植物名称:三七(Panax notoginseng)。材料类别:茎、叶柄、叶。培养条件:(1)诱导愈伤组织和胚状体发生培养基为MS 2,4-D0.5—1.0mg/L(单位下同)(2)胚状体成苗培养基为M8 BAI GA1。培养温度26士1℃,愈伤组织诱导至胚状体发生过程为暗培荞;胚状体成苗为光培养,每日光照10小时,光照度1000~1500lx。生长与分化情况:取还未完全展开幼叶的小苗,按常规消毒后,以茎、总叶柄、小叶柄和叶外植体分别切成小段或小块,切段长度为0.5~0.7cm,叶片切块面积为0.7~1.0cm~2,接种于培养基(1)中,  相似文献

菜薹花药培养诱导胚状体的研究   总被引:1,自引:0,他引:1  
分别以特早50、青梗菜心、9872和四九菜心(19)为供试材料,进行了基因型、温度、碳源等因素对菜心花药培养诱导胚状体影响的研究.结果 表明:不同基因型菜薹花药诱导率的差异显著,在改良B5固体培养基上特早50、9872、四九菜心(19)的诱导率依次降低,而青梗菜心诱导不出胚状体;不同温度的前处理对9872花药的诱导率也存在影响,其中以4℃低温1预处理幼花序1~2d和33℃(2温预培养24~48h的处理效果最佳;碳源对胚状体的诱导也有较大影响,其中蔗糖效果好于麦芽糖,且质量浓度为13%的蔗糖对特早50诱导率最高,而13%的麦芽糖诱导不出胚状体.  相似文献

对大蒜组织培养中胚状体的发生进行了研究。在分化培养中观察到了不同形态阶段的胚状体。经多次继代培养,颗粒状细胞团仍保持不断产生胚状体的能力并分化成苗。在同一颗粒状细胞团中,可以观察到单个的原胚细胞,2—4细胞和多细胞的原胚细胞团。  相似文献

甘蓝花药培养胚状体诱导形成影响因子研究   总被引:1,自引:0,他引:1  
用甘蓝F1、F2和自交系S33个世代6种基因型材料进行甘蓝花药培养诱导胚状体形成影响因子研究。结果显示:(1)高浓度蔗糖对甘蓝胚状体形成具有显著的诱导作用,6%蔗糖浓度是甘蓝花药培养的最适浓度,其胚状体的诱导率最高达12.2%;(2)材料基因型是影响花药培养的主要因素,F2和F1代材料胚状体诱导效果好,且胚状体诱导率F2代(F2P192和F2P194)18.9%比F1代(F1S17和F1S13)17.1%较高,但差异不显著,自交系S3代材料很难诱导出胚状体;(3)B5培养基比MS培养基更适合甘蓝花药胚状体的诱导培养。结果表明,甘蓝F2代是其花药诱导培养胚状体的最佳基因型材料,B5 2.0 mg/L 2,4-D 2.0 mg/L KT 6%Suc是甘蓝花药诱导培养胚状体的最适培养基。  相似文献

金花茶子叶在离体培养中胚状体的发生和小植株的形成   总被引:3,自引:0,他引:3  
研究了金花茶(Camellia chrysantha(Hu)Tuyama)子叶在离体培养中体细胞胚状体发生的条件。在MS基本培养基中附加苄基嘌呤(BA)或苄基嘌呤与萘乙酸(NAA)组合,诱导了胚状体发生。组织学观察表明,胚状体起源于子叶的表皮细胞。在增添细胞分裂素和生长素的MS或改良B_5液体培养基里振荡培养,明显地促进了胚状体根的生长和茎的发育。胚状体在继代培养中能保持旺盛的再生能力。已得到两个繁殖率较高的胚状体无性系。在合适的条件下,胚状体能长成正常的小植株。  相似文献

在甘蔗体细胞组织培养中,发现胚状体的发生较普遍,我们已从六个甘蔗品种的体细胞培养诱导出胚状体。 选出一号、八号、九号三个胚性细胞无性繁殖系,经1年10个月16次继代培养,仍能保持旺盛的繁殖能力,并能正常分化成植株。 高浓度的 2,4-D(2~3毫克/升)能促进胚性细胞团的繁殖和抑制绿芽的产生,低浓度或除去2,4-D,则有利胚性细胞团产生绿芽。  相似文献

在毛主席的革命科技路线指引下,我国从1975年首次成功地从玉米离体培养的花药诱导出单倍体植株以来,许多单位培养玉米花粉植株都已成功,并报道了从离体培养的花药中有成熟胚状体的产生。我们在1976年玉米花药培养的实验中也见到有较多胚状体的产生,并从胚状体形成绿苗有较高的频率。这是一个值得注意的问题。 我们的实验材料是大田春播的金黄113(墩子黄×W591)及金黄,号(太181×黄204)两个杂交种。诱导培养基为N_6,附加不同配比的激素(表1),蔗糖12%。分化培养基为N_6 K 1ppm BA 1ppm IAA 0.1ppm,蔗糖6%。  相似文献

以10个黄瓜品种为材料,取成熟胚外植体先经MS 2,4—D 1.0mg/L(单位下同) KT0.5诱导后转入MS IAA 0.1 KT0.5上分化,胚状体发生频率高并有植株再生。材料的基因型差异显著,“农大春光”分化率最高(40%)。双层培养有效地促进畸形胚状体的正常发育,5%蔗糖和1.0mg/L ABA促进胚性愈伤组织的继代增殖和胚性保持。  相似文献

Seven proteins each contain 8 to 52 tandem repeats of a unique class of oligopeptide. The consensus peptide for each is rhodopsin Tyr Pro Pro Gln Gly synaptophysin Tyr Gly Pro Gln Gly synexin Tyr Pro Pro Pro Pro Gly gliadin Tyr Pro Pro Pro Gln Pro RNA polymerase II Tyr Ser Pro Thr Ser Pro Ser hordein Phe Pro Gln Gln Pro Gln Gln Pro gluten Tyr Pro Thr Ser Pro Gln Gln Gly Tyr Although there is obvious variation of sequence and of length, the penta- to nonapeptides share an initial Tyr (or Phe) and have high Pro contents and abundant Gly, Gln, and Ser. We have evaluated helical models that both recognize the uniqueness of these sequence repeats and accommodate variations on the basic theme. We have developed a group of related helical models for these proteins with about three oligopeptide repeats per turn of 10-20 A. These models share several common features: Most of the phi dihedral angles are -54 degrees, to accommodate Pro at all positions except the first (Tyr). Except for the beta-turns, most psi dihedral angles are near +140 degrees as found in polyproline. Each oligopeptide has at least one beta-turn; several have two. Some contain a cis-Tyr, Pro peptide bond; a few have a cis-bond plus one beta-turn. Tyr side chains vary from totally exposed to buried within the helices and could move to accommodate either external hydrophobic interactions or phosphorylation. The several related structures seem to be readily interconverted without major change in the overall helical parameters, and therein may lie the key to their functions.  相似文献

动态测定烧伤患者血浆及红细胞内游离氨基酸的含量 ,探讨输入外源性氨基酸后对血及红细胞内游离氨基酸的影响。以日立 835— 5 0型氨基酸自动分析仪测定烧伤患者血浆及红细胞内游离氨基酸含量。结果发现烧伤患者血浆总游离氨基酸浓度从伤后到 2 1天均显著降低 (P <0 .0 5~ 0 .0 1) ;赖、苯丙和苯丙 酪氨酸比值显著升高 (P <0 .0 5~ 0 .0 1) ;色、组、精、丙、甘、苏、脯和丝氨酸比值显著降低 (P <0 .0 5~ 0 .0 1) ;缬、亮、异亮、酪、胱和支链氨基酸伤后早期降低。烧伤患者红细胞内总游离氨基酸含量不同程度降低 ,其中 1、3、7天降低显著 (P <0 .0 5~ 0 .0 1) ;红细胞内苯丙和苯丙 酪氨酸比值未见显著性升高 ;色、蛋、精、脯氨酸含量很低或基本未测出。输注复合氨基酸注射液后未能显著改善患者血及红细胞内游离氨基酸含量。结果提示烧伤患者红细胞内游离氨基酸含量的变化趋势与血浆游离氨基酸变化趋势基本一致 ;烧伤后红细胞内苯丙氨酸及苯丙 酪氨酸比值有别于血浆变化。本研究条件下补充外源性氨基酸未能显著改变烧伤患者血浆及红细胞内游离氨基酸的含量  相似文献

The action of 1 mM solutions of L-amino acids in 5 mM phosphate buffer, pH 7.4, on the production of hydrogen peroxide and hydroxyl radicals under the action of X-rays and heating has been studied. Hydrogen peroxide was estimated by the method of enhanced luminescence in a system luminol-paraiodophenol-peroxidase and hydroxyl radicals were determined by using the fluorescence probe coumarin-3-carboxylic acid. It was shown that amino acids can be divided by their influence on H202 formation into three groups: those that reduce the yield of H202, that do not influence it, and that increase it. A similar action of amino acids was observed upon heating, but the composition of the groups was different. All amino acids lowered the formation of hydroxyl radicals under the action of X-rays, and the most effective among them were Cys > His > Phe = Met = Trp > Tyr. Met, His and Phe lowered the amount of hydroxyl radicals by heating, Ser raised it, whereas Tyr and Pro did not change it. Thus, amino acids differently influence the formation of reactive oxygen species by the action of X-rays and heat, and some of amino acids reveal themselves as effective natural antioxidants.  相似文献

The complete amino acid sequence of the β-subunit of protocatechuate 3,4-dioxygenase was determined. The β-subunit contained four methionine residues. Thus, five peptides were obtained after cleavage of the carboxymethylated β-subunit with cyanogen bromide, and were isolated on Sephadex G-75 column chromatography. The amino acid sequences of the cyanogen bromide peptides were established by characterization of the peptides obtained after digestion with trypsin, chymotrypsin, thermolysin, or Staphylococcus aureus protease. The major sequencing techniques used were automated and manual Edman degradations. The five cyanogen bromide peptides were aligned by means of the amino acid sequences of the peptides containing methionine purified from the tryptic hydrolysate of the carboxymethylated β-subunit. Specific combinations of amino acids or purine ribonucleosides and amino acids are required for efficient germination of endospores of Bacillus anthracis DeltaSterne, a plasmidless strain, at ligand concentrations in the low-micromolar range. The amino acid L-alanine was the only independent germinant in B. anthracis and then only at concentrations of >10 mM. Inosine and L-alanine both play major roles as cogerminants with several other amino acids acting as efficient cogerminants (His, Pro, Trp, and Tyr combining with L-alanine and Ala, Cys, His, Met, Phe, Pro, Ser, Trp, Tyr, and Val combining with inosine). An ortholog to the B. subtilis tricistronic germination receptor operon gerA was located on the B. anthracis chromosome and named gerS. Disruption of gerS completely eliminated the ability of B. anthracis endospores to respond to amino-acid and inosine-dependent germination responses. The gerS mutation also produced a significant microlag in the aromatic-amino-acid-enhanced-alanine germination pathways. The gerS disruption appeared to specifically affect use of aromatic chemicals as cogerminants with alanine and inosine. We conclude that efficient germination of B. anthracis endospores requires multipartite signals and that gerS-encoded proteins act as an aromatic-responsive germination receptor.  相似文献

Twenty tryptic peptides were isolated from the performic acid-oxidized He chain of ricin D by Dowex 1 × 2 column chromatography followed by paper chromatography. The amino acids contained in these peptides accounted for 218 out of 266 residues in the whole protein. The amino acid sequences of nine peptides were determined by manual liquid phase or automatic solid phase Edman degradation, and N- and C-terminal sequences of the He chain of ricin D were established to be NH2–Ile–Phe–Pro–Lys–Gln–Tyr–Pro–Ile–Ile– and Cys–Ala–Pro–Pro–Pro–Ser–Ser–Gln–Phe, respectively.  相似文献

以药用植物宁夏枸杞愈伤组织为材料,离体培养诱导体细胞胚发生。采用多重示踪剂和γ射线能谱分析法研究不同浓度AgNO_3处理的枸杞体细胞胚发生过程中对多种痕量金属元素离子的吸收。结果表明:(1)当AgNO_3的浓度小于50mg/L时,随着AgNO_3浓度的增加,多种痕量金属离子的吸收率也随之增加,而超过此浓度后,对多种痕量金属离子的吸收影响不同。Ag~ 对痕量金属离子的吸收有协同,拮抗或竞争的作用。(2)适当浓度的AgNO_3对细胞分化及体细胞胚发生有促进作用。当AgNO_3的浓度小于50mg/L时,随着AgNO_3浓度的增加,体细胞胚的发生频率随之增加。Ag~ 对枸杞体细胞胚发生表现促进作用,当AgNO_3的浓度为50mg/L时,可大大提高愈伤组织中体细胞胚发生,是对照(不加AgNO_3)组的3倍左右。而超过此浓度后,Ag~ 对枸杞体细胞胚发生表现毒害作用,体细胞胚的发生受到明显抑制。  相似文献

The sequence of 96 amino acid residues from the COOH-terminus of the active subunit of cholera toxin, A1, has been determined as PheAsnValAsnAspVal LeuGlyAlaTyrAlaProHisProAsxGluGlu GluValSerAlaLeuGlyGly IleProTyrSerGluIleTyrGlyTrpTyrArg ValHisPheGlyValLeuAsp GluGluLeuHisArgGlyTyrArgAspArgTyr TyrSerAsnLeuAspIleAla ProAlaAlaAspGlyTyrGlyLeuAlaGlyPhe ProProGluHisArgAlaTrp ArgGluGluProTrpIleHisHisAlaPro ProGlyCysGlyAsnAlaProArg(OH). This is the largest fragment obtained by BrCN cleavage of the subunit A1 (Mr 23,000), and has previously been indicated to contain the active site for the adenylate cyclase-stimulating activity. Unequivocal identification of the COOH-terminal structure was achieved by separation and analysis of the terminal peptide after the specific chemical cleavage at the only cysteine residue in A1 polypeptide. The site of self ADP-ribosylation in the A1 subunit [C. Y. Lai, Q.-C. Xia, and P. T. Salotra (1983) Biochem. Biophys. Res. Commun.116, 341–348] has now been identified as Arg-50 of this peptide, 46 residues removed from the COOH-terminus. The cysteine that forms disulfide bridge to A2 subunit in the holotoxin is at position 91.  相似文献

小麦品种崇阳红麦和鄂思一号杂种一代幼胚培养具有再生植株的潜力。从一个幼胚经200天左右的连续培养获得530多株再生植株,并从中获得了典型的具有两极性的与愈伤组织块仅局部相连的胚状体。体细胞胚胎发生是小麦幼胚培养的主要途径,但受培养条件的影响,以MS培养基作基本培养基,低浓度2,4-D(0.4mg/1)和水解酪蛋白(1000mg/l)有利于体细胞胚胎发生。  相似文献
