A novel pale-yellow-pigmented, moderately halophilic, facultatively alkaliphilic, non-motile, non-spore-forming, catalase- and oxidase-positive, obligately aerobic Gram-positive coccus, strain YIM-C678T was isolated from a saline soil sample collected from a hypersaline habitat in the Qaidam basin, northwest China. The organism grew at 4–37°C and pH 6.0–11.0, with optimum growth at 25°C and pH 8.0. Strain YIM-C678T grew optimally in the presence of 10–12% (w/v) NaCl and growth was observed in 1–25% (w/v) NaCl. The cell wall murein type was l-Lys-Gly5. Major cellular fatty acids were anteiso-C15:0, iso-C15:0, iso-C16:0 and C16:0. Menaquinone 6 (MK-6) was the major respiratory quinone. The DNA G + C content was 46.5 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that the strain YIM-C678T belonged to the family Staphylococcaceae and was most closely related to the eight described species of the genus Salinicoccus with sequence similarities from 92.2 (S. luteus YIM 70202T) to 97.5% (S. kunmingensis YIM Y15T). The DNA–DNA relatedness between strain YIM-C678T and S. kunmingensis YIM Y15T was 35.4%. Chemotaxonomic data and 16S rRNA gene sequence analysis supported the affiliation of strain YIM-C678T with the genus Salinicoccus. The combination of phylogenetic analysis, phenotypic characteristics, chemotaxonomic differences and DNA–DNA hybridization data supported the view that the bacterium represents a novel species of the genus Salinicoccus, for which the name Salinicoccus salitudinis sp. nov. is proposed, with YIM-C678T (=DSM 17846 = CGMCC 1.6299) as the type strain.  相似文献

A novel bacterium B9T was isolated from tidal flat sediment. Its morphology, physiology, biochemical features, and 16S rRNA gene sequence were characterized. Colonies of this strain are yellow and the cells are Gram-negative, rod-shaped, and do not require NaCl for growth. The 16S rRNA gene sequence similarity indicated that strain B9T is associated with the genus Lysobacter (≤ 97.2%), Xanthomonas (≤ 96.8%), Pseudomonas (≤ 96.7%), and Luteimonas (≤ 96.0%). However, within the phylogenetic tree, this novel strain shares a branching point with the species Luteimonas composti CC-YY255T (96.0%). The DNA-DNA hybridization experiments showed a DNA-DNA homology of 23.0% between strain B9T and Luteimonas mephitis B1953/27.1T. The G+C content of genomic DNA of the type strain is 64.7 mol% (SD, 1.1). The predominant fatty acids are iso-C11:0, iso-C15:0, iso-C16:0, iso-C17:0, iso-C17:0 ω9c, and iso-C11:0 3-OH. Combined analysis of the 16S rRNA gene sequences, fatty acid profile, and results from physiological and biochemical tests indicated that there is genotypic and phenotypic differentiation of the isolate from other Luteimonas species. For these reasons, strain B9T was proposed as a novel species, named Luteimonas aestuarii. The type strain of the new species is B9T (= KCTC 22048T, DSM 19680T).  相似文献

A novel actinomycete, designated as strain YIM 75980T, was isolated from a soil sample collected from a dry-hot river valley in Dongchuan county, Yunnan province, south-west China and was subjected to polyphasic taxonomic characterization. The organism produced circular, smooth, red to black coloured colonies comprising coccoid-shaped cells. Colonies on agar medium lacked mycelia and cells adhered to the agar. Strain YIM 75980T contained meso-diaminopimelic acid as the diagnostic diamino acid in the cell-wall peptidoglycan and contained galactose, arabinose and glucosamine as the main sugars in the whole-cell hydrolysates. The predominant menaquinone was MK-9 (H4) and the major fatty acids were iso-C15:0, iso-C16:0 and C16:0. The DNA G + C content of strain YIM 75980T was 73.1 mol%. Phylogenetic analyses based on 16S rRNA gene sequences clearly showed that strain YIM 75980T formed a distinct clade within the genus Geodermatophilus and was closely related to Geodermatophilus obscurus DSM 43160T (level of similarity, 97.9%). Furthermore, the result of DNA–DNA hybridization between strain YIM 75980T and G. obscurus 43160T demonstrated that this isolate represented a different genomic species in the genus Geodermatophilus. Moreover, the physiological and biochemical data showed the differentiation of strain YIM 75980T from its closest phylogenetic neighbour. Therefore, it is proposed that strain YIM 75980T represents a novel species of the genus Geodermatophilus, for which the name Geodermatophilus nigrescens sp. nov. is proposed. The type strain is YIM 75980T (=CCTCC AA 2011015T =JCM 18056T).  相似文献

A Gram-negative, aerobic, rod shaped, non-spore-forming bacterial strain, designated Dae08T, was isolated from sediment of the stream near Daechung dam in South Korea, and was characterized in order to determine its taxonomic position, using a polyphasic approach. Comparative 16S rRNA gene sequence analysis showed that strain Dae08T belongs to the family Xanthomonadaceae of the Gammaproteobacteria, and is related to Lysobacter brunescens ATCC 29482T (97.3%). The phylogenetic distances from any other species with validly published names within the genus Lysobacter were greater than 3.7%. The G+C contents of the genomic DNA of strain Dae08T was 69.3 mol%. The detection of a quinone system with Q-8 as the predominant compound and a fatty acid profile with iso-C15:0, iso-C17:1, ω9c, iso-C17:0, iso-C16:0, and iso-C11:0 3-OH as the major acids supported the affiliation of strain Dae08T to the genus Lysobacter. DNA-DNA relatedness between strain Dae08T and its phylogenetically closest neighbour was 28%. On the basis of its phenotypic properties and phylogenetic distinctiveness, strain Dae08T (= KCTC 12600T) should be classified in the genus Lysobacter as the novel species, for which the name Lysobacter daecheongensis sp. nov. is proposed.  相似文献

A Streptomyces-like actinomycete strain, designated as YIM 78087T, was isolated from a sediment sample collected from Hehua hot spring in Tengchong, Yunnan province, south-west China. The taxonomic position of strain YIM 78087T was investigated by a polyphasic approach. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain YIM 78087T belongs to the genus Streptomyces and is closely related to Streptomyces fimbriatus DSM 40942T, Streptomyces marinus DSM 41968T and Streptomyces qinglanensis DSM 42035T (97.18, 97.05 and 97.1 % similarity, respectively). Combined with the low values of DNA–DNA hybridization between strain YIM 78087T and its closest neighbours, these analyses indicated that this new isolate represents a different genomic species in the genus Streptomyces. The predominant menaquinones of strain YIM 78087T were identified as MK-9 (H4) and MK-9 (H6). The major fatty acids were identified as anteiso-C15:0 (28.4 %), anteiso-C17:0 (23.0 %) and iso-C16:0 (15.1 %). The whole-cell hydrolysates found to contain glucose, mannose and ribose. The DNA G+C content was determined to be 73.0 mol%. Based on the comparative analysis of phenotypic and genotypic characteristics, it is proposed that strain YIM 78087T represents a novel species of the genus Streptomyces, for which the name Streptomyces calidiresistens sp. nov., is proposed. The type strain is YIM 78087T (=BCRC 16955T=DSM 42108T=JCM 19629T).  相似文献

Cells were observed to be rod-shaped and the colonies convex, circular, smooth, yellow and non-transparent. Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain YIM 77924T belongs to the genus Thermus. The 16S rRNA gene sequence similarity values between strain YIM 77924T and other species of the genus Thermus were all below 97 %. The polar lipids of strain YIM 77924T were determined to be aminophospholipid, phospholipid and glycolipid. The predominant respiratory quinone was determined to be MK-8 and the G+C content was 66.64 mol%. The major fatty acids identified were iso-C16:0, iso-C15:0, iso-C17:0 and C16:0. On the basis of the morphological and chemotaxonomic characteristics as well as genotypic data, strain YIM 77924T is proposed to represent a novel species, Thermus tengchongensis sp. nov., in the genus Thermus. A Gram-positive, moderately halophilic, facultatively alkaliphilic, catalase- and oxidase-positive, obligately aerobic, filamentous actinomycete strain, designated YIM 90022T, was isolated from saline soil collected from the Qaidam Basin, north-west China. Phylogenetic analysis based on 16S rRNA gene sequences indicated that the new isolate was a member of the genus Nocardiopsis and the sequence similarities between the isolate and the type strains of members of the genus Nocardiopsis were in the range of 95.1–98.7%. Phenotypic and chemotaxonomic properties of this organism also indicated that strain YIM 90022T was a member of the genus Nocardiopsis. The strain grew well on most of the media tested, producing yellow-white to deep brown substrate mycelium and white aerial mycelium. Light gray to deep brown diffusible pigments were produced. The substrate mycelium was well developed and fragmented with age; the aerial mycelium produced long, straight to flexuous spore chains with non-motile, smooth-surfaced, rod-shaped spores on them. The strain grew in the presence of 1–15% (w/v) total salts (optimum, 3–5%) and at pH 6.0–10.5 (optimum, pH 8.5) and 10–45°C (optimum, 30°C). Whole-cell hydrolysates of strain YIM 90022T contained meso-diaminopimelic acid and no diagnostic sugars. The predominant menaquinones were MK-10(H4), MK-9(H8), MK-10(H6) and MK-10(H8). Polar lipids comprised diphosphatidylglycerol, phosphatidylcholine, phosphatidylglycerol and phosphatidylmethylethanolamine. The major cellular fatty acids were iso-C16:0, anteiso-C17:0, 10-methyl-C18:0 and 10-methyl-C17:0. The DNA G + C content of strain YIM 90022T was 71.5 mol%. The combination of phylogenetic analysis, DNA–DNA relatedness data, phenotypic characteristics and chemotaxonomic data supported the suggestion that strain YIM 90022T represents a new species of the genus Nocardiopsis, for which the name Nocardiopsis terrae sp. nov. is proposed. The type strain is YIM 90022T (=CCTCC AA 208011T =KCTC 19431T).  相似文献

A strain YIM 33098T (= CCTCC AA001027T = DSM 41831T) was isolated from a forest soil sample collected from Nanning in Guangxi Province, China, in the course of screening for producers of new drug lead compounds. This strain was identified by using a polyphasic approach. The results showed that it should be assigned to the genus Streptomyces. An almost complete 16S rRNA gene sequence of the strain was determined and compared with those of representative Streptomyces species. Strain YIM 33098T was clustered in the same subclade with Streptomyces tendae ATCC19812T and Streptomyces eurythermus ATCC14975T. Similarities of strain YIM 33098T with the two strains were 97.35% and 97.42%, respectively. Based on the phenotypic and genotypic evidence, it is therefore proposed that strain YIM 33098T should be classified in the genus Streptomyces as a new species under the name of Streptomyces nanningensis sp. nov.  相似文献

A novel cold-resistant bacterium, designated YIM 016T, was isolated from a peat bog sample collected from Mohe County, Heilongjiang Province, Northern China and its taxonomic position was investigated using a polyphasic approach. The strain was Gram-positive, aerobic, endospore-forming, motile and rod-shaped. Phylogenetic analyses based on the 16S rRNA gene sequence clearly revealed that strain YIM 016T is a member of the genus Paenibacillus. The strain is closely related to Paenibacillus alginolyticus DSM 5050T, Paenibacillus chondroitinus DSM 5051T and Paenibacillus pocheonensis Gsoil 1138T with similarities of 99.0 %, 97.0 % and 96.3 %, respectively. Meanwhile, the low DNA–DNA relatedness levels between strain YIM 016T and its closely related phylogenetic neighbours demonstrated that this isolate represents a new genomic species in the genus Paenibacillus. Phenotypic and chemotaxonomic tests showed that growth of strain YIM 016T occurred at 4–37 °C, pH 6.0–8.0 and with a NaCl tolerance up to 0.5 % (w/v). The peptidoglycan contained meso-diaminopimelic acid, alanine and glutamic acid. The whole-cell hydrolysates mainly contained glucose, galactose and ribose. The predominant menaquinone was MK-7 and the major fatty acids were anteiso-C15:0 and iso-C16:0. The DNA G+C content of strain YIM 016T was 51.7 mol %. On the basis of phylogenetic, phenotypic and chemotaxonomic characteristics, strain YIM 016T could be clearly distinguished from other species of the genus Paenibacillus. It is therefore concluded that strain YIM 016T represents a novel species in the genus Paenibacillus, for which the name Paenibacillus frigoriresistens sp. nov. is proposed. The type strain is YIM 016T (= CCTCC AB 2011150T = JCM 18141T).  相似文献

A novel actinomycete, designated strain YIM CF22T, was isolated from a desert soil sample collected from Turpan in Xinjiang Uyghur Autonomous Region, north-western China. The taxonomic position of the strain YIM CF22T is described based on a polyphasic approach. Strain YIM CF22T was found to form irregular sporangia on agar media. It contains meso-diaminopimelic acid in the cell wall peptidoglycan. The major menaquinone was identified as MK-9(H4); the polar lipids were identified as diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, two unidentified phospholipids and two unidentified glycolipids. The whole cell sugars were found to be ribose, mannose, galactose, glucose and xylose. The major cellular fatty acids were found to be (>?5%) iso-C16:0 (43.5%), anteiso-C17:0 (10.2%), iso-C15:0 (7.1%), C17:1 ω8c (6.3%) and iso H-C16:1 (5.9%). The G+C content was determined to be 70.8%. 16S rRNA gene sequence analysis of strain YIM CF22T showed high similarity (97.0%) to Actinoplanes rishiriensis NBRC 108556T. The strain also showed high 16S rRNA gene sequence similarities to Verrucosispora sediminis CGMCC 4.3550T (96.9%) and Micromonospora tulbaghiae DSM 45142T (96.8%). Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain YIM CF22T clusters with A. rishiriensis NBRC 108556T, Actinoplanes globisporus JCM 3186T and Actinoplanes rhizophilus NEAU-A-2T. Based on the differential phenotypic characteristics and the results of DNA–DNA relatedness and phylogenetic analysis, it is proposed that strain YIM CF22T represents a novel species of the genus Actinoplanes, for which the name Actinoplanes deserti sp. nov. is proposed. A Gram-positive, aerobic, non-motile actinobacterial strain, designated YIM C01235T, was isolated from a soil sample collected from the Swallow Cave, Yunnan province, south-west China. The isolate grew at 10–30 °C, pH 6.0–9.0 and 0–8 % (w/v) NaCl. Phylogenetic analysis based on 16S rRNA gene sequences revealed highest similarity to Saccharopolyspora gloriosae YIM 60513T (96.8 %), and lower 16S rRNA gene sequence similarities (95.1–96.7 %) with the other species of the genus Saccharopolyspora. The whole-cell hydrolysates contained meso-diaminopimelic acid (meso-DAP), arabinose and galactose. The polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylcholine, phosphatidylethanolamine, phosphatidylmethylethanolamine, two unknown glycolipids, two unknown phospholipids and one polar lipid. MK-9(H4) was the predominant menaquinone. The major fatty acids were iso-C15:0, iso-C16:0, anteiso-C15:0 and anteiso-C17:0. Mycolic acids were not detected. The genomic DNA G+C content was 69.1 mol%. These chemotaxonomic data, together with its morphological properties, were consistent with the assignment of strain YIM C01235T to the genus Saccharopolyspora. The results of physiological and biochemical tests allowed strain YIM C01235T to be differentiated phenotypically from all the recognized Saccharopolyspora species. On the basis of evidence from this polyphasic study, the novel species Saccharopolyspora cavernae sp. nov. is proposed. The type strain is YIM C01235T (=DSM 45825T = CCTCC AA 2012022T).  相似文献

A novel actinomycete strain, designated TRM 40133T, was isolated from a hypersaline habitat of Tarim basin in Xinjiang Province, north-west China. Its taxonomic status was determined using a polyphasic approach. Phylogenetic analysis based on an almost-complete 16S rRNA gene sequence of the strain showed that it formed a well-seperated sub-branch within the radiation of the genus Saccharopolyspora. The highest levels of 16S rRNA gene sequence similarity was found between the strain TRM 40133T and Saccharopolyspora qijiaojingensis YIM 91168T (96.5%). The chemotaxonomic characteristics of the isolate are typical for the genus Saccharopolyspora. It contained meso-DAP as the diagnostic diamino acid. Whole cell hydrolysate contained arabinose, xylose, ribose and glucose. The diagnostic phospholipids were phosphatidylcholine, phosphatidylglycerol, phosphatidylinositol and two unknown phospholipids. The main menaquinone was MK-9(H6) and MK-9(H4). No mycolic acid was detected. The predominant cellular fatty acids were iso-C16:0 and anteiso-C17:0. The G+C content of the genomic DNA was 68.2 mol%. In addition, the strain TRM 40133T had a phenotypic profile that readily distinguished it from the recognized representatives of the genus Saccharopolyspora. The strain TRM 40133T therefore represents a novel species of the genus Saccharopolyspora, for which the name Saccharopolyspora lacisalsi sp. nov. is proposed. The type strain is TRM 40133T (=KCTC 19987T =CCTCC AA 2010012T).  相似文献

Two actinomycete strains, designated YIM M11168T and YIM M11177, were isolated from marine sediment samples from Little Andaman, Indian Ocean, and their taxonomic position was determined by a polyphasic approach. The two Gram-positive, aerobic strains were observed to produce branched substrate mycelium and aerial hyphae but did not fragment, and no diffusible pigment was produced on the media tested. At maturity, spores were formed singly or in pairs on aerial hyphae and substrate mycelium, and occasionally the single ones were borne on long sporophores. The optimum growth was determined to occur at 28 °C, 0–4 % (w/v) NaCl and pH 7.0–8.0. Whole-cell hydrolysates of both strains contained meso-diaminopimelic acid and the diagnostic sugars were determined to be galactose, glucose and arabinose. Their predominant menaquinone was found to be MK-9(H4). The polar lipids detected in the two strains were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol mannoside, phosphatidylinositol, phosphatidylmethylethanolamine, phosphatidylethanolamine and two unknown phosphoglycolipids. The major fatty acids (>10 %) identified were iso-C16:0, iso-C16:1 H, iso-C16:0, C17:1 ω6c for strain YIM M11168T, iso-C16:0 and Summed Feature 3 for strain YIM M11177. The G + C contents of the genomic DNAs of both strains were determined to be 71.4 %. DNA–DNA hybridization relatedness values (78.4 ± 3.7 %) of these two isolates supported the conclusion that they belong to the same species. Based on phylogenetic analysis, phenotypic and genotypic data, it is concluded that the two isolates belong to a novel species of the genus Saccharomonospora of the family Pseudonocardiaceae. The name Saccharomonospora oceani sp. nov. (Type strain YIM M11168T = DSM 45700T = JCM 18128T) is proposed for the novel species.  相似文献

A novel aerobic, Gram-positive, non-motile halotolerant actinobacterium, designated strain YIM 96448T, was isolated from a saline soil sample collected from the south bank of Aiding Lake in Xinjiang Province, northwest China. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain YIM 96448T is closely related to Phytoactinopolyspora endophytica EGI 60009T (98.0%) and Phytoactinopolyspora alkaliphila EGI 80629T (97.0%). Optimum growth of the strain was observed at 28-37 °C, pH 7–9 and in the presence of 5–8% NaCl (w/v). The respiratory quinone of strain YIM 96448T was determined to be MK-9(H4). The major fatty acids (>10%) were identified as anteiso-C15:0, iso-C16:0, anteiso-C17:0 and summed feature 4 (iso-C17:1 I and/or anteiso-C17:1B). The polar lipid profile was found to consist of diphosphatidylglycerol, an unidentified aminophospholipid, phosphatidylglycerol, two phosphatidylinositol mannosides, three unidentified phosphoglycolipids and four unidentified phospholipids. The DNA G+C content was determined to be 68.7 mol%. On the basis of phenotypic, chemotaxonomic and phylogenetic analysis, strain YIM 96448T should be classified as a novel species in the genus Phytoactinopolyspora, for which the name Phytoactinopolyspora halotolerans sp. nov. is proposed. The type strain is YIM 96448T (=CCTCC AB 2017055T = KCTC 39924T).  相似文献
