Variation in the inheritance of expression among subclones for an unselected (uidA) and a selected (bar) transgene was analyzed in two individual transformation events in maize. The unselectable gene (uidA) and the selectable gene (bar), on two separate plasmids, were transferred to maize (Hi-II derivative) by particle bombardment of embryogenic calli or suspension cells. A total of 188 fertile T1 plants were obtained from one transformant (transformation event BG which integrated uidA and bar). A total of 98 fertile T1 plants were obtained from a second transformant (transformation event B which integrated bar). Through self-pollination and/or cross-pollination in the greenhouse, approximately 10 000 T2 progeny were obtained from event BG, and more than 1000 T2 progeny were obtained from event B. Segregation of transgene expression was analyzed statistically in a total of 2350 T2 progeny from 40 T1 subclones of event BG and in 217 T2 progeny from six T1 subclones from event B. Variation in the inheritance of expression among subclones for the two transgenes (uidA and bar) was observed in the two transformants. A significant difference was observed between the use of the female or male as the transgenic parent in the inheritance of expression for the two transgenes in event BG. No inheritance through the pollen was observed in two of four T1 subclones analyzed in event B. Co-expression analysis of event BG showed that both transgenes were co-expressed in 67.7% of the T2 plants which expressed at least one of the two transgenes. Of the T2 expressing plants, 30.4% expressed only bar, and 1.9% expressed only uidA. Inactivation of the unselected (uidA) and the selected (bar) transgenes was observed in individual T2 plants.  相似文献

Summary A procedure suitable for the extraction and mapping of total proteins from the basidiomycete, Agrocybe aegerita, was developed. A. aegerita mycelia were fragmented either with a Dangoumeau grinder, an X-press bomb or a sonicator and the efficiency of these three disruption methods were compared. The extraction buffer composition was optimized to avoid proteolytic activities. 2D-SDS-PAGE analysis of protein extracts showed that the rate of reproducibility depending on extractions and electrophoretic separations was always greater than 96% for all strains. The differences in efficiency observed between the breaking procedures indicate that the A. aegerita cell wall is more mechanically resistant than that of other basidiomycetes. The efficient action of protease inhibitors (PMSF and SDS) showed that A. aegerita mycelia contains numerous and/or highly active proteases. Reproducibility of protein extraction and separation methods allowed the establishment and the comparison of standard maps. Qualitative and quantitative variations in gene products between a wild dikaryotic strain and 11 homokaryotic strains from its progeny were examined. The genetic diversity, determined by comparing the distribution of proteic variations in 11 homokaryons from the same progeny, was comparable to that observed between co-isogenic homokaryons of another basidiomycete.  相似文献

In Calliphora vicina from Belomorsk (North Karelia) the pattern of reactivation of diapausing larvae at different temperatures (4, 12·5, 15·5, 20, and 25°C) was studied experimentally. Most favourable in this respect are the first and last temperatures. Photoperiodic reactivation was established. The rate of pupation under a long-day régime is two to four times greater when compared with that observed under a short-day régime. In C. vicina maternally operating photoperiods determined not only the ratio of diapausing and non-diapausing larvae in the progeny but also the peculiarities of the diapause. A more intensive diapause is typical for the progeny of short-day-treated flies. The diapause of the progeny of long-day-treated flies is less intense and can be easily terminated as a result of the action of the above-mentioned factors.  相似文献

Summary Previous experiments have revealed that the maize transposable element Activator (Ac) may become active during tissue culture. The objective of the present study was to determine whether a second transposable element, Suppressor-mutator (Spm), could also be activated in tissue culture and detected in regenerated maize plants. Approximately 500 R1 progeny of 143 regenerated plants (derived from 49 embryo cell lines) were crossed as males onto an Spm-responsive tester stock. Spm activity was observed in two R1 progeny of a single regenerated plant. This plant had been regenerated from Type II (friable embryogenic) callus of an A188 × B73 genetic background after 8 months in culture; the absence of Spm activity in four other plants regenerated from this same callus demonstrates that Spm activity was not present before culturing. Approximately 20 Spm-homologous DNA sequences were detected in each of the inbreds used to initiate the tissue cultures; it is presumed that one of these became active to give rise to Spm activity.  相似文献

Triple-testcross experiments (Kearsey and Jinks 1968) were employed to investigate the mode of gene action affecting pupa weight in Tribolium castaneum. Their experimental design involves two inbred lines, the F1 progeny and a segregating population derived from the cross of the inbred lines. In the present experiments, four segregating populations were used. These populations included the F2 generation, a select line (SEL) and two relaxed select lines (RSI and RSII). In addition, all possible reciprocal crosses were made among the RSI, RSII, and SEL populations. It was observed that: (1) additive, dominant and epistatic gene effects all made significant contributions to the pupa weight of the progeny from all four segregating populations; (2) there was no evidence of either accumulation of epistasis as a result of selection in the SEL population or decline in epistasis as a result of removing selection pressure from the RSI and RSII populations; and (3) significant negative heterosis and maternal effects contributed to the pupa weight of the crossbred progeny of the RSI, RSII and SEL populations.  相似文献

Morphology and meiosis are described in four progeny plants resulting from tetraploid Grindelia camporum Greene (2n = 24) from California pollinated by diploid G. grandiflora Hook. (2n = 12) from Coahuila, Mexico. Three of the four progeny were tetraploid, morphologically like the pistillate parent, and had metaphase I chromosome configurations which included quadrivalents and a complementary number of bivalents. They are considered to have resulted from selfing. The fourth plant was triploid (2n = 18) andmorphologically intermediate between the parents. Chromosome configurations in the triploid were variable with univalents, ring and rod bivalents, trivalents and pentavalents. These two species are considered related through an ancestor with a basic genome, but are separated cytologically by polyploidy and by two distinct chromosomal interchanges that explain the configurations observed in the triploid hybrid.  相似文献

Plant species from unproductive or adverse habitats are often characterized by a low potential relative growth rate (RGR). Although it is generally assumed that this is the result of selection for specific trait combinations that are associated with a low rate of net biomass accumulation, few studies have directly investigated the selective (dis-)advantage of specific growth parameters under a set of different environmental conditions. Aim of the present study was to quantify the impact of inherent differences in growth parameters among phenotypes of a single plant species, Lychnis flos-cuculi, on their performance under different soil nutrient conditions. Growth analysis revealed significant variation in RGR among progeny families from a diallel cross between eight genotypes originating from a single population. Differences in RGR were due to variation in both leaf area ratio (LAR) and in net assimilation rate (NAR). A genetic trade-off was observed between these two components of growth, i.e. progeny families with high investment in leaf area had a lower rate of net biomass accumulation per unit leaf area. The degree of plasticity in RGR to nutrient conditions did not differ among progeny families. Inherent differences in growth parameters among progeny families had a significant impact on their yield in competition with Anthoxanthum odoratum and Taraxacum hollandicum. In nutrient-rich conditions, progeny families with an inherently high leaf weight ratio (LWR) achieved higher yield in competition, but variation in this trait could not explain differences in competitive yield under nutrient-poor conditions. Inherent differences in growth parameters among progeny families were poorly correlated with differences in survival and average rosette biomass (a good predictor of fecundity) among these progeny families sown in four field sites along a natural gradient of soil fertility. In the more productive sites none of the growth parameters was significantly correlated with rosette biomass, but in the least productive site progeny families with an inherently high specific leaf area (SLA) tended to produce smaller rosettes than low-SLA families. These results are consistent with the view that a selective advantage may accrue from either high or low values of individual RGR components, depending on habitat conditions, and that the selective advantage of low trait values in nutrient-poor environments may results in indirect selection for low RGR in these habitats.  相似文献

Natural populations of Thymelaea hirsuta have previously been shown to comprise four distinct sexual morphs: males, females, protogynous individuals, i.e., first female then male, and protandrous individuals, i.e., first male then female. The objective of the present study has been to confirm the genetic basis of this sexual tetramorphism by quantifying morph ratios in the open-pollinated progeny of the four sexual phenotypes growing in a natural population. All four phenotypes were recovered in the progeny of each morph. All observed plants displayed a single sexual phenotype, thus confirming the genetic basis of the tetramorphism. The progeny sex ratios indicate that the genetic determination of sex in this species may be influenced by cytoplasmic factors, while the observed levels of functional female fertility suggest a near-dioecious system. The evolutionary significance of this tetramorphism as a transitional stage in the evolution of dioecy is discussed.  相似文献

A laboratory experiment was conducted to determine the effect of the pupal age of Calliphora erythrocephala (Meigen) on the reproductive biology (in terms of number, size, developmental time and longevity of progeny) of the parasitoid Melittobia acasta Walker. Melittobia acasta females of uniform size were given five C. erythrocephala pupae from one of four experimental age groups: 17–24 h, 24–48 h, 48–72 h and 72–96 h, for parasitization. The mean number of progeny produced from the experimental age groups for a 24 h period were 2, 7.6, 15.6 and 13.6, respectively. The parasitoids preferred hosts that were 48–72 h old. There were no significant differences in the mean development time (18.2 days) and size of progeny (mean head width = 0.38 ± 0.01 mm) produced from the experimental host age groups. The longevity of progeny from the four host age groups varied (range: 4–39 days), with those from the 48–72 h group living longest (mean = 25 days). The F1 females from the 48–72 h group were reproductively more successful than those from the other groups, producing a mean F2 progeny of 912 individuals when compared with 867, 801 and 757 individuals from the 24–48 h, 72–96 h and 17–24 h age groups, respectively. These findings make significant contributions to our knowledge of the breeding and utilization of this parasitoid for the biological control of dipteran flies in pigsties and poultry houses.  相似文献

The effect of Ac copy number on the frequency and timing of germinal transposition in tobacco was investigated using the streptomycin phosphotransferase gene (SPT) as an excision marker. The activity of one and two copies of the element was compared by selecting heterozygous and homozygous progeny of transformants carrying single SPT::Ac inserts. It was observed that increasing gene copy not only increases the transposition frequency, but also occasionally alters the timing of transposition such that earlier events are obtained. The result is that some homozygous plants generate multiple streptomycin resistant progeny carrying the same transposed Ac (trAc) element. We have also investigated the effect of modification of the sequence in the region around 82 bp downstream of the polyadenylation site and 177 bp from the 3 end of the element on germinal excision frequencies. Alteration of three bases to create a BglII site at this location caused a minor decrease in germinal excision events, but insertion of four bases to create a Cla I site caused a 10-fold decrease in the transposition activity of the Ac element.  相似文献

The variability of pharyngeal dentition in a natural population of B. intermedius and effects of genetic, hormonal and environmental factors on the number of tooth rows in the pharyngeal dentition in offspring from wild‐caught parents have been investigated. It was revealed that: (i) about 10% of fish from natural population have four‐rowed dentition instead of three‐rowed dentition characteristic for this species; (ii) the presence of the additional tooth row is not an abnormality of tooth replacement since it occurs symmetrically on both sides; (iii) occurrence of the fourth row of teeth is heritable since laboratory‐reared offspring from parents with four‐rowed dentition have the same dentition. Even if one of the parents had four‐rowed dentition the percentage of four‐rowed individuals in the progeny was significantly higher than in progeny from parents with normal (three rowed) dentition; (iv) the number of tooth rows appears to be hormonally controlled: high levels of thyroid hormone result in a decrease in the number of tooth rows to two. In contrast, deficiency of this hormone results in an increase to four rows; (v) no differences in the number of tooth rows were found in fish reared under 17°C, 24°C, 30°C and room temperature (20–26°C).  相似文献

The conventional way to drive modifications in old forest tree seed orchards is to establish progeny trials involving each parent tree and then evaluate its contribution to the performance of the progeny by estimating its general and specific combining ability (GCA and SCA). In this work, we successfully applied an alternative parent selection tactic based on paternity testing of superior offspring derived from a hybrid seed orchard established with a single Eucalyptus grandis seed parents and six E. urophylla pollen parents. A battery of 14 microsatellite markers was used to carry out parentage tests of 256 progeny individuals including two independent samples of selected trees and one control unselected sample, all derived from 6-year-old forest stands in eastern Brazil. Paternity determination was carried out for all progeny individuals by a sequential paternity exclusion procedure. Exclusion was declared only when the obligatory paternal allele in the progeny tree was not present in the alleged parent tree for at least four independent markers to avoid false exclusions due to mutation or null alleles. After maternity checks to identify seed mixtures and selfed individuals, the paternity tests revealed that approximately 29% of the offspring was sired by pollen parents outside the orchard. No selfed progeny were found in the selected samples. Three pollen parents were found to have sired essentially all of the offspring in the samples of selected and non-selected progeny individuals. One of these three parents sired significantly more selected progeny than unselected ones (P0.0002 in a Fisher exact test). Based on these results, low-reproductive-successful parents were culled from the orchard, and management procedures were adopted to minimize external pollen contamination. A significant difference (P<0.01) in mean annual increment was observed between forest stands produced with seed from the orchard before and after selection of parents and revitalization of the orchard. An average realized gain of 24.3% in volume growth was obtained from the selection of parents as measured in forest stands at age 2–4 years. The marker-assisted tree-breeding tactic presented herein efficiently identified top parents in a seed orchard and resulted in an improved seed variety. It should be applicable for rapidly improving the output quality of seed orchards, especially when an emergency demand for improved seed is faced by the breeder.Communicated by D.B. Neale  相似文献

The polymerase chain reaction with arbitrary primers (RAPD–PCR) was used to study and to evaluate the genetic variation in the hybrid progeny of two Cyprinidae species, common bream Abramis brama L. and roach Rutilus rutilusL. Genetic polymorphism was studied in 20 fishes (young of the current year) obtained in four individual crosses: R. rutilus × R. rutilus (RR), A. brama × A. brama (AA), R. rutilus × A. brama (RA), and A. brama × R. rutilus (AR). Amplification spectra obtained with eight primers contained 288 fragments, 97.6% of which proved to be polymorphic. The proportion of polymorphic fragments was 75.0% in the RR progeny, 58.1% in the AA progeny, 84.9% in the AR progeny, and 77.8% in the RA progeny. Classification analysis in the space of principal components was performed with the first four components, which together accounted for 64% of the total variance of the character under study. The individual contributions of components I, II, III, and IV were 26.8, 16.8, 11.5, and 8.9%, respectively. Fishes of the two pure species and the hybrid progeny (direct and reverse hybrids together) were clearly differentiated in the space of principal components I and II. The best differentiation of the four samples (RR, AA, RA, and AR) was observed in the space of principal components II and IV. Possible causes of high genetic variation in interspecific hybrids are discussed.  相似文献

Bulk seedlots of two unpedigreed multiprovenance seed production areas (SPAs) each of Eucalyptus camaldulensis and Eucalyptus tereticornis and one pedigreed seedling seed orchard (SSO) of E. tereticornis were planted in genetic gain trials at three southern Indian trial sites. At the time of seed collection, fewer than 30% trees flowered in these orchards, except in one E. camaldulensis SPA where 73% of the trees flowered, which had an estimated outcrossing rate of 86%. The E. tereticornis SSO was dominated by pollen from five highly fecund families of the Indian Mysore gum land race, which contributed 59% of the fruits produced. The SPA and SSO seedlots were compared with a bulked natural-provenance seedlot of E. camaldulensis (Morehead, Laura, and Kennedy Rivers, Queensland), another natural-provenance seedlot (Petford, Queensland), commercial eucalypt clones at two sites, and a Mysore gum seedlot at one site. At 3 years, progeny from all the four SPAs displayed good survival (79–93%) and performance similar to that of the natural provenances and the commercial clones. Progeny from the E. tereticornis SSO had significantly lower growth (at two sites) and lower survival at all three test sites. The Mysore gum seedlot displayed high fecundity and lower growth but better survival than the SSO progeny. Seed orchard genetic composition and flowering contributions thus affected progeny performance and the extent to which orchard genetic diversity was captured in the progeny. SPA progeny displayed greater fecundity than the natural provenances, indicating a response to selection for fertility.  相似文献

The evolution of selfing taxa from outcrossing ancestors has occurred repeatedly and is the subject of many theoretical models, yet few empirical studies have examined the immediate consequences of inbreeding in a population with variable expression of self-incompatibility. Because self-incompatibility breaks down with floral age in Campanula rapunculoides, we were able to mate outbred and selfed maternal plants in a crossing design which produced progeny with inbreeding coefficients of 0, 0.25, 0.50 and 0.75. Cumulative inbreeding depression in plants that were selfed for one generation was very high in families derived from strongly self-incompatible plants (average δ = 0.98), and somewhat lower in families derived from plants with weaker expression of self-incompatibility (average δ = 0.90). Relative to outbred progeny, inbred progeny produced fewer seeds, had lower rates of germination, less vegetative growth and fewer flowers per plant. Inbred progeny also took longer to germinate, and longer to produce a first leaf and to flower. Interestingly, inbred plants also produced 40% fewer seeds than outcrossed plants (t-test P < 0.001) even when mated to the same, unrelated pollen donor, suggesting that inbreeding can produce profound maternal effects. Most importantly, our results demonstrate that progeny derived from plants with stronger expression of self-incompatibility exhibited greater levels of inbreeding depression than progeny from plants with weaker expression of self-incompatibility. Moreover, the decline in fitness (cumulative, ln-transformed) over the four inbreeding levels was steeper for the progeny of the strongly self-incompatible lineages. These empirical results suggest that inbreeding depression and mating system phenotype have the potential to coevolve.  相似文献

We evaluated the degree of selfing and inbreeding depression at the seed and seedling stages of a threatened tropical canopy tree, Neobalanocarpus heimii, using microsatellite markers. Selection resulted in an overall decrease in the level of surviving selfed progeny from seeds to established seedlings, indicating inbreeding depression during seedling establishment. Mean seed mass of selfed progeny was lower than that of outcrossed progeny. Since the smaller seeds suffered a fitness disadvantage at germination in N. heimii, the reduced seed mass of selfed progeny would be one of the determinants of the observed inbreeding depression during seedling establishment. High selfing rates in some mother trees could be attributed to low local densities of reproductive individuals, thus maintenance of a sufficiently high density of mature N. heimii should facilitate regeneration and conservation of the species.  相似文献

The four loci Gabra3, DXPas8, CamL1, and Bpa, located near the murine X-linked visual pigment gene (Rsup), have been ordered using 248 backcross progeny from an interspecific mating of (B6CBA-Aw-J/A-Bpa) and Mus spretus. One hundred twenty backcross progeny have been analyzed at seven anchor loci spanning the X chromosome and form a regional mapping panel. An additional 128 progeny have been screened for recombination events between Cf-9 and Dmd. Eighteen recombinants between these loci have been detected in the 248 animals; all of the recombinants were screened at the other anchor loci to identify any double crossovers. Pedigree analysis using these recombinants strongly favors a gene order of (Cf-9)-Gabra3-(DXPas8, Bpa)-CamL1-(Rsvp, P3, Cf-8)-Dmd for the loci studied. Synteny with human Xq27–Xq28 is retained, although the relative order of some loci may differ between the two species.  相似文献

Rare and endangeredplants of Silene (Caryophyllaceae) occuron four continents, including North America. In Oregon, inbred and outbred progeny weremonitored for three years after experimentalreintroduction of a narrow endemic, S.douglasii var. oraria, into formerlygrazed habitat within its presumed historicalrange. Survival and reproduction were comparedfor progeny that were derived from the seeds ofself- versus cross-pollinated flowersproduced in situ at Cascade Head, aUNESCO Biosphere Reserve where the largest ofthree extant populations occurs. Progeny ofcross-pollinated flowers had significantlygreater field survival in all years than didoffspring of selfed or open-pollinated flowers(P < 0.01). Outbred progeny alsosignificantly exceeded other treatment cohortsin canopy area, and produced more reproductivestems and flowers than other progeny types ofthe same maternity. For plots varying in plantdensity, mortality was greatest in thehigh-density competitive regime but thesurvivors reached significantly larger sizesand reproductive capacities than in low densityplots (P < 0.05). In all, successfulconservation plans involving reintroduction mayrequire genetically diverse progeny to offsetinbreeding depression as well as suitableplanting densities and source populations.  相似文献

I used a discontinuous population ofScleranthus annuus (Caryophyllaceae) to study the effect of crossing distances on flower morphology of the progeny. Four types of progeny were produced by artificial selfing, crossing with pollen-donors from the same patch in the population, crossing with pollen from donors from other patches in the population and inter-population crosses. The size of gynoecium parts and 12 sepal characters in this petal-lacking species were significantly influenced by the type of cross and the patch in the population from where the seed-parents originated. All comparisons of progeny types except selfed vs progeny produced by within-patch crosses were significantly separated from each other, while all four seed-parent patches used were significantly separated in a multidimensional space.  相似文献

We analyzed the inheritance of mitochondrial DNA (mtDNA) species in matings of the grass symbiont Epichloe typhina. Eighty progeny were analyzed from a cross in which the maternal (stromal) parent possessed three linear plasmids, designated Callan-a (7.5 kb), Aubonne-a (2.1 kb) and Bergell (2.0 kb), and the paternal parent had one plasmid, Aubonne-b (2.1 kb). Maternal transmission of all plasmids was observed in 76 progeny; two progeny possessed Bergell and Callan-a, but had the maternal Aubonne-a replaced with the related paternal plasmid Aubonne-b; two progeny lacked Callan-a, but had the other two maternal plasmids. A total of 34 progeny were analyzed from four other matings, including a reciprocal pair, and in each progeny the plasmid transmission was maternal. The inheritance of mitochondrial genomes in all progeny was analyzed by profiles of restriction endonuclease-cleaved mtDNA. In most progeny the profiles closely resembled those of the maternal parents, but some progeny had nonparental mtDNA profiles that suggested recombination of mitochondrial genomes. These results indicate that the fertilized stroma of E. typhina is initially heteroplasmic, permitting parental mitochondria to fuse and their genomes to recombine.  相似文献
