Summary The paper deals with some cultural and physiological studies of a strain ofPhytophthora palmivora Butler, causing fruit rot ofAchras sapota L. Among the various culture media studied, oat meal, corn meal, lima bean and rice meal agars were the best ones, with abundant growth and sporulation of the organism. Lactose, galactose, dextrin and glycogen were the best sources of carbon. Among the various nitrogenous compounds studied, ammonium phosphate (monobasic), ammonium sulphate, ammonium tartrate, ammonium lactate, asparagin, glycine and glutamic acid were the best ones utilized by the organism. A pH of 5.9 was found to be the optimum for the growth and sporulation.Part of Senior Author's M.Sc. The effect of different carbon and nitrogen compounds on growth and sporulation ofC. microcephala (Wallr.)Tul. causing ergot disease of Bajra has been studied. Nine different sources of carbon were used but cane sugar was found to be the best source for both, growth and sportulation of the fungus. Glucose, sucrose and maltose gave good growth but fair sporulation. Lactose and sorbitaol proved to be the poor sources. However, fungus failed to utilize starch, dextrin and mannitol.Nineteen nitrogen compounds were tried for the growth and sporulation of the fungus. Best growth and sporulation were supported by peptone and glycine. L-asparagine, DL-valine, Urea, magnesium nitrate and L-proline supported good growth and fair sporulation except DL-valine where it was excellent. Poor growth was obtained on L-isoleucin, ammonium sulphate, potassium nitrate,-alanine, ammonium chloride, DL-aspartic acid and DL-methionine. Fungus failed to utilize thio-urea.  相似文献

Summary Nitrogen and sulphur requirements ofColletotrichum inamdarii Lal isolated from the leaves ofCarissa carandas L. have been studied. DL-serine, L-asparagine and L-phenylalanine have been found to be of good nitrogen source followed by potassium nitrate, calcium nitrate, magnesium nitrate, DL-alanine, ammonium nitrate, glutamic acid, ammonium sulphate, DL-valine, aspartic acid, ammonium chloride, ammonium hydrogencarbonate, L-histidine and potassium nitrite. There was no growth in the absence of nitrogen.Sporulation was excellent on calcium nitrate and sodium nitrate, Very good on DL-serine, potassium nitrate, and magnesium nitrate. Good on L-asparagine, L-phenylalanine and ammonium oxalate. Fair on DL-alanine, DL-leucine, ammonium sulphate, DL-valine, ammonium chloride and L-histidine whereas poor on glutamic acid, aspartic acid, ammonium tartarate and ammonium nitrate. Few spores were observed on ammonium hydrogencarbonate but potassium nitrite did not show any sporulation.Amongst the sulphur compounds sodium bisulphate gave the best growth and good sporulation, followed by sodium thiosulphate, magnesium sulphate, ammonium sulphate and potassium sulphate. Thiourea gave negligible growth whereas it failed to grow on zinc sulphate and potassium persulphate.  相似文献

Three isolates ofA. tenuis isolated from the diseased leaves ofMangifera indica l. Musa paradisiaca l. andPsidium guajava l. were investigated. They were grown on different sources of nitrogen viz., potassium nitrate, sodium nitrate, calcium nitrate, ammonium nitrate, sodium nitrite, ammonium sulphate, ammonium chloride, glycine, DL-valine, L-glutamic acid, urea, thiourea, L-asparagine and peptone. They were also grown on the medium lacking nitrogen. A wide variation was observed in the growth and reproduction of the different isolates. The growth of all of them was good on potassium nitrate, calcium nitrate, glycine, DL-valine, L-glutamic acid, L-asparagine and peptone but the sporulation was satisfactory on calcium nitrate only. Sodium nitrite supported moderate growth of banana leaf isolate whereas there was no growth of the other two isolates. None of the organisms could grow on the medium lacking nitrogen as well as on thiourea. The results obtained with the isolates under study have been compared with those of earlier investigators and it has been clearly established that the different isolates ofA. tenuis could show marked differences in their nitrogen requirements.  相似文献

Nine inorganic nitrogenous compounds and sixteen amino acids were used to study the growth and sporulation of highly and weakly virulent isolates ofColletotrichum falcatum Went. Potassium nitrate, sodium nitrate and calcium nitrate supported good growth, while ammonium phosphate, potassium nitrate and sodium nitrate supported good sporulation of both isolates. Among the amino acids tested, glycine and DL-alanine were best for growth, and DL-threonine, L-leucine, DL-tyrosine, DL-serine, DL-phenylalanine and L-cystine for sporulation. No correlation existed between virulence and growth but the highly virulent isolate sporulated significantly more than the weakly virulent isolate.This work is a part of the author's Ph. D. The effect of different sulphur and phosphorus compounds on the growth and sporulation ofCurvularia pallescens Boed. has been studied. Nine different sulphur sources were tried but among them only magnesium sulphate yielded the best dry weight of the fungus. Zinc sulphate, sodium sulphate, sodium thiosulphate, potassium sulphate and calcium sulphate supported good growth. Poor growth was recorded on sodium bisulphite, ammonium sulphate, sodium sulphide and control. Sporulation was excellent on magnesium sulphate. It was good on zinc sulphate, sodium sulphate and potassium sulphate. On sodium thiosulphate, calcium sulphate, sodium bisulphite and control it was fair. Sodium sulphide and ammonium sulphate had inhibitory effect as sporulation was poor and nil on these two compounds respectively.Six phosphorus compounds were studied. Tripotassium phosphate gave best growth and excellent sporulation. Good growth and excellent sporulation was recorded on monobasic potassium phosphate and magnesium phosphate. Growth and sporulation were good on dibasic potassium phosphate and sodium dihydrogen phosphate. Ammonium phosphate was poorly utilized.  相似文献

Summary The paper deals with some physiological studies of two typical strains ofPhytophthora palmivora Butl. andP. parasitica Dast. var.macrospora Ashby causing severe fruit rots ofAchras sapota L. andAnona squamosa L. respectively. The effect of temperature on the germination of sporangia had a marked effect. Maximum percentage of indirect germination (by formation of zoospores) occurred at 20° C, whereas direct germination of sporangia (by formation of germ tube) was at 30° C, which further continued upto 37° C. Maximum indirect germination at 20° C shows the favourable cool temperature for the epidemic of the diseases (Fruit rots) in nature. The best substrate (medium) for germination of sporangia was found to be tap water. Next to this was the host decoction or extract. Glucose solution also accelerated sporangial germination. The effect of two dyes viz., Malachite green and crystal violet was also studied in relation to growth and sporulation ofPhytophthora isolates. Their addition to the medium in various fractional dilutions had a profound influence in the rate of growth and sporulation. An interesting observation noted was that growth of the isolates was inversely porportional to the various concentrations of the dyes, under study. An attempt was also made to study the influence of various vitamins. In all, six vitamins were included in the study. Out of these, thiamine and riboflavin were found to be the best sources promoting good growth and sporulation of both the species ofPhytophthora under study.Forms a part of Senior author's M. Sc. Aspergillus tamarii Kita grew and sporulated best at 30°C. The best pH for growth and sporulation were 5.5 and 6.5, respectively. Among the carbon sources employed, glucose supported the highest growth and sporulation. Best growth was obtained with sodium nitrate as nitrogen source and best sporulation with ammonium tar-tarate. When glucose was used as carbon source, the carbohydrates found in the mycelium included myoinositol. dulcitol, fructose, arabinose and ribose.  相似文献

Mycelial growth of an isolate ofT. bakamatsutake was tested in media with C/N ratio ranging from 0 to 50 and with 32 carbon and 12 nitrogen sources. The isolate grew best at the C/N ratio of 30. It utilized the monosaccharidesd-glucose,d-mannose, andd-fructose, the disaccharide trehalose, and polysaccharide pectin among the carbon sources; and yeast extract,l-glutamic acid, and ammonium compounds among the nitrogen sources. The growth of ten isolates and secretion of gluconic and oxalic acids were compared ind-glucose, trehalose, and pectin media. The utilization ofd-glucose, trehalose, and pectin differed among the ten isolates, but all the isolates secreted gluconic acid in thed-glucose media and oxalic acid in the pectin media.  相似文献

Summary Colletotrichum gloeosporioides isolated from the diseased leaves ofPolyscias balfuriana could grow and sporulate on a wide range of pH (viz. from 3.0 to 9.0). Maximum growth was recorded at pH 5.5. Mannitol was the best carbon source for growth. Good growth as well as good or excellent sporulation was also recorded on glucose, fructose, maltose and starch. Organic acids (malic and tartaric) supported poor growth.Present organism could utilize a number of nitrogen sources. Nitrates in general were comparatively better sources than ammonium compounds. Aspartic acid was found to be the best nitrogen source for growth. Nitrites were toxic at lower pH values though they supported growth at alkaline medium. Best growth of the organism was obtained on MgSO4, 7H2O. The urea supported poor growth. ZnSO4 inhibited the growth completely. The present organism was incapable of growing in media lacking carbon, nitrogen or sulphur.  相似文献

A strain ofBacillus cereus var.mycoides isolated from Burdwan soil producesl-glutamate in the medium. The strain is able to grow and produce in a synthetic medium but supplementation with casamino acid or yeast extract improves the yield. Maintenance of pH of the fermentation medium near neutrality prolongs the active growth period and improves the yield. Glucose and ammonium nitrate were found to be most suitable carbon and nitrogen sources, respectively. Cane sugar molasses (as a substitute for glucose) significantly stimulated the growth but glutamate production was less. Various B vitamins stimulate the growth and glutamate yield. The yield of glutamate under optimal condition is 5.2 g/l.  相似文献

Effect of fourteen different nitrogen sources on five species ofSphaeropsis i. e.S. cactinae, S. cycadis, S. jodhpurensis, S. muehlenbeckiae andS. punicae, obtained respectively from the infected parts ofCitrus medica, Cycas circinalis, Cordia gharaf, Muehlenbeckia platycladoes andPunica granatum was studied, under cultural conditions on Asthana and Hawker's Medium A by substituting its nitrogen with equal quantity of an other nitrogen source. All the present species reproduced well on potassium nitrate, sodium nitrate, ammonium sulphate and ammonium nitrate. None of the present species could grow on sodium nitrite. Out of all amino acids, no one was found to be excellent for the growth and sporulation of the present fungi. Most of the amino acids supported poor growth and sporulation of three of the five species ofSphaeropsis, S. cactinae, S. cycadis andS. punicae on L-asparagine, Glycine, L-cystine and L-alanine. Maximum and minimum pH for growth and sporulation ranged from 6.5 to 7.8 and 5.3 to 4.2 respectively. Decrease in adjusted pH was also observed for all the species on L-asparagine and DL-valine.  相似文献

An Arthrobacter sp. (strain 9006), isolated from lake water, accumulated nitrite up to about 15 mg N/l, but no nitrate. In a mineral medium supplemented with tryptone, yeast extract, acetate and ammonium, the cells released nitrite into the medium parallel to growth or when growth had virtually ceased. The nitrite formed was proportional to the initial acetate concentration, indicating an involvement of acetate metabolism with nitrification. The organism grew with a wide variety of organic carbon sources, but washed cells formed nitrite from ammonium only in the presence of citrate, malate, acetate or ethanol. Magnesium ions were required for nitrification of ammonium and could not be replaced by other divalent metal ions. Analysis of the glyoxylate cycle key enzymes in washed suspensions incubated in a minimal medium revealed that isocitrate lyase and malate synthase were most active during the nitrification phase. Nitrite accumulation but not growth was inhibited by glucose, tryptone and yeast extract. The influence of various inhibitors of hyphal growth, sporulation and biosynthesis of aflatoxin B1 in Aspergillus parasiticus NRRL 2999 was studied. 6-Thioguanine, dl-ethionine, fluoroacetic acid and phenylboric acid, inhibitors of maturation of fungal conidiophores and of conidiogenesis, were added at various concentrations to malt extract agar. Lower concentrations of 6-thioguanine and dl-ethionine did not inhibit the growth of hyphae and the sporulation. Phenylboric acid reduced conidiogenesis more than hyphal growth. The yields of aflatoxin B1 were significantly reduced. Additions of fluoroacetic acid did not greatly affect the growth of hyphae but totally inhibited the production of conidia and concurrently significantly reduced the formation of aflatoxin B1. An interrelation between conidiogenesis and onset of secondary metabolism in A. parasiticus is evident.  相似文献

Summary The paper deals with some physiological studies viz., (1) Production of enzymes (2) Effect of chemicals and sugar solutions and (3) Utilization of amino acids, by two species ofPhytophthora i.e.,Phytophthora palmivora Butler andP. parasitica Dast. var.macrospora Ashby., isolated from rotten fruits ofAchras sapota andAnona squamosa respectively.In the study of production of extra-cellular enzymes, it was found that both the isolates ofPhytophthora produced enzymes like inulase, amidase, emulsin and diastase in small quantity.Among the various chemicals and salt solutions used to study their effects in the formation of sporangia; it was observed that these were remarkably produced in abundance by both the species ofPhytophthora specially in the chemical solutions like sodium chloride, potassium nitrate, sodium thiosulphate and potassium permanganate. On the other hand, chemical solutions like strontium sulphate, potassium chromate, ammonium oxalate, zinc sulphate, copper sulphate, potassium sulphate, strontium nitrate and ferrous sulphate completely ceased the sporangial development. Among the various sugar solutions tried, dextrose, galactose, glucose, laevulose and fructose accelerated sporangial formation.An attempt was also made to study the effect of various amino acids singly or in combination on growth and sporulation. Both the isolates under study, utilized L-Arganine monohydrochloride and DL-Aspartic acid, hence these were growth promoters. It is interesting to note that the growth of the isolates was good, only when DL-Norleucine and DL-Methionine (both growth inhibitors) were provided in combination.Forms a part of Senior author's M.Sc. Summary The physiological conditions governing growth and sporulation ofSaksenaea vasiformis Saksena, a fungus with outstanding morphological features quite peculiar for Mucorales, were determined. Earlier studies made byTiwari (1955) on a strain of the same species had shown that this fungus is incapable of sporulating on any synthetic medium normally employed for growing fungi.The fungus had been found to have a high tolerance for very low and high pH values. It showed maximum growth at two pH values, one near neutral point, at pH 6, and another at high alkaline pH value, i.e., pH 11. Reason for this behaviour of this fungus has already been discussed. The most suitable temperature for the growth of the fungus was found to be between 30–35° C.Nearly all the carbon, nitrogen and sulphur sources which generally favour growth of fungi were found to support vegetative growth of this fungus as well. However, sporulation in this fungus had peculiar nutritional needs. Only some of the carbon sources, viz., arabinose, rhamnose, sorbose, galactose, lactose and citric acid which supported poor growth, were found to support good or excellent sporulation. But it may be stated that not all carbon sources supporting poor growth could induce sporulation of the organism. Also none of the nitrogen or sulphur sources could induce the fungus to sporulate in presence of glucose as carbon source.  相似文献

Summary Four isolates ofCytospora cincta Fr. and 2 ofC. leucostoma Fr. were cultured on media containing 7 different nitrogenous compounds. Maltose served as a constant source of carbon. All experiments were carried out at 25° C.Total growth as determined by mycelial weights, and degree of sporulation as determined by an arbitrary system, revealed that 1) potassium nitrate was most satisfactory as a source of nitrogen, 2) response of the isolates tended to segregate them along species lines, and 3) the pH of the medium appeared to be a factor in the degree of response.Growth-habit experiments emphasized 1) extreme variation in colony characteristics and 2) the need for standardization of laboratory environments for comparative studies of the fungi.Approved by the Director of the Idaho Agricultural Experiment Station as Research Paper No. 518.  相似文献

Phaeodactylum tricornutum Bohlin grew well withL-methionine-DL-sulfoximine (MSX) as sole nitrogen source. Such growth helps to explain the lack of effect of MSX on ammonium assimilation by this organism. Methylammonium inhibited growth with nitrate or MSX as sole nitrogen source but not growth on ammonium. Methylammonium could not be metabolised byP. tricornutum but was accumulated in the cells, the concentration factor sometimes approaching 25,000. Ammonium addition, but not that of MSX or nitrate, displaced methylammonium from the cells and this displacement was followed by resumption of growth. Both methylammonium and ammonium inhibited the uptake of nitrate and nitrite by the cells but inhibition by methylammonium, in comparison with that by ammonium, required a higher concentration and a longer time to develop. Inhibition by methylammonium is shown to be associated with its accumulation by the cells. Methylammonium also prevented the disappearance of nitrate from the interior of the cells (presumably by nitrate assimilation) whereas ammonium did not. It is concluded that methylammonium and ammonium differ in the ways in which they inhibit nitrate metabolism inP. tricornutum.Abbreviation MSX L-methionine-DL-sulfoximine  相似文献

Summary A medium consisting of agar plus yeast extract contained the necessary metabolites for rapid growth and sporulation ofHistoplasma capsulatum andBlastomyces dermatitidis. H. capsulatum when harvested after 10 or 30 days incubation period from this medium was shown to have a similar number of spores as well as total particle viability for each period of growth.The growth characteristics ofH. capsulatum and four different isolates ofB. dermatitidis on yeast extract medium were similar to that obtained previously using starling (Sturnis vulgaris) manure extract medium. These characteristics are rapid growth consisting of many viable spores and a low ratio of vegetative mycelium.Several isolations ofH. capsulatum from naturally contaminated soil specimens were made using yeast extract medium.From the Communicable Disease Center, Public Health Service, U. Thirty-nine strains from the genus Monascus were cultivated aerobically to study the relation between nitrogen nutrition and sporulation and pigment production. The effects of yeast extract, nitrate, ammonium, and ammonium nitrate have been compared. During cultivation the pHs of the different media are not the same, resulting in the formation of different coloured pigments. When the source of nitrogen is yeast extract or nitrate the pH is around 6.5 and red pigments are formed, whereas with ammonium or ammonium nitrate the pH is around 2.5 and the pigments are orange. It is proposed that only the orange pigments, monascorubrin and rubropunctatin, are produced biosynthetically and that the other pigments are formed from these by chemical transformations depending on the cultural conditions. The presence of organic nitrogen is optimal for growth and unfavourable for pigment production. Reduced growth and best pigment formation occurs with the three other nitrogen sources. Nitrate stimulates conidiation and sexual reproduction, while ammonium is inhibitory. Pigment production is better when conidiation is reduced. A mechanism is proposed for the control of sporulation and pigment production.  相似文献
