Response of 13 alfalfa (Medicago sativa L.) genotypes to varied Zn supply (+Zn: 2 mg kg−1 soil, −Zn: no added Zn) was studied in a pot experiment under controlled environmental conditions. Plants were grown for
four weeks in a Zn-deficient siliceous sandy soil. Plants grown at no added Zn showed typical Zn deficiency symptoms i.e.
interveinal chlorosis of leaves, yellowish-white necrotic lesions on leaf blades, necrosis of leaf margins, smaller leaves
and a marked reduction in growth. There was solute leakage from the leaves of Zn-deficient plants, while no solute leakage
from Zn-sufficient plants. The ratios of P:Zn, Fe:Zn, Cu:Zn and Mn:Zn in Zn-deficient plants were extremely high compared
with Zn-sufficient plants indicating disturbance of P:Zn, Fe:Zn, Cu:Zn and Mn:Zn balance within plant system by Zn deficiency.
Genotypes differed markedly in Zn efficiency based on shoot dry matter production. Alfalfa genotypes also differed markedly
in P:Zn ratio, Cu:Zn ratio and Fe:Zn ratio under —Zn treatment. The shoot dry weight, shoot:root ratio, chlorophyll content
of fresh leaf tissue, solute leakage from the leaves, Zn uptake and distribution of Zn in shoots and roots were the most sensitive
parameters of Zn efficiency. Zn-efficient genotypes had less solute leakage but higher shoot:root ratio and higher Zn uptake
compared with Zn-inefficient genotypes. Under —Zn treatment, Zn-inefficient genotypes had less Zn partitioning to shoots (33–37%)
and more Zn retained in roots (63–67%), while Zn-efficient genotypes had about equal proportions of Zn in roots (50%) and
shoots (50%).

The effect of boron (B), zinc (Zn), and manganese (Mn) deficiency in the medium on eight soybean genotypes was evaluated using friable callus after a 21 day growth period. Of the three micronutrients evaluated, Zn and B deficiencies had the greatest effect on callus weight, while Mn had only a slight effect. Despite this, significant differences in callus weight reduction of the eight genotypes tested were observed on the three different media. These results indicate that genotypic variation for response to B, Zn, and Mn deficiency is present in soybean at the cellular level. If the resistant genotypes identified in this study are also resistant at the whole plant level, then they could be used in soils which are deficient in the above micronutrients. 相似文献

Antioxidant responses and nodule function of Medicago truncatula genotypes differing in salt tolerance were studied. Salinity effects on nodules were analysed on key nitrogen fixation proteins such as nitrogenase and leghaemoglobin as well as estimating lipid peroxidation levels, and were found more dramatic in the salt-sensitive genotype. Antioxidant enzyme assays for catalase (CAT, EC 1.11.1.6), superoxide dismutase (EC 1.15.1.1), ascorbate peroxidase (EC 1.11.1.11) and guaiacol peroxidase (EC 1.11.1.7) were analysed in nodules, roots and leaves treated with increasing concentrations of NaCl for 24 and 48 h. Symbiosis tolerance level, depending essentially on plant genotype, was closely correlated with differences of enzyme activities, which increased in response to salt stress in nodules (except CAT) and roots, whereas a complex pattern was observed in leaves. Gene expression responses were generally correlated with enzymatic activities in 24-h treated roots in all genotypes. This correlation was lost after 48 h of treatment for the sensitive and the reference genotypes, but it remained positively significant for the tolerant one that manifested a high induction for all tested genes after 48 h of treatment. Indeed, tolerance behaviour could be related to the induction of antioxidant genes in plant roots, leading to more efficient enzyme stimulation and protection. High induction of CAT gene was also distinct in roots of the tolerant genotype and merits further consideration. Thus, part of the salinity tolerance in M. truncatula is related to induction and sustained expression of highly regulated antioxidant mechanisms. 相似文献

Zinc deficiency and salinity are well-documented soil problems and often occur simultaneously in cultivated soils. Usually, plants respond to environmental stress factors by activating their antioxidative defense mechanisms. The antioxidative response of wheat genotypes to salinity in relation to Zn nutrition is not well understood. So, we investigated the effect of Zn nutrition on the growth, membrane permeability and sulfhydryl group (–SH groups) content of root cells and antioxidative defense mechanisms of wheat plants exposed to salt stress. In a hydroponic experiment, three bread wheat genotypes (Triticum aestivum L. cvs. Rushan, Kavir, and Cross) with different Zn-deficiency tolerance were exposed to adequate (1 μM Zn) and deficient (no Zn) Zn supply and three salinity levels (0, 60, and 120 mM NaCl). The results obtained showed that adequate Zn nutrition counteracted the detrimental effect of 60 mM NaCl level on the growth of all three wheat genotypes while it had no effect on the root and shoot growth of ‘Rushan’ and ‘Kavir’ at the 120 mM NaCl treatment. At the 0 and 60 mM NaCl treatments, Zn application decreased root membrane permeability while increased –SH group content and root activity of catalase (CAT) and superoxide dismutase (SOD) in ‘Rushan’ and ‘Kavir’. In contrast, Zn had no effect on the root membrane permeability and –SH group content of ‘Rushan’ and ‘Kavir’ exposed to the 120 mM NaCl treatment. At all salinity levels, ‘Cross’ plants supplied with Zn had lower root membrane permeability and higher –SH group content compared to those grown under Zn-deficient conditions. At the 0 and 60 salinity levels, Zn-deficient roots of Kavir and Rushan genotype leaked significantly higher amounts of Fe and K than the Zn-sufficient roots. In contrast, at the 120 mM treatment, Zn application had no effect or slightly increased Fe and K concentration in the root ion leakage of these wheat genotypes. For ‘Cross’, at all salinity levels, Zn-deficient roots leaked significantly higher amounts of Fe and K compared with the Zn-sufficient roots. The differential tolerance to salt stress among wheat genotypes examined in this study was related to their tolerance to Zn-deficiency, –SH group content, and root activity of CAT and SOD. Greater tolerance to salinity of Zn-deficiency tolerant genotype ‘Cross’ is probably associated with its greater antioxidative defense capacity. 相似文献

Summary Nine genotypes of sorghum were screened for their relative susceptibility to zinc deficiency in soil. All the varieties were highly responsive to zinc application, but behaved differently under zinc stress conditions in respect of dry-matter production, zinc concentration and uptake. The effects of nitrogen source on iron deficiency responses were investigated in two Vitis genotypes, one tolerant to limestone chlorosis Cabernet Sauvignon (Vitis vinifera cv.) and the other susceptible Gloire de Montpellier (Vitis riparia cv.). Plants were grown with or without Fe(III)-EDTA, and with NO3− alone or a mixture of NO3− and NH4+. Changes in pH of the nutrient solution and root ferric chelate reductase (FC-R) activity were monitored over one week. We
carried out quantitative metabolic profiling (1H-NMR) and determined the activity of enzymes involved in organic acid metabolism in root tips. In iron free-solutions, with
NO3− as the sole nitrogen source, the typical Fe-deficiency response reactions as acidification of the growth medium and enhanced
FC-R activity in the roots were observed only in the tolerant genotype. Under the same nutritional conditions, organic acid
accumulation (mainly citrate and malate) was found for both genotypes. In the presence of NH4+, the sensitive genotype displayed some decrease in pH of the growth medium and an increase in FC-R activity. For both genotypes,
the presence of NH4+ ions decreased significantly the organic acid content of roots. Both Vitis genotypes were able to take up NH4+ from the nutrient solution, regardless of their sensitivity to iron deficiency. Reactive oxygen species (ROS) and reactive nitrogen species (RNS) are generated throughout the human body. Enzymatic and nonenzymatic antioxidants detoxify ROS and RNS and minimize damage to biomolecules. An imbalance between the production of ROS and RNS and antioxidant capacity leads to a state of "oxidative stress" that contributes to the pathogenesis of a number of human diseases by damaging lipids, protein, and DNA. In general, lung diseases are related to inflammatory processes that generate increased ROS and RNS. The susceptibility of the lung to oxidative injury depends largely on its ability to upregulate protective ROS and RNS scavenging systems. Unfortunately, the primary intracellular antioxidants are expressed at low levels in the human lung and are not acutely induced when exposed to oxidative stresses such as cigarette smoke and hyperoxia. However, the response of extracellular antioxidant enzymes, the critical primary defense against exogenous oxidative stress, increases rapidly and in proportion to oxidative stress. In this paper, we review how antioxidants in the lung respond to oxidative stress in several lung diseases and focus on the mechanisms that upregulate extracellular glutathione peroxidase. 相似文献

Plants are routinely subjected to multiple environmental stresses that constrain growth. Zinc (Zn) deficiency and high bicarbonate are two examples that co-occur in many soils used for rice production. Here, the utility of metabolomics in diagnosing the effect of each stress alone and in combination on rice root function is demonstrated, with potential stress tolerance indicators identified through the use of contrasting genotypes. Responses to the dual stress of combined Zn deficiency and bicarbonate excess included greater root solute leakage, reduced dry matter production, lower monosaccharide accumulation and increased concentrations of hydrogen peroxide, phenolics, peroxidase and N-rich metabolites in roots. Both hydrogen peroxide concentration and root solute leakage were correlated with higher levels of citrate, allantoin and stigmasterol. Zn stress resulted in lower levels of the tricarboxylic acid (TCA) cycle intermediate succinate and the aromatic amino acid tyrosine. Bicarbonate stress reduced shoot iron (Fe) concentrations, which was reflected by lower Fe-dependent ascorbate peroxidase activity. Bicarbonate stress also favoured the accumulation of the TCA cycle intermediates malate, fumarate and succinate, along with the non-polar amino acid tyrosine. Genotypic differentiation revealed constitutively higher levels of D-gluconate, 2-oxoglutarate and two unidentified compounds in the Zn-efficient line RIL46 than the Zn-inefficient cultivar IR74, suggesting a possible role for these metabolites in overcoming oxidative stress or improving metal re-distribution. 相似文献

The antioxidant response of four freshwater microalgal species, Chlorella vulgaris Beij., Scenedesmus platydiscus (G. M. Smith) Chod., Scenedesmus quadricauda(Turp.) Bréb., and Selenastrum capricornutum Printz without pyrene addition (control) and at two pyrene concentrations (0.1 and 1.0 mgL−1) were investigated. Under the control condition, the values of the antioxidant parameters differed significantly among species and the difference was seemed not to be related to their susceptibility to pyrene. The antioxidant response to pyrene treatments also varied from species to species. Pyrene led to a significant increase in total glutathione (GSH) content in all species except C. vulgaris, a species did not exhibit any ability to metabolize pyrene. The glutathione-S-transferase (GST) activities also remained unchanged in pyrene treated C. vulgaris, increased greatly in S. platydiscus and Se. capricornutum (the two species with higher pyrene metabolism ability), but inhibited remarkably in S. quadricauda (the only species sensitive to pyrene toxicity). On the other hand, the glutathione reductase (GR) activities increased in C. vulgaris but remained at a similar level as the control in the other three species. The malondialdehyde (MDA) content, an indicator of lipid peroxidation, declined in S. quadricauda but showed no significant change in the other three species. The activities of glutathione peroxidase (GPX), superoxide dismutase (SOD) and catalase (CAT) in pyrene treated cells remained almost the same as the controls for all microalgal species. Results suggested that the alterations of antioxidant systems in microalgae might not be useful indicators of pyrene exposure but pyrene-enhanced GSH metabolism might be important in pyrene biotransformation. 相似文献

Ten genotypes from Pisum sativum and Pisum arvense were screened for their regeneration abilities. Most of them were created
through experimental mutagenesis from Bulgarian varieties and have various valuable agronomic traits. Embryonic axes from
immature embryos were plated on modified Murashige and Skoog medium, containing different concentrations of 2,4-dichlorophenoxyacetic
acid (2,4-d), α-naphthaleneacetic acid (NAA) and benzyladenine (BA). Two schemes for direct and indirect organogenesis were
established. Callus and shoot formation were induced on media containing 0.2 mM 2,4-d or 5 mM BA, respectively. Embryonic
axes formed buds directly when plated on medium with 10 mM BA and 1 mM NAA. Organogenesis and adventitious bud formation were
maintained on medium supplemented with BA and NAA. Rhizogenesis was induced on Gamborgs' B5 medium. All screened genotypes
were able to regenerate plants with a high efficiency (50–100%) although some differences in their organogenetic response
were observed.

Chickpea genotypes differ in their sensitivity to Zn deficiency 总被引:3,自引:1,他引:3
Zinc (Zn) deficiency is common in most of the chickpea growing areas of the world and growing Zn-efficient genotypes on Zn-deficient soil is a benign approach of universal interest. Response of 13 chickpea genotypes (10 desi and 3 kabuli) to Zn nutrition was studied in a pot experiment under glasshouse conditions. Plants were grown in a Zn-deficient siliceous sand for 6 weeks and fertilized with 0 (Zn–) and 2.5 mg Zn per kg soil (Zn+). When grown with no added Zn, Zn deficiency symptoms (chlorosis of younger leaves and stipules followed by necrosis of leaf margins) appeared 3–4 weeks after planting and were more apparent in cultivars Tyson, Amethyst and Dooen than Kaniva and T-1587. Zn deficiency reduced shoot growth, but it was less affected in breeding lines T-1587 and CTS 11308 than cultivars Tyson, Dooen, Amethyst and Barwon. Among all genotypes, Tyson produced the lowest root dry weight in Zn– treatment. Zinc efficiency based on shoot dry weight showed marked differences among genotypes; breeding lines CTS-60543, CTS-11308 and T-1587 were 2-fold more Zn-efficient than cultivars Tyson and Dooen. A higher Zn accumulation per plant and higher Zn uptake per g. of root dry weight were recorded in T-1587 and CTS-11308 when compared with Tyson. Root:shoot ratio was increased and proportionally more Zn was transported to the shoot when the soil was deficient. Cultivars that were very sensitive to Zn deficiency tended to have their root:shoot ratio increased by Zn deficiency more than less sensitive cultivars. The insensitive lines T-1587 and CTS-11308 transported more than 70% of the total absorbed Zn to the shoot. It is concluded that chickpea genotypes vary in their sensitivity to Zn deficiency. Advanced breeding lines T-1587 and CTS-11308 are relatively more Zn-efficient compared with Australian chickpea cultivar Tyson. Zn efficiency in chickpea genotypes is probably related to an efficient Zn absorption coupled with a better root to shoot transport. 相似文献

Gas exchange rates, chlorophyll fluorescence, pressure–volume relationships, photosynthetic pigments, total soluble sugars,
starch, soluble proteins and proline concentrations were investigated in five Olea europaea L. cultivars with different geographical origins (Arbequina, Blanqueta, Cobran?osa, Manzanilla and Negrinha) grown under
Mediterranean field conditions. We found considerable genotypic differences among the cultivars. Comparing the diurnal gas
exchange rates, we observed that Cobran?osa, Manzanilla and Negrinha had high photosynthetic rate than Arbequina and Blanqueta.
The first group reveals to be better acclimated to drought conditions, and appears to employ a prodigal water-use strategy,
whereas Blanqueta and Arbequina, with high water-use efficiency, appear to employ a conservative water-use strategy. The degree
of midday depression in photosynthesis was genotype dependent, with a maximum in Arbequina and a minimum in Negrinha. The
reductions in the photosynthetic rate were dependent from both stomatal and non-stomatal limitations. Elastic adjustment plays an important role as drought tolerance mechanism. The group of cultivars that employ a prodigal
water-use strategy revealed high tissue elasticity, whereas Arbequina and Blanqueta revealed high tissue rigidity. We also
identified the existence of drought tolerance mechanisms associated with soluble proteins accumulation in the foliage. The
high levels of soluble proteins in Arbequina may represent an increased activity of oxidative stress defence enzymes and may
also represent a reserve for post stress recovery. In all cultivars, especially in Manzanilla, free proline was accumulated
in the foliage. The discussed aspects of drought stress metabolism may have an adaptative meaning, supporting the hypothesis
that olive cultivars native to dry regions, such as Cobran?osa, Manzanilla and Negrinha, have more capability to acclimate
to drought conditions than cultivars originated in regions with a more temperate climate, like Arbequina and Blanqueta. 相似文献

The variability of the developmental responses of two contrasting cultivars of pea (Pisum sativum) was studied in relation to the genetic diversity of their nitrogen-fixing symbiont Rhizobium leguminosarum bv. viciae. A sample of 42 strains of pea rhizobia was chosen to represent 17 genotypes predominating in indigenous rhizobial populations, the genotypes being defined by the combination of haplotypes characterized with rDNA intergenic spacer and nodD gene regions as markers. We found contrasting effects of the bacterial genotype, especially the nod gene type, on the development of nodules, roots and shoots. A bacterial nod gene type was identified that induced very large, branched nodules, smaller nodule numbers, high nodule biomass, but reduced root and aerial part development. The plants associated with this genotype accumulated less N in shoots, but N concentration in leaves was not affected. The results suggest that the plant could not control nodule development sustaining the energy demand for nodule functioning and its optimal growth. The molecular and physiological mechanisms that may be involved are discussed. 相似文献

Zinc has been shown to effect--in vitro--a number of processes associated with neurotransmission. We have tested whether the rate of impulse conduction--in vivo--as measured from the latencies of auditory brainstem responses (ABR), is influenced by dietary zinc deficiency in the rat. Dietary zinc deficiency for up to 26 wk had no effect on the wave I-IV interval compared to zinc-adequate fed animals. The results are discussed in relation to the observed constancy of brain overall and extracellular fluid zinc concentrations under conditions of dietary zinc deficiency. 相似文献

Summary In order to examine the differential response of tropical maize genotypes to Zn and Mn nutrition, a pot experiment was conducted
and the results subsequently evaluated in a field trial. Zn was rendered ‘physiologically immobile’ in the root tissue as
revealed by a substantial reduction in shoot concentration as compared to root concentration. This effect was most pronounced
in the inbred line CM-111. Quite contrasting to Zn, the root concentration of Mn was substantially lower while the shoot concentration
was comparable with that of Zn indicating that under identical supply of Zn and Mn to substrate, more of Zn is root absorbed
and also ‘root immobilized’, while, of the quantities absorbed by root, relatively more of Mn is translocated to shoot. Maximum
root and shoot dry matter yields were obtained at specific Zn/Mn ratio in each of the genotype studied. Available Mn estimated
at periodic interval (15 days) during plant growth showed consistent increase corresponding to higher rates applied while
available Zn showed an initial (on 15th day of sampling) decrease corresponding to higher rates (possibly initial reversion?)
and subsequent (on 45th day of sampling) increase (possibly later release through plant root activity in rhizosphere?). Significant
and positive correlation coefficients were obtained between available Zn and root Zn while in the case of available Mn it
was so only with shoot Mn. In field experiment Ganga-5 outyielded all the other genotypes and showed a positive response to
Mn application.

Spring wheat (Triticum aestivum L.) and pea (Pisum spp.) genotypes were tested for reaction to root inoculation with rhizosphere bacteria affecting plant growth. Plant response was studied in greenhouse experiments after treatment of seedlings with bacteria suspended in nutrient broth. Significant genotype variation was found in both wheat and pea in terms of shoot dry weight and severity of bacteria-induced leaf symptoms. For most bacterial isolates tested, there was good correlation between ratings of leaf symptoms 7 to 14 days after inoculation and growth inhibition measured after four weeks. Interactions between isolates and plant genotypes were significant in both wheat and pea (P=0.0024 and 0.0001, respectively), but genotypes with sensitivity or tolerance to most isolates could be distinguished. In an outdoor pot experiment, two of the bacterial isolates caused delayed plant development and differential decreases in grain yield of wheat genotypes. The hypothesis that the reaction of wheat genotypes to the tested becteria was related to their influence on bacterial establishment in the rhizosphere could not be substantiated. 相似文献
