The permeability of the inner membrane of turnip mitochondria to H+ and OH- ions has been investigated using an acid pulse technique. The rate of decay of a H+ pulse across the inner membrane is exponential having first-order kinetics and gives t 1/2 values of approx 54 s at neutral pH and at 25° C. Valinomycin or 1799 alone have little effect on t 1/2 values, whereas in combination, values of <15 s are observed. Nigericin produces a similar effect. The effective proton conductance of the inner membrane near pH 7 at 25° C is 0.27 nmol H+ min-1 mg protein-1 mV-1. The results suggest that at neutral pH, the inner membrane of plant mitochondria is relatively impermeable to H+ and OH- ions.  相似文献

Mitochondria isolated from spinach leaves (Spinacia oleracea L.) and potato tubers (Solanum tuberosum L.) were partly injured when subjected to freezing for 2 to 4 h at-25°C in salt solutions in the absence of cryoprotectants. Damage was manifested by the inactivation of respiratory properties and increase in the permeability of the mitochondrial membranes. Decrease in respiratory control indicated that the control mechanism of the electron transport chain was influenced by freezing. Oxidative phosphorylation was only slightly more affected than electron transport. The inactivation of the membrane systems was caused by an increase in the concentration of membrane-toxic solutes. This was confirmed by treatment of the organelles at 0°C in solutions of high salt concentrations. When sugar was present in the course of freezing, mitochondria were partly or completely protected. On a molar basis, sucrose was more effective in membrane protection than glucose. Summary Carnitine was found to stimulate fatty acid oxidation by pea (Pisum sativum L.) cotyledon mitochondria. The stimulation was at a maximum for long chain (C16:0) and short chain (C4:0 and C6:0) fatty acids. Evidence was also provided which indicated that mid-chain (C10:0 and C12:0) fatty acid oxidation by mitochondria was stimulated by carnitine. In the present study we have observed a higher state of coupling in respiring mitochondriaisolated from green as compared to red tomatoes (Lycopersicon esculentum, Mill.). Greentomato mitochondria produced a membrane potential () high enough to phosphorylate ADP,whereas in red tomato mitochondria, BSA and ATP were required to restore to the levelof that obtained with green tomato mitochondria. This supports the notion that such uncouplingin red tomato mitochondria is mediated by a plant uncoupling mitochondrial protein (PUMP;cf. Vercesi et al., 1995). Nevertheless, mitochondria from both green and red tomatoes exhibitedan ATP-sensitive linoleic acid (LA)-induced decrease providing evidence that PUMP isalso present in green tomatoes. Indeed, proteoliposomes containing reconstituted green or redtomato PUMP showed LA uniport and LA-induced H+ transport. It is suggested that the higherconcentration of free fatty acids (PUMP substrates) in red tomatoes could explain the lowercoupling state in mitochondria isolated from these fruits.  相似文献

Mitochondria isolated from 7-day old darkgrown Avena sativa L. (var. Arnold) laminae given 5 min illumination of red light, followed by varying lengths of darkness up to 3 h, showed at least a twofold increase in the rates of both NADH-dependent oxygen consumption and respiratory chain phosphorylation over those of mitochondria isolated from unilluminated tissue. Similar organelles, isolated from tissue given either far-red or red followed by far-red pretreatment, exhibited rates of both functions of between 25% and 75% below those of the mitochondria from unilluminated tissue. The induction-reversion criteria for phytochrome control of respiration and oxidative phosphorylation were satisfied under all experimental conditions during the greening process.Treatment with continuous far-red light, acting presumably through the high irradiance reaction of phytochrome, served to disengage phytochrome activity from photosynthesis. A radioimmunoassay for the detection of as little as 0.5–1 pmol indole-3-acetic acid (IAA) in unpurified or partially purified plant extracts is described. The assay makes use of either IAA[125I]tyrosine methyl ester or [3H]IAA methyl ester as radioactive antigens and IAA methyl ester as the assay standard (measuring range: 1–200 pmol). The sialidase secreted byClostridium chauvoei NC08596 was purified to apparent homogeneity by ion-exchange chromatography, gel filtration, hydrophobic interaction-chromatography, FPLC ion-exchange chromatography, and FPLC gel filtration. The enzyme was enriched about 10 200-fold, reaching a final specific activity of 24.4 U mg–1. It has a relatively high molecular mass of 300 kDa and consists of two subunits each of 150 kDa. The cations Mn2+, Mg2+, and Ca2+ and bovine serum albumin have a positive effect on the sialidase activity, while Hg2+, Cu2+, and Zn2+, chelating agents and salt decrease enzyme activity. The energy-dependent transport of phenylalanine into isolated vacuoles of barley (Hordeum vulgare L.) mesophyll protoplasts has been studied by silicone-layer floatation filtering. The uptake of this aromatic amino acid into the vacuolar compartment is markedly increased by MgATP, showing saturation kinetics; the K m values were 0.5 mM for MgATP and 1.2 mM for phenylalanine. V max for phenylalanine transport was estimated to 140 nmol phenylalanine·(mg·Chl)-1·h-1. The transport shows a distinct pH optimum at 7.3 and is markedly inhibited by 40 mM nitrate. Azide (1 mM) and vanadate (400 M) had no or little effect on rates of transport while p-fluorophenylalanine seemed to be an effective inhibitor, indicating a possible competition at an amino-acid carrier. Using the technique of silicone oil filtration of organelles and the inhibitor stop method, the kinetics of transport of inorganic phosphate across the inner mitochondrial membrane were tested in relation to different stages of greening (0 to 24 h) of etiolated laminae of Avena sativa L., and compared to the rates of oxygen consumption and ATP formation. The results demonstrate that there is a pronounced increase in phosphate transport after 3 h of greening, reaching values for Vmax (about 17 mol mg protein-1 h-1) that are three times as high as those measured with mitochondria from etiolated tissue. This is also mirrored by the rates of respiration and oxidative phosphorylation. After 24 h of light treatment (4 Klx), respiration and ATP formation, as well as V decreased again to levels below those of the etiolated stage. In contrast to V, there was no change in the affinity between inorganic phosphate and the binding sites of the transporting systems involved, as indicated by a rather constant Km (0.23 mM) for phosphate transport. Of the inhibitors of phosphate transport tested, mersalyl and methyl mercuric iodide were most efficient with identical characteristics of inhibition; but compared to animal mitochondria, the concentrations needed to result in similar amounts of inhibition, were more than ten times higher. Mitochondria from etiolated corn possess a much greater Ca2+ uptake capacity per mg protein than microsomes from the same source. Differences in energy requirements, sensitivity to specific inhibitors, and sedimentation properties enabled us to study both Ca2+ uptake mechanisms without mutual contamination. The microsomal Ca2+ uptake does not vary much among different plants as compared to the mitochondrial Ca2+ uptake; this is also true for different organs of the same plant. Mitochondrial Ca2+ uptake is more dependent on the age of the seedlings than microsomal uptake, because of changes in active Ca2+ uptake activity rather than of changes in efflux. Intactness and the oxidative and phosphorylative properties of the mitochondria remained unchanged during this time period. Different methods were tested to isolate protoplasts from etiolated, partially greened, and light-grown leaves of Avena sativa. Preparations with high yields and high photosynthetic capacities (time of illumination 4 h) were obtained when small transverse leaf segments were incubated for 2 h at 30°C in 2% cellulysin (Calbiochem), 0.6 M mannitol, and 0.5% bovine serum albumin (BSA) at pH 5.6, without shaking. As measured by light-dependent O2 evolution or fixation of labeled bicarbonate, protoplasts exhibited rates of up to 124 mol per mg of chlorophyll per h at 20°C and saturating bicarbonate, which were nearly identical to those found with intact leaves. The assay conditions necessary for this activity were 0.6 M sorbitol, 50 mM N-2-hydroxy-ethylpiperazine-N-2-ethane sulfonic acid (pH 7.6), and 10 mM NaHCO3. If plastids were isolated from these protoplasts, sorbitol was 0.45 M, including 10 mM ethylenediaminetetraacetate (EDTA). under these conditions, rates of photosynthesis were up to 125 (light-grown) and 71 (6 h illuminated) mol O2 evolved or 14CO2 fixed per mg of chlorophyll per h, compared to 3.5 mol·mg chl-1·h-1 obtained with mechanically isolated plastids. With this system, CO2-dependent O2 evolution was already detected after 3 h of illumination of etiolated tissue, but could only be observed at pH values between 7.6 and 8.6, in the presence of EDTA. At lower pH (7.3) or at pH 7.6 in the absence of EDTA, light-dependent O2 evolution up to 24 h of greening was only measurable with 3-phosphoglycerate as the substrate. Summary Earlier whole-animal experiments have shown that the California killifish (Fundulus parvipinnis) from tidal marshes is highly tolerant to sulfide while the speckled sanddab (Citharichtys stigmaeus) from the open coast is intolerant to sulfide. In the present paper, we demonstrate that the liver mitochondria of the California killifish detoxify sulfide by oxidizing it to thiosulfate and produce ATP in the process. Sulfide oxidation is obligately and stoichiometrically linked to mitochondrial electron transport to oxygen. Concentrations up to 20 M sulfide stimulate mitochondrial respiration while 50 M sulfide causes half-inhibition. Sulfide oxidation by mitochondria is adversely affected at pH<7.4. ATP production is maximal at 10 M sulfide. The finding of sulfide oxidation coupled to ATP production by killifish mitochondria is unprecedented among vertebrates. In comparison, mitochondria of the specked sanddab oxidize sulfide at a much lower rate. Phosphatase activities were measured in preparations of vacuoles isolated from storage roots of red beet (Beta vulgaris L.). The vacuoles possessed both acid phosphatase and ATPase activities which could be distinguished by their susceptibility to inhibition by low concentrations of ammonium molybdate [(NH4)6Mo7O24·4H2O]. The acid phosphatase was completely inhibited by 100 M ammonium molybdate but the ATPase was unaffected. The acid phosphatase was a soluble enzyme which hydrolysed a large number of phosphate esters and had a pH optimum of 5.5. In contrast, the ATPase was partially membrane-bound, had a pH optimum of 8.0 and hydrolysed ATP preferentially, although it was also active agianst PPi, GTP and GDP. At pH 8.0 both the ATPase and PPase activities were Mg2+-dependent and were further stimulated by KCl. The ATPase and PPase activities at pH 8.0 may be different enzymes. The recovery and purification of the ATPase during vacuole isolation were determined. In the culture supernatant ofTrypanosoma rangeli, strain El Salvador, a sialidase was present with an activity of 0.1 U/mg protein as determined with the 4-methylumbelliferyl glycoside of -N-acetylneuraminic acid as substrate. This enzyme was purified about 700-fold almost to homogeneity by gel chromatography on Sephadex G-100 and Blue Sepharose, and affinity chromatographies on 2-deoxy-2,3-didehydroneuraminic acid and horse submandibular gland mucin, both immobilized on Sepharose. The pH optimum is at 5.4–5.6, and the molecular weight was determined by gel chromatography, high performance liquid chromatography and sodium dodecyl sulphate gel electrophoresis to be 70 000. The substrate specificity of the enzyme is comparable to bacterial, viral and mammalian sialidases with cleavage rates for the following substrates in decreasing order: N-acetylneuraminyl-(2–3)-lactose> N-glycoloylneuraminy-(2–3)-lactose> N-acetylneuraminyl-(2–6)-lactose >sialoglycoproteins>gangliosides>9-O-acetylated sialoglycoproteins.4-O-Acetylated derivatives are resistant towards the action of this sialidase. Cells of the purple non-sulphur bacterium Rhodobacter sphaeroides express a high-affinity K+ uptake system when grown in media with low K+ concentrations. Antibodies againts the catalytic KdpB protein or the whole KdpABC complex of Escherichia coli crossreact with a 70.0 kDa R. sphaeroides protein that was expressed only in cells grown in media with low K+ concentrations. In membranes derived from R. sphaeroides cells grown with low K+ concentrations (induced cells), a high ATPase activity could be detected when assayed in Tris-HCl pH 8.0 containing 1 mM MgSO4. This ATPase activity increased upon addition of 1 mM KCl from 166 to 289 mol ATP hydrolysed x min-1 x g protein-1 (1.7-fold stimulation). The K+-stimulated ATPase activity was inhibited approximately 93% by 0.5 mM vanadate but hardly by N,N-dicyclohexylcarbo-diimide (DCCD). These results indicate that the inducible K+-ATPase in R. sphaeroides resembles the Kdp K+-translocating ATPase of Escherichia coli. The membrane orientation and symmetry of protein thiol group(s) necessary for transport of Pi in rat liver mitochondria have been assessed by comparing inhibition of transport in intact mitochondria to that in inverted vesicles of purified inner membrane. The permeability characteristics of a variety of inhibitors have been determined under specified conditions. The sensitivities of the uptake pathways in mitochondria and in inverted vesicles appear thus far to be identical. Digitonin was applied to permeabilize the plasma membrane of Bothrops alternatus erythrocytes to study respiration, oxidative phosphorylation and Ca2+ transport by mitochondria in situ. These mitochondria oxidized added NAD-linked substrates, succinate and N,N,N, N-tetramethyl-p-phenylenediamine. Respiration was sensitive to rotenone and cyanide but not to antimycin A. This indicates that Bothrops mitochondria possess the respiratory complexes NADH-ubiquinone, succinate-ubiquinone, and ferrocytochrome c-oxygen oxidoreductases, although the lack of sensitivity to antimycin A raises doubt about the composition of the ubiquinol cytochrome c-reductase complex. An ability to build up and sustain a membrane potential was documented by their capacity to accumulate tetraphenylphosphonium and Ca2+ through an uncoupler-sensitive mechanism. Addition of ADP caused a transient decrease in the membrane potential, indicating that this is the predominant driving force for ATP synthesis as in most types of mitochondria. Purified intact protoplasts were isolated from etiolated and greening leaves of Avena sativa. They were ruptured by forcing them through a 20-m aperture nylon net and immediately thereafter fractionated into a pure pellet of plastids (well above 70% of total plastids), a layer of mitochondria only slightly contaminated by other cellular constituents (about 50% of total mitochondria), and a cytoplasmic supernatant. This was achieved within 60 s by an integrated method of homogenation of protoplasts and centrifugal filtration of the homogenate on a gradient of silicone oils, contained together with the nylon net in 450 l microtubes, and verified by comparing the levels of activity of specific markers within the three fractions obtained. With appropriate modifications to immediately quench metabolic reactions within the fractions, this method allows the determination of metabolite levels within plastids, mitochondria, and the cytoplasmic compartment of intact protoplasts. The applicability of this technique is demonstrated by the determination of ATP in the plastids, mitochondria, and the cytoplasm of protoplasts obtained from etiolated and greening primary leaves of Avena. Treatment of spheroplasts of Nostoc museorum with hypotonic buffer results in membranes depleted of cytochrome c-553, but still active in photosynthetic and respiratory electron transport. These membranes retain full photosystem II activity (H2ODADox). Complete linear electron transport (H2ONADP+), however, is decreased as compared with untreated spheroplasts. Treatment of intact potato (Solanum tuberosum L.) tubers with acetaldehyde, ethanol or acetic-acid vapors led to a respiratory upsurge which was further increased when the volatiles were applied in 100% O2. Mitochondria from tubers held in 100% O2 (O2 control) displayed a substrate state, state 3, and state 4 in respiration, whereas in mitochondria from the volatile-treated tubers the respiratory rate of the different states was virtually indistinguishable. This respiratory pattern was companied by the development of a cyanide-resistant respiration since these mitochondria exhibited resistance to CN and sensitivity to CN+salicylhydroxamic acid.
