The conventional way to drive modifications in old forest tree seed orchards is to establish progeny trials involving each parent tree and then evaluate its contribution to the performance of the progeny by estimating its general and specific combining ability (GCA and SCA). In this work, we successfully applied an alternative parent selection tactic based on paternity testing of superior offspring derived from a hybrid seed orchard established with a single Eucalyptus grandis seed parents and six E. urophylla pollen parents. A battery of 14 microsatellite markers was used to carry out parentage tests of 256 progeny individuals including two independent samples of selected trees and one control unselected sample, all derived from 6-year-old forest stands in eastern Brazil. Paternity determination was carried out for all progeny individuals by a sequential paternity exclusion procedure. Exclusion was declared only when the obligatory paternal allele in the progeny tree was not present in the alleged parent tree for at least four independent markers to avoid false exclusions due to mutation or null alleles. After maternity checks to identify seed mixtures and selfed individuals, the paternity tests revealed that approximately 29% of the offspring was sired by pollen parents outside the orchard. No selfed progeny were found in the selected samples. Three pollen parents were found to have sired essentially all of the offspring in the samples of selected and non-selected progeny individuals. One of these three parents sired significantly more selected progeny than unselected ones (P0.0002 in a Fisher exact test). Based on these results, low-reproductive-successful parents were culled from the orchard, and management procedures were adopted to minimize external pollen contamination. A significant difference (P<0.01) in mean annual increment was observed between forest stands produced with seed from the orchard before and after selection of parents and revitalization of the orchard. An average realized gain of 24.3% in volume growth was obtained from the selection of parents as measured in forest stands at age 2–4 years. The marker-assisted tree-breeding tactic presented herein efficiently identified top parents in a seed orchard and resulted in an improved seed variety. It should be applicable for rapidly improving the output quality of seed orchards, especially when an emergency demand for improved seed is faced by the breeder.Communicated by D.B. Neale  相似文献

Eighteen black spruce (Picea mariana) stands, representing postfire ages of 26 to 120 yr, were surveyed for understorey vegetation and site/microsite characteristics at two spatial scales. This enabled comparison of within- versus among-stand compositional variation.Detrended correspondence analysis (DCA) ordination among the 18 stands revealed a complex age/moisture gradient. DCA ordination among 1 800 quadrats within the stands indicated a similar gradient with much compositional overlap. Quadrats were grouped, using two-way indicator species analysis (TWINSPAN), into 9 classes each representing a phase in understorey vegetation composition. These phases shifted in abundance from young to old stands with a high degree of concordance among replicates in the same age class. 为了探讨不同红树林林分土壤有机碳(soil organic carbon,SOC)和全氮(total nitrogen,TN)储量空间的分布特征以及与C/N的相关性,该研究以广西防城珍珠湾红树林湿地为对象,通过样地调查取样和实验室分析,测定了SOC和TN的含量以及土壤碳储量的计量,揭示了广西北仑河珍珠湾秋茄、木榄和混交林三种红树林林分SOC和TN储量空间的分布特征以及C/N与SOC和TN的相关性。结果表明:(1)秋茄、木榄和混交林的SOC储量分别为140.73、124.94、144.71 t·hm-2,三者无显著性差异(P>0.05);木榄和混交林垂直分布特征表现为20~40 cm>0~20 cm>40~60 cm,秋茄表现为随着土层深度的增加而递减。(2)秋茄、木榄和混交林的TN储量分别为6.49、5.01、5.87 t·hm-2,表现为随着土层深度的增加而减少的趋势。(3)秋茄、木榄和混交林的SOC与TN储量之间的相关性极显著(P<0.01),相关系数分别为0.924、0.971和0.844,说明SOC与TN之间存...  相似文献

The aim of this study was to determine if individual ticks among the progeny of a single female Rhipicephalus (Boophilus) microplus tick removed from cattle under natural conditions are the result of mating with one or several males. To this end, simulations were run using an existing dataset of genotypes from 8 microsatellite loci to predict the number of samples required and the best locus. Subsequently, 14–22 progeny from each of 15 engorged female ticks removed from three cows, and the engorged females themselves, were genotyped for the BmM1 locus and the minimum number of potential male parents was determined for each progeny group. Of the 15 progeny groups, 10 must have been sired by more than one male, as indicated by the presence of five unique alleles among the progeny or three unique alleles that could not have been contributed by the female. This finding demonstrates multiple paternity in R. microplus.  相似文献

Summary We studied the chondriomes (the mitochondrial genomes) of sexual-progeny plants derived from eleven Nicotiana cybrids which resulted from donor-recipient protoplast fusions. The recipients were either N. tabacum or N. sylvestris and the donor (of the cytoplasm) was N. bigelovii. The chondriomes were characterized by the mitochondrial DNA (mtDNA) restriction-patterns. The differences in mtDNA restriction patterns were revealed after Sal I digestions and probing the respective Southern-blots with three mtDNA fragments. The hybridization patterns of mtDNAs from 35 second-generation plants (i.e. the sexual progeny derived from the cybrid plants) indicated only minor variations between plants derived from the same cybrid but pronounced variations among sibs derived from different cybrids. The mtDNA of 32 second-generation plants varied from both original fusion partners but the mtDNA of one (male-sterile) plant was apparently identical with the mtDNA of one of the original donor (N. bigelovii) and the mtDNA of two other (male-fertile) plants was apparently identical to the mtDNA of an original recipient (N. sylvestris). Generally, the mtDNAs of male-fertile, second-generation plants were similar to the mtDNAs of the original recipients while the mtDNAs of the male-sterile second-generation plants were similar to the mtDNA of the donor (N. begelovii). The analyses of mtDNAs from the thirdgeneration plants indicated stabilization of the chondriomes; no variations were detected between the mtDNAs of plants derived from a given second-generation plant.  相似文献

Variability in recombination frequency was reported in the Iowa Stiff Stalk Synthetic. The objectives of the present research were to verify the differences in recombination frequency among individuals in the Iowa Stiff Stalk Synthetic maize population and to determine if the recombination frequency differences persisted among the S1 progeny. Testcrosses to measure male recombination frequency on three chromosomes (4, su1-c2; 5, a2-bt1-pr1; 9, sh1-bz1-wx1) were repeated for eight S0 individuals. Recombination frequencies were repeatably divergent among those individuals which were selected based on high or low recombination frequencies on specific chromosomes. Individuals which had been selected for long and short total map distances across the three chromosome regions produced repeatably divergent recombination frequencies only at the su1-c2 region. The recombination frequencies of the S1 lines, derived from the S0 individuals which had the most divergent recombination frequencies on a single chromosome, were significantly different. The broadsense heritability estimates derived from the regression of six S1 lines on six S0 individuals ranged from 0.69 to 0.20 for the five chromosome regions. We conclude that genetic differences for recombination frequency exist in this population and that modification by selection should be possible.  相似文献

Abstract. We compared the species composition and species density of vascular plants in the understorey vegetation of boreal forest between Picea mariana (Black spruce) and Populus tremuloides (Trembling aspen) stands in British Columbia, Canada, and related differences in species composition and species density between the two forest types to dominant canopy tree species as well as a wide variety of environmental factors. We analysed 231 stands, distributed in three different climatic regions representing drier, wetter, and milder variations of montane boreal climate. Of these stands 118 were dominated by P. mariana and 113 by P. tremuloides. P. tremuloides stands had higher species density than P. mariana stands in all climatic regions, but species density of each dominance type varied among climatic regions. The floristic composition of the understorey vegetation was markedly different for P. mariana and P. tremuloides dominated stands. A detailed study on the effect of canopy dominance and local environmental factors on the understorey vegetation of the boreal forest was conducted using 88 stands from one of the three climatic regions. Using a combination of ordination and variation partitioning by constrained ordination we demonstrated a small but unique effect of canopy dominance type on the understorey vegetation, while a larger amount of compositional variation was shared with other factors. Our results accord with a scenario in which differences in primary environmental factors and humus form properties, the latter accentuated by the canopy dominants themselves, are the most important causes of higher species density in P. tremuloides stands than in P. mariana stands, as well as differences in species composition among the two canopy dominance types. Processes and time scales involved in the small but significant direct and indirect effects of the canopy dominant on understo‐ rey species composition are discussed.  相似文献

为探究森林群落树种组成对苔藓植物分布的影响,利用多元统计方法研究粤东亚热带地区苔藓植物的组成和分布对林分类型的响应。根据森林乔木层的树种组成划分为翻白叶树(Pterospermum heterophyllum)、木荷(Schima superba)+米锥(Castanopsis carlesii)和米锥3种森林类型。结果表明,3种森林类型中苔类(liverworts)和藓类(mosses)植物群落组成特征总体上存在显著差异;双齿裂萼苔(Chiloscyphus latifolius)和细指苔(Kurzia gonyotricha)在3种类型林分中的重要值变化指示了苔类植物群落的差异。3种类型林分中,均以东亚拟鳞叶藓(Pseudotaxiphyllum pohliaecarpum)为优势种。不同林分中藓类植物种类组成主要表现为亚优种分布的不同。这表明森林群落树种组成作为重要的生物因子,对林内苔藓植物的分布和种类组成有重要影响。  相似文献

1. Spatial variation of methane (CH4) efflux from the littoral zone of a meso‐eutrophic boreal lake was studied with a closed‐chamber technique for three summer days in 22 vegetation stands, consisting of three emergent and three floating‐leaved species. 2. Between‐species differences in CH4 emission were significant. The highest emissions were measured from the emergent Phragmites australis stands (0.5–1.7 mmol m?2 h?1), followed by Schoenoplectus lacustris > Equisetum fluviatile > Nuphar lutea > Sparganium gramineum > Potamogeton natans. Within‐species differences between stands were not significant. 3. In P. australis stands, the stand‐specific mean CH4 emission was significantly correlated with solar radiation, probably indicating the role of effective pressurised ventilation on CH4 fluxes. The proportion of net primary production emitted as CH4 was significantly higher in P. australis stands (7.4%) than in stands of S. lacustris and E. fluviatile (both 0.5%). 4. In N. lutea stands, CH4 efflux was negatively correlated with the mean fetch and positively with the percentage cover of leaves on the water surface. There were no differences in CH4 efflux between intact N. lutea leaves and those grazed by coleopteran Galerucella nymphaeae. In S. graminaeum and P. natans stands, CH4 effluxes were not related to any of the measured environmental variables. 5. For all vegetation stands, the biomass above water level explained about 60% of the observed spatial variation in CH4 emission, indicating the important role of plants as gas conduits and producers of substrates for methanogens in the anoxic sediment.  相似文献

We conducted a microcosm experiment for studying the decomposition of Altingia obovata leaf litter by the decomposer community at 20 and 30°C from three forest stands (namely a 35-year-old secondary forest, a 47-year-old secondary forest, and a primary forest) of a tropical montane rainforest. Our results showed that rank-order of the litter decomposition among the three forest stands was not parallel to the stand age. At 20°C, the mass loss of A. obovata leaf litter from the primary forest was higher than those from the two secondary forests, of which the younger stand showed higher mass loss than did the older one. However, there were no differences in mass loss among these three stands at 30°C. The mass loss for the two secondary forest stands, but not for the primary forest stand, increased significantly from 20 to 30°C. The level of lignin decomposition among the three stands at 20°C corresponded to their forest stand age, i.e., the primary forest > the 47-year-old secondary forest > the 35-year-old secondary forest. A rise of 10°C in temperature significantly increased lignin decomposition for the two secondary forests, while the reverse was true for the primary forest. Carbohydrate decomposition was positively related to the temperature but not to the stand age. The different responses of litter decomposition to the forest stand age and temperature might be due to the differences in the microbial activities among the three forest stands.  相似文献

造林再造林作为新增碳汇的一种有效途径,受到国际社会的广泛关注。如何通过改变林分树种组成,优化造林模式提高人工林生态系统碳贮量已成为国内外学者关注的重点。通过样方调查和生物量实测相结合的方法,对南亚热带26年生红锥纯林(PCH)、杉木纯林(PCL)及红锥×杉木混交林(MCC)生态系统各组分碳含量、碳贮量及其分配特征进行了比较研究。结果表明:杉木、红锥各器官平均碳含量分别为492.1—545.7 g/kg和486.7—524.1 g/kg。相同树种不同器官以及不同树种的相同器官间碳含量差异显著(P0.05)。红锥各器官碳含量的平均值(521.3 g/kg)高于杉木(504.7 g/kg)。不同林分间地被物碳含量大小顺序为PCHMCCPCL;不同树种之间的土壤碳含量差异显著(P0.05),0—100 cm土壤平均碳含量为PCLMCCPCH。生态系统碳贮量大小顺序为PCL(169.49 t/hm2)MCC(141.18 t/hm2)PCL(129.20 t/hm2),相同组分不同林分以及相同林分的不同组分碳贮量均存在显著差异(P0.05)。造林模式对人工林碳贮量及其分配规律有显著影响,营建混交林有利于红锥生物量和土壤碳的累积,而营建纯林有利于杉木人工林生物量碳的吸收,也有利于土壤碳的固定。因而,混交林的固碳功能未必高于纯林,在选择碳汇林的造林模式时,应以充分考虑不同树种的固碳特性。  相似文献

Progeny produced by inbreeding were compared to progeny derived from outcrosses for gynodioecious Schiedea salicaria and subdioecious S. globosa to assess fitness consequences of breeding system on parental fecundity (seeds per capsule) and progeny measures of fitness (germination, survival, biomass, and number of flowers). Results from both species indicated that inbreeding depression occurred at all measured stages of the life history. In both species, different females showed different levels of inbreeding depression. Multiplicative fitness functions of the ratio of values for selfed and outcrossed progeny in S. salicaria resulted in inbreeding depression values of 0.62–0.94. Within- vs. between-family crosses of S. globosa also resulted in inbreeding depression values as high as 0.49. These values suggest that inbreeding depression may promote the evolution of dioecy within S. globosa and S. salicaria, depending on the levels of natural outcrossing.  相似文献

摘要 目的:比较同源肿瘤细胞来源的不同单克隆表型差异。方法:采用极限稀释法,在悬浮培养条件下获取HCT116结肠癌细胞系的单个细胞,对每孔含单个的细胞进行扩增培养,获得子代单克隆,并以同样方法继续挑取单克隆,连续获得子三代克隆。根据单克隆形态特点,选取第三代的三株代表性的单克隆,采用Western blot和免疫荧光法比较其SOX2、EpCAM和Vimentin蛋白表达差异。采用放疗观察三株单克隆的Vimentin蛋白的动态变化,研究其放疗干预的时间异质性,Transwell体外侵袭实验比较克隆侵袭力的差异。结果:三株由单细胞扩增培养的同源第三代子克隆依然存在明显生物学差异。形态有明显区别的球形与不规则的克隆形态。不规则形态克隆更表现为SOX2低表达及Vimentin的高表达。并且在单个细胞水平上,同个单克隆群体内也存在个体细胞间蛋白的表达差异(Vimentin; EpCAM)。通过观察放疗前后Vimentin蛋白在不同时间点上的荧光强度,发现肿瘤单克隆细胞存在时间异质性。Transwell体外侵袭实验也显示三个同源克隆间存在明显的差异性。结论:同源的、连续单细胞扩增获得的第三代单克隆依然存在明显生物学差异,提示肿瘤内部异质性是其固有特征,并且在治疗干预下,也会引起肿瘤时间异质性的产生。  相似文献

Abstract. Species composition, detritus, and soil data from 97 boreal forest stands along a transect in central Canada were analysed using Correspondence Analysis to determine the dominant environmental/site variables that differentiate these forest stands. Picea mariana stands were densely clustered together on the understorey DCA plot, suggesting a consistent understorey species composition (feather mosses and Ericaceae), whereas Populus tremuloides stands had the most diverse understorey species composition (ca. 30 species, mostly shrubs and herbs). Pinus banksiana stands had several characteristic species of reindeer lichens (Cladina spp.), but saplings and Pinus seedlings were rare. Although climatic variables showed large variation along the transect, the CCA results indicated that site conditions are more important in determining species composition and differentiating the stand types. Forest floor characteristics (litter and humus layer, woody debris, and drainage) appear to be among the most important site variables. Stands of Picea had significantly higher average carbon (C) densities in the combined litter and humus layer (43530 kg‐C.ha‐1) than either Populus (25 500 kg‐C.ha‐1) or Pinus (19 400 kg‐C.ha‐1). The thick surface organic layer in lowland Picea stands plays an important role in regulating soil temperature and moisture, and organic‐matter decomposition, which in turn affect the ecosystem C‐dynamics. During forest succession after a stand‐replacing disturbance (e.g. fires), tree biomass and surface organic layer thickness increase in all stand types as forests recover; however, woody biomass detritus first decreases and then increases after ca. 80 yr. Soil C densities show slight decrease with ages in Populus stands, but increase in other stand types. These results indicate the complex C‐transfer processes among different components (tree biomass, detritus, forest floor, and soil) of boreal ecosystems at various stages of succession.  相似文献

Seeds collected from five Amaranthus dubius plants of the same population were weighed and tested in continuous light or darkness at three temperatures in controlled conditions. Different seed families differed in weight, germination percentages and rate, and responded differently to an increase in temperature from 25 to 38 C. Plants derived from these seed families and grown outdoors, either singly or in mixtures, differed in several growth characteristics and in time to flowering. Significant interactions between growth characteristics and growth conditions, as measured with a two-way ANOVA, suggest that plants derived from different individuals differ in their competitive ability. The progeny of different individuals may thus be favored in different environmental conditions.  相似文献

Recent changes in environmental conditions in populations of peat-bog pine (Pinus uliginosa Neumann) caused rapid decline or even extinction of the species in several stands in Central Europe. Conservation strategies forP. uliginosa require information about the evolutionary history and genetic structure of its populations. Using isozymes we assessed the genetic structure ofP. uliginosa from four isolated stands in Poland and compared the results to genetic structures of other closely related pine species including eight populations ofPinus mugo, ten ofPinus sylvestris and one ofPinus uncinata. The level of genetic variability ofP. uliginosa measured by the mean number of alleles per locus and average heterozygosity was similar to others related toP. uliginosa taxa from the reference group but it differs among populations. High genetic similarity was found between two populations ofP. uliginosa from Low Silesian Pinewood. The populations were genetically distinct as compared to other populations includinglocus classicus of the species from the peat bog at Batorów Reserve. Very low genetic distance (Dn = 0.002) and small genetic differentiation (GST = 0.003) were found betweenP. uliginosa andP. mugo in the sympatric populations of the species from Zieleniec peat bog suggesting the ongoing natural hybridisation and genetic contamination of peat-bog pine from this area. Some evidence for skew in allele frequency distribution potentially due to recent bottleneck was found in population from Low Silesian Pinewood. The analysed open pollinated progeny derived from twoP. uliginosa stands from Low Silesian Pine-wood showed the excess of homozygotes as compared to the maternal trees indicating high level of inbreeding (F = 0.105,F = 0.081). The results are discussed in the context of evolution ofP. uliginosa populations, taxonomic relationships between the analysed species and conservation strategies for active protection of peat-bog pine.  相似文献

We estimated rates of self-fertilization and inbreeding depression in the hermaphroditic perennial Arabis fecunda. Crosses were made on mesh-bagged wild plants, yielding 12 plants producing self-, outcross-, and naturally fertilized seeds that were then grown in a greenhouse. Analysis of variance indicated aboveground dry weight at 160 days differed among pollination treatments, but mean seed weight, number of seeds per fruit, and days to germination did not. For dry weight, selfed progeny have 9.4% reduction and naturally pollinated progeny a 3.7% reduction compared to outcrossed progeny, suggesting a 39% selling rate in Arabis. Significant inbreeding depression and a mixed mating system are evidence that the deleterious effects of self-fertilization are maintained through high rates of mildly deleterious mutation. Significant maternal-parent-by-pollination-treatment interactions for mean seed weight and dry weight are consistent with inbreeding depression caused by deleterious recessives and varying past maternal inbreeding.  相似文献

This study compares survival and growth of progeny derived from chasmogamous (CH) and cleistogamous (CL) flowers in Impatiens capensis, a forest annual. When progeny were grown in the field, CH seeds had significantly higher survival rates over winter (64% versus 56%), and the survival advantage of outcrossed progeny was not attributable to seed weight differences. No differences in seedling growth were detected. Greenhouse comparisons revealed no difference in seed survival but a 30% growth advantage to CH seedlings. We found no changes in developmental homeostasis of three leaf shape characters between inbred and outbred progeny, nor was there any difference in variability within CH and CL families. The outcrossing advantage observed in these experiments could not have been caused by avoidance of sib competition. Theory predicts that self-pollinated progenies may be more variable than outcrossed progenies if rare, recessive alleles are important contributors to genetic variances. Electrophoretic markers indicate that progeny derived from CH flowers are predominantly outcrossed (at least 54-97%).  相似文献

Summary When conducting tree breeding experiments, geneticists often assume that individuals from open-pollinated families are halfsibs. The reliability of this assumption was tested using data from enzyme electrophoresis to estimate the genetic relatedness among progeny within 22 open-pollinated families of Robinia pseudoacacia L. (black locust) and 34 open-pollinated families of Gleditsia triacanthos L. (honey locust) from natural stands. An algorithm employing population estimates of fixation indices, pollen allele frequencies, and selfing rates was used to calculate the mean expected number of alleles in common across loci under assumptions of either full-sib (i.e., a single pollen parent) or half-sib (i.e., random mating) relationships. For each open-pollinated family, the average coefficient of relationship among progeny was calculated by linear interpolation from the observed number of alleles in common. For most families of both species, coefficients were significantly higher than 0.25 (half-sib relation), but were significantly lower than 0.50 (full-sib relation). These results suggest that the assumption of a half-sib relationship among progeny of open-pollinated families is violated for these tree species. More critical to the estimation of heritabilities and the prediction of genetic gains was the observation that estimates of the coefficient of relationship varied widely among open-pollinated families (for R. pseudoacacia r 0=0.20–0.43, mean=0.34; for G. triacanthos r 0=0.29–0.55, mean=0.36).  相似文献

Nearly all published rates of secondary forest (SF) regrowth for Amazonia are inferred from chronosequences. We examined SF regrowth on abandoned pastures over a 4‐year period to determine if measured rates of forest recovery differ from chronosequence predictions. We studied the emergence, development and death of over 1300 stems in 10 SFs representing three age classes (<1–5, 6–10 and 11–14 years old). Mean tree biomass accumulation in both the <1–5 and 6–10 years old (4.4 and 5.7 Mg ha−1 yr−1, respectively) abandoned pastures was lower than predicted and deviated significantly (57% and 41%) from rates estimated from the chronosequence. The older SFs, with a mean growth rate of 9.9 Mg ha−1 yr−1 followed the rate predicted by the chronosequence. Understocking was the primary cause of low biomass recovery rates in the youngest forests; although the youngest stands had a diameter at breast height increment three times the oldest stands, the youngest stands lacked sufficient density to cumulatively produce high biomass accumulation rates. Four years of measurement indicated that the youngest stands had developed 59% of the stems measured in the older stands during the same time period. The 6–10‐year‐old stands were rapidly self‐thinning and approached stem density values measured in the same aged stands at the onset of the study. Mortality was high for all stands, with 54% of the original stems remaining after 4 years in intermediate‐aged stands. The forests were dominated by the tree Vismia, which represented 55–66% of the biomass in all stands. The Vismia share of the biomass was decreasing over time, with other genera replacing the pioneer. Our measured rates of regrowth indicate that generalized estimates of forest regrowth through chronosequence studies will overestimate forest regrowth for the youngest forests that were under land use for longer time‐periods before abandonment. Certified Emission Reductions under the Clean Development Mechanism of the Kyoto protocol should consider these results when predicting and compensating for carbon sequestered under natural forest management.  相似文献
