The tribe Bovini comprises cattle and cattle-like species. Reconstructions of their phylogeny have so far been incomplete and have yielded conflicting conclusions about the relationship of American bison and wisent (European bison). We have compared the sequences of three mitochondrial and two Y-chromosomal DNA segments. Mitochondrial DNA indicates that four distinct maternal lineages diverged after an early split-off of the buffalo species, leading to (1) taurine cattle and zebu, (2) wisent, (3) American bison and yak, and (4) banteng, gaur, and gayal, respectively. At a higher level, lineages (1) and (2) and lineages (3) and (4) are probably associated. In contrast, Y-chromosomal sequences indicate a close association of American and European bison, which is in agreement with their morphological similarity, complete fertility of hybrid offspring, and amplified fragment length polymorphism (AFLP) fingerprints of nuclear DNA. One explanation for the anomalous divergence of the mitochondrial DNA from the two bison species is lineage sorting, which implies that two distinct mitochondrial lineages coexisted in the bison-yak branch until the recent divergence of American bison and wisent. Alternatively, the wisent may have emerged by species hybridization initiated by introgression of bison bulls in another ancestral species. This "transpatric" mode of species formation would be consistent with the recent appearance of the wisent in the fossil record without clearly identifiable ancestors.  相似文献

Between April 2008 and March 2009, we analyzed the pattern of coccidian oocysts present in the feces of the European bison (Bison bonasus L., 1758) and found 4 species (Eimeria bovis , E. canadensis, E. ellipsoidalis, E. zuernii) previously reported from this host and 3 species (Eimeria alabamensis, E. cylindrica, E. pellita) that are new host and locality records. All the species occurred in bison females, and only 4 occurred in males; E. bovis was the most prevalent in both sexes. The overall prevalence of Eimeria spp. invasion reached 34.7% in cows and 13.9% in bulls. The highest prevalence was noted in early spring, with a peak in April, and the lowest in late autumn and winter. The oocyst count per gram of feces (OPG) varied from 50 to 1,350; no symptoms of clinical coccidiosis were observed. We found a significant influence of winter aggregations of bison on shedding of coccidian oocysts. The prevalence and OPG values were higher in bison congregating in large numbers around winter-feeding sites in comparison to other sites. We suggest that the coming together of cows during the growing season impacts the gender-related differences in prevalence and the number of coccidian species involved. This observation probably results from an increased production of oocysts by sub-clinically infected individuals in high-density bison populations.  相似文献

牦牛的起源与属级分类学地位至今仍然存在一定的争议。我们测定了家养牦牛和野生牦牛线粒体控制区(D-loop)序列,并以此构建牦牛和牛属、野牛属、水牛属以及非洲水牛属相关种的系统发育树。研究结果表明线粒体D-loop区与Cytb基因序列在构建牛族的系统发育具有同样重要的价值。系统发育关系显示野牛属的灭绝种草原野牛与现存种美洲野牛先聚合为一单系群,然后再和牦牛形成一单系分支,表明牦牛与野牛属的草原野牛、美洲野牛亲缘关系最近,具有最近的共同祖先,而与牛属的其它亚洲物种亲缘关系较远。因此,本研究不支持将牦牛独立为牦牛属———Poephagus,牛属与野牛属在分类上也应合并为一个属。基于上述研究结果和化石证据,我们进一步对牦牛起源的历史背景进行了讨论,认为牦牛与野牛属的分化是由于第四纪气候变化在欧亚大陆发生的,野牛通过白令陆桥进入北美;冰期结束后,由于欧亚大陆其它地区温度升高,牦牛只能局限分布在较为寒冷的青藏高原;而野牛属在北美先后分化为草原野牛和美洲野牛,前者可能是后者的直接祖先。  相似文献

A free-living population of European bison Bison bonasus in the Białowieża Primeval Forest originated from only seven founder animals after a severe bottleneck that occurred at the beginning of the 20th century. Consequently, the contemporary population of the species is characterized by low genetic diversity. We studied a total of 195 individuals (127 males and 68 females). A 1429 bp fragment of mitochondrial DNA (mtDNA) including the D-loop region was analyzed in 87 individuals and revealed only three distinct haplotypes. Nucleotide ( π ) and haplotype ( H d) diversity values were estimated for the European bison and were compared with π and H d estimated from three individuals of American bison Bison bison . Very low diversity values were found in the European bison in comparison with the diversity values found in the American bison. The low mtDNA variability in the European bison is in concordance with theoretical expectations for a species that has undergone a severe and recent bottleneck. A management strategy for the preservation of the rare and very rare haplotypes present in the Białowieża population of the European bison is discussed. Furthermore, all 195 individuals were investigated for heteroplasmy involving these three haplotypes, in order to detect a possible association between heteroplasmy and the incidence of males affected by posthitis , a disease that affects the male reproductive organs, leading to necrotic lesions. Heteroplasmy was found in 15 females, in 17 males affected by posthitis and in 11 non-affected males, and no significant association was found.  相似文献

The genetic integrity of crossfertile bovine- or cattle-like species may be endangered by species hybridization. Previously, amplified fragment length polymorphism, satellite fragment length polymorphism and microsatellite assays have been used to analyze the species composition of nuclear DNA in taurine cattle, zebu, banteng and bison populations, while mitochondrial DNA reveals the origin of the maternal lineages. Here, we describe species-specific markers of the paternally transmitted Y-chromosome for the direct detection of male-mediated introgression. Convenient PCR-restriction fragment length polymorphism and competitive PCR assays are shown to differentiate the Y-chromosomes of taurine cattle, American bison and European bison, and to detect the banteng origin of Indonesian Madura and Bali cattle bulls.  相似文献

Despite moose (Alces americanus) likely being both an ecological and cultural keystone species in the North American boreal forest, few studies have examined niche overlap between moose and other ungulates. In response to concerns from local people about the potential for competition for habitat between moose and reintroduced bison (Bison bison), and the potential for disturbance to moose by bison and bison hunters, we investigated overlap in winter habitat selection by these species in southwestern Yukon, Canada. We used available geo-referenced data of animal locations, and associated environmental covariates, to develop resource selection probability function (RSPF) models of early-winter and late-winter habitat selection by moose and bison. In early-winter, moose selected sub-alpine shrub-dominated habitats, whereas bison extensively selected wet sedge meadows in lowland valleys. Both species used a greater variety of habitats in late-winter, with moose selecting drainages above tree line or river valleys with deciduous forest cover, while bison selected either south-facing grassy slopes or wet sedge meadows. Given observed differences in seasonal habitat selection between moose and bison, these species are predicted to overlap on only 0.5 and 6.6% of our 12,818-km2 study area during early-winter and late-winter, respectively. The lack of demonstrated winter habitat overlap between moose and reintroduced bison, coupled with low diet overlap, points to an overall low potential for competition for winter habitat between these species, or for disturbance to moose attributable to bison or bison hunters. Resource partitioning that facilitates coexistence on a shared landscape suggests that concern over the impact of reintroduced bison on resident populations of moose is likely unwarranted.  相似文献

Introgressive hybridization is one of the major threats to species conservation, and is often induced by human influence on the natural habitat of wildlife species. The ability to accurately identify introgression is critical to understanding its importance in evolution and effective conservation management of species. Hybridization between North American bison (Bison bison) and domestic cattle (Bos taurus) as a result of human activities has been recorded for over 100 years, and domestic cattle mitochondrial DNA was previously detected in bison populations. In this study, linked microsatellite markers were used to identify domestic cattle chromosomal segments in 14 genomic regions from 14 bison populations. Cattle nuclear introgression was identified in five populations, with an average frequency per population ranging from 0.56% to 1.80%. This study represents the first use of linked molecular markers to examine introgression between mammalian species and the first demonstration of domestic cattle nuclear introgression in bison. To date, six public bison populations have been identified with no evidence of mitochondrial or nuclear domestic cattle introgression, providing information critical to the future management of bison genetic resources. The ability to identify even low levels of introgression resulting from historic hybridization events suggests that the use of linked molecular markers to identify introgression is a significant development in the study of introgressive hybridization across a broad range of taxa.  相似文献

Grazing by large ungulates often increases plant species richness in grasslands of moderate to high productivity. In a mesic North American grassland with and without the presence of bison (Bos bison), a native ungulate grazer, three non-exclusive hypotheses for increased plant species richness in grazed grasslands were evaluated: (1) bison grazing enhances levels of resource (light and N) availability, enabling species that depend on higher resource availability to co-occur; (2) spatial heterogeneity in resource availability is enhanced by bison, enabling coexistence of a greater number of plant species; (3) increased species turnover (i.e. increased species colonization and establishment) in grazed grassland is associated with enhanced plant species richness. We measured availability and spatial heterogeneity in light, water and N, and calculated species turnover from long-term data in grazed and ungrazed sites in a North American tallgrass prairie. Both regression and path analyses were performed to evaluate the potential of the three hypothesized mechanisms to explain observed patterns of plant species richness under field conditions. Experimental grazing by bison increased plant species richness by 25% over an 8-year period. Neither heterogeneity nor absolute levels of soil water or available N were related to patterns of species richness in grazed and ungrazed sites. However, high spatial heterogeneity in light and higher rates of species turnover were both strongly related to increases in plant species richness in grazed areas. This suggests that creation of a mosaic of patches with high and low biomass (the primary determinant of light availability in mesic grasslands) and promotion of a dynamic species pool are the most important mechanisms by which grazers affect species richness in high productivity grasslands.  相似文献

Understanding and documenting the process of hybridization and introgression between related species is a major focus of recent evolutionary research using molecular techniques. Many North American bison herds have cattle ancestry introduced by crossbreeding over a century ago. Molecular estimates of this ancestry have shown much higher levels for cattle mtDNA than for autosomal cattle genes. A large part of this difference appears to be the result of partial reproductive isolation between the two species where only bison bull × domestic cow crosses are successful, and all the surviving progeny are females. In addition, selection against autosomal cattle genes in bison may have contributed to differential levels of cattle ancestry. The impact of selection against cattle mtDNA and gene flow of bison mtDNA are examined to explain particular combinations of mtDNA and autosomal cattle ancestry. A bottleneck, after the level of cattle ancestry in bison was reduced to a low level, is consistent with the high variance over autosomal loci observed for cattle ancestry, and differential selection among cattle loci in bison does not need to be invoked. Further examination of the cattle genome in bison may shed light on whether these markers, or their associated regions, are indeed neutral.  相似文献

North American bison (Bison bison) are becoming increasingly important to both grassland management and commercial ranching. However, a lack of quantitative data on their diet constrains conservation efforts and the ability to predict bison effects on grasslands. In particular, we know little about the seasonality of the bison diet, the degree to which bison supplement their diet with eudicots, and how changes in diet influence gut microbial communities, all of which play important roles in ungulate performance. To address these knowledge gaps, we quantified seasonal patterns in bison diet and gut microbial community composition for a bison herd in Kansas using DNA sequencing-based analyses of both chloroplast and microbial DNA contained in fecal matter. Across the 11 sampling dates that spanned 166 days, we found that diet shifted continuously over the growing season, allowing bison to take advantage of the seasonal availability of high-protein plant species. Bison consumed more woody shrubs in spring and fall than in summer, when forb and grass intake predominated. In examining gut microbiota, the bacterial phylum Tenericutes shifted significantly in relative abundance over the growing season. This work suggests that North American bison can continuously adjust their diet with a high reliance on non-grasses throughout the year. In addition, we find evidence for seasonal patterns in gut community composition that are likely driven by the observed dietary changes.  相似文献

The American Bison (Bison bison Linnaeus) in the Henry Mountains are one of the last free-roaming, genetically pure herds of bison remaining in North America. Anecdotal evidence indicates that this herd is utilising a cattle winter range during the summer and fall, creating a conflict between the state agency that manages the bison, and the Bureau of Land Management (BLM) and local ranchers. In theory, the addition of bison grazing pressure could reduce forage availability in the short term and lead to undesired changes in the plant community in the long term. Our objective was to determine whether bison have altered the plant species composition of the cattle winter range. We characterised plant species composition, percent cover, and grazing intensity on three adjacent, geomorphologically similar mesas. Grazing regimes were different on the three mesas, one with bison and cattle present, one with cattle only present, and the third with neither cattle nor bison present. Vegetation surveys were accompanied by a 28-year remote sensing time series to test for temporal shifts in an index of primary productivity. We found a higher grazing intensity on two dominant forage species on the bison plus cattle grazed mesa in fall, before the cattle were turned out to winter pasture. Despite this difference in grazing intensity, we found few differences in species composition, percent cover, or NDVI across the three grazing regimes. Our results suggest that high intensity summer bison grazing, while likely creating short-term reductions in forage availability, has not caused differences in plant community composition or productive potential. Shifts in community composition can take years to unfold and just as long to correct; therefore, continued monitoring of the combined effects of cattle and bison is needed.  相似文献

Wild American plains bison (Bison bison) populations virtually disappeared in the late 1800s, with some remnant animals retained in what would become Yellowstone National Park and on private ranches. Some of these private bison were intentionally crossbred with cattle for commercial purposes. This forced hybridization resulted in both mitochondrial and nuclear introgression of cattle genes into some of the extant bison genome. As the private populations grew, excess animals, along with their history of cattle genetics, provided founders for newly established public bison populations. Of the US public bison herds, only those in Yellowstone and Wind Cave National Parks (YNP and WCNP) appear to be free of detectable levels of cattle introgression. However, a small free-ranging population (~350 animals) exists on public land, along with domestic cattle, in the Henry Mountains (HM) of southern Utah. This isolated bison herd originated from a founder group translocated from YNP in the 1940s. Using genetic samples from 129 individuals, we examined the genetic status of the HM population and found no evidence of mitochondrial or nuclear introgression of cattle genes. This new information confirms it is highly unlikely for free-living bison to crossbreed with cattle, and this disease-free HM bison herd is valuable for the long-term conservation of the species. This bison herd is a subpopulation of the YNP/WCNP/HM metapopulation, within which it can contribute significantly to national efforts to restore the American plains bison to more of its native range.  相似文献

This study provides endocrine data in relation to behavioral events during the transition of the non-breeding into the breeding season in American bison (Bison bison). Fecal progesterone metabolite patterns (20-oxo-P) were obtained in 13 adult female American bison and hormonal data were correlated with behavioral observations; i.e. copulation, male tending, female tail-up behavior and gestation length. Based on fecal progesterone metabolite patterns, the breeding season started between the middle of July and early August. Predictable short cycles reflected the transition from non-breeding to the breeding season; the luteal phase of these cycles was 4.10+/-0.86 days. Copulations and female tail-up behavior were reliably associated with the hormonally detected ovulation. Male tending behavior was more loosely associated with hormonally detected ovulation. The observed hormonal pattern in the study females indicated that 9 of 10 pregnant cows conceived during the second ovulatory period in the breeding season. One other cow conceived during her third ovulatory period, and one cow did not conceive until later in the breeding season by beginning of October. Gestation duration was on average 266.30+/-1.00 days. In summary, this study confirmed that the bison is a seasonally polyestrous species; the transition from the non-breeding into the breeding season was characterized by short cycles with low progesterone metabolite values.  相似文献

Ruminal contents from 79 slaughtered bison and 2 ruminally cannulated bison were collected to obtain information on total numbers and species distribution of ciliated protozoa. The bison originated from numerous herds throughout the Great Plains and were grouped into three dietary categories: (i) only forage; (ii) forage with moderate levels of supplementation; and (iii) feedlot concentrate-silage diet. Total ciliate counts were highest in bison receiving grain supplementation (210.1 x 10(4)/g) and lowest in bison consuming only forage (27.1 x 10(4)/g). All protozoan species found in bison have been reported in domestic livestock, although Ophryoscolex sp., a relatively common protozoan in cattle, was detected at low concentrations in only eight bison. The uncommon holotrich Microcetus lappus was present in five bison in concentrations reaching 8.4% of the total ciliate population. Charonina ventriculi, another infrequently observed species, was present in 18 bison, with the highest concentrations in forage-fed animals. Thirty bison possessed a type B protozoan population, characterized by Epidinium sp., Eudiplodinium maggii, and Eudiplodinium bovis. Thirty-eight bison possessed a mixed A-B population, characterized by Polyplastron sp. coexisting with low numbers of Eudiplodinium maggii or Epidinium sp. or both. Thirteen bison possessed populations lacking any remnant type B ciliate species. At least 29 of the bison possessing Polyplastron sp. were known to have been in contact with cattle, whereas all bison isolated from cattle had type B populations. The reduction of type B populations in bison becomes increasingly likely as bison production expands into areas inhabited by domestic livestock.  相似文献

Selected host defense functions of neutrophils isolated from American bison (Bison bison) were characterized and compared with those of cattle (Bos taurus). Bison neutrophils had a robust chemotactic response to both IL-8 and LTB(4), with maximal responses occurring at 10(-7) M (IL-8) and 10(-8) M (LTB(4)). The magnitude of the chemotactic response to IL-8 was similar in bison and bovine neutrophils (except at 10(-7) M IL-8, where bison had a stronger response). In response to LTB(4), bison neutrophils had a much stronger chemotaxis at both 10(-8) and 10(-7) M than did bovine cells. Production of reactive oxygen species (ROS) in response to phorbol myristate acetate (PMA) and opsonized zymosan (OpZ) was similar between bison and bovine neutrophils. However, the production of ROS in bison neutrophils stimulated with OpZ was primarily intracellular, while extracellular release of ROS was evident in bovine neutrophils stimulated with OpZ. Like bovine neutrophils, bison neutrophils did not generate a respiratory burst in response to fMLF. Granules prepared from bison neutrophils had potent direct killing action on the Gram-negative bacteria Escherichia coli but failed to kill the Gram-positive bacteria Staphylococcus aureus and, at intermediate doses, actually had a permissive effect for this bacteria. Thus, bison neutrophils have potent host defense capabilities similar in quality to those of bovine neutrophils; however, unique differences are present, which may allow bison neutrophils to respond to the distinct immunological challenges that bison encounter.  相似文献

Blood was collected at slaughter from 132 adult American bison (Bison bison) raised in an anaplasmosis endemic area where the vector Dermacentor andersoni (equals venustus) is indigenous. Hematologic studies revealed no indication of clinical anaplasmosis. Card agglutination and complement-fixation tests on all bison serums were negative. Eleven anaplasmosis-susceptible calves each inoculated with 204 ml of blood pooled from 12 bison did not develop anaplasmosis. Results of this study indicate American bison have resistance to natural A. marginale infection.  相似文献

Babesia bigemina was experimentally transmitted from cattle to bison and back to cattle. One spleen-intact and two splenectomized American bison (Bison bison) inoculated with a B. bigemina stabilate exhibited clinical and hematological signs of babesiosis within 10 days of exposure. Blood from the infected bison produced disease in a splenectomized bovine steer.  相似文献

One of the most informative sources which allow the drawing of far-reaching conclusions about the origins and phylogenetics of many species, including domestic animals and humans, is mitochondrial DNA (mtDNA). One of the important research targets should include the identification of similarities between wild and domestic species. The analysis involved the nucleotide sequences of mtDNA of wisent, auroch, bison, yak, bovine reference sequence (BRS) T3, T3a, T3b, T1, T1a, T1'2'3, T2, T3, T4, T5, Q, Q1, P, R, I1, and I2 bovine haplotypes. The non-coding D-loop regions were excluded from the evolutionary analysis and 15,419-bp coding sequences were used in the final dataset. Trees constructed on the basis of whole mitochondrial genomes or on total mtDNA coding sequences alignment were generally in agreement with previous studies on the Bovini tribe. American bison shows stronger maternal relationships to yak than to wisent. It seems that the isolation and divergence of wisent took place early, almost 2 to 1.6 million years ago. This appears to be compatible with the paleontological date, indicating Late Pleistocene speciation of Bison bonasus. The yak/bison mitochondrial transfer model is in agreement with our mutation analysis and phylogenetic tree. The bison/yak mutations were collected in the bison mitochondrial genome before the transfer. After the transfer, the parallel accumulation of unique mutations took place. According to our assessment, the transfer took place at about 700 ky. The characteristic feature of the wisent and bison evolution is the maintenance of mtDNA variability, despite the fact that both species underwent population bottlenecks. Our studies did not reveal any impact of these phenomena populations in the analyzed mitochondrial genomes.  相似文献

Sexual size dimorphism has been related to a high variance in male reproductive success. Research on American bison (Bison bison bison), a species that shows moderate sexual dimorphism, has put forward the assumption that under wild conditions the largest part of the female herd is bred by a limited number of males. Thus far, this hypothesis has never been supported by genetic data. The aim of this study is therefore to test the hypothesis of extreme male reproductive variance among bison through the determination of the reproductive success of the bulls in private herds. We analysed genetic paternity in four private herds (n=19, 33, 39 and 54) kept under semi-natural conditions. By analysing blood, hair and tissue samples, using 8-10 polymorphic microsatellites, the reproductive success of the bulls was quantified. Additional genetic data were gathered through a survey distributed among bison breeders with private herds over the world. Results show a highly differential reproductive success for bison males in private herds kept under semi-natural conditions. In each study group, a single male sires at least 70% of the calves. In the survey groups, a single male sires 81-100% of the calves. Only one survey group displayed a deviating pattern showing a more evenly distribution of paternity (28.57, 33.33 and 38.10%) among three same aged subadult males.  相似文献
