Neuronal nuclei were isolated from rabbit cerebral cortex, and lipid acetylation reactions were studied because of the high nuclear concentration of acetyltransferases that generate platelet activating factor (PAF) and its acyl analogue AcylPAF. The neuronal nuclear acetylation of 1-palmitoyl lysophosphatidylcholine (lyso PC) was found to be increased more than two fold when low concentrations of lyso PC were incubated in acetylation assays in the presence of 1-palmitoyl lysophosphatidic acid (lyso PA) or 1-hexadecyl glycerophosphate (AGP). This effect was not found for a variety of other acidic and neutral 1-acyl lysoglycerophospholipids. At 4 M concentrations, AGP was the more effective in increasing rates of lyso PC acetylation, while lyso PA was more effective at 25-35 M. 1-Stearoyl, 1-alkenyl and 1-decanoyl analogues of lyso PA were all less effective than 1-palmitoyl lyso PA. Phosphatidic acid was considerably less effective than lyso PA, while the acetylated analogue of AGP, AAcGP (alkylacetylglycerophosphate), increased rates of lyso PC to maxima similar to those seen with lyso PA or AGP. In addition, AAcGP promoted these maxima at considerably lower concentrations (2-4 M). A mechanism for these effects was suggested when nuclear envelopes (NE), isolated in the presence of PMSF, showed these maximal acetylation rates at low lyso PC concentrations, and these rates were not elevated by the presence of lyso PA. PMSF is a protease inhibitor but can also inhibit lysophospholipase activity. We found a nuclear lysophospholipase that degraded lyso PC at rates more than 13 times those of nuclear lyso PC acetylation. PMSF did inhibit this nuclear lysophospholipase, as did lyso PA, AGP and AAcGP. Kinetic analyses of the effects of lyso PA, AGP and AAcGP on lyso PC lysophospholipase indicated that these three lipids acted as competitive inhibitors for the lyso PC substrate. It is possible that low rates of lyso PC acetylation seen in neuronal nuclei at low lyso PC concentrations, are caused by lyso PC loss mediated by a very strong nuclear lysophospholipase. The effects of lyso PA, AGP and AAcGP in boosting rates of lyso PC acetylation likely come from the inhibition of nuclear lysophospholipase and a preservation of lyso PC concentrations. Competing neuronal nuclear reactions for low endogenous levels of lyso PC may regulate the formation of AcylPAF, and rising lyso PA, AGP or AAcGP concentrations can increase rates of nuclear AcylPAF synthesis.  相似文献

Summary Rainbow trout (Salmo gairdneri) acclimated to 5°C or 20°C were administered 2-3H-glycerol and 1-14C-acetate (63 Ci of each isotope/100 g body weight) via intraperitoneal injection, and subsequently maintained at their respective acclimation temperatures. Total lipid extracts (>80% phospholipid) were prepared from isolated microsomes of liver, gill and muscle tissue at various times over a three week period. Half-lives were determined independently for the fatty acyl and glycerol moieties from slopes of regression lines relating dpm/nmole phospholipidP i vs time. In liver tissue, rates of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) turnover were also determined. Membrane turnover was most rapid in liver followed by gill and muscle. In liver, membrane fatty acids turned over more rapidly in warm-(t 1/2=3.4 days) than in cold-(t 1/2=6.8 days) acclimated fish, whereas in gill, rates of fatty acid turnover, did not differ significantly between acclimation groups. In contrast, rates of glycerol turnover were independent of acclimation temperature in liver, but faster (t 1/2=6.7 days) in warm- than cold- (t 1/2=15.1 days) acclimated fish in gill. In total lipid extracts, rates of fatty acid and glycerol turnover were equivalent in warm-acclimated fish, however, in cold-acclimated trout, there was a tendency for fatty acids (t 1/2=9.1 days) to turnover more rapidly than glycerol (t 1/2=15.1 days) in gill tissue, but more slowly (t 1/2=6.82 days) than glycerol (t 1/2=4.1 days) in liver. Although rates of glycerol turnover were equivalent in PC and PE of liver microsomes, the fatty acyl component turned over significantly more rapidly in PC at both acclimation temperatures. In cold-acclimated trout, rates of fatty acid and glycerol turnover were equivalent in PE, but the fatty acyl moiety of PC (t 1/2=4.7 days) turned over significantly more rapidly than glycerol (t 1/2=7.5 days). These results were interpreted as indicating that: (1) acclimation temperature independently influenced rates of fatty acid and glycerol turnover in a tissue specific manner, (2) a deacylation-reacylation pathway was activated in both liver and gill as a consequence of cold acclimation, but that liver tissue was more effective than gill in reutilizing the fatty acids released by phospholipase activity, and (3), in liver microsomes, patterns of turnover were phospholipid specific, with PC and PE differing either in the susceptibility of their acyl groups to degradation, or in their ability to reutilize fatty acids cleaved during membrane turnover at cold temperatures.  相似文献

The effect of amyloid (A), the major constituent of the Alzheimer's (AD) brain on lipid metabolism was investigated in cultured nerve cells and in a fetal rat brain model. Differentiated (NGF) and undifferentiated PC12 cells or primary cerebral cell cultures were incubated with [14C]acetate in the absence or presence of A1–40. Incorporation of label into lipid species was determined after lipid extraction and TLC separation. Phosphatidylcholine (PC) and phosphatidylserine (PS) synthesis was increased by A1–40, in a dose dependent manner, an effect which was more pronounced in differentiated PC12 cells. A significant proportion of radioactivity (5–6%) was released into the medium with a radioactivity distribution similar to that of the cellular lipids. Cholesterol and PC were the highest labeled medium lipids. Increasing A1–40 concentration up to 0.1 g/ml in cerebral cells but not in PC12 cells, caused a relative increase (1.5 fold) in release of PS, while that of PE decreased. Stimulation of PS release may possibly be associated with apoptotic cell death. A1–40 peptide (5 g) was administered intraperitonealy into rat fetuses (18 days gestation) along with [14C]acetate (2Ci/fetus). After 24 h, the maternal-fetal blood supply was occluded for 20 min (ischemia) followed by 15 min reperfusion. Fetuses were killed and liver and brain tissue subjected to lipid extraction and radioactivity determination after TLC. A1–40 peptide increased synthesis of different classes of lipids up to 20–40% in brain tissue compared to controls. Labeling of liver lipids was decreased by A1–40 by 20–30%. A general decrease in synthesis of lipids was observed after ischemia/reperfusion. Our data suggest that A1–40 peptide regulates normal lipid biosynthesis but under ischemia it compromises it. The latter finding may confirm the oxidative stress etiology in AD and suggests that A1–40 modulation of lipid metabolism may have Alzheimer's pathological relevance, particularly at high peptide concentrations.  相似文献

Summary Gibberellic acid (GA3) accelerated abscission when applied, in a wide concentration range, to excised abscission zones of cotton. Abscission was promoted equally by distal or proximal applications of from 10-3 to 100 g. A slight, but inconsistent, abscission retardation was obtained with distal applications of 10-6 and 10-7 g.Seven different gibberellins accelerated abscission equally when applied distally at amounts of 5×10-4 to 5×10-1 g per abscission zone. At 5×10-5 g there were great differences in effectiveness; their activities can be ranked: A3>A5A4>A7=A8>A1=A9.The ready translocatability of GA3 was suggested when 1.0 or 0.01 g was applied to one petiole, and the opposite untreated petiole abscised at the same time as the treated one. However, 0.001 g was not effective in moving across the stem and inducing abscission of the untreated petiole.The rate of abscission of petioles treated with 1.0 g GA3 was not affected by increasing the length of the petiole from 3 to 9 mm. However, abscission of petioles treated with smaller amounts is inversely proportional to petiole length.The rate of abscission of petioles treated with GA3 decreased with increasing seedling age; there was a simultaneous increase in abscission rate of the controls.Part of this research was based on a portion of a thesis submitted by the senior author to the Graduate Division, University of California, Davis, in partial fulfillment of the requirements for the M.S. degree.  相似文献

Synaptoneurosomes isolated from cerebral cortices of male Sprague-Dawley rats were used for studying GABAA receptor-regulated chloride influx. The in vitro effects of GABA antagonists, SR 95531 (a pyridazinyl GABA derivative) and bicuculline, on pentobarbital-stimulated, muscimol-stimulated or flunitrazepam-enhanced, muscimol-stimulated chloride uptake were studied. The chloride uptake was determined at 30°C, for 5 sec. Pentobarbital and muscimol produced a maximal stimulation of chloride uptake in cortical synaptoneurosomes at 500 M and 50M, respectively. SR 95531 as well as bicuculline had no effect on the basal uptake of chloride. Whereas, SR 95531 (0.3–30 M) and bicuculline (0.1–100 M), when added 5 min before muscimol (50 M), produced a significant concentration-dependent inhibition of muscimol (50 M)-stimulated chloride uptake (IC50 s of 0.89±0.11 M and 13.45±2.10M, respectively). In studies of the inhibitory effects of SR 95531 and bicuculline on pentobarbital (500 M)-stimulated chloride uptake, the IC50 s were 0.81±0.12 M and 3.86±1.14 M, respectively. SR 95531 exhibited a more potent inhibitory effect than bicuculline on flunitrazepam-enhanced, muscimol-stimulated chloride uptake. The results revealed that SR 95531 has a more potent antagonistic effect than bicuculline on GABAA-regulated chloride flux.  相似文献

The aim of this study was to examine the effect of hypo- and hyperthyroidism on the phospholipid composition in developing rat heart. The hypothyroid state (PTU) was induced by 0.05% 6-n-propyl-2-thiouracil in drinking water given to nursing mothers from the postnatal day 2–21. The hyperthyroidism (T3) was made by daily injection of 3,3,5-triiodo-L-thyronine (10 g/100 g body wt) to newborns in the same time period. Age matched intact littermates were taken as euthyroid controls. PTU decreased the concentration of total phospholipids (PL), choline phosphoglycerides (PC), ethanolamine phosphoglycerides (PE) and diphosphatidylglycerol (DPG) and increased the proportion of plasmalogen component of PE (PLPE). T3 increased the concentration of PL, PC, PE, DPG and decreased PLPE in comparison with euthyroid controls. The ratio of saturated/unsaturated fatty acids (FA) in PE was decreased in PTU and increased in T3 group. The ratio of n-6/n-3 polyunsaturated FA in PC, PE and phosphatidylinositol (PI) was increased in PTU due to increase of 18:2n-6 and decrease of 22:6n-3 proportion. T3 decreased this ratio because of decline in 20:4n-6 and rise in 22:6n-3 proportion. Both hypo- and hyperthyroidism decreased the ratio of 20:4n-6/18:2n-6 in the majority of phospholipids. PTU decreased the unsaturation index in PC, PI and phosphatidylserine. It is concluded that thyroid state plays an essential role in the development of membrane phospholipid components in cardiac membranes during the early postnatal period.  相似文献

Besides playing an important role in the maintenance of cell membrane phospholipids, phospholipases A2 (PLA2) are responsible for the release of arachidonic acid (AA) which is a precursor for prostaglandin biosynthesis. The cytosolic PLA2 has been the focus of recent studies, probably due to its ability to respond to protein kinases and changes in intracellular calcium levels. In this study, we examined agents for stimulation of the cytosolic phospholipase A2 in immortalized astrocytes (DITNC). Incubation of DITNC cells with [14C]arachidonic acid (AA) resulted in a time-dependent uptake of the label into phospholipids (PL) and neutral glycerides. In prelabeled cells, release of labeled AA could be stimulated by calcium mobilizing agents such as calcium ionophore A23187 (4–20 M) and thimerosal (100 M), and by phorbol myristate acetate (PMA, 100 nM), an agent for activation of protein kinase C. The release of AA could also be stimulated by ATP (200 M), probably through activation of the purinergic receptor but not by glutamate (1 mM). The stimulated release of AA was dependent on extracellular Ca2+ and was inhibited by mepacrine (50 M), a non-specific PLA2 inhibitor. Western blot analysis further confirmed the presence of an 85 kDa cPLA2 in both membrane and cytosol fractions of these cells and stimulation by A23187 resulted in translocation of this protein to the membrane fraction. Besides labeled fatty acids, A23187 also stimulated the concomitant release of labeled PL into the culture medium and this event was accompanied by the increased release in lactate dehydrogenase (LDH). Results thus revealed that besides activation of cPLA2, the calcium ionophore A23187 is capable of perturbating cell membrane integrity.  相似文献

Midbrain slices containing the dorsal and medial raphe nuclei were prepared from rat brain in order to study serotonergic-GABAergic interaction. The slices were loaded with either [3H] serotonin or [3H]GABA, superfused and the electrically induced efflux of radioactivity was determined. The GABAA receptor agonist muscimol (3 to 30 M) and the GABAB receptor agonist baclofen (30 and 100 M) inhibited [3H]serotonin and [3H]GABA release. These effects of muscimol were reversed by the GABAA antagonists bicuculline (100 M). The GABAB antagonist phaclofen (100 M) also antagonized the baclofen-induced inhibition of [3H]serotonin and [3H]GABA release. Phaclofen by itself increased [3H]serotonin release but it did not alter [3H]GABA overflow. Muscimol (10 M) and baclofen (100 M) also inhibited [3H]serotonin release after depletion of GABAergic neurons by isoniazid pretreatment. These findings indicate the presence of postsynaptic GABAA and GABAB receptors located on serotonergic neurons. The 5-HT1A receptor agonist 8-OH-DPAT (0.01 to 1 M) and the 5-HT1B receptor agonist CGS-12066A (0.01 to 1 M) inhibited the electrically stimulated [3H]serotonin and [3H]GABA release. The 5-HT1A antagonist WAY-100135 (1 M) was without effect on [3H]serotonin and [3H]GABA efflux by itself but it reversed the 8-OH-DPAT-induced transmitter release inhibition. During KCl (22 mM)-induced depolarization, tetrodotoxin (1 M) did not alter the inhibitory effect of CGS-12066A (1 M) on [3H]GABA release, it did blocked, however, the ability of 8-OH-DPAT (1 M) to reduce [3H]GABA efflux. After depletion of raphe serotonin neurons by p-chlorophenylalanine pretreatment, CGS-12066A (1 M) still inhibited [3H]GABA release whereas in serotonin-depleted slices, 8-OH-DPAT (1 M) was without effect on the release. We conclude that reciprocal influence exists between serotonergic projection neurons and the GABAergic interneurons or afferents in the raphe nuclei and these interactions may be mediated by 5-HT1A/B and GABAA/B receptors. Both synaptic and non-synaptic neurotransmission may be operative in the 5-HTergic-GABAergic reciprocal interaction which may serve as a local tuning in the neural connection between cerebral cortex and midbrain raphe nuclei.  相似文献

Previous studies have suggested that the binding of mouse glucuronidase to endoplasmic reticulum membrane is stabilized by the membrane protein egasyn. Using a radioimmunoassay for egasyn, we have now examined the inheritance of egasyn levels in mice. Mice of the ibred strain C57BL/6J, which have normal levels of microsomal glucuronidase, contained 56±10 g egasyn per gram of liver. Mice of the inbred strain YBR, which carry the Eg 0 mutation resulting in the absence of microsomal glucuronidase, did not contain detectable levels of egasyn. The F1 progeny of these two strains contained intermediate levels of egasyn, 25±4 g egasyn per gram of liver. Progeny from the backcross of these F1 animals to YBR were distributed equally into two discrete phenotypic classes. One class lacked both egasyn and microsomal glucuronidase, while the other class contained 25±3 g egasyn per gram of liver and contained normal levels of microsomal glucuronidase. Thus egasyn levels are determined by the Eg locus and show additive inheritance. These results suggest that the Eg gene codes for egasyn and that it is the inability to produce egasyn that results in a deficiency of microsomal glucuronidase in the Eg 0 mutant.This work was supported in part by USPHS Grant GM-19521.  相似文献

Clozapine and several other antipsychotic/antidepressant drugs that fully or partially block GABAA receptors were tested at concentrations that reversed the inhibitory effect of 1 M GABA on 35S-t-butylbicyclophosphorothionate ([35S]TBPS) binding to rat forebrain membranes only about 20–30%, here designated core fractions. Clozapine at 10 M reverses 1 M GABA 25 ± 4.0% (n = 23) (its core fraction). Fourty three compounds were tested alone, and pairwise together with 10 M Clozapine. The core fractions of some of the compounds yielded significant additive reversals together with 10 M Clozapine, while others did not. A group of 14 compounds of which 7 are clinically effective antipsychotic drugs, including Chlorprothixene, Clomacran, Clopipazan, Fluotracen, Sulforidazine, Thioproperazine, and cis-Thiothixene, were statistically non-additive with 10 M Clozapine, suggesting that all of these drugs selectively block the same core population of GABAA receptors as Clozapine. These non-additivities also suggest that Clozapine at 10 M fully saturates a subset of GABAA receptors blocked by 1 M GABA. Therefore, Clozapine probably blocks 2 or more types of GABAA receptors, but only half of the receptors that are sensitive to 1 M GABA. A second group of 12 compounds of which 6 are clinically active antidepressant/antipsychotic drugs including Amoxapine, Clothiapine, Dibenzepine, Inkasan (Metralindole), Metiapine and Zimelidine were slightly, but significantly, additive with Clozapine suggesting that these compounds block most of Clozapine's core fraction, plus a small additional fraction. A third group consisted of ten compounds that yielded larger (R > 80) and statistically highly significant additivities with Clozapine. Complete additivity was obtained with Bathophenanthroline disulfonate, and Isocarboxazid, suggesting that they block GABAA receptors other than those blocked by 10 M Clozapine. Seven classical GABAA receptor blockers, also tested at concentrations yielding 21 to 33% reversal alone, were all significantly additive with 10 M Clozapine, but in no case was the additivity complete. The largest additivity was obtained with Pitrazepine (21%) and the smallest with Tubocurarine (9%). These results provide further support for the notion that selective blockade of the same subset of GABAA receptors may contribute to the clinical antipsychotic/antidepressant effects of Clozapine. The Bopt values for Clozapine are 50 ± 1.7% and 26 ± 2.6% ( n = 3) in whole rat forebrain and cerebellum, respectively, confirming that clozapine-sensitive GABAA receptors are unevenly distributed in the brain. The sedative and anxiolytic properties of Clozapine and other antipsychotic drugs may be due to selective blockade of GABergic disinhibition at certain interneurons.  相似文献

The effect of individual unsaturated fatty acids on the release of tumour necrosis factor (TNF) and interleukin 6 (IL6) was investigated in thioglycollate — induced rat peritoneal macrophages. The intracellular mechanisms associated with the changes of cytokine production in response to fatty acids were also studied. Incubation of macrophages with 100 M docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) increased TNF (21% and 15% respectively) and IL6 (69% and 40% respectively) production. Linoleic acid (LA) diminished TNF production by 16%. At 100 M oleic acid (OA), LA and EPA concentration an increase in macrophage adenylate cyclase activity (110%, 72% and 39% respectively) and a decrease (14%) in the presence of DHA was observed. PGE2 production in the presence of 100 M DHA was reduced by 36%, whereas in the presence of 100 M LA an increase (75%) was observed. Phospholipase A2 (PLA2) activity was also found to be modified in the presence of EPA and DHA at 50 M (20% and 60% respectively) and 100 M (34% and 62% respectively) concentrations. The activities of both protein kinase A (PKA) and protein kinase C (PKC) were effected by the different fatty acids. At 50 M all fatty acids suppressed PKA activity except OA which enhanced PKA activity by 14%. At 100 M fatty acid concentration, EPA suppressed PKA activity by 40%. PKC activity was enhanced by LA and OA, by 18% and 21% respectively. However, at 100 M EPA and DHA, PKC activity was suppressed by 37% and 17% respectively, whereas PKC activity was enhanced by 146% in the presence of 100 M LA. These results show for the first time that unsaturated fatty acids have an effect on macrophage PLA2 activity and that PGE2 may be a potent modulator of IL6 production. From these studies it is tempting to speculate that macrophage TNF and IL6 release may, in part, occur via a PKC and PKA independent pathway and that PLA2 activity and PGE2 concentration are inversely related to production of TNF and IL6.  相似文献

A model study of the microfiltration of fatty acids (C2 to C8) dissolved in a synthetic ultrafiltrate was performed. Ceramic membranes of 0.1 and 0.2 m were used. Results showed that minerals from the ultrafiltrate were involved in membrane fouling, while fatty acids were poorly rejected.On 0.1 m membranes, modelisation of fouling exhibited two different steps of fouling, a first one due to adsorption of solutes, and a second one involving particles deposit onto the membrane. On 0.2 m membranes, only fouling due to particles deposit was observed in modelisation. Results of fouling and rejection pointed out the great difference between 0.1 and0.2 m membranes.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Rainbow trout were used to characterize the direct influence of growth hormone on hepatic lipid mobilization in vitro. Liver was removed from fish fasted 72 h, sliced into 1-mm3 pieces and incubated in Hanks' medium containing ovine or salmon growth hormone (0.28 ng·ml-1–28 g·ml-1). Lipid mobilization, resulting from triacylglycerol hydrolysis, was evaluated by fatty acid and glycerol release into culture medium. Control rates of fatty acid release and glycerol release were 0.95±0.16 (mean ± SE) and 0.88±0.28 mol·l-1·mg fresh weight, respectively. Both ovine growth hormone (28 ng·ml-1) and salmon growth hormone (28 ng·ml-1) stimulated fatty acid release into culture medium, increasing rates by 112% and 97%, respectively, during the course of a 24-h incubation. Glycerol release was similarly augmented by growth hormone treatment. Growth hormone-stimulated metabolite release became evident within 12 h and persisted for up to 72 h. The presence of amino acids in the culture medium potentiated the lipolytic action of growth hormone. Ovine growth hormone (28 ng·ml-1) in the presence of amino acids stimulated a 53% increase in fatty acid, and a 108% increase in glycerol, release over rates observed in the absence of amino acids. Salmon growth hormone (28 ng·ml-1) in the presence of amino acids stimulated a 53% increase in fatty acid, and a 44% increase in glycerol, release over rates observed in the absence of amino acids. Ovine growth hormone (28 ng·ml-1) also stimulated gluconeogenesis, as indicated by a 75% increase in phosphoenolpyruvate carboxykinase activity in liver pieces incubated in the presence of amino acids. Burning landscapes under controlled conditions to reduce the risk of wildfires is a controversial land management practice. The health risks of smoke generated from controlled burning relative to wildfire remain uncertain. Recent work in the Australian monsoon tropics provided a unique opportunity to study the health effects of smoke pollution at and well below national air quality standards. It found that for each increase in the atmospheric mass of particles 10 g or less in aerodynamic diameter (PM10) per cubic meter of air per 24-hour period, there was a 26% increase in daily asthma presentations to the emergency department of the Royal Darwin Hospital, with an apparent threshold at 40 g/m3 PM10 (lower than the Australian PM10 air quality standard of 50 g/m3). This finding was unaffected by adjusting for weekly rates of influenza, weekday vs. weekends, and school holiday periods. The response of Acinetobacter strain 210A to low phosphate concentrations was investigated in P- or C-limited chemostat cultures. The organism accumulated poly--hydroxybutyric acid under P-deprivation, at phosphate concentrations ranging from 0.1 to 0.7 mM. The amount of biomass was proportional to the phosphate concentration in the medium and no polyphosphate was formed. When shifting a culture from P- to C-limitation phosphate was accumulated as polyphosphate. No poly--hydroxybutyrate could be detected in these cells. The amount of polyphosphate in the cell showed a hysteresis. When cultures were shifted from low to high phosphate concentrations, polyphosphate reached a maximum of about 60 mg P per gram of dry weight at about 3 times excess phosphate (ca. 2.5 mM Pi). It decreased to 45 mg P per gram dry weight at approximately 5 times the phosphate needed for growth (ca. 3.5 mM Pi). In the reverse case (high to low) polyphosphate did never exceed 45 mg P per gram dry weight. The specific activities of alkaline phosphatase and the phosphate uptake system were induced at residual Pi concentrations below the detection limit (<10 M). The specific uptake rate followed also a hysteresis. Signal-activated phospholipase A2 cleavesphosphatidylcholine (PC) into free fatty acids and LPC, respectively.Using bis-BODIPY-PC as an indicator substrate for phospholipaseA2 which is taken up by parsley cells, active auxins atconcentrations as low as 1 M and a fungal elicitor induced fattyacid-accumulation. Nordihydroguajaretic acid inhibited the accumulationof fatty acid induced by the elicitor. In addition to this, theelicitor, but not auxin, decreased the pool size of diacylglycerol,which seemed to originate from a PC-splitting phospholipase C, whichwould be a new enzyme in plant signal transduction. However, thiselicitor is known to rapidly increase cytosolic calcium in parsley cellsand this activates phospholipase C. Thus, activation of phospholipase Cshould lead to an increase of diacylglycerol and not to a decrease whichmight indicate a discrepancy between animal and plant phospholipidsignal transduction.  相似文献

The concentration of the C2–C5 carboxylic acids in the water column and sediment of shallow, eutrophic, drainage channels (Lewes Brooks, UK) were measured by gas chromatography. The concentrations of these acids were negatively correlated with dissolved oxygen. The highest concentrations of acetate (up to 200 M), propanoate (up to 38 M) and butanoate (up to 1.2 M) were measured during the summer in the water above the sediment, which became hypoxic during this period. Both acetate and propanoate concentrations declined at night. High concentrations of the acids were also recorded in reedbeds, which were also hypoxic. Only acetate was detected in the sediment pore water (up to 168 M). Summary The pancreatic islets of the guinea pig have been studied by light and electron microscopy. The B granules in glutaraldehyde-fixed tissue often are cup-shaped with an indentation visible on one side of the granule. Phosphotungstic acid hematoxylin (PTAH) positive cells have been characterized by electron microscopy as three subtypes based on the size of the secretory granules. Aa cells are the most common and have secretory granules around 200 m in diameter. Ab cells have large secretory granules around 300 m in diameter and are relatively infrequent. Ac cells are the least common and have small (160 m) granules. DOPA synthesis from phenylalanine was studied in PC12 cells incubated with m-hydroxybenzylhydrazine, to inhibit aromatic L-amino acid decarboxylase. DOPA synthesis rose with increasing concentrations of either phenylalanine or tyrosine; maximal rates (~55 pmol/min/mg protein for tyrosine; ~40 pmol/min/mg protein for phenylalanine) occurred at a medium concentration of ~10 M for either amino acid. The Km for either amino acid was about 1 M (medium concentration). At tyrosine concentrations above 30 M, DOPA synthesis declined; inhibition was observed at higher concentrations for phenylalanine (300 M). These effects were most notable in the presence of 56 mM potassium. Measurements of intracellular phenylalanine and tyrosine suggested the Km for either amino acid is 20–30 M; maximal synthesis occurred at 120–140 M. In the presence of both phenylalanine and tyrosine, DOPA synthesis was inhibited by phenylalanine only at a high medium concentration (1000 M), regardless of medium tyrosine concentration. The inhibition of DOPA synthesis by high medium tyrosine concentrations was antagonized by high medium phenylalanine concentrations (100, 1000 M). Together, the findings indicate that for PC12 cells, phenylalanine can be a significant substrate for tyrosine hydroxylase, is a relatively weak inhibitor of the enzyme, and at high concentrations can antagonize substrate inhibition by tyrosine.  相似文献
