Two new species of Diorfilaria, Dirofilaria macrodemos and Dirofilaria panamensis, are described from the subserosa of Central and South American sloths. Dirofilaria macrodemos, described from the three-toed sloth, Bradypus tridactylus, in Guyana, is characterized and differentiated from other dirofilarias by its length (female 214mm, male 64mm), relatively long tail (female 140 micron, male 110 micron), and the number and distribution of the caudal papillae (7 pairs) in the male. Dirofilaria panamensis, parasite of the two-toed sloth, Cholopeus hoffmanni, in Panama, was previously described as Dirofilaria incrassata by Caballero (1947), but can be distinguished from both D. macrodemos and D. incrassata on the basis of body size (female 66 mm long by 360 micron wide, male 34 mm long by 250 micron wide), tail length (female 80 micron, male 80 micron), and number (6 pairs) of caudal papillae in the male.  相似文献

Comparisons were made of in vivo labeled polypeptides from Aedes aegypti strains refractory to either Brugia malayi or Dirofilaria immitis. There does not seem to be a generalized "anti-parasite" polypeptide response that mosquitoes refractory to filarial worm infection produce following bloodfeeding. Instead, it seems that any response produced by these mosquitoes is localized to the tissue in which the filarial parasite develops.  相似文献

Two new human cases of dirofilariasis, both due to Dirofilaria (Nochtiella) repens, recently occurring in Nuoro province (Sardinia) on people permanently resident in the island, are described. The parasite was located in one case into the uterine round ligament, in the other it was subconjunctival. Some observations about the diagnostic difficulties in the first one and the dramatic symptoms in the second, whose parasite was yet alive during the surgical ablaction, are discussed. These cases are the 9th and the 10th, respectively, reported in man in Sardinia, but it is possible that other cases are undiagnosed and consequently are not published.  相似文献

The present study deals with the discovery and partial characterization of specific binding proteins for retinol and retinoic acid from filarial parasites (worms of the superfamily Filarioidea), including those from two species of Onchocerca. These binding proteins, which are distinct in their physicochemical properties and in the mode of ligand interactions from the host-tissue retinoid-binding proteins, may be involved in the mediation of the putative biological roles of retinoids in the control of parasitic growth, differentiation and reproduction. Parasite retinol-binding protein and retinoic acid-binding protein exhibited specificity for binding retinol and retinoic acid respectively. Both the binding proteins showed an s20,w value of 2.0 S. On gel filtration, both proteins were retarded to a position corresponding to the same molecular size (19.0 kDa). On preparative columns, the parasite binding proteins exhibited isoelectric points at pH 5.7 and 5.75. Unlike the retinoid-binding proteins of mammalian and avian origin, the parasite retinoid-binding proteins showed a lack of mercurial sensitivity in ligand binding. The comparative amounts of retinoic acid-binding protein in five parasites, Onchocerca volvulus, Onchocerca gibsoni, Dipetalonema viteae, Brugia pahangi and Dirofilaria immitis, were between 2.7 and 3.1 pmol of retinoic acid bound/mg of extractable protein. However, the levels of parasite retinol-binding protein were between 4.8 and 5.8 pmol/mg, which is considerably higher than the corresponding levels of cellular retinol-binding protein of mammalian and avian origin. Both retinol- and retinoic acid-binding-protein levels in O. volvulus-infected human nodules and O. gibsoni-infected bovine nodules were similar to their levels in mammalian tissues. Also, these nodular binding proteins, like the host-binding proteins, exhibited mercurial sensitivity to ligand interactions.  相似文献

The purpose of this study was to identify and characterize soluble parasite antigens present in sera from humans infected with the filarial nematode Wuchereria bancrofti. Affinity chromatography and immunoblot methods were used to demonstrate a 200,000 m.w. circulating parasite antigen in sera from infected humans which corresponded to an antigen released by adult W. bancrofti during in vitro culture. Two monoclonal antibodies were produced to this antigen by immunizing mice with antigens from Dirofilaria immitis, a filarial parasite that is closely related to W. bancrofti, and screening cell fusion supernatants by enzyme immunoassay and counterimmunoelectrophoresis inhibition. The antibodies bound to a single repeated epitope (not phosphorylcholine) that was resistant to heat, acid, and protease treatments but sensitive to periodate oxidation. Immunoperoxidase studies showed that the epitope was concentrated in the cuticle and reproductive organs in D. immitis, and it was released in relatively large amounts by adult female D. immitis in vitro. The epitope is also present in antigens of other species of filarial and nonfilarial nematodes, but on the basis of preliminary studies, its presence in human serum appears to be specific for W. bancrofti infection.  相似文献

Parasitism is an important factor in conservation worldwide, especially for endangered species, as it can affect host populations by reducing growth rates, fecundity and affecting nutritional status. The maned wolf Chrysocyon brachyurus, a near-threatened species, is distributed across different habitats from the south Amazonian forest in Brazil to northern Argentina, Paraguay and eastern Bolivia. Most studies on gastrointestinal parasites in wild maned wolves are based on coprological findings, and there is little information about adult parasites and parasite species richness. Therefore, this study aims to expand the knowledge about the helminth infracommunity of a maned wolf, describe adult parasites, and explore parasitic coinfections. We performed a necropsy of an adult individual found road killed in Chaco province, Argentina. We collected adult worms from subcutaneous tissues, from the right ventricle, kidneys, and intestine. The parasites were morphologically identified as Dirofilaria immitis, Dioctophyma renale and Spirometra sp. respectively. To our knowledge, this is the first record for adults of Dirofilaria immitis in maned wolves. Filarioids were located within the heart as well as in subcutaneous tissues with mature females containing uterine microfilariae; this study also adds C. brachyurus as a new host for Spirometra sp. in Argentina. Considering that the three helminths found herein are zoonotic and that the maned wolf is an endangered canid, it is essential provide baseline information to determine the risk factors involved in the transmission and extend the studies to sympatric wild and domestic canids in the area.  相似文献

Two new cases of human dirofilariasis, occurred in women aged 46 and 52 years, respectively, both living in Lombardy (Northern Italy) are reported. Dirofilaria repens nematodes were localised in the breast in both cases. In one of them the parasite was accidentally extracted while the patient underwent a fine needle aspirate. The international literature records 30 cases of breast Dirofilariasis. Except in rare cases, the parasite was located subcutaneously in the breast, and nested in a nodule. The clinical diagnoses were consistently wrong, the nodule being diagnosed as a suspected tumor of the breast.  相似文献

Although the filarial nematode parasite Onchocerca volvulus is an important human pathogen in large areas of Africa and Latin America, little is known of the molecular interactions that govern the clinical status of patients with this chronic, debilitating disease. As a step toward defining the parasite molecules important to the immunobiology of host-parasite interactions, we have identified and cloned a major surface-associated antigen expressed by O. volvulus microfilariae. Radiolabeling experiments demonstrated that O. volvulus microfilariae have a limited repertoire of peptides at the surface. Prominent among these labeled peptides is an 18-kDa component. Immunological cross-reactivity between a surface-associated component of Dirofilaria immitis microfilariae and the 18-kDa surface-associated molecule from O. volvulus was exploited in a strategy to clone this potentially important O. volvulus microfilarial antigen. The cross-reacting antibodies were used to immunoscreen O. volvulus cDNA expression libraries. One clone, M2f.e, contained an open reading frame of 495 bp encoding an 18.1-kDa protein (OVMS18). Antibodies produced against the expression product of M2f.e recognized an 18-kDa component in extracts of O. volvulus microfilariae and bound to the surface of intact O. volvulus and D. immitis microfilariae. Southern blot analyses showed that M2f.e-like sequences are present in the genomic DNA of a number of filarial nematode species, but not in DNAs from nonfilarial nematode species.  相似文献

Cuticular markings of five species of Dirofilaria were examined by SEM. Variations in the patterns on the en face view were observed among the species. Transverse striations were observed on the cuticles of all five species, but longitudinal ridges were lacking on Dirofilaria immitis, the type species of the subgenus Dirofilaria. They were present on the other four species, Dirofilaria repens, Dirofilaria tenuis, Dirofilaria corynodes, and Dirofilaria magnilarvatum, members of the subgenus Nochtiella. Longitudinal ridges and other cuticular structures and patterns were seen to vary among the species and with the portion and aspect of the worm examined. It is suggested that SEM would be a good tool for a detailed study of specific cuticular morphology of all species of Dirofilaria, thus contributing toward their differentiation and recognition.  相似文献

Dirofilaria immitis is a worldwide filarial nematode causing heartworm disease in dogs and cats. Several mosquito species, which are able to feed both on humans and animals, can transmit this parasite. Inflammatory progression of host tissues induced by parasites are mediated by several molecules, including nitric oxide (NO), which usually exerts deleterious effects on parasites and occasionally on the host. We analyze the in vitro effect of total D. immitis adult worm somatic antigens on na?ve rat alveolar macrophage NO production and further separation of parasite proteins to define specific D. immitis somatic molecules influencing host cell NO secretion. Additionally, we address the possible influence of Wolbachia spp. on the in vitro production of NO by macrophages. Our results demonstrate that D. immitis adult worm soluble antigens are able to specifically induce NO production from host macrophages. Furthermore, we demonstrated that this effect is due to nematode antigens rather than to defined components (LPS and metabolic molecules) derived from its endosymbiont, Wolbachia spp. In addition, we were able to isolate and identify one of the parasite specific components from the DiSo extract, denominated DiID35.3 and putatively belonging to the Immunoglobulin Superfamily Protein (ISP) group, triggering NO release from macrophages in a dose-dependent and specific manner.  相似文献

A method for the identification of circulating parasite antigens in filarial nematode infections was developed using canine infections with Dirofilaria immitis as a model. Filarial antigens ranging in molecular weight from 211 to 13 kDa were extracted from the sera of microfilaremic dogs by a solid phase immunobinding procedure and identified by immunostaining of Western blots. A major antigen of 104 kDa was selected for further characterization. The 104 kDa circulating antigen showed antigenic and biochemical identity with 104 kDa peptides found in extracts of adult male and microfilarial stages of the parasite. The 104 kDa peptide was antigenically stable under a variety of storage conditions. Its potential as a diagnostic target is discussed.  相似文献

The molecules associated with the surface of adult Dirofilaria immitis were identified and characterized employing IODO-GEN-mediated surface labeling methods. D. immitis female and male parasites were found to have a limited number of surface-associated proteins (17.5, 16, and 14.5 kDa) and glycoproteins (49 and 20 kDa) which were readily extracted from parasite homogenates in the absence of detergent. The major surface labeled proteins and glycoproteins were antigenic in rabbits, but appeared to elicit only a weak humoral response in dogs with patent dirofilariasis. In addition, a 10- to 6-kDa surface-associated glycolipid was identified which may form a coat on the outside of the parasite and play a role in immune evasion. In immunoprecipitation experiments, the glycolipid was not recognized by the antibodies from rabbits exposed to the glycolipid or by antibodies in the sera of patently infected animals. The glycolipid and the 14.5-kDa surface protein were selectively released by the adult parasite during in vitro culture.  相似文献

Lamellodiscus sanfilippoi n. sp. takes place, among the other species of Lamellodiscus, in the "ergensi" sub-group (Amine et Euzet, 2005) characterized by the morphology of the dorsal lateral bars of the haptor. This sub-group comprises, in the Mediterranean, L. ergensi Euzet and Oliver, 1966, L. kechemirae Amine and Euzet, 2005, L. tomentosus Amine and Euzet, 2005, all parasite of Diplodus sargus, and L. baeri Oliver, 1974 parasite of Pagrus pagrus. L. sanfilippoi can be distinguished from the previous species by the morphology and size of the dorsal lateral bars. The new species is close to Lamellodiscus furcillatus Kritsky, Jiménez-Ruiz and Sey, 2000, a parasite of Diplodus noct in the Persian Gulf, but differs by the size of the haptoral sclerotised pieces and the morphology of the male copulatory apparatus. Lamellodiscus gussevi Sanfilippo (1978) et Lamellodiscus abbreviatus Sanfilippo (1978) are considered as nomina nuda.  相似文献

The flesh-burrowing parasitic isopod Ichthyoxenus fushanensis was found infecting the body cavity of a freshwater fish, Varicorhinus bacbatulus , in heterosexual pairs. Herein we investigate the question of how the host body size may constrain the parasite size and clutch size by analyzing the interactions among the body size and clutch size of the parasite, and host size. Due to the low transmission rate of I. fushanensis to its host and the positive relationship between clutch size and female size, selection may favor larger females with larger clutch sizes to compensate for massive losses of manca (the free-living juveniles). The path model reveals that clutch size depends not only directly on female size, but also on the sizes of her host and mate. Female size also depends on the sizes of the host and her mate. A negative correlation exists between the body sizes of the paired males and females. This negative correlation may be regarded as a consequence of competition for limited available space or other resources provided by the host. The effects of host size on parasite size, however, act on the total volume of both sexes as a whole, not specifically on either the female or the male. In this case, the available space/resources may not allow both individuals of different sexes to evolve toward a larger size simultaneously. Under the constraint of host size, a strategy of reducing the body size of the paired male may provide a way to increase the body size of the paired female and achieve a larger clutch size.  相似文献

Polystoma nacialtuneli n. sp. is described from the urinary bladder of the eastern spadefoot, Pelobates syriacus from Turkey. This is the fifth polystome species known from Turkey and the third species in Pelobates. We show that this new parasite species can be distinguished from other polystome species in the area by a combination of characteristics, including parasite size and the shape and size of the hamuli. Polystoma pelobatis from Pelobates cultripes has a pair of well-developed hamuli, while P. fuscus from Pe. fuscus characteristically has a pair of underdeveloped hamuli barely larger than the marginal hooklets. Polystoma nacialtuneli n. sp. has well-developed hamuli that vary significantly in shape. Phylogenetic relationships of P. nacialtuneli n. sp. within Polystoma, supplemented with molecular divergences estimated from internal transcribed spacer 1 (ITS1) sequences, indicate that they are well separated from their closest relatives, i.e. P. fuscus and P. pelobatis from Pe. fuscus and Pe. cultripes, respectively.  相似文献

The parasite faunas of the Patagonian toothfish Dissostichus eleginoides from six locations around the Southern Ocean were studied and compared. Thirty-two parasite taxa were found. Ten parasite species are reported from D. eleginoides for the first time and some other previously reported species were new locality records. Sample size at Shag Rocks was sufficient to examine the effect of intrinsic host factors, including sex and length, on the parasite fauna and these results are discussed here. Some parasite species were found only in certain areas. Sørensens similarity index indicated that the parasite faunas at Heard, Maquarie and Prince Edward Islands were the most similar, while those from the Ross Sea was the most dissimilar. There may be a gradual decrease in parasite diversity the further east the samples were collected around the Southern Ocean.  相似文献

Aim  Comparative studies have revealed strong links between ecological factors and the number of parasite species harboured by different hosts, but studies of different taxonomic host groups have produced inconsistent results. As a step towards understanding the general patterns of parasite species richness, we present results from a new comprehensive data base of over 7000 host–parasite combinations representing 146 species of carnivores (Mammalia: Carnivora) and 980 species of parasites.
Methods  We used both phylogenetic and non-phylogenetic comparative methods while controlling for unequal sampling effort within a multivariate framework to ascertain the main determinants of parasite species richness in carnivores.
Results  We found that body mass, population density, geographical range size and distance from the equator are correlated with overall parasite species richness in fissiped carnivores. When parasites are classified by transmission mode, body mass and home range area are the main determinants of the richness of parasites spread by close contact between hosts, and population density, geographical range size and distance from the equator account for the diversity of parasites that are not dependent on close contact. For generalist parasites, population density, geographical range size and latitude are the primary predictors of parasite species richness. We found no significant ecological correlates for the richness of specialist or vector-borne parasites.
Main conclusions  Although we found that parasite species richness increases instead of decreases with distance from the equator, other comparative patterns in carnivores support previous findings in primates, suggesting that similar ecological factors operate in both these independent evolutionary lineages.  相似文献

Impact of a Novel, Feminising Microsporidium on its Crustacean Host   总被引:1,自引:0,他引:1  
We describe the transmission and pathogenic effects of a novel, feminising microsporidium, probably a Nasema species, on its crustacean host Gammarus duebeni. The parasite prevalence in the field was high (46% of females were infected) and the parasite was transovarially transmitted to 91% of embryos of infected females. The impact of the parasite on the host was assessed by means of a host breeding experiment. The parasite feminised 66% of infected host young and was transovarially transmitted by these individuals to the next host generation. The parasite differed from other feminising microsporidia in G. duebeni in that early embryos had a high parasite burden (288 parasites per embryo) and the infection was pathogenic, causing a reduction in both the growth rate of young hosts and in adult size. This study suggests that feminising microsporidia are a diverse group in which a variety of host/pathogen relationships have evolved.  相似文献

A method of affinity chromatography developed for the purification of species-specific antigens from Toxocara canis adult worms is described. Immunochemical analyses by polyacrylamide gel electrophoresis, immunoelectrophoresis and immunodiffusion showed that ‘pure’ antigen contained fewer but more specific proteins than ‘crude’ antigen. Purified antigens and parasite sections from four parasite species (Toxocara canis, Dirofilaria immitis, Angiostrongylus cantonensis and Ascaris lumbricoides) were used in immunofluorescence tests to measure serum antibody levels in animals with natural or experimental T. canis infections and people with zoonotic toxocariasis. ‘Pure’ antigen showed higher specificity and sensitivity than ‘crude’ antigen in serological testing.  相似文献
