The phasmatid species Didymuria violescens comprises ten distinct chromosome races parapatrically distributed such that adjacent races meet in narrow zones of overlap. The interracial karyotypic variation is remarkable and involves both diploid number differences (in the range 26–40) and differences in the sex-chromosome mechanism. Karyotypic comparisons and analyses of the meiotic pairing relationships in interracial hybrids have shown that the differences derive in large part from a series of centric fusion events and X-autosome fusions, which together contribute to the reduction in chromosome number within the species. The origin and development of the current racial pattern can best be interpreted in terms of the stasipatric hypothesis of White.This paper is affectionately dedicated to Professor Spencer Smith-White on the occasion of his 66th birthday.  相似文献

Populations may diverge at fitness-related genes as a result of adaptation to local conditions. The ability to detect this divergence by marker-based genomic scans depends on the relative magnitudes of selection, recombination, and migration. We survey rat (Rattus norvegicus) populations to assess the effect that local selection with anticoagulant rodenticides has had on microsatellite marker variation and differentiation at the warfarin resistance gene (Rw) relative to the effect on the genomic background. Initially, using a small sample of 16 rats, we demonstrate tight linkage of microsatellite D1Rat219 to Rw by association mapping of genotypes expressing an anticoagulant-rodenticide-insensitive vitamin K 2,3-epoxide reductase (VKOR). Then, using allele frequencies at D1Rat219, we show that predicted and observed resistance levels in 27 populations correspond, suggesting intense and recent selection for resistance. A contrast of F(ST) values between D1Rat219 and the genomic background revealed that rodenticide selection has overwhelmed drift-mediated population structure only at Rw. A case-controlled design distinguished these locus-specific effects of selection at Rw from background levels of differentiation more effectively than a population-controlled approach. Our results support the notion that an analysis of locus-specific population genetic structure may assist the discovery and mapping of novel candidate loci that are the object of selection or may provide supporting evidence for previously identified loci.  相似文献

Fish of the genus Selene, known as lookdowns or moonfish, are one of the most morphologically derived groups of the family Carangidae, whose phylogenetic relationships are still largely unknown. In this study, we discuss karyoevolutionary aspects of three representatives of this genus from the Western Atlantic: Selene brownii (2n = 48; FN = 48), Selene setapinnis (2n = 46; FN = 48), and Selene vomer (2n = 48; FN = 50). Their body patterns were also investigated and compared to one another and in relation to two other species of different genera. Two mechanisms of karyotypic evolution seem to have acted in the diversification of this genus, namely pericentric inversions and centric fusions. Mapping of rDNA sequences showed that chromosome pairs bearing 5S rDNA sites are similar, whereas those bearing 18 rDNA sites are morphologically distinct while apparently also exhibiting interspecies synteny. Although the nucleolar organizer-bearing chromosomes are extremely efficient cytotaxonomic markers among Selene species, others cytogenetic patterns of these species are relatively conserved. Hybridization with telomeric probes (TTAGGG)n did not exhibit interstitial telomeric sites (ITS), especially in S. setapinnis, where, along with a reduction in diploid number, a large metacentric pair derived from centric fusion is present. Data obtained by geometric morphometrics enable a clear morphological distinction among the three species, as well as in relation to two other species of the genus Caranx and Oligoplites. Data obtained suggest that morphologic evolution in Selene species was primarily dissociated from visible changes that occurred at the chromosomal level.  相似文献

Karyotypes of 13 Rattus species collected in Asia and Oceania were analysed with special interest to karyotype evolution and species differentiation. They were classified into three groups according to their karyotype similarity. Four species (R. annandalei, R. exulans, R. muelleri and R. norvegicus) with 2n=42 and a karyotype similar to some of the polymorphic karyotypes in the Asian black rats (R. rattus) are classified into the first group. Pericentric inversion of some acrocentrics seemed to have caused the differentiation of these species. The other four species (R. bowersii, R. fuscipes, R. leucopus and R. conatus) with similar karyotypes as the above group, but lower chromosome numbers than 2n=42 are classified into the second group. Robertsonian fusion in some acrocentrics observed in the first group are suggested to have caused the development of the species in this group. The remaining four species (R. sabanus, R. canus, R. huang and R. niviventer) with karyotypes markedly different from the above two in having a fewer number of small metacentrics are classified into the third group. They seemed to be more primitive karyotypes than the other Rattus species. By the comparison between the polymorphic karyotypes in the black rat, and karyotypes in its related species it was suggested that the former had occurred as primary events to the differentiation of the latter. Biochemical analyses of partially purified preparations of APH-4 and -6 (common allelic forms) and APH-2 and -10 (rare allelic forms) of D. melanogaster reveal that the two common forms are similar in all properties investigated except for pH optimum (8.0 vs. 8.5). The common and rare forms share certain properties in common but differ in that the common forms are more stable to heat and more sensitive to inhibition by inorganic phosphate. With respect to such properties as substrate preferences and K i values for inorganic phosphate, the common forms and APH-2 are similar to one another, whereas APH-10 is distinctly different. All four activities show preference for a phosphoaromatic compound as substrate, with O-phosphotyrosine being the best substrate of biological origin. In this study, the evolutionary history of the variable second exon of RT1.Ba and its adjoining intron b are compared across a number of species and subspecies of the Australian RATTUS: Three lineages are identified in the second intron across a range of Rattus species. Two of these lineages, separated by the insertion of a probable rodent short interspersed nucleotide element and by point mutations outside the indel region, are both found in each of the major clades of the endemic Australian RATTUS: This pattern of ancestral polymorphism is reflected in the adjoining exon 2 sequences, although phylogenetic constraints confirm that the clustering is not identical to that of the associated intron sequences. In addition, the coding sequences show evidence of the retention of ancestral polymorphism, with identical exon sequences found in two divergent species, and some indication of gene conversion detected for the exon sequences.  相似文献

The diets of sympatric rodents partially define their realized niches. Identifying items in stomachs of introduced rodents helps determine rodents’ trophic positions and species most at risk of consumption. In the Hawaiian Islands, which lacked rodents prior to human arrival, three rodents (Rattus rattus or black rat, R. exulans or Pacific rat, Mus musculus or house mouse) commonly coexist in native habitats where they consume a wide range of plants and animals. These three rodent species were trapped in montane forest for 2.5 years; their stomach contents were analyzed to determine short-term diets (n = 12–95 indiv. per species), and isotopic fractions of δ15N and δ13C in their bone collagen were analyzed to further estimate their trophic positions (n = 11–20 indiv. per species). For all three species, >75 % of individuals had plants and >90 % had arthropods in their stomachs, and significant differences in mean relative abundances were found for food items in stomachs among all three rodents. Rodents may be dispersing some native and non-native seeds, including the highly invasive Clidemia hirta. Most identifiable arthropods in rodent stomachs were non-native, and no stomachs contained birds, snails, or lizards. The δ15N and δ13C signatures were consistent with trophic feeding differences revealed from stomach contents. Dietary niche differentiation by coexisting rodent species is evident in this forest, with Pacific rats being intermediate between the mostly carnivorous house mouse and the mostly herbivorous black rat; such findings can help forecast rodent impacts and direct management efforts in ecosystems where these invasive animals coexist.  相似文献

Four reproductively isolated populations of the Australian bush-rat, Rattus fuscipes greyii , are polymorphic for electrophoretic variants of the mitochondrial enzyme, NADP-dependent iso-citrate dehydrogenase (M-Idh). The electrophoretic patterns of M-Idh and a small amount of breeding data are in agreement with the hypothesis that the variation is controlled by two alleles at an autosomal locus, Idh-2. In three relatively large populations, the proportion of heterozygotes at the Idh-2 locus ranges from 0.07 to 0.23, while on a small off-shore island it is 0.60. In this latter population there is an excess of heterozygotes which is on the borderline of statistical significance, suggesting that heterotic selection may be maintaining the polymorphism. Populations on eight other small islands are monomorphic for the allele which is the most frequent in the large populations. In most species M-Idh is less variable than most other enzymic proteins and the results presented here constitute one of the few published examples of an extensive M-Idh polymorphism.  相似文献

A genetic locus controlling the electrophoretic mobility of a methylglyoxal dehydrogenase (EC 1.2.1.23) in the rat is described. The locus, designatedMgd1, is expressed in liver and kidney. Inbred rat strains have fixed either alleleMgd1 a or alleleMgd1 b . Codominant expression is observed in heterozygotes, providing evidence for a tetrameric enzyme structure. Backcross progenies showed the expected 1:1 segregation ratio, and there is evidence thatMgd1 is linked toPep3 andFh1 on chromosome 13. 多重PDZ结构域蛋白1型(MUPP1)是一种存在于上皮细胞和神经细胞内含有13个PDZ结构域的重要支架蛋白.在上皮细胞中,MUPP1蛋白在紧密连接结构的形成和上皮细胞的极化过程中发挥重要作用.而在中枢神经系统中,MUPP1基因的1个提前终止突变导致了其最后12个PDZ结构域的缺失,以及严重的先天性脑积水.此外,MUPP1蛋白的表达水平与酒精依赖性和药物戒断的严重性也具有显著的相关性.因此,对MUPP1蛋白所含的PDZ结构域进行纯化和性质鉴定,将有助于深入研究MUPP1蛋白的功能和分子机制.在本文研究中,利用亲和纯化和分子筛技术,对大鼠来源的MUPP1蛋白的第8个PDZ结构域进行了表达和纯化.多角度激光光散射的数据表明: MUPP1-PDZ8结构域在溶液中为单体,分子量为16.4 kD.圆二色谱结果表明,MUPP1-PDZ8结构域具有较好的二级结构折叠,测得其熔解温度为71.6摄氏度,暗示该PDZ结构域在溶液中非常稳定.最后,MUPP1-PDZ8结构域的晶体结构显示,该结构域属于I 型PDZ 结构域,包含3个α螺旋和6个β折叠.其中GLGL模块、β折叠B上的1 351位亮氨酸,以及α螺旋B上的1 405位异亮氨酸/1 398位组氨酸形成的PDZ结合口袋,可以特异性地与其目标蛋白质的羧基末端相结合.综上所述,本文的研究提供了MUPP1-PDZ8结构域的生化特性,以及该结构域与其目标蛋白质相互作用的分子机制,这将为MUPP1蛋白的功能研究提供生物化学与结构生物学的理论基础.  相似文献

The distribution of genetic diversity in a local population of the trematode Schistosoma mansoni was determined within and between individual wild rats at a microspatial geographic scale of a standing water transmission site. Using RAPD markers, molecular variance and canonical correspondence analysis were performed to test the significance of genetic differentiation between infrapopulations. Of total gene diversity, 8 and 11% was partitioned between hosts trapped at few metres distance from each other. Significant temporal differentiation (2%) was also detected among schistosomes sampled at 6 month intervals with more infrapopulation pairs differentiated during the dry season of parasite transmission than during the rainy season (45 and 12%, respectively). A combination of factors such as restricted displacement of rats, patchy spatial aggregation of infected snails and limited cercarial dispersion in standing water are likely to promote the genetic differentiation observed between infrapopulations at this microgeographic scale.  相似文献

Abstract This review provides a synthesis, from published and unpublished sources, of records of mycophagy (fungus-feeding) by Australian mammals. Mycophagy is shown to be widespread among Australian mammals, paralleling the previously well-documented situation in North America. Mycophagy appears to be most prevalent within the Potoroidae (rat-kangaroo family) but has also been recorded for a variety of other mammals. Information is presented on the classification, morphology and ecology of the fungi consumed, on the nutritional benefits (or otherwise) of mycophagy, and on the role of mammals in spore dispersal. Fungi whose sporocarps are hypogeal (truffles, false-truffles and sporocarpic Endogonaceae) and which enter into mycorrhizal relationships with plants predominate among the species recorded in mammal diets.  相似文献

Twenty-five unique CfoI-generated whole-cell DNA profiles were identified in a study of 30 Paenibacillus alvei isolates cultured from honey and diseased larvae collected from honeybee (Apis mellifera) colonies in geographically diverse areas in Australia. The fingerprint patterns were highly variable and readily discernible from one another, which highlighted the potential of this method for tracing the movement of isolates in epidemiological studies. 16S rRNA gene fragments (length, 1,416 bp) for all 30 isolates were enzymatically amplified by PCR and subjected to restriction analysis with DraI, HinfI, CfoI, AluI, FokI, and RsaI. With each enzyme the restriction profiles of the 16S rRNA genes from all 30 isolates were identical (one restriction fragment length polymorphism [RFLP] was observed in the HinfI profile of the 16S rRNA gene from isolate 17), which confirmed that the isolates belonged to the same species. The restriction profiles generated by using DraI, FokI, and HinfI differentiated P. alvei from the phylogenetically closely related species Paenibacillus macerans and Paenibacillus macquariensis. Alveolysin gene fragments (length, 1,555 bp) were enzymatically amplified from some of the P. alvei isolates (19 of 30 isolates), and RFLP were detected by using the enzymes CfoI, Sau3AI, and RsaI. Extrachromosomal DNA ranging in size from 1 to 10 kb was detected in 17 of 30 (57%) P. alvei whole-cell DNA profiles. Extensive biochemical heterogeneity was observed among the 28 P. alvei isolates examined with the API 50CHB system. All of these isolates were catalase, oxidase, and Voges-Proskauer positive and nitrate negative, and all produced acid when glycerol, esculin, and maltose were added. The isolates produced variable results for 16 of the 49 biochemical tests; negative reactions were recorded in the remaining 30 assays. The genetic and biochemical heterogeneity in P. alvei isolates may be a reflection of adaptation to the special habitats in which they originated.  相似文献

Metaphase chromosomes from neuroblasts of strains from both laboratory stocks and natural populations of D. serido, D. meridionalis, D. borborema and D. buzzatii have been studied using colcemid pretreatment, and air-drying followed by Giemsa staining. The two latter species both show a uniform ‘basic’ metaphase karyotype. D. serido and D. meridionalis, on the other hand, both include a number of different, geographically distinct, metaphase karyotypes involving differences in the major blocks of constitutive heterochromatin present on the sex chromosomes and/or the 6th chromosome (microchromosome). These chromosomal differences are largely due to the acquisition of extra heterochromatin though pericentric inversions appear to be responsible for some of the Y-chromosome variants in D. serido. Moreover, the cytological evidence demonstrates that populations of both species are far from continuous in distribution. The extent to which such cytological differences reflect the existence of subspecific or specific complexes with minimal morphological differentiation is under investigation.  相似文献
