Growth regulators, indole-3-acetic acid (IAA), gibberellic acid (GA3), and kinetin (KIN), were used in different concentrations to stimulate the initiation and further development of the fruiting bodies of the fungusLentinus tigrinus. Vegetative mycelium of the fungus was cultivated on cellulose cylinders soaked with a synthetic nutrient solution or with a 3% malt extract. When the mycelium covered the surface of the cylinders, further cultivation was carried out in graduated concentrations of the growth regulators mentioned above. The number of developed fruiting bodies showed that the optimum IAA and GA3 concentrations were in both media 300 p.p.m. The optimum concentration of kinetin was 400 p.p.m. The individual growth regulators influenced characteristically also the shape of the fruiting bodies and changed in them the natural level of endogenous growth regulators. The addition of IAA into the medium raised the level of endogenous auxins in the cap. The presence of gibberellic acid and of kinetic in the medium resulted in an increase in the level of these regulators in the fruiting bodies.  相似文献

The effect of indol-3-ylacetic acid, gibberellie acid, kinetin, abscisic acid and Ethrel on the growth of mycelium, sporulation and germination of spores ofFusarium culmorum of different pathogenicity to wheat seedlings was studied. The production of gibberellins, auxins, cytokinins, ethylene and growth inhibitors by these isolates was determined as well. It has been found that most pronounced and explicit effect on growth and development in fungi was produced by Ethrel which strongly inhibited these processes. ABA proved to be a strong growth and development stimulator, though to a different extent in different isolates. GA3 strongly stimulated sporulation and spore germination in some isolates. The effect of IAA and K on growth and development in fungi was slight. More sensitive to growth regulators were the fungi in earlier stages of growth. No correlation between the pathogenicity of the isolates and their ability to produce growth regulators as well as between their susceptibility to exogenous growth substances in the processes of fungal growth and development was stated. Summary The response of groundnut cotyledons to the presence of various growth regulators in concentrations from 0.1 to 5 mg/l has been studied in detail using several genotypes of groundnut on two different media. Cotyledons with embryo axis, cultured on Blaydes' medium with cytokinins, produced shoots, in the axils of which 2–7 flower buds could be seen. The frequency of flower bud induction in general increased with increasing concentrations of cytokinins, the optimal levels being 3 mg/l of KN or 4 mg/l of BAP. Cotyledons without embryo axis, cultured on Blaydes' medium with BAP (0.5 mg/l), produced a cluster of flower buds directly, ranging in number from 8–28, without any vegetative growth. Excised embryo axes cultured on the same medium gave plantlets without flower buds. The growth regulators IAA, NAA, GA3 and ABA failed to induce flower buds in independent treatments. However, lower concentrations of IAA and NAA in combination with cytokinins exerted a positive influence on flowering. The blooming of the flower buds was facilitated on media supplemented with low concentrations of cytokinins. Six percent of the induced flowers resulted in gynophore development and ultimately formed pods when cultured under complete dark conditions in modified MS medium supplemented with kinetin.  相似文献

Summary On a simple nutrient medium in explants from roots of Cichorium intybus form shoots visible after about 14 days. Gibberellic acid (GA3) does not influence the spontaneous development of the chicory explants. GA3 in combination with kinetin inhibits shoot formation whereas kinetin alone promotes the process. On the other hand high concentrations of IAA inhibit the regeneration of shoots.The soluble proteins of chicory cultures treated with growth regulators were examined by disc-electrophoresis. It was shown that the proteins detected by staining with Amido black, phosphatases, esterases and glutamate dehydrogenase (GDH) present in the original root tissue remained constant under the different culture conditions during a period of 12 days. The quantitative changes of some of the proteins, phosphatases and esterases observed during the culture period were identical for all the different cultures in spite of the different morphogenetic behaviour. Only the activities of GDH and peroxidase changed after treatment with different growth regulators; however, in these cases, there was also no direct connection with the morphogenetic responses of the cultures.The specific activity of the GDH-band was promoted by IAA and at the same time the formation of peroxidases was inhibited. Kinetin delayed the formation of peroxidases during the first days of the culture period but promoted it later on. There was a repression by IAA of a specific kationic peroxidase. In the tissues treated with GA3 the activity of peroxidases was always higher than in the control tissue. This effect of GA3 can be partly explained by the fact that GA3 inhibits the release of peroxidases of the explants into the nutrient medium.  相似文献

The fungusLentinus tigrinus was cultivated in a synthetic medium. During vegetative growth of mycelium and differentiation of fruiting bodies samples serving for determination of endogenous growth regulators were collected. The material was extracted with methanol, separated by chromatography and the isolated fractions were tested. Substances of auxin type were assayed with coleoptile segments of oat plants, gibberellins by measuring the hypocotyl growth in the lettuce and cytokinins from the degree of chlorophyll decomposition in the leaf segments of barley. Very low content of auxins was found in all phases of growth and differentiation of the fungus. The content of gibberellin substances was relatively high during the whole life of the fungus. The highest content is found in the vegetative mycelium and in the differentiated fruiting body, especially in the cap; it decreases during the differentiation of fruiting bodies. The content of cytokinins is equally high and the pattern of its change resembles that of the gibberellins. Its maximum is reached in young vegetative mycelium, it decreases when fruiting bodies are formed and when they are growing. Minimum is reached in the differentiated stalk, where also the amount of inhibitions is the highest. Experimental results show that during differentiation of fruiting bodies and their morphogenesis not only gibberellins and cytokinins are of importance, but also substances of an inhibitory character. A relatively high amounts of these substances was secreted into the cultivation medium. In a synthetic nutrient solution they may be easily demonstrated.  相似文献

Phytohormones such as auxins, cytokinins, gibberellins, andabscisic acid differentially affect gametangial induction inmale and female clones of Bryum argenteum. Both IAA and GA3increased the percentage of fertile gametophores in the maleclone, and inhibited the response in the female clone. GA3 wasmore effective than IAA in eliciting the response in the maleclone. Cytokinins, on the other hand, increased the productionof fertile gametophores in the female clone, and inhibited itslightly in the male clone. The two cytokinins tested (kinetinand DMAAP) were almost equally effective for the female clone. An Interaction of IAA and kinetin nullified their individualinhibitory effects on the female and male clones, respectively.Cyclic AMP prevented the inhibitory effect of kinetin in themale clone; whereas, in the female clone, it stimulated theresponse elicited by kinetin. Abscisic acid (ABA) acted as ageneral inhibitor of vegetative growth and gametangial inductionin this moss. However, the inhibition of gametangial inductionwas greater in the female clone which is also more sensitiveto ABA than the male clone.  相似文献

The character of endogenous growth regulators in different stages of growth and development ofLentinus tigrinus was followed by means of bioassays. The methanolic extract of differently old mycelium and fruiting body was chromatographically separated and tested for auxins by means of Avena coleoptile segments, for gibberellins by using lettuce seedlings and for cytokinins by measuring the content of chlorophyl in leaf segments of barley. Auxins were found only in young vegetative mycelium and in the growing young stalk. The level of gibberellins is more significant and it reaches the highest values in the vegetative mycelium, in the growing young stalk and in the differentiated cap. The content of auxin and gibberellin substances did not increase even in the medium after cultivation of mycelium. Bioassays of cytokinins show a slight stimulation in the vegetative mycelium and in the primordia of caps. Significant inhibition was ascertained in the fully developed stalk. The high quantities of cytokinins were found in caps when basidiospores were formed. The results show a close connection of endogenous growth substances with the growth and the development of the fungus and indicate their participation in differentiation processes.  相似文献

The aim of the experiments presented here was twofold. On the one hand, to study the influence of plant growth regulators, i.e. auxins (IAA and NAA), cytokinins (kinetin, zeatin and zeatin riboside), gibberellins: (GA3) and polyamines (spermidine, spermine and putrescine) on the generative development of winter wheat plants, while on the other to study the interaction of plant growth regulators with electric current in affecting the efficiency of the flowering of wheat plants. Winter wheat var. Grana was subjected to a short vernalization (2 weeks at 5°C + 10 days at 10°C) on Murashige and Skoog medium containing growth regulators at different concentrations. For selected substances, seedlings were additionally treated with electric current by applying a constant voltage for a given period of time (30 V for 30 s or 1.5 V for 1 h) with the anode or the cathode inserted into apical leaves and the reference electrode into the media. After that, the seedlings were transferred to a glasshouse (20/17°C) where they were grown until heading was achieved. All the substances studied stimulated the generative development of winter wheat, in contrast to non-treated plants (control), especially at their highest concentrations. Spermidine, kinetin and GA3 were the most effective in this process as they stimulated 100–80% of generative plants (whereas for the control the percentage stood at only 30% plants). The studied regulators also increased the rate of generative development, i.e. shortening the length of the vegetative phase (by about 30% in comparison with the control). Moreover, these substances treatments decreased the fresh mass of both seedlings and flowering plants and increased the number of spikelets in the ear. The electric current treatment interacted with the applied substances: the anode insertion into the leaves generally increased, whereas the cathode insertion decreased, the inductive effect of the growth regulators on generative development. However, the inhibition of the flowering process was more pronounced than the induction. Moreover, the passage of electric current shortened the time to heading. This decrease in the length of the vegetative phase was more visible after the anode was inserted into the leaves. The least noticeable effect of the electric current interaction with plant growth regulators on both the generative development and the time of heading was observed for auxins.  相似文献

Unfertilized and fertilized ovules of Gossypium barbadense Giza 45 (extra long staple variety) were used to study the effect of plant growth substances (auxins, gibberellins and cytokinins) on in vitro fiber initiation and development. Kinetin, alone did not increase total fiber unit (TFU) of unfertilized ovules, while an increase in TFU value occurred when a constant level of IAA and GA3 were used either separately or in combination in the liquid medium. GA3 used alone, produced a higher TFU value than that produced by IAA, whilst, IAA with a constant level of GA3 (5 M) produced the highest value of TFU. GA3 with a constant level of IAA (5 M) produced a lower TFU value. Kinetin reduced the stimulatory effect of IAA and GA3 on TFU value when used in combination with either substance. In fertilized ovules, the highest level of TFU was reached when IAA, with a constant level of GA3, was added to the medium, whilst its lowest level was obtained when IAA was used alone. Estimation of in vitro fiber production, as well as the effect of growth substances used in different concentrations on in vitro fiber initiation and development from unfertilized and fertilized ovules of Egyptian cotton varieties Gossypium barbadense Giza 45 are discussed.  相似文献

Treatment of different plant materials, seeds of Phaseolus vulgaris, Zea mays and Pinus silvestris and young plants of Phaseolus, with kinetin increased the level of extractable IAA. For seeds this increase was most pronounced in bean seeds, which contained the lowest amount of endogenous IAA and cytokinins, and lower in maize seeds with high endogenous content of IAA and cytokinins. – For young bean plants the kinetin treatment significantly increased the extractable amounts of IAA from all parts of the plant, hypocotyls, cotyledons, epicotyls and primary leaves, when the cut plants were placed for 24 h in kinetin solution. For plants sprayed with kinetin solution only the primary leaves showed a significantly higher level of extractable IAA, which could be explained by the fact that the plants were growing very close together, so that the primary leaves received most of the kinetin during spraying.  相似文献

Summary The effect of 3-indoleacetic acid (IAA), 6-furfurylaminopurine (kinetin), and gibberellic acid (GA3) on germination of the orchid Comparettia falcata was evaluated in a factorial experiment (4×4×4) with Murashige and Skoog (1962) basal medium. It was established that seeds of this orchid could be maintained under aseptic conditions as long as the necessary nutrients and appropriate concentrations of growth regulators were provided. Of the three growth regulators used, IAA significantly decreased seed germination of Comparettia falcata. There was a synergistic effect in the kinetin:GA3 combination that produced a positive response in both percentage seed germination and development of seedlings. This study describes a single medium-based protocol able to achieve more than 160000 seedlings within 21 wk, starting from a single capsule, sufficient for both large-scale propagation and in vitro conservation of this threatened orchid.  相似文献

The effect of GA3 on somatic embryogenesis from petiole fragments excised from micropropagated fennel plantlets was studied. Explants were maintained for 4 weeks on an induction medium containing, 2,4-d and kinetin and were then transferred to a medium devoid of these growth regulators to allow embryo development. The addition of autoclaved or filter-sterilized GA3 to the induction medium or to the embryo development medium increased the number of embryogenic explants. No positive effect was observed when GA3 was added to the micropropagation medium of the mother plantlets. Gibberellic acid also counteracted the inhibiting effect of continuous light on the number of embryogenic explants. The percent germination and seedling growth of chickpea (Cicer arietinum L. cv. PBG-1) decreased with increasing concentrations of exogenous polyethylene glycol 6000 (PEG). With 15% PEG in the growth medium germination was only 33% while with 10% PEG it was 58% as compared to 93% in control. Addition of gibberellic acid (GA3) and kinetin to medium containing 10% PEG increased the germination and seedling growth and the effect was maximum with 6 µM GA3 which was a better inducer of growth and germination under reduced water potential than kinetin. However, indole acetic acid (IAA) inhibited germination and growth of stressed seedlings. The activity of amylase in cotyledons under stress was significantly increased with GA3 while kinetin and IAA were less effective. Gibberellic acid also enhanced the mobilization of starch from cotyledons of stressed seedlings resulting in low starch levels in cotyledons compared with stressed seedlings.  相似文献

The interrelationship between ethylene and growth regulators in the senescence of romaine lettuce (Lactuca sativa L.) leaves was studied. Gibberellic acid (GA3), kinetin, and 3-indoleacetic acid (IAA) retarded chlorophyll loss from leaf discs which were floated on hormone solutions. Abscisic acid (ABA) and ethephon enhanced chlorophyll loss and antagonized the senescence-retarding effect of GA3 and kinetin. A high concentration of IAA (10–4 M) caused accelerated chlorophyll loss, whereas a similar concentration of kinetin neither retarded nor promoted chlorophyll loss. The ineffectiveness of IAA and kinetin at their supraoptimal concentrations in retarding leaf senescence was related to increased production of ethylene induced in the treated leaf discs. GA3 was the most effective in retarding chlorophyll loss and did not stimulate ethylene production at all. The senescence-enhancing effect of ABA was not mediated by ethylene. However, the moderately increased production of ethylene, induced by relatively high concentrations of ABA, could act synergistically with the latter to accelerate chlorophyll loss. The effect of growth regulators on induction of androgenesis in coconut was investigated using seven different growth regulators at various concentrations and combinations. Three auxins (1-naphthalene acetic acid—NAA, indoleacetic acid—IAA, picloram) and three cytokinins (2-isopentyl adenine-2-iP, kinetin, zeatin) were tested either alone or in combination with 2,4-dichlorophenoxyacetic acid (2,4-D), using modified Eeuwens Y3 liquid medium as the basal medium. Among the tested auxins, 100 μM NAA in combination with 100 μM 2,4-D enhanced the production of calli/embryos (123) whereas IAA and picloram showed negative and detrimental effects, respectively, for androgenesis induction over 100 μM 2,4-D alone. Kinetin and 2-iP enhanced the production of calli/embryos when 100 μM 2,4-D was present in the culture medium. Both cytokinins at 10 μM yielded the highest frequencies of embryos (113 and 93, respectively) whereas zeatin (1 or 2.5 μM) had no impact on microspore embryogenesis. When calli/embryos (produced from different treatments in different experiments) were sub-cultured in somatic embryo induction medium (Y3 medium containing 66 μM 2,4-D), followed by maturation medium (Y3 medium without growth regulators) and germination medium (Y3 medium containing 5 μM-6-benzyladenine—BA and 0.35 μM gibberellic acid—GA3), plantlets were regenerated at low frequencies (in most treatments ranging from 0% to 7%).  相似文献

The antagonistic effects of some growth regulators [i.e. indol-3-yl-acetic acid (IAA), gibberellic acid (GA3) or kinetin] on stress imposed by sea water on leaf area, pigment and photosynthetic activity in leaves of broad bean plants at different stages of development were investigated. Seed priming with GA3 alleviated either partially or completely the effects induced by the two levels of sea water (10 and 25 %) used on leaf area at all experimental stages. However, IAA, GA3 and kinetin inhibited leaf growth by themselves in almost all measurements. Seed pretreatment with kinetin alleviated the inhibition of pigment production in sea water-irrigated plants. Furthermore, GA3 or kinetin nullified the deleterious effects imposed by irrigation with sea water particularly the high level (25 %) on photosynthetic14CO2 fixation.  相似文献

In Nigella damascena, where “doubleness” behaves as a single gene character, recessive to the normal wild “single” form, we detected a number of differential responses of cultured floral apices of the two forms to plant growth regulators. “Single” buds initiated all organ types on a defined medium lacking any plant growth regulators. “Double” buds, however, required either kinetin or GA3 for the development of a normal organ complement. Kinetin enhanced the growth and development of buds of both genotypes, whereas GA3, at all concentrations, specifically inhibited the initiation and growth of stamens and nectaries in “single” buds. No combination of IAA or kinetin overcame this inhibition of “single” buds by GA3. In the presence of both IAA and GA3, “single” buds, following the initiation of normal carpels, initiated a few abnormal carpelloid structures from regions which had failed to produce stamens and nectaries. These observations, coupled with other studies including those on the endogenous levels of gibberellin in the two genotypes, suggest that “doubleness” correlates strongly with a modification of gibberellin metabolism. The causal connection between the two phenomena is as yet to be discovered.  相似文献

The data obtained characterize the changes in the contents of endogenous phytohormones (IAA, cytokinins, GA, and ABA) in germinating pollen grains and growing pollen tubes of a self-compatible clone of petunia (sPetunia hybrida L.) within an 8-h period under in vitro conditions. The hydration and initiation of germination of pollen grains brought the ABA content down to a zero level, while the levels of GA, IAA, and cytokinins increased 1.5–2-fold. Later, in the growing pollen tubes, the GA content increased twofold, while the levels of IAA and cytokinins decreased. The exogenous ABA and GA3 considerably promoted pollen germination and pollen tube growth; however, only the treatment with GA3 produced the maximum length of pollen tubes. The exogenous IAA promoted and the exogenous cytokinins hindered the growth of pollen tubes. The membrane potential, as assessed with a potential-sensitive dye diS-C3-(5), considerably increased in the pollen grains treated with ABA and benzyladenine, whereas IAA and GA3 did not practically affect it. The authors conclude that the mature pollen grains contain the complete set of hormones essential for pollen germination and pollen tube growth. American ginseng (Panax quinquefolium L.) is a perennial medicinal herb originally grown in Canada and USA, and recently also in China, Australia, Holland and Poland. Several commercial preparations are produced from ginseng roots, that are known for their antifatigue, antitumor, antistress and immune system stimulating functions. The medicinal properties are due mainly to the active components – ginsenosides. In this work, the results of field cultivation experiments are presented that examine the effects of foliar application of several growth regulators on quality parameters and ginsenoside content of P. quinuefolium roots. The growth regulators tested, i.e., kinetin, daminozide, mixture of gibberellic acid (GA3) with potassium salt of α-naphthalene acetic acid (kNAA) and new preparation – IPO-1 – benzimidazole derivative (obtained from the Institute of Organic Industry in Warsaw – at present during the process of patent), were applied at a concentration of 100 or 200 mg l−1 in the middle of June in the 2nd year of vegetation. After 4 years of cultivation, the roots were dug up and dried, and subsequently the quantitative analysis of individual saponins (Rb1, Rb2, Rc, Rd, Re, Rg1) by HPLC was performed. Growth regulators significantly affected quality parameters, morphological features and accumulation of individual and total ginsenosides in ginseng roots. Regardless of doses, the plant roots treated with growth regulators had a higher content of total ginsenosides in comparison to the control. The growth regulators also affected individual ginsenosides level and narrowed the ratio of Rb:Rg group. The application of kinetin, daminozide and benzimidazole derivative for foliar spray during 2nd year of American ginseng vegetation caused a significant increase in air dry weight of roots and aboveground parts whereas the mixture of GA3 and kNAA showed a decreasing effect. An increase of roots size was observed using higher doses (200 mg l−1) of kinetin and daminozide while a decreasing tendency appeared with the application of the other preparations.  相似文献

A field experiment was conducted to investigate the effects of presoaking the wheat grains (Triticum aestivum L.) in different levels of salinity (33 or 66 mM) and in growth regulators (indolyl-3-acetic acid, IAA at SO g m-3, gibberellic acid, GA3 at 100 g m-3, or kinetin at 100 g m-3) on the shoot growth and pigment content of the developing wheat flag leaf. Salinity at 33 or 66 mM led to an insignificant increase in the fresh and dry masses as well as in the shoot diameter and shoot length, but it attenuated the flag leaf area. In the majority of cases, salinity increased the chlorophyll (Chla, Chlb) and carotenoid contents as well as the number of chloroplasts per a mesophyll cell. The growth in the wheat shoot of the saline-treated plants was, in general, stimulated in response to presoaking the grains in kinetin or GA3. On the other hand, IAA + salinity led to a negligible effect on the growth in the wheat plants particularly at the early stages of growth. The presoaking of grains in NaCl at 33 mM + IAA or 66 mM + kinetin induced a marked increase in the pigment content of the wheat flag leaf particularly at the early stages of growth. The interaction between salinity and phytohormones increased the number of chloroplasts; kinetin was the most effective.  相似文献
