The pattern of distribution of cholinergic and adrenergic nerves in the uterus of albino rats and guinea pigs was examined histochemically. In the albino rat, the uterus was found well-innervated by both adrenergic and cholinergic nerves with a clear regional variation. Dense innervation was demonstrated at the tubal and cervical ends of the uterus and in the cervix. Cholinergic nerves supplying the glands were more numerous than the adrenergic nerves which were relatively few. In the guinea-pigs, the uterus was richly innervated by adrenergic nerves with a clear regional variation. No cholinesterase-positive nerves or nerve cells were demonstrated.  相似文献

The ovarian vascular pedicle and ovarian suspensory ligament were briefly frozen to destroy the nerves. Examination of sections from the ovary, oviduct and utero-tubal junction by fluorescence histochemistry showed that they were usually devoid of adrenergic nerves. Measurement of noradrenaline in segments of the uterine horn by high-performance liquid chromatography showed that the transmitter was eliminated from the upper third but not the middle or lower thirds of the uterine horn. Unilateral or bilateral denervations at metoestrus in cyclic rats did not affect either the number of ovulations or the numbers or spacing of conceptuses at Day 7 of pregnancy. Bilateral denervations on Days 4, 7 or 11 of pregnancy did not affect ovarian weights or numbers of conceptuses observed 1 week later. Plasma progesterone concentrations were at least as high in the denervated groups as in the sham-operated control groups. After unilateral or bilateral denervations at Day 15, pregnancy continued normally and birth of normal young occurred, without apparent problems, at the same time as for sham-operated rats. The mothers tended their young and allowed them to suck. It is concluded that the adrenergic innervation of the ovary in the rat is not required for its normal function during pregnancy; that of the oviduct and mesosalpinx is not required for ovum pick-up or for transport of oocytes, spermatozoa or early embryos; and that of the utero-tubal junction is not required for uterine motility involved in embryo spacing and parturition.  相似文献

Uterine adrenergic and cholinesterase (AChE)-positive innervation of the sheep uterus during anestrus and at 4 stages of pregnancy were examined by histochemical methods. In addition, uterine and cervical myometrium concentrations of norepinephrine (NE) and dopamine (DA) were determined using high-performance liquid chromatography. During anestrus, adrenergic and AChE-positive nerve fibers in the uterine myometrium and endometrium were primarily associated with the vasculature. Innervation of myometrial smooth muscle was almost exclusively by adrenergic fibers. In the endometrium, fibers of both types were observed closely associated with endometrial glands, and adrenergic fibers were observed in the connective tissue beneath the luminal epithelium. Density of uterine innervation decreased by day 65 of pregnancy with an additional decrease by day 105. Myometrial NE concentrations were higher in the cervix than the uterus. Uterine NE concentrations generally were not affected by pregnancy. Although cervical NE per gram of tissue decreased during pregnancy, this effect of pregnancy was not detected when NE was expressed per microgram of DNA. Myometrial DA concentrations were higher in uterine segments than in the cervix. DA concentrations decreased during pregnancy in all tissues except the posterior uterine segment. The DA to NE ratio in the uterus was greater than that for the cervix and was not generally affected by the stage of pregnancy. These results demonstrate that cholinergic and adrenergic nerves supply the sheep uterus. Decreasing fiber density during pregnancy suggests that a majority of the innervation to the sheep uterus is supplied by 'short' nerve fibers whose activity is regulated by steroids of pregnancy. The possible role of DA as a neurotransmitter in the sheep uterus is discussed.  相似文献

Summary The adrenergic innervation of the juxtaglomerular complex was studied in kidneys from mice, rats, guinea-pigs, rabbits, cats, dogs, pigs, monkeys, and humans using fluorescence histochemistry of neuronal nor-adrenaline and autoradiography of 3H-noradrenaline. The localization of the nerves was established by phase contrast optics or by perfusing the vascular system with India ink. Adrenergic nerve terminals, exhibiting a formaldehyde-induced fluorescence and having the ability to take up and accumulate 3H-noradrenaline, were easily identified when they enclosed the glomerular afferent arteriole. They continued in between and close to the macula densa and lacis cells to supply the glomerular efferent arteriole. The nerves could be seen to accompany this arteriole for a considerable distance until they branched off to the vasa recta in the juxtamedullary region and to adjacent cortical veins. This innervation pattern was found to be a constant feature except in kidneys from guinea-pigs and cats, in which post-glomerular adrenergic nerves were not found in some of the superficial glomerular units. The fluorescence in all adrenergic fibres supplying the juxtaglomerular complex disappeared after removal of the aortico-renal ganglion, showing that they belong to a common system of renal sympathetic nerves.This work is dedicated to Professor Wolfgang Bargmann in honour of his seventieth birthday, January 26, 1976  相似文献

Summary In the guinea-pig uterus, by use of an indirect immunofluorescence method, S-100 immunoreactivity was found to be restricted to nerves that corresponded in number, distribution and type to adrenergic axons and preterminals. With advancing pregnancy S-100 immunoreactivity completely disappeared in uterine tissue adjacent to a fetus, in parallel with an ultrastructural degeneration of the adrenergic innervation. In the cervix and the uterine horn devoid of a fetus, however, the number and distribution of S-100-immunoreactive nerves was seemingly unchanged and no ultrastructural changes were found in adrenergic nerves. In contrast, chemical sympathectomy produced by 6-hydroxydopamine did not change S-100 immunoreactivity of uterine nerves. These findings suggest that there are differences in the denervation effected by chemical and by pregnancy-induced sympathectomy. The latter probably represents a special type of adrenergic denervation by inducing a degeneration of Schwann cells in addition to destruction of neuronal structures. This may explain the differences in the speed of regeneration of uterine adrenergic nerves following the two types of denervation.  相似文献

The pattern of uterine innervation by noradrenergic (NA) and acetylcholinesterase-positive (AChE) nerves in different reproductive stages of the adult Japanese long-fingered bats were investigated histochemically and immunohistochemically. In the non-pregnant bat, the uterine horn was supplied with abundant NA and AChE nerves. These two types of nerves were closely associated with the uterine arteries and myometrial smooth muscles. In the pregnant bat, NA and AChE nerves supplying the uterus did not degenerate much during hibernating period, but reduced markedly after arousal. In the postpartum bat, the density of nerves recovered progressively. The significant change in the innervation pattern of uterine NA and AChE nerves in the pregnant bats under and after hibernation, and in the postpartum bat must be considered in relation to the adrenergic and cholinergic controlling mechanisms on the uterine function that is matched for the unique reproductive cycle of this bat.  相似文献

The paper describes the effect of exogenous oestradiol and progesterone on noradrenalin (NA) content in the ovary, tuba uterina, uterus, and vagina, and on the morphological features of adrenergic nerves in these organs in sexually immature female pigs. 30 animals were used, of body weight 37 to 43 kg, and age 115 to 125 d. The animals were divided randomly into 4 groups. Group I (n = 6) constituted the control, group II (n = 8) was given oestradiolum benzoicum, group III (n = 8) was given progesterone, and group IV (n = 8) was given oestradiol and progesterone. Oestradiol injections resulted in a 1.5-fold increase of NA content in the ovary, a 4.5-fold increase in the tuba uterina, and a decrease of NA content in the uterine horn, cervix, and in the vagina, to 0.6 of the control NA level. Progesterone injections did not cause changes in NA content in the ovary, while there was 8.5-fold increase of NA in the oviduct, a 4-fold increase in the uterile horn, a 6-fold increase in the cervix, and a 3-fold increase in the vagina compared to the control. Joint injections of oestradiol and progesterone resulted in a decrease of NA content in the ovary to about 0.5 of the control level, and in an increase of NA content of about 5-fold in the tuba uterina, 7-fold in the uterine horn, 8-fold in the cervix, and 2-fold in the vagina. The increase of NA content in the organs was coupled with a slight increase in the density of the adrenergic innervation and the intensity of fluorescence of these nerves compared to the control. The results were compared to the results of similar but not identical studies, carried out on other mammals.  相似文献

The innervation of the uterus in the ovoviviparous urodele Salamandra salamandra was studied. In whole mount preparations of the thin-walled uterus of pregnant females, a dense adrenergic network was demonstrated using a modified glyoxylic acid fluorescence technique. Based on vesicle type and cytochemical reactivity after chromate/dichromate fixation for electron microscopy at least two types of neural process were distinguished and classified as adrenergic and cholinergic. Both types are preferentially situated above or between the smooth muscles of the uterine tissue. Adjacent to the muscles in the walls of arterioles mainly adrenergic fibers are seen. Using high performance liquid chromatography with electrochemical detection in tissue homogenates of uterus a considerable amount of noradrenaline could be identified. The significance of the dense innervation is discussed with respect to the function of the uterus during pregnancy and birth.  相似文献

The present study was undertaken to investigate the structural changes in both cholinesterase (ChE)-positive nerve fibers and adrenergic nerves with formaldehyde-induced fluorescence in pregnant and postpartum uteri of both the albino rat and guinea pig. Particular attention was directed to the relationship between these changes and the local factors associated with the growing fetus. ChE reaction was absent in the control and pregnant uterus of the guinea pig. In the albino rat, there were signs of degeneration in pregnancy. These were evidenced by vacuolation of large nerve trunks and the presence of focal segments with very faint reaction along the course of the nerve bundles. Myometrial segments from fetus-containing horns showed some fragmented nerve fibers, but at the same time some other normal ones. Most of the fine nerve bundles gave a weak reaction. Three weeks after delivery, multiple ChE fibers were found in the uterus of the albino rat. The normal appearance was, however, not regained and some nerve fibers were still fragmented. Noradrenergic (NA) nerve fibers were disintegrated and markedly reduced in number in the myometrium of the pregnant uterus of both the guinea pig and albino rat, particularly in the uterine horns that were distended by fetuses. The number of NA fibers was not significantly reduced in the tubal ends of the albino rat uterus. Three weeks after delivery, normal NA fibers were seen in the myometrium of both the albino rat and guinea pig uterus. Nerves with reduced fluorescence reaction were observed less frequently.  相似文献

Summary The occurrence and distribution of peptidergic nerves in the guinea pig uterus was studied by means of immunocytochemistry using numerous neuropeptide antisera. Neuropeptide Y (NPY)-immunoreactive (IR) nerves were the most abundant, whereas substance P (SP)-, calcitonine gene-related peptide (CGRP)-, and neurokinin A (NKA)-IR nerves were less frequent, and peptide histidine isoleucine (PHI)-IR nerves were the most sparse. Chemical sympathectomy by means of 6-hydroxydopamine, and capsaicin treatment revealed the division of the peptidergic nerves into three separate populations: (1) NPY-IR nerves, which co-existed with adrenergic nerves, (2) SP-, CGRP-and NKA-IR nerves, which mutually co-existed, and (3) PHI-IR nerves. Parallel-running adrenergic/NPY-IR and SP-IR nerves could be found with very similar although not completely identical morphological appearance. Paracervical ganglia contained neurotensin-and dynorphin A-IR cell bodies in addition to cell bodies with immunoreactivities similar to those in prevertebral ganglia. Combined retrograde tracing with True blue and immunocytochemistry showed that the adrenergic and NPY-IR uterine nerves originate in paracervical and prevertebral ganglia. In the prevertebral ganglia the cellular origin was the same for adrenergic and NPY-IR nerves. In contrast, SP-, CGRP-,and NKA-IR nerves originated in dorsal root ganglia. At full-term pregnancy all the neuropeptide immunoreactivities had vanished, probably reflecting a fetus-induced general nerve degeneration.  相似文献

The adrenergic innervation of the urinary bladder of normal female and pregnant rats has been studied using a fluorescence histochemical method. The bladder is richly innervated by adrenergic nerve fibres as is evidenced by the presence of numerous adrenergic nerves in the adventitia, musculosa and submucosa. However, adrenergic nerve cells could not be observed. During pregnancy, adrenergic nerve fibres showed signs of degeneration, as most of the nerve fibres disappeared and the surviving fibres were much swollen. 10 days after parturition the pattern and density of adrenergic innervation became almost similar to those of the control animals.  相似文献

The indirect immunofluorescence technique was used to determine the distribution of peptide-containing axons in the gall bladder of the cane toad, Bufo marinus. In addition, the adrenergic innervation of the gall bladder was examined by use of immunoreactivity to the catecholamine-synthesizing enzyme, tyrosine hydroxylase, and glyoxylic acid-induced fluorescence. On the basis of peptide coexistence, two intrinsic populations of neurones and their projecting fibres could be distinguished substance P neurones and vasoactive intestine peptide neurones. Neither of these two types of neurones contained any other colocalized neuropeptides. Four populations of nerve fibres arising from cell bodies outside the gall bladder were identified: nerves containing colocalized galanin, somatostatin and vasoactive intestinal peptide; nerves containing colocalized calcitonin gene-related peptide and substance P; adrenergic nerves containing neuropeptide Y; and nerves containing only adrenaline.  相似文献

The local destination transfer of prostaglandin E2 (PGE2) from the uterine lymph to arterial blood supplying the ovary and its retrograde transfer to arterial blood supplying the uterine horn and the effect of additional delivery of PGE2 into the ovary on the secretion of steroid hormones was studied in early pregnant gilts. The injection of PGE2 under the perimetrium caused an increase (P<0.001) in PGE2 concentration in both uterine venous effluent and ovarian and uterine arterial blood. The infusion of PGE2 into the ovarian artery increased the concentration of progesterone in ovarian venous blood on day 13 of pregnancy during (P<0.05) and after (P<0.001) infusion, and on day 14 of pregnancy after infusion (P<0.01). In conclusion, local destination transfer of PGE2 from uterine lymph and venous blood to the ovary may affect luteal function, and retrograde transfer of PGE2 to the arterial blood supplying the uterus may contribute to the prevention of regressive changes of the endometrium in early pregnant gilts.  相似文献

Both technics in nonpregnant animals revealed a rich network of acetylcholinesterase and norepinephrine positive nerves around the blood vessels while the muscular innervation was moderately positive. Acetylcholinesterase fibers were also concentrated beneath the surface epithelium forming a plexus-like arrangement where isolated ganglion cells could be seen. At mid pregnancy cholinergic and adrenergic fibers decreased in density. The intensity of fluorescence was weaker and nerve fiber morphology was modified. We endeavoured to relate our findings to the problem of the neural control of contractions and the opening of the cervix of the ewe which is poorly supplied in nerve fibers, particularly at mid pregnancy.  相似文献

Summary The adrenergic and acetylcholinesterase-containing nerves of the hearts of mice, rats, guinea-pigs, rabbits, and cats were studied. The fluorescence technique of Falck and Hillarp was used for the demonstration of adrenergic nerves, whereas a modified Koelle cholinesterase technique was used for the cholinesterase-containing nerves. The inhibitors used were Mipafox, iso-OMPA and Nu 683. Microspectrofluorometry was used to identify the structures containing dopamine.Adrenergic as well as acetylcholinesterase-containing fibres were found in all parts of the heart, most abundantly in the atria. Dense nerve plexa supplied the sinoarial and atrioventricular nodes. There was a plexus of both fibre types in the endocardium and on the atrial side of the valves. In the valves, it could be shown that adrenergic and cholinesterase-containing fibres ran closely parallel to each other. Indirect evidence suggested that this applies also to the myocardium.No nerve fibres containing dopamine were revealed in the microspectrofluorometer. The dopamine previously found in the atria seems, instead, to be situated in so-called small intensely fluorescent cells.No adrenergic ganglion cells were found in the heart despite extensive search. The vagus of rabbits was found to contain only few adrenergic preterminals.  相似文献

Summary The adrenergic and cholinergic innervation to the rat iris has been studied at a light and electron microscopic level. Catecholamine fluorescence histochemistry showed adrenergic nerves to be present in both the dilatator and the constrictor pupillae regions. At a fine structural level the terminal innervation of the iris was studied and criteria for the differentiation between presumptive adrenergic and presumptive cholinergic axon terminals were examined. To aid this examination presumptive adrenergic axons were either labelled with the false adrenergic transmitter, 5-hydroxydopamine, or chemical sympathectomy performed using 6-hydroxydopamine. The value of using acetylcholinesterase staining as a marker for cholinergic nerve terminals was also studied.Results showed a mixed adrenergic/cholinergic innervation to the dilatator pupillae. In the constrictor pupillae an exclusively cholinergic innervation was found although adrenergic and cholinergic nerves were found supplying the blood vessels and at the dilatator-constrictor interface. These findings are discussed with regard to innervation-function relationships in the iris.  相似文献

Summary Problems associated with the low intensity and fast fading of the formaldehydeinduced fluorescence of adrenergic nerves in young foetal human and rat gut have been overcome by loading the nerves with a primary catecholamine. This has been achieved by incubating the tissue, before freeze-drying, in Krebs solution containing added noradrenaline or -methyl-noradrenaline. With this method fluorescent nerves have been revealed at a stage before any were demonstrable by normal procedures.This work was supported by the Australian Research Grants Committee and the National Heart Foundation of Australia. The relationship between nutrition and reproduction in sheep has been the subject of research in several international groups. This review will particularly focus on the effects of undernutrition on the potential causes of reproductive failure including abnormalities of the ovum or the embryo, luteal inadequacy and failure of the supply of progesterone to the uterus, or the mechanisms involved in maternal recognition of pregnancy. The level of nutrition and peripheral progesterone concentrations are inversely related, and increased rates of embryo loss, associated with higher progesterone concentrations in ewes with low levels of nutrition have been reported. Undernutrition may act through changes in the distribution of progesterone in the endometrium. Thus, lower endometrial levels on day 5 of the cycle in ewes fed half of their maintenance requirements have been observed, providing a link between the known role of progesterone in embryo survival by the modulation of uterine function and the higher embryo losses found in undernourished ewes. The evidence of an effect of maternal nutrition on IFNtau secretion from the conceptus and of PGF2alpha production from the uterus is presented. Moreover, undernutrition provokes a reduction in the sensitivity of the endometrium to progesterone that may affect embryo survival. Finally, a state of undernutrition induces changes in the endometrial sensitivity to steroid hormones at early stages of pregnancy that could adversely alter uterine environment to the detriment of embryo survival.  相似文献

Summary In the guinea pig myometrium, the adrenergic nerves selectively demonstrated at the ultrastructural level after treatment with 5-OH-DA, show varying degree of degeneration during pregnancy. The changes are more extensive in a late gestational stage (40–45 days) than in an early one (20–25 days), and are particularly evident in the uterus overlying the conceptus as compared to the regions between the fetuses. Scattered degenerative changes were also observed in myometrial specimens from virgin animals, but probably reflect the normal continuous turnover of axons.  相似文献
