Summary
Aspergillus niger SZ was grown under citric acid accumulation conditions in a 1l bioreactor. Radioactive citric acid was added during the fermentation to the culture medium. The accumulation of radioactive citric acid in the mycellium was measure by measuring radioactive CO2 in the offgas. Evidence was obtained of significant turnover and metabolism of exogenous citric acid even during the phase of maximum accumulation into the medium. 相似文献

In view of the often-cited theory that citric acid accumulation is caused by an inhibition of aconitase activity, the equilibrium of the reaction of aconitase was investigated by comparing in vivo steady-state concentrations of citrate and isocitrate in Aspergillus niger grown under various citric acid-producing conditions. With the equilibrium catalyzed by the A. niger enzyme in vitro, similar values were obtained. The validity of our in vivo measurements was verified by the addition of the aconitase inhibitor fluorocitrate, which appreciably elevated the citrate:isocitrate ratio. The results strongly argue against an inhibition of aconitase during citric acid fermentation. 相似文献

Highest yields were obtained with the cane juice and synthetic medium. The rate of stirring had a pronounced influence on the morphology of A. niger. 相似文献

Citric acid production by Aspergillus niger NCIM 548 and Candida lipolytica NCIM 3472 has been studied in shake culture using glucose and molasses as carbon sources. Methanol addition (3% v/v) at 40 h of fermentation enhanced the production of citric acid by Aspergillus niger whereas a reduction in citric acid production by Candida lipolytica was observed with addition of methanol. Maximum citric acid concentration of 12 kg/m3 was obtained with Aspergillus niger using molasses in the presence of methanol, while maximum citric acid concentration of 8.4 kg/m3 was obtained with Candida lipolytica using glucose without methanol. It appears that product formation by Aspergillus niger is either non-growth associated or partially growth associated depending on the substrate. Methanol addition changes the nature of product formation in case of Candida lipolytica. 相似文献

Summary Quantitative balances have been made for sugar and oxygen uptake rates during citric acid accumulation by Aspergillus niger: during the first phase of citric acid accumulation (up to 130 h) more sugar is taken up than the production of biomass, CO2 and citric acid account for. In contrast, during later phases of fermentation more citric acid, CO2 and biomass are formed than sugar uptake would theoretically allow. A similar pattern is obtained for oxygen uptake, where less uptake occurs during the early phase of fermentation than needed for complete balance, and the reverse is observed during the late stage of fermentation. It could subsequently be shown that this is caused by the intermediate accumulation and partial re-consumption of a number of polyhydric alcohols (glycerol, arabitol, erythritol and mannitol) during citric acid fermentation.Dedicated to Professor H. Summary Two strains of Aspergillus niger showed a 57% and 40% reduction in growth in a shaken mineral medium with 5% glucose, at the initial pH 6.5–6.0 where ammonium acetate (0.25% and 0.2%) was a source of nitrogen. Uptake of glucose was not inhibited and mycelium kept its acid-forming activity. Growth inhibition was abolished in the presence of 12% glucose or increased content of ammonium and sodium acetate up to 765 mg acetate anion per 100 cm3.This work was a part of Project MR II, 17, topic no. 2.3.2. 相似文献

The effect of nitriloacetic acid (NTA) and 8-hydroxyquinoline on the production of citric acid by Aspergillus niger was investigated. The complexing agents showed an effect only during inoculation of the microorganism. Subsequent addition after inoculation did not produce any significant increase in citric acid yield. When 200 ppm of NTA was added during inoculation, an increase of 10.9 g·dm−3 citric acid over that produced by the control culture was observed. 8-Hydroxyquinoline, on the other hand, produced a higher concentration of citric acid which was 34 g·dm−3 over that of the control culture. The use of 8-hydroxyquinoline is therefore suggested for the production of citric acid from molasses using Aspergillus niger. 相似文献

Experiments performed in batch fermentation under phosphate-limited growth conditions showed that the citric acid yield was inversely related to the excess nitrogen concentration in the medium. Results from chemostat culture confirmed a negative relationship between the citric acid yield and both the specific growth rate and the nitrogen consumption rate. This is evidence for nitrogen catabolite repression. A fed-batch fermentation performed under dual phosphate/nitrogen limitation produced results very similar to those from a culture limited by nitrogen alone. There is no advantage in maintaining an excess of phosphate during citric acid production and the process will therefore be more economic when operated under dual limitation conditions. 相似文献

Stoichiometric modeling of the early stages of the citric acid fermentation process by Aspergillus niger revealed that ammonium ions combine with a carbon-containing metabolite inside the cell, in a ratio 1:1, to form a nitrogen compound which is then excreted by the mycelium. High-performance liquid chromatography analysis identified glucosamine as the product of the relationship between glucose and ammonium during the early stages of the citric acid fermentation process. Slightly acidic internal pHs, extremely low ammonium ion concentrations inside the cell, and glucosamine synthesis come into direct contradiction with the earlier theory of the ammonium pool inside the cell, regarded as responsible for inhibition of the enzyme phosphofructokinase. At later fermentation stages, when the mycelium is involved in a process of fragmentation and regrowth, the addition of ammonium sulfate leads to a series of events: the formation and secretion of glucosamine in elevated amounts, the short inhibition of citrate synthesis, growth enhancement, the utilization of glucosamine, and finally, the enhancement of citric acid production rates. Obviously, the enzymatic processes underlining the phenomena need to be reexamined. As a by-product of the citric acid fermentation, glucosamine is reported for the first time here. Suitable process manipulations of the system described in this work could lead to successful glucosamine recovery at the point of its highest yield before degradation by the fungus occurs. 相似文献

Summary This article introduces an easy to handle immobilization apparatus for the entrapment of microbial cells, organelles and enzymes in spherical gel beads.Ca-alginate beads with entrapped cells of Aspergillus niger showed typical shrinking behaviour (from 3.00 mm to 2.25 mm particle diameter). A loss of stability down to 20% of the initial strength during precultivation of the fungus and within the following citric acid production occurred. The observed particle shrinkage was due to the increasing acidification of the medium, whereas the decreased mechanical strength was caused by the entrapped growing microorganism. This was confirmed by electron scanning micrographs, indicating a sponge-like gel structure within the region of enhanced mycelium growth which reduced diffusional resistance of the matrix. Therefore no differences were found between citric acid production of Ca-alginate entrapped Aspergillus niger at 3 mm and 1.5 mm initial particle size. 相似文献

The effect of glucose concentration on glycolytic metabolism under conditions of citric acid accumulation by Aspergillus niger was studied with 13C-labelled glucose. The results show that during cultivation at high glucose (14%, w/v), most of the label in citric acid is in C-2/C-4, and is thus due to the pyruvate carboxylase reaction. However, a significant portion is also present in C-1/C-5, whose origin is less clear but most likely due to reconsumption of glycerol and erythritol. Formation of trehalose and mannitol is high during the early phase of fermentation and declines thereafter. The early fermentation phase is further characterized by a high rate of anaplerosis from oxaloacetate to pyruvate, which also decreases with time. At low glucose concentrations (2%, w/v), which lead to a significantly reduced citric acid yield and formation rate, labelling of citrate in C-2/C-4 is decreased and C-l/C-5 labelling increased. Growth on 2% glucose is also characterized by an appreciable scrambling of mannitol and considerable backflux from mannitol to trehalose (indicating tight glycolytic control at the fructose-6-phosphate step) and an increased anaplerotic formation of pyruvate from oxaloacetate. These data indicate that cultivation on high sugar concentrations shifts control of glycolysis from fructose-6-phosphate to the glyceraldehyde-3-phosphate dehydrogenase step. 相似文献

A study was performed to understand the physiology and biochemical mechanism of citric acid accumulation during solid state fermentation of sweet potato using Aspergillus niger Yang No.2. A low citrate-producing mutant was isolated followed by a comparative study of the fermentation process and selected physiological and biochemical parameters. In contrast with the parent strain, the mutant strain displayed lower concentrations, yields and production rates of citric acid, accompanied by higher concentrations, yields and production rates of oxalic acid. In addition, the mutant utilized starch at a lower rate although higher concentrations of free glucose accumulated in the cultures. Biochemical analyses revealed lower rates of glucose uptake and hexokinase activity of the mutant strain in comparison with the parent strain. It is proposed that, in common with submerged fermentation, over-production of citric acid in solid state fermentation is related to an increased glucose flux through glycolysis. At low glucose fluxes, oxalic acid is accumulated. 相似文献

Stationary cultures of Aspergillus niger grown on a synthetic medium have been used to study the effect of some metabolic inhibitors on citric acid production. Addition of 0.05 to 1 mM sodium malonate or 0.01 to 0.1 mM potassium ferricyanide, iodoacetate, sodium azide, sodium arsenate or sodium fluoride stimulated citric acid production (3.6 to 45%), but not total titratable acids. Addition of higher concentrations (0.2 to 10 mM) of later inhibitors caused a marked inhibition of fungal growth and citric acid production. Natural oils with high unsaturated fatty acids content when added at concentrations of 2% and 4% (v/v) to beet molasses (BM) medium caused a considerable increase in citric acid yield from Aspergillus niger. Summary We report the derivation of Aspergillus niger strains having a lower standard deviation in citric acid production by the use of 60Co gamma radiation and by the parasexual cycle. On the basis of the results obtained by isolating diploids and segregants, diploidization seems to be sufficient for reducing variability.
