The effects of adrenalectomy on growth, some enzyme activities in the liver and kidney, and urinary excretion of urea, creatinine and creatine were investigated in rats fed the 10% casein diets containing 7% glycine with or without l-arginine and l-methionine (10C, 10C7G and 10C7ArgMet).

Body weight gains of the intact 10C and 10C7GArgMet groups were almost same as the corresponding adrenalectomized groups. The body weight of the adrenalectomized 10C7G group was extremely decreased though that of the intact 10C7G group was maintained almost constant; but the decrease was recovered by the administration of hydrocortisone. The activities of liver arginase and carbamylphosphate synthetase were not affected by those diets. Liver serine dehydratase and ornithine δ-aminotransferase activities were increased in the intact 10C7G and 10C7GArgMet groups, but these increases were depressed by adrenalectomy. Glutamate-pyruvate transminase activities in the liver of intact 10C7G and 10C7GArgMet groups were also enhanced, but were extremely decreased in the corresponding adrenalectomized groups. Kidney transamidinase activity was not affected by adrenalectomy. The amount of urinary excreted urea was almost unchanged by adrenalectomy, but was increased by hydrocortisone administration. The amounts of excreted creatine of the adrenalectomized groups were generally larger than the corresponding intact groups, but slightly decreased by the administration of hydrocortisone. The amount of excreted creatinine was not generally affected by adrenalectomy.  相似文献

The adaptive liver growth response was investigated in intact and adrenalectomized rats. When adult male rats were given a single oral dose of mirex (100 mg/kg body weight) there was a 72% increase in relative liver weight (RLW) in 72 hr. Based on [3H]-thymidine ([3H]TdR) incorporation into hepatic DNA, there was also a wave of DNA synthesis which peaked at 48 hr and decreased to essentially control values by 96 hr post mirex dose. In mirex-dosed adrenalectomised (Adx) animals, the RLW was increased by only 38% and there was sustained DNA synthesis. When mirex-dosed Adx rats were given corticosterone supplements, the RLW response was similar to the RLW response in intact mirex-dosed rats. However, the 48-hr DNA synthesis peak seen in intact mirex-dosed rats was eliminated. From these data it is suggested that mirex-induced adaptive liver growth has two components: a hypertrophic component which is mediated by corticosterone, and a hyperplastic component which is independent of corticosterone.  相似文献

Summary A technique for the organ culture of postnatal and adult rat liver has been developed. Liver slices, 0.3 mm thick, were maintained in Conway units at the interphase between medium and a 95% O2:5% CO2 atmosphere. Postnatal liver in culture for up to 72 h had healthy hepatocytes throughout the explants; if adult liver was used the upper 0.2 mm was healthy after 24 h. These slices incorporated tritiated orotate and leucine into trichloroacetic acid-precipitable material. Incorporation of orotate was shown to be spread over the entire slice of neonatal liver. Culturing did not alter the potassium ion content of postnatal liver. Tyrosine aminotransferase activity in liver slices from postnatal, adult, and adrenalectomized adult rats was stimulated by glucocorticoids and dibutyryl cyclic AMP. Cycloheximide and actinomycin D prevented this response. Further, cortisol exerted a permissive effect on the stimulation of tyrosine aminotransferase activity by dibutyryl cyclic AMP in slices from adrenalectomized rats. Induction of urea cycle enzymes by cortisol was demonstrated in cultures of liver from adrenalectomized adult animals. Deceased October 1, 1983. Low molecular weight polar complexes were shown to be formed in vivo from 3H-aldosterone in both kidney and liver subcellular fractions, the majority being present in the cytosol fractions. Significant differences were observed between the quantities of polar complexes present in kidney subcellular fractions from intact and adrenalectomized male rats and also between the quantities of these kidney polar complexes from spironolactone treated male rats. 3H-aldosterone macro-molecule complexes were shown to exist in appreciable quantities only in the kidney cytosol fractions of adrenalectomized male rats. These gel filtration studies also showed the 3H-aldosterone labeled macromolecule complexes to consist of two protein peaks; one of high molecular weight and the other of lower molecular weight (~50,000 mol. wt.). The amount of 3H-aldosterone labeled protein complexes in kidney cytosol was greatly reduced when adrenalectomized rats were pretreated in vivo with spironolactone.  相似文献

Summary The effect of different phosphorus supplies on the uptake, translocation and accumulation of14C-urea by orchard grass was investigated. Phosphorus starvation inhibits the uptake, translocation and accumulation of the carbon of urea similarly to the nitrogen of urea. As compared with the uptake process the reduction of the accumulation is much more effected by the inhibition of the carbon translocation from roots to the aboveground parts. Lack of phosphorus also decreases the incorporation of the14C of urea into high-molecular compounds. The effect of phosphorus deficit on the accumulation of14C-urea increases with time of starvation.  相似文献

1. Sodium, K and Cl excretion and changes in blood volume were studied after parenteral injection of Cd2+ (1 μmol/100g body weight) in intact and adrenalectomized rats.2. The final systolic arterial pressure and mean arterial pressure of adrenalectomized rats increase significantly after Cd2+ administration compared with values from control group and initial systolic arterial pressure of each group.3. Cadmium administration induced a significant inhibition of Na+ excretion in urine and a decrease in blood volume in ADX rats compared with values from control or sham operated rats.4. The possibility that hypertension induced by Cd may be explained by inhibition of Na excretion and changes in blood volume is discussed.  相似文献

1. The administration of dexamethasone to intact fed rats by intraperitoneal injection for 3h was associated with a 6-fold increase in the time for which mitochondria subsequently isolated from the liver retain a given load of exogenous Ca2+. This effect was blocked by the co-administration of cycloheximide with dexamethasone, and partially blocked by the co-administration of puromycin. Daily administration of dexamethasone for periods of 4--7 days resulted in liver mitochondria that exhibited a decreased ability to retain exogenous Ca2+. 2. When glucagon was administered to fed adrenalectomized rats, the increase in mitochondrial Ca2+-retention time that results from the action of this hormone was reduced by 50% when compared with its effect on intact animals. The administration of dexamethasone to adrenalectomized rats partially restored the full effect of glucagon. 3. Dexamethasone did not enhance the effect of glucagon on mitochondrial Ca2+-retention time when administered to intact fed rats. 4. It is concluded that these data support the hypothesis that the hormone-induced modification of liver mitochondria, which results in an increase in the time for which exogenous Ca2+ is retained, involves a step in which new protein is synthesized.  相似文献

The effects of intravenous administration of PGE1 on the glycogen synthase and phosphorylase system in rat heart were studied.Unlike the consistent effects of PGE1 on glycogen synthase in liver, the response in heart was variable. A significant decrease in the per cent synthase occurred in fasted intact rats while a significant increase was seen in adrenalectomized hydrocortisone treated fasted rats. No significant effect was seen on the synthase system in either fed intact or fasted adrenalectomized rats.Phosphorylase activity was increased significantly following PGE1 administration in fed intact rats and slightly increased in adrenalectomized fasted rats. The phosphorylase system was not affected in fasted intact and fasted adrenalectomized rats given glucocorticoid replacement. With our present state of knowledge an adequate explanation for the response of these heart enzymes to PGE1 under the various conditions of this study does not appear possible.  相似文献

Experiments on Wistar male rats revealed depression of labeled amino acid incorporation into the tissue proteins as a result of swimming. When prolonged swimming was accompanied by an enhanced adrenocortical activity the block of the protein synthesis in the liver proved to disappear. Adrenalectomy excluded the activation of alanine-aminotranspherase and a drop of the free amino acids in the liver during physical exercise: corticosterone administration to adrenalectomized animals restored these shifts.  相似文献

Spinal cord section brings about early and late changes in rat liver tyrosine-alpha-ketoglutarate aminotransferase activity. Early effects (4 h after surgery): spinal cord section at C7 level causes an unreactiveness of rat liver tyrosine-alpha-ketoglutarate aminotransferase both to endogenously and exogenously elevated plasma glucocorticoid levels. Induction of tyrosine–alpha-ketoglutarate aminotransferase by hydrocortisone administration is almost completely inhibited. This unreactiveness of the rat liver enzyme to hydrocortisone is not due to delayed resorption of hydrocortisone by the peritoneum as tested with [3H]hydrocortisone, to changes in the secretion of hypophyseal hormones or to changes in the levels of glucose in blood or liver. L3 level section of the spinal cord or sham operation results in a stress-like enzyme pattern (an increase at 4 h with return to normal level at 24 h). The stress elevation of tyrosine–alpha-ketoglutarate aminotransferase at 4 h after the operation is absent in C7 level sectioned rats. This effect is not due to a decreased plasma corticosterone level since it is 4.1-fold higher in C7 level sectioned rats and 2.7-fold higher in sham-operated controls (as measured 2.5 h after the operation). Late effects (24 h after the surgery): C7 level section of the spinal cord brings about a nine-fold increase in a tyrosine–alpha-ketoglutarate aminotransferase activity in animals with intact adrenals and three-fold increase in adrenalectomized rats at 24 h after the operation. This increase is abolished almost completely by cycloheximide, irrespective of the time of administration. Experiments with actinomycin D, injected at different times after C7 level section have shown that there exists a period of higher sensitivity of the amino-transferase toward the antibiotic (lasting about 3 h), followed by a period of lower sensitivity (lasting 16 h or longer). These results can be explained by assuming the existence of two tyrosine-alpha-ketoglutarate aminotransferase mRNAs with different lifetime. A direct participation of the CNS in the changes in enzymic activity observed after section of the spinal cord above the segments innervating the liver is suggested.  相似文献

1. Fatty acid synthesis, measured in the perfused liver of fed adrenalectomized rats with 3H2O and 14C-labelled precursors, was less than in control sham-operated rats. 2. This defect was more extensive for synthesis of fatty acids incorporated into triacylglycerols than into phospholipids. 3. There was impairment in desaturation and export of newly synthesized fatty acid. 4. Fatty acid synthesis and desaturation were restored to normal rates 5h after treatment with cortisol in vivo. 5. Fatty acid synthesis was seasonally variable, being highest in the winter; the impairment after adrenalectomy was observed in all seasons. 6. In perfusions with oleate (0.7 mM), no further impairment in fatty acid synthesis was discerned in livers from adrenalectomized rats, in which the rate resembled that in control livers. 7. No defect in the incorporation of oleate into glycerides was discerned in livers from adrenalectomized rats. 8. Cortisol exerted no stimulatory effect on fatty acid synthesis when added to perfusion media. 9. The impairment in hepatic lipogenesis, demonstrable after adrenalectomy, shows that adrenal glucocorticoids promote hepatic capacity for fatty acid synthesis de novo, at least in intact non-diabetic rats. It is suggested that this effect is mediated by insulin, perhaps through direct action on the liver.  相似文献

During germination a decrease in both soluble and protein argmine occurs in pumpkin cotyledons (Cucurbita moschata Poir.) and this decrease parallels the decrease in total nitrogen. Arginine is readily metabolized to urea and ornithine and the metabolism of the latter compounds were studied by allowing cotyledons of intact plants to absorb urea?14C and by incubating cotyledon discs with ornithine-2?14C. Of the urea absorbed, 90 % of the label was metabolized to 14CO2 with little free urea remaining in the plant. Urease activity in the cotyledons reached a maximum at 4 to 6 days of germination. Ornithine was extensively metabolized with 37 % of the label being incorporated into protein by 3 h. Soluble and protein arginine accounted for 23 % of the label suggesting that Krebs-Henseliet cycle enzymes were present.  相似文献

Rats were injected with combinations of morphine-N-14CH3 and morphine-63H and the isotope content of the brain and liver was measured by combustion in a tissue oxidizer. The liver of intact male rats showed a significant increase in the 3H to 14C isotope ratio relative to the blood reflecting the existence of N-demethylation in this organ. This increase was not observed in the liver of either intact females or castrated males or females. Centrally, the hypothalamus, medial thalamus, and corpus striatum of both intact and castrated male and female rats exhibited increases in 3H to 14C isotope ratios indicating the presence of N-demethylation in these tissues. These results indicate that testicular hormones serve to increase the hepatic N-demethylation of morphine, but apparently reduce the comparable reaction in the CNS.  相似文献

The aim of the study was to investigate the effects of zinc deficiency and supplementation on lipid peroxidation and glutathione levels in blood and in some tissues of rats performing swimming exercise. Forty adult male Sprague-Dawley rats were divided into four groups: group 1, zinc-deficient consisted of swimming rats; group 2 consisted of zinc-supplemented swimming rats; groups 3 and 4 were the swimming and nonswimming controls, respectively. The levels of malondialdehyde and glutathione were measured after 4 wk of zinc-deficient or zinc-supplemented diet and 30 min of swimming exercise daily. The erythrocyte glutathione levels of groups 2 and 4 were significantly higher than those of groups 1 and 3 (p<0.01). The plasma malondialdehyde level of group 1 was significantly higher than all other groups. The glutathione levels in liver, kidney, striated muscle, and testes of group 2 were higher than in the other groups (p<0.01) and higher in kidney and striated muscle of group 3 than in groups 1 and 4 (p<0.01). The tissue malondialdehyde levels of striated muscle, liver, kidney, and testes of group 1 were significantly higher than for all other groups (p<0.01). Our findings suggest that both swimming exercise and zinc deficiency result in an increase of lipid peroxidation in tissues and that zinc supplementation prevents these alterations by the activation of the antioxidant system.  相似文献

Summary Regulation of urea transport by vasopressin in inner medullary collecting duct (IMCD) cells is thought to be important for the urinary concentrating mechanism. Isolated tubule perfusion studies suggest the existence of a saturable urea carrier. We have measured14C-urea efflux in IMCD cells which were freshly isolated and grown in primary culture. Cells were isolated from rat papilla by collagenase digestion and hypotonic shock. In suspended cells,14C-urea efflux (J urea from loaded cells was exponential with time constant 59±3 sec (sem,n=6, 23°C).J urea had an activation energy of 4.1 kcal/mole and was inhibited 42±7% by 0.25mm phloretin and 30–40% by the high affinity urea analogues dimethylurea and phenylurea.J urea was increased 40–60% by addition of vasopressin (10–8 m) or 8-bromo-cAMP (1mm); stimulatedJ urea was inhibited 55±8% by the kinase A inhibitor H-8. Phorbol esters and epidermal growth factor did not alterJ urea. IMCD cells grown in primary culture were homogeneous in appearance with>fivefold stimulation of cAMP by vasopressin. The exponential time constant for urea efflux was 610±20 sec (n=3).J urea was not altered by vasopressin, cAMP or phloretin. Another function of in vivo IMCD cells, vasopressin-dependent formation of endosomes containing water channels, was absent in the cultured cells. These results demonstrate presence of a urea transporter on suspended IMCD cells which is activated by cAMP and inhibited by phloretin and urea analogues. The urea transporter and its regulation by cAMP, and cAMP-dependent apical membrane endocytosis, are lost after growth in primary culture.  相似文献

The objective of the present study is to explore the effects of acute swimming exercise on plasma levels of some elements in rats, immediately after the exercise, and 24 and 48 h later. The study included 40 adult male rats of Spraque Dawley species, which were equally allocated to four groups. Group 1: General Control Group; Group 2: Swimming Group, the group that was decapitated immediately after 30-min acute swimming exercise; Group 3: Swimming Group, the group that was decapitated 24 h after 30-min acute swimming exercise; Group 4: Swimming Group, the group that was decapitated 48 h after 30-min acute swimming exercise. Plasma copper (Cu), iron (Fe), magnesium (Mg), phosphorus (P), selenium (Se), zinc (Zn) levels were determined according to atomic emission method in the blood samples collected from the animals by decapitation method. Measurements conducted immediately after acute swimming exercise (group 2) showed a significant decrease in Se and Zn levels (p < 0,01) and a significant increase in P levels (p < 0,01), when compared to group 1. Measurements carried out 24 h after the exercise (group 3) demonstrated a significant increase in all parameters except for Mg, in comparison to groups 1 and 2 (p < 0,01). It was seen in the measurements made 48 h after the exercise (group 4) that all parameters were restored to control values. The results of our study show that acute swimming exercise significantly changes plasma Cu, Fe, P, Se, and Zn levels.  相似文献

The present study was performed to examine the effect of angiotensin II on hepatic angiotensinogen production in adrenalectomized rats. The hepatic angiotensinogen mRNA levels in rats without adrenal glands increased 2.8-fold 4 h after the start of angiotensin II infusion. In intact rats with adrenal glands, the hepatic angiotensinogen mRNA levels increased 2.7-fold 4 h after the start. The angiotensin II infusions did not only increase angiotensinogen mRNA levels in intact rats but also increased those in adrenalectomized rats. The results suggest that the angiotensinogen response to ANG II was not dependent on adrenal glucocorticoid.  相似文献

[Purpose]

The purpose of this study is to examine the effects of six-weeks of endurance swim training and short-term intake of Rg1 on the expression of related proteins as well as improvement of aerobic exercise capacity in 8-week-old male SD rats.

[Methods]

The groups were divided into placebo (NP, n=6), Rg1 (NRG, n=6), exercise+placebo (EP, n=7), and exercise+Rg1 (ERG, n=7). On completion of the 6-week swimming exercise, Rg1-intake groups were treated with acute uptakes (3 times within 24hrs) of Rg1. After the treatment, all groups were subjected to a swim to exhaustion test, and then the mass of muscle tissue, mRNA expression level and activity of citrate synthase (CS) were analyzed on plantaris.

[Results]

There were no differences in the effect of 6-week swimming exercise and short-term intake of Rg1 on body weight and muscle mass between groups. Although the CS mRNA expression was elevated in the exercise group and combined treatment group, there was no significant difference in CS activity. Acute uptakes of Rg1 did not affect swimming time to exhaustion, but it was increased by 235% and 314% by the 6-weeks of exercise and combined treatment of exercise and Rg1, respectively, which suggests that the combined treatment increased the effect on the capacity of aerobic exercise.

[Conclusion]

Based on these results, it was confirmed that even a short-term treatment of Rg1 can give an additive effect for improvement of exercise function, and additional studies are needed for the mechanisms and modes of its working.  相似文献

Abstract

The process of muscular atrophy following denervation has been tentatively ascribed to the influence of glucocorticoids (G) because of the rapid increase of cytosolic G receptors (RG) after sciatic nerve section. It appears however that the level of muscular atrophy is similar: 1. in slow or fast-twitch muscles in spite of huge variations in RG; 2. in intact or adrenalectomized (ADR-X) rats. Moreover, the protein muscle profile of intact or ADR-X rats after gel electrophoresis is similar but drastically decreased after 3 weeks of denervation. We conclude that there is no causeeffect relationship between muscle atrophy and RG elevation after nerve section.  相似文献
