Now that the Clinton Administration has overturned the ban on federal funding for fetal tissue transplantation, old ethical issues renew their relevance and new ethical issues arise. Is fetal tissue transplantation necessary and beneficial? Are fetal rights violated by the use of fetal tissue in research? Is there a moral danger that the potential of fetal tissue donation will encourage elective abortions? Should pregnant women be allowed to designate specific fetal transplant recipients? What criteria should be used to select fetal tissue transplants? Whose consent should be required for the use of fetal tissue for transplantation? We review the current state of clinical research with fetal tissue transplantation, the legal history of fetal tissue research, the major arguments against the use of fetal tissue for transplantation, and the new postmoratorium ethical dilemmas. We include recommendations for guidelines to govern the medical treatment of fetal tissue in transplantation.  相似文献

The recovery of viable follicles from cryopreserved ovarian tissue would be of benefit in many areas of assisted reproduction. Structural integrity needs to be maintained following cryopreservation of ovarian tissue in order to retrieve healthy follicles which can then be cultured in vitro to produce viable oocytes. We have assessed the effect of in vitro culture of bovine tissue for 0, 1, 4, 24, or 48 h after exposure to, or cryopreservation in, dimethylsulphoxide. Immediately after freezing, normality of primary and preantral follicles within the tissue was significantly lower than for tissue exposed to the cryoprotectant without freezing or for control tissue. After 4 h in culture, cryopreserved tissue appeared to have recovered from damage caused by freezing, although the percentage of tissue with normal morphology declined after 24 and 48 h of culture. There was no significant difference between percentage normality in control tissue and tissue exposed to the cryoprotectant without freezing for any of the culture times studied. These data indicate that it is possible to freeze/thaw bovine ovarian tissue while retaining a reasonable yield of morphologically intact follicles and that a short period of post-thaw culture may enhance follicle recovery.  相似文献

目的探讨经体外实验筛选出的降胆固醇乳酸菌对肉鸡血清胆固醇及胸肌组织、腿肌组织和肝脏组织胆固醇含量的影响。方法将肉鸡随机分为2组,对照组喂食普通饲料,实验组喂食含有DM86056菌的饲料;喂食56d后分离其血清及肝脏等组织,并应用硫酸铁铵法测定其胆固醇含量,观察对照组与实验组结果差异是否有显著性。结果对肉鸡血清、胸肌组织、腿肌组织胆固醇含量测定后,对照组与实验组结果差异有显著性(P〈0.05),而对肝脏组织胆固醇含量测定后,对照组与实验组结果差异无显著性(P〉0.1)。结论菌株DM86056可明显降低肉鸡血清、胸肌组织、腿肌组织中胆固醇含量,而对肝脏组织胆固醇含量没有明显改变。  相似文献

The concentration of antibacterial agents in the interstitial tissue fluid has been studied in an experimental model using implanted perforated Silastic capsules (tissue cages). Tissue fluid concentrations were always lower than the initial peak concentration in the serum, but for those drugs that were more slowly excreted the tissue fluid was similar to the serum concentration after six hours. In contrast the concentration in the tissue fluid for those drugs that are excreted rapidly was unpredictable, being either negligible or maintained at concentrations significantly better than in the serum. There was no evidence of accumulation in tissue fluid with regular dosage. These results indicate that the tissues may never be exposed to an adequate concentration of some antibacterial agents. This may be of clinical importance in tissue infections. Antibiotics which are rapidly excreted should be given in large enough doses and often enough for adequate blood levels to be maintained, so allowing equilibrium with tissue fluid to be achieved.  相似文献

The aim of this study is to determine the relation between diabetes and vaginal candidiasis in terms of oxidative biomarker levels in a vaginal candidiasis model of the diabetic rats by evaluating malondialdehyde (MDA), sulphydrile groups or glutathione (RSH), and ascorbic acid (C vit) levels. All rats were randomly divided into five groups. All of the groups were observed for 21 days. In the treated diabetes groups, MDA (0.90, 0.68 nmol/ml and 3.78, 3.79 nmol/g tissue, plasma and vaginal tissue, respectively) and RSH (227, 171 nmol/100 ml 0.38, 0.37 μmol/g tissue, plasma and vaginal tissue, respectively) levels were found to be decreased while the levels of C vit were found to be increased (0.49, 0.37 μmol/l 2.39, 2.01 nmol/g tissue plasma, and vaginal tissue, respectively) (P < 0.05). In the groups of untreated diabetes, vaginal candidiasis were found to be more serious and oxidative biomarkers were found to be increased (MDA 1.30, 1.26 nmol/ml and 7.82, 2.37 nmol/g tissue and RSH 258, 145 nmol/100 ml and 0.31, 0.46 μmol/g tissue) while the antioxidant C vit levels were found to be decreased (0.24, 0.17 μmol/l 1.33, 2.66 nmol/g tissue) (P < 0.05). RSH, plasma MDA, blood glucose, and tissue MDA levels of vaginal candidiasis embedeled diabetic rats, were found to be higher than those in untreated diabetic and untreated vaginitis enbedeled rats ‹P < 0.05’. Vaginal candidiasis caused oxidative stress in diabetic rats working together. Systemic oxidative stress biomarkers were found to be affected from vaginal candidiasis although it was a local mucosal infection. 形体指标的检测是鱼类常规营养评定的重要组成部分。目前,分析鱼类组织体重比的方法会对鱼体造成损伤,且取样过程复杂,迫切需要无损快速检测技术分析鱼体组织占比。低场核磁共振技术具有快速无损检测的特点,已有研究利用低场核磁共振T1谱技术检测小鼠内脂肪组织体积,但尚未有研究应用低场核磁共振T1-T2谱技术对鱼类各组织器官进行定性和定量分析。基于此,利用低场核磁共振T1-T2谱技术,将分离后的罗非鱼的肌肉组织、腹腔脂肪组织、肝脏组织、肠道组织单独以及混合后进行扫描分析,结果发现利用低场核磁共振T1-T2谱技术可以分离罗非鱼肌肉组织和腹腔脂肪组织,但是无法区分肝脏组织和肠道组织。进一步对能够实现分离的肌肉组织和脂肪组织建立定量分析的模型,分析罗非鱼组织信号强度与组织重量相关性,结果显示,肌肉组织相关性R2=0.974 3,腹腔脂肪组织相关性R2=0965 0。并利用罗非鱼活体验证了肌肉组织定量分析模型的可靠性,将活体扫描肌肉信号大小转换成肌肉组织重量并分析其与全鱼体重相关性,结果显示肌肉组织重量与体重相关性R2=0.806 9。研究表明,在鱼类营养代谢研究中,可以利用低场核磁共振T1-T2谱技术快速无损地定量分析鱼体内肌肉组织含量。  相似文献

This paper presents an animal model [the kangaroo], a quantitative anatomical dissection procedure, and a mathematical model [two-phase linear regression] which illustrate that body tissues grow at varying rates relative to each other. An argument is developed that biochemists interested in tissue chemical activity need to be able to sample tissue of known [predicted] growth rate. It is assumed that the ability to select, say muscle tissue samples, from any one animal at a stage of its growth where the individual selected pieces of tissue have known [predicted] low, average and high growth rates would allow comparisons to be made between the sampled tissues that may elucidate the underlying biochemical mechanisms involved in the growth process. It is asserted that to establish standards for tissue samples used in biochemical growth studies, the growth rate of the sampled tissue should be one of the criteria incorporated into the definition of what is "standard" for a tissue sample.  相似文献

Following injury, tissue repair process takes place involving inflammation, granulation tissue formation and scar constitution. Granulation tissue develops from the connective tissue surrounding the damaged area and contains vessels, inflammatory cells, fibroblasts and myofibroblasts. Myofibroblasts play an important role in many tissue injuries and fibrocontractive diseases. The process of normal wound repair after tissue injury follows a closely regulated sequence including the activation and the proliferation of fibroblastic cells. In pathological situations, the normal resolution stages are abrogated and the proliferation of myofibroblasts continues, inducing excessive accumulation of extracellular matrix. The differentiation of fibroblastic cells into myofibroblasts is an early event in the development of tissue fibrosis. Myofibroblastic cells express smooth muscle cytoskeletal markers (alpha-smooth muscle actin in particular) and participate actively in the production of extracellular matrix. The evaluation of myofibroblast differentiation in renal biopsies would be useful for histopathologists to appreciate the intensity of tissue injury and particularly to predict the long term outcome of some nephropathies. Immunohistochemical studies for alpha-smooth muscle actin should be made systematically in renal tissue biopsies. Myofibroblastic differentiation appears to play a significant role in the progression of renal failure and seems to be a useful marker of progressive disease.  相似文献

Electrocautery and directed energy devices (DEDs) such as lasers, which are used in surgery, result in tissue damage that cannot be readily detected by traditional histological methods, such as hematoxylin and eosin staining. Alternative staining methods, including 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) to stain live tissue, have been reported. Despite providing superior detection of damaged tissue relative to the hematoxylin and eosin (H&E) method, the MTT method possesses a number of drawbacks, most notably that it must be carried out on live tissue samples. Herein, we report the development of a novel staining method, "antigen destruction immunohistochemistry" (ADI), which can be carried out on paraffin-embedded tissue. The ADI method takes advantage of epitope loss to define the area of tissue damage and provides many of the benefits of live tissue MTT staining without the drawbacks inherent to that method. In addition, the authors provide data to support the use of antibodies directed at a number of gene products for use in animal tissue for which there are no species-specific antibodies commercially available, as well as an example of a species-specific direct antibody. Data are provided that support the use of this method in many tissue models, as well as evidence that ADI is comparable to the live tissue MTT method.  相似文献

The effect of fixation technique and post mortem-to-fixation interval in immersion-fixed tissue from the central nervous system on immunocytochemical staining for the presence of an immunoglobulin was determined in mice. Immersion-fixed tissue was found to be inferior to perfusion-fixed tissue for immunocytochemical staining of this serum protein. Unlike what has been observed for other antigens, the quality of staining for IgG in immersion-fixed tissue decreased to unacceptable levels if the post mortem-to-fixation interval was increased to more than a few hours. This effect may be secondary to the rapid post-mortem disintegration of the blood-brain barrier and a resulting diffusion of serum proteins into surrounding tissue from the vasculature.  相似文献

Knowledge about mechanical tissue properties is required for functional modelling and simulating of tissue and organ responses to external mechanical stress. To get the right properties especially for functional modelling of organs, tissue properties have to be determined in vivo. There are only few described methods for characterization of internal organ's tissue mechanics that can be applied in vivo. We introduce and evaluate a method to determine mechanical tissue properties, especially those of lung tissue, endoscopically. Inverse finite element analysis (utilizing a Neo-Hookean model for hyperelastic materials) and image processing algorithms are used to determine the shear modulus of a soft tissue. The resulting values for shear moduli were normally distributed. The shear modulus of the artificial tissue sample was determined with a relative error of 0.47% compared to the value obtained by uniaxial tensile test.  相似文献

Tissue engineering aims to develop functionalized tissues for organ replacement or restoration. Biodegradable scaffolds have been used in tissue engineering to support cell growth and maintain mechanical and biological properties of tissue constructs. Ideally cells on these scaffolds adhere, proliferate, and deposit matrix at a rate that is consistent with scaffold degradation. However, the cellular rearrangement within these scaffolds often does not recapitulate the architecture of the native tissues. Directed assembly of tissue-like structures is an attractive alternative to scaffold-based approach for tissue engineering which potentially can build tissue constructs with biomimetic architecture and function. In directed assembly, shape-controlled microstructures are fabricated in which organized structures of different cell types can be used as tissue building blocks. To fabricate tissue building blocks, hydrogels are commonly used as biomaterials for cell encapsulation to mimic the matrix in vivo. The hydrogel-based tissue building blocks can be arranged in pre-defined architectures by various directed tissue assembly techniques. In this paper, recent advances in directed assembly-based tissue engineering are summarized as an emerging alternative to meet challenges associated with scaffold-based tissue engineering and future directions are addressed.  相似文献

A tissue bank is accountable before the community in fulfilling the expectations of tissue donors, their families and recipients. The expected output from the altruistic donation is that safe and high quality human tissue grafts will be provided for the medical treatment of patients. Thus, undertakings of tissue banks have to be not only authorised and audited by national competent health care authorities, but also comply with a strong ethical code, a code of practices and ethical principles. Ethical practice in the field of tissue banking requires the setting of principles, the identification of possible deviations and the establishment of mechanisms that will detect and hinder abuses that may occur during the procurement, processing and distribution of human tissues for transplantation. The opinions and suggestions manifested by the authors in this paper may not be necessarily a reflection of those within the institutions or community they are linked to.  相似文献

BACKGROUND: Characterization of tissues can be based on the topographical relationship between the cells. Such characterization should be insensitive to distortions intrinsic to the acquisition of biological preparation. In this paper, a method for the robust segmentation of tissues based on the spatial distribution of cells is proposed. MATERIALS AND METHODS: The neighborhood of each cell in the tissue is modeled by the distances to the surrounding cells. Comparison with an example or prototype neighborhood reveals topographical similarity between tissue and prototype. Processing of all cells in the tissue extracts the regions with tissue architecture similar to the given example. RESULTS: Comparison with other topographical-segmentation methods shows that the proposed method is better suited for partitioning tissue architecture. As an example, the quantification of the structural integrity in rat hippocampi after ischemia is demonstrated. In contrast to other methods, the algorithm correlates well with expert evaluation. CONCLUSIONS: The present method reduces the nonbiological variation in the analysis of tissue sections and thus improves confidence in the result. The method can be applied to any field where regular patterns have to be detected, as long as the directional distribution of neighbors may be neglected.  相似文献

Adipose tissue is a structure highly specialized in energy storage. The adipocyte is the parenchymal component of adipose tissue and is known to be mesoderm or neuroectoderm in origin; however, adipocyte development remains poorly understood. Here, we investigated the development of adipose tissue by analyzing postnatal epididymal adipose tissue (EAT) in mouse. EAT was found to be generated from non-adipose structure during the first 14 postnatal days. From postnatal day 1 (P1) to P4, EAT is composed of multipotent progenitor cells that lack adipogenic differentiation capacity in vitro, and can be regarded as being in the 'undetermined' state. However, the progenitor cells isolated from P4 EAT obtain their adipogenic differentiation capacity by physical interaction generated by cell-to-matrix and cell-to-cell contact both in vitro and in vivo. In addition, we show that impaired angiogenesis caused by either VEGFA blockade or macrophage depletion in postnatal mice interferes with adipose tissue development. We conclude that appropriate interaction between the cellular and matrix components along with proper angiogenesis are mandatory for the development of adipose tissue.  相似文献

Indirect immunofluorescence is used to detect and locate proteins of interest in a tissue. The protocol presented here describes a complete and simple method for the immune detection of proteins, the mouse lactating mammary gland being taken as an example. A protocol for the preparation of the tissue samples, especially concerning the dissection of mouse mammary gland, tissue fixation and frozen tissue sectioning, are detailed. A standard protocol to perform indirect immunofluorescence, including an optional antigen retrieval step, is also presented. The observation of the labeled tissue sections as well as image acquisition and post-treatments are also stated. This procedure gives a full overview, from the collection of animal tissue to the cellular localization of a protein. Although this general method can be applied to other tissue samples, it should be adapted to each tissue/primary antibody couple studied.  相似文献
