FN distribution observed by immunocytochemistry after RA treatment may be due to the retention of FN on the surface of the blastemal cells rather than to any effect on the levels of synthesis of this molecule.  相似文献

When blastemas of several stages of differentiation were grafted in normal orientation to stump levels proximal or distal to their level of origin, normal limbs regenerated. Histological and autoradiographic studies of the development of these regulated limbs showed that the grafted blastemas formed only structures normally distal to their level of origin. In the case of a blastema transplanted proximally, regulation occurred by intercalary regeneration from the stump, whereas, when blastemas were transplanted distally, regulation appeared to take place within the blastema itself by a distal shift in its pattern of organization. The results suggest that the proximal limit of the limb regenerate is determined by level-specific properties of the limb cells but that these properties allow for interactions leading to regulation when different levels of stump and blastema are brought together.  相似文献

Interactions between the limb stump and the developing regenerate were studied in the limbs of adult newts, Notophthalmus viridescens. Forelimb blastemas at various stages were transplanted to the contralateral forelimb such that the anterior-posterior axes of stump and blastema were opposed. The blastemas were transplanted either from a proximal to distal, distal to proximal, proximal to proximal, or distal to distal level limb stump. The results indicate that at the earliest stage studied the anterior-posterior axis of the blastema is established but is not stable. An interection between the stump and blastema at this early stage results in the production of a variety of limbs intermediate in polarity between the graft and the stump. At all later stages, the original anterior-posterior axis of the blastema can be retained, although under certain grafting conditions the stump can still exert considerable influence over the anterior-posterior organization of the final regenerate. In those circumstances in which the blastema retains its original handedness, the interaction between stump and blastema results in the production of separate anterior and posterior supernumerary regenerates.The results of transplanting proximal blastemas to a distal limb level indicate that the proximal boundary of the blastema has been established by the earliest stage studied, leading to the production of limbs with serially duplicated segments. However, irrespective of the stage of a blastema transplanted from a distal to proximal level, there are no deleted structures in the proximal-distal axis of the resulting limb. From both histological examination of transplanted regenerates and the arrangement of skeletal elements of the resulting limbs, it is postulated that the stump plays an important role in the production of the intercalary regenerate.  相似文献

After tail and limb amputation in lizard, injection of 5BrdU for 6 days produces immunolabelled cells in most tissues of tail and limb stumps. After further 8 and 16 days, and 14 and 22 days of regeneration, numerous 5BrdU-labelled cells are detected in regenerating tail and limb, derived from most stump tissues. In tail blastema cone at 14 days, sparse-labelled cells remain in proximal dermis, muscles, cartilaginous tube and external layers of wound epidermis but are numerous in the blastema. In apical regions at 22 days of regeneration, labelled mesenchymal cells are sparse, while the apical wound epidermis contains numerous labelled cells in suprabasal and external layers, indicating cell accumulation from more proximal epidermis. Cell proliferation dilutes the label, and keratinocytes take 8 days to migrate into corneous layers. In healing limbs, labelled cells remain sparse from 14 to 22 days of regeneration in wound epidermis and repairing tissues and little labelling dilution occurs indicating low cell proliferation for local tissue repair but not distal growth. Labelled cells are present in epidermis, intermuscle and peri-nerve connectives, bone periosteum, cartilaginous callus and sparse fibroblasts, leading to the formation of a scarring outgrowth. Resident stem cells and dedifferentiation occur when stump tissues are damaged.  相似文献

The mechanisms by which cells obtain instructions to precisely re-create the missing parts of an organ remain an unresolved question in regenerative biology. Urodele limb regeneration is a powerful model in which to study these mechanisms. Following limb amputation, blastema cells interpret the proximal-most positional identity in the stump to reproduce missing parts faithfully. Classical experiments showed the ability of retinoic acid (RA) to proximalize blastema positional values. Meis homeobox genes are involved in RA-dependent specification of proximal cell identity during limb development. To understand the molecular basis for specifying proximal positional identities during regeneration, we isolated the axolotl Meis homeobox family. Axolotl Meis genes are RA-regulated during both regeneration and embryonic limb development. During limb regeneration, Meis overexpression relocates distal blastema cells to more proximal locations, whereas Meis knockdown inhibits RA proximalization of limb blastemas. Meis genes are thus crucial targets of RA proximalizing activity on blastema cells.  相似文献

Regeneration blastemas at the stages of medium bud and palette were transplanted to contralateral limb stumps so that either their anterior and posterior positions or their dorsal and ventral positions were apposed to those of the stumps. Grafts were shifted from distal levels to proximal levels, or from proximal levels to distal levels, or remained at either a proximal or a distal level. When anterior and posterior positions of graft and stump were apposed, supernumerary limbs were produced at the graft-stump junction in anterior and posterior positions relative to the stump. All analyzable supernumerary limbs were of stump handedness. Apposition of dorsal and ventral positions of graft and stump led to the formation of supernumerary limbs at dorsal and ventral positions relative to stump tissues. All analyzable supernumerary limbs were once again of stump handedness. Shifts from distal levels to proximal levels never resulted in skeletal deletions, as potential deletions in the proximal-distal axis were always filled in. Shifts from proximal levels to distal levels resulted in a low frequency of serial duplications. The results are discussed in view of a recently presented formal model for pattern regulation in epimorphic fields.  相似文献

Amphibians have a remarkable capacity for limb regeneration. Following a severe injury, there is complete regeneration with restoration of the patterning and cellular architecture of the amputated limb. While studies have focused on the structural anatomical changes during amphibian limb regeneration, the signaling mechanisms that govern cellular dedifferentiation and blastemal progenitors are unknown. Here, we demonstrate the temporal and spatial requirement for hedgehog (Hh) signaling and its hierarchical correlation with respect to Wnt signaling during newt limb regeneration. While the dedifferentiation process of mature lineages does not depend on Hh signaling, the proliferation and the migration of the dedifferentiated cells are dependent on Hh signaling. Temporally controlled chemical inactivation of the Hh pathway indicates that Hh-mediated antero-posterior (AP) specification occurs early during limb regeneration and that Hh is subsequently required for expansion of the blastemal progenitors. Inhibition of Hh signaling results in G0/G1 arrest with a concomitant reduction in S-phase and G2/M population in myogenic progenitors. Furthermore, Hh inhibition leads to reduced Pax7-positive cells and fewer regenerating fibers relative to control tissue. We demonstrate that activation of Wnt signaling rescues the inhibition of Hh pathway mainly by enhancing proliferative signals, possibly mediated through TCF4 activity. Collectively, our results demonstrate coordinated signaling of Hh and Wnt activities in regulating blastemal progenitors and their hierarchical positioning during limb regeneration.  相似文献

A spatiotemporal pattern of cell death occurred in the chick wing and leg bud mesoderm after removal of apical ectodermal ridge at stages 18–20. Cells died in a region extending from the limb bud distal surface to 150–200 μm into the mesoderm. Limb buds from which ridge was removed at later stages in development did not exhibit a spatiotemporal pattern of cell death. In control experiments in which dorsal ectoderm was removed, a pattern of cell death did not occur. Removal of the ridge and part of the 150- to 200-μm zone of prospective cell death resulted in cell death in an area approximately equal to the amount of the zone remaining. After removal of all of the prospective zone of cell death plus the apical ridge, cell death was observed in the remaining limb bud mesoderm. In these limb buds, cell death occurred in a region in which it had not been seen in limb bud with apical ridge alone removed. We conclude that at stages 18–20 the mesodermal cells 150–200 μm beneath the ridge require the apical ridge to survive. More proximal mesodermal cells do not die after ridge removal alone, but apparently require the presence of the more distal mesoderm to survive. Whether this is a requirement for something intrinsic to the distal mesoderm or something it possesses by way of the ridge is unknown. After stage 23, the limb mesoderm cells do not die when the apical ridge is removed. Nevertheless, at the later stages, ridge continues to be required for limb bud proximal-distal elongation and the differentiation of distal limb elements.  相似文献

It has been established that X-ray irradiation localized to a forelimb or entire irradiation of premetamorphic Pleurodeles larvae prevented limb regeneration. Transplantation of non-irradiated skin, dermis or muscle to limb stumps of locally irradiated newts was sufficient to allow a blastema to develop. Transplantation of the same tissues to limb stumps of entirely irradiated newts yielded different results with the different graft types. Skin graft allowed a normal blastema to be established but dermis or muscle grafts did not. In order to define more precisely the role played by the epidermis in the establishment of a blastema, and in the growth of a regenerate, different combinations of limb tissues, either irradiated or not, were carried out at the level of amputated limb stumps. At four different times (8-10 days; 13-15 days; 20-23 days; 30 days or more) after amputation the stumps were examined in histological longitudinal sections to study the first events of regeneration, that is dedifferentiation and growth. Dedifferentiation occurred in both normal and irradiated tissues of mesodermal origin. The healthy mesenchymal cells began dividing and formed a growing blastema only when associated with a non-irradiated epidermis. Healthy mesenchymal cells covered with an irradiated epidermis exhibited a few mitoses after dedifferentiation, but the mitotic figures became rarer and rarer until the animals died. The lack of dense accumulation of blastemal cells in such limb stumps suggested that the healthy epidermis allows the mesenchymal cells to divide actively to constitute a growing blastema. Hence, X-ray irradiation seems to be responsible for the loss of such an epidermal mitogenic influence on the underlying mesenchymal cells.  相似文献

Summary Embryonic and adult field do not exhibit the same properties because they are formed by cell populations whose activities cannot be compared in all respects. Dominant cells are not distributed in the same manner; the same mechanisms are not involved for remodelling of the field, especially after an amputation.In adult organisms, cell equilibrium is disturbed a first time by elimination of differentiated tissues which provokes dedifferentiation and proliferation, and a second time by healing, which restores cell equilibrium of a distal end. Undifferentiated cells begin to differentiate immediately after their production and they reform the missing levels, following a sequence either basipetal or basifugal according to the place of their production (the stump or an apical proliferation center). The differentiating regenerate temporarily exhibits embryonic field properties in various experimental conditions which suppress the dominance of the stump or supplies additional undifferentiated material.  相似文献

Urodele amphibians are unique adult vertebrates because they are able to regenerate body parts after amputation. Studies of urodele limb regeneration, the key model system for vertebrate regeneration, have led to an understanding of the origin of blastema cells and the importance of positional interactions between blastema cells in the control of growth and pattern formation. Progress is now being made in the identification of the signaling pathways that regulate dedifferentiation, blastema morphogenesis, growth and pattern formation. Members of the Wnt family of secreted proteins are expressed in developing and regenerating limbs, and have the potential to control growth, pattern formation and differentiation. We have studied the expression of two non-canonical Wnt genes, Wnt-5a and Wnt-5b . We report that they are expressed in equivalent patterns during limb development and limb regeneration in the axolotl ( Ambystoma mexicanum ), and during limb development in other tetrapods, implying conservation of function. Our analysis of the effects of ectopic Wnt-5a expression is consistent with the hypothesis that canonical Wnt signaling functions during the early stages of regeneration to control the dedifferentiation of stump cells giving rise to the regeneration-competent cells of the blastema.  相似文献

Distal and proximal mesoderm of chick limb bud was respectively dissociated and cultured in the medium containing various concentrations of retinoic acid (RA). At low concentrations (5-50 ng/ml), RA promoted proliferation and chondrogenesis in the distal mesodermal cells. The distal cells of stage 20-24 limb buds were responsive to RA, although those of stages 25-27 were unresponsive. Both the cells of anterior and posterior regions of the distal mesoderm were responsive to RA, while the cells of proximal mesoderm were unresponsive. At higher concentrations, the growth-promoting effect of RA was reduced and chondrogenesis in the distal cells was rather inhibited. These results were discussed in relation to the role of RA as the morphogen in normal limb development and experimental duplicate formation.  相似文献

The salamander limb regenerate develops from a mesenchymatousbud of cells called the blastema. This review briefly considersthe local origin and major features of blastemal cells and thendiscusses their role in the histogenesis of the regenerate.The capacity of blastemal cells to form the various cell typesof the regenerate is first considered in general terms, withthe discussion focusing on three major alternatives: blastemalcells form (a) cartilage only, (b) connective tissues only,or (c) essentially all internal tissues of the regenerate. Thedifferentiative properties of blastemal cells originating fromknown cell types are then considered in detail. Cartilage isof particular interest in this regard, because it has been utilizedas a pure cell type. Results from the study of mixed tissues,including muscle and connective tissue of the fin, are alsopresented. Information gained from the study of known cell typesparticipating in regeneration is discussed in terms of the mechanismsresponsible for the stability of cellular differentiation andin terms of the flow of cells from stump tissues into the regenerate.  相似文献

Summary 1. Undifferentiated fore limb blastemas were denuded of their epidermis and grafted to the flank musculature; each transplant consisted of four such blastemas. Outgrowth of the mesenchyme and subsequent digit formation were prevented by covering the transplants with whole flank skin with its dermis intact. 2. Notwithstanding the absence of digits, 26 out of the 36 differentiated transplants formed one or two oblong, stout cartilages resembling proximal skeletal elements of the limb. Considering that four blastemas had been grafted in random orientation, this indicates considerable intrinsic morphogenetic capacities of the blastemal mesenchyme in the absence of organizing influences of the epidermis. 3. Thus, although permanent contact with wound epidermis has previously been shown to be necessary for blastemal mesenchyme to form distal limb structures, such contact is not required for the formation of proximal skeletal elements.
