A comparative study was made of the fatty acid composition of the total lipids extracted from the cotyledons and the callus cultures derived from cotyledon segments of six species of Cucurbitaceae. Conditions for callus induction and growth of cultures were identical. The difference between the two systems was in the reversal of the ratio of total unsaturated to saturated acids in all callus cultures. In callus cultures, instead of linoleic, linolenic was the major unsaturated fatty acid. In Momordica charantia, α-elaeostearic acid present in the cotyledon was not detected in callus and oleic acid was the major unsaturated fatty acid.  相似文献

Callus tissues were derived from cotyledon segments of Cucumis melo and Cucumis sativus. Four combinations of growth factors, i.e., naphthalene acetic acid (NAA) plus coconut water (CW); NAA plus kinetin; NAA plus 6-benzylaminopurine (BAP) and indole 3-butyric acid plus BAP, were incorporated in the medium for callus initiation as well as for growth of excised callus in culture for six passages. The proportion of total saturated to unsaturated acids and the ratio of linoleic to linolenic acid was influenced by the change in the type of auxin and cytokinin in the combinations used. A many fold increase of myristic acid was recorded for the indole 3-butyric acid plus BAP combination.  相似文献

It is believed that free fatty acids contribute to the pathogenesis of type 2 diabetes in humans. We have recently shown that lipoapoptosis of human beta-cells is specifically induced by saturated fatty acids while unsaturated had no effect. In the present study we tested the effect of co-incubation of different saturated and unsaturated free fatty acids on lipoapoptosis in beta-cells. RIN1046-38 cells and isolated human beta-cells were incubated with combinations of saturated fatty acids (palmitate, stearate) and mono- or polyunsaturated fatty acids (palmitoleate, oleate, and linoleate). Cells were incubated for 24-72 h with 1mM fatty acids. All unsaturated fatty acids tested completely prevented palmitate- or stearate-induced apoptosis of rat and human beta-cells as assessed by flow cytometric cell cycle analysis and TUNEL assay. This might suggest that apoptosis in vivo is predominantly determined by the content of unsaturated fatty acids in a mixed fatty acid pool.  相似文献

Root, hypocotyl, cotyledon, stem and leaf of Cucumis melo var. utilissimus seedlings were used for callus induction. Comparison was made between these parts, between callus tissues originating from all the parts and between each part and its callus, with respect to the fatty acid composition of total lipids. In all the parts there was a greater proportion of unsaturated fatty acids, the predominant fatty acid in root, stem and leaf being linolenic acid whilst in the cotyledon linoleic predominated. In the hypocotyl these two acids were present in equal amounts. In callus cultures the proportion of saturated acids was greater and the predominant fatty acid was palmitic. The major unsaturated fatty acid in callus cultures was linolenic. The analysis showed that callus tissue and its respective plant part had different fatty acid patterns and that all the callus cultures had very similar patterns irrespective of their origin.  相似文献

The increasing bacterial resistance against conventional antibiotics has led to the search for new antimicrobial drugs with different modes of action. Cationic antimicrobial peptides (AMPs) and lipopeptides are promising candidates to treat infections because they act on bacterial membranes causing rapid destruction of sensitive bacteria. In this study, a decapeptide named A2 (IKQVKKLFKK) was conjugated at the N‐terminus with saturated, unsaturated, methoxylated and methyl ‐branched fatty acids of different chain lengths (C8 – C20), the antimicrobial and structural properties of the lipopeptides being then investigated. The attachment of the fatty acid chain significantly improved the antimicrobial activity of A2 against bacteria, and so, endowed it with moderated antifungal activity against yeast strains belonging to genus Candida. Lipopeptides containing hydrocarbon chain lengths between C8 and C14 were the best antibacterial compounds (MIC = 0.7 to 5.8 μM), while the most active compounds against yeast were A2 conjugated with methoxylated and enoic fatty acids (11.1 to 83.3 μM). The improvement in antimicrobial activity was mainly related to the amphipathic secondary structure adopted by A2 lipopeptides in the presence of vesicles that mimic bacterial membranes. Peptide conjugation with long hydrocarbon chains (C12 or more), regardless of their structure, significantly increased toxicity towards eukaryotic cells, resulting in a loss of selectivity. These findings suggest that A2‐derived lipopeptides are potential good candidates for the treatment of infectious diseases caused by bacteria and opportunistic pathogenic yeast belonging to genus Candida. In the studies described here rat liver microsomes containing labeled palmitic, stearic, oleic or linoleic acids were incubated with fatty acid binding protein (FABP) and the rate of removal of14C-labeled fatty acids from the membrane by the soluble protein was measured using a model system. More unsaturated than saturated fatty acids were removed from native liver microsomes incubated with similar amounts of FABP. Thein vitro peroxidation of microsomal membranes mediated by ascorbate-Fe++, modified its fatty acid composition with a considerable decrease of the peroxidizability index. These changes in the microsomes facilitated the removal of oleic and linoeic acids by FABP, but the removal of palmitic and stearic acids was not modified. This effect is proposed to result from a perturbation of membrane structure following peroxidation with release of free fatty acids from susceptible domains.Abbreviations BSA bovine serum albumin - FABP fatty acid binding protein  相似文献

Administration of various supposed precursors to the callus cultures of Lithospermum erythrorhizon grown on the Linsmaier—Skoog medium supplemented with IAA and kinetin established that the constituent shikonin is formed via shikimic acid, p-hydroxybenzoic acid, m-geranyl-p-hydroxybenzoic acid and geranylhydroquinone. In a strain of callus culture lacking the capacity to synthesize shikonin and in callus cultures which have had this capacity but lost it due to cultivation on a medium supplemented with 2,4-D, substances up to m-geranyl-p-hydroxybenzoic acid in the biosynthetic sequence have been detected. Although illumination with white light also arrested shikonin production, traces of pigment were still formed presumably because light did not reach the innermost part of the callus cultures.  相似文献

Acetone-soluble extractives of “blue” and “green” strain Pinus muricata D. Don were found to consist of free and “combined” fatty acids, resin acids, and phenols. The composition of the extractives from the two strains was similar though “green” strain P. muricata contained more Δ8(9),15 isopimaric acid than “blue” strain. This difference may be used to identify these muricata strains if the age of the wood precludes a monoterpene examination.  相似文献

The fatty acid (FA) composition of Demospongiae species from the Sea of Okhotsk was studied. Fifteen sponge species were investigated for the first time, and the previously studied species Desmacella rosea and Myxilla incrustans were reexamined for their FA composition. Gas chromatography-mass spectrometry revealed 150 different fatty acids, of which 15 have not been identified in sponge lipids previously. The relative content of saturated FAs varied from 7.6 in Melonachora kobjakovae to 29.6% in Amphilectus digitata, with an average of 14.6% of total FAs. The relative content of monoenic FAs ranged from 12.8 in T. dirhaphis to 27.0% in Polymastia sp., with an average of 20.6% of total FAs. Non-methylen-interrupted, primarily unsaturated Δ5,9-FAs contributed a significantly to the amount of polyunsaturated fatty acids of sponges; this being a distinguishing feature of the FA composition of the investigated group of organisms.  相似文献

The quantity of total fatty acids in soybean cotyledons during aging, senescence and regreening has been studied. The greatest change in the fatty acid profile during the initial greening of the cotyledons (4–7 days after germination) was a 130% increase in the content of linolenate. Linoleate, as in the case of the other fatty acids, declined in the first 4 days and then increased by 7 days. Following the 10th day after germination, the quantity of palmitate, linoleate, and linolenate decreased continuously through senescence to 20–28% of the maximum quantity of each. When the cotyledons were regreened by removal of the epicotyl 15 or 16 days after germination, linolenate was present in quantities substantially higher than in the senescing cotyledon. On the 22nd day after germination, the quantity of linolenate in regreened tissue was 140% greater than that in senescing tissue of the same age. By contrast, the quantity of linoleate was only 30–40% greater in regreening tissue and the quantity of most of the other fatty acids was similar in both tissues. Similar changes in the quantity of chloroplast fatty acids were observed during this period. Removal of the epicotyl resulted in a higher level of chloroplast linolenate. During aging, the total chlorophyll and the number of chloroplasts reached a maximum on the 10th day and decreased rapidly during senescence. The amount of chlorophyll per chloroplast remained relatively constant during this period whereas the quantity of linolenate per chloroplast decreased during senescence. It is suggested that major structural changes observed in chloroplast membranes may be related to changes in fatty acid composition, but are not dependent on changes in chlorophyll concentration.  相似文献

Larvae of the three important Central American malaria vectors, Anopheles albimanus, An. vestitipennis, and An. darlingi, are found in distinctly different habitats broadly defined by hydrology and aquatic vegetation, but little is known about the actual food quality and quantity of these habitats. Polyunsaturated fatty acids (PUFA) are of special interest, because mosquitoes require 20:5ω3 (EPA), 20:4ω6 (ARA), and 22:6ω3 (DHA) and without an adequate supply of these PUFAs they are not able to complete their life cycle. We collected samples of larvae and their corresponding habitats and analyzed their fatty acid (FA) composition to reveal if there are any species‐specific and habitat‐specific differences in FA composition, and if habitat FA differences can be linked to differences in the mosquito FA pattern and, ultimately, mosquito performance. We also assessed how FA of wild larvae compare to the laboratory‐reared larvae. Habitats were generally low in essential PUFAs and there were no significant differences among the FA composition of habitat samples. There were significant differences in FA composition of larvae. An. darlingi contained significantly higher amounts of FA, specifically a higher content of ω‐6 PUFA, represented mainly by the linoleic acid (18:2ω‐6). Large differences were found between field‐collected and laboratory‐reared An. vestitipennis larvae, especially in the content of PUFAs. The laboratory‐reared larvae contained significantly more of the total FA, ω3 PUFA, and MUFA. The laboratory‐reared larvae contained three to five times more essential PUFAs, EPA, and DHA. However, there were no differences in the total dry weight of the 4th instar larvae between the wild vs laboratory‐reared larvae. Total FA in both larvae and habitats of An. albimanus and An. darlingi were positively correlated with the concentration of particulate organic carbon and nitrogen (POC, PON) in their respective habitats, but no such correlation was found for An. vestitipennis. PUFA are a good indicator of nutritional quality, although factors controlling the success of anopheline development from larval habitats are likely to be more complex and would include the presence of predators, pathogens, and toxins as interacting factors.  相似文献

The Inuit undergo substantial changes in their lifestyle, but few data exist on how these changes occur in biomarkers, such as polyunsaturated fatty acids (PUFAs). Here, we report data from a cross-sectional survey conducted in 2004 among 861 representative Nunavik Inuit adults, in whom FAs were measured in red blood cells (RBCs). FAs were also measured in plasma phospholipids (n=452) to assess temporal trend by comparing plasma PUFAs measured in 1992. Food intakes were estimated using a validated food frequency questionnaire. In 2004, marine food intake was 84±4 g/d (±SEM). Adjusted-mean of RBC omega-3 was significantly higher, and omega-6 lower, in older age groups (Ptrend<0.001). In 2004, plasma omega-3 was 25% lower, while omega-6 was 9% higher, compared to 1992. Our study revealed that Nunavik Inuit adults still have high RBC omega-3, but show signs of nutritional transition – as indicated by lower omega-3 and higher trans-fats in RBCs of young compared to older.  相似文献

脂肪酸不仅是细菌细胞膜组分,还是许多生物活性物质的合成原料。不饱和脂肪酸(unsaturated fatty acid, UFA)具有更低的相变温度,是细菌调节细胞膜流动性的重要分子,因此UFA合成途径是重要的抗菌药物筛选靶点。细菌可利用厌氧途径合成UFA,其中模式生物大肠杆菌利用经典的FabA-FabB途径合成UFA,但不同细菌中UFA合成的厌氧途径具有多样性,相关催化酶类也不尽相同;细菌还可以利用需氧途径合成UFA,利用脂肪酸脱饱和酶直接将饱和脂肪酸(saturated fatty acid, SFA)转化为不饱和脂肪酸,而不同脱饱和酶会生成不同结构的UFA,在逆境耐受、致病力等多方面发挥重要作用;细菌还可以利用单加氧酶,将脂肪酸合成途径中癸酰酰基载体蛋白(acyl carrier protein, ACP)转化为顺-3-癸烯酰ACP,并最终合成UFA。细菌脂肪酸合成相关的其他酶类在UFA合成或不同种类UFA调节中也发挥着重要作用。本文系统地总结了细菌UFA合成途径与相关酶类的多样性研究进展,旨在为进一步了解细菌UFA合成机制,并以此为靶点开发抗菌药物等方面提供理论支撑。  相似文献

The total fatty constituents of slash pine (Pinus elliottii) tissue cultures, seeds and seedlings were examined by GLC and MS. Qualitatively, the fatty acid composition of these tissues was very similar to that reported for other pine species. The fatty acid contents of the tissue cultures resembled that of the seedling tissues. In addition to the fatty acids common to botanical materials, Δ5-C18 and -C20 nonmethy lene-interrupted polyunsaturated acids were present in low relative abundances. The branched-chain C17 acid reported for several other Pinus species was confirmed as the anteiso isomer.  相似文献

The phyto-constituents of Gymnema sylvestre are used in the treatment of diabetes and obesity. The present work reports on the extraction of gymnemic acid through gymnemagenin from callus cultures of G. sylvestre. Components were separated on pre-coated silica gel 60 GF254 plates with chloroform:methanol (8:2) and scanned using a densitometric scanner at 205 nm in the near-UV region. Linearity of determination of gymnemagenin was observed in the range 2-10 microg. The average percentage recovery of gymnemagenin from leaf callus extracts was 98.9+/-0.3.  相似文献

Abstract The polar lipids of 5 species of Thiobacillus were extracted and purified. An analysis of the fatty acid composition of the polar lipids documented the presence of methoxy, cyclopropyl, monounsaturated and hydroxycyclopropyl fatty acids of sufficiently unusual structure to serve as 'signatures' for the presence of these organisms in environmental samples. The structures of the unusual fatty acids of the polar lipids were confirmed by mass spectrometry (MS) after isolation by capillary gas chromatography (GC).  相似文献

The absolute rates of reaction of the trichloromethylperoxy radical, CCl3OO, derived from carbon tetrachloride and the halothane peroxy radical, CF3CHClOO, with oleic, linoleic, linolenic and arachidonic acids have been determined using the fast reaction technique of pulse radiolysis. In general, the rates of reaction of the radical derived from carbon tetrachloride are approximately five times greater than those for the halothane related radical. In both cases the rate constant increases with increasing unsaturation of the fatty acid in agreement with the known greater susceptibility of polyunsaturated fatty acids to peroxidative decomposition.  相似文献

The cell suspension culture of Cryptomeria japonica contains volatile oils, the yield of which was 0.005–0.01% of the fresh cells. In the volatiles, five aldehydes, ten fatty acids and their esters, and two diterpenes of abietatriene and ferruginol have been found. Of these, palmitic acid is present as the most predominant component, amounting to ca 40% of the volatiles.  相似文献
