Detailed male meiosis, critical morphological observations and distribution pattern of diploid as well as tetraploid cytotypes of the Western Himalayan species, Bupleurum lanceolatum have been evaluated at present. A diploid (n = 8) cytotype is reported from Kashmir, whereas, both diploid (n = 8) and tetraploid (n = 16) cytotypes are available from two districts Kangra and Sirmaur of Himachal Pradesh. Out of these, the tetraploid cytotype makes new addition for the species on a worldwide basis. As per behavior, a tetraploid cytotype is characterized by abnormal meiosis leading to high pollen sterility and size variation of the pollen grains. Morphologically, tetraploids are noted to be luxuriant in comparison to the diploids.  相似文献

Chromosome number variations play an important role in the genus Medicago. In addition to polyploidy there are cases of dysploidy as evidenced by two basic numbers, x = 8 and x = 7, the latter limited to five annual species having 2n = 14. Annuals are diploid with the exception of Medicago scutellata and Medicago rugosa which have 2n = 30 and are considered the result of crosses between the 2n = 16 and 2n = 14 species. However, this hypothesis has never been tested. This study was carried out to investigate the 2n = 14 and 2n = 30 karyotypes and verify the allopolyploid origin of M. scutellata and M. rugosa. Fluorescence in situ hybridization (FISH) of rDNA probes and genomic in situ hybridization (GISH) were performed. FISH showed that all five diploids with 2n = 14 have one pair of 45S and one pair of 5S rDNA sites. M. scutellata displayed four sites of 45S and four sites of 5S rDNA, while in M. rugosa only one pair of each of these sites was found. GISH did not produce signals useful to identify the presumed progenitors with 14 chromosomes. This result suggests alternative evolutionary pathways, such as the formation of tetraploids (2n = 32) and subsequent dysploidy events leading to the chromosome number reduction.  相似文献

A new halophilous–gypsicolous species of Corynopuntia, C. halophila (Cactaceae), is described and illustrated from the state of Coahuila, north-eastern Mexico. It is morphologically similar to the diploid Corynopuntia moelleri, but it differs by some traits, such as body size and some unusual morphological features which are here reported for the first time; there are also ecological and karyological differences, as the latter study revealed that C. halophila is a polyploid with 2n = 44 chromosomes. Diagnostic morphological characters from similar taxa are discussed and arranged in a taxonomic key. Knowledge of the species ecology, biogeography, demography and conservation status is also provided.  相似文献

The Solanum brevicaule complex contains about 20 species of diploids (2n = 2x = 24), tetraploids (2n = 4x = 48) and hexaploids (2n = 6x = 72), distributed from central Peru south to northwestern Argentina. The complex is defined entirely by morphological similarity of its constituent members, which are very similar to each other and to some landraces of the cultivated potato, Solanum tuberosum. Conflicting taxonomic treatments are common among authors. Species boundaries within the complex have been studied with morphological phenetics from germplasm accessions planted in a field plot in the north central US, and with molecular marker data from RAPDs, low-copy nuclear RFLPs, and AFLPs. The present study compares these results with an additional replicated morphological study of the same germplasm accessions in a greenhouse environment in the high Andes of central Peru. The results support extensive reduction of species in the complex.  相似文献

Detailed ecological, morphological and molecular analyses were performed in mixed populations of diploid and autotetraploid Dactylorhiza maculata s.l. in Scandinavia. Comparisons were made with pure populations of either diploid ssp. fuchsii or tetraploid ssp. maculata. It was shown that mixed populations are the result of secondary contact between ssp. fuchsii and ssp. maculata. No patterns of recent and local autopolyploidization were found. Morphology and nuclear DNA markers (internal transcribed spacers of nuclear ribosomal DNA) showed that diploids and tetraploids from mixed populations have similar levels of differentiation to diploids and tetraploids from pure populations. Vegetation analyses, as well as analyses of environmental variables, revealed that diploid and tetraploid individuals in mixed populations are ecologically well differentiated on a microhabitat level. Diploids and tetraploids in pure populations have wider ecological amplitudes than they do in mixed populations. Triploid hybrids grew in intermediate microhabitats between diploids and tetraploids in the mixed populations. Plastid DNA markers indicated that both diploids and tetraploids may act as the maternal parent. Based on morphology and nuclear markers triploids are more similar to tetraploids than to diploids. There were indications of introgressive gene flow between ploidy levels. Plastid markers indicated that gene flow from diploid to tetraploid level is most common, but nuclear markers suggested that gene flow in opposite direction also may occur. Similar patterns of differentiation and gene flow appeared in localities that represented contrasting biogeographic regions. Disturbance and topography may explain why hybridization was slightly more common and the differentiation patterns somewhat less clear in the Scandinavian mountains than in the coastal lowland. Chromosome counts on the progeny of crosses between diploid and tetraploid races ofDactylis show that tetraploid hybrids are produced as well as the expected triploids. The relative proportions of 4x and 3x hybrids vary greatly in different crosses, and the data suggest that parental geno-type influences the result. Overall, the frquencies of 3x and 4x hybrids are about equal, with no indication of a difference between the reciprocal 2x×4x and 4x×2x cross-combinations except perhaps in the case of diploids and autotetraploids of the same subspecies. Rare triploid hybrids are found in crosses between diploid subspecies ofDactylis. The mechanisms by which a diploid plant could donate two genomes to its offspring are discussed in relation to theDactylis situation, and the evolutionary significance of 4x hybrids formed in this way is considered.  相似文献

Adult triploid zebrafish Danio rerio has previously been reported to be all male. This phenomenon has only been reported in one other gonochoristic fish species, the rosy bitterling Rhodeus ocellatus, despite the fact that triploidy is induced in numerous species. To investigate the mechanism responsible, we first produced triploid zebrafish and observed gonad development. Histological sections of juvenile triploid gonads showed that primary growth oocytes were able to develop in the juvenile ovary, but no cortical alveolus or more advanced oocytes were found. All adult triploids examined were male (n = 160). Male triploids were able to induce oviposition by diploid females during natural spawning trials, but fertilization rates were low (1.0 ± 3.1%) compared with diploid male siblings (67.4 ± 16.6%). The embryos produced by triploid sires were aneuploid with a mean ploidy of 2.4 ± 0.1n, demonstrating that triploid males produce aneuploid spermatozoa. After confirming that adult triploids are all male, we produced an additional batch of triploid zebrafish and exposed them (and a group of diploid siblings) to 100 ng/L estradiol (E2) from 5 to 28 dpf. The E2 treated triploids and nontreated triploids were all male. The nontreated diploids were also all male, but the E2 treated diploids were 89% female. This demonstrates that triploidy acts downstream of estrogen synthesis in the sex differentiation pathway to induce male development. Based on this and the observations of juvenile gonad development in triploids, we suggest that triploidy inhibits development of oocytes past the primary growth stage, and this causes female to male sex reversal.  相似文献

The cultivated potato, Solanum tuberosum L. (2n=4x=48), has a very large number of related wild and cultivated tuber-bearing species widely distributed in the Americas. These species, grouped in 16 taxonomic series, range from the diploid to the hexaploid level. Polyploid species are either disomic or polysomic, and sexual polyploidization via genetically controlled 2n gametes has played a major role in their evolution. Species are separated in nature by geographical and ecological barriers. However, there are several examples of sympatric species that share the same niches but do not readily cross (i.e., the diploids S. commersonii and S. chacoense in certain areas of Argentina). External barriers alone are, therefore, not sufficient to explain species integrity. In addition, there is no strong evidence indicating that genome differentiation is important in the group. In this review we present evidence supporting the assertion that interspecific pollen-pistil incompatibility, nuclear-cytoplasmic male sterility, and the endosperm are major forces that strengthen the external hybridization barriers allowing, at the same time and under specific circumstances, a certain amount of gene exchange without jeopardizing the integrity of the species.  相似文献

Genetic differentiation, physiological homeostasis, and heterozygosity were contrasted in the closely related octoploid species Fragaria chiloensis and F. virginiana, and in one of their putative diploid progenitors F. vesca. Plants from 27 natural sites supporting strawberry colonies were collected and grown in a common greenhouse. They were analyzed for 18 quantitative traits and three enzyme loci. Intra- and inter-populational variation was measured using analysis of variance techniques. Micro- and macro-environmental measurements were made at each site. Phenotypic stability of octoploid and diploid individuals was contrasted across four environmental regimes. The octoploids displayed more morphological variation, were more heterozygous, had greater physiological homeostasis, and were found across a broader range of environments. Octoploid strawberries may have broader ecological ranges than the diploids, both because they have higher levels of physiological homeostasis, and also because they have undergone more genetic differentiation. The higher levels of physiological homeostasis may be due to the fact that they are more heterozygous than the diploids.  相似文献

Heuchera micrantha (Saxifragaceae) is a morphologically variable species comprising five varieties: diversifolia, erubescens, hartwegii, micrantha, and pacifica. Both diploids (2n = 14) and tetraploids (2n = 28) occur within the species. The tetraploid cytotype occupies the central portion of the geographic range of the species, whereas diploids occur primarily in the southern and northern portions of the range. Both diploids and tetraploids have been detected within vars. diversifolia, pacifica, and hartwegii. All counts for vars. erubescens and micrantha are diploid and tetraploid, respectively. Several lines of evidence suggest that tetraploid H. micrantha is of autopolyploid origin. The species is distinct morphologically and is also well separated geographically from other closely related species in subsection Micranthae. The two cytotypes are karyologically identical, possess nearly the same suite of allozymes, and have a very high genetic identity (Ī = 0.971). Significantly, an earlier study documented tetrasomic inheritance in the tetraploid cytotype. Following theoretical expectations, the mean number of alleles per locus, proportion of loci that are polymorphic, and observed heterozygosity are significantly higher for the autotetraploid than for the diploid. The occurrence of both cytotypes in three of the varieties suggests that autopolyploidy may have occurred several times independently in H. micrantha. This was further substantiated by discriminant analysis using morphological characters, which provided evidence for a minimum of two separate origins for the autopolyploid cytotype.  相似文献

This study investigated the pattern of variation in nuclear DNA content at different ploidy levels in Fragaria (Strawberry, Rosaceae) using flow cytometry based on mean fluorescent intensity (MFI) reflected by propidium-iodide-stained nuclei. On average, MFI values were 237 for diploids F. vesca, F. viridis, and F. nubicola, 416.5 for tetraploid F. orientalis, 621.5 for hexaploid F. moschata, and 798 for octoploids F. × ananassa, F. virginiana, and F. chiloensis. Within diploids MFI ranged from 225.9 in F. vesca ssp. vesca to 255.4 in F. nubicola, and within octoploids varied from 766 in F. × ananassa to 808 in F. virginiana. The nuclear DNA variation was significant among diploid species (N = 21, P < 0.008), but not across octoploid species (N = 17, P>0.386). MFI values were also variable among different genotypes of a given species though not significant. The values of mean basic genome DNA (MFI divided by ploidy level) were 118.5, 104, 103.5, and 99.8, respectively, for diploids, tetraploid, hexaploid, and octoploid species. This indicates that relative genomic size decreases by increasing ploidy level, and that there is no direct proportional relationship between DNA content and ploidy levels in Fragaria, supporting the idea of genome downsizing during polyploidization in plants.  相似文献

Fish of the genus Selene, known as lookdowns or moonfish, are one of the most morphologically derived groups of the family Carangidae, whose phylogenetic relationships are still largely unknown. In this study, we discuss karyoevolutionary aspects of three representatives of this genus from the Western Atlantic: Selene brownii (2n = 48; FN = 48), Selene setapinnis (2n = 46; FN = 48), and Selene vomer (2n = 48; FN = 50). Their body patterns were also investigated and compared to one another and in relation to two other species of different genera. Two mechanisms of karyotypic evolution seem to have acted in the diversification of this genus, namely pericentric inversions and centric fusions. Mapping of rDNA sequences showed that chromosome pairs bearing 5S rDNA sites are similar, whereas those bearing 18 rDNA sites are morphologically distinct while apparently also exhibiting interspecies synteny. Although the nucleolar organizer-bearing chromosomes are extremely efficient cytotaxonomic markers among Selene species, others cytogenetic patterns of these species are relatively conserved. Hybridization with telomeric probes (TTAGGG)n did not exhibit interstitial telomeric sites (ITS), especially in S. setapinnis, where, along with a reduction in diploid number, a large metacentric pair derived from centric fusion is present. Data obtained by geometric morphometrics enable a clear morphological distinction among the three species, as well as in relation to two other species of the genus Caranx and Oligoplites. Data obtained suggest that morphologic evolution in Selene species was primarily dissociated from visible changes that occurred at the chromosomal level.  相似文献

Three endemic Centaurea species from North Africa are investigated for the first time by chromomycin fluorochrome banding for GC-rich DNA distribution, fluorescence in situ hybridization for physical mapping of rRNA genes, and flow cytometry for genome-size assessment. Investigated species belong to three different sections and possess three basic chromosome numbers: C. tougourensis subsp. tougourensis 2n = 4x = 36 (x = 9), C. musimonum 2n = 2x = 20 (x = 10), and C. maroccana 2n = 2x = 24 (x = 12). The number and distribution of chromomycin positive bands (CMA+) and 18S-5.8S-26S (35S) rDNA loci were different among investigated species and ranged from 6 to 80 chromomycin bands and from 2 to 6 35S rDNA loci. The three species have just one 5S rDNA locus at intercalary position on a separate chromosome pairs, except in the case of C. musimonum in which both rDNA loci were localized on the same chromosome. All rDNA loci were co-localized with CMA+ bands, except three 35S in C. musimonum. Genome size ranged from 2C = 1.66 to 2C = 2.86 pg in diploid species (C. musimonum and C. maroccana, respectively) and to 2C = 4.51 pg in tetraploid C. tougourensis subsp. tougourensis.  相似文献

Nuclear genome size, as measured by flow cytometry with propidium iodide, was used to investigate the relationships within the genus Eucomis L’Hér. (Hyacinthaceae). Most species of Eucomis have the same basic chromosome number, x = 15. However, the somatic DNA 2C-value (2C) is shown to range from 21 to 31 pg for the diploids. The largest genome contains roughly 1010 more base pairs than the smallest. Genome sizes are evaluated here in combination with available morphological and geographical data. Therefore, the taxonomy proposed here is not based on genome size alone. The genus Eucomis, as here determined, has 12 species. These can be divided into two groups: mainly dwarf diploid species and large-sized, tetraploid species. A small diploid plant, Eucomis (autumnalis subsp.) amaryllidifolia, is restored to species status, as a diploid subspecies seems incongruent with an allotetraploid Eucomis autumnalis. Moreover, as a diploid it is separated reproductively from the allotetraploid E. autumnalis. A new diploid species that has the lowest C value, E. grimshawii, is described here. On the basis of DNA content and other morphological characters, possible parents are suggested for all tetraploid species. Nuclear DNA content as measured by using flow cytometry may conveniently be used to produce systematic data. It is applicable even in dormant bulbs or sterile plants for the monitoring of the trade in bulbous species.  相似文献

Cytogenetic characterization by karyotyping and determination of DNA content by flow cytometry of five species of Chrysolaena (Vernonieae, Asteraceae) was performed. This is the first study of nuclear DNA content realized in the genus. The 2C-values were compared with the ploidy level and the total karyotype length (TKL) of each species. Mitotic analysis revealed a base chromosome number x = 10 for all entities and different ploidy levels, from diploid (2n = 2x = 20) to octoploid (2n = 8x = 80). All species showed bimodal karyotypes composed of metacentric and submetacentric chromosomes. The average chromosome size (ML) varied from 1.86 μm to 2.70 μm, while the TKL ranged from 18.65 μm to 80.55 μm. The intrachromosomal asymmetry index (A1) varied from 0.27 to 0.38, while the interchromosomal asymmetry index (A2) ranged from 0.19 to 0.25. A new cytotype is reported for the first time for C. propinqua. Accessory chromosomes found in C. verbascifolia, C. cognata, C. flexuosa, and C. propinqua are also reported as new.  相似文献

Morphological variation, ploidy level and genetic diversity have been studied on 10 populations of the Pilosella hoppeana aggr. from the Alps, Abruzzo, Calabria and Sicily. Chromosome counts showed that the plants from Abruzzo and those from Sicily are tetraploid (2n = 36); they are assigned to P. hoppeana subsp. macrantha. The plants from the Alps (P. hoppeana subsp. hoppeana) and those from Calabria are diploid. The Calabrian populations, previously included in P. hoppeana subsp. macrantha, are shown to belong to a separate species, P. leucopsilon. The principal component analysis, based on 25 morphological characters, allowed distinguishing clearly four groups. An allozyme study using 10 enzyme systems revealed 7 polymorphic loci with a total of 20 alleles, some of them exclusive at regional level, others shared between populations showing similar morphological features. The genetic differentiation between populations was relatively high. The obtained dendrogram supports recognition of the morphologically defined taxa.  相似文献

Cytological studies were carried out on two populations of Trigonobalanus verticillata in Yinggeling, Hainan Province, China (YGL), and Fraser’s Hill, Malaysia (FH). In the two populations, the pattern of interphase nuclei was of the simple chromocentre type, the mitotic prophases were of the proximal interstitial type and chromosome numbers were 2n = 2x = 14 (YGL) and 2n = 6x = 42 (FH), representing diploid and hexaploid respectively. The basic chromosome number of the species was x = 7; this is a unique number in the Fagaceae and in the Fagales as well. The characteristic karyotype of the diploid population in China was 2n = 14 = 8m + 4sm + 2st. Based on the new finding of the chromosome number in this study, we clarify the previous dispute about the basic chromosome number and propose that the two Asian Trigonobalanus species have closer affinity.  相似文献

The potatoes, Solanum L. section Petota Dumortier, are a group of species that possess a very broad range of biological diversity, and a wide geographical distribution in the Americas. These species constitute euploid series with somatic chromosome numbers ranging from 2n = 2x = 24 to 2n = 6x = 72. Although special attention has been given to the origin of polyploid potato species, principally the cultivated forms, that are major food crops, and hybridization has been accepted as an important evolutionary force in the section, the mechanisms involved in the origin and evolution of the diploid species have not been elucidated. Herein, we propose that homoploid hybridization is the main mechanism involved in the origin and evolution of the diploid potato species, and discuss the evidences that support our proposal.  相似文献

Morphological relationships were investigated among diploidStellaria porsildii, polyploidS. longipes, and diploidS. longifolia. Canonical discriminant analysis, based on a priori assumptions to maximize differences among groups, showed thatS. longipes clusters equally distant between the two diploid species along an axis connecting the diploids' centroids, but it differs along an axis perpendicular to this axis. The intermediacy along the former axis is evidence thatS. longipes is an amphiploid derived fromS. longifolia andS. porsildii. The divergence along the latter axis may be attributable to adaptively valuable heterotic traits which were retained following amphidiploidization. The only morphological discontinuity occurred between the two diploids, whereas the morphological range ofS. longipes overlapped the range of both diploids forming a continuum. The lack of discrete clusters is likely due to hybridization and introgression withS. longifolia on one hand, and convergence of traits betweenS. longipes var.monantha andS. porsildii on the other. High a posteriori assignments in classificatory discriminant analysis supports the separation ofS. longipes var.monantha from otherS. longipes specimens. AlthoughS. longipes var.monantha grouped close toS. porsildii, the two groups separate based on leaf shape traits. Overall results support, firstly, the hypothesis thatS. porsildii is a diploid parent species which by hybridizing withS. longifolia gave rise to polyploidS. longipes. Secondly, results suggest thatS. longipes var.monantha converged morphologically towardsS. porsildii relatively recently due to ecological specialization, and merits distinction at least as a variety ofS. longipes.  相似文献
