干旱荒漠白刺灌丛植被演替过程土壤种子库变化特征   总被引:7,自引:0,他引:7

土壤种子库是反映气候与土地利用变化的指示器,也是荒漠植被更新与恢复的基础。典型荒漠白刺灌丛植被广泛分布于我国西北绿洲边缘地带,对保护绿洲免受风沙危害发挥着重要作用。但是,对干旱荒漠白刺灌丛植被土壤种子库仍未见研究报道。应用空间代替时间的方法,选择石羊河下游白刺灌丛植被演替的初始阶段、稳定阶段、衰退阶段与严重衰退阶段样地,研究了白刺灌丛植被演替过程的土壤种子库物种组成、时空分布和数量变化特征。结果表明:石羊河下游白刺灌丛植被演替过程中,土壤种子库共出现9科18种植物,其中草本植物占到55%—80%,是白刺灌丛植被土壤种子库的主体。不同演替阶段土壤种子库密度以稳定阶段最高,达到660.7粒/m2,是初始阶段、衰退阶段和严重衰退阶段的5.6、14.5倍和6.2倍。不同沙堆部位,土壤种子库密度以迎风坡中部与背风坡中部最高,分别达到329粒/m2和309粒/m2;沙堆土壤种子库密度是堆间地的1.9倍,土壤种子库主要分布于灌丛沙堆上。不同演替阶段土壤种子库间的相似性系数均在0.6以下,除稳定阶段外,其他阶段间均属于中等相似水平;土壤种子库与地上植被的相似性系数均在0.5以上,达到中等相似或极相似水平。该研究进一步证明了灌丛沙堆和土壤种子库在白刺灌丛植被中的重要性,也说明人为干扰损害了白刺灌丛地下潜在植被及其恢复潜力。因此,需要开展人工种子库或人工植被建设以控制干旱荒漠白刺灌丛沙堆活化,并实现退化白刺灌丛植被恢复。  相似文献

Allelopathy is an important process in plant communities, but the role of seed allelopathy in natural ecosystems remains poorly understood. In the present study, we examined the potential allelopathic effects of Ligularia virgaurea (a dominant species in degraded Tibetan grasslands) seeds on the germination of four native grass species (Festuca sinensis, Agrostis gigantean, Bromus inermis, and Elymus nutans). The results showed that L. virgaurea seeds can have potential allelopathic effects on seed germination, mean time to germination and root growth rates of native grass species. We further demonstrate that these effects are driven by a water-soluble seed leachate. Species with smaller seeds were generally more sensitive than larger seeded species. The results suggest that seed-to-seed allelopathic potential may be an important mechanism driving the dominance of L. virgaurea in degraded alpine grasslands on the Tibetan Plateau. Further studies are required to demonstrate effects of seed-to-seed potential allelopathy in a field setting.  相似文献

Summary Temperate grassy ecosystems are amongst Australia's most endangered ecosystems. Most remnants are small, fragmented and highly degraded. Practical methods for restoring native understorey species are urgently required. Dominant native grasses such as Kangaroo Grass (Themeda triandra Forssk.) and Tussock‐grasses (Poa species) have been eliminated from many remnants by heavy grazing in the past. The reintroduction of these grasses is a critical step for understorey restoration. This paper (i) reviews the literature on Themeda seed biology and seedling establishment; (ii) summarizes the lessons learnt from three major attempts to establish Themeda stands in south‐east Australia; and (iii) identifies the research needed to enhance Themeda restoration. Considerable information is available on Themeda seed and establishment biology, and restoration exercises have shown that Themeda stands can be readily established by surface‐spreading awned seeds in seed‐bearing hay. However, many practical challenges remain, including the need to identify optimal sowing periods, create better seedbed conditions, develop practical mulching techniques, and improve weed control. The use of seed‐bearing hay has constrained restoration to relatively small areas in the past. Future trials may benefit by using more concentrated seed products such as seed‐bearing florets and pure seeds which permit larger areas to be restored at one time.  相似文献

Natural seed deposition patterns and their effects on post-dispersal seed fate are critical to tropical tree recruitment. The major dispersal agents of the large-seeded tree Canarium euphyllum in Khao Yai National Park, Thailand, are large frugivorous birds such as hornbills, which generated spatially heterogeneous seed deposition patterns because they regurgitated seeds at perching trees and conspecific and heterospecific feeding trees. We investigated the fate of seeds dispersed in this manner using seed removal experiments and automatic camera trapping. Seeds placed experimentally around conspecific feeding trees had higher removal rates than seeds placed elsewhere. These effects were likely mediated by two seed-eating rodents, the Indochinese ground squirrel (Menetes berdmorei) and the giant long-tailed rat (Leopoldamys sabanus). Consequently, the spatial patterns generated by hornbills had consequences for post-dispersal seed fates, particularly whether or not the seeds were removed by rodents. Primary dispersal by hornbills does alter seed fate by altering the probability of rodent–seed interaction, but the ultimate impact of dispersal by hornbills will depend on how important rodent scatterhoarding is to seed germination and seedlings. Given that major seed dispersers of C. euphyllum are now absent or rare in degraded forests in tropical Asia, it is becoming increasingly important to understand the roles of scatterhoarding rodents in these altered habitats in this region.  相似文献

Phaseolin, the major seed storage protein of Phaseolus vulgaris L., is degraded in the cotyledons in the first 7–10 d following seed germination. We assayed cotyledon extracts for protease activity by using [3H]phaseolin as a substrate and then fractionated the digestion mixtures by sodium dodecyl sulfate-polyacrylamide gel electrophoresis in order to identify the cleavage products. The cotyledons of 4-d-old seedlings contain an endopeptidase which cleaves the polypeptides of [3H]phaseolin (apparent molecular weights=51 000, 48 000, 46 000 and 43 000) into three discrete clusters of proteolytic fragments (M rs=27 000, 25 000 and 23 000). Endopeptidase activity is not detected in the cotyledons until the protein content of these organs starts to decline, shortly after the first day of seedling growth. Endopeptidase activity increases to a maximum level in the cotyledons of 5-d-old seedlings and then declines to a minimum value by day 10. The enzyme was purified 335-fold by ammonium-sulfate precipitation, organomercurial-agarose chromatography, gel filtration and ion-exchange chromatography. The endopeptidase constitutes 0.3% of the protein content in the cotyledons of 4-d-old seedlings. It is a cysteine protease with a single polypeptide chain (M r=30 000). Optimum hydrolysis of [3H]phaseolin occurs at pH 5. The enzyme is irreversibly inactivated at pH values above 7 and at temperatures above 45° C. The endopeptidase attacks only a limited number of peptide bonds in [3H]phaseolin, without causing any appreciable change in the native molecular weight of the storage protein. The endopeptidase is also able to hydrolyze the bean-seed lectin, phytohemagglutinin. Fifty-two genotypes of Eleusine coracana collected from Uttarakhand hills were subjected to simple sequence repeat (SSR), random amplified polymorphic DNA (RAPD)-PCR and protein profiling analysis to investigate the variation in protein content. The main objective of the present study was to detect variability among E. coracana and also assess the discriminating ability of these three molecular methods. A total of 21 RAPD and 24 SSR primers were assayed for their specificity in detecting genetic variability in E. coracana, of which 20 RAPD and 21 SSR primers were highly reproducible and were found suitable for use in PCR analysis. Assessing genetic diversity among E. coracana genotypes by RAPD-PCR using 20 polymorphic primers yielded 56 different RAPD markers which clustered the genotypes into different groups on the basis of protein content. Similarly, SSR-PCR with 21 polymorphic primers clustered the genotypes into different groups. On the other hand, biochemical typing of E. coracana using whole seed proteins generated profiles that showed no major difference indicating the technique to be not useful in typing genotypes of this crop. However, a few of the genotypes showed the presence of a unique band of 32 kDa that needs to be further investigated to understand the role of the protein from nutritional point of view, if any. In the present study, significant negative correlation (r = −0.69*) was found between the protein and calcium content of finger millet genotypes. Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis based seed storage proteins generated profiles showed no major differences in banding pattern among 52 finger millet genotypes while quantitative estimation of seed storage protein fractions using Lowry method revealed that glutelin was highest followed by prolamin, globulin and albumin.  相似文献

We have analyzed the intracellular behavior of the human transferrin receptor (TfR) in Saccharomyces cerevisiae. The major part of the heterologously expressed TfR, which has previously been used as a model for heterologous expression of membrane proteins in yeast, is localized in the endoplasmic reticulum (ER) membranes; a minor fraction is present in the plasma membrane (PM). The stability of the TfR depends on vacuolar proteases, implying that it is degraded in the vacuolar compartment. Degradation is further dependent on favorable transport conditions to this compartment. The main bottleneck of transport seems to be the transition from the ER to the PM. The chaperone Cne1p, which is involved in quality control in the ER, plays a role in regulating the amount of heterologous TfR, as deletion of CNE1 leads to significant accumulation of the protein. Intensive large-scale revegetation programs are a major activity in degraded landscapes. The collection and deployment of seed for revegetation is primarily based on local seed sources to preclude concerns associated with local adaptation as well as outbreeding depression in small remnant populations. For most species however, little is known about the levels of genetic diversity being sourced and deployed, or even of the levels of adaptively significant variation present across the species range. Acacia acinacea (Gold Dust wattle) is a key revegetation species widely distributed across south eastern Australia. Levels of variation in terms of morphology, fitness, ploidy level and AFLPs were assessed using seed lots from 35 populations across the Murray Darling Basin in south eastern Australia. Multivariate analysis indicated that three of the populations were misidentified while the remaining populations were differentiated into three groups. One of these groups (Bendigo) was highly geographically localised and tetraploid while the other two (Group1 and Group2) were both diploid with overlapping distributions. Fitness differences were also evident among these three groups while AFLPs indicated that they were genetically differentiated and that lower levels of diversity were captured from some populations. A major finding of this study was that no one technique was able to fully describe the variation present in A. acinacea and seed sourcing guidelines based on any single approach could have resulted in erroneous decisions being made. Information generated from this study has been used to refine seed sourcing guidelines.  相似文献

Summary Electrophoretic profiles of crude protein extracts from seed of F1 hybrids and reciprocal crosses among diploid, tetraploid and hexaploid wheats were compared with those of their respective parental species. The electrophoretic patterns within each of three pairs of reciprocal crosses, T.boeoticum X T.urartu, T.monococcun X T. urartu and T.dicoccum X T. araraticum, were different from one another but were identical with those of their respective maternal parents. Protein bands characteristic of the paternal parents were missing in F1 hybrid seed suggesting that the major seed proteins in wheat were presumably regulated by genotype of the maternal parent rather than by the seed genotype. However, in another three pairs of reciprocal crosses, T.boeoticum X T. durum, T.dicoccum X T.aestivum and T. zhukovskyi x T. aestivum, protein bands attributable to the paternal parents were present in the F1 hybrid seeds indicating that the seed proteins were not always exclusively regulated by the maternal genotype. The expression of paternal genomes is presumably determined by dosage and genetic affinity of the maternal and paternal genomes in the hybrid endosperm. The maternal regulation of seed protein content is probably accomplished through the maternal control over seed size. The seed protein quality may, however, depend upon the extent of expression of the paternal genome.  相似文献

The Restionaceae is one of the three major families defining fynbos, the characteristic vegetation type of the Cape Floristic Region. Periodic fires with a frequency of 5 to 40 years are a natural phenomenon in fynbos vegetation. Fire-stimulated seed germination has been reported for a variety of fynbos species, and species in the Asteraceae, Ericaceae, Proteaceae and Restionaceae have shown a germination response to smoke and/or aqueous smoke extracts. In the present study seed of 32 species was screened to obtain an indication of how important the smoke cue is for germination in the South African Restionaceae. The results of the present study represents the first occasion that comparative germination data for South African species in this family have ever been obtained. Twenty-five of the 32 species tested showed a statistically significant improvement in germination following smoke treatment. Untreated seeds of 18 of the species responding, showed a high degree of dormancy with only 0.1% to 2.0% germination. These results suggest that under natural conditions smoke from fynbos fires may provide an important cue for triggering seed germination in this family. The degree of improvement in germination following smoke treatment ranged from 147% in the case of Restio festuciformis to 25300% in the case of Rhodocoma capensis. It is suggested that the 16 species which showed a 1000% or more increase in germination following smoke treatment form a group in which smoke is likely to be the major cue for germination. In those species in which there is a lesser response, smoke may be one of a number of germination cues which include heat, and possibly alternating high and low incubation temperatures. The four species that did not germinate were all myrmecochorus, nut-fruited species. More information is needed concerning the fire survival strategy of South African Restionaceae species and many more will have to be investigated in the nursery and in the field, before the full pattern of response within the family and its significance can be elucidated.  相似文献

Desertification is a major environmental problem in arid and semiarid regions. Tree plantation has been commonly employed to foster the recovery of degraded areas. However, this technique is costly, and their outcomes are often uncertain. Therefore, we evaluated an alternative method for the restoration of degraded semiarid steppes that involved the construction of branch piles to attract frugivores as potential seed‐dispersing birds, promoting seed rain, and fostering the formation of woody patches. We measured the success of branch piles in terms of the number of bird visits and seed input compared to naturally occurring shrub patches. Generally, frugivorous birds visited branch piles less frequently than shrub patches. Yet, branch piles accumulated seeds of patch‐forming shrub species. Seed rain was higher under patches of the dominant shrub Rhamnus lycioides than under branch piles. In contrast, woody patches and branch piles did not differ in seed input of the less abundant Pistacia lentiscus shrub. Our study demonstrates that branch piles are used by frugivorous birds and accumulate seeds of patch‐forming shrubs. Branch piles may be a suitable method to promote the expansion of bird‐dispersed plant species and restore semiarid wooded steppes. However, their efficiency largely depends on pile persistence and economic cost.  相似文献

Lack of seed dispersal can be an important obstacle to natural regeneration (NR) of degraded pastures in the humid tropics. Tree plantations can facilitate secondary forest succession by attracting seed dispersal agents from nearby forests. We studied seed rain and seed dispersal agents in 12–13 years old pure and mixed native tree plantations at La Selva Biological Station, Costa Rica from July to December 2004. Plantations of Balizia elegans (5,522), Dipteryx panamensis (2,263), and Jacaranda copaia (2,091) had the greatest total seed abundance; treatments with the least total seed abundance were Calophyllum brasiliense (56), nonplanted abandoned pasture control 2 (353), Mixed Species 2 (389), and control 1 (836). Plantations of J. copaia and Hyeronima alchorneoides had the greatest seed species richness density, whereas the lowest seed species richness was found in the control treatments. The NR plots had more seeds dispersed by wind, whereas in the plantations, the most important dispersal agents were birds and mammals. The most abundant seeds were those of Miconia spp. (14,492), Psychotria bracheata (2,252), and the Poaceae family (1,346), all species from early successional stages. Plantations of native species are effective in attracting seed dispersal agents and thus facilitating regeneration of degraded pasturelands in the region.  相似文献

Summary We have examined mitochondrial (mt) ribosome assembly and-function in five nuclear and six extranuclear mutants of Neurospora crassa which had previously been characterized as deficient in cytochromes b and aa 3. All six extranuclear mutants showed phenotypes similar to that previously described for the extranuclear [poky] mutant: small subunit-deficient with 19 S rRNA rapidly degraded. The nuclear mutants have the following phenotypes: 297-24 is mt small subunit deficient with 19 S RNA rapidly degraded. 289-56 is mt small subunit deficient but contains normal ratios of 19 S to 25 S RNA in whole mitochondria. 289-67 and 299-9 show defects in the processing of 25 S RNA leading to accumulation of a large precursor RNA. 289-4 is deficient in large subunits although a substantial, but less than normal, amount of 25 S RNA is present in the mitochondria.The present work provides new insight into the phenotypes of mt small subunit-deficient mutants. Previous studies using chloramphenicol suggest that some defects in the assembly of mt small subunits may arise secondarily as a result of inhibition of mt protein synthesis (LaPolla and Lambowitz, 1977; Lambowitz et al., 1979). Three mutants (289-56, 289-67 and 299-9) appear to show such defects. These strains contain incomplete mt small subunits which sediment more slowly than normal and are deficient in at least two proteins, S-5 and S-9. Correlation of mutant phenotypes with rates of mt protein synthesis in the different strains suggests that mt protein synthesis must be decreased to less than one half of the wild-type rate before secondary defects in mt small subunit assembly are observed. This threshold value is much lower than that which leads to gross deficiencies of cytochromes b and aa 3. Although several mutants have phenotypes suggestive of alterations in mt ribosomal proteins, no such alterations could be identified by two dimensional gel electrophoresis.  相似文献

Seed rain is a crucial element in vegetation regeneration, but has been rarely studied in high altitude regions, particularly degraded Kobresia meadow. Weed infestation is a distinctive feature of pasture degradation in Kobresia meadows on the Tibetan plateau, the ecological mechanism of which is closely related with vegetation’s seed rain. In this paper we assess the effect of vegetation degradation on seed rain and consider its implication for restoration of degraded Kobresia meadows in the headwater area of Yellow river, through analysis of seed species composition, number of seeds landing per m2 of soil surface, and their relationship with above ground vegetation. Vegetation degradation had an impact on the species composition and numbers of seeds in seed rain and their relationship with above-ground vegetation. Within the un-degraded meadow, which provided a closed vegetation cover, 35 % of the seed rain was of sedge and gramineae species. However, within the degraded meadows, as the extent of degradation increased, so the total number of seeds m?2 increased, with those derived from sedge and gramineae species forming a declining proportion of the total. Degradation of Kobresia meadow on the Tibetan plateau is exacerbated by the seed input of weed species (such as Oxytropis ochrocephala, Carum carvi, Aconitum pendulum, Pedicularis kansuensis in this study). Therefore, a major priority for the restoration of such degraded meadows should be the elimination of these weeds from the above ground vegetation by human intervention.  相似文献

Summary A suppressor mutation of a temperature-sensitive mutant of ribosomal protein L24 (rplX19) was mapped close to the lon gene by genetic analysis and was shown to affect protease LA. The degradation and the synthesis rates of individual ribosomal proteins were determined. Proteins L24, L14, L15 and L27 were found to be degraded faster in the original rplX19 mutant than in the rplX19 mutant containing the suppressor mutation. Other ribosomal proteins were either weakly or not at all degraded in both mutants. Temperature-sensitive growth was also suppressed by the overproduction of mutant protein L24 from a plasmid. Our results suggest that (1) either free ribosomal proteins or proteins bound to abortive assembly precursors are highly susceptible to the lon gene product and (2) the mutationally altered protein L24 can still function at the nonpermissive growth temperature of the mutant, if it is present in sufficient amounts.  相似文献

Forest succession on degraded tropical lands often is slowed by impoverished seed banks and low rates of seed dispersal. Within degraded landscapes, remnant forests are potential seed sources that could enhance nearby forest succession. The spatial extent that forest can influence succession, however, remains largely unstudied. In abandoned agricultural lands in Kibale National Park, Uganda, recurrent fires have helped perpetuate the dominance of tall (2–3 m) grasses. We examined the effects of distance from forest and grassland vegetation structure on succession in a grassland having several years of fire exclusion. At 10 and 25 m from forest edge, we quantified vegetation patterns, seed predation, and survival of planted tree seedlings. Natural vegetation was similar at both distances, as was seed (eight species) and seedling (six species) survival; however, distance may be important at spatial or temporal scales not examined in this study. Our results offer insight into forest succession on degraded tropical grasslands following fire exclusion. Naturally recruited trees and tree seedlings were scarce, and seed survival was low (20% after 7 mo). While seedling survival was high (95% after 6 to 8 mo), seedling shoot growth was very slow (x?= 0.5 cm/100 d), suggesting that survivorship eventually may decline. Recurrent fires often impede forest succession in degraded tropical grasslands; however, even with fire exclusion, our study suggests that forest succession can be very slow, even in close proximity to forest.  相似文献

Three major hemolymph proteins of Papilio polyxenes larvae were isolated and characterized. Density gradient ultracentrifugation of hemolymph resulted in flotation of the major lipoprotein, lipophorin. P. polyxenes larval lipophorin is composed of two apoproteins, apolipophorin-I and apolipophorin-II, plus a mixture of lipids, to give a density of 1.13 g/ml. Immunoblotting experiments using antisera directed against Manduca sexta apolipophorin-I and apolipophorin-II, respectively, revealed cross-reactivity of apoLp-I with Manduca sexta apoLp-I, and apoLp-II with M. sexta apoLp-II. Gel permeation chromatography of the subnatant obtained following density gradient ultracentrifugation revealed the presence of a major protein peak which was shown to contain three major serum proteins, two of which were isolated and characterized. One of these proteins was purified by lectin affinity chromatography. Both proteins have native molecular weights in the range of 450,000 and appear to be hexamers of a single subunit type. Major serum protein-1 is nonglycosylated and has a subunit molecular weight of 75,000. Major serum protein-2 is glycosylated and has a subunit molecular weight of 74,000. Amino acid analysis of this protein revealed a tyrosine plus phenylalanine content of 20 mole percent, characteristic of the arylphorin class of insect storage proteins. Using antibodies against M. sexta larval hemolymph proteins, both the P. polyxenes major serum proteins were shown to be immunologically related to serum proteins of other lepidopteran species.  相似文献

Two abscisic acid (ABA)-responsive seed proteins, ABR17 and ABR18 (ABA-responsive 17000-Mr and 18000-Mr, respectively), previously found to be induced in cultured embryos of pea (Pisum sativum L.) are major components synthesised during normal seed desiccation. The ABR17 and ABR18 proteins showed different patterns of accumulation. The ABR18 protein was abundant in the testa during early seed development but in desiccating seed it was synthesised in the embryo, indicating spacial as well as temporal regulation of expression. The ABR18 protein was undetectable soon after germination but reappeared after adding ABA. The ABR17 protein was not detected in the testa but appeared in the embryo just prior to maximum fresh weight. The ABR17 protein continued to be synthesised during germination and was also present in non-stressed leaves. A high level of endogenous ABA or added ABA increased levels of translatable ABR17 mRNA. The ABR17 and ABR18 proteins were further characterised so as to help determine their structure and function. Neither protein appeared to contain a signal peptide but both proteins appeared to be glycosylated. The proteins had similar amino-acid compositions and limited Nterminal analysis showed 56% sequence identity. Neither protein had any significant N-terminal sequence homology to any of the late embryogenesis-abundant (LEA) proteins or dehydrins.
