An improved method for the isolation of chloroplasts from Poteriochromonasmalhamensis is described. Poteriochromonas cells were brokenby passage through a nylon mesh with pores of 6µ in diameterat a flow rate of about 5 ml/15 s. After centrifugation thecrude chloroplast fraction was purified by centrifugation ina step gradient of Percoll. The isolated chloroplasts were enclosedby envelope membranes and were still surrounded in part by cytoplasmicresidues. The chloroplasts had the capacity for translation,which was both chloramphenicol-sensitive and cycloheximide-insensitive.The properties of these isolated chloroplasts from Poteriochromonasare discussed in relation to experiments on the transport intothe chloroplasts of nucleus-encoded proteins. 2 Present address: Bundesgesundheitsamt, Zulassungsstelle furGentechnologie, Columbiadamm 3, D-1000 Berlin, F.R.G. The thylakoid membranes of isolated Euglena chloroplasts wereseparated into two fractions by aqueous two-phase-partitioning(mixture of dextran 500 and poly(ethylene glycol) 4000) followingpress disruption. These two fractions differ in many respectsduring most of the cell cycle of this alga in comparison withthe thylakoid characteristics of higher plants or green algae.The amount of thylakoid membranes with separation characteristicscomparable with inside-out-vesicles of higher plant chloroplastschanges depending on the cell cycle stage of Euglena gracilis.Photosystems II and I are not restricted to one fraction. Boththylakoid membrane fractions evolve oxygen photosynthetically.When chloroplast differentiation in Euglena gracilis is complete(i.e. at the end of the light-time) the composition and thephotosynthetic efficiency of the two thylakoid fractions aregenerally equal. Photosystem I-related LHCI is present in bothfractions. Photosystem II-related CP29, however, was only detectedin unfractionated thylakoid membranes. During the ageing and senescence of leaves of the birch tree,Betula verrucosa Ehrb., from the fully expanded green stateto the yellow terminal condition, the shape of the chloroplastsalters and their volume decreases to less than one-fifth. Associatedwith these changes is an initial increase in the number of chloroplastlamellae and size of the grana but this is followed by the breakdownof the membrane systems. The chloroplast lipid globules increaseconsiderably in size but show little increase in number. Thestructure of the chloroplast membranes is described in detail.Small particles approximately 8 x 5 nm, become visible on theinner surfaces of the thylakoid membranes during chloroplastbreakdown. The chloroplasts are the first organelles to showsigns of senescence and are the last to remain after the otherorganelles have all broken down. The observations are discussedin relation to the senescence of chloroplasts of herbaceousplants and the development of chromoplasts of fruits.  相似文献

In C4 plants, bundle sheath (BS) chloroplasts are arranged inthe centripetal position or in the centrifugal position, althoughmesophyll (M) chloroplasts are evenly distributed along cellmembranes. To examine the molecular mechanism for the intracellulardisposition of these chloroplasts, we observed the distributionof actin filaments in BS and M cells of the C4 plants fingermillet (Eleusine coracana) and maize (Zea mays) using immunofluorescence.Fine actin filaments encircled chloroplasts in both cell types,and an actin network was observed adjacent to plasma membranes.The intracellular disposition of both chloroplasts in fingermillet was disrupted by centrifugal force but recovered within2 h in the dark. Actin filaments remained associated with chloroplastsduring recovery. We also examined the effects of inhibitorson the rearrangement of chloroplasts. Inhibitors of actin polymerization,myosin-based activities and cytosolic protein synthesis blockedmigration of chloroplasts. In contrast, a microtubule-depolymerizingdrug had no effect. These results show that C4 plants possessa mechanism for keeping chloroplasts in the home position whichis dependent on the actomyosin system and cytosolic proteinsynthesis but not tubulin or light.  相似文献

Leaf alcohol (cis-3-hexenol) and leaf aldehyde (trans-2-hexenal)are responsible for the green odor in leaves and fruits. cis-3-Hexenal,a precursor of cis-3-hexenol and trans-2-hexenal, was producedfrom linolenic acid by a homogenate of Farfugium japonicum (Japanesesilver) leaves. n-Hexanal was produced from linoleic acid bya homogenate of the leaves. The enzyme system catalyzing formationof C6-aldehydes from linolenic and linoleic acids was localizedin chloroplast lamellae, and required oxygen for reaction. C18-unsaturatedfatty acids such as linolenic acid, linoleic acid and -linolenicacid, which have carboxyl groups and cis-1, cis-4-pentadienesystems including a double bond at C-12, acted as substrates,and C6-aldehydes (cis-3-hexenal or n-hexanal), but not C9-aldehydes,were produced from them. The properties of the enzyme systemin chloroplasts were as follows: optimal pH 7.0; stable at pH5 to 7; thermolabile and no activity at 50?C. These propertieswere very similar to those of tea chloroplasts. The enzyme systemcould be solubilized from chloroplasts by 2% Triton X-100, butwas very unstable in solubilized form. Twelve cDNAs corresponding to mRNAs inducible by ethylene were isolated by differential screening of a cDNA library from ethylene-treated Citrus sinensis fruits. Northern analysis of RNA extracted from flavedo of ethylene-treated fruits and from fruits at different maturation stages showed that some of the mRNAs corresponding to these cDNAs were regulated both by ethylene treatment and during fruit maturation. The effect of exogenous ethylene on leaves and of endogenous ethylene on flowers showed that gene induction was not restricted to the flavedo tissue. The possible role of ethylene during maturation of the non-climacteric Citrus fruit is discussed.  相似文献

The presence of nucleus, chloroplasts, mitochondria, microbodiesand a vacuole are confirmed in Cyanidium caldarium strain CCAP1355/1. Chloroplasts usually contain parallel rows of unstackedthylakoids surrounded by a peripheral thylakoid, although theconcentric arrangement has also been observed. The chloroplastenvelope consists of two closely appressed membranes which canonly be resolved after gluteraldehyde—osmium fixation.The chloroplast of Rhodosorus marinus also contains parallel,unstacked thylakoids surrounded by a peripheral thylakoid but,in addition, a prominent, stalked pyrenoid projects into thecytoplasm and is bounded by both the peripheral thylakoid andthe chloroplast envelope. This structure is covered by a capof starch grains and contains membranous vesicles in the matrix.Phycobilisomes are absent from the chloroplasts of both algae.The recognition of C. caldarium as a rhodophyte is supportedby these observations. The hyphae of Lophodermella sulcigena are usually surroundedby an electron-opaque matrix when growing in the needles ofCorsican pine (Pinus nigra var. maritima). The host-parasiteinterface is of a type which has been little studied; the intercellularhyphae kill the mesophyll cells of the host ahead of themselvesand their walls are separated from those of the host by a matrix.The first signs of injury to the host are disorganized membranesystems, particularly the thylakoid and bounding membranes ofthe chloroplasts break down. Changes in fine structure of fruit tissues were examined byelectron microscopy for the following host-parasite relationships:Sclerotinia fructigena on apple and pear, S. sclerotiorum oncucumber, and Phytophthora palmivora on egg plant. While S.fructigena caused localized degradation of the wall region,S. sclerotiorum caused extensive wall degradation, and infectionby P. palmivora was characterized by disintegration of plasmalemmaand tonoplast. These effects appeared to be correlated respectivelywith pectolytic enzyme activity in vivo with S. fructigenaycellulolytic activity with S. sclerotiorum, and the in vitrolysis of membranes by culture filtrates of P. palmivora.  相似文献

Saplings of Azadirachta indica Juss. were exposed to sulphurdioxide (SO2) and some of the exposed saplings were treatedwith ascorbic acid (AA). The SO2 exposure alone inflicted heavydamage to the chloroplasts and cytoplasm in palisade cells.The degeneration of chloroplasts was followed by the rupturingof the outer envelope and the extrusion of plastoglobuli andstarch into the cytoplasm. Chloroplasts were isolated from the diatom Phaeodactylum tricornutumby French press treatment and centrifugation. Electron micrographsof the isolated chloroplasts indicated that they lacked mostof the envelope membranes but retained the lamellar structurecharacteristic of the diatom chloroplast; three thylakoids weregrouped to form a band which transversed the chloroplast. Agirdle lamella also composed of three thylakoids surroundedthese transversal lamellae. Studies on the histology and on effects of growth substancesand phenols as well as changes in activities of pectinmethylesterase indicated that the mechanism of abscission of Hevealeaflets infected with Microcyclus ulei differed from the mechanismof abscission of debladed, ethylene treated and senescent leaves.An abscission layer which was formed during abscission of debladed,ethylene-treated and senescent leaves was absent during abscissionof heavily diseased leaves. The ratio of pectinmethyl esteraseactivities in tissues distal to the abscission zone to activitiesin tissues proximal to the zone decreased in debladed and ethylenetreated leaves but such decreases were not detected during abscissionof Hevea leaves infected with M. ulei. Farnesol accumulates in water-stressed plants of Sorghum, andhas been shown to induce closure of stomata in the Commelinabioassay. In the present study we have found that it inhibitsbicarbonate- and PGA-stimulated O2 evolution of isolated Pisumchloroplasts. Farnesol apparently disrupts the membranes ofisolated plastids and most of the effects that we have observedcan probably be attributed to this. Apart from nerolidol, relatedisoprenoid alcohols had no comparable effects, but the fattyacid linoleic acid appeared to cause damage of a similar nature.These compounds all caused stomata of Commelina to close. It is suggested that, while farnesol causes irreversible damageto membranes when applied exogenously in in vitro systems, itmight act in vivo as a regulator of membrane structure and soaffect the solute movements on which the turgor changes of guardcells depend.  相似文献

The photoreduction of dioxygen in spinach thylakoid membraneswas enhanced about 10-fold by the FAD-enzyme monodehydroascorbateradical (MDA) reductase at 1 µM. The primary photoreducedproduct of dioxygen catalyzed by MDA reductase was the superoxideradical, as evidenced by the inhibition of photoreduction ofCyt c by superoxide dismutase. The apparent Km for dioxygenof the MDA reductase-dependent photoreduction of dioxygen was100 µM, higher by one order of magnitude than that observedwith thylakoid membranes only. Glutathione reductase, ferredoxin-NADP+reductase, and glycolate oxidase also mediated the photoproductionof superoxide radicals in thylakoid membranes at rates similarto those with MDA reductase. Among these flavoenzymes, MDA reductaseis the most likely mediator stimulating the photoreduction ofdioxygen in chloroplasts; its function in the protection fromphotoinhibition under excess light is discussed. Outer and inner envelope membranes of spinach chloroplasts wereisolated using floatation centrifugation followed by sedimentationsucrose density gradient centrifugation after disruption ofintact chloroplasts by freezing and thawing. Two major fractionswith buoyant densities of 1.11 and 1.08 g cm–3 and a minorfraction with a density of 1.15 g cm–3 were obtained.They were identified as innei and outer envelope and thylakoidfractions, respectively, by analyzing their polypeptide compositionby high-resolution SDS-PAGE and the N-terminal sequences oftheir protein components. Due to the refinement of the isolation procedure, most of theribulose-l,5-bisphosphate carboxylase/oxygenasc (RuBisCO), whichhad always been observed as a contaminant, was eliminated fromthe outer envelope fraction. Application of high-resolutionSDS-PAGE revealed that this fraction was rich in the low-molecular-massouter envelope protein, E6.7 [Salomon et at. (1990) Proc. Natl.Acad. Sci. USA 87: 5778] and a protein with a molecular massof 15 kDa which is homologous to the 16 kDa outer envelope proteinof pea [Pohlmeyer et al. (1997) Proc. Natl. Acad. Sci. USA 94:9504]. The two proteins account for 90% of the total proteinspresent in outer envelope membranes. Proteins which are suggestedto function in translocation of nuclear-encoded polypeptideswere not identified in the envelopes from spinach in the presentstudy. The induction curve of delayed luminescence emitted from 0.5to 2.5 ms after excitation of dark-adapted intact chloroplastsof the green alga, Bryopsis maxima, showed three transient peaks,L1, L2 and L3 (in order of appearance), at about 0.1, 1 and5 s after theonset of intermittent illumination. Intact chloroplastswere needed for L2 to appear, whereas L1 and L3 were presentin hypotonically treated chloroplasts. L1 and L2 are related to the electric field generated acrossthe thylakoid membranesbecause the two peaks parallelled theappearance of the first and second peaks of electrochromic absorptionchanges at 560 nm and they were totally abolished by valinomycinand CCCP. A smaller contribution to the L1 and L2 of the protonactivity gradient across the membranes, or of pH changes insideor outside the membranes, was suggested by the partial suppressionof the transient by NH4CI. L3 is related to the proton gradient or pH changes because thetransient was inhibited by NH4CI and CCCP but enhanced by N,N'-dicyclohexylcarbodiimide.In the presenceof valinomycin, which somewhat lowered the peakheight of L3, the kinetics of delayed luminescence parallelledthat of fluorescence. Electrogenic reactions which occur sequentiallyduring the dark to light transition of the photosynthetic machineryin intact chloroplasts is discussed in connection with transientchanges in delayed luminescence. The chloroplasts in dark-grown, 5-day-old seedlings of Piceaabies contained doubly stacked primary thylakoid membranes connectedwith prolamellar bodies. The photo-system II reaction centerand photosystem II-associated electron transfer were assembledin the primary thylakoid membranes, but the oxygen-evolvingsystem remained latent unless the seedlings were exposed tolight. The photoactivation of this system was strongly dependenton temperature during pre-illumination. The amount of total chlorophyll, chlorophylls a and b as wellas the ratio of a to b decreased in chloroplasts isolated fromQuercus rotundifolia leaves, kept for 17 d in a solution of35.5 µM evernic acid in 1 mM Na HCO3, when compared withthe chloroplasts of control leaves (kept in NaHCO3). The chloroplastsin the spongy parenchyma were smaller and the amount of starchand plastoglobuli lower. The number of grana per chloroplastsection, the number of thylakoids per grana and the height ofgrana stacks were also less in the chloroplasts of leaves treatedwith evernic acid. Factors concerning the chloroplast disposition in bundle sheathcells were investigated in finger millet (Eleusine coracanaGaertn.), and NAD malic enzyme type C4 plant with the centripetalarrangement of bundle sheath chloroplasts. Segments were cutfrom immature regions of emerging leaves in which the centripetalarrangement of bundle sheath chloroplasts had not yet been established.The leaf segments were floated on solutions with or withoutreagents. Sections were made of the segments at time intervalsand the distribution of bundle sheath chloroplasts was observedby light microscopy. The bundle sheath chloroplasts migratedto the vascular bundle and established a centripetal arrangementby 12-16 h in control solutions. Auxins, cycloheximide and cytochalasinB inhibited the disposition of bundle sheath chloroplasts whilechloramphenicol and colchicine had no effect. The inhibitoryeffect of auxins appeared only at early stages of chloroplastmigration while cycloheximide and cytochalasin B were effectiveeven at later stages. The division of chloroplasts in a green alga, Pediastrum duplex,was studied by electron microscopy. Cells were treated for observationwith the freeze-substitution method. Fibrils, or fibrous belts,which we had observed previously at the dividing constrictionsof chloroplasts in Trebouxia potteri were not visible in Pediastrum,even though the method of preparation was the same for bothsets of samples. Microtubules (MTs) and the septum seem notto participate directly in the division of the single chloroplastin Pediastrum cells. Many thin fibrils, 7–20 nm in diameter,attached to, or protruding from, the surface of the dividingconstriction were seen. These fibres were less densely distributedat the constrictions of non-dividing chloroplasts. It is suggestedthat these fibrils are involved in the divison of chloroplastsin Pediastrum duplex.
