Serine proteases form one of the most important families of enzymes and perform significant functions in a broad range of biological processes, such as intra- and extracellular protein metabolism, digestion, blood coagulation, regulation of development, and fertilization. A number of serine proteases have been identified in parasitic helminths that have putative roles in parasite development and nutrition, host tissues and cell invasion, anticoagulation, and immune evasion. In this review, we described the serine proteases that have been identified in parasitic helminths, including nematodes (Trichinella spiralis, T. pseudospiralis, Trichuris muris, Anisakis simplex, Ascaris suum, Onchocerca volvulus, O. lienalis, Brugia malayi, Ancylostoma caninum, and Steinernema carpocapsae), cestodes (Spirometra mansoni, Echinococcus granulosus, and Schistocephalus solidus), and trematodes (Fasciola hepatica, F. gigantica, and Schistosoma mansoni). Moreover, the possible biological functions of these serine proteases in the endogenous biological phenomena of these parasites and in the host-parasite interaction were also discussed.  相似文献

The objectives of this study was to conduct a survey on schistosomiasis and soil-transmitted helminth (STH) infections in order to come up with feasible control strategies in Lake Victoria basin, Tanzania. Depending on the size of the school, 150-200 schoolchildren were recruited for the study. Duplicate Kato-Katz stool smears were prepared from each child and microscopically examined for Schistosoma mansoni and STHs. Urine specimens were examined for Schistosoma haematobium eggs using the filtration technique. After the survey, mass drug administration was done using praziquantel and albendazole for schistosomiasis and STHs infections, respectively. A total of 5,952 schoolchildren from 36 schools were recruited for the study and had their stool and urine specimens examined. Out of 5,952 schoolchildren, 898 (15.1%) were positive for S. mansoni, 754 (12.6%) for hookworms, 188 (3.2%) for Ascaris lumblicoides, and 5 (0.008%) for Trichuris trichiura. Out of 5,826 schoolchildren who provided urine samples, 519 (8.9%) were positive for S. haematobium eggs. The results revealed that intestinal schistosomiasis, urogenital schistosomiasis, and STH infections are highly prevalent throughought the lake basin. The high prevalence of intestinal and urogenital schistosomisiasis in the study area was a function of the distance from Lake Victoria, the former being more prevalent at localities close to the lake, whilst the latter is more so away from it. Control of schistosomiasis and STHs in the study area requires an integrated strategy that involves provision of health education to communities, regular treatments, and provision of adequate safe water supply and sanitation facilities.  相似文献

Isotopic studies of nitrogen and sulphur inputs to plant/soil systems commonly rely on limited published data for the 15N/14N and 34S/32S ratios of nitrate, ammonium and sulphate in rainfall. For systems with well-developed plant canopies, however, inputs of these ions from dry deposition or particulates may be more important than rainfall. The manner in which isotopic fractionation between ions and gases may lead to dry deposition and particulates having 15N/14N or 34S/32S ratios different from those of rainfall is considered. Data for rainfall and throughfall in coniferous plantations are then discussed, and suggest that: (1) in line with expectations, nitrate washed from the canopy has 15N/14N ratios higher than those in rainfall; (2) the 15N/14N ratios of ammonium washed from the canopy are variable, with high ratios being found for canopies of higher pH in conditions of elevated ambient ammonia gas concentrations; and (3) in accord with expectations and previous work, 34S/32S ratios of sulphate washed from the canopy are not substantially different from those in rainfall. The study suggests that if atmospheric inputs are relevant to isotopic studies of the sources of nitrogen for canopied systems, then confident interpretation will require analysis of these inputs. The objective of this study was to carry out a community survey on schistosomiais and soil-transmitted helminth (STH) infections in order to suggest feasible and effective intervention strategies in Lake Victoria basin, Tanzania. A total of 37 communities selected from 23 districts of the 4 regions in the Lake Victoria basin of Tanzania were involved in the study. From each of the selected locality, 50 adult community members, 25 males and 25 females, were recruited for the study. Each study participant was requested to submit stool and urine specimens. From each stool specimen, duplicate Kato-Katz thick smears were prepared and microscopically examined for Schistosoma mansoni and STH eggs. Urine specimens were processed by the filtration technique and microscopically examined for Schistosoma haematobium eggs. Ultrasound examination for morbidity due to schistosomiasis was performed. Mass treatment was done using praziquantel and albendazole for schistosome and STHs infections, respectively. Out of 1,606 adults who provided stool specimens, 199 (12.4%) were positive for S. mansoni, 349 (21.7%) for hookworms, 133 (8.3%) for Ascaris lumbricoides, and 33 (2.0%) for Trichuris trichiura. Out of 1,400 participants who provided urine specimens, 25 (1.8%) were positive for S. haematobium eggs. Because of the co-endemicity of these afflictions and their impact on vulnerable population groups, the helminthiasis could be simultaneously treated with 2 drugs, praziquantel for schistosomiasis and albendazole for STHs.  相似文献

Cognitive behavior therapy (CBT), as exemplified by the model of psychotherapy developed and refined over the past 40 years by A.T. Beck and colleagues, is one of the treatments of first choice for ambulatory depressive and anxiety disorders. Over the past several decades, there have been vigorous efforts to adapt CBT for treatment of more severe mental disorders, including schizophrenia and the more chronic and/or treatment refractory mood disorders. These efforts have primarily studied CBT as an adjunctive therapy, i.e., in combination with pharmacotherapy. Given the several limitations of state‐of‐the‐art pharmacotherapies for these severe mental disorders, demonstration of clinically meaningful additive effects for CBT would have important implications for improving public health. This paper reviews the key developments in this important area of therapeutics, providing a summary of the current state of the art and suggesting directions for future research.  相似文献

In many pathological situations, tissue damage is caused by cellular generation of superoxide free radicals (O2-). These active species are generated during post-ischemic reperfusion of organs, in hyperoxic tissue, during acute and chronic inflammation and during exposure to ionizing radiation. Exogenous superoxide dismutase (SOD) was shown to significantly prevent such damage.

The genes for human cytosolic Cu/ZnSOD and mitochondrial MnSOD were cloned and introduced into an E. coli expression system. The proteins were expressed in high yields and purified to homogeneity, yielding pharmaceutical-grade materials. These enzymes were used in a variety of in vivo animal models for the demonstration of their protective effects against oxidative damage. Comparative pharmacokinetic studies in rats have revealed that the half-life of Cu/ZnSOD was 6–10min., while that of MnSOD was 5–6 hours, thus indicating that MnSOD may be superior to Cu/ZnSOD for the treatment of chronic diseases. Indeed, MnSOD was found to be erective as an anti-inflammatory agent in the rat carrageenan induced paw edema acute inflammation model. Both enzymes were also effective in ameliorating post-irradiation damage in mice exposed to whole-body or localized chest X-ray radiation.  相似文献

白藜芦醇(resveratrol,Res.)是一种潜在的抗HIV药物,在体外与AZT、3Tc、ddI等核苷类抗病毒药物联用时表现出很强的协同增强作用。在本篇论文中我们在Friend-MuLV病毒感染的BALB/c鼠艾滋病模型(MAIDS)上研究了用白藜芦醇、AZT、3Tc不同组方及白藜芦醇单独使用治疗后的抗逆转录病毒作用,分析白藜芦醇在联合治疗中可能的潜在协同增强作用。72只BALB/c小鼠随机分成9组(每组8只):正常对照组(未感染和未治疗),病毒对照组(感染但未治疗)和7个其他药物治疗组(感染病毒后并用白藜芦醇、AZT、3Tc不同组方治疗)。药物通过口服给药方式在病毒感染4 h后给药,每天一次,每周7 d,持续3周。感染后3周,所有实验组处死,对表现疾病病理特征的一些指标进行检测评价。结果表明:在体内鼠艾滋病模型上,单独运用白藜芦醇仅仅只有部分能够减少疾病的进展,未表现出所预期的显著的抗逆转录病毒作用(只有20.78%的脾肿抑制率)。白藜芦醇和AZT3、Tc的联合用药有较好的抗病毒效果(减少白细胞数83.34%,脾肿抑制率91.79%,升高CD4 /CD8 比值等)。然而,这些结果与AZT和3Tc联合给药的结果是重叠的,AZT和3Tc联合给药有类似的结果(脾肿抑制率87.65%和白细胞数减少82.25%),排除了AZT3、Tc和白藜芦醇可能的协同增效作用。因此在MAIDS模型中白藜芦醇和AZT3、Tc之间没有协同作用,AZT、3Tc并不会因为和白藜芦醇的联用而提高抗病毒效果。  相似文献

自闭症谱系障碍(Autism spectrum disorder, ASD)是一类常见神经发育疾病,以社会交往障碍、刻板重复行为与狭隘的兴趣为主要临床特征。在过去40年间,ASD患病率呈不断上升趋势,因而日益受到人们关注。近年来由于大规模外显子测序的应用,发现了许多新的ASD易感基因。这些易感基因富集在几个共同的遗传信号通路中,参与突触形成和染色质重构等。最新的动物模型研究表明,ASD的发病机制包括神经突触可塑性异常和神经回路兴奋性-抑制性平衡紊乱。本文从ASD遗传病因的高度异质性、众多致病基因突变影响的共同生物学过程以及遗传诊断方法和药物研发的进展等几个方面进行了综述,以期帮助人们深入了解ASD的遗传基础和转化研究现状。  相似文献

Summary The statoliths of Sepia officinalis, Octopus vulgaris, Alloteuthis subulata and Taonius megalops have a smooth outline, but an irregular shape. They have projections and indentations. The statoliths from a pair of statocysts are usually quite similar in size and shape, and the general pattern is probably maintained throughout the size range of the species. Statoliths from large animals are marginally larger than those from smaller ones. The statolith usually occupies only a small part of the cavity of the statocyst, and it is situated in the anterior part of the statocyst. They are joined to the macula by hairs extending from it. These hairs are very delicate and easily broken during preparation of the specimens. The hairs are much longer and narrower than the receptor cilia of the macula. The receptor cilia are enclosed within holes in the tangled hairlike anchoring fibrils.The statolith is made up of crystalline subunits, the statoconia. The crystals vary in size, they are usually elongated, hexagonal with pointed ends. The statolith consists of a closely packed mass of these crystals, sometimes they are irregularly arranged, where in others they are stacked with their long axes parallel. In Sepia officinalis and Taonius megalops, the crystals are arranged in regular shaped packets and these packets of crystals are stacked together. These larger subunits are not always arranged in a regular way, and their major axes can be organised in several different ways. The size and outline of these large subunits do vary in different parts of the statolith.The external surface of the statolith is macroscopically smooth. Over some parts there is a surface layer covering the rod-like crystals that make up the major bulk of the stone. In other regions, the surface is rough at a microscopic level, the roughness is produced by the exposed ends of the filamentous crystals. Extracts enriched for globulin proteins were prepared from the seeds of a large number of legume species and were tested for homology to antisera prepared against the glycosylated 7S seed storage protein of the soybean (Glycine max). Electrophoretic identification and subsequent analysis of proteins precipitated with 7S antisera was useful at relatively short taxonomic distances, particularly within the tribe Phaseoleae, to which G. max belongs. Glycine and most other members of the subtribe Glycininae are unusual within the Phaseoleae in having high molecular weight (> 70 000 dalton) subunit polypeptides. Seeds from other plants representing other subtribes of the Phaseoleae also contained proteins that cross-reacted with the G. max antisera; the molecular weights of these proteins varied from 30 000 to nearly 90 000 daltons. Homology was detected across a wider range of legume tribes within the subfamily Papilionoideae by enzyme-linked immunosorbent assay (ELISA). The results of these experiments suggest both that the 7S proteins of these tribes are evolutionarily related and that at least some features of these apparently rapidly-evolving proteins are under relatively strong selectional constraint.  相似文献

The genomic structure surrounding a T-DNA integration site in a transgenic petunia plant, which shows deregulation of a root-specific promoter, was investigated. We have already demonstrated that T-DNA integration in this transformant (P13) had occurred close to a scaffold/matrix attachment region (S/MAR). A major question regarding the observed promoter leakiness was whether the T-DNA had integrated into the centre or at the border of the Petun-SAR and whether other regulatory elements are located within this genomic region. While small rearrangements were shown to occur during T-DNA integration in agreement with other reports, we find indications of the presence of a SINE retroposon – an apparent landmark for recombinogenic targets – at the integration site. Binding assays to both plant and animal nuclear scaffolds, supported by biomathematical analyses, reveal that the T-DNA is definitely located at the border of a strong S/MAR, which is in agreement with current models on the structure of integration sites. These results, together with a developmentally regulated leaf-specific enhancer effect of the Petun-SAR on gene expression in transgenic tobacco plants, indicate that the Petun-SAR demarcates the right border of a chromatin domain with genes predominantly active in leaves.  相似文献

A family of universal expression vectors based on the pUC8 and pUC9 plasmids has been constructed. These vectors cover all three possible reading frames in both directions, allowing any synthetic DNA, genomic DNA or cDNA to be expressed under control of the lac promoter. The four new vectors retain the useful features of the pUC plasmids, including the blue to white color change on X-gal plates indicating the presence of an insert. This family of expression vectors is expected to be quite useful in allowing direct immunological screening of cDNA or genomic DNA banks.  相似文献

将猪瘟病毒的E2基因克隆入酵母分泌型表达载体pPIC9K中,酶切线性化后电穿孔导入Pichia pastoris进行整合,经G418筛选得到高拷贝转化子,甲醇诱导表达。SDS-PAGE和Western blit结果证实了酵母培养上清液中含有E2蛋白。免疫活性研究证明P.pastoris表达的E2蛋白能刺激动物产生抗猪瘟病毒的抗体。  相似文献

根据柑橘黄龙病亚洲种23S/5S的DNA序列设计一对引物对不同地理来源的6个柑橘黄龙病样品DNA进行扩增,扩增片段大小均为1 654 bp包括一个假定细胞壁水解酶假基因(putative cell wall hydrolase pseudogene)和5S rRNA 基因.序列同源性分析结果表明;6个柑橘黄龙病病原菌样品与柑橘黄龙病病原菌亚洲种Sihui样品的同源性为99%,然而与土壤杆菌,布鲁氏菌,根瘤菌,中华根瘤菌,巴通体菌和中慢生根瘤菌的同源性只有89%~95%,说明在23S/5S rDNA序列上黄龙病病原菌亚洲种与α变形菌纲根瘤菌目的其他病原菌相差较大.对黄龙病病原菌亚洲种种内的23S/5S rDNA序列进行比较分析,结果发现黄龙病病原菌亚洲种种内之间putative cell wall hydrolase pseudogene和5S rRNA的基因序列非常保守,但不同地理来源的柑橘黄龙病样品碱基序列间确实存在差异,差异的大小与地理的远近无关.利用简约法对黄龙病病原菌亚洲种及α变形菌纲其它病原菌的23S/5S rDNA序列构建的系统发育树显示黄龙病病原菌亚洲种单独聚为一类,其他细菌聚为另一类,该结果与基于rplJ基因及16S rRNA基因的DNA序列构建的分子系统进化树结果一致.  相似文献

Summary Using T7 RNA polymerase and specific constructs derived from 5S rRNA and RNA I genes, we generated substrates for the RNA processing enzyme RNase E. Using these substrates we have shown that a 3.2 kb DNA fragment that complements the rne-3071 mutation can express RNase E activity. We also found that T7 RNA polymerase terminates within the 5S rRNA gene.  相似文献
