The kinetics of extension induced by GA31 in the hypocotyl ofintact seedlings of Lactuca sativa are similar in the dark andin the light, and differs fundamentally from the kinetics ofelongation in the dark without GA3. Both in continuous lightand in the dark, GA3-induced promotion starts 24 h after incubation.In the dark, even low concentrations of GA3, which do not affectthe length measured after 6 d when the extension of hypocotylalmost ceases, remove the lag period of 48 h which precedesextension, and prolong the high rate of elongation. FollowingGA3 supply the hypocotyl length in the dark and in the lightdoes not differ until 48 h; thereafter the rate of elongationin the light is less, so that the final length of the hypocotylis 40 per cent shorter than that of the dark-grown seedlingswithout GA3. IAA supplied apically to light-grown seedlings induces a weakpromotion at a concentration of 1 mg l–1 only. With anincreasing concentration of GA3 supplied simultaneously, theconcentration of IAA inducing a significant promotion decreases.A combined supply of both these regulators, however, does notrestore the light-mediated inhibition of hypocotyl elongationcompletely. The maximum decrease in hypocotyl length induced by the growthretardants AMO-1618, CCC, and B-9 supplied from the beginningin the dark does not exceed 70 per cent. Saturating doses ofGA3 supplied in combination with any one of the retardants compensateonly a fraction of the decrease. The results have been interpreted to show that native GAs arenot involved in extension growth in the dark.  相似文献

Four 13-hydroxygibberellins, gibberellin A1 (GA1), 3-epi-GA1,GA19 and GA20 were identified by full-scan GC/MS in extractsof lettuce seedlings (Lactuca sativa L. cv. Grand Rapids). Theresults suggest that the early-13-hydroxylation biosyntheticpathway to GA1 functions in the lettuce seedlings. It was alsofound that GA1 is active per se in the control of hypocotylelongation in lettuce seedlings. To investigate the relationshipbetween control by light of hypocotyl elongation and levelsof endogenous GAs in lettuce, endogenous levels of GAs werequantified by radioimmunoassay in seedlings that had been grownfor 5 days in the dark (5D) and in those that had been grownfor 4 days in the dark and then under white light for 1 day(4D1L). The endogenous level of GA1 in the upper and lower partsof hypocotyls in 5D seedlings was about four times higher thanthat in 4D1L seedlings. The response of explants (hypocotylsegments with cotyledons) from dark-grown seedlings to GA1 isknown to be similar in the dark and under white light when theexplants are treated with inhibitors of the biosynthesis ofGA. Therefore, the above information suggests that the highlevel of GA1 in hypocotyls of dark-grown seedlings is responsiblefor the rapid elongation of hypocotyl, while irradiation bywhite light decreases the endogenous level of GA1 in the hypocotylswith a resultant decrease in the rate of hypocotyl elongation. The interaction of light, gibberellic acid (GA3), and phlorizinin the growth of lettuce (Lactuca sativa L. cv. ‘GrandRapids’) hypocotyls was investigated. At all concentrationsof GA3, phlorizin enhanced GA3-induced growth at luminous intensitiesabove 50 ft-c (continuous light). Without GA3, phlorizin hadno effect on hypocotyl growth in the light but it inhibitedgrowth in the dark. Both seedlings and hypocotyl sections respondedto phlorizin in the presence of GA3. There was no iteractionbetween phlorizin and KCl. Water-growth was severly inhibitedby light. GA3,-induced growth was slightly inhibited by light,and then only at luminous intensities above 50 ft-c. Thus, relativeto H2O-growth, GA3-induced growth increased with increasingluminous intensity up to 450 ft-c, where it reached saturation.It seems that a synergism may exist between light and GA3 aswell as between phlorizin and GA3. Germination and growth of wild-type and two mutant strains (aux-1and Dwf) of Arabidopsis thaliana L. have been examined. Seedlingsof aux-1 exhibit agravitropic roots whereas Dwf display bothagravitropic roots and shoots. Wild-type seedlings retained the seed coat at the root-hypocotyltransition zone and developed hypocotyl hooks. In contrast,aux-I and Dwf seedlings did not retain their seed coats andlacked hypocotyl hooks. A positive gravitropic response of theroots was essential for the retention of the seed coat at theroot—hypocotyl transition zone by the attachment of roothairs to the seed coat. The development of the hypocotyl hookwas aided by the retention of the seed coat. The apical regionof the hypocotyl apparently remained agravitropic during formationand maintenance of the hypocotyl hook. Individuals ofArabidopsis thaliana, collected in different naturalpopulations, were grown in controlled and elevated CO2in a glasshouse.Following germination, root growth of progeny of different linesof these populations was studied in control and elevated atmosphericCO2. No significant direct effect of atmospheric CO2concentrationcould be demonstrated on root growth. An important parentaleffect was apparent, namely that root length and branching weredecreased in seeds collected from a mother plant which had beengrown in elevated CO2. This was correlated with smaller seeds,containing less nitrogen. These parental effects were geneticallyvariable. Variation in osmotic adjustment (OA) among chickpea (Cicer arietinumL.) cultivars has been observed when exposed to terminal drought,but some studies suggest that this benefits yield while otherssuggest it does not benefit yield in water-limited environments.In the present study, parents differing in OA were crossed anda set of advanced breeding lines (ABLs) developed for yieldtesting. The variation in OA during podding was measured underterminal drought in the F2, F3, F7, and F8 progeny and in theparents by either rehydrating the leaves before sampling forosmotic potential (OP) or by measuring the relative water content(RWC) and OP on adjacent leaves for the calculation of the OPat full turgor. Yields were measured in the F8 progeny underterminal drought in Australia and India. While differences inOA were measured in the chickpea lines and parents, OA variedfrom year to year and did not consistently benefit yield whenmeasured in the field under terminal drought. In Australia,differences in OA were not associated with any yield benefitin any year, while in India early flowering resulted in higheryields at three of the four sites, and OA had an inconsistenteffect on seed yields. A comparison of OP at full turgor measuredafter rehydration and from measurements of RWC and OP showedthat the rehydration technique underestimated OA. The high osmotic concentration of the viscid layer and non-volatilegermination inhibitors cannot account for the prolonged dormancyof the seeds of Amyema preissii while they remain within theintact fruit. Once excised, however, germination could not beprevented by depriving the seed of an external supply of water,oxygen or light. Elongation of the hypocotyl, bearing the primaryhaustorium of the young seedling, was enhanced by conditionsthat favoured photosynthesis as well as respiration. Imbibition of seeds of oil seed rape (Brassica napus cv Jetneuf)in 10–3 M aminoethoxyvinylglycine (AVG) or 10–2silver thiosulphate (Ag+) had no effect on germination nor onthe emergence of seedlings from uncompacted or lightly compressedsoil, but significantly reduced emergence from moderately compressedsoil of 68.4 or 143.3 N cm–2 impedance. Exertion of force by emerging control seedlings against a staticcantilever bar fitted with strain gauges reached a maximum (Fmax)of 6 g over 10 h. Higher axial forces were achieved when theseedlings were emerging from compressed soil, without any changein the time required to reach Fmax, so that the build-up offorce was considerably (1.8 fold) faster than in uncompressedsoil. This adaptive response to soil impedance was modified by theseed pretreatments employed. Seedlings from AVG or Ag+pretreatedseeds produced lower axial forces than controls, and neitherFmax nor the rate at which force developed showed any responseto soil compression. After pretreatment in 10–3 ethephon or 10–4 naphthaleneacetic acid (NAA) the seedlings achieved similar Fmax to controlseedlings, but responded more rapidly to soil compression sothat the rate of build up of emergence force was 2.3 fold (NAA)or 2.8 fold (ethephon) faster in compressed than in uncompressedsoil. The results suggest that the exertion of force by a seedlingagainst a barrier involves a dynamic response to impedance onthe part of the seedling. This response can be either enhancedor suppressed by substances which affect ethylene productionor ethylene action. Such compounds may have promise for modifyingseedling emergence from impeding soils. Using detached leaves, two cultivars of pearl millet [Pennisetumamericanum (L.) Leeke], B282 and Serere 39, were assessed forvariation in the capacity to accumulate ABA in response to waterstress. Significant differences in ABA accumulation were detectedbetween cultivars and between different inbred lines withina cultivar, but within lines there was much less variation inthis character. In crosses between individual lines of B282(low ABA) and Serere 39 (high ABA), ABA accumulation in theF1 was mid-way between parental values, indicating additivegenetic control and lack of dominance. Selfed progeny of a B282 x Serere 39 cross were selected forcontrasting ABA accumulation in the F2 to F4 generations. Asixfold range in ABA accumulation was found amongst 207 F2 progeny.This increased to nearly ninefold at F3 and F4. Regression analysisindicated high heritability of ABA accumulation and rapid approachto homozygosity. As the cross studied involved a dwarf (B282) and a tall (Serere39) parent, segregation occurred for height as well as for ABA,though not entirely independently. Tall F3 progeny had significantlyhigher ABA contents than dwarf progeny and high ABA was thereforeassociated with other traits (e.g. large leaves, high leaf percent d. wt) characteristic of tall plants. Nevertheless, therewas a substantial range of ABA content within both groups whichwas uncorrelated with height and other characters. The potential use of the selections in studies on drought responseis briefly discussed. Hypocotyl elongation under white fluorescent light was aboutdoubled in dwarf watermelon (Citrullus lanatus0 (Thunb.) Matsu.and Nakai) seedlings treated with 0.1 to 0.3 µg apicaland 3 x 10–6 to 10.3 M root applications of 6-benzyladenine(BA). BA-enhancement of growth occurred primarily during thefirst 48 h after treatment. Increased hypocotyl length in BA-treatedseedlings was attributed more to longer cells than to an increasein cell number. Early hypocotyl growth of normal seedlings wasalso significantly enhanced by BA although final hypocotyl lengthwas not substantially affected. Benzyladenine caused expansion of cotyledons and, at higherdoses, lateral expansion of hypocotyls. BA-induced increasesin fresh weight of cotyledons and hypocotyls were accompaniedby an increase in dry weight of hypocotyls at the expense ofroots which had less dry matter than untreated seedlings.  相似文献

The anatomy of wood was studied inQuercus roburL. andQuercussuberL. seedlings exhibiting 3–8 units of extension, eachwith a tier of photosynthesizing leaves in their upper parts,generated as the result of rhythmic shoot growth under favourableconditions. At all the axis levels examined (i.e. the base ofeach of the different units of extension, four other equidistantlevels within the first unit of extension and the upper partof the taproot), the wood displayed rings when treated withWiesner reagents. This indicated cinnamaldehyde groups presentin lignins. No rings appeared when the Maüle reaction wasused for specific detection of syringyl subunits in lignins.A trend towards a periodical arrangement of xylem parenchymabands was also found when sections were treated with I2/KI.The number of rings coincided with the number of leaf tiersabove the level of measurement, and did not vary inside thefirst unit of extension. Thus, the rings are called rhythmicgrowth rings. In sections of the first and the second unitsof extension, and in the taproot, the area and width of a givenrhythmic growth ring were highly correlated with the total areaof leaves present above the level of measurement at the presumedtime of growth ring formation. Moreover, stem diameter at thebase of the units of extension was highly correlated with theleaf area above. These results indicate that differentiationof xylem, particularly its lignification, varies rhythmicallyin oak seedlings. Growth of seedling oil palms (Elaeis guineensis), in full daylightand under three levels of shade, was studied using growth analysistechniques. In full day-light, net assimilation rates (EA) betweeno.15 and 0.32 g./dm.2/week were obtained associated with lowleaf-area ratios (F) giving relative growth-rates (RW) rangingfromI I.8 to 3.2 per cent, per day. There were no indicationsof seasonal differences within the small range of values found. The plants take about 90 days to adapt to shade conditions becausethe mean plastochron is 24 days, and shading effects are beststudied on plants grown since germination under the shade treatments.Very different pictures of response to shade were obtained usingplants grown initially in full light followed by 90 days' shadebefore sampling compared with plants grown under shade sincegermination. In the latter, except at the lowest light levelused, I I.I per cent, of full day-light, there was very littleeffect of light on EA or Rw, although the F values decreasedas light increased. Extrapolation of the F values to the extinctionpoint gave values similar to those obtained in another experimenton the effect of a number of shade levels on F. The physiological and ecological implications of these findings,particularly the low growth-rates and shade tolerance, are discussed.  相似文献

The effects of low temperature and the Rht3 dwarfing gene onthe dynamics of cell extension in leaf 2 of wheat were examinedin relation to gibberellin (GA) content and GA-responsivenessof the extension zone. Leaf 2 of wild-type (rht3) wheat closelyresembled that of the Rht3 dwarf mutant when seedlings weregrown at 10C. The maximum relative elemental growth rate (REGR)within the extension zone in both genotypes was lower at 10Cthan at 20C, but the position with respect to the leaf basewas unaffected by temperature. The size of the extension zoneand epidermal cell lengths were similar in both genotypes at10C. Growth at 20C, instead of 10C, increased the lengthof the extension zone beyond the point of maximum REGR in thewild type, but not in the Rht3 mutant. Increasing temperatureresulted in longer epidermal cells in the wild type. Treatingwild-type plants at 10C with gibberellic acid (GA3) also increasedthe length of the extension zone, but the Rht3 mutant was GA-non-responsive.However, the concentrations of endogenous GA1 and GA3 remainedsimilar across the extension zone of wild-type plants grownat both temperatures, despite large differences in leaf growthrates. The period of accelerating REGR as cells enter the extensionzone, and the maximum REGR attained, are apparently not affectedby GA. It is proposed that GA functions as a stimulus for continuedcell extension by preventing cell maturation in the region beyondmaximum REGR and that low temperature increases the sensitivitythreshold for GA action. Plant dry matter (DM) partitioning, survival rates, stem anatomy,and stem water conductivity were investigated in wild-type (WT)and long-hypocotyl (lh) mutant seedlings of cucumber (Cucumissativus) grown as isolated individuals under natural radiation.The lh mutant is severely deficient in phytochrome B. Wild-typeseedlings accumulated more DM than lh seedlings over a 4-weekgrowth period in the glasshouse. Leaf and root DM were higherin the WT but stem DM was higher in the lh mutant. Stem DM perunit length was larger in WT than in lh mutant seedlings, evenwhen the two genotypes were compared at equal whole plant DM,which was achieved by growing the plants under different irradiance.In WT seedlings, the hypocotyl was shorter but thicker, withlarger average cell diameter than the lh mutant. In hypocotyltransverse sections the area occupied by load-bearing tissues(xylem and phloem fibres) and the number and diameter of xylemvessels were larger in WT than lh seedlings. Survival ratesof the lh mutant were normal in the glasshouse but very lowoutdoors due to hypocotyl fracture. The water conductivity ofhypocotyl sections was higher in WT than lh seedlings, but nosignificant differences in water conductivity were observedwhen the root remained attached to the hypocotyl. Seeds of two lettuce cultivars (Lactuca sativa L., cv. Meikoninginand cv. Grand Rapids) were hermetically stored with constantmoisture contents ranging between 3.6 and 17.9 per cent (freshweight basis) at constant temperatures ranging between 5 and75 °C. The decline with time in percentage germination andpercentage normal seedlings was determined for each storagetreatment. The data were fitted to an equation which containsthe constants: K1, the probit of the initial percentage germinationor normal seedlings; KE, a species constant; CW, the constantof a logarithmic moisture term; CH, the constant of a lineartemperature term and CQ, the constant of a quadratic temperatureterm. Regression analysis of data from storage periods up to5.5 years at temperatures of 5–75 °C and seed moisturecontents of 3.6–13.6 per cent yielded the following values:KE= 8.218, CW=4.797±0.163, CH=0.0489±0.0050 andCQ=0.000365±0.000056. Although this equation consistentlyprovided a better fit, simplified equations, assuming eithera log-linear relationship between seed longevity and temperature,or a log-linear relationship between seed longevity and bothmoisture content and temperature, accounted for more than 94per cent of the variation at the restricted temperature rangeof 5–40 °C. Longevity of the same seed lots at sub-zero temperatures (–5,–10 and –20 °C) was studied in separate tests.Freezing damage, resulting in abnormal seedlings in the germinationtest, occurred at –20 °C when the moisture contentof the seeds exceeded 12 per cent. No decline in percentagenormal seedlings was observed after a storage period of 18 monthsor longer at –20 °C, provided the seed moisture contentdid not exceed 9.5 per cent. For seeds stored at –5 and–10 °C with 9.6–12.5 per cent moisture content,the observed rate of decline of percentage normal seedlingswas adequately predicted by the viability equation, using theabove values for the constants. This suggests that for low moisturecontents the viability equation can be applied to estimate longevityat sub-zero temperatures. The tomato fruit was compared to a sphere with a radius R. Radialgrowth rates in the fruit (FIW and FID) due to water importor to dry matter import, respectively, which are also the waterimport rate or dry matter import rate per unit surface areaof fruit, were calculated from two sets of published results.This data referred to fruits which swelled in such a way thatthe availability of assimilates had little effect on growth.Two varieties differentiated the two series of results and inone series, three trials were differentiated by the salinityof the nutrient solution. In all trials, it was found that FIW and FID decreased whenR increased. Two phases were observed for FIW: after a firstphase, FIW decreased more quickly and almost linearly when Rincreased. FID was constant or decreased with respect to R.Except at the beginning of growth at the greatest salinity,there were clearly linear regressions between FIW and FID suchas FID = aFIW-b; where b was lower with higher salinity. Thechanges of the concentration of imported dry matter (FID/FIWwere examined in terms of R and FIW/R. The mechanisms controllingthe changes in FIW and FID were discussed. The second leaf of wheat was used as a model system to examinethe effects of the Rht3 dwarfing gene on leaf growth. Comparedto the rht3 wild type, the Rht3allele decreased final leaf length,surface area and dry mass by reducing the maximum growth rates,but without affecting growth duration. Gibberellic acid (GA3)increased final leaf length and maximum growth rate in the rht3wild type, but was without effect on the Rht3 mutant, whichis generally regarded as being non-responsive to gibberellin(GA). Paclobutrazol, an inhibitor of GA biosynthesis, decreasedfinal leaf length and maximum growth rate in the rht3 wild typeto values similar to those in the untreated Rht3 mutant. NeitherGA3 nor paclobutrazol affected the duration of leaf growth.The decrease in leaf length was produced by reduction of celllength rather than cell number. The maximum relative elementalgrowth rate (REGR) for cell extension was essentially the samein all treatments, as was the time between the cells leavingthe meristem and achieving maximum extension rate. The differencesbetween the genotypes and treatments were all almost entirelydue to differences in the time taken from the attainment ofmaximum REGR of cell extension to the cessation of extension.This was reflected in the length of the extension zone, whichwas approximately 6–8 per cent of final leaf length. Theeffects of the Rht3 allele, GA3 and paclobutrazol all appearto be on the processes which promote the cessation of cell elongation. Lolium temulentum seedlings were grown on a nutrient mediumcontaining NH4NO2 at 0, 0·1, 0·5, 1·0 and4·3 mmoll–1 as the sole N source. Relative andabsolute extension rates, maximal leaf size, duration of extensiongrowth, rate of leaf appearance and plastochron index were determinedfrom the parameters of Richards functions fitted to lengthsof laminae measured at intervals after sowing. The final lengthof leaf I was relatively insensitive to N whereas mean relativeextension rate was increased and duration of growth decreasedwith increasing NH4NO2 concentration. Leaves 2 and 3 enlargedprogressively with N at concentrations up to 1·0 mmoll–1but were unresponsive thereafter. There was no significant correlationbetween final length and mean relative extension rate for leaves1 to 3. Leaves 4 to 6 continued to show increasing length beyond1·0 mmoll–1 N and final length was significantlycorrelated with mean relative extension rate. Increasing N increasedthe rate of leaf appearance by decreasing the duration of leafextension and plastochron. These results are discussed in relationto the control of leaf and N turnover. The effect of ultraviolet A (UVA) on growth and photosyntheticrate was studied in diatoms (Melosira spp.) of the phytoplanktonof a eutrophic lake and a cultured green alga Chloretla ellipsoidea.The cells were incubated under photosynthetically active radiation(PAR) (–UVA) or PAR + UVA conditions (+UVA). Growth ofC.ellipsoidea was retarded under +UVA, as shown by an increasein the lag period, but the rate of exponential growth was almostthe same in + and –UVA conditions. The photosyntheticrate was depressed markedly by UVA in Chlorella cells grownunder –UVA. In contrast, cells grown in +UVA showed onlyslight inhibition by UVA and after exposure to UVA for 6 daysthere was no inhibition. During the growth experiment, the cellularchlorophyll a content was higher in +UVA than +UVA grown cells.A similar effect was observed in diatoms from the eutrophicLake Suwa. In vivo fluorescence with (Fa) and without 3-(3,4-dichloropheny)-l,l-dimethylurea (DCMU) (Fb) and the photosynthetic rate were measured forC.ellipsoidea and the diatoms for 5 h under + and –UVAconditions. Soon after C.ellipsoidea had been subjected to +UVA,Fb and Fa / Fb decreased quickly and reached minima after 40min and 1 h, respectively. The suppressed in vivo fluorescenceresumed and full recovery was achieved after 4 h. This suggeststhat reactivation of the photosystem is acquired under prolongedexposure to UVA. A similar shift of Fa + Fb, but no change inFb, was found in diatoms by exposure to UVA. Changes in photosyntheticoxygen evolution by C.ellipsoidea under +UVA were similar tochanges in Fa + Fb. Degradation of chlorophyll a extracted inmethanol was enhanced by UVA. The rate of degradation by UVAwas independent of temperature from 15 to 34°C, suggestinga photochemical reaction. The results indicate that C.ellipsoideaand Melosira spp. acclimatize to prolonged UVA exposure by reactivationof the photosystem and enhanced cellular chlorophyll a synthesis.The ecological importance of these results to phytoplanktonproductivity in natural aquatic environments is discussed.  相似文献

 以砂培菊芋(Helianthus tuberosus)幼苗作为试验材料,分别进行不同浓度NaCl (50、 100、150、200、250 mmol&;#8226;L-1)和Na2CO3 (25、50、 75、100、125 mmol&;#8226;L-1)胁迫处理,以1/2全营养液作为对照,处理7 d后研究NaCl和Na2CO3胁迫处理对菊芋幼苗叶片光合作用及叶绿素动力学 参数的影响。结果表明:1)在NaCl处理下,当浓度小于150 mmol&;#8226;L-1时,增加了菊芋的叶绿素含量、净光合速率(Net photosynthetic rate, Pn)和气孔导度(Stomatal conductivity, Gs),对荧光参数PSⅡ的电子传递情况( Fm/Fo)、PSⅡ原初光能转换效率(Fv/Fm)、PSⅡ量子效率 (Actual quantum yield of PSⅡ under actinic irradiation,φPSⅡ)和光化学猝灭系数(Photochemical quenching coefficient, qP)和非 光化学猝灭系 数(Non-photochemical quenching coefficient, NPQ)没有显著影响,随着浓度的增加,各项生理指标与对照相比除了NPQ显著 增加,其余均显著降低;2)在Na2CO3胁迫处理下,随着Na2CO3浓度的增加,与对照相比菊芋幼苗叶绿素含量、Pn、Gs以及叶绿素a荧光诱导动力 学参数Fm/Fo、Fv/Fm、φPSⅡ和qP均显著降低,NPQ显著增加;3)就NaCl和Na2CO3相比而言,在相同Na+浓度情况下,处于Na2CO3胁迫下的菊芋 幼苗的叶绿素含量、Pn、Gs以及叶绿素a荧光诱导动力学参数Fm/Fo、Fv/Fm、φPSⅡ和qP下降幅度和NPQ的增加幅度均显著大于NaCl,这说明 NaCl和Na2CO3胁迫均对菊芋幼苗造成不同程度的伤害,但在相同Na+浓度情况下,Na2CO3的伤害程度大于NaCl。由此说明菊芋对盐的忍耐程度高 于碱。  相似文献
