Effects of experimental ventilation and ambient Po2 on cutaneous O2 uptake in vitro were studied in the carp, Cyprinus carpio. Oxygen uptake rate of the isolated cutaneous tissue was determined by ventilating the epidermis side of the skin with normoxic water in flow-through respirometers. Oxygen uptake rate of the skin increased with ventilation rate across the skin between 2.5 and 40 ml/min and became 3.2 nmol/cm2/min at a flow rate of 40 ml/min, which corresponds to an apparent water velocity of 1.1 cm/sec. At a ventilation rate of 10 ml/min, oxygen uptake rate of the skin increased with the ambient Po2 between 115 and 230 Torr and became constant (3.8 nmol/cm2/min) between 230 and 295 Torr. When both sides of the skin were ventilated with normoxic water, oxygen uptake rate of the skin increased and became 3.7 nmol/cm2/min at a flow rate of 20–40 ml/min. These results suggest that the oxygen requirement of the skin is 3.7–3.8 nmol/cm2/min at 21.3°C and that cutaneous O2 uptake in vitro depends on experimental ventilation and ambient Po2, consistent with values measured in vitro in the carp (ref).  相似文献

Models of diving optimality qualitatively predict diving behaviours of aquatic birds and mammals. However, none of them has been empirically tested. We examined the quantitative predictions of optimal diving models by combining cumulative oxygen uptake curves with estimates of power costs during the dives of six tufted ducks, Aythya fuligula. The effects of differing foraging costs on dive duration and rate of oxygen uptake (VO2up) at the surface were measured during bouts of voluntary dives to a food tray. The birds were trained to surface into a respirometer after each dive, so that changes in VO2up over time could be measured. The tray held either just food or closely packed stones on top of the food to make foraging energetically more costly. In contrast to predictions from the Houston & Carbone model, foraging time (tf) increased after dives incorporating higher foraging energy costs but surface time (ts) remained the same. While optimal diving models have assumed that the cumulative oxygen uptake curve is fixed, VO2up increased when the energy cost of the dive increased. The optimal breathing model quantitatively predicted ts in both conditions and oxygen consumption during foraging (m2tf) in the control condition, for the mean of all ducks. This offers evidence that the ducks were diving optimally and supports the fundamentals of optimal diving theory. However, the model did not consistently predictts or m2tf for individual birds. We discuss the limits of optimal foraging models for air-breathing divers caused by individual variation. To evaluate the efficiency of oxygen (O2) uptake from water through the fish gill lamellar system, a cost function (CF) representing mechanical power expenditure for water ventilation and blood circulation through the gill was formulated, by applying steady-state fluid mechanics to a homogeneous lamellar-channel model. This approach allowed us to express CF as the function of inter-lamellar water channel width (w) and to derive an analytical solution of the width (wmin) at the minimum CF. Morphometric and physiological data for rainbow trout in the literature were referred to calculate CF(w) curves and their wmin values at five intensity stages of swimming exercise. Obtained wmin values were evenly distributed around the standard measure of the width (ws = 24 μm) in this fish. Individual levels of CF(wmin) were also fairly close to the corresponding CF(ws) values within a 10% deviation, suggesting the reliability of approximating [CF(wmin) = CF(ws)]. The cost-performance of O2 uptake through the gill (ηg) was then assessed from reported data of total O2 uptake/CF(ws) at each intensity stage. The ηg levels at any swimming stage exceeded 95% of the theoretical maximum value, implying that O2 uptake is nearly optimally performed in the lamellar-channel system at all swimming speeds. Further analyses of O2 transport in this fresh water fish revealed that the water ventilation by the buccal/opercular pumping evokes a critical limit of swimming velocity, due to confined O2 supply to the peripheral skeletal muscles, which is avoided in ram ventilators such as tuna.  相似文献

We measured oxygen uptake in 3- to 13-day-old Heliothis virescens (tobacco budworm) larvae. The “test” group was infected with cytoplasmic polyhedrosis virus (CPV) while the “control” group was not. The healthy budworms had an average oxygen uptake of 7.64 μl of O2/mg body weight, while those infected with CPV had an average uptake of 47.11 μl of O2/mg body weight/hr. The weights of larvae from the two groups were likewise recorded. Budworms from the “control” group showed an average weight of 186.9 mg, while larvae infected with CPV had an average weight of 18.3 mg.  相似文献

The effects of time after exposure to acetylene and of nodule excision were examined using a flow-through system. After a transient depression in the rate of acetylene reduction that began about 1.5 min after exposure to acetylene, the rate recovered to 98% of the initial maximum value after 40 min. After nodule excision the rate stabilized to 90% of the initial maximum value observed in the intact plant.Excised nodules, measured at 6-min intervals in a closed system, with frequent changes of the gas mixture, were used for the remaining experiments. Acetylene reduction by the nodules increased rapidly as temperature was increased between 6 and 26°C. Between 26 and 36°C there was relatively little effect of temperature on acetylene reduction.Nodules and cultures ofFrankia were compared with respect to the effect of temperature and pO2 (partial pressure of oxygen) on oxygen uptake. Cultures ofFrankia were grown on a nitrogen-free medium at either 0.3 kPa O2 (vesicles absent) or 20 kPa O2 (vesicles present). Oxygen uptake by nodules (vesicles absent) and by vesicle-containing cultures was strongly dependent on pO2 at values below 20 kPa. This suggests the presence of a barrier to oxygen diffusion. Oxygen uptake was dependent on temperature as well as on pO2, but the Q10 was much larger for the cultures than for the nodules. This suggests that vesicles or related structures are not the source of the diffusion barrier in Casuarina nodules. Respiration by cultures ofFrankia lacking vesicles became O2-saturated at low pO2 values. Thus these cultures did not have a significant diffusion barrier. From these results it is concluded that nodules ofCasuarina cunninghamiana have a barrier to oxygen diffusion supplied by the host tissue and not byFrankia.  相似文献

Numerous reports show that both transfer and uptake of poorly-water soluble substrates are significantly enhanced in two-phase partitioning bioreactors (TPPBs). A number of hypotheses have been put forward to explain these enhancements and among them, the occurrence of direct substrate or oxygen uptake from the vector/water interface has been suggested. The objective of this paper was to quantify the direct oxygen uptake from the vector/water interface in a culture of Pseudomonas putida, performed in a stirred tank reactor, using glucose as substrate and silicone oil as vector. Despite of a sufficient dissolved O2 concentration in the vector phase (17 mg l−1) and a significant vector surface area (4,000 m−1) no significant direct O2 uptake from the vector/water interface was observed, compared to O2 uptake from the aqueous phase. From these results it was concluded that, direct O2 or substrate uptake from the vector/water interface might not be significant in TPPBs.  相似文献

A Haxo-Blinks rate-measuring oxygen electrode together with a modulated light source gave an average current signal (change in net O2 exchange) and a modulated current signal (photosynthetic O2 evolution). Using this apparatus, net O2 exchange and photosynthetic O2 evolution at low intensities have been studied in the green alga, Chlamydomonas reinhardi. At both 645 nm and 695 nm, the curves of net O2 exchange as a function of light intensity were steeper at lowest intensities than about compensation, indicative of the Kok effect. The effect was greater at 695 nm than at 645 nm. The corresponding curves of photosynthetic O2 evolution, on the other hand, showed no Kok effect; here, the slope was lowest at lowest intensity. The absence of the Kok effect in O2 evolution, together with its sensitivity to monofluoroacetic acid, show that it is due to an interaction of photosynthesis and respiration. The effect was exaggerated by 3-(3,4-dichlorophenyl)-1,1-dimethylurea. In the presence of concentrations of this inhibitor sufficient to inhibit O2 evolution completely, a light-induced change in net O2 exchange remained. This was interpreted as a system I dependent depression of respiratory O2 uptake. The Kok effect remained undiminished in concentrations of carbonyl cyanide m-chlorophenylhydrazone and 2,4-dinitrophenol which partially uncoupled either oxidative phosphorylation alone or both oxidative and photosynthetic phosphorylations. The above results can be explained within a model of the Kok effect in which O2 uptake is depressed by diversion of reductant away from respiratory electron transport and into photosystem I. The same photodepression of O2 uptake also appears to account for a transient in net O2 exchange seen in several algae upon turning off the light.  相似文献

Addition of 1mM ascorbate to isolated chloroplasts with methyl viologen (MV) as electron acceptor trebled the rate of oxygen uptake and decreased the ADPO ratio to a third of that with no ascorbate present. These effects of ascorbate were reversed by superoxide dismutase (SOD), which in the absence of ascorbate had little effect on O2 uptake or ADPO ratio. A chloroplast-associated SOD activity equivalent to 500 units/mg chlorophyll was detected. The effects of ascorbate and SOD on O2 uptake were similar in both coupled and uncoupled chloroplasts. The results are consistent with the hypothesis that ascorbate stimulates O2 uptake by reduction of superoxide, which is formed by autoxidation of the added electron acceptor (MV), and which dismutates in the absence of ascorbate. Ascorbate does not seem to stimulate O2 uptake by replacing water as the photosystem II donor.  相似文献

Tumor hypoxia can be identified by [18F]FAZA positron emission tomography, or invasively using oxygen probes. The impact of anesthetics on tumor hypoxia remains controversial. The aim of this comprehensive study was to investigate the impact of isoflurane and ketamine/xylazine anesthesia on [18F]FAZA uptake and partial oxygen pressure (pO2) in carcinoma and muscle tissue of air- and oxygen-breathing mice.

Methods

CT26 colon carcinoma-bearing mice were anesthetized with isoflurane (IF) or ketamine/xylazine (KX) while breathing air or oxygen (O2). We performed 10 min static PET scans 1 h, 2 h and 3 h after [18F]FAZA injection and calculated the [18F]FAZA-uptake and tumor-to-muscle ratios (T/M). In another experimental group, we placed a pO2 probe in the tumor as well as in the gastrocnemius muscle to measure the pO2 and perfusion.

Results

Ketamine/xylazine-anesthetized mice yielded up to 3.5-fold higher T/M-ratios compared to their isoflurane-anesthetized littermates 1 h, 2 h and 3 h after [18F]FAZA injection regardless of whether the mice breathed air or oxygen (3 h, KX-air: 7.1 vs. IF-air: 1.8, p = 0.0001, KX-O2: 4.4 vs. IF-O2: 1.4, p < 0.0001). The enhanced T/M-ratios in ketamine/xylazine-anesthetized mice were mainly caused by an increased [18F]FAZA uptake in the carcinomas. Invasive pO2 probe measurements yielded enhanced intra-tumoral pO2 values in air- and oxygen-breathing ketamine/xylazine-anesthetized mice compared to isoflurane-anesthetized mice (KX-air: 1.01 mmHg, IF-air: 0.45 mmHg; KX-O2 9.73 mmHg, IF-O2: 6.25 mmHg). Muscle oxygenation was significantly higher in air-breathing isoflurane-anesthetized (56.9 mmHg) than in ketamine/xylazine-anesthetized mice (33.8 mmHg, p = 0.0003).

Conclusion

[18F]FAZA tumor uptake was highest in ketamine/xylazine-anesthetized mice regardless of whether the mice breathed air or oxygen. The generally lower [18F]FAZA whole-body uptake in isoflurane-anesthetized mice could be due to the higher muscle pO2-values in these mice compared to ketamine/xylazine-anesthetized mice. When performing preclinical in vivo hypoxia PET studies, oxygen should be avoided, and ketamine/xylazine-anesthesia might alleviate the identification of tumor hypoxia areals.  相似文献

Summary Simultaneous measurements of ventilatory frequency, tidal volume, O2 uptake, CO2 output and cardiac frequency were made in the diamondback water snake,Natrix rhombifera while breathing hypoxic (15% to 5% O2 in N2) or hypercarbic (2% to 10% CO2 and 21% O2 in N2) gases. The snakes responded to hypoxia by increasing tidal volume and decreasing ventilatory frequency resulting in little change in ventilation (50% increase at 5% inspired O2), or O2 uptake and only a light increase in CO2 output. Hypercarbia to 4.2% inspired CO2 resulted in a slight hyperventilation but ventilation was depressed at 6.3% inspired CO2 and became erratic at higher concentrations. The resting rate of O2 uptake was maintained throughout hypercapnia. Heart rate increased during hypoxia and decreased during hypercapnia. Cutaneous O2 uptake increased during extreme hypoxia (5% inspired O2) and cutaneous CO2 output increased during hypercapnia, probably due to changes in the body-to-ambient gas gradients (Crawford and Schultetus, 1970). Both pulmonary oxygen uptake and ventilation were dramatically increased immediately following 10–15 min experimental dives. The increased ventilation was achieved primarily through an increased tidal volume.  相似文献

In spinach thylakoids prepared from intact chloroplasts by shocking in the presence of ascorbate to preserve the operation of ascorbate peroxidase, the rate of oxygen uptake with methyl viologen as acceptor decreased in response to the addition of H2O2. Such a decrease was not observed in the presence of KCN or when the thylakoids lost ascorbate peroxidase activity. Illumination of intact chloroplasts in the presence of H2O2 and methyl viologen showed an initial rate of oxygen exchange, which is intermediate between the initial rate of oxygen evolution in the presence of H2O2 alone and steady-state oxygen uptake in the presence of methyl viologen. The data showed that monodehydroascorbate radical generated in ascorbate peroxidase reaction could compete with methyl viologen for electrons supplied by the electron transport chain in both thylakoids and intact chloroplasts. During the illumination of intact chloroplasts the rate of oxygen uptake increased. The presence of nigericin swiftly led to steady-state oxygen uptake, and to a clear-cut 1:1 relationship between the electron transport rate estimated from fluorescence assay and the electron transport rate determined from oxygen uptake, taking the stoichiometry 1O2:4e. The increase in oxygen uptake was attributed to the cessation of monodehydroascorbate radical generation brought about by consumption of intrachloroplast ascorbate in the peroxidase reactions, and the effects of nigericin were explained by acceleration of such consumption. The competition between methyl viologen and monodehydroascorbate radical in the intact chloroplasts was estimated under various conditions.  相似文献

It is postulated that the burst of oxygen consumption and H2O2 formation following phagocytosis by polymorphonuclear leukocytes is due to the action of an oxidase located in the plasma membrane. The cyanide-resistant oxygen consumption of resting polymorphonuclear leukocytes was also found to be stimulated by 2,4-dichlorophenol with H2O2 being the sole product formed. NADH and NADPH added to the leukocytes greatly enhanced the oxygen consumption and were oxidized in the process without penetrating the leukocytes. Mn2+ stimulated this oxidase activity. The apparent Km values for added NADH and NADPH were 50 and 40 μm, respectively, with a V of 300 nmol/mg protein/min. A stoichiometry of 1 mol H2O2 formed per mol of NAD(P)H was found. Whilst the oxidase is similar to the oxidase properties of a peroxidase, myeloperoxidase is not responsible for the activity.  相似文献

The nature of the process responsible for the stationary O2 uptake occurring in the light under saturating CO2 concentration in Chlamydomonas reinhardii has been investigated. For this purpose, a mass spectrometer with a membrane inlet system was used to measure O2 uptake and evolution in the algal suspension. First, we observed that the O2 uptake rate was constant (about 0.5 micromoles of O2 per milligram chlorophyll per minute) during a light to dark transition and was not affected by 3-(3,4-dichlorophenyl)-1,1-dimethylurea. Salicylhydroxamic acid had no effect on O2 uptake in the dark or in the light, but was found to have the same inhibitory effect either in the dark or in the light when added to cyanide-treated algae. The stimulation of the O2 uptake rate due to the uncoupling effect of carbonyl cyanide m-chlorophenylhydrazone was about the same in the dark or in the light. From these results, we conclude that mitochondrial respiration is maintained during illumination and therefore is not inhibited by high ATP levels. Another conclusion is that in conditions where photorespiration is absent, no other light-dependent O2 uptake process occurs. If Mehler reactions are involved, in Chlamydomonas, under conditions where both photosynthetic carbon oxidation and reduction cycles cannot operate (as in cyanide-treated algae), their occurrence in photosynthesizing algae either under saturating CO2 concentration or at the CO2 compensation point appears very unlikely. The comparison with the situation previously reported in Scenedesmus (R. J. Radmer and B. Kok 1976 Plant Physiol 58: 336-340) suggests that different O2 uptake processes might be present in these two algal species.  相似文献

The basis of inhibition of photosynthesis by single acute O3 exposures was investigated in vivo using analyses based on leaf gas exchange measurements. The fully expanded second leaves of wheat plants (Triticum aestivum L. cv Avalon) were fumigated with either 200 or 400 nanomoles per mole O3 for between 4 and 16 hours. This reduced significantly the light-saturated rate of CO2 uptake and was accompanied by a parallel decrease in stomatal conductance. However, the stomatal limitation, estimated from the relationship between CO2 uptake and the internal CO2 concentration, only increased significantly during the first 8 hours of exposure to 400 nanomoles per mole O3; no significant increase occurred for any of the other treatments. Analysis of the response of CO2 uptake to the internal CO2 concentration implied that the predominant factor responsible for the reduction in light-saturated CO2 uptake was a decrease in the efficiency of carboxylation. This was 58 and 21% of the control value after 16 hours at 200 and 400 nanomoles per mole O3, respectively. At saturating concentrations of CO2, photosynthesis was inhibited by no more than 22% after 16 hours, indicating that the capacity for regeneration of ribulose bisphosphate was less susceptible to O3. Ozone fumigations also had a less pronounced effect on light-limited photosynthesis. The maximum quantum yield of CO2 uptake and the quantum yield of oxygen evolution showed no significant decline after 16 hours with 200 nanomoles per mole O3, requiring 8 hours at 400 nanomoles per mole O3 before a significant reduction occurred. The photochemical efficiency of photosystem II estimated from the ratio of variable to maximum chlorophyll fluorescence and the atrazine-binding capacity of isolated thylakoids demonstrated that photochemical reactions were not responsible for the initial inhibition of CO2 uptake. The results suggest that the apparent carboxylation efficiency appears to be the initial cause of decline in photosynthesis in vivo following acute O3 fumigation.  相似文献

The respiratory responses to increasing temperature and progressive hypoxia were examined relative to temperature acclimation in the nonindigenous, brown mussel, Perna perna (Mytilidae) from the Gulf of Mexico. When oxygen uptake rate (V?O2) was recorded at near full air O2 saturation, rate-temperature curves for Texas specimens of P. perna were sigmoidal, V?O2 generally increasing with increasing temperature but becoming suppressed as temperatures approached 10 and 30 °C, corresponding closely to this species' incipient thermal limits. At each tested temperature, V?O2 did not differ among individuals acclimated to 15, 20, or 25 °C. Lack of thermal acclimation was also reflected in acclimatory Q10 values>1.0 (range=1.34-2.14) recorded across acclimation groups at test temperatures equivalent to acclimation temperature. Low acute respiratory Q10 values in all acclimation groups across 15-20 °C indicated a limited capacity for thermal regulation of V?O2 within this temperature range. The ability of P. perna to regulate O2 uptake with progressive hypoxia was temperature-dependent, increasing from poor O2 regulation at 10 °C to good regulation at 30 °C. The O2 regulatory ability of P. perna and other open-water mytilids in declining O2 concentrations does not greatly differ from that of estuarine heterodont bivalves, suggesting that it is not a major factor preventing open-water species, such as P. perna, from invading estuarine environments. However, P. perna's inability to regulate O2 uptake at temperatures>25 °C combined with its relatively low upper thermal limit of 30 °C will likely prevent it from establishing permanent estuarine populations on Gulf of Mexico shores.  相似文献

Summary The uptake of carbohydrates and oxygen by cell suspension cultures of the plant Eschscholtzia californica (California poppy) was studied in relation to biomass production in shake flasks, a 1-1 stirred-tank bioreactor and a 1-1 pneumatically agitated bioreactor. The sequence of carbohydrate uptake was similar in all cases, with sucrose hydrolysis occurring followed by the preferential uptake of glucose. The uptake of fructose was found to be affected by the oxygen supply rate. Carbohydrate utilization occurred at a slower rate in the bioreactors. Apparent biomass yields, Y X/S, ranged from 0.42 to 0.50 g biomass/g carbohydrate, while true biomass yields, Y X/S, were about 0.69 g/g. The maintenance coefficient for carbohydrate, m S, ranged between 0.002 and 0.008 g/dry weight (DW) per hour. The maximum measured specific oxygen uptake rate was 0.56 mmol O2/g DW per hour and occurred early in the growth stage. The decline in specific uptake rate coincided with a decline in cell viability. The oxygen uptake rate was faster in shake flasks, corresponding to the higher growth rate obtained. The true growth yield on oxygen, YX/O2, was calculated to range from 0.83 to 1.23 g biomass/g O2, while the maintenance coefficient, mO2, ranged from 0.15 to 0.25 mmol O2/g DW per hour. Glycolate and ammonia excretion plus oxygen exchanges were measured in the light in l-methionine-dl-sulfoximine treated air-grown Chlamydomonas reinhardii. At saturating CO2 (between 600 and 700 microliters per liter CO2) neither glycolate nor ammonia were excreted, whereas at the CO2 compensation concentration (<10 microliters per liter CO2) treated algae excreted both glycolate and ammonia at rates of 37 and 59 nanomoles per minute per milligram chlorophyll, respectively. From the excretion values we calculate the amount of O2 consumed through the glycolate pathway. The calculated value was not significantly different from the component of O2 uptake sensitive to CO2 obtained from the difference between O2 uptake of the CO2 compensation point and at saturating CO2. This component was about 40% of stationary O2 uptake measured at the CO2 compensation point. From these data we conclude that glyoxylate decarboxylation in air-grown Chlamydomonas represents a minor pathway of metabolism even in conditions where amino donors are deficient and that processes other than glycolate pathway are responsible for the O2 uptake insensitive to CO2.  相似文献
