The susceptibility of Spodoptera litura to SlMNPV infection was markedly affected by phyto-chemicals ingested during the acquisition of viral inoculum on foliage of tomato and cauliflower. The LD50 values computed for second, third and fourth instar larvae assayed on tomato leaves were 254, 819 and 23395 PIBs/larva, respectively whereas, it was 326, 1719 and 43843 PIBs/larva for respective instars when assayed on cauliflower leaves. Thus LD50 values for second, third and fourth instar larvae were 1.28-, 2.09- and 1.87- fold lower, respectively in tomato leaves. Similarly, LT50 values for second, third and fourth instar larvae assayed on tomato leaves were 7.1 and 7.5 days, respectively at inoculum dose of 2.7×104 PIBs/larva whereas, it was 7.7 and 8.0 days for respective instars when assayed on cauliflower leaves at same inoculum. This result also showed that the S. litura were more susceptible on tomato leaves in comparison to cauliflower leaves as the time required for mortality was lower in tomato leaves. The possible biochemical bases for differential level of mortality of S. litura larvae on tomato and cauliflower crops needs to be investigated.  相似文献

The parasitic effectiveness of Cotesia plutellae Kurdjumov for the control of diamondback moth (DBM), Plutella xylostella L., was investigated without spraying pesticides in the greenhouse, and the damage degrees of Brassica rapa cv. Pakchoi grown in parasitoid-applied plot and control plot were evaluated. Among the 961 fourth instar DBM larvae collected in the common cabbage fields at Kwanshan region in Taiwan, 336 cocoons of C. plutellae were harvested from DBM larvae, which showed a parasitism rate of 35.0%. A total of 650 DBM adults and 600 C. plutellae adults were released according to the releasing scheme in the experiment plot. As a result, 56.7% of the DBM larvae were parasitized by C. plutellae adults 12 days before harvest, and the parasitism reached 80.8% at six days before harvest. At the harvest time, a population density of C. plutellae cocoons in the experiment plot was 2.2/plant, and that of normal DBM larvae was 0.5/plant. The density of DBM in the control plot was much higher than in the parasitoid-released plot by 7.7/plant. In the first survey, the density of DBM larvae on a Pakchoi plant was quite low, 0.3 ∼ 0.8 larvae. However, the DBM population increased dramatically after six days later, 5.7-10.1 larvae/plant. The population density of DBM larvae showed clear difference between the parasitoid-released plot and control plot. The parasitism increased to nearly two fold within a week after the release of DBM adults. As a further study, a combining use of two or more species para-sitoids could be considered to improve a parasitism against DBM in the fields.  相似文献

The present study relates to a methanol extract of the seed coat of Ginkgo biloba, and tested particularly on the third instar larvae of Spodoptera exigua. The extract was found to have an inhibitory effect on the growth of the larvae besides bringing a change in the nutrient reserves in the body of the insect. Topical application of five different doses of the methanol extract resulted in a mortal effect to third instar larvae of S. exigua that is very much dependent on the dose as well as duration of exposure. Lower doses revealed lower mortality after 24 h of application. At doses of 1.00, 2.00, 4.00, 8.00 and 16.00 ng/larva, mortalities were 9.25, 26.07, 50.32, 56.28 and 92.44%, respectively. The dose for 50% mortality (LD50) of methanol extracts by applied by a topical method with 1 µL of acetone solution was 1.92 ng/larva. Nutrient reserves like protein, glycogen and lipid are known to regulate pupation and adult emergence. These reserves have been found to be lower in treated larvae, indicating the insecticidal role of methanol extracts from G. biloba against third instar larvae of S. exigua.  相似文献

The fumigant toxicity of 66 plant essential oils to Plutella xylostella (L.) larvae and Cotesia glomerata (L.) adults was examined using a vapor-phase toxicity bioassay and compared with that of dichlorvos. Responses varied according to oil and insect species used. Based on 24 h LD50 values, pennyroyal oil [10.77 mg/filter paper (4.25 cm diameter)] was the most toxic fumigant, followed by rosemary and sage (Dalmatin) oils (15.15 mg/paper). Potent fumigant toxicity was also produced from armoise, buchu leaf, cedarleaf, coriander, eucalyptus, howood, lavender, myrtle, niaouli, peppermint, and rosewood oils (LD50, 21.29–27.31 mg/paper). All essential oils were less effective than dichlorvos (LD50, 0.52 mg/paper). Against adult C. glomerata, dichlorvos (LD50, 0.03 mg/paper) was the most toxic fumigant, whereas the LD50 values of the 14 essential oils ranged from 1.59 to 8.51 mg/paper. Based on selective toxicity ratio (STR, P. xylostella LD50/C. glomerata LD50), the 14 essential oils (STR, 2.5–14.5) are more selective than dichlorvos (STR, 17.3). The essential oils tested merit further study as potential fumigants for the control of P. xylostella in greenhouses because of their selective toxicity to adult C. glomerata and their much greater activity as a fumigant.  相似文献

The diamondback moth (DBM), Plutella xylostella (L.) (Lepidoptera: Plutellidae), is oligophagous on plants in the Brassicaceae, and is considered one of the most significant insect pests of canola (Brassica napus L.), a major oilseed crop grown in the Prairie Provinces of Canada. The bertha armyworm (BAW), Mamestra configurata Walker (Lepidoptera: Noctuidae), is a generalist herbivore that preferentially feeds on canola plants. In the canola growing season in the Prairie Provinces of Canada, DBM feeding occurs prior to BAW herbivory. In this study, we test the effect of DBM herbivory on subsequent host use by BAW. Oviposition by female BAW was not influenced by previous DBM‐herbivory or mechanical damage of canola plants. Bertha armyworm larvae were deterred from feeding on canola previously damaged by DBM and larvae developed into lighter pupae when reared on DBM‐damaged plants. Bertha armyworm pupae that developed from larvae fed on mechanically damaged plants had similar pupal weights to those fed undamaged plants. Adult BAW reared on canola with previous DBM feeding damage had marginally smaller wings than moths reared on canola treated differently. The combination of these results suggests that herbivory by the brassicaceous specialist, DBM, negatively influences host use and larval performance by the generalist, BAW, on canola.  相似文献

In this research work, the susceptibility of egg and four larval instars of Leptinotarsa decemlineata (Say) (Col.: Chrysomelidae) to Insect Growth Regulators (IGRs) compounds (lufenuron 25% EC and fenoxycarb 25% WP) was determined. Different larval instar groups were separated by measuring the head capsule width and were used in all bioassays. The data were analysed with log-probit transformation using the SPSS software. The LC50 for egg was determined by dipping egg masses in different concentration of either compound for 10 s, and LC50 values for each group of larvae was estimated by using treated potato plants. The LC50 values of lufenuron on egg, first, second and third instars of larvae were 682.65, 40.58, 47.83 and 261.38 ppm, respectively, and for fenoxycarb, these were estimated as 897.50, 35.60, 57.91 and 355.23 ppm, respectively. The LD50 values of lufenuron and fenoxycarb on second instar larvae were 139.56 and 228.42 ppm, respectively.  相似文献

Preliminary screening of extracts of the leaves and stems of four Turraea plant species (Meliaceae) on second instar larvae of Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) showed that the methanolic extract of Turraea abyssinica leaves possess good toxic potential with LD50 of 270.7 ppm. Fractionation of this extract led to isolation of a new limonoid derivative, 12α‐diacetoxywalsuranolide (1) and three other known limonoids [1α,7α,12α‐triacetoxy‐4α‐carbomethoxy‐11β‐hydroxy‐14β,15β‐epoxyhavanensin (2), 11‐epi‐21‐hydroxytoonacilide (3), 11β,12α‐diacetoxycedrelone (4)]. The structures of the compounds were established by IR, UV, mass spectrometry and 1D/2D NMR analyses, and in the case of the known compounds, also by comparison with reported data. All the isolates were tested for their larvicidal activities at different dose range against second instar larvae of T. absoluta. They were found to be more active (with LD50 < 7.0 ppm) compared to the reference compound azadirachtin (LD50 value of 7.8 ppm).  相似文献

South American tomato pinworm, Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) is a devastating invasive global insect pest of tomato, Solanum lycopersicum (Solanaceae). In nature, pests face multiple overlapping environmental stressors, which may significantly influence survival. To cope with rapidly changing environments, insects often employ a suite of mechanisms at both acute and chronic time-scales, thereby improving fitness at sub-optimal thermal environments. For T. absoluta, physiological responses to transient thermal variability remain under explored. Moreso, environmental effects and physiological responses may differ across insect life stages and this can have implications for population dynamics. Against this background, we investigated short and long term plastic responses to temperature of T. absoluta larvae (4th instar) and adults (24–48 h old) from field populations. We measured traits of temperature tolerance vis critical thermal limits [critical thermal minima (CTmin) and maxima (CTmax)], heat knockdown time (HKDT), chill coma recovery time (CCRT) and supercooling points (SCP). Our results showed that at the larval stage, Rapid Cold Hardening (RCH) significantly improved CTmin and HKDT but impaired SCP and CCRT. Heat hardening in larvae impaired CTmin, CCRT, SCP, CTmax but not HKDT. In adults, both heat and cold hardening generally impaired CTmin and CTmax, but had no effects on HKDT, SCP and CCRT. Low temperature acclimation significantly improved CTmin and HKDT while marginally compromising CCRT and CTmax, whereas high temperature acclimation had no significant effects on any traits except for HKDT in larvae. Similarly, low and high temperature acclimation had no effects on CTmin, SCPs and CTmax, while high temperature acclimation significantly compromised adult CCRT. Our results show that larvae are more thermally plastic than adults and can shift their thermal tolerance in short and long timescales. The larval plasticity reported here could be advantageous in new envirnments, suggesting an asymmetrical ecological role of larva relative to adults in facilitating T. absoluta invasion.  相似文献

We examined mortality and feeding inhibition response of Lymantria dispar L. (Lepidoptera: Lymantriidae) larvae to ingested doses of Bacillus thuringiensis subsp. kurstaki as a function of dose, instar and temperature. We developed generalized (logistic) linear mixed models and a mixture survival model, commonly used in medical statistics, to analyze the complex data set. We conducted bioassays of Foray 48B with larvae from the NJSS laboratory stock, using droplet imbibing or force-feeding to ensure dose ingestion. The dose causing mortality in 50% of the test population (LD50) under standard test conditions (22 °C) ranged from 0.019 International Units (IU)/larva for first instar larvae (L1) to 1.6 IU/larva for L4. Temperature affected larval mortality in two ways. Mortality occurred sooner and progressed more rapidly with increasing temperature (13-25 °C) at each dose level and instar, while the maximum level of mortality attained by each instar decreased with increasing rearing temperature. The mechanisms underlying this effect are being investigated. Larvae that survived exposure to B. thuringiensis resumed feeding after a period that was dependent on instar, dose, and temperature. The equations describing observed mortality and feeding recovery responses were used to construct a simulation model, which was able to predict both processes, and which forms the basis for a process-oriented model that can be used as a decision support tool in aerial sprays.  相似文献

Injections into 4th instar larvae of Galleria mellonella using either in vitro or in vivo inoculum of the BR-6 isolate of Spiroplasma citri, propagated for one to nine passages, caused 5.7 to 24.7% mortality. Weight gain of the larvae injected at their 4th, 5th, and 6th instar was reduced in the first 4 days after inoculation but final pupal weight of the survivors was not significantly affected. Fourth instar larvae pupated within 10 days after the injection, but more larvae (6–13%) injected with 5th- to 9th-passage cultures pupated 5 or more days later than did larvae injected with 1st to 4th-passage cultures (0–3%). As many as one-third of the injected larvae developed into deformed pupae, with some external appendages missing or with a reduced and distorted thorax or abdomen with uneven tanning of the integument. Spiroplasma multiplied to dense concentrations (108 to 109/ml) in hemolymph smears from injected larvae incubated under oil. Larvae of Tenebrio molitor were not susceptible to S. citri by injection or feeding and G. mellonella were not susceptible by feeding. Transmissibility of S. citri by leafhopper vector to celery and periwinkle plants was retained after propagation for nine successive passages during 7 months in a nonhost insect such as Galleria.  相似文献

在研究比较了11种农药对斜纹夜蛾Spodoptera litura细胞(简称SL细胞)的毒杀活性基础上,选择毒杀活性最高的杀菌剂丙环唑,对其毒理学机理进行进一步研究。结果表明,丙环唑的细胞毒力最高,在100 μg/mL浓度下处理后48 h,SL细胞的死亡率为98.08%。处理后36 h,丙环唑对SL细胞的LC50值为20.31 μg/mL。丙环唑能明显降低SL细胞的蛋白质含量。以0.5 μg/头的丙环唑注射斜纹夜蛾4龄幼虫,处理后72 h,试虫血淋巴总含量及血细胞数分别下降了26.80%和25.26%;在1.0 μg/头的剂量下,则分别下降了37.67%和36.32%。以0.5 μg/头和1.0 μg/头的丙环唑注射处理后,斜纹夜蛾幼虫体重显著降低。此外,丙环唑能降低斜纹夜蛾幼虫血淋巴含糖量及血淋巴蛋白质含量。在注射处理后96 h和120 h,丙环唑对斜纹夜蛾4龄幼虫的LD50值分别为0.59 μg/头和0.45 μg/头。丙环唑对SL细胞和斜纹夜蛾幼虫均具有较好的毒杀活性,显示出丙环唑类似物控制害虫的可能性。  相似文献

To determine the effect of sublethal doses of Helicoverpa armigera single nucleocapsid nucleopolyhedrovirus (HearSNPV) on the metabolic rate of H. armigera, the respiration rates of third instar H. armigera larvae inoculated with sublethal doses of HearSNPV were evaluated. Respiration rates, measured as the rate of CO2 production (VCO2), were recorded daily using closed-system respirometry. By 4 days post-inoculation (dpi), the metabolic rates of LD25 or LD75 survivors were significantly higher than that of uninoculated controls. When dose data were pooled, the VCO2 values of larvae that survived inoculation (0.0288 ml h−1), the uninoculated controls (0.0250 ml h−1), and the larvae that did not survive inoculation (0.0199 ml h−1) differed significantly from one another. At 4 dpi, the VCO2 of the uninoculated controls were significantly lower than the VCO2 of inoculation survivors, but significantly higher than the VCO2 of inoculation non-survivors. Inoculation survivors may have had high metabolic rates due to a combination of viral replication, organ damage, and an energy-intensive induced cellular immune response. The high 4 dpi metabolic rate of inoculation survivors may reflect an effective immune response and may be seen as the metabolic signature of larvae that are in the process of surviving inoculation with HearSNPV.  相似文献

Spodoptera frugiperda is a highly invasive pest species that recently invaded Africa and Asia causing severe economic losses, primarily related to corn and rice crops. Temperature is one of the most important environmental factors that influence the invasion of pests into new habitats. However, little is known regarding the thermal tolerance characteristics of invasive S. frugiperda. Thus, we investigated the response of four developmental stages of S. frugiperda (i.e., eggs, third and sixth instar larvae, and pupae) to cold acclimation (CA) and rapid cold-hardening (RCH). All individuals suffered high mortality with 24-h temperature treatments at <?5°C and >35 °C. The CA treatment significantly increased the survival rate of the eggs and third instar larvae, although it did not affect the sixth instar larvae and it decreased the pupation rate. The RCH treatment at 5 °C for 5 h or 2 °C for 2 h increased the cold tolerance capabilities of the third and sixth instar larvae, respectively. Thus, the larval stage appears to be crucial for the cold tolerance of S. frugiperda. Our findings improve the current understanding of the cold tolerance characteristics of S. frugiperda and indicate its potential for survival in the newly invaded temperate regions of Asia.  相似文献

The diamondback moth (DBM), Plutella xylostella (L.), is a globally destructive pest of cruciferous vegetables. Excessive use of synthetic pesticides to control this species results in negative effects on the environment, human health, and nontargeted organisms. The essential oils (EOs) derived from plants may be developed as effective alternatives to conventional pesticides. In this study, thirteen EOs were extracted by hydrodistillation, respectively. Their chemical compositions were identified by gas chromatography-mass spectrometry (GC–MS). Furthermore, the biological activities of EOs such as toxicity, antifeedant activity, and growth inhibition effect, toward DBM larvae were investigated. Against DBM second-instar larvae, the most toxic EO was Pelargonium graveolens (LC50 = 0.36 μg/μl) after 72 hr of exposure, followed by Polygonum hydropiper (LC50 = 0.53 μg/μl). The Ocimum basilicum EO exhibited the highest antifeedant effect to third-instar larvae at at all set concentrations. At 15 μg/μl, the EOs of Acorus calamus, O. basilicum, and P. graveolens completely inhibited the feeding activities of larvae (100%). The Ruta graveolens EO showed the lowest relative growth rate and the highest growth inhibition rate towards third-instar larvae at diverse concentrations. And the EOs of P. hydropiper, A. calamus, and O. basilicum showed promising growth inhibition activities. Overall, the five EOs (P. graveolens, O. basilicum, R. graveolens, P. hydropiper, and A. calamus) showed moderate to high bioactivity, whereas eight EOs were found to be less active against DBM larvae. These results indicate that the five tested EOs are promising to be developed as novel botanical insecticides to control DBM population.  相似文献

Numbers of viable bacteria in second instar Culex pipiens quinquefasciatus larvae were determined following ingestion of pathogenic strain SSII-1 and nonpathogenic Bacillus sphaericus. Numbers of nonpathogenic B. sphaericus recovered from larvae declined rapidly after cessation of feeding, as did numbers of pathogenic SSII-1 cells fed at LD20 dosage. When pathogenic cells were fed at LD70 dosage, the number of B. sphaericus in larvae increased following initial decline. When chloroformtreated SSII-1 cultures, in which all bacteria except spores were dead, were fed at LD10 and LD98 dosages, no viable B. sphaericus were recovered from larvae. In all SSII-1 treatments, other bacterial flora multiplied rapidly in larvae following onset of mortality; the role of this multiplication in the pathogenesis was not determined. It is proposed that toxic material is released when SSII-1 cells are digested and that multiplication of B. sphaericus in the larval gut is not essential in the pathogenesis. There appears to be no difference in the pathogenesis when differing numbers of B. sphaericus. i.e., LD10–20 or LD70–98 dosages, are ingested. Possible nature of the toxic material is discussed.  相似文献

Wiseana spp. are lepidopterous, soil-dwelling, pasture pests in New Zealand. Three viruses, Wiseana nuclear polyherosis (WNPV), Wiseana entomopox (WEpox), and Wiseana granulosis virus, have been found to infect this insect. WNPV, and, to a lesser extent WEpox, have been shown to be effective in controlling this pest to subeconomic levels. It has been estimated that 6 infected larvae/4 m2 are enough to produce an LD50 of WNPV for Wiseana spp. Provided that stock movement spreads the virus over the soil surface during the early larval instars, the virus maintains itself from one year to the next at a level ensuring high larval mortality. The fact that total insect control is not necessary in this situation means that a residual larval population can exist in equilibrium with virus to give continued virus production.  相似文献

A nucleopolyhedrovirus (SpliMNPV-Az) was isolated from diseased larvae of Spodoptera littoralis, collected at the Island of S. Miguel in Azores. The virulence of this isolate was tested against S. littoralis larvae in laboratory. LD50 against 2nd and 3rd instars were not significantly different, 1.44 × 104, 3.89 × 104 OBs per larvae, respectively, but both were significantly different from that against 4th instar, which was 61.3 × 104 OBs per larvae. The complete codons sequence of SpliMNPV-Az Polh gene obtained was 750 bp (NCBI GenBank Accession No. AY600451). This sequence was compared to other 38 polyhedrin genes from NPVs and to 6 granulin genes from GVs and resulted to be identical to the sequence of a SpliMNPV previously published, thus indicating that the natural host of SpliMNPV-Az must be S. littoralis. Genetic distances estimated from restriction enzymes profiles showed SpliMNPV-Az is close to the Egyptian SpliMNPV type B, despite some degree of genetic divergence suggested by slight differences observed on PstI profile.  相似文献

To determine the impact of Nomuraea rileyi on consumption by the green cloverworm, Plathypena scabra, larvae were reared from eggs obtained from field-collected moths, inoculated with conidia, and placed individually in separate plastic vials with a piece of surface-sterilized soybean leaflet. No significant differences in consumption rates were found between N. rileyi-inoculated and control larvae until after 144 hr post-treatment. After this period, consumption by larvae inoculated as first, second, third, or fourth instars was significantly less than that of control larvae because of mortality. First or second instars inoculated with N. rileyi conidia had significantly longer subsequent stadia than did control larvae. The average LT50 of all inoculated instar groups was 6.5 days, and there was little evidence of significantly different N. rileyi susceptibility among instar groups. In general during the N. rileyi infection period leading to their deaths, inoculated P. scabra larvae had consumption patterns that were very similar to those of healthy larvae.  相似文献

Biological control offers potentially effective suppression of the diamondback moth (DBM), Plutella xylostella, a serious pest of Brassica crops. Little is known of whether multiple natural enemies have additive, antagonistic, or synergistic effects on DBM populations. No-choice and choice tests were conducted to assess predation by Podisus maculiventris on DBM larvae parasitized by Cotesia plutellae and unparasitized larvae. In no-choice tests, P. maculiventris preyed on greater numbers of parasitized than unparasitized larvae and greater numbers of young larvae than old larvae. In choice tests with early third instar DBM, there was no difference in predation between parasitized or unparasitized larvae. However, in choice tests with older prey, P. maculiventris preyed on more parasitized than unparasitized larvae. Two field studies were conducted to test if this predator and parasitoid have additive, antagonistic or synergistic effects on DBM populations and plant damage in cabbage (Brassica oleracea var. capitata). In 2002, DBM populations were significantly lower in the presence of C. plutellae but not in the presence of P. maculiventris. There was not a significant interaction between the natural enemies. Plant damage was reduced only with C. plutellae. In 2003, DBM populations were significantly lower in the presence of C. plutellae and P. maculiventris, although the combination of natural enemies did not lead to a non-additive interaction. Plant damage was unaffected by the presence of either natural enemy. Because of its greater predation on parasitized larvae, P. maculiventris could be an intraguild predator of C. plutellae. Yet, their overall combined effect in the field was additive rather than antagonistic.  相似文献

棉铃虫Helicoverpa armigera(Hubner)中肠谷胱甘肽S-转移酶(GSTs)对甲基对硫磷和灭多威的代谢能力明显高于对马来酸二乙酯(DEM)和两个混剂。LD5剂量的对硫磷和灭多威对棉铃虫3龄幼虫GSTs的活性均没有诱导增加的影响,用LD50的选择剂量仅对硫磷组GSTs活性增加15%。用含0.01%的芸香苷、2-十三烷酮和槲皮素的人工饲料饲养棉铃虫经1~4代后,GSTs活性提高4~18倍。3种植物次生性物质诱导组对灭多威和溴氰菊酯的敏感度均没有明显的变化,而槲皮素组对甲基对硫磷的敏感度则降低近一半,芸香苷和2-十三烷酮组对甲基对硫磷的敏感度略有降低。这种对甲基对硫磷敏感度的变化可能与上述GSTs活性的变化有关。  相似文献
