This study was conducted to evaluate the inhibitory potential of P. harmala leaf, stem and root extract on germination and growth of Avena fatua L. and Convolvulus arvensis L., as well as identification of the phytotoxic substances responsible for this activity. According to our results, the degree of toxicity of different P. harmala plant parts can be arranged in the following order: leaves > stems > roots. The two test species differed in their sensitivity to P. harmala extracts. Inhibitory effect on shoot length and seedling dry weight was more pronounced in C. arvensis, whereas higher reduction in germination, root length and total chlorophyll content occurred in A. fatua. A significant amount of water-soluble phenolic acids were found in P. harmala plant extracts. Total phenolic acids content was higher in leaf extracts when compared to that of stem or root extracts. Seven phenolic acids including gallic acid, vanillic acid, 4-hydroxybenzoic acid, 3,4-dihydroxybenzoic acid, caffeic acid, syringic acid and ferulic acid were found in P. harmala leaf extracts. On the other hand, we identified four phenolic acids from stem (galllic acid, vanillic acid, 3,4-dihydroxybenzoic acid and caffeic acid) and root (galllic acid, 4-hydroxybenzoic acid, syringic acid and cinnamic acid) extracts. The greater number of growth inhibitors detected in the leaves might explain the stronger inhibitory activity. Overall, our results suggest that P. harmala might be used as a natural herbicide for weed control and consequently reduce dependence on synthetic herbicides.  相似文献

Peganum harmala L. is a perennial herbaceous plant and can be a future drug due to its wide medicinal purposes. Despite its economic importance, the molecular genetics of P. harmal have not yet been studied in detail. Genetic diversity of 12 P. harmala genotypes were investigated by using Inter-Simple Sequence Repeats (ISSR), PCR-RFLP of rDNA-ITS, PCR-SSCP of rDNA-ITS and Simple Sequence Repeat (SSR) markers. The level of polymorphism revealed by ITS-SSCP is the lowest, followed by ITS-RFLP then ISSR and the highest polymorphism level was reported for SSR marker. The AMOVA analysis implied that most of the variation occurred within the Populations. A value of inbreeding coefficient Fis estimated by the three co-dominant markers was nearly equal and offer an indication of the partial out-crossing reproductive system of P. harmala. Principal Coordinate Analysis (PCOA) plot revealed a clear pattern of clustering based on the locations of collected plants which coincide with the isolation by distance. The study revealed that ITS-SSCP and ISSR markers respectively were more informative than the other used markers in the assessment of genetic diversity of P. harmala. The results reflect the great diversity of P. harmala and data obtained from this study can be used for future collecting missions.  相似文献

In the current study; insecticidal, growth regulation, oviposition deterrence and repellency of petroleum ether extracts of Azadirachta indica, Penganum harmala, Datura stramonium, Tribulus terrestris and Chenopodium murale against 2nd instar larvae of housefly was investigated. Five different concentrations (5%, 10%, 15%, 20% and 25%) were used through larval feeding and the mortality data was recorded after 24, 48 and 72 hrs. Highest mortality was induced by P. harmala (63.87%) followed by D. stramonium (62.78%), A. indica (53.84%), T. terrestris (41.86%) and C. murale (4.09%) after 72 h at 25% concentration, respectively. Increased mortality was observed with increased time duration and concentration. Longest larval duration (9.33 ± 0.33 days) and pupal duration (7.33 ± 0.33 days) days) was recorded in larvae treated with 25% concentration of P. harmala which also caused a decrease in the activity of AChE, ACP, AKP, α-Carboxyl, and β-Carboxyl enzymes. However, at 25% concentration, C. murale showed highest oviposition deterrence activity (81.88%) followed by D. stramonium (79.58%). In comet assay test, at highest concentration (25%) the mean comet tail lengths represented by Penganum harmala, Datura stramonium and Azadirachta indica (Reference plant) were 10.20 ± 0.49, 9.20 ± 0.37 and 7.80 ± 0.49 μm while percent DNA damage was 10.56 ± 0.77, 10.67 ± 1.62 and 8.11 ± 0.85% respectively compared to controls cells. Phytochemical analysis indicated the presence of flavonoids, steroids, saponins, cardiac glycosides, tannins, alkaloids, terpenoids and anthraquinones. Fourier Transform Infrared spectroscopy (FTIR) analysis revealed the presence of phenolic flavonoids, saponins, tannins as major functional groups. Further studies are needed to explore and thus, to incorporate weed plant extracts for the management of house flies.  相似文献

Halogeton glomeratus (M. Bieb.) C.A. Mey., Lepidium latifolium Linn. and Peganum harmala Linn. are distributed in temperate salt playa habitats of Upper Hunza, Pakistan. Seeds were germinated under various salinity (0–500 mM NaCl), light (12 h-light:12 h-dark and 24 h-dark) and temperature (5/15, 10/20, 15/25, 20/30, and 25/35 °C, dark/light) regimes for 20 days to determine the optimal conditions for germination and recovery of seeds from these factors when exposed to less than optimal conditions. Seeds that failed to germinate in dark were transferred successively to 12 h-photoperiod, salinity to distilled water and from various temperature regimes to 20/30 °C, to determine the effect of these stresses and the ability of these seeds to recover respectively. Highest seed germination (H. glomeratus and L. latifolium: 100%; P. harmala: 80%) was obtained in non-saline control at 20/30 °C in 12 h-photoperiod, however, increase in salinity progressively inhibited seed germination. Seed germination of H. glomeratus and P. harmala was substantially inhibited and that of L. latifolium was prevented in dark. Salinity and dark treatments have a synergistic effect in inhibiting seed germination of all species. No seed of any species germinated at 5/15 °C; germination was substantially inhibited at 25/35 °C both for H. glomeratus and P. harmala while L. latifolium failed to germinate at 25/35 °C. Rate of germination also decreased with an increase in salinity at all temperature regimes but this effect was minimal at optimal temperature regime of 20/30 °C. After successive elimination of light, salinity and temperature stresses, final seed germination was identical to respective controls. The results indicate that seeds of these temperate halophytes could endure environmental stresses without losing viability and germinate readily when these stresses are removed. Under the extremely variable conditions of the playa habitat these species are highly opportunistic exploiting the windows of opportunity available during spring or early summer.  相似文献

The inhibitory effect of phenolic compounds and alkaloids of Inonotus hispidus and Peganum harmala on Candida rugosa lipase was investigated, also, their antioxidant activities using DPPH, ABTS and phosphomolybdenum were studied in this paper. The phenolic extracts have shown a stronger antiradical activity than the alkaloids extracts. The enzymatic inhibition produced by these extracts is described here for the first time. The results have shown that the phenolic and the alkaloid extracts are good inhibitors of C. rugosa lipase. Thus, the inhibitor molecules (harmaline and hispidin) have been isolated from P. harmala and I. hispidus. Their structures were elucidated by 1H NMR analysis. Molecular docking has been achieved using AutoDock Vina program to discuss the nature of interactions and the mechanism of inhibition. Therefore, these isolated molecules could be used in the treatment of candidiasis.  相似文献

The use of traditional medicines as a diuretic agent has been increasing in recent years. The diuretic activity of a number of plant extracts used as diuretic agents in ethnomedicine has been confirmed in experimental animals. However, despite the widespread use of Peganum harmala in traditional medicine, there is a paucity of data supporting its use as a diuretic agent. Therefore, the present study aimed to envisage the true effect and magnitude of diuresis of methanolic extract of P. harmala (MEPH) in comparison with a well-known diuretic drug furosemide using Wistar albino rats. MEPH was administered orally in three different doses (150, 300 and 450 mg/kg) to experimentally dehydrated rats. Furosemide (10 mg/kg orally) was used as a reference drug. The diuretic effect of the MEPH was evaluated by measuring urine volume, urine pH, urinary electrolyte levels, natriuretic and saliuretic effects. The urine volume (in mL) measured at 5 h and 24 h and electrolyte excretion (Na+, K+, and Cl) at 24 h duration were measured. The urine output and urinary electrolyte excretion were found to be significantly higher in rats treated with MEPH as compared to normal rats in a dose dependent manner (P < 0.05). The results of our study were comparable to furosemide drug. Based on observed results, we can recommend that P. harmala may be an effective diuretic, however, toxicity studies should be conducted before administration.  相似文献

Fatty acid contents of the Peganum harmala plant as a result of hexane extraction were analyzed using GC–MS. The saturated fatty acid composition of the harmal plant was tetradecanoic, pentadecanoic, tridecanoic, hexadecanoic, heptadecanoic and octadecanoic acids, while the saturated fatty acid derivatives were 12-methyl tetradecanoic, 5,9,13-trimethyl tetradecanoic and 2-methyl octadecanoic acids. The most abundant fatty acid was hexadecanoic with concentration 48.13% followed by octadecanoic with concentration 13.80%. There are four unsaturated fatty acids called (E)-9-dodecenoic, (Z)-9-hexadecenoic, (Z,Z)-9,12-octadecadienoic and (Z,Z,Z)-9,12,15-octadecatrienoic. The most abundant unsaturated fatty acid was (Z,Z,Z)-9,12,15-octadecatrienoic with concentration 14.79% followed by (Z,Z)-9,12-octadecadienoic with concentration 10.61%. Also, there are eight non-fatty acid compounds 1-octadecene, 6,10,14-trimethyl-2-pentadecanone, (E)-15-heptadecenal, oxacyclohexadecan-2 one, 1,2,2,6,8-pentamethyl-7-oxabicyclo[4.3.1]dec-8-en-10-one, hexadecane-1,2-diol, n-heneicosane and eicosan-3-ol.  相似文献

This article reviews our present knowledge on the formation of tryptophan derived secondary metabolites in tissue cultures of Peganum harmala. With the presence of -carboline alkaloids and serotonin, P. harmala contains two rather simple, interrelated biosynthetic pathways. The long term disadvantage of low and unstable productivity of P. harmala suspension culture has recently been overcome by establishing highly productive hairy root cultures. The first -carboline alkaloid biosynthetic enzymes, specific for the O-methylation of harmalol and harmol as well as for the oxidation of harmaline to harmine, have been detected in these cultures, and they should thus provide a suitable source for studying the yet unknown initial two enzymatic steps of -carboline alkaloid biosynthesis. Seedlings of P. harmala have also been successfully transformed with constructed strains of Agrobacterium, as demonstrated by the overexpression of a tryptophan decarboxylase gene from Catharanthus roseus in cultures of P. harmala. In such transgenic cultures a large overproduction of serotonin was observed. The relative simplicity of these pathways and the rather easy handling of the cultures could make P. harmala a useful and attractive model system for studying the interaction, regulation and manipulation of secondary pathways in cultured cells.Abbreviations TDC tryptophan decarboxylase - tdc gene of tryptophan decarboxylase  相似文献

Nitraria retusa (Forssk.) Asch. was subjected to botanical and chemical studies and compared against four selected taxa of Zygophyllaceae viz., Fagonia arabica L., Peganum harmala L., Tribulus terrestris L. and Zygophyllum album L.f. The botanical section included the study of morphological and anatomical characters of both vegetative and reproductive organs using LM and scanning electron microscope. The chemical section included a detailed study of the flavonoids present in N. retusa. Twelve compounds were isolated and identified as flavone, flavonol O-glycosides and flavone C-glycosides. Flavonoid patterns from N. retusa (Forssk.) Asch. of the selected taxa Zygophyllaceae were surveyed through HPLC and correlated with morphological and anatomical criteria.  相似文献

The present study showed the toxicity caused by heavy metal and its detoxification responses in two desert plants: perennial Peganum harmala and annual Halogeton glomeratus. In pot experiments, 1-month-old seedlings were grown under control and three levels of combined heavy metal stress. Seedling growth as well as heavy metal accumulation, antioxidative enzymes [superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX)] activities and the contents of malondialdehyde (MDA), and hydrogen peroxide (H2O2) in leaves was examined after 2 months of heavy metal exposure. Compared with H. glomeratus, growth of P. harmala was more severely inhibited. In leaves, the heavy metal accumulation pattern in both the plants was dose-dependent, being more in H. glomeratus. H. glomeratus exhibited a typical antioxidative defense mechanism, as evidenced by the elevated activities of all the three enzymes tested. P. harmala exhibited a different enzyme response pattern, with a significant reduction in CAT activity, and elevated SOD and APX activities, but significantly elevated APX activity was only at the lowest heavy metal concentration. MDA and H2O2 contents were significantly enhanced in leaves of heavy metal-treated P. harmala, but in H. glomeratus were elevated only at the highest heavy metal treatment. These results indicated that H. glomeratus had a greater capacity than P. harmala to adapt to oxidative stress caused by heavy metal stress, and antioxidative defense in H. glomeratus might play an important role in heavy metal tolerance.  相似文献

Peganum harmala L. is a traditional Chinese and Uygur medicine used to treat cancer. Bioactivity‐guided fractionation was applied to determine the cytotoxic constituents from P. harmala. A novel triterpenoid and a phenolic glycoside were isolated and identified, as well as seven known compounds. The novel metabolites were elucidated to be 3α‐acetoxy‐27‐hydroxyolean‐12‐en‐28‐oic acid methyl ester ( 1 , OA) and N‐acetyl‐9‐syringinoside ( 9 ). Some compounds exhibited potent cytotoxicity against human tumor cells. Among them, OA showed the highest cytotoxicity against human lung cancer cells A549 with an IC50 value of 8.03 ± 0.81 μm . OA had a potent anti‐NSCLC cell activity by interfering with the epidermal growth factor receptor (EGFR) activation and its downstream signaling, and could exert an antiproliferative effect by inactivation of EGFR‐driven antiapoptotic pathway followed by the release of mitochondrial cytochrome c, which might prove to be a promising leading compound for the development of an anti‐lung cancer drug.  相似文献

Feeding and trapping experiments to Peganum harmala callus cultures were limited by compartmentation; exogenous substrates were detoxified by precipitation, presumably as polymers or conjugates, or by conversion to water-soluble products, such as phenols and glucosides, easily stored in vacuoles. Alkaloid-producing and non-alkaloid-producing callus cultures were readily able to convert tryptamine to 5-hydroxytryptamine and harmaline to dihydroruine (8-hydroxyglucosylharmaline). Phenolic substrates, including 5- and 6-hydroxy-tryptophan, 5- and 6-hydroxytryptamine and harmalol, were not metabolized. In alkaloid-producing callus cultures, radioactivity from [methylene-14C]-l-tryptophan and [methyl-14C-]-harmaline was incorporated into harmine. The dilution of radioactivity was 30000- and 2-fold respectively.  相似文献

Summary. Exine ontogeny is studied in five taxa of the family Zygophyllaceae sensu lato (Peganum harmala L., Zygophyllum album L., Fagonia cretica L., Tribulus terrestris L., and Nitraria retusa [Forsk.] Asch.). In the beginning of the tetrad stage, the plasmalemma is attached to the callose wall, except in T. terrestris, where it describes crests and hollows. The primexine matrix is fibrillar, bilayered in T. terrestris and unilayered in the other taxa. In all species except P. harmala, the procolumellae are heterogeneous with clear zones and they become compact later. In T. terrestris, they are built on the crests. In Z. album and T. terrestris, a primordial nexinic lamella is set up. It is tripartite with a white line seen at some levels; on its external leaflet, the foot layer is observed, and on its internal leaflet, there is the endexine with numerous lamellae. This white line disappears often in the mature exine. In T. terrestris, there is a thick nexine that is coarsely lamellate inside. In the aperture zone, the columellae are lacking, the tectum and the foot layer get thinner; they unite and form the apertural membrane with the external part of the endexine. There is a granulolamellar endexinic zone well developed in P. harmala, whereas it is threelayered and weakly developed in T. terrestris. Protozoic infections caused by genus Leishmania pose an enormous public health threat in developing countries, compounded by the toxicity and resistance to current therapies. Under the aegis of our ongoing program on drug discovery and development on antileishmanial agents from plants, we carried out bioassay guided fractionation on Peganum harmala seeds which resulted in the isolation of 1 as an antileishmanial agent. 2D-NMR spectral data and single crystal X-ray crystallography data indicated 1 as peganine hydrochloride in dihydrated form. The compound 1 exhibited in-vitro activity against both extracellular promastigotes as well as intracellular amastigotes residing within murine macrophages in Leishmania donovani. Furthermore, 1 also exhibited in-vivo activity, 79.6 (±8.07)% against established VL in hamsters at a dose of 100 mg/kg b.wt.  相似文献

The aim of this study was to assess the phytotoxic potential of Cleome arabica L, as well as to isolate the main bioactive compounds. Phytotoxicity was evaluated on germination and seedling growth of Lactuca sativa, Raphanus sativus, Peganum harmala and Silybum marianum, through testing aqueous and organic extracts of different C. arabica organs (roots, shoots, siliquae and seeds). Results showed that siliquae methanol extract caused the greatest negative effect on lettuce germination and growth. For the bioactive subfractions (petroleum ether, ethyl acetate and methanol–water), the ethyl acetate induced highly significant reduction, showing 100% inhibition of lettuce growth at 6 g/L. The bioactive ethyl acetate subfraction was chromatographed and subjected to NMR techniques. Based on bio-guided chromatographic fractionation, five bioactive allelochemical compounds were isolated from ethyl acetate extract of siliquae of C. arabica. The most inhibitory compound on lettuce seedling growth was elucidated as 11-α-acetylbrachy-carpone-22(23)-ene.  相似文献

The antimicrobial activity of plant extract of Peganum harmala, a medicinal plant has been studied already. However, knowledge about bacterial diversity associated with different parts of host plant antagonistic to different human pathogenic bacteria is limited. In this study, bacteria were isolated from root, leaf and fruit of plant. Among 188 bacterial isolates isolated from different parts of the plant only 24 were found to be active against different pathogenic bacteria i.e. Escherichia coli, Methicillin-resistant Staphylococcus aureus (MRSA), Enterococcus faecium, Enterococcus faecalis and Pseudomonas aeruginosa. These active bacterial isolates were identified on the basis of 16S rRNA gene analysis. Total population of bacteria isolated from plant was high in root, following leaf and fruit. Antagonistic bacteria were also more abundant in root as compared to leaf and fruit. Two isolates (EA5 and EA18) exhibited antagonistic activity against most of the targeted pathogenic bacteria mentioned above. Some isolates showed strong inhibition for one targeted pathogenic bacterium while weak or no inhibition for others. Most of the antagonistic isolates were active against MRSA, following E. faecium, P. aeruginosa, E. coli and E. faecalis. Taken together, our results show that medicinal plants are good source of antagonistic bacteria having inhibitory effect against clinical bacterial pathogens.  相似文献

Peganum harmala is a herb grows spontaneously in arid and rocky areas. From ancient time, it has been claimed to be an important medicinal plant in rough environment. In this study, we developed 12 microsatellite loci from P. harmala by the combining biotin capture method for the first time. A total of 31 microsatellite sequences were recovered through screening the library and 12 of them are polymorphic. The number of alleles per locus in 36 sampled individuals ranged from 3 to 8, expected heterozygosity and observed heterozygosity ranged from 0.1381 to 0.6821 and from 0.3573 to 0.8739, respectively. In addition, all markers have been crossly checked in the other congeneric species. These microsatellite markers would provide a useful tool for investigating the genetic diversity and study the population genetic structure in detail.  相似文献

Seventeen quinazoline alkaloids and derivatives, containing two pairs of new epimers, named as (S)- and (R)-1-(2-aminobenzyl)-3-hydroxypyrrolidin-2-one β-d-glucopyranosyl-(1?→?6)-β-d-glucopyranoside (1, 2), (S)- and (R)-vasicinone β-d-glucopyranosyl-(1?→?6)-β-d-glucopyranoside (3, 4), and a new enantiomer (12b), together with six known ones (58, 10, and 12a), and three pairs of known enantiomers (9, 11, and 13), were isolated from the ethanol extracts of the seeds of Peganum harmala L.. Their structures including the absolute configuration were elucidated by using 1D and 2D NMR, and ECD calculation approaches. The cytotoxic activities of all isolated compounds were evaluated. 11 showed moderate cytotoxicity against PC-3 cells with an IC50 value of 15.41?μM.  相似文献
