When adding alpha-ketoglutarate and glutamate the intensity of respiration by the myocardium mitochondria increases gradually from the 15th day of embryonic development till the chicken hatching out. In the presence of succinate respiration of mitochondria of 15- and 20-day embryos and 5-day chickens is almost the same and decreases noticeably in adult chickens. When the above-mentioned substrates are added the value of P/O gradually decreases during the chicken development.  相似文献

The distribution and ultrastructure of myotomal muscle fibres was studied in larvae and early juveniles of the curimatã-pacú Prochilodus marggravii , a tropical freshwater fish endemic to the São Francisco River system, Brazil. At 26°C, larvae hatched 15 h post-fertilization at a relatively early stage of development with the head still curved around the yolk-sac (head-trunk angle greater than 45°), and prior to pigmentation of the eyes and formation of the jaws, gut and pectoral fins. Although motile the swimming muscles of newly-hatched larvae were largely undifferentiated. The myotomes were made up of a single layer of superficial muscle fibres containing six to eight myofibrils and abundant mitochondria, surrounding an inner core of myoblasts, myotubes and immature muscle fibres. The volume densities of mitochondria and myofibrils in the immature inner muscle fibres of 1-day-old lavae were 14.5 and 6.4% respectively. The body axis straightened within 24 h of hatching and the yolk sac was completely absorbed by 72 h. Larval development was rapid with gill filaments, a muscular stomach, liver and swimbladder present after 7 days. The inner muscle fibres were well differentiated in 7-day-old larvae; the volume density of myofibrils had increased to 63.1% whereas the volume density of mitochondria had decreased to 3.5%. In 14-day-old juveniles the superficial muscle had thickened to a layer two to three fibres thick in the region of the lateral line nerve and capillaries were present in the inner muscle. Muscle growth until 14 days was largely due to the hypertrophy of the fibres present at hatching.  相似文献

Antibodies against chicken erythrocyte vimentin and gizzard desmin were affinity purified and then cross-absorbed with the heterologous antigen. They were used to study the in vivo distributions of these proteins in developing and mature myotubes by immunofluorescence microscopy of 0.5-2-micron frozen sections of iliotibialis muscle in 7- 21-day chick embryos, neonatal and 1-d postnatal chicks, and adult chickens. The distributions of vimentin and desmin were coincidental throughout the development of myotubes, but the concentration of vimentin was gradually reduced as the myotubes matured and became largely undetectable at the time of hatching. The process of confining these proteins to the level of Z line from the initial uniform distribution occurred subsequent to the process of bringing myofibrils into lateral registry: in-register lateral association of several myofibrils was occasionally seen as early as in 7-11-d embryos, whereas the cross-striated immunofluorescence pattern of desmin and vimentin was only vaguely discerned in myotubes of 17-d embryos, just 4 d before hatching. In some myotubes of 21-d embryos, myofibrils were in lateral registry as precisely as in adult myofibers but desmin was still widely distributed around Z line in an irregular manner. Nevertheless, in many other myotubes of prenatal or neonatal chicks, desmin became confined to the level of Z line in a manner similar to that seen in adult myofibers, thus essentially completing its redistribution to the confined state of adult myofibers in coincidence with the time of hatching. In extracts from iliotibialis and posterior latissimus dorsi muscles of adult chickens, we detected a hitherto unidentified protein that was very similar to vimentin in molecular weight but did not react with our antivimentin antibody. Summary The ultrastructure of the myocyte at all phases of mitosis as well as of early postmitotic cells has been studied in the myocardia of 14- and 18-day rat embryos and 5- and 7-day old rats. The myofibrils remain unchanged up to the late prophase. In prometaphase the majority of Z-disks in embryo myocyte myofibrils and considerable part of these disks in myofibrils of suckling rats are drastically disintegrated.This is followed by a progressive isolation and scattering of the myofilament bundles and of the whole sarcomeres during the subsequent phases of mitosis. Thick myofilaments seem to be unchanged but thin ones become frequently poorly outlined (mainly in embryos). The sarcoplasmic reticulum, including its typically differentiated subsarcolemmal cisternae, exhibits relatively few changes during mitosis. In the early postmitotic period there is a gradual restoration of contrast-rich Z-bands, interconnecting the previously isolated sarcomeres. Patterns of this process have much in common with early stages of myofibrillogenesis (appearance of subsarcolemmal Z-bodies, formation of skeins of thin filaments etc.). The cleavage furrow formation is either absent or considerably retarded up to the postmitotic period.Behaviour of some other organelles during myocyte mitosis has been described. The myocardium of red-cheeked sousliks was studied at different seasons with the use of stereological analysis. The volume and surface densities of the myofibrils, mitochondria, T system, sarcoplasmic reticulum as well as the surface-volume ratios of the main organelles were measured. Pronounced seasonal changes in physiological activity of the heterothermal animals were accompanied by marked reorganization of the spatial ultrastructure of the cardiomyocytes. The seasonal regression of the heart weight and diminution of the cardiomyocyte diameter were recorded during hibernation. The authors believe that the increased volume ratio of the cytoplasmic organelles to the myofibrils forms the basis for a rapid adaptive reaction of the heart during hibernation and waking up.  相似文献

To follow the intracellular distribution of calcium in the breast muscles of developing chickens, Ca45 was injected into the albumen of predeveloped eggs. Since the embryos were grown in a radioactive medium, a complete exchange of the isotope for its non-radioactive counterpart in muscles was accomplished. Subcellular particulates of the muscle cells were separated by the method of differential centrifugation. Analysis of the separated fractions showed that in the muscles of the 13-day embryo, when the nuclear-myofibrillar ratio is high, 65 per cent of the muscle calcium is in the nuclei. With the increased synthesis of myofibrils, the nuclear-myofibrillar ratio decreases with a concomitant fall in radioactivity. Thus, calcium was not associated with the developing myofibrils. At the time of hatching, when myofibrils perform physiological work, the highest level of calcium is in the mitochondria. This suggests that the mitochondria play a key role in the physiological activities of calcium in the cell. The microsomal fraction reaches a maximal level of calcium when the adult composition of muscle is attained. Results of investigations on dystrophic muscles show changes in the calcium distribution of the fractions as early as the 3rd week of embryonic development, which are interpreted to indicate an alteration in the protein metabolism of the cell, or an early destruction of muscle tissue. Further, alterations in the calcium content of fractions which seem to regulate the movements of this ion in the cell are discussed. A new technique for homogenizing tissues from embryos of different ages is presented.  相似文献

Electron-microscopy study of rat myocardium 2 weeks after a heart attack revealed significant alterations in the ultrastructure of cardiomyocytes than for the control. The location of myofibrils was less regular than for normal cells. The population of interfibrillar mitochondria decreased. Mitochondrial cristae were located less densely and formed cellated structures. Swollen mitochondria were observed in the periinfarction and intact areas, indicating the development of ischemia in the myocardium as a whole. Six months after the occlusion of coronary vessels alterations in the location of myofibrils and mitochondria were mainly observed in the peri-infarction area. Mitochondria also formed cellated structures. A 30% decrease in the density of the arrangement of the inner membranes of mitochondria on an area unit was found in the periinfarction zone. The ratio between the relative volumes of mitochondria and myofibrils in the cardiomyocytes of the peri-infarction area was increased by 20%. The area of mitochondria in the intact zone of the left ventricle was 30% greater than for the control. A study of isolated living cardiomyocytes revealed that the mitochondrial- membrane potential in the rats subjected to myocardial infarction half a year ago previously was significantly lower than for the mitochondrial-membrane potential in the control rats. Thus, cardiomyocytes that were similar to healthy cardiomyocytes in their morphology exhibited lower total mitochondrial-membrane potential, indicating their decreased energy state.  相似文献

Carp show a partial compensation in metabolic rate and activity following temperature acclimation. In the present study crucian carp, Carassius carassius , were acclimated for eight weeks to either 2deg; C or 28deg; C. The effects of temperature acclimation on muscle fibre ultrastructure has been investigated. The fractional volume (%) of each fibre type occupied by mitochondria and myofibrils was determined using a point counting morphometric method. Mitochondrial density was found to be higher in the muscles of cold (red fibres 25%; pink fibres 20% and white fibres 4%) than in those of warm acclimated fish (red fibres 14%, pink fibres 11%, white fibres 1%). The proportion of subsarcolemmal to intra-myofibrillar mitochondria was significantly lower in the red fibres of cold acclimated fish. Metabolic compensation to low temperatures are therefore associated with an increase in the number of mitochondria per cell. In contrast, the fractional volume occupied by myofibrils actually decreased following cold acclimation. Evidence is reviewed that temperature compensation of contractile activity results from qualitative rather than quantitative changes in myofibrillar proteins.  相似文献

Timed pregnancies were obtained in Sprague-Dawley rats and early ultrastructural differentiation of myocardium of embryos of 10, 11, 12, 13, and 14 days was investigated and compared with that of newborn. Ten-day myocardium is characterized by loosely packed cells; the cytoplasm is typified by a dearth of organelles. Both thick (myosin) and thin (actin) filaments become identifiable for the first time in the 10-day myocardium where the heart is pulsating but circulation is not established. These filaments are not visible in the embryos of 9-day-old myocardium. The formation of these filaments is observed to continue throughout the period covered in this investigation. Concomitant with the appearance of the myofilaments is the synthesis of Z band material. By the eleventh day of gestation and during the subsequent days there is a rapid proliferation and differentiation of most of the organelles. The myofilaments become organized into fully formed striated fibrils. Intercalated discs appear as. small wavy lines on the eleventh day and become plicated in later stages and serve as cell boundaries and points of attachment for myofilaments and fibrils. There is a perceptible change in the number and morphology of mitochondria from the tenth to eleventh day and later stages of development when the heart becomes functional. Similarly, there is a rapid proliferation and differentiation of granular endoplasmic reticulum and Golgi bodies. Large quantities of free ribosomes are dispersed in the cytoplasm of 10-day myocardium; however, in later stages there is a progressive reduction in the distribution of these particles. An intimate association of ribosomes and polysomes with the developing myofibrils is discernible. The T -system and sarcoplasmic reticulum begin to appear in II-day myocardium. The embryonic myocardium displays intense mitotic activity throughout its development and a unique feature of embryonic myocardial cells is the simultaneous occurrence of myofilament synthesis and mitotic activity within the same cells.  相似文献

The ultrastructure and the appearance of glycogen were studied in the extraocular muscles of 14 externally normal human embryos (Carnegie stages 13-21). At stage 16, myofibrils with an immature Z line and glycogen granules appeared in the cytoplasm of the myoblast. The myoblasts came into cluster at stage 18, and fusion between the myotubes was observed at stage 20. At this stage, an M line appeared in the myofibrils. At stage 21, an A band with a Z line and an H band with an M line were observed, the sarcoplasmic reticulum appeared in the cytoplasm of the muscle fibers and glycogen increased in volume in the cytoplasm. In the previous study, we showed that the muscle-specific isoenzymes, such as creatine kinase, beta-enolase and glycogen phosphorylase, appeared from stage 18 to 20 in the extraocular muscles. The previous findings and the present results suggest that the fusion of the muscle cells occurs in the period when some molecular markers of muscle differentiation are expressed in vivo.  相似文献

Proteins of inner mitochondrial membranes of the albino rat myocardium during postnatal development of 1, 3 and 6 months old animals were electrophoretically separated in 10% polyacrylamide gel. The rate of 14C-amino acids incorporation into examined proteins was determined in vitro. Specific radioactivity of the total mitochondrial fraction decreased in the course of the postnatal development. That of outer membranes remained unchanged, though it sharply increased in inner membranes of mature animals as compared with animals aged one month. Levels of radioactive precursor incorporation in separate protein fractions of inner membranes of the myocardium mitochondria were estimated.  相似文献

Pacific bluefin tuna are active teleost fish with a large capacity for heat conservation and endothermy. They have a high metabolism, and hence the myocardium must be capable of sustaining elevated levels of cardiac output over a wide range of temperatures. To examine the way that the myocardial cells of bluefin tuna respond to their unique cardiac physiology, we have studied the ultrastructure of the internal membrane system and mitochondria of atrial and ventricular myocytes by light and electron microscopy. Our results reveal that cardiomyocytes of juvenile bluefin tuna posses a relatively high content of sarcoplasmic reticulum (SR), together with a large volume of mitochondria within the two (compact and spongy) ventricular compartments and in the atrial myocardium. The mitochondrial structure and distribution in bluefin tuna myocardium follow specific metabolic zonation resulting in a higher volume and lower cristae density in the compact ventricular layer than in atrium and spongy layer. The presence of junctional SR profiles and an extensive network of free SR within cells may ensure a rapid delivery of Ca(2+) to the myofibrils. This, in conjunction with transarcolemmal Ca(2+) entry, might contribute to a faster excitation-contraction-relaxation cycle and thus enhance cardiac performance, cardiac output, and the maintenance of excitability at low temperatures. We propose that the mitochondrial configuration together with the developed SR ultrastructure of bluefin tunas myocardium are important evolutionary steps for the maintenance of high heart rates and endothermy in this teleost fish.  相似文献

Diadenosine tetraphosphate (AP4A) belongs to a wide group of naturally derived endogenous purine compounds that have recently been considered as new neurotransmitters in the autonomic nervous system. It has been shown that AP4A induces inhibitory effects and modulates adrenergic control in the heart of adult mammals. Nevertheless, the physiological significance of AP4A in early postnatal development, when sympathetic innervation remains yet immature, has not been investigated. The aim of the present study was to elucidate the effects of AP4A on the heart bioelectrical activity in early postnatal ontogenesis. Action potentials (AP) were recorded using the standard microelectrode technique in multicellular isolated right atrial (RA), left atrial (LA), and ventricle (RV) preparations from male Wistar rats at postnatal days 1, 14, and 21 and from 60-day animals that were considered as adults. The application of AP4A caused significant reduction of AP duration in atrial (RA and LA) preparations from rats of all ages. Also, AP4A caused significant AP shortening in RV preparations from rats of various ages; however, the effect was more pronounced in 21-day-old and adult rats. AP4A failed to alter automaticity of RA preparations from the rats at postnatal days 1, 14, and 21 and weakly decreased spontaneous rhythm in RA preparations from the adult rats. The effect of AP4A was partially abolished by P2-receptor blocker PPADS in LA preparations from both 21-dayold and adult rats, while it failed to suppress AP4A-caused AP shortening in preparations from 1- and 14-dayold animals. Thus, extracellular AP4A causes shortening of AP both in the atrial and ventricular myocardium in the rats of early postnatal ontogenesis and in adults. The effect of AP4A depends on age only for ventricular myocardium where it may be attributed with growing contribution of diadenosine polyphosphates to the control of myocardium inotropy.  相似文献

The AMP deaminase activity measured in crude chicken liver extract did not change significantly during development. The livers of 10- and 14-day chick embryos, 1-day, 5-, 10- and 16-week-old chickens and adult hens were examined for the existence of multiple forms of AMP deaminase. Phosphocellulose column chromatography revealed the existence of two peaks of enzyme activity in the liver of 10- and 16-week-old chickens and adult hens. Kinetic studies with the preparations of AMP deaminase revealed sigmoid-shaped substrate-saturation curves at all developmental stages and hyperbolic-shaped saturation curves for the enzyme form appearing in 10-week-old chickens. All AMP deaminases investigated were susceptible to activation by ATP and inhibition by Pi. Kinetic and regulatory properties as well as pH optima of all the enzyme preparations tested indicate that AMP deaminase isolated from the embryos and from 1-day-old chicks was similar to the form I isolated from adult hens and differed significantly from the form II of this enzyme.  相似文献

Terminal deoxynucleotidyl transferase activity begins to be detectable in the thymus of 14-day old chicken embryos. It reaches a maximum 3 weeks after hatching, and persits at a fairly high level in 21-weeks old chickens. Separation of different molecular species of hemoglobin from developing chickens by starch gel electrophoresis has revealed the appearance of early embryonic (embryonic), late embryonic (fetal) and adult hemoglobin (Hb) type during development. In 5-day embryos, there are 3 or 4 forms of embryonic Hb type. They begin to decrease in 6-day embryos and cannot be detected in embryos after 10 days of incubation. In 6-day embryos, two forms of adult Hb type appear, and one of them, which is a major form in adults, becomes t o be a major one in 7-day embryos. One or two forms of fetal Hb type first appear in 10-day embryos and are still present in 5-day posthatching chickens.
Ultracentrifugation of carbonmonoxyhemoglobins from embryos at early and at later stages (fetuses), from newly hatched and from adult chickens has shown that they have a single monodisperse peak. Some heterogeneity, however, has been detected after starch gel electrophoresis, probably owing to aggregation or polymerization.
Subunit analysis of embryonic, fetal and adult Hb type by starch gel electrophoresis in formate buffer at pH 1.9 has indicated that embryonic Hb type contains total 5 subunits, C, D, E, F and G; fetal Hb type, total 2, A and H; and adult Hb type, total 3, B, F and H.  相似文献

Maturation of the cardiac cell is characterized by increasing diversity of isozymic expression of creatine kinases. Expression of the M-CK isozyme always precedes that of mitochondrial isozyme (mi-CK), however the expression of an isoform does not inform about its localization or cellular function. The functional role of isozymes binding to sites of energy utilization and production characteristic of the adult myocardium can be evidenced by the functional coupling of M-CK to myofibrillar ATPase and mito-CK to translocase in Triton X-100 and saponin skinned fibers. Functional activity of M-CK and mito-CK were investigated during perinatal development. Both functional activities appear during late fetal life in species mature at birth like guinea pig, and in the first postnatal weeks in immature species like rat or rabbit. Thus, the functional activity of bound CK isozymes is not associated with birthper se but with the general process of cell maturation. Localization of CK in the cytosol appears optimal for the transfer of glycolytic production of ATP to sites of utilization in an immature heart. During cell maturation, the increasing contribution of oxidative phosphorylation to ATP production, the apparition and binding of mi-CK to mitochondria, the binding of M-CK to myofibrils, turn the cell in a compartmentalized system of energy production. This provides the cellular basis for energy transfer by the PCr-Cr-CK system between sites of ATP production and utilization. Compartmentation of both Ca handling and energy turnover leads to a highly structured cell organization and could be essential for the efficiency of heart function.  相似文献

The embryonic chick heart ventricle of day 11 was studied electron microscopically to learn the structural changes that develop in lead poisoning. The chick embryos were administered with 0.015 mg/egg of lead acetate at day 2. The most pronounced changes observed in the ventricle were: malformed mitochondria, disorganized, short and scanty myofibrils and abundance of swollen vacuoles. The ultrastructure of the ventricle from the control chick embryos was normal. The most frequent change noted in the ventricular tissue was an alteration in the myofibrils. This study indicates that electron microscopic changes can be induced in the embryonic chick heart ventricle by lead poisoning.  相似文献

The common ventricle in the heart of the Thunnus alalunga was studied. The ventricular myocardium consists of an outer compact layer and a thick inner spongy layer. The compact layer has slightly larger cells (4-6 microns diameter) than the spongy layer (2.5-5 microns diameter). Ultrastructurally the myocardium displays normal arrangements of myofibrils and mitochondria. The sarcoplasmic reticulum is poorly developed. The intercalated discs are simple with the fascia adherens being the most frequent junctional type observed; occasionally a desmosome was seen. Nexus type junctions are present but are unassociated with the intercalated discs. There are no t-tubules evident but the plasmalemma exhibits numerous caveolae which rarely form couplings with the sarcoplasmic reticulum. A morphometric analysis of the volume percent of mitochondria and myofibrils showed that the myocardial cells in the spongy layer of the heart have a significantly greater volume percentage of mitochondria than the compact layer. No significant differences were found between myocardial regions when the volume percentages of myofibrils were compared. The physiological studies revealed that the albacore tuna has heart rates (120 bpm) and ventricular blood pressures (100 mmHg) that are among the highest reported for fish.  相似文献

The myocardium of Wistar rats was studied by electron microscopy after ligation of the renal artery resulting in a stable elevation of blood pressure. The ultrastructural data on the development of heart hypertrophy were described within 5 to 35 days after operation using the morphometric and stereological methods and correlation analysis. The hypertrophied cardiomyocytes showed a reduction in the ratio of the total volume density of the mitochondria, sarcoplasmic reticulum and T system to the volume density of myofibrils. It was discovered that hypertrophy of cardiomyocytes is marked by an increase in the sarcoplasmic reticulum rather than in myofibrils and other cell compartments, whereas the relative volume of mitochondria decreases.  相似文献
