The cacao mealybug, Planococcus lilacinus Cock, is an important quarantine pest. Infested commodity should be subject to appropriate phytosanitary treatment, while irradiation is recommended for the cacao mealybug. Radio-tolerance comparison tests were conducted on the crawler, nymphs, and adult females of P. lilacinus at the X-ray radiation doses of 40, 80, and 120 Gy, respectively. The results showed that irradiation had a strong effect on preventing of development and reproduction; the adult female stage was identified as the most tolerant. During the following dose–response tests, among young and late females X-ray-irradiation (20–100 Gy), the late females were most tolerant when preventing F1 generation 2nd instars emergence was used as the evaluation criterion. Minimum absorbed dose and its 95 % fiducial limits to provide probit 9 efficacy at 95 % confidence level (100 % mortality/inhibition in an estimated population of 93,616 individuals) were 131.5 Gy (122.5, 142.6 Gy) and 144.4 Gy (132.7, 159.4 Gy), estimating from the probit analysis on dose-mortality data of 1–30 and 1–10-day-old neonates laid by late females, respectively. In the large-scale confirmatory tests, a total of estimating 97,384 late females of P. lilacinus rearing on the pumpkins fruits were irradiated with gamma-ray at the target dose of 135 or 145 Gy (measured doses 126.1–163.0 Gy), which resulted in no F1 generation 2nd nymphs developing during a 6-week post-treatment period. The treatment efficacy calculated is 99.9969 % at the 95 % confidence level. Therefore, a minimum absorbed dose of 163.0 Gy is recommended for phytosanitary treatment of P. lilacinus in infested commodity.  相似文献

Early adverse life stress has been associated to behavioral disorders that can manifest as inappropriate or aggressive responses to social challenges. In this study, we analyzed the effects of artificial rearing on the open field and burial behavioral tests and on GFAP, c‐Fos immunoreactivity, and glucose metabolism measured in anxiety‐related brain areas. Artificial rearing of male rats was performed by supplying artificial milk through a cheek cannula and tactile stimulation, mimicking the mother's licking to rat pups from the fourth postnatal day until weaning. Tactile stimulation was applied twice a day, at morning and at night, by means of a camel brush on the rat anogenital area. As compared to mother reared rats, greater aggressiveness, and boldness, stereotyped behavior (burial conduct) was observed in artificially reared rats which occurred in parallel to a reduction of GFAP immunoreactivity in somatosensory cortex, c‐Fos immunoreactivity at the amygdala and primary somatosensory cortex, and lower metabolism in amygdala (as measured by 2‐deoxi‐2‐[18fluoro]‐d ‐glucose uptake, assessed by microPET imaging). These results could suggest that tactile and/or chemical stimuli from the mother and littermates carry relevant information for the proper development of the central nervous system, particularly in brain areas involved with emotions and social relationships of the rat. © 2017 Wiley Periodicals, Inc. Summary Complementation of two metabolic deficiences — nitrate reductase and tryptophan synthase — was used to select for somatic fusion hybrids between tobacco (Nicotiana tabacum) and henbane (Hyoscyamus muticus) with prior X-irradiation of one partner. Using species specific, radioactively labelled DNA probes it could be shown that a) irradiation significantly reduced the amount of chromosomal DNA of the irradiated fusion partner in the somatic hybrid, b) irradiation with doses which completely inhibit protoplast division did not pevent transfer of substantial amounts of chromosomal DNA into the fusion hybrids (so called cybrids) and c) this method transfers functional nuclear genes together with the partial genome from the irradiated partner.  相似文献

Invitro phosphorylation and acetylation of nonhistone chromosomal (NHC) proteins and their modulation by Ca++ and estradiol were studied by incubating slices of cerebral cortex of 2-, 15- and 84-week female rats with 32Pi and 14C-Na-acetate. Phosphorylation pattern of NHC proteins is unique for each age. Ca++ and estradiol stimulate phosphorylation of different NHC proteins which is also age-specific. Acetylation of NHC proteins decreases precipitously with age. No unique NHC protein is acetylated preferentially at any age, nor does Ca++ stimulate acetylation. Estradiol, however, stimulates acetylation of a few NHC proteins. It is suggested that phosphorylation of NHC proteins and its modulation by effectors may be more important for gene expression than their acetylation.  相似文献

We studied the effects of i.p. injection of melatonin in pharmacotherapeutic doses and of constant illumination (a melatonin synthesis-suppressing factor) on the behavior of rats in the open-field test and on the content of the main isoforms of neural cell adhesion molecule (NCAM) in the hippocampus, cerebellum, and neocortex of these animals. In the studied brain structures of the rats kept under conditions preventing the melatonin synthesis, we observed suppression of the behavioral activity of animals and a decrease in the expression of the NCAM180 isoform. In rats injected with 10 mg/kg melatonin, changes in the behavioral activity were insignificant. In the hippocampus and neocortex of rats of this group, the NCAM180 content increased. Our experiments showed that melatonin can be involved in the control of balance of the expression of different NCAM isoforms. Such a balance is a crucial factor determining plastic rearrangements of the synaptic contacts.  相似文献

We studied changes in behavior of Mongolian gerbils (Meriones unguiculatus) after ischemia-reperfusion of the brain (7-min-long occlusion of both arteriae carotis) and structural alterations in the hippocampus of such animals. Behavioral manifestations were observed under conditions of the open field test within 7 days of the postischemic period; immunofluorescent staining of brain sections with antibodies against specific proteins of neurons and glial cells was used. Motor hyperactivity reaching its maximum a day after ischemization and gradually decreasing within the postocclusion period was found in ischemized animals. On the 7th day, the level of locomotor activity in experimental gerbils was practically equal to that in the control group. In contrast, a postischemic decrease in the duration of episodes of resting was preserved for a longer time. A week after ischemia-reperfusion, intense delayed neuronal death and activation of glial cells were observed in the CA1 hippocampal area. Thus, cerebral ischemia-reperfusion results in significant transient disorders of behavioral phenomena in gerbils; at the same time, a clear correlation is observed between structural changes of neurons and the level of reactivity of glial cells in the hippocampus. These events can be significant aspects of the dynamics of postischemic damage to the above structure. 目的观察X线全身照射对2型糖尿病模型KKAy小鼠的造血免疫系统功能的损伤作用,并与对照C57小鼠进行比较。方法KKAy小鼠,分为对照组和照射组,照射组小鼠经X线全身照射,剂量4Gy,C57小鼠作为对照。照射后15d检测小鼠的外周血常规,流式细胞术检测骨髓中造血祖细胞、造血干细胞和长期造血干细胞的比例,脾中B细胞和T细胞的比例,胸腺中CD4CD8双阳性T细胞、CD4单阳性T细胞和CD8单阳性T细胞的比例。通过粒细胞集落形成能力实验评价小鼠造血祖细胞的功能。结果照射前KKAy小鼠的HSC和LT-HSC的比例低于C57小鼠。4Gy全身照射后,KKAy小鼠的外周血WBC、RBC、PLT、HGB和LYM%分别下降了68.42%、12.17%、8.78%、30.12%、70.84%;骨髓中HPC、HSC和LT-HSC的比例分别下降了34.02%、29.49%、35.74%;脾B细胞和T细胞的比例分别下降了57.85%、58.81%;胸腺CD4CD8双阳性细胞的比例下降了51.70%。KKAy小鼠的骨髓HSC、LT-HSC、外周血RBC和HGB的降低幅度显著低于C57小鼠。结论4Gy全身照射损伤KKAy小鼠的造血免疫系统功能,KKAy小鼠可能比C57小鼠表现出对电离辐射较强的耐受性。  相似文献

采用旷场行为测试方法, 测定204条德国牧羊犬、拉布拉多犬、史宾格犬二月龄幼犬在新异环境下的兴奋性和探索活动, 同时应用RFLP-PCR方法检测单胺氧化酶B(Monoamine Oxidase B, MAOB)基因的多态性, 分析MAOB基因的基因型和基因频率在品种间的分布差异以及基因多态性与旷场测验中行为参数的相互关系, 发现MAOB基因型频率与基因频率在犬品种之间差异极显著(P<0.01), MAOB基因型与幼犬在旷场中的走动时间、趴卧时间、跨格次数、站立扒墙次数有关(P<0.01或P<0.05), 对运动姿势改变次数也有一定影响(P=0.064)。其中, TT基因型犬的走动时间和跨格次数均高于TC型和CC型犬(P<0.05), 运动姿势改变次数和站立扒墙次数高于CC型犬(P<0.05); 而CC型个体的趴卧时间高于TT型个体(P<0.05)。MAOB基因对走动时间和跨格次数的加性遗传效应达极显著(P<0.01), 对运动姿势改变次数、站立扒墙次数和趴卧时间有显著的加性效应(P<0.05)。实验结果表明, MAOB基因与幼犬在旷场测验中的运动、兴奋性和探索活动有关, TT基因型对运动、兴奋性和探索活动具有正遗传效应。  相似文献

During purification of branched chain ketoacid dehydrogenase from Pseudomonasputida, large losses in enzyme activity occurred. Much of the activity was restored by the addition of a heat-treated, soluble fraction. The factor had a molecular weight less than 1000 and, of several potential effectors tested, the branched chain amino acids were the only compounds which stimulated enzyme activity, with valine being the most effective. The concentration of valine in the heat-treated fraction was found to be sufficient to account for all of the stimulation produced by this fraction. Valine had no effect on either pyruvate or 2-ketoglutarate dehydrogenases.  相似文献

IL-10 is known to suppress the inflammatory responses in a variety of experimental models. Because we previously found that whole-body X-irradiation causes massive apoptosis in the thymus and transient infiltration of neutrophils, in this study, we examined whether or not IL-10 is involved in the regulation of neutrophil infiltration upon whole-body X-ray irradiation using IL-10 knockout mice. Although IL-10 was induced in the thymus on whole-body X-ray irradiation, apoptosis of thymocytes, neutrophil infiltration, and MIP-2 and KC production in the thymus were not affected by an IL-10 deficiency. Coculturing of bone marrow-derived macrophages with late apoptotic cells caused MIP-2 production, which was also not affected by an IL-10 deficiency. These results suggest the uniqueness of the inflammatory response induced by whole-body X-ray irradiation, which does not seem to be regulated by IL-10.  相似文献

We studied the effects of i.p. injections of L-DOPA (100 mg/kg) on the behavioral activity of Wistar rats and spontaneously hypertensive rats (SHR) in the open-field test, as well as on the content of catecholamines in the blood plasma of these animals. Prior to the administration of L-DOPA, the total level of locomotor/research activity in SHR was higher, as compared with that in Wistar rats. This was manifested in significantly greater values of the duration of moving of the animals in the center and on the periphery of the field and also in a greater number of rearings on the periphery of this field. At the same time, the episodes of grooming and sitting in Wistar rats were longer. After injections of L-DOPA, interstrain differences increased and became significant for most (9/10) indices of behavioral activity examined in our study. Injections of L-DOPA resulted in significant modifications of the behavioral activity of rats of the above strains, which is evidenced by changes in the number of visits to the peripheral squares of the open field and of rearings in the same field zones. Over repetitive test sessions, interstrain differences between most indices of behavioral activity (except the duration of research activity on the periphery and that of sitting) decreased. Injections of L-DOPA resulted in a significant increase in the content of this agent and dopamine in the blood plasma of rats of both strains; the level of noradrenaline in SHR also increased. The importance of a hereditary factor-determined increase in the activity of catecholaminergic brain systems (first of all, the dopaminergic system) in SHR for the specificity of locomotor behavior of these animals is discussed.  相似文献

A major challenge in structural biology is to determine the configuration of domains and proteins in multidomain proteins and assemblies, respectively. All available data should be considered to maximize the accuracy and precision of these models. Small-angle X-ray scattering (SAXS) efficiently provides low-resolution experimental data about the shapes of proteins and their assemblies. Thus, we integrated SAXS profiles into our software for modeling proteins and their assemblies by satisfaction of spatial restraints. Specifically, we modeled the quaternary structures of multidomain proteins with structurally defined rigid domains as well as quaternary structures of binary complexes of structurally defined rigid proteins. In addition to SAXS profiles and the component structures, we used stereochemical restraints and an atomic distance-dependent statistical potential. The scoring function is optimized by a biased Monte Carlo protocol, including quasi-Newton and simulated annealing schemes. The final prediction corresponds to the best scoring solution in the largest cluster of many independently calculated solutions. To quantify how well the quaternary structures are determined based on their SAXS profiles, we used a benchmark of 12 simulated examples as well as an experimental SAXS profile of the homotetramer d-xylose isomerase. Optimization of the SAXS-dependent scoring function generally results in accurate models if sufficiently precise approximations for the constituent rigid bodies are available; otherwise, the best scoring models can have significant errors. Thus, SAXS profiles can play a useful role in the structural characterization of proteins and assemblies if they are combined with additional data and used judiciously. Our integration of a SAXS profile into modeling by satisfaction of spatial restraints will facilitate further integration of different kinds of data for structure determination of proteins and their assemblies.  相似文献

Oxidative stress, an imbalance between oxidants and antioxidants, contributes to the pathogenesis of traumatic brain injury (TBI). Oxidative neurodegeneration is a key mediator of exacerbated morphological responses and deficits in behavioral recoveries. The present study assessed early hippocampal sequential imbalance to possibly enhance antioxidant therapy. Young adult male Sprague-Dawley rats were subjected to a unilateral moderate cortical contusion. At various times post-TBI, animals were killed and the hippocampus was analyzed for antioxidants (GSH, GSSG, glutathione peroxidase, glutathione reductase, glutathione-S-transferase, glucose-6-phosphate dehydrogenase, superoxide dismutase, and catalase) and oxidants (acrolein, 4-hydroxynonenal, protein carbonyl, and 3-nitrotyrosine). Synaptic markers (synapsin I, postsynaptic density protein 95, synapse-associated protein 97, growth-associated protein 43) were also analyzed. All values were compared with those for sham-operated animals. Significant time-dependent changes in antioxidants were observed as early as 3 h posttrauma and paralleled increases in oxidants (4-hydroxynonenal, acrolein, and protein carbonyl), with peak values obtained at 24-48 h. Time-dependent changes in synaptic proteins (synapsin I, postsynaptic density protein 95, and synapse-associated protein 97) occurred well after levels of oxidants peaked. These results indicate that depletion of antioxidant systems following trauma could adversely affect synaptic function and plasticity. Early onset of oxidative stress suggests that the initial therapeutic window following TBI appears to be relatively short, and it may be necessary to stagger selective types of antioxidant therapy to target specific oxidative components.  相似文献

We examined, using a Western blot technique, the contents and compositions of a specific neuronal protein, NCAM, and of an astrocyte marker, GFAP, in the hippocampus and cortex of rats with streptozotocin (STZ)-induced diabetes and compared these indices with those in control (intact) animals and STZ-diabetic rats treated with melatonin. Behavioral cognitive indices manifested in the passive avoidance test (PAT) and Morris water maze (MWM) learning performance were also estimated in the above groups of animals. As was found, STZ-diabetic rats demonstrated clear cognitive deficits according to the values of the retention latency in the PAT and time of reaching the escape platform in the MWM performance. In these animals, the GFAP content was elevated, and the amount of degraded products of this protein increased, as compared with the control. Simultaneously, considerable down-regulation of the NCAM expression and modifications of NCAM isoform composition were found in diabetic animals. In addition, significantly increased levels of lipid peroxidation (according to the amounts of malondialdehyde + 4-hydroxyalkenals) were measured in the cortex and hippocampus of rats with stable diabetic hyperglycemia. All the above-mentioned shifts were significantly smoothed or even nearly completely compensated in the case of treatment of STZ-diabetic rats with melatonin (10 mg/kg per day). The role of diabetes-related changes in the amount and composition of specific neural and glial proteins in the development of cognitive deficits, the involvement of oxidative stress in the mechanisms of the respective shifts, and possible mechanisms of the neuroprotective effect of melatonin with respect to diabetes-related pathological biochemical and behavioral shifts are discussed. The paper deals with changes in the activity of transaldolase in tissues of the liver, spleen and bone marrow of rats 4.24 h and on the 3 d, 7th, 18th, 30th, 45th, and 60th day after total X-ray irradiation in a dose of 600 R. The ionizing irradiation causes a stable inhibition of the enzyme activity in All tissues. The observed changes are homogeneous in their direction but have their own peculiarities for each tissue. Differences in the degree and extent of the enzyme activity changes at various stages of the X-ray lesion development in the tissues under study may be explained by their different sensitivity to the penetrating radiation.  相似文献

A study was made of the influence of gamma-irradiation of rats (14.4, 9.6, 7.2 and 4.8 Gy) on the number of nucleate cells, and the concentration of triacylglycerides (TG) and phospholipids (PL). Cellularity and concentration of PL decreased while that of TG increased under the effect of radiation. The degree of the changes in the above parameters was a function of dose and time after irradiation.  相似文献

The macrocycles L1-L3 having N2S2O-, N2S2-, and N2S3-donor sets, respectively, and incorporating the 1,10-phenanthroline unit interact in EtOH and MeCN solutions with CuII to give 1:1 [M(L)]2+ complex species. The compounds [Cu(L1)(ClO4)]ClO4 (1), [Cu(L2)(ClO4)]ClO4 ·  (2) and [Cu(L3)](ClO4)2 (3) were isolated at the solid state and the first two also characterised by X-ray diffraction studies. The conformation adopted by L1 and L2 in the cation complexes reveals the aliphatic portion of the rings folded over the plane containing the heteroaromatic moiety with the ligands encapsulating the metal centre within their cavity by imposing, respectively, a square-based pyramidal and a square planar geometry. In both complexes, the metal ion completes its coordination sphere by interacting with a ClO4 ligand. The compound [Cu(L3)2](PF6)2 (4) containing a 1:2 cation complex was also isolated at the solid state: EPR spectroscopy measurements suggest the presence of a CuN4 chromophore in this complex. The EPR and electronic spectral features of 1-4 have been studied and their redox properties examined in comparison with those observed for Type-1 blue copper proteins.The reactivity of L1-L3 has also been tested toward stoichiometric amounts of the CuI salt [CuCl(PPh3)3].  相似文献

实验采用原子吸收分光光度法,快速周期伏安法,高效液相电化学检测等方法,研究以6-羟基多巴(6-OHDA)制备的帕金森病(PD)模型大鼠黑质内铁含量的变化。铁对多巴胺(DA)能神经元的直接毒性作用以及铁离子螯合剂甲磺酸去铁胺的神经保护作用。结果发现:(1)PD大鼠损毁侧黑质内铁含量为非标准PD大鼠的3倍左右;(2)PD大鼠损毁侧纹状体内铁含量无明显改变;(3)单纯注射6-OHDA的大鼠其损毁侧纹状体(CPu)DA的释放量和含量均明显降低;(4)侧脑室预先注射甲磺酸去铁胺,再重复上述实验,损毁侧CPu DA释放量和含量均无明显改变;(5)单侧黑质内注射40ug FeCl3后,大鼠损毁侧CPu内DA释放量和含量显著降低。上述结果提示,6-OHDA可导致CPu DA释放量及含量减少,此过程有铁的参与。由于铁可导致DA神经元死亡,因此铁含量的增加可能是DA含量减少的原因之一,甲磺酸去铁胺具有保护DA神经元的作用。  相似文献

A study was made of the incorporation of 35S-methionine into nuclear matrix proteins of hepatic cells of pregnant rats and their embryos subjected to single gamma-irradiation (60Co, 1 and 2 Gy, 0.0233 Gy/s) on days 3, 13 and 17 of pregnancy and embryogenesis. On day 21 of pregnancy and embryogenesis a decrease in the rate of incorporation of 35S-methionine into nuclear matrix proteins was shown to be a function of radiation dose and time of pregnancy and embryogenesis on the moment of exposure.  相似文献
