地下深部油藏通常为高温、高压以及高盐的极端环境,含有非常丰富的本源嗜热厌氧微生物,按代谢类群可分为发酵细菌、硫酸盐还原菌、产甲烷古菌和铁还原菌。从油田环境已经分离出90株铁还原微生物,如热袍菌目、热厌氧杆菌目、脱铁杆菌目、δ-变形菌纲脱硫单胞菌目、γ-变形菌纲希瓦氏菌属和广古菌门栖热球菌属等,这些菌株生长温度范围为4-85°C,生长盐度范围为0.1%-10.0%NaCl,还未见到文献报道油藏铁还原菌的耐压性研究。在油藏环境中存在微生物、矿物和流体(油/水)三者之间的相互作用,油藏中的粘土矿物能够作为微生物生命活动的载体,也能为微生物代谢作用提供电子受体。本文综述了油藏铁还原菌分离和表征的研究进展,简述了油藏铁还原菌的环境适用性,并展望了铁还原菌在提高原油采收率方面的应用前景。  相似文献

Ontogenesis of osmotic regulation in the striped mullet, Mugil cephalus L.   总被引:1,自引:0,他引:1  
The osmoregulatory capabilities of juvenile striped mullet, Mugil cephalus L ., of three size-groups (20–29, 30–39 and 40–9 mm s.l.) were compared in a series of six salinities ranging from fresh water to full sea water. The two smaller size-groups were able to tolerate instantaneous transfer from the brackish water in which captured, to all salinities but fresh water, while the 40–69 mm group were tolerant of instantaneous transfer to all experimental salinities. At high environmental salinities, the osmotic regulatory capabilities improved with growth in size to an apparently definitive condition in fish of 40–69 mm s.l. The smallest of these individuals were estimated to be about 7.5 months old. The osmotic regulatory capability in waters of low salinity had reached a definitive state with the development of tolerance to such salinities. The ontogenetic pattern for Mugil cephalus is virtually identical to that of the Atlantic salmon, Salmo salar .  相似文献

Temperature and salinity tolerances of juvenile Rhabdosargus holubi (Steindachner) were determined experimentally. Results indicated that they are tolerant over a wide range of salinities (0.7–70‰) and temperatures (10–30°C) and that little interaction between temperature and salinity occurs. R. holubi is restricted to the warm temperate waters of southeastern Africa. It is absent from the subtropical Indian Ocean and from temperate Atlantic Ocean waters. The juveniles occur mainly in the inshore waters and estuaries. Evidence from tolerance experiments and salinity and temperature data from southern African estuaries suggested that the distribution of R. holubi is controlled by temperature.  相似文献

Cyanobacterial mats of the Arabian Gulf coast of Saudi Arabia experience extreme conditions of temperature and salinity. Because they are exposed to continuous oil pollution, they form ideal models for biodegradation under extreme conditions. We investigated the bacterial diversity of these mats using denaturing gradient gel electrophoresis and 16S rRNA cloning, and tested their potential to degrade petroleum compounds at various salinities (fresh water to 16%) and temperatures (5 to 50 degrees C). Cloning revealed that c. 15% of the obtained sequences were related to unknown, possibly novel bacteria. Bacteria belonging to Beta-, Gamma- and Deltaproteobacteria, Cytophaga-Flavobacterium-Bacteroides group and Spirochetes, were detected. The biodegradation of petroleum compounds at different salinities by mat microorganisms showed that pristine and n-octadecane were optimally degraded at salinities between 5 and 12% (weight per volume NaCl) whereas the optimum degradation of phenanthrene and dibenzothiophene was at 3.5% salinity. The latter compounds were also degradable at 8% salinity. The same compounds were degraded at temperatures between 15 and 40 degrees C but not at 5 and 50 degrees C. The optimum temperature of degradation was 28-40 degrees C for both aliphatics and aromatics. We conclude that the studied microbial mats from Saudi Arabia are rich in novel halotolerant and thermotolerant microorganisms with the potential to degrade petroleum compounds at elevated salinities and temperatures.  相似文献

The diversity of thermophilic microbial assemblages detected within two neighboring high temperature petroleum formations was shown to closely parallel the different geochemical regimes existing in each. A high percentage of archaeal 16S rRNA gene sequences, related to thermophilic aceticlastic and hydrogenotrophic methanogens, were detected in the natural gas producing Rincon Formation. In contrast, rRNA gene libraries from the highly sulfidogenic Monterey Formation contained primarily sulfur-utilizing and fermentative archaea and bacteria. In addition to the variations in microbial community structure, microbial activities measured in microcosm experiments using high temperature production fluids from oil-bearing formations also demonstrated fundamental differences in the terminal respiratory and redox processes. Provided with the same suite of basic energy substrates, production fluids from the South Elwood Rincon Formation actively generated methane, while thermophilic microflora within the Monterey production fluids were dominated by hydrogen sulfide producing microorganisms. In both cases, molecular hydrogen appeared to play a central role in the stimulation of carbon and sulfur cycling in these systems. In methanogenic production fluids, the addition of sulfur compounds induced a rapid shift in the terminal electron accepting process, stimulating hydrogen sulfide formation and illustrating the metabolic versatility of the subsurface thermophilic assemblage. The high similarity between microbial community structure and activity corresponding with the prevalent geochemical conditions observed in deep subsurface petroleum reservoirs suggests that the resident microflora have adapted to the subsurface physicochemical conditions and may actively influence the geochemical environment in situ.  相似文献

Physiological response of tropical organisms to salinity changes was studied for some marine, estuarine and freshwater fishes (Astyanax bimaculatus, Petenia karussii, Cyprinodon dearborni, and Oreochromis mossambicus), marine and freshwater crustaceans (Penaeus brasiliensis, Penaeus schmitti and Macrobrachium carcinus), and marine bivalves (Perna perna, Crassostrea rhizophorae, and Arca zebra) collected from Northeast Venezuela. They were acclimated for four weeks at various salinities, and (1) placed at high salinities to determine mean lethal salinity, (2) tested by increasing salinity 5@1000 per day to define upper lethal salinity tolerance limit, or (3) observed in a saline gradient tank to determine salinity preference. Acclimation level was the most significant factor. This phenomenon is important for tropical aquatic organisms in shallow waters, where they can adapt to high salinity during the dry season and cannot lose their acclimation level at low salinity during abrupt rain. For saline adaptation of tropical organisms, this behavior will contribute to their proliferation and distribution in fluctuating salinity environments.  相似文献

Temperature and salinity tolerances of Tilapia rendalli were determined experimentally. Results indicate that they are tolerant over a wide range of temperatures (11-37°C), but are incapable of surviving in salinities above 19 ‰ The maximum salinity tolerance is at temperatures between 20–28°C. The osmotic concentration of the blood rises from 255 mosmol/1 in freshwater to 340 mosmol/1 in a salinity of 19‰ T. rendalli is restricted to the warmer waters of the Zambezi river system in central Africa and southwards to the Pongolo river, as well as certain tropical and subtropical brackish water lagoons and lakes. Evidence from these experiments suggest that the distribution of T. rendalli isgoverned by both temperature and salinity.  相似文献

Methanogenesis was investigated in formation waters from a North Sea oil rimmed gas accumulation containing biodegraded oil, which has not been subject to seawater injection. Activity and growth of hydrogenotrophic methanogens was measured but acetoclastic methanogenesis was not detected. Hydrogenotrophic methanogens showed activity between 40 and 80°C with a temperature optimum (ca. 70°C) consistent with in situ reservoir temperatures. They were also active over a broad salinity range, up to and consistent with the high salinity of the waters (90 g l−1). These findings suggest the methanogens are indigenous to the reservoir. The conversion of H2 and CO2 to CH4 in methanogenic enrichments was enhanced by the addition of inorganic nutrients and was correlated with cell growth. Addition of yeast extract also stimulated methanogenesis. Archaeal 16S rRNA gene sequences recovered from enrichment cultures were closely related to Methanothermobacter spp. which have been identified in other high-temperature petroleum reservoirs. It has recently been suggested that methanogenic oil degradation may be a major factor in the development of the world’s heavy oils and represent a significant and ongoing process in conventional deposits. Although an oil-degrading methanogenic consortium was not enriched from these samples the presence and activity of communities of fermentative bacteria and methanogenic archaea was demonstrated. Stimulation of methanogenesis by addition of nutrients suggests that in situ methanogenic biodegradation of oil could be harnessed to enhance recovery of stranded energy assets from such petroleum systems.  相似文献

Twenty-five samples of stratal fluids obtained from a high-temperature (60–84°C) deep subsurface (1700–2500 m) petroleum reservoir of Western Siberia were investigated for the presence of dissimilatory Fe(III)-reducing microorganisms. Of the samples, 44% and 76% were positive for Fe(III) reduction with peptone and H2 respectively as electron donors. In most of these samples, the numbers of culturable thermophilic H2-utilizing iron reducers were in the order of 10–100 cells/ml. Nine strains of thermophilic anaerobic bacteria and archaea isolated from petroleum reservoirs were tested for their ability to reduce Fe(III). Eight strains belonging to the genera Thermoanaerobacter, Thermotoga, and Thermococcus were found capable of dissimilatory Fe(III) reduction, with peptone or H2 as electron donor and amorphous Fe(III) oxide as electron acceptor. These results demonstrated that Fe(III) reduction may be a common feature shared by a wide range of anaerobic thermophiles and hyperthermophiles in deep subsurface petroleum reservoirs. Remane's Artenminimum at the horohalinicum is a fundamental concept in ecology to describe and explain the distribution of organisms along salinity gradients. However, a recent metadata analysis challenged this concept for protists, proposing a species maximum in brackish waters. Due to data bias, this literature-based investigation was highly discussed. Reliable data verifying or rejecting the species minimum for protists in brackish waters were critically lacking. Here, we sampled a pronounced salinity gradient along a west–east transect in the Baltic Sea and analysed protistan plankton communities using high-throughput eDNA metabarcoding. A strong salinity barrier at the upper limit of the horohalinicum and 10 psu appeared to select for significant shifts in protistan community structures, with dinoflagellates being dominant at lower salinities, and dictyochophytes and diatoms being keyplayers at higher salinities. Also in vertical water column gradients in deeper basins (Kiel Bight, Arkona and Bornholm Basin) appeared salinity as significant environmental determinant influencing alpha- and beta-diversity patterns. Importantly, alpha-diversity indices revealed species maxima in brackish waters, that is, indeed contrasting Remane's Artenminimum concept. Statistical analyses confirmed salinity as the major driving force for protistan community structuring with high significance. This suggests that macrobiota and microbial eukaryotes follow fundamentally different rules regarding diversity patterns in the transition zone from freshwater to marine waters.  相似文献

The microbial community and its diversity in production water from a high-temperature, water-flooded petroleum reservoir of an offshore oilfield in China were characterized by 16S rRNA gene sequence analysis. The bacterial and archaeal 16S rRNA gene clone libraries were constructed from the community DNA and, using sequence analysis, 388 bacterial and 220 archaeal randomly selected clones were clustered with 60 and 28 phylotypes, respectively. The results showed that the 16S rRNA genes of bacterial clones belonged to the divisions Firmicutes, Thermotogae, Nitrospirae and Proteobacteria, whereas the archaeal library was dominated by methanogen-like rRNA genes (Methanothermobacter, Methanobacter, Methanobrevibacter and Methanococcus), with a lower percentage of clones belonging to Thermoprotei. Thermophilic microorganisms were found in the production water, as well as mesophilic microorganisms such as Pseudomonas and Acinetobacter-like clones. The thermophilic microorganisms may be common inhabitants of geothermally heated specialized subsurface environments, which have been isolated previously from a number of high-temperature petroleum reservoirs worldwide. The mesophilic microorganisms were probably introduced into the reservoir as it was being exploited. The results of this work provide further insight into the composition of microbial communities of high-temperature petroleum reservoirs at offshore oilfields.  相似文献

The growth of 1-year-old Arctic cisco ( Coregonus autumnalis ) was monitored under laboratory conditions for fish acclimated to one of two temperatures (5 and 10° C) and one of five salinities (6, 12, 18,24, 30‰). Fish were maintained for 43 days at rations of 3% wet body weight per day at 5° C and 5% wet body weight per day at 10° C, with rations adjusted for weight gain every 7–12 days. Fish increased 9–11% in length and 55–71% in weight at 5° C, and 23–27% in length and 141–161% in weight at 10° C. Length and weight increased linearly over 43 days. There was a statistically significant effect of temperature on growth but no statistically significant effect of salinity. Higher growth rates at 10° C were partially attributable to significantly greater gross conversion efficiency at the higher temperature. Over the course of the experiment, the condition (weight per unit length) of all fish increased by 3·2 to 63·6% at 5° C and by 5·6 to 46·0% at 10° C. There was no discernible effect of salinity on condition at either temperature. These results demonstrate that, with salinity acclimation and high food ration, 1-year-old Arctic cisco can grow at equivalent rates across salinities ranging from 6 to 30‰. The ecological implications of the results are discussed.  相似文献

Emperor angel fish Pomacanthus imperator were acclimated to various salinities (33, 22, 15, 10 and 7%) for 1 month and changes in serum chemistry, body composition and liver enzyme activities were monitored. No mortality was observed in fish adapted throughout the salinity range of 7–33% and the limit of survival was around 5%. Serum Na+; and Cl exhibited a stepwise decrease as the salinity was reduced from 33–7% but no concomitant tissue hydration could be observed. Branchial Na-K-ATPase activity was generally higher in hypoosmotic (7–15%) than in hyperosmotic salinities (22–33%o). Serum concentrations of glucose, lipid, α-amino acids and protein as well as the haematocrit value were all elevated in the salinity range of 15–22%. Serum cortisol was markedly lower but serum thyroxine was significantly elevated at 15–22%. Fish adapted to 15–22% had higher hepatosomatic indices, liver glycogen and muscle lipid contents and hepatic glucose-6-phosphate dehydrogenase activity. These data indicate that Pomacanthus imperator is a' physiologically euryhaline' species and metabolism in isoosmotic salinities has been fundamentally re-organized to favour carbohydrate and lipid retention.  相似文献

The number of microorganisms of major metabolic groups and the rates of sulfate-reducing and methanogenic processes in the formation waters of the high-temperature horizons of Dagang oilfield have been determined. Using cultural methods, it was shown that the microbial community contained aerobic bacteria oxidizing crude oil, anaerobic fermentative bacteria, sulfate-reducing bacteria, and methanogenic bacteria. Using cultural methods, the possibility of methane production from a mixture of hydrogen and carbon dioxide (H2 + CO2) and from acetate was established, and this result was confirmed by radioassays involving NaH14CO3 and 14CH3COONa. Analysis of 16S rDNA of enrichment cultures of methanogens demonstrated that these microorganisms belong to Methanothermobacter sp. (M. thermoautotrophicus), which consumes hydrogen and carbon dioxide as basic substrates. The genes of acetate-utilizing bacteria were not identified. Phylotypes of the representatives of Thermococcus spp. were found among 16S rDNAs of archaea. 16S rRNA genes of bacterial clones belong to the orders Thermoanaerobacteriales (Thermoanaerobacter, Thermovenabulum, Thermacetogenium, and Coprothermobacter spp.), Thermotogales, Nitrospirales (Thermodesulfovibrio sp.) and Planctomycetales. 16S rDNA of a bacterium capable of oxidizing acetate in the course of syntrophic growth with H2-utilizing methanogens was found at high-temperature petroleum reservoirs for the first time. These results provide further insight into the composition of microbial communities of high-temperature petroleum reservoirs, indicating that syntrophic processes play an important part in acetate degradation accompanied by methane production.  相似文献

Shallow-water coastal areas suffer frequent reductions in salinity due to heavy rains, potentially stressing the organisms found there, particularly the early stages of development (including pelagic larvae). Individual adults and newly hatched larvae of the gastropod Crepipatella peruviana were exposed to different levels of salinity stress (32(control), 25, 20 or 15), to quantify the immediate effects of exposure to low salinities on adult and larval behavior and on the physiological performance of the larvae. For adults we recorded the threshold salinity that initiates brood chamber isolation. For larvae, we measured the impact of reduced salinity on velar surface area, velum activity, swimming velocity, clearance rate (CR), oxygen consumption (OCR), and mortality (LC50); we also documented the impact of salinity discontinuities on the vertical distribution of veliger larvae in the water column. The results indicate that adults will completely isolate themselves from the external environment by clamping firmly against the substrate at salinities ≤24. Moreover, the newly hatched larvae showed increased mortality at lower salinities, while survivors showed decreased velum activity, decreased exposed velum surface area, and decreased mean swimming velocity. The clearance rates and oxygen consumption rates of stressed larvae were significantly lower than those of control individuals. Finally, salinity discontinuities affected the vertical distribution of larvae in the water column. Although adults can protect their embryos from low salinity stress until hatching, salinities <24 clearly affect survival, physiology and behavior in early larval life, which will substantially affect the fitness of the species under declining ambient salinities.  相似文献

The spring benthos of 22 lakes ranging from 1–88 gl−1 salinity contained 58 species of macroinvertebrates, but only 23 species occurred in waters >3 gl−1. The amphipod Hyalella azteca and the chironomids Procladius freemani, Chironomus nr. muratensis and Cryptochironomus spp. were important at lower salinities (1–12 gl−1) whereas the chironomids Tanypus nubifer, Cricotopus ornatus and Chironomus nr. annularis dominated at moderate salinities (5–30 gl−1) and dolichopodid and ephyrid dipterans were the only species in hypersaline lakes (>50 gl−1). Diversity decreased significantly with increased salinity. Mean dry biomass ranged from 0–9.12 gm−2, showing little correlation with salinity, though hyposaline lakes often had elevated values and hypersaline lakes very low values. Shallow lakes (<5 m) had significantly lower standing crops. There were long term changes in biomass (over 45 years) in some lakes due to cultural eutrophication or secular changes in salinity. Chironomids were by far the dominant contributors to biomass at salinities to 50 gl−1, above which dolichopodid and ephyrid dipterans dominated. The lakes were classified into four groups—subsaline, hyposaline, shallow hypo-mesosaline and hypersaline, reflecting the importance of salinity and also relative depth as major controlling factors.  相似文献

When mineral oil, hexadecane, and glutamate were added to natural samples of varying salinity (3.3 to 28.4%) from salt evaporation ponds and Great Salt Lake, Utah, rates of metabolism of these compounds decreased as salinity increased. Rate limitations did not appear to relate to low oxygen levels or to the availability of organic nutrients. Some oxidation of l-[U-C]glutamic acid occurred even at extreme salinities, whereas oxidation of [1-C]hexadecane was too low to be detected. Gas chromatographic examination of hexane-soluble components of tar samples from natural seeps at Rozel Point in Great Salt Lake demonstrated no evidence of biological oxidation of isoprenoid alkanes subject to degradation in normal environments. Some hexane-soluble components of the same tar were altered by incubation in a low-salinity enrichment culture inoculated with garden soil. Attempts to enrich for microorganisms in saline waters able to use mineral oil as a sole source of carbon and energy were successful below, but not above, about 20% salinity. This study strongly suggests a general reduction of metabolic rate at extreme salinities and raises doubt about the biodegradation of hydrocarbons in hypersaline environments.  相似文献

Salinity is one of the most important drivers of the distribution, abundance and diversity of organisms. Previous studies on the evolution of saline tolerance have been mainly centred on marine and terrestrial organisms, while lineages inhabiting inland waters remain largely unexplored. This is despite the fact that these systems include a much broader range of salinities, going from freshwater to more than six times the salinity of the sea (i.e. >200 g/L). Here, we study the pattern and timing of the evolution of the tolerance to salinity in an inland aquatic lineage of water beetles (Enochrus species of the subgenus Lumetus, family Hydrophilidae), with the general aim of understanding the mechanisms by which it was achieved. Using a time‐calibrated phylogeny built from five mitochondrial and two nuclear genes and information about the salinity tolerance and geographical distribution of the species, we found that salinity tolerance appeared multiple times associated with periods of global aridification. We found evidence of some accelerated transitions from freshwater directly to high salinities, as reconstructed with extant lineages. This, together with the strong positive correlation found between salinity tolerance and aridity of the habitats in which species are found, suggests that tolerance to salinity may be based on a co‐opted mechanism developed originally for drought resistance.  相似文献

Salinity varies widely in coastal areas that often have a high abundance of Pseudo‐nitzschia H. Peragallo. Pseudo‐nitzschia is abundant in Louisiana waters, and high cellular domoic acid has been observed in natural samples but no human illness has been reported. To assess the threat of amnesic shellfish poisoning (ASP), we examined the effect of salinity on Pseudo‐nitzschia occurrence in the field and growth in the laboratory with special emphasis on the salinity range where oysters are harvested (10–20 psu). In Louisiana coastal waters, Pseudo‐nitzschia spp. occurred over a salinity range of 1 to >35 psu, but they occurred more frequently at higher rather than lower salinities. Seven species were identified, including toxigenic species occurring at low salinities. In culture studies, seven clones of three species grew over a salinity range of 15 to 40 psu, some grew at salinities down to 6.25 psu, and most grew at salinities up to 45 psu. Tolerance of low salinities decreased from Pseudo‐nitzschia delicatissima (Cleve) Heiden to P. multiseries (Hasle) Hasle to P. pseudodelicatissima (Hasle) Hasle emend. Lundholm, Hasle et Moestrup. In conclusion, although Pseudo‐nitzschia was more prevalent in the field and grew better in the laboratory at higher salinities, it grew and has been observed at low salinities. Therefore, the probability of ASP from consumption of oysters harvested from the low salinity estuaries of the northern Gulf of Mexico is low but not zero; animal mortality events from toxin vectors other than oysters at higher salinity on the shelf are more likely.  相似文献

Petrochemical and geological evidence suggest that petroleum in most reservoirs is anaerobically biodegraded to some extent. However, the conditions for this metabolism and the cultivation of the requisite microorganisms are rarely established. Here, we report on microbial hydrocarbon metabolism in two distinct oilfields on the North Slope of Alaska (designated Fields A and B). Signature anaerobic hydrocarbon metabolites were detected in produced water from the two oilfields offering evidence of in situ biodegradation activity. Rate measurements revealed that sulfate reduction was an important electron accepting process in Field A (6–807 µmol S l?1 day?1), but of lesser consequence in Field B (0.1–10 µmol S l?1 day?1). Correspondingly, enrichments established at 55°C with a variety of hydrocarbon mixtures showed relatively high sulfate consumption but low methane production in Field A incubations, whereas the opposite was true of the Field B enrichments. Repeated transfer of a Field B enrichment showed ongoing methane production in the presence of crude oil that correlated with ≥ 50% depletion of several component hydrocarbons. Molecular‐based microbial community analysis of the methanogenic oil‐utilizing consortium revealed five bacterial taxa affiliating with the orders Thermotogales, Synergistales, Deferribacterales (two taxa) and Thermoanaerobacterales that have known fermentative or syntrophic capability and one methanogen that is most closely affiliated with uncultured clones in the H2‐using family Methanobacteriaceae. The findings demonstrate that oilfield‐associated microbial assemblages can metabolize crude oil under the thermophilic and anaerobic conditions prevalent in many petroleum reservoirs.  相似文献
