The essential oil from the fruits of Trachyspermum ammi exhibited toxicity at 800 ppm against Aspergillus flavus and A. niger, the nature of toxicity being cidal. The toxicity of the oil was not affected by autoclaving, temperature treatment and storage upto 365 days. The oil killed the test fungi within 50 seconds; withstood heavy inoculum density and was inhibitory to as many as 21 fungi at its minimum inhibitory concentration. However the seeds of Arachis hypogea whentreated with oil at 5000 ppm and stored for 12 months did not show the appearance of any fungi indicating thereby the grain protectant activity of the oil. The oil was characterized by various physico chemical properties and on chemical investigation Thymol and p-cymene were isolated as antifungal principles of the oil exhibiting toxicity against the test fungi at 1000 ppm.  相似文献

Summary The essential oil extracted from the epicarp of Citrus sinensis exhibited absolute fungitoxicity against the 10 post-harvest pathogens. GC–MS studies of the oil revealed the presence of 10 chemical constituents, of which limonene was found to be the major component (84.2%). The activity of the oil was tested by the poisoned food technique (PF) and the volatile activity (VA) assay and the oils showed greater toxicity in the VA assay than in the poisoned food assay. The nature of the toxicity was studied in the VA assay and it was observed that the oil was fungicidal for the 10 pathogens in the 700 ppm (mg/l) to 1000 ppm range. The oil was extremely toxic for spore germination and it was found that at 700 ppm, spore germination was inhibited in the 10 test fungi out of the 12 tested. Treatment at 300 ppm concentration exhibited 70–100% inhibition of spore germination in most of the fungi tested. Scanning electron microscopy (SEM) was done to study the mode of action of the oil in Aspergillus niger and it was observed that treatment with the oil leads to distortion and thinning of the hyphal wall and the reduction in hyphal diameter and absence of conidiophores.  相似文献

Summary Leaves ofChenopodium ambrosioides exhibited strong fungitoxicity against the mycelial growth ofRhizoctonia solani causing damping off diseases of some seedlings. Minimum inhibitory concentration of the fungitoxic constituent isolated in form of essential oil, was found to be 1000 ppm at which it was fungicidal in nature. It exhibited broad range of antifungal activity and did not show any phytotoxicity on the germination and seedling growth ofPhaseolus aureus.  相似文献

In isolation tests on different parts of sound Hinoki cypress (Chamaecyparis obtusa), the coelomyceteCryptosporiopsis abietina was isolated, mostly from immature cone scales and inner bark areas of branches and main stems. In isolation tests using branches or stems of 3-yr-old seedlings to 35-yr-old adult cypresses from various localities,C. abietina was isolated in 28 to 100% of the cypresses tested. It was also confirmed that the fungus rarely infects seeds of the cypress, and can survive in dried seeds for several years, though it is not a seed-borne fungus. No symptoms appeared in inoculation tests with the fungus on 8-yr-old cypress. In inoculation test in a greenhouse, reliable infection without symptoms was obtained on wounded cypress seedlings. These results reveal thatC. abietina is a common endophytic fungus in Hinoki cypress. Confrontation and volatile antagonistic tests in Petri dishes indicated that some isolates ofC. abietina produce antifungal substances in vitro. On these isolates, the reproduction of a mycophagous nematodeBursaphelenchus xylophilus was retarded. The results of inoculation tests suggest that the possibility ofC. abietina as a primary causal agent of the resinous stem canker of Hinoki cypress is low. The role ofC. abietina in the cypress is briefly discussed.  相似文献

The insecticidal effect of the entomopathogenic fungus Beauveria bassiana (Balsamo) Vuillemin (Ascomycota: Hypocreaes) in combination with three diatomaceous earth (DE) formulations against adults of the granary weevil, Sitophilus granarius (L.) (Coleoptera: Curculionidae) was tested in the laboratory. The three DEs were Insecto™, SilicoSec® and PyriSec®. The fungus was applied at 400 ppm alone, or in combination with 200 ppm of each of the three DEs. Mortality was measured after 7 d of exposure. Bioassays were conducted at three temperatures 20, 25 and 30 °C and two relative humidities (rh) 55% and 75%. On wheat treated with B. bassiana alone, mortality was higher at 55% than at 75% rh. Also, the fungus alone was less effective at 20 °C than at the other two temperatures tested, but mortality did not exceed 52% for any of the conditions tested. Similar mortality levels were also noted on wheat treated with each of the three DEs alone. The simultaneous presence of B. bassiana and DE increased weevil mortality. In this combination, mortality was higher at high temperatures and low rh, and this effect was similar for all DEs tested. Progeny production on wheat treated with B. bassiana was higher that the respective progeny counts in the DE-treated wheat. The results indicate that a combination of B. bassiana and DEs is effective against S. granarius, under a broad range of temperature and rh levels in stored wheat.  相似文献

The essential oil of Caesulia axillaris has exhibited its fungitoxicity against Aspergillus flavus at its minimum inhibitory concentration of 1300 mg/l. It showed the potentiality of an ideal fungitoxicant because of its long shelf life, thermostable nature, broad fungitoxic spectrum and persistence of fungitoxicity even on introduction of high inoculum density of the test fungus. The fungitoxic principle of the oil was standardized as -asarone which showed fungitoxicity against the test fungus at 500 mg/l.  相似文献

Various plant parts ofIndigofera tinctoria L. were collected separately at different growth stages and analysed for their rotenoid content. The total rotenoid content decreased with age; among the plant parts, maximum content was in leaves and minimum in stem. The identity of different rotenoids was confirmed by melting point, mixed melting point, UV and infrared spectral studies, and gas-liquid chromatography. Six rotenoids (deguelin, dehydrodeguelin, rotenol, rotenone, tephrosin and sumatrol) were isolated, identified and quantified invivo. The static cultures ofIndigofera tinctoria were established from seeds on RT medium, and maintained for a period of six months by frequent subculturings. Only four rotenoids were present in callus cultures; sumatrol and tephrosin were absent. The maximum content was found in eight week old tissue after fresh subculturings and minimum at 2 weeks. The toxicological studies ofin vivo andin vitro extract against the pulse beetle(Callosobruchus chinensis) and mosquito(Anopheles stephensi) larvae, showed that rotenoids were more effective against mosquito larvae thanCallosobruchus chinensis. Extracts from callus was more effective against both the test animals than that from plant parts.  相似文献

An ascomycete fungus causing an undescribed disease was found on an oak,Quercus phillyraeoides. Naked hymenia appeared on the lower side of leaves, causing slightly swollen spots or blisters that exhibited chlorosis. Ascogneous cells were thick-walled, oval and convex. Asci were cylindric or clavate, without a stalk cell. Ascospores were rarely observed, and numerous blastospores were formed in asci. Mycelium habit was intercellular. Blastospores grew well on PDA and formed pinkish yeast-like colonies. Morphological aspects of the fungus matched well with those ofTaphrina caerulescens. Quercus phillyraeoides is recorded as a new host ofT. caerulescens, and the disease ofQ. phillyraeoides caused by this fungus is termed “leaf blister.”  相似文献

AnAscochyta fungus was obtained during a survey of leaf spot diseases ofFagus crenata in the mountains of Aomori prefecture in 1995. The pathogenicity of the fungus toF. crenata was confirmed by inoculation. Its teleomorph was first found on artificially infected leaves after the leaves were placed in an incubator at 5°C with a 12-h photoperiod (approximately 500lx, daylight strip lamps) for 4 mo. The fungus was found to be the causal agent of yellow leaf spots ofF. crenata andQuercus mongolica var.grosseserrata in the field. Comparison with similar fungi so far described from Fagaceae indicated that the anamorph isAscochyta fagi and the teleomorph is an undescribed species ofDidymella. The nameDidymella fagi is introduced for the teleomorph. Correlation between the two morphs has been proved by cultural, morphological and pathological studies.  相似文献

The leaves of Ocimum gratissimum (Clocimum) exhibited strong volatile fungitoxicity against betelvine (Piper betle L.) pathogens—Alternaria alternata, Colletotrichum capsici and Sclerotium rolfsii. Fifteen compounds could be identified from the fungitoxic constituents—the essential oil. The oil at its minimum inhibitory concentrations of 50, 250 and 500 ppm against S. rolfsii, A. alternata and C. capsici, respectively, was fungistatic, although, fungicidal at higher concentrations. Eugenol was found to be the major fungitoxic principle in the oil. The oil was either equally effective or superior to synthetic commercial fungicides and was non-phytotoxic to the host plants. Thus, the oil can be used as a valuable indigenous and biodegradable agent against fungi that cause losses to the betelvine industry.  相似文献

Puccinia kraussiana, a rust fungus of Africa, was found in Taiwan for the first time in eastern Asia. It was parasiting on leaves ofHeterosmilax gaudichaudiana, on which all four stages were characteristically found. The host species,H. gaudichaudiana, newly added to the host range ofP. kraussiana, constitutes also a new entry to the phanerogamic flora of Taiwan.  相似文献

A leaf spot disease called frosty mildew was observed onChaenomeles sinensis throughout Japan. Small brown spots with white tufts occurred followed by successive defoliation. On the fallen leaves, minute black dots are formed. The causal fungus was regarded as a new species ofMycosphaerella, M. chaenomelis, andCercosporella chaenomelis in anamorph. Pathogenicity of the fungus was confirmed only inC. sinensis by inoculation experiments. Colonies of the fungus grew well on potato sucrose agar, and grew at 10–30°C with an optimum temperature of 25°C. The fungus overwintered on the fallen diseased leaves in the form of pseudothecia, and ascospores served as the primary infection source from April to June.  相似文献

The causal fungus of frosty mildew inQuercus acutissima was taxonomically studied. Brown spots with conical white to yellowish tufts of the causal, fungus developed on the leaves, followed by successive defoliation. The fungus was regarded as a new species ofMycopappus, M. quercus, from the morphology of the multicellular propagules. The ascogenous stage and true conidia of the fungus were not observed, although sclerotia were formed on the fallen diseased leaves.  相似文献

The repellency of the essential oil of the shrubCleome monophylla (Family: Capparidaceae) and identified constituents of the oil were evaluated against the livestock tick,Rhipicephalus appendiculatus and the maize weevil,Sitophillus zeamais. In a tick climbing repellency bioassay, the oil ofC. monophylla exhibited repellency which, at the highest dose, was comparable to that of the commercial arthropod repellent N,N-diethyl toluamide (DEET). In a Y-tube olfactometer bioassay,C. monophylla oil showed higher or comparable repellency againstS. zeamais relative to DEET at all the doses tested. 14 Compounds were identified in theC. monophylla oil by GC, GC-MS and coinjection with authentic samples. Terpenolene was found to occur in largest quantity (14%) followed by 1-α-terpeneol (10%), pentacosane (9%), (α+β)-humulene (8%), phytol (5%) and 2-dodecanone (4%). The most repellent components againstR. appendiculatus andS. zeamais were 1-α-terpeneol and 2-dodecanone. The overall pattern of repellency activity of theC. monophylla constituents with respect to the two arthropods was, however, different. The potential ofC. monophylla in tick and maize weevil control is discussed.  相似文献

Potassium sorbate, sodium benzoate, sulfur dioxide, and diethylpyrocarbonate (DEPC) were tested for their effectiveness in preventing the outgrowth ofByssochlamys nivea Westling ascospores. Sulfur dioxide was the most inhibitory of the test antimycotics, complete inhibition of colony formation occurring in acidified (pH 3.5) potato dextrose agar containing 50 ppm of the preservative. Complete inhibition ofB. nivea ascospore outgrowth in grape juice stored for 60 days was noted in the presence of 300 ppm sulfur dioxide, 400 ppm potassium sorbate, and 600 ppm DEPC. Growth was observed in grape juice containing 1000 ppm sodium benzoate. The presence of up to 100 ppm potassium sorbate in grape juice during heat activation appears to have a stimulatory effect on breaking dormancy, while the other test preservatives at this concentration decrease the heat resistance ofB. nivea ascospores. The time elapsed between heat shock and exposure to DEPC or sodium benzoate is critical with respect to the sensitivity of ascospores to these preservatives.  相似文献

Camellia leaves were most effective for recoveringPhytophthora cinnamomi from soils followed by azalea leaves and cedar needles. A total of 131 isolates ofP. cinnamomi was obtained from soils and roots collected in Shanghai. Among them 125 were A1 and 6 were A2 type. There was little variation in morphological and physiological characteristics among 82 isolates tested. It is suggested that the fungus may have been a recent settler in Shanghai.  相似文献

A species ofMycosphaerella with aPseudocercospora anamorph was collected on overwintered fallen leaves of Japanese beech,Fagus crenata. Based on comparison of morphology withMycosphaerella species on Fagaceae, the fungus was newly described asMycosphaerella buna. Beauveria bassiana strain EABb 04/01-Tip isolated from stem-borer larvae of Timaspis papaveris (Hymenoptera: Cynipidae), a serious pest of opium poppy in Spain, was shown to be able to become established endophytically in this pharmaceutical crop. Microbiological, molecular and light and electron microscopic methods were used to study fungal colonisation and to describe its mode of penetration. After inoculation with a foliar spray of conidia, microbiological methods showed 100% of plants examined 24, 48, 72 and 144 h after treatment to be colonised endophytically by the fungus, although the percentage of previously surface sterilised leaf pieces showing fungal growth was 100% at 24 and 48 h, and 80 and 75% at 72 and 144 h after treatment, respectively. The fungus was also observed in leaf pieces obtained from newly formed leaves, indicating that it could spread from treated leaves to leaves formed after fungal application. For molecular studies, a polymerase chain reaction (PCR) protocol was used to amplify the ITS1-5.8S-ITS2 regions of the rDNA of the plant and the fungus. This procedure allowed the detection of the fungus on the surface of the leaves and also endophytically, but only at 72 h after treatment. A nucleotide BLAST search revealed that the ITS1-5.8S-ITS2 sequence of strain EABb 04/01-Tip showed 100% homology with a similar sequence from Cordyceps bassiana. SEM images revealed that although numerous conidia were observed on the leaf surface, few germinated and penetrated. Intracellular colonisation by B. bassiana was not observed, but hyphae were detected growing into the xylem vessels. The fungus was found to colonise 40.5 ± 4.3% of seedlings (with two cotyledons and the two first real leaves) from seeds dressed with a fungal spore suspension. These results may have implications in the biological control of T. papaveris, including the possible systemic protection of the plant against this cynipid.  相似文献

To exploit natural products for plant disease control, the essential oil of Syzygium aromaticum (L.) Merr. & Perr. (clove) was investigated for its antifungal activity against Rhizopus stolonifer and Fusarium solani, the postharvested yam tuber rot pathogens. The essential oil was obtained by hydrodistillation using a Clevenger‐type apparatus. The chemical composition of the oil was determined by gas chromatography and gas chromatography coupled with mass spectrometry. Antifungal activities of the oil were tested in vitro against mycelia growth and spores germination. In situ tests were conducted on healthy yam tubers, and necrosis symptoms were assessed. Results showed that eugenol (79.4%), eugenylacetate (9.2%) and isocaryophyllene (7.0%) were the major components. The oil exerted antifungal activities with total inhibition (TI) of the mycelia growth of R. stolonifer and F. solani was recorded at 200 and 300 ppm, respectively, while TI of spores germination was recorded at 31.25 and 250 ppm, respectively. For the standard fungicide (Ridomil®), TI value of mycelia growth was 1600 ppm for the both pathogens, while TI of spores germination were 200 ppm and 1600 ppm, respectively, for Rhizopus and Fusarium. In situ tests showed complete inhibition of yam tuber rot when the essential oil was applied at 2000 ppm for preventive tests. This oil also reduced significantly (P ≤ 0.05) necrosis development on yam tuber for curative test at the same concentration. Total inhibition of the necrosis by Ridomil (3000 ppm) was observed only for Rhizopus on preventive test. There were positive correlations between the oil concentration and the reduction of necrosis cause by R. stolonifer and F. solani. These findings showed that clove essential oil may serve as environmental friendly bio‐fungicide for the management of postharvest yam tuber rot.  相似文献

β-Glucosidase and β-galactosidase activity profile tested in different seeds during 24 h germination revealed reasonably high levels of activity inVigna radiata, Cicer arietinum, andTrigonella foenum-graecum. In all seeds tested, β-galactosidase activity was, in general, higher than that of β-glucosidase.T. foenum-graecum seedlings exhibited maximal total and specific activities for both the enzymes during 72 h germination. Se supplementation as Na2SeO3 up to 0.75 ppm was found to be beneficial to growth and revealed selective enhancement of β-galactosidase activity by 40% at 0.5 ppm Se. The activities of both the enzymes drastically decreased at 1.0 ppm level of Se supplementation. On the contrary, addition of Na2SeO3 in vitro up to 1 ppm to the enzyme extracts did not influence these activities. Hydrolytic rates of β-glucosidase in both control and Se-supplemented groups were enhanced by 20% with 0.05M glycerol in the medium and 30% at 0.1M glycerol. The rates were marginally higher in Se-supplemented seedlings than the controls, irrespective of added glycerol in the medium. In contrast, hydrolysis by β-galactosidase showed a trend of decrease in Se-supplemented seedlings compared to the control, when glycerol was present in the medium. Addition of Se in vitro in the assay medium showed no difference in the hydrolytic rate by β-galactosidase when compared to control, while the activity of β-glucosidase declined by 50%. Se-grown seedlings showed an enhancement of transglucosidation rate by 40% in the presence of 0.1M glycerol. The study reveals a differential response to Se among the β-galactosidase and β-glucosidase ofT. foenumgraecum with increase in the levels of β-galactosidase activity.  相似文献
