祁连山两侧中国沙棘不同居群的遗传多样性研究   总被引:1,自引:0,他引:1  
应用RAPD技术研究了祁连山中段中国沙棘(Hippophae rhamnoidoes ssp.sinensis)4个居群和1个对照居群的遗传多样性水平,探讨中国沙棘边缘居群的遗传变异以及天然屏障祁连山对中国沙棘居群遗传结构的影响。结果显示,祁连山地区中国沙棘居群水平的Nei’基因多样度(h)和Shannon多态信息指数(I)分别为0.2070和 0.297 4,基因分化系数(Gst)为0.319 3,均高于中国沙棘整个分布区的平均值,表明该地中国沙棘边缘居群遗传多样性水平及居群间的遗传分化都有增加的趋势;居群间每代迁移数(Nm=1.065 9)显著低于异交风媒植物(Nm= 5.380),表明祁连山的隔离对中国沙棘居群间的基因交流有限制作用,造成了该地中国沙棘边缘居群间较高的遗传分化。  相似文献

We investigated adaptive differences among three geographically separate populations of coho salmon (Oncorhynchus kisutch) by forming first generation intercrosses (hybrid lines) and comparing them to parental types (control lines). Broodstock for the experiment came from the Gastineau Hatchery in Juneau, Hidden Falls Hatchery on Baranof Island, and Neets Bay Hatchery near Ketchikan, Alaska. All were isolated hatchery populations separated by 220–400 km and derived 15–20 years previously from single local wild populations. For each population, gametes were taken from 50 mature salmon of each sex and combined to form nine lines (three control and six hybrid); each line had 50 full-sibling families which were assigned to separate cells of an incubator at Gastineau Hatchery. Embryo survival and development times were measured as indicators of locally adapted fitness traits. Two of the control lines had higher survival rates than hybrid lines formed between either of their parental populations and other populations. Differences (p < 0.05) were found between development times for control and hybrid groups, which varied by as many as 20 days between families and as many as 30 days between control lines. The intermediate expression of development time of intercrossed lines is consistent with additive genetic variation of development time between the ancestral populations of coho salmon and indicates that important genetic divergence exists between the populations. A loss of local adaptation through a change in seasonal timing of completion of embryonic development would occur in intercrosses between the populations.  相似文献

Xerothermic species are rare and threatened in central and eastern Europe. In light of the continuing loss of steppe‐like habitats due to anthropogenic fragmentation and degradation, the evaluation of genetic variation in populations inhabiting them is of immediate importance if appropriate conservation measures are to be undertaken. Here we report on the genetic diversity of the rare leaf beetle Crioceris quatuordecimpunctata, whose populations in central and eastern Europe inhabit highly geographically isolated areas. All of the studied populations (in Poland, Ukraine, and Slovakia) were differentiated at the mitochondrial marker COI. However, with respect to the nuclear marker ITS1, Polish populations were monomorphic, but distinct from all other populations. The distinctiveness of the studied populations was confirmed by Wolbachia screening, which showed that all populations carried different strains (one or two), which were probably transferred independently from other insects. On the other hand, no diversity was found in any marker within particular populations, which could be caused (at least for mtDNA) by a Wolbachia selective sweep. Crioceris quatuordecimpunctata probably consists of isolated populations, which went through narrow bottlenecks leading to a drastic reduction in their genetic diversity. As these populations are reciprocally monophyletic for mtDNA haplotypes and show a significant divergence of allele frequencies at nuclear loci, they could be classified as evolutionarily significant units (ESUs). In addition, DNA barcodes were used to identify Asparagus officinalis as the host plant for members of all studied populations. These data should be valuable in efforts to conserve populations of C. quatuordecimpunctata (e.g., for guiding reintroductions).  相似文献

In the Northern Hemisphere, global warming has been shown to affect animal populations in different ways, with southern populations in general suffering more from increased temperatures than northern populations of the same species. However, southern populations are also often marginal populations relative to the entire breeding range, and marginality may also have negative effects on populations. To disentangle the effects of latitude (possibly due to global warming) and marginality on temporal variation in population size, we investigated European breeding bird species across a latitudinal gradient. Population size estimates were regressed on years, and from these regressions we obtained the slope (a proxy for population trend) and the standard error of the estimate (SEE) (a proxy for population fluctuations). The possible relationships between marginality or latitude on one hand and slopes or SEE on the other were tested among populations within species. Potentially confounding factors such as census method, sampling effort, density-dependence, habitat fragmentation and number of sampling years were controlled statistically. Population latitude was positively related to regression slopes independent of marginality, with more positive slopes (i.e., trends) in northern than in southern populations. The degree of marginality was positively related to SEE independent of latitude, with marginal populations showing larger SEE (i.e., fluctuations) than central ones. Regression slopes were also significantly related to our estimate of density-dependence and SEE was significantly affected by the census method. These results are consistent with a scenario in which southern and northern populations of European bird species are negatively affected by marginality, with southern populations benefitting less from global warming than northern populations, thus potentially making southern populations more vulnerable to extinction.  相似文献

The house finch (Carpodacus mexicanus) is a native songbird of western North America that was introduced to the eastern United States and Hawaiian Islands in historic times. As such, it provides an unusually good opportunity to test the ability of molecular markers to recover recent details of a known population history. To investigate this prospect, genetic variation in 172 individuals from 16 populations in the western and eastern United States, southeastern Canada, Hawaiian Islands, and Mexico, as well as genetic variation in the closely related purple finch (Carpodacus purpureus) and Cassin's finch (Carpodacus cassinii) was studied by a semi-automated fluorescence-labeled amplified fragment length polymorphism (AFLP) marker system. A total of 363 markers were generated, of which 258 (71.2%) were polymorphic among species, 166 (61.4%) polymorphic among house finch subspecies, and 157 (60.2%) polymorphic among populations within the frontalis subspecies complex. Heterozygosities and interpopulation divergences revealed by the analysis appeared relatively low at all taxonomic levels, but there are few similar studies in avian populations with which to compare results. Whereas the known population history predicts that both eastern and Hawaiian finches should have been derived from within western populations, tree analysis using both populations and individuals as units suggests weak monophyly of eastern populations and indicates that Hawaiian populations are not clearly derived from California populations. However, the genetic distinctiveness of native and recently founded populations was disclosed by analyses of molecular variance as well as by a model-based assignment approach in which 98%, 94%, and 99% individuals from western, Hawaiian, and eastern regions, respectively, were assigned correctly to their populations without using prior information on population of origin, suggesting that these recent introductions have resulted in detectable differentiation without substantial loss of AFLP diversity. Our results indicate that AFLPs are a useful tool for population genetic and evolutionary studies of birds, particularly as a prelude to finding molecular markers linked to traits subjected to recent adaptive evolution.  相似文献

Abstract.— The role of reinforcement in speciation can be explained by two distinct models. In model I, two diverged populations hybridize and produce fertile hybrids that successfully backcross (hybridization with gene flow). In model II, two populations hybridize but succeeding backcrosses are unproductive (hybridization without gene flow). Using Drosophila persimilis and D. pseudoobscura , we have tested model I by comparing the extent of heterospecific introgression in sympatric versus allopatric populations. We show that certain expectations of this particular model of reinforcement, which is based on hybridization and gene flow between divergent populations after secondary contact, are not realized in these two species. The evidence consists of the similarity of genetic distances as well as proportions of unique/rare alleles between sympatric and allopatric heterospecific populations and a negative correlation between genetic distance and geographical distance between heterospecific populations, which suggests ecological differentiation. This approach in quantifying differential gene flow has important consequences to studies that compare sympatric and allopatric isolation using genetic distance. Following model I, one would expect a pattern of higher prezygotic isolation in sympatric species compared to allopatric species of the same genetic distance simply as a result of an underestimation of genetic distance due to introgression between sympatric populations. We suggest more parsimonious explanations such as reinforcement without genetic exchange (model II) and ecological differentiation, which require high levels of preexisting reproductive isolation between populations.  相似文献

The Mekong River is known to act as a boundary between a number of terrestrial and freshwater species, including various parasites and their intermediate hosts as well as endangered mammal species. Little information is available, however, on the genetic differentiation between terrestrial invertebrates to the east and the west of this wide river. The genetic diversity among eight natural populations of Brachytrupes portentosus (Lichtenstein, 1796) (Orthoptera: Gryllidae) collected from Thailand and the Lao People's Democratic Republic (PDR) were analyzed by multilocus enzyme electrophoresis. The allelic profiles of 20 enzymes encoding 23 loci were analyzed. An average of 41% fixed differences was detected between the populations from Thailand and Lao PDR, which are separated by the Mekong River. The percent fixed differences ranged between 4% and 26% within the populations from Thailand and between 4% and 22% within the populations from Lao PDR. A phenogram shows that the eight populations fell into two major clusters based on the Thai and Lao sampling sites. The genetic distance between the samples within Thailand and within Lao PDR was related to the distances between sampling areas. The genetic variability between populations of this cricket indicates that genetic relationships are influenced by a natural barrier as well as by the geographical distance between these allopatric populations.  相似文献

红砂(Reaumuria soongarica, 柽柳科)是广泛分布于亚洲内陆干旱区温带荒漠的建群灌木, 该物种内部经历过分化和杂交事件, 是研究荒漠植物多样性发生和杂交物种形成的理想模型。然而, 红砂不同群体的倍性水平尚不清楚, 制约了对其物种形成机制的深入研究。为了确定红砂不同支系的倍性及基因组大小变异模式, 本研究以红砂幼苗根尖为实验材料, 以野生番茄(Solanum pimpinellifolium)作为内标物种, 利用流式细胞术对分别来自北疆支系及其可能的亲本支系(东部和西部支系)的共8个红砂群体进行了DNA 1C-值测定和分析。结果显示: 红砂东部支系的DNA 1C-值(1.149 ± 0.012 pg)略小于西部支系的1C-值(1.195 ± 0.031 pg)。北疆支系中阜康(FK)和沙湾(SW)群体的DNA 1C-值处于东、西支系的1C-值之间, 而火烧山(HSS)和五彩城(WCC)群体的1C-值接近东、西支系1C-值的2倍。结合之前的分子标记结果, 我们推测北疆支系中阜康和沙湾群体为东、西部支系的同倍体杂交种群, 而火烧山和五彩城群体属于异源四倍体杂交种群。不同倍性的北疆群体起源于不同的杂交事件, 可以划归为不同的物种。  相似文献

A survey of French natural populations of Drosophila melanogaster has been systematically performed, concerning their status of infection by the sigma virus and the characteristics of viral clones. These investigations, which were not as extensive as those performed in the Languedoc region (Fleuriet et al., 1990) nevertheless give a good representation of the evolution of this system because of the long period involved (almost 20 years). Some trends were observed in all French populations such as (1) a decrease in the high efficiency of transmission by males (which is an important parameter for the viral invading ability); (2) high frequency of a best adapted viral Type. These high frequencies might be due to a recent invasion which is expected to spread to other European populations. However, the frequency of infected flies remained low in northern and central France, unlike in Languedoc. The complexity of this, apparently simple, system of two well-known coevolving organisms should once again be stressed. It is impossible with the known parameters to arrive at a general interpretation of observations made in Languedoc and the rest of France. These data may also throw some light on the structure of French wild populations of D. melanogaster which appear to be subdivided into local populations between which gene flow might be low.  相似文献

The data are reviewed on the population structure and evolutionary dynamics of the nodule bacteria (rhizobia) which are among the most intensively studied microorganisms. High level of the population polymorphism was demonstrated for the rhizobia populations using the enzyme electrophoresis (MLEE profiles). The average value of Nei's coefficient of heterogeneity (H = 1 - sigma pi2 [n/(n - 1)]) were: 0.590 for rhizobia (Rhizobium, Bradyrhizobium), 0.368 for enterobacteria (Escherichia, Salmonella, Shigella) and 0.452 for pathogenic bacteria (Bordetella, Borrelia, Erysipelothrix, Haemophilus, Helicobacter, Listeria, Mycobacterium, Neisseria, Staphylococcus) populations. In spite of being devoid of the effective systems for the gene conjugative transfer, many rhizobia populations possess an essentially panmictic structure. However, the enterobacteria populations in which the gene transfer may be facilitated due to the conjugative F- and R-factors, usually display the clonal population structure. The legume host plant is proved to be a key factor that determines the high levels of polymorphism and of panmixis as well as high evolutionary rates of the symbiotic bacteria populations. The host may ensure: a) an increase in mutation and gene transfer frequencies; b) stimulation of the competitive (selective) processes in both symbiotic and free-living rhizobia populations. A "cyclic" model of the rhizobia microevolution is presented which allows to assess the inputs the interstrain competition for the saprophytic growth and for the host nodulation into evolution of a plant-associated rhizobia population. The nodulation competitiveness in the rhizobia populations is responsible for the frequency-dependent selection of the rare genotypes which may arise in the soil bacterial communities as a result of the transfer of symbiotic (sym) genes from virulent rhizobia strains to either avirulent rhizobia or to the other (saprophytic, phytopathogenic) bacteria. Therefore, the nodulation competitiveness may ensure: a) panmictic structure of the natural rhizobia populations; b) high taxonomic diversity of rhizobia which was apparently caused by a broad sym gene expansion in the soil bacterial communities. The kin selection models are presented which explain evolution of the "altruistic" (essential for the host plant, but not for the bacteria themselves) symbiotic traits (e.g., the ability for symbiotic nitrogen fixation and for differentiation into non-viable bacteroids) in the rhizobia populations. These models are based on preferential multiplication of the nitrogen-fixing clones either in planta (due to an elevated supply of the nitrogen-fixing nodules with photosynthates) or ex planta (due to a release of the rhizopines from the nitrogen-fixing nodules). Speaking generally, interactions with the host plants provide a range of mechanisms increasing a genetic heterogeneity and an evolutionary potential in the associated rhizobia populations.  相似文献

Possible cell sources for restorative processe in adult vertebrates have been considered: (1) differentiated cells undergoing a certain reconstruction after the organ damage; (2) reserve of little-differentiated cells which can be represented by heterogenous populations. Experimental data obtained on a few most studied models of restoration of the damaged organs or their parts have been analyzed. Each particular restorative process can be realized by cell populations which differ from each other by a number of features: origin, potencies, way of formation, localization of cambium, etc. Participation of heterogenous cell populations is most pronounced upon stimulation of restorative processes and in the extreme conditions. Specific involvement of every possible cell source in a particular case of restoration depends on the organism's properties, peculiarities of the damaged organ, as well as on conditions of damage. The existence of diverse cell sources provides for certain reserves of restoration what appears to be indispensable for reliability of restorative processes in varying conditions of damage.  相似文献

The effect of colonization on the distribution of genetic diversity within and among populations in relation to species characteristics remains an open empirical question. The objective of this study was to contrast genetic diversity within and among established and colonizing populations of Nassauvia lagascae var. lanata on Volcán Lonquimay (Araucanía Region, Chile), which erupted on 25 December 1988, and relate genetic diversity to biological characteristics of the populations. We analyzed a total of 240 individuals from 15 populations distributed along the Andes Cordillera using AFLP and obtained a total of 307 AFLP bands, of which 97.7% are polymorphic. Values of population differentiation (F(ST)) did not differ significantly among established and colonizing populations, but colonizing populations did have reduced levels of genetic divergence (as indicated by private and rare bands) and genetic variation (e.g., Shannon index). We conclude that a founder effect through limited numbers of founding propagules derived from nearby source populations has not yet been compensated for by subsequent population growth and migration. Low rates of secondary dispersal via running water, kin-structure within populations, and slow population growth seem to contribute to the slow recovery of genetic diversity.  相似文献

We analyzed the distribution frequency of HLA-A and-B antigens in populations of the major Balkan area in an attempt to study the various hypotheses on the genetic history of populations of the area. Among these hypotheses are these for genetic change between northern and southern populations of Greece (Barbujani and Sokal 1990) and also those for genetic similarities between North Italy and Greece as a result of colonization of Italy by Greeks (“Magna Grecia”) in classical times (Piazza et al 1988). It seems that three main groups are grossly formed within the populations studied, which is in agreement with historical data.  相似文献

Genetic variation of forest coffee trees (Coffea arabica L.) from four regions of Ethiopia was investigated using inter-simple sequence repeat (ISSR) markers. A total of 160 individuals representing 16 populations were sampled. Eleven ISSR primers amplified a total of 123 fragments of which 31 fragments (25%) were polymorphic. Estimate of total gene diversity (H T), and the coefficient of genetic differentiation (G ST) were 0.37 and 0.81, respectively. This indicates that most of the variability is between populations than within populations. The partitioning of genetic variation into within and between populations based on Shannon’s information index also revealed more differentiation between populations (0.80) than within populations (0.20). In the phenogram most of the coffee tree samples were clustered on the basis of their regions of origin but failed to cluster according to their respective populations, which could be attributed to the presence of substantial gene flow between adjacent populations in each region assisted by man in the process of transplantation or by wild animals such as monkeys, which eat the berries and defecate the seeds elsewhere. On the other hand, the inter-regional clustering of some coffee tree samples from Bale and Jimma regions could be due to the transport of coffee seeds across regions and their subsequent planting. Although ISSR markers detected lower polymorphic loci than previously reported results with random amplified polymorphic DNA (RAPD) markers on the same materials, it can be used as an alternative method for molecular characterization of C. arabica populations. The results may provide information to select sites for in situ conservation.  相似文献

Analysis of allozyme data of the European freshwater fish Cottus gobio showed marked genetic differentiation across drainage basins in northeastern Bavaria, which points to the existence of at least two cryptic taxa. Genetic variability within populations differed significantly between these two taxa, which could be due to historical (bottlenecks) or ecological reasons (population size). To distinguish between these two hypotheses we sampled 12 distinct populations from Rhine, Elbe and Danube drainages. Using allozyme data we examined the influence of population size and isolation on genetic variability within populations. We used spatial extent of populations (patch size) as a measure for population size. To estimate isolation we calculated a compound measure which took into account patch size and distance to all neighbouring populations. Both patch size and isolation were highly correlated with genetic variability, explaining ≈95% of the variance of genetic variability within populations. Furthermore, analysis of covariance suggests that the difference in genetic variability between taxa may be explained by differences in population size.  相似文献

Greenhouse and growth chamber studies were established to determine if there are pathological and physiological differences among Meloidogyne hapla populations from California (CA), Nevada (NV), Utah (UT), and Wyoming (WY) on alfalfa cultivars classified as resistant or susceptible to root-knot nematodes. In the greenhouse, plant survival was not consistent with resistance classifications. While all highly resistant Nevada Synthetic germplasm (Nev Syn XX) plants survived inoculation with all nematode populations, two cultivars classified as moderately resistant (''Chief'' and ''Kingstar'') survived (P ≤ 0.05) inoculation with M. hapla populations better than did ''Lobo'' cultivar, which is classified as resistant. Plant growth of Nev Syn XX was suppressed by only the CA population, whereas growth of the other alfalfa cultivars classified as M. hapla resistant or moderately resistant was suppressed by all nematode populations. Excluding Nev Syn XX, all alfalfa cultivars were severely galled and susceptible to all nematode populations. Except for Nev Syn XX, reproduction did not differ among the nematode populations on alfalfa cultivars. Nev Syn XX was not as favorable a host to CA as were the other cultivars; but, it was a good host (reproductive factor [Rf] = 37). Temperature affected plant resistance; the UT and WY populations were more pathogenic at 15-25 C, and CA was more pathogenic at 30 C. Nev Syn XX was susceptible to all nematode populations, except for CA, at only 30 C, and all other alfalfa cultivars were susceptible to all nematode populations at all temperatures.  相似文献

In previous work (Betancourt, Genetics 181:1535, 2009), I propagated three large laboratory populations of an RNA phage (MS2) as they adapted to a controlled laboratory environment. These populations were large enough so that evolution might be expected to be mostly repeatable, but they nevertheless fixed different suites of mutations over the course of the experiment. Here, I investigate one possible explanation for these results: epistasis, in which the effect of a mutation depends on its genetic background, may have prevented populations with different initial substitutions from fixing the same set of subsequent mutations. I show that two mutations that previously occurred in different genetic backgrounds are beneficial on either background. This result suggests that sign epistasis-in which a mutation is beneficial on one background, but deleterious on another-is not the cause of different evolutionary trajectories observed in the Betancourt (2009) experiment. However, they can be explained by either magnitude epistasis-in which mutations have stronger or weaker beneficial effects depending on the background-or by the simultaneous fixation of multiple beneficial mutations. Surprisingly, the large populations of the previous experiment showed less parallel evolution than the small populations of this experiment, which lends support to the fixation of multiple beneficial mutations contributing to the patterns seen in both experiments.  相似文献

Largemouth Bass (Micropterus salmoides) have been introduced on a global scale for sport fishing but represent a conservation concern given their documented negative impacts on native faunal diversity and abundance. Recent research using molecular data to characterize invasive Largemouth Bass populations elsewhere has demonstrated that populations are typically characterized by limited genetic diversity, or represent a combination of Largemouth Bass and Florida Bass (Micropterus floridanus). To test whether these traits were consistent with invasive populations in Brazil, we generated mitochondrial sequence data from four established populations of Largemouth Bass collected in southern Brazil as well as a local aquaculture facility to confirm species identity and quantify levels of genetic diversity. We identified the exclusive presence of Largemouth Bass in the region and observed limited levels of haplotype (haplotype diversity = 0.0684, SE = 0.038) and nucleotide diversity (0.0003, SE = 0.0002) which suggested the presence of a founder effect associated with introduction. Each of the four populations were dominated by a single haplotype that was identical to one recovered from a nearby aquaculture facility, which identified this facility as a potential introduction source.  相似文献

Ethiopia is considered to be a putative migratory corridor for both Near-East Bos taurine and Arabian and Indian B. indicus cattle into East Africa. African pastoralism, which is associated with adaptation to specific habitats and farming systems, has contributed to the composite constitution of Ethiopian cattle. We analyse, for the first time, five Y-chromosome microsatellite markers from seven north Ethiopian cattle populations, using a European Holstein-Friesian population as a reference, to assess the paternal gene pool and to explore the mechanisms behind the genetic structure. Our results reveal that the indicine alleles predominate in the present populations, with only one animal in the Arado carrying the taurine alleles. The north Ethiopian cattle populations with one exception (Abergelle) are characterized by a general low Y-chromosome haplotype diversity, as well as by a reduced interpopulation variance (Phi(ST)=4.0%), which can be a result of strong male-mediated selective sweeps. Population structure revealed by multidimensional-scaling analysis differentiates two populations (Arado and Abergelle) from the rest. Analysis of molecular variance does not lend support to the traditional classification for the populations, which is mainly based on physical characteristics. A network analysis indicates two closely related founding haplotypes accounting for a large proportion (50.0% in Abergelle and 85.0-94.7% in others) of north Ethiopian cattle Y-chromosomes. Our findings point to a common, but limited, paternal origin of the north Ethiopian cattle populations and strong male-mediated gene flow among them. The findings also provide insight into the historical immigration of cattle into East Africa.  相似文献

Greater prairie-chickens (Tympanuchus cupido pinnatus) were once found throughout the tallgrass prairie of midwestern North America but over the last century these prairies have been lost or fragmented by human land use. As a consequence, many current populations of prairie-chickens have become isolated and small. This fragmentation of populations is expected to lead to reductions in genetic variation as a result of random genetic drift and a decrease in gene flow. As expected, we found that genetic variation at both microsatellite DNA and mitochondrial DNA (mtDNA) markers was reduced in smaller populations, particularly in Wisconsin. There was relatively little range-wide geographical structure (FST) when we examined mtDNA haplotypes but there was a significant positive relationship between genetic (FST) and geographical distance (isolation by distance). In contrast, microsatellite DNA loci revealed significant geographical structure (FST) and a weak effect of isolation by distance throughout the range. These patterns were much stronger when populations with reduced levels of genetic variability (Wisconsin) were removed from the analyses. This suggests that the effects of genetic drift were stronger than gene flow at microsatellite loci, whereas these forces were in range-wide equilibrium at mtDNA markers. These differences between the two molecular markers may be explained by a larger effective population size (Ne) for mtDNA, which is expected in species such as prairie-chickens that have female-biased dispersal and high levels of polygyny. Our results suggest that historic populations of prairie-chickens were once interconnected by gene flow but current populations are now isolated.
