While integration of ColE1 had not been observed previously by ordinary suppressive integration, a dnaA (Ts) E. coli strain with Tn5 at various sites of the chromosome and ColE1 or its mini-derivative, pAO3, but not pSC101, inserted by the same transposon produced integratively suppressed strains depending on the RecA function. In contrast to Hfr strains made with a stringently controlled plasmid, they contained the plasmid not only in an integrated but in an autonomous state at an amount comparable to the strain containing the plasmid only autonomously. Introduction of a RecA-deficient mutation to the strain with an integrated ColE1 derivative through conjugation failed. This is likely to be due to lethality of such a strain without RecA-dependent excision of the integrated high copy number plasmid or to quantitative deficiency of DNA polymerase I in addition to the recA mutation.  相似文献

During the process of transformation Haemophilusinfluenzae cells bind its own DNA but little or no foreign DNA. This specificity for recognition of DNA was studied by cloning Haemophilus DNA in E. coli. Haemophilus DNA fragments were cloned using plasmid pBR322 as a vector. The fragment cH7 cloned in pBR322 was found to be homologous to Haemophilus DNA and shown to bind irreversibly to competent Haemophilus cells. The fact that cH7 isolated from E. coli lacks Haemophilus modification leads to the conclusion that modification does not play a role in the uptake mechanism. Uptake specificity is a function of recognition sequences that reside in DNA itself.  相似文献

We report the construction of recombinant plasmids containing part of the mitochondrial DNA of Drosophilamelanogaster. Of the four fragments of this DNA generated by the restriction endonuclease HindIII, two were successfully cloned into the HindIII site of the plasmid pCM2. Unexpectedly the other two fragments could not be isolated by cloning into the HindIII site of either pCM2 or pBR322. Part of a third fragment, containing the gene for the large ribosomal RNA, was incorporated into the PstI site of pBR322. We show that this recombinant plasmid contains sequences complementary to an abundant RNA species which is present in Drosophila embryos and which binds to oligo-dT-cellulose.  相似文献

DNA reassociation kinetics have been partly elucidated for the brine shrimp Artemia salina, using calf thymus DNA as a standard. The Artemia single-copy DNA sequences comprise 45% of the genome; sequences having a repetition frequency of about 2–90 are not detectable. The average repetition frequency of the intermediately redundant DNA component is about 5,000 copies. Reassociation kinetic data are consistent with a unit genome size of 1.5 pg.  相似文献

Mitochondrial DNA from Drosophila contains high “A+T”-rich region. Its DNA replication starts in the “A+T”-rich region and proceeds unidirectionally around the molecule. In order to determine precise location of the DNA replication origin and elucidate unique feature of its nucleotide sequence, the “A+T”-rich region of mitochondrial DNA from Drosophilavirilis has been cloned in Escherichiacoli. The chimeric plasmid DNA containing the “A+T”-rich region stimulates invitro DNA replication system from Drosophilavirilis mitochondria about ten fold higher than the parental plasmid DNA, as does native mitochondrial DNA.  相似文献

Pyridoxal 5′ phosphate at concentrations < 0.5 mM inhibits polymerization of deoxynucleoside triphosphate catalysed by variety of DNA polymerases isolated from type C RNA tumor viruses, as well as E.coli, but doesnot affect the polymerase associated RNase H activity. Both phosphate and aldehyde groups of pyridoxal phosphate are essential for the inhibition which appears to be mediated through the reversible Schiff base.  相似文献

Hybrid genes were constructed between the β-lactamase gene of plasmid pBR322 and enzymatically synthesized DNA sequences coding for urokinase-like material. The recombinant plasmids, pULB1013 and pULB1010, contain DNA inserts of 220 and 535 base pairs respectively, which appear to be in the correct reading frame. The hybrid genes are expressed in Escherichia coli and their products are specifically immunoprecipitated with antiserum to urokinase. In addition injection into a rabbit of a crude extract from the clone pULB1010 leads to the production of specific antibodies against urokinase.  相似文献

FlacS is a thermostable extrachromosomal element isolated in Salmonella typhimurium which is altered in its replication as compared to its precursor Fts114lac. Sedimentation of both these plasmids in alkaline sucrose gradients has indicated a difference in their sizes. Contour length measurements of open circular plasmid DNA molecules photographed in the electron microscope have revealed the estimated molecular weight of Fts114lac to be 81 × 106 daltons while that of FlacS is 109 × 106 daltons. FlacS may carry a segment of S. typhimurium chromosomal or cryptic plasmid DNA.  相似文献

Clones of hybrid plasmids containing moderately and highly repeated sequences of Drosophila hydei exhibit positive autoradiographic signals if hybridized to labeled whole genome DNA. Such clones were screened with labeled male (XY) and female (XX) DNA, and male-specific fragments were identified. Further hybridization of male-specific clones to female XXY, and male X0 DNAs established them as containing Y-specific moderately repeated sequences. Further verification of one particular cloned fragment as Y-specific is presented and possible applications of this procedure are briefly discussed.  相似文献

The pyrene-like fluorescence of the covalent benzo(a)pyrene diol-epoxide-DNA complex prepared by reacting 7,8,-dihydrodiol 9,10-epoxy benzo(a)pyrene (BPDE) with DNA in aqueous solution in vitro, has been investigated. It is shown that this fluorescence is sensitive to molecular oxygen, to the concentration of native DNA and to the ionic strength (KCl concentration), but is insensitive to the concentration of denatured DNA. These effects are related to the conformation of the pyrene-like chromophore of BPDE. Most of the fluorescence of a dilute solution of the DNA-bound benzo(a)pyrene derivative originates from binding sites in which the pyrene moiety is not intercalated between the DNA base pairs, but is located on the outside of the DNA double helix.  相似文献

The squirrel monkey (Saimiri,sciureus), a New World primate, contains multiple copies of endogenous type D retroviral gene sequences in the cellular DNA of all its tissues. Gene sequences partially homologous to these type D virus genes are also found in the cellular DNA of normal tissues of the New World carnivore, the skunk (Mephitis,mephitis and Spilogale,putorius). We there-fore conclude that this class of viruses has, under natural conditions, been transmitted between the germ lines of these evolutionarily distant species. The example of interspecies transmission described here is the first that has been described among New World species and also the first that has been demonstrated for retroviruses other than type C viruses.  相似文献

The invitro DNA dependent synthesis of ribosomal protein L12 and the β subunit of RNA polymerase has been investigated using DNA from a plasmid which contains the genetic information for ribosomal protein L12 and the β subunit of RNA polymerase. This DNA, however, lacks the promoter region and the genetic information for the first 26 amino acids of ribosomal protein L10. It was found that L12 and the β subunit of RNA polymerase are efficiently synthesized invitro from this DNA. These results suggest that L12 and the β subunit of RNA polymerase can be synthesized from a promoter situated within the L10 gene.  相似文献

Novobiocin (and its related drug, coumermycin A1) and nalidixic acid are specific inhibitors of DNA gyrase in bacteria. These drugs inhibit many enzymatic activities in yeast; such as DNA polymerase activity in crude extracts, invitro 2-μm plasmid DNA replication, purified DNA polymerase I and II, and topoisomerase I. Therefore, the inhibition by these inhibitors in yeast is not specific for a particular enzyme.  相似文献

The 1.672 g/cm3 satellite DNA of Drosophila melanogaster was purified by successive equilibrium centrifugations in a CsCl gradient, an actinomycin DCsCl gradient, and a netropsin sulfate/CsCl gradient. The resulting DNA was homogeneous by the physical criteria of thermal denaturation, renaturation kinetics and equilibrium banding in each of the gradients listed above. In addition, the complementary strands could be separated in an alkaline CsCl gradient. Despite this rigorous purification procedure, nucleotide sequence analysis indicates the presence of two different DNA species in this satellite, poly A-A-T-A-TT-T-A-T-A and polyA-A-T-A-T-A-TT-T-A-T-A-T-A. Further physical, chemical and template properties of the isolated complementary strands demonstrate that these two repeating sequences are not interspersed with each other. This result has biological significance since sequences of this particular satellite are known to be located primarily on two different chromosomes, Y and 2. These results further suggest that the sequence heterogeneity observed in satellite DNA of higher eukaryotes may result from mixtures of very closely related but molecularly homogeneous repeated sequences each restricted to a particular chromosome or chromosomal region.  相似文献
