In vitro and in planta sensitivity of an indirect enzyme-linked immunoassaytechnique, using a monoclonal antibody specific for the lipopolysaccharide (LPS) of Xanthomonas campestris pv. vesicatoria , was increased 10-foldby using a newextraction buffer (gl of : KH2PO4, 2; NaHPO4, 11·5; EDTAdisodium, 0·14; thimerosal, 0·02; and lysozyme, 0·2). The procedure improvedsensitivity without increasing background levels. In vitro , the limit of detection wasbetween 1×107 and 1×108 cells ml−1 with the conventionalextraction buffer phosphate-buffered saline (PBS) and less than 1×106 cells ml−1 when lysozyme extraction buffer was substituted for PBS. In comparing 22 X. c.vesicatoria strains, absorbance readings were increased close to three-fold with the lysozymeextraction buffer as opposed to PBS. When leaf tissue extract was spiked with the bacterium, thelimit of detection was 1×107 cfu ml−1 and 1×108 cfu ml−1 with the lysozyme solution and PBS, respectively, as the extraction buffers. Whenusing the lysozyme extraction buffer in combination with a commercial amplification system, thelimit of detection was decreased to less than 1×105 cfu ml−1 in leaftissue. The addition of the lysozyme and EDTA to the phosphate buffer resulted in release of asignificant quantity of LPS and concomitant dramatic increase in sensitivity. The new procedure,termed lysozyme ELISA (L-ELISA), should increase sensitivity of ELISA reactions where LPS isthe reacting epitope.  相似文献

By using two polyclonal antisera against WH 7803 strain (Synechococcus sp.) and WH 5701 strain (Synechococcus bacillaris) it is possible to detect and to enumerate cells of the two cyanobacterial serogroups. The immunofluorescence technique was used to study the distribution of the two serogroups in the estuarine, coastal and upwelling waters of the Mediterranean Sea surrounding Messina. In the estuarine waters of the Alcantara River (Ionian Sea), the WH 7803 serogroup was present at a concentration in the order of 102 cells ml−1 and the WH 5701 serogroup at a concentration of 5·5 × 102 cellsml−1. In the coastal waters of Messina, where urban and industrial wastes are usuallydumped, the concentration of total phycoerythrin- Synechococcus ranged from 1·3 × 102 to 4·1 × 103 cells ml−1; the WH 7803 serogroup accounted for 50–94% of the totalpopulation in Ionian stations, whereas the WH 5701 serogroup ranged from1·4 × 101 to6·7 × 102cells ml−1. In the upwelling area (Straits of Messina) bothserogroups were found. Vertical distribution of two Synechococcus strains had anopposite trend and their concentrations were of the order of 101–102cells ml−1. Theuse of the Scan laser system allows both autofluorescent and labelled organismsto be distinguished in a preparation for optical microscopy. The efficacy of high-temperature, short-time (HTST) pasteurization (72 °C/15 s) when low numbers (≤ 103 cfu ml −1 ) of Mycobacterium paratuberculosis are present in milk was investigated. Raw cows' milk spiked with Myco. paratuberculosis (103 cfu ml−1, 102 cfu ml−1, 10 cfu ml−1, and 10 cfu 50 ml−1) was subjected to HTST pasteurization using laboratory pasteurizing units. Ten bovine strains of Myco. paratuberculosis were tested in triplicate. Culture in BACTEC Middlebrook 12B radiometric medium detected acid-fast survivors in 14·8% and 10% of HTST-pasteurized milk samples at the 103 and 102 cfu ml−1 inoculum levels, respectively, whereas conventional culture on Herrold's egg yolk medium containing mycobactin J detected acid-fast survivors in only 3·7% and 6·7% of the same milk samples. IS900-based PCR confirmed that these acid-fast survivors were Myco. paratuberculosis . No viable Myco. paratuberculosis were isolated from HTST-pasteurized milk initially containing either 10 cfu ml−1 or 10 cfu 50 ml−1.  相似文献

Reproductive characteristics of the male herring in the northern Baltic Sea   总被引:1,自引:0,他引:1  
The gonad weight and gonadosomatic index of the male Baltic herring Clupea harengus membras L., were highest at the beginning of the reproductive season (April/May), the values decreasing towards the end of it (July/August) during 1988–1991. The decline could not be explained by fish size but may have been due to fish condition. A high individual variation was typical for both gonad weights and gonadosomatic indices in fish of the same size and maturity stage. The mean density of sperm cells was significantly higher in June (34·9 × 109 ml−1) than in July (19·2 × 109 ml−1, Mann-Whitney U= 17; P<0·05), the variation among the males being high in both groups. Electron microscope analysis showed a severe disruption of the mitochondrial elements in males spawning at 22°C.  相似文献

Abstract:  In laboratory bioassays, the efficacy of the entomopathogenic fungus Beauveria bassiana against the spruce bark beetle, Ips typographus , was tested under various conditions. Four of the tested isolates and the commercial product Boverol® caused 99–100% mortality when tested at a concentration of 1.0 × 107 conidia/ml at 25°C. Using B. bassiana isolate 138 at a concentration of 1.0 × 106, the median survival time (MST) was 6.1 d and significantly longer compared with the MST of 4.2 and 4.0 d at 1.0 × 107 and 1.0 × 108 conidia/ml, respectively. In the next experiment, the beetles were maintained on spruce bark, filter paper or artificial diet during the bioassay with Boverol®, and significant differences in the MST of 3.6, 2.5 and 5.3 d, respectively, were noticed. The experiment with Boverol® at different temperatures showed that the beetles lived significantly longer at 15°C (MST 8.7 d) than at 20, 25, 30 and 35°C. At 25°C, the beetles died most rapidly (MST 3.5 d). At different relative humidities (RH) of 40, 70 and 100%, nearly all beetles were dead after treatment with a suspension of Boverol® at 1.0 × 107 conidia/ml. At 40% RH, 49% of the untreated beetles died after 7 d. The best effects were achieved with the following bioassay: beetles were fed for three days on artificial diet, then dipped into a solution of 1.0 × 107 conidia/ml and transferred on a piece of spruce bark in Petri dishes at 25°C and 70% RH.  相似文献

A cell-free crude extract containing the white line inducing principle (WLIP), a lipodepsipeptide produced by Pseudomonas 'reactans' , could inhibit browning of mushrooms caused by Pseudomonas tolaasii . Mushrooms inoculated with Ps. tolaasii at concentrations of 2·7 × 106 cfu ml−1 or higher showed the symptoms of the disease after 2 d of incubation. Mushroom caps treated with various concentrations of a crude WLIP preparation, and later inoculated with bacterial concentrations higher than the threshold value, did not develop the symptoms of the disease. One milligram of a crude WLIP preparation could block 50% of the symptoms caused by 1·2 × 107 cfu. The inhibition of browning was effective when incubating at low temperatures for 4 d. A suspension containing 1·6 mg ml−1 of pure WLIP was also able to inhibit the symptoms of brown blotch disease induced by 7·6 × 106 cfu ml−1 of Ps. tolaasii .  相似文献

The amylase-producing ability of the intestinal microflora in cultured specimens of ayu, carp, channel catfish, Japanese eel and tilapia was determined. Mean viable counts of aerobes and anaerobes ranged from 1·1×106 to 3·7×108 cfu g−1 and from 1·3×103 to 1·6×108 cfu g−1, respectively. Aeromonas spp. and Bacteroidaceae were predominant in four to five fish species. Of 206 strains examined, 65 (31·6%) produced ≥0·01 U amylase ml−1. The percentage of producers differed among families and genera of bacteria and fish species. While 56% of the anaerobes produced amylase, only 20% of the aerobes did. More than 50% of Aeromonas , Bacteroidaceae and Clostridium strains produced amylase efficiently while Acinetobacter , coryneforms, Enterobacteriaceae, Moraxella , Plesiomonas and Streptococcus strains did not. High amylase production (≥0·05 U ml−1) was found in 12 strains, 11 from Aeromonas and one Pseudomonas . The percentage of high amylase producers in Japanese eel was lower than the other four fish (2–30%). These results strongly suggest that the amylase produced by the intestinal microflora play an important role in the digestion of starch in freshwater fish to some extent.  相似文献

H. SUGITA, J. KAWASAKI, J. KUMAZAWA AND Y. DEGUCHI. 1996. The amylase-producing ability of intestinal bacteria in one marine crab and seven fish species was determined. Mean total viable counts ranged from 1.3 × 105 to 1.5 × 108 cfu g−1, and Vibrionaceae were predominant in all specimens. Of 1585 strains examined, 341 (21.5%) produced ≥0.01 U amylase ml−1. Percentage of producers (≥0.01 U ml−1) differed among genera/families. High abilities (≥0.05 U ml−1) were found in 1.4-3.6% of Enterobacteriaceae, Pseudomonas and Vibrionaceae strains. On the other hand, percentage of producers varied with animal species. These results reveal that the amylase producers were widely distributed in the digestive tracts of coastal animals including crabs and fish, irrespective of their food habitats.  相似文献

Abstract: The feeding of the marine ciliate Euplotes mutabilis was studied using bacteria ( Vibrio natriegens ) doubly labelled with 3H-thymidine and 14C-leucine. In the presence of abundant bacteria (30 × 106 bacteria ml−1), an average Euplotes cell (initially without food vacuoles) with a protein content of 12 ng consumed 16 × 103 bacteria in the first hour and 27 × 103 bacteria over four hours, accumulating about 60% of the bacterial protein into ciliate macromolecules. Euplotes which had been starved or under-fed to reduce cell protein biomass to 7 or 9 ng consumed significantly fewer bacteria, but the gross growth efficiency for protein did not change. The rate of consumption of bacteria by large Euplotes of protein content 15 ng was initially less than that of 12 ng cells, and it decreased markedly before the end of a 4-hour experiment. Recently divided cells ingested bacteria rapidly, but showed a reduced gross growth efficiency of about 40%. At low bacterial concentrations (6 × 106 bacteria ml−1) the rates of ingestion were markedly reduced to between     and     of maximal levels; the smallest cells could not sustain feeding activity at the low prey concentration and gross growth efficiency fell from 43 to 20% during a 4-hour experiment. The strategy adopted by Euplotes in response to local fluctuations in food supply involves rapid consumption with high growth efficiency in times of plenty, but slow shrinkage without cell division to survive in times of shortage.  相似文献

The main blood parameters of the swordtail, Xiphophorus helleri , were studied. Morphology, granulation staining and cytochemistry of leucocytes in peripheral blood, kidney, spleen and gills were investigated by light microscopy. Blood parameters are similar to other fish species: Red blood cell count (4.5 × 106μl), white blood cell count (15.2 × 103μl), haema-tocrit (33.8%) haemoglobin (7.8 mg ml−1), MCV (mean corpuscle volume, 75.1 μm3). MCH (mean content of haemoglobin, 17.3 pg), MCHC (mean percentage haemoglobin/erythrocyte, 23.1%/100 ml erythrocytes). Leucocytes can be classified into lymphocytes, thrombocytes, neutrophilic and eosinophilic gra-nulocytes, monocytes macrophages and melanomacrophages.
Morphological and cytochemical features of the cells are described and compared with results from other fish species.  相似文献

Cow's milk was inoculated with ca 103 and 107 cfu ml−1 Escherichia coli O157 : H7. After fermentation at 42°C for 0–5 h, the yoghurt was stored at 4°C. Two kinds of yoghurt were used : traditional yoghurt (TY), made with Streptococcus thermophilus and Lactobacillus bulgaricus starter cultures, and 'bifido' yoghurt (BY), made with the two starter cultures plus Bifidobacterium bifidum . After 7 d E. coli O157 : H7 decreased from 3·52 to 2·72 log10 cfu ml−1 and from 7·08 to 5·32 log10 cfu ml−1 in TY, and from 3·49 to 2·73 log10 cfu ml−1 and from 7·38 to 5·41 log10 cfu ml−1 in BY. The pH values of yoghurt dropped from 6·6 to 4·5 and 4·4 in TY (for low and high pathogen inocula, respectively), and from 6·6 to 4·6 and 4·5 in BY (for low and high pathogen inocula, respectively).  相似文献

The antimicrobial activity of the indoloquinoline alkaloid, cryptolepine, isolated from Cryptolepis sanguinolenta (Fam. Periplocaceae) was determined against selected micro-organisms. The minimum inhibitory concentration (MIC) ranges obtained, expressed as μg ml−1, were: 5–10 for Saccharomyces cerevisiae NCPF 3139; 10–20 for S. cerevisiae NCPF 3178; 20–40 for Escherichia coli NCTC 10418; 40–80 for E. coli NCTC 11560, Candida albicans ATCC 10231 and C. tropicalis NCPF; and 80–160 for C. albicans NCPF 3242 and NCPF 3262.
Biocidal effects were noted at concentrations 2–4 times those of the MIC of the alkaloid following challenge with 106 cfu ml−1 of micro-organisms. Time-kill studies showed a reduction in viable count from 106 to < 10 cfu ml−1 in 4 h in C. albicans ATCC 10231 exposed to 320 μg ml−1 of the agent; 3 log cycle reductions were recorded for the 6 h counts of E. coli NCTC 10418 and S. cerevisiae NCPF 3139 exposed to 40μg ml−1 and 160 μg ml−1 of the alkaloid respectively.
These results were consistent with findings using scanning electron microscopy. Exposure of cells to biocidal concentrations of cryptolepine produced filamentation prior to lysis in E. coli NCTC 10418 and extreme disturbance of surface structure, including partial and total collapse, followed by lysis in C. albicans ATCC 10231 and S. cerevisiae NCPF 3139.  相似文献

Abstract The four species of Bordetella differed in their ability to grow at 37°C in membrane-filtered tracheobronchial washings (TBW) from seven vertebrate species, including their natural hosts. From washed inocula of approximately 2×103 colony-forming units per ml (cfu ml−1), Bordetella bronchiseptica and B. avium grew much better than the other two bordetellae and yielded stationary-phase cultures containing 108−109 cfu ml−1 in most of the TBW samples. These counts were only moderately lower than those attained in CL medium which contains about a 450-times higher concentration of amino acids. B. bronchiseptica and B. avium also grew to a limited extent in phosphate-buffered saline without nutrient supplements. B. parapertussis grew in TBW from man, sheep, rabbit, mouse and chicken, but not in TBW from a dog and a horse or in PBS. B. pertussis grew well in CL medium, but not in PBS or in any of 13 samples of TBW from the seven vertebrate species, which included three samples of lung lavage fluid from human patients. Analysis of the TBW samples for known Bordetella nutrients revealed concentrations of amino acids and nicotinic acid averaging 0.35 mM and 0.56 μg ml respectively.  相似文献

Abstract The distribution of oil-degrading microorganism in samples of surface water and sediment from North Eastern Japanese coastal waters was studied. Modified natural sea water (NSW) agar supplemented with emulsified crude oil (Arabian light, 5 g 1−1) was used to enumerate oil-degrading bacteria. In addition, filtered samples were inoculated into NSW broth containing weathered crude oil. Incubation was carried out at 20°C for 7–10 days. Populations of oil-degrading microorganisms ranged from 3–230 CFU 100 ml−1 in surface waters and 2.9 × 103 to 1.2 × 105 CFU g in sediment samples. Analysis of variance showed that oil-degraders were heterogenously distributed. Six mixed populations selected from 20 samples were studied to determine which of the constituent microflora were capable of crude oil biodegradation. Among 51 strains selected for identification, only 61% could be identified which formed 17 different bacterial species. Acinetobacter species (14 strains), Psychrobacter immobilis (9 strains) and Gram-positive cocci (10 strains) were the predominant types. Oil-degrading activity by various mixed populations (three each from water and sediment samples) was determined by using a conventional total weight reduction technique. Reduction in amount of various aliphatic and aromatic hydrocarbon substrates was verified using gas chromatography and high pressure liquid chromatography. Biodegradation of crude oil ranged from 35–58%. One mixed population of the sediment samples degraded more hydrocarbon (both aliphatic and aromatic) and the biodegradation of the aromatic hydrocarbon reached as high as 48%.  相似文献

A commercial fluorochromic system was evaluated for the rapid detection of lactic acid bacteria in fortified wines by the epifluorescent filter technique (DEFT). The viability test used, employing the fluorescence dyes SYTO 9 and propidium iodide, was able to detect and clearly differentiate viable from non-viable cells (killed with a 50% v/v ethanol solution). A good overall agreement ( r = 0·92) was obtained between the DEFT count and the plate count in the range studied (5 × 102–4 × 109 cells ml−1). Wine components which might otherwise interfere with the method could be removed by including simple wash steps in the protocol. This measure proved critical to the success of the procedure. For practical purposes, the rapid method studied seems to be a good alternative to the traditional cultural methods as part of quality control programmes in wine making. It may also be useful when studying the efficacy of certain treatments in the elimination of wine bacterial contaminants.  相似文献

Abstract: The rates of ingestion of bacteria and of accumulation of bacterial biomass by hungry Pteridomonas danica and Paraphysomonas imperforata were measured using dual radioactive-labelled bacteria in experiments lasting 4–8 h. Pteridomonas continuously consumed 4–5 bacteria h−1 throughout experiments lasting 8 h, irrespective of bacterial concentration above a threshold of about 5 × 105 bacteria ml−1, and continued to catch bacteria even below this density. The clearance rate of about 1 nl cell−1 h−1 at higher bacterial concentrations increased three or four times as bacterial numbers fell. Paraphysomonas cells, with only half the biomass of Pteridomonas , ingested up to 10 bacteria h−1 at high bacterial concentrations, and gradually reduced the feeding rate, effectively ceasing to feed at 106 bacteria ml−1; their initial clearance rate of 1–2.5 nl cell−1 h−1 subsequently fell as low as 0.1 nl cell−1 h−1. Estimation of feeding rate by extrapolation from short-term experiments on such flagellates requires extreme caution. These flagellates, starved to levels typical of the natural environment, accumulated ingested bacterial biomass at an efficiency of between 16 and 21%, indicating that in nature they would recycle 80% or more of the nutrients contained in their food.  相似文献

The sludge from hospital waste treatment facilities is a potential source of infectious organisms. The average numbers of micro-organisms in the sludge of hospital wastewater in Taiwan were as follows: total count 8·1 × 107 cfu g−1 (dry weight of sludge), and 1·4 × 106, 3·6 × 105, 1·6 × 105, 2·2 × 105 and 5·5 × 104 cfu g−1 (dry weight of sludge) for total coliforms, faecal coliforms, faecal streptococci, Pseudomonas aeruginosa and Salmonella spp., respectively . Salmonella spp. were detected in 37% (10 of 27) of the sludges from hospital wastewaters. Therefore, the treatment of such sludge to reduce pathogenic micro-organisms should be considered.  相似文献

SUMMARY: Experiments are described in which minced chicken meat, packed anaerobically, was irradiated at room temperature and in the frozen state with a wide range of doses of 4 MeV cathode rays. Sterility was achieved in 14 out of 15 samples which had received 2 × 106 rads or more. Doses of 0·5 and 1·0 × 106 rads allowed survival of a few bacteria/g, usually spore formers. Bacterial counts indicated an approximately logarithmic decrease in numbers at lower doses, while freezing reduced the bactericidal effect.
The storage life at 5° was prolonged only slightly by doses of 5 × 104 and 10 × 104 rads, and highly variable results were obtained with 17·5 × 104 rads. A dose of 25 × 104 rads, however, increased the storage life very considerably. The types of bacteria present initially, and after irradiation with low doses and storage at 5°, were studied. After storage for 12 days or more various types of nonsporing Gram-positive rods were predominant in almost all samples, both control and irradiated. Streptococci were also important where irradiation with 17·5 × 104 and 25 × 104 rads was followed by long storage.  相似文献

Blood samples from healthy adult Atlantic salmon fed an optimal diet in net sea pens were collected at intervals from October to May. Haematological determinations and biochemical serum analyses were carried out on 20 fish in each of seven samples. The ranges of haemato-logical values for sample means were: haematocrit 44–49%, haemoglobin 8.9–10.4 g 100ml−1, red blood cell count 0.85–1.10 × 1012 l−1, MCV 441–553 × 10−15 1, MCH 94–106 × 10−6 g, MCHC 19.4–21.7 g 100ml−1 and leucocrit 0.43–0.96%. The ranges of enzyme activities in serum, for sample means, were: alkaline phosphatase 647–988Ul−1, aspartate aminotrans-ferase 202–351 Ul−1 and alanine aminotransferase 4–8 Ul−1. The ranges of the other parameters analyzed in serum were: total protein 41.6–56.6 gl−1, albumin 18.3–24.3 gl−1, albumin/total protein ratio 39.3–44.0%, creatinine 26–46 μmol, triglycerides 2.53–4.98 μmol and cholesterol 9.3–12.8 μmol. These values are considered to be the normal ranges in healthy fish. Variations due to seasonal changes, and the clinical significance of the selected parameters, are discussed. Data showing the reproducibility of the biochemical analyses in serum are presented.  相似文献

The structure of six Irish kefir samples was studied in the electron microscope, and the microbial composition and fermentation kinetics during growth in 10% reconstituted skim milk at 21°C. The microbial composition of the six samples was similar; at the end of the fermentation the counts of lactococci, leuconostocs, lactobacilli, acetic acid bacteria and yeasts were 109, 108, 5 × 106, 105 and 106 ml−1 respectively; the levels of acetic acid bacteria and lactobacilli showed some intersample differences. Lactate was the major metabolite followed in order by ethanol, acetate and acetoin. The final concentrations of L-lactate produced (66–90 mmol kg−1) were 10-fold higher than those of D-lactate. Acetate and ethanol concentrations varied from 4 to 14 and 2 to 40 mmol kg−'1 respectively. The rates of citrate utilization and concentration of acetoin produced during growth differed between samples. Scanning electron microscopy showed not only variation between the interior and exterior of the sample but also large variation between different sections of the interiors and exteriors of the same sample. Long and short, and straight and curved rods and yeasts were seen in all samples, the curved rods observed in the interior, but lactococci were seen on the surface of only one sample. There were no gross differences in structure between samples.  相似文献
