Background

Aluminium (Al) toxicity is a major agricultural constraint for crop cultivation on acid soils, which comprise a large portion of the world''s arable land. One of the most widely accepted mechanisms of Al tolerance in plants is based on Al-activated organic acid release into the rhizosphere, with organic acids forming stable, non-toxic complexes with Al. This mechanism has recently been validated by the isolation of bona-fide Al-tolerance genes in crop species, which encode membrane transporters that mediate Al-activated organic acid release leading to Al exclusion from root apices. In crop species such as sorghum and barley, members in the multidrug and toxic compound extrusion (MATE) family underlie Al tolerance by a mechanism based on Al-activated citrate release.

Scope and Conclusions

The study of Al tolerance in plants as conferred by MATE family members is in its infancy. Therefore, much is yet to be discovered about the functional diversity and evolutionary dynamics that led MATE proteins to acquire transport properties conducive to Al tolerance in plants. In this paper we review the major characteristics of transporters in the MATE family and will relate this knowledge to Al tolerance in plants. The MATE family is clearly extremely flexible with respect to substrate specificity, which raises the possibility that Al tolerance as encoded by MATE proteins may not be restricted to Al-activated citrate release in plant species. There are also indications that regulatory loci may be of pivotal importance to fully explore the potential for Al-tolerance improvement based on MATE genes.  相似文献

MATE (multidrug and toxic compound extrusion) transporter proteins mediate metabolite transport in plants and multidrug resistance in bacteria and mammals. MATE transporter NorM from Vibrio cholerae is an antiporter that is driven by Na+ gradient to extrude the substrates. To understand the molecular mechanism of Na+‐substrate exchange, molecular dynamics simulation was performed to study conformational changes of both wild‐type and mutant NorM with and without cation bindings. Our results show that NorM is able to bind two Na+ ions simultaneously, one to each of the carboxylic groups of E255 and D371 in the binding pocket. Furthermore, this di‐Na+ binding state is likely more efficient for conformational changes of NorM_VC toward the inward‐facing conformation than single‐Na+ binding state. The observation of two Na+ binding sites of NorM_VC is consistent with the previous study that two sites for ion binding (denoted as Na1/Na2 sites) are found in the transporter LeuT and BetP, another two secondary transporters. Taken together, our findings shed light on the structure rearrangements of NorM on Na+ binding and enrich our knowledge of the transport mechanism of secondary transporters. Cell-to-cell directional flow of the phytohormone auxin is primarily established by polar localization of the PIN auxin transporters, a process tightly regulated at multiple levels by auxin itself. We recently reported that, in the context of strong auxin flows, activity of the vacuolar ZIFL1.1 transporter is required for fine-tuning of polar auxin transport rates in the Arabidopsis root. In particular, ZIFL1.1 function protects plasma-membrane stability of the PIN2 carrier in epidermal root tip cells under conditions normally triggering PIN2 degradation. Here, we show that ZIFL1.1 activity at the root tip also promotes PIN1 plasma-membrane abundance in central cylinder cells, thus supporting the notion that ZIFL1.1 acts as a general positive modulator of polar auxin transport in roots.  相似文献

In rice, there are five members of the auxin carrier AUXIN1/LIKE AUX1 family; however, the biological functions of the other four members besides OsAUX1 remain unknown. Here, by using CRISPR/Cas9, we constructed two independent OsAUX3 knock‐down lines, osaux3‐1 and osaux3‐2, in wild‐type rice, Hwayoung (WT/HY) and Dongjin (WT/DJ). osaux3‐1 and osaux3‐2 have shorter primary roots (PRs), decreased lateral root (LR) density, and longer root hairs (RHs) compared with their WT. OsAUX3 expression in PRs, LRs, and RHs further supports that OsAUX3 plays a critical role in the regulation of root development. OsAUX3 locates at the plasma membrane and functions as an auxin influx carrier affecting acropetal auxin transport. OsAUX3 is up‐regulated in the root apex under aluminium (Al) stress, and osaux3‐2 is insensitive to Al treatments. Furthermore, 1‐naphthylacetic acid accented the sensitivity of WT/DJ and osaux3‐2 to respond to Al stress. Auxin concentrations, Al contents, and Al‐induced reactive oxygen species‐mediated damage in osaux3‐2 under Al stress are lower than in WT, indicating that OsAUX3 is involved in Al‐induced inhibition of root growth. This study uncovers a novel pathway alleviating Al‐induced oxidative damage by inhibition of acropetal auxin transport and provides a new option for engineering Al‐tolerant rice species.  相似文献

Four maize cultivars, which differ in tolerance to acid soils under field conditions ( Zea mays L., acid soil-tolerant C 525 M, BR 201 F and Adour 250, and acid soil-sensitive HS 7777) were used to study the influence of pH (4.3 and 6.0) and Al (0, 20 and 50 μ M ) on the elongation of seminal roots in nutrient solution. Root elongation was inhibited by high H+ concentrations (pH 4.3) in cultivars C 525 M, Adour 250 and HS 7777 but not in BR 201 F. After 20 h exposure to Al, root elongation rates were more inhibited in cultivars BR 201 F and HS 7777 than in C 525 M and Adour 250. The use of a computerized linear displacement transducer system with high resolution (1 μm) allowed the monitoring of short-term responses of root elongation to Al. In the three cultivars affected by H+ toxicity, but not in the acid-tolerant BR 201 F, Al supply caused an immediate, but transient increase of relative root elongation rates. This result supports the hypothesis that Al-induced growth stimulation is caused by amelioration of proton toxicity. The time required for 20 μ M Al to induce a 5% decrease of root elongation rates was shorter in the Al-sensitive BR 201 F (33 min) and HS 7777 (86 min) than in the Al-tolerant C 525 M (112 min) and Adour 250 (146 min) cultivars. However, the response-time to Al may be overestimated in the proton-sensitive cultivars, due to the transient stimulation of root elongation rates induced by Al. According to our results, experiments intended to investigate primary mechanisms of Al toxicity should be started after less than 30 min exposure to toxic Al concentrations, using pH conditions which avoid Al-induced growth stimulation due to amelioration of proton toxicity.  相似文献

Al-activated organic acid anion efflux from roots is an important Al resistance mechanism in plants. We have conducted homologous cloning and isolated Vigna umbellata multidrug and toxic compound extrusion (VuMATE), a gene encoding a de novo citrate transporter from rice bean. Al treatment up-regulated VuMATE expression in the root apex, but neither in the mature root region nor in the leaf. The degree of up-regulation of VuMATE was both partially Al concentration and time dependent, consistent with the delay in the onset of the Al-induced citrate efflux in rice bean roots. While La(3+) moderately induced VuMATE expression, Cd(2+) and Cu(2+) did not induce the expression. Electrophysiological analysis of Xenopus oocytes expressing VuMATE indicated this transporter can mediate significant anion efflux across the plasma membrane. [(14) C]citrate efflux experiments in oocytes demonstrated that VuMATE is a H(+) -dependent citrate transporter. In addition, expression of VuMATE in transgenic tomato resulted in increased Al resistance, which correlated with an enhanced citrate efflux. Taken together, these findings suggest that VuMATE is a functional homolog of the known citrate transporters in sorghum, barley, maize and Arabidopsis. The similarities and differences of all the known citrate transporters associated with Al stress in the MATE family are also discussed.  相似文献

Auxin signaling mediated by various auxin/indole‐3‐acetic acid (Aux/IAAs) and AUXIN RESPONSE FACTORs (ARFs) regulate lateral root (LR) development by controlling the expression of downstream genes. LATERAL ROOT PRIMORDIUM1 (LRP1), a member of the SHORT INTERNODES/STYLISH (SHI/STY) family, was identified as an auxin‐inducible gene. The precise developmental role and molecular regulation of LRP1 in root development remain to be understood. Here we show that LRP1 is expressed in all stages of LR development, besides the primary root. The expression of LRP1 is regulated by histone deacetylation in an auxin‐dependent manner. Our genetic interaction studies showed that LRP1 acts downstream of auxin responsive Aux/IAAs‐ARFs modules during LR development. We showed that auxin‐mediated induction of LRP1 is lost in emerging LRs of slr‐1 and arf7arf19 mutants roots. NPA treatment studies showed that LRP1 acts after LR founder cell specification and asymmetric division during LR development. Overexpression of LRP1 (LRP1 OE) showed an increased number of LR primordia (LRP) at stages I, IV and V, resulting in reduced emerged LR density, which suggests that it is involved in LRP development. Interestingly, LRP1‐induced expression of YUC4, which is involved in auxin biosynthesis, contributes to the increased accumulation of endogenous auxin in LRP1 OE roots. LRP1 interacts with SHI, STY1, SRS3, SRS6 and SRS7 proteins of the SHI/STY family, indicating their possible redundant role during root development. Our results suggested that auxin and histone deacetylation affect LRP1 expression and it acts downstream of LR forming auxin response modules to negatively regulate LRP development by modulating auxin homeostasis in Arabidopsis thaliana.  相似文献

The effect of NAA (naphthaleneacetic acid) on the development of cluster roots in members of the Proteaceae and Leguminosae was investigated. The exogenous addition of NAA led to initiation of cluster roots in phosphate conditions normally inhibitory for their development, but initiation took place within the limits of the cluster pattern under –P conditions. There was no change in spacing within the cluster root nor between cluster roots in Grevillea robusta Cunn. ex R. Br. or in rootlet length or cluster root length. In Lupinus albus L., change in rootlet length and cluster root length was noted at 10-10 and 1012 M NAA. In L. albus, the length of time that roots were exposed to NAA does not appear to be important, with similar levels of cluster root initiation after 48 h and 7 days. Cluster root production in G. robusta differed from that in L. albus in terms of the concentration of NAA needed to induce initiation, and in the effects of extremely low levels of NAA on rootlet numbers and lengths. L. arboreus L. does not produce cluster roots under –P conditions. Furthermore, neither L. arboreus L., L. angustifolius L., L. luteus L. nor L. mutabilis L. were induced to produce cluster roots under –P conditions, nor under +P conditions in the presence of exogenous NAA. Thus, exogenous NAA only leads to the induction of cluster roots, at levels of P normally inhibitive of their development, in species of Lupinus that produce them under –P conditions. Auxin-induced cluster roots develop within the same constraints as those developing under –P conditions. NAA does not induce cluster roots in species of Lupinus that do not produce them under –P conditions.  相似文献

Crop yields are significantly reduced by aluminum (Al) toxicity on acidic soils, which comprise up to 50% of the world’s arable land. Al‐activated release of ligands (such as organic acids) from the roots is a major Al tolerance mechanism in plants. In maize, Al‐activated root citrate exudation plays an important role in tolerance. However, maize Al tolerance is a complex trait involving multiple genes and physiological mechanisms. Recently, transporters from the MATE family have been shown to mediate Al‐activated citrate exudation in a number of plant species. Here we describe the cloning and characterization of two MATE family members in maize, ZmMATE1 and ZmMATE2, which co‐localize to major Al tolerance QTL. Both genes encode plasma membrane proteins that mediate significant anion efflux when expressed in Xenopus oocytes. ZmMATE1 expression is mostly concentrated in root tissues, is up‐regulated by Al and is significantly higher in Al‐tolerant maize genotypes. In contrast, ZmMATE2 expression is not specifically localized to any particular tissue and does not respond to Al. [14C]‐citrate efflux experiments in oocytes demonstrate that ZmMATE1 is a citrate transporter. In addition, ZmMATE1 expression confers a significant increase in Al tolerance in transgenic Arabidopsis. Our data suggests that ZmMATE1 is a functional homolog of the Al tolerance genes recently characterized in sorghum, barley and Arabidopsis, and is likely to underlie the largest maize Al tolerance QTL found on chromosome 6. However, ZmMATE2 most likely does not encode a citrate transporter, and could be involved in a novel Al tolerance mechanism.  相似文献

外源茉莉酸处理野生型拟南芥能够促进侧根的形成,而在asa1-1突变体中茉莉酸抑制侧根的形成,这与在该突变体背景下茉莉酸显著降低PIN2蛋白水平密切相关。为了进一步研究茉莉酸诱导PIN2蛋白水平下调的分子机制,文章采用正向遗传学的方法筛选asa1-1抑制子soa,期望获得茉莉酸处理后侧根发育恢复的突变体。通过筛选鉴定获得2个突变体:soa563和soa856。这2个突变体在10μmol/L茉莉酸甲酯处理条件下都能够恢复侧根发育,而且茉莉酸处理后PIN2蛋白水平降低的现象在soa563中被完全抑制,在soa856中被部分抑制。这些结果表明这两个突变基因可能影响了茉莉酸调控的PIN2蛋白水平下调途径,并且参于了茉莉酸对侧根发生的调控。对这两个基因的分离和功能研究将为阐明茉莉酸与生长素互作调控侧根发生的分子机制提供新的知识积累。  相似文献

Root growth relies on both cell division and cell elongation, which occur in the meristem and elongation zones, respectively. SCARECROW (SCR) and SHORT-ROOT (SHR) are GRAS family genes essential for root growth and radial patterning in the Arabidopsis root. Previous studies showed that SCR and SHR promote root growth by suppressing cytokinin response in the meristem, but there is evidence that SCR expressed beyond the meristem is also required for root growth. Here we report a previously unknown role for SCR in promoting cell elongation. Consistent with this, we found that the scr mutant accumulated a higher level of reactive oxygen species (ROS) in the elongation zone, which is probably due to decreased expression of peroxidase gene 3, which consumes hydrogen peroxide in a reaction leading to Casparian strip formation. When the oxidative stress response was blocked in the scr mutant by mutation in ABSCISIC ACID 2 (ABA2) or when the redox status was ameliorated by the upbeat 1 (upb1) mutant, the root became significantly longer, with longer cells and a larger and more mitotically active meristem. Remarkably, however, the stem cell and radial patterning defects in the double mutants still persisted. Since ROS and peroxidases are essential for endodermal differentiation, these results suggest that SCR plays a role in coordinating cell elongation, endodermal differentiation, redox homeostasis and oxidative stress response in the root. We also provide evidence that this role of SCR is independent of SHR, even though they function similarly in other aspects of root growth and development.  相似文献

Quantitative trait loci (QTL) analysis of aluminium (Al) tolerance was performed using Ler/Cvi recombinant inbred (RI) lines of Arabidopsis thaliana. Relative root length (RRL) (root length with 4 µm Al/root length with no Al at pH 5.0) on day 5 was used as the Al tolerance index for QTL analysis. Al tolerance judged by RRL was well correlated to tolerance judged by other indexes, including accumulation of callose, reactive oxygen species in the root apex and growth performance on acid soil containing a large amount of exchangeable Al. Using data sets with an hb2 of 0.91, two QTLs were detected at the top of chromosome 1 and bottom of chromosome 3. These QTLs explained 40 and 16% of the phenotypic variation of Al tolerance, respectively, and the positive effect of the Cvi allele. The QTL on chromosome 1 overlapped with a major QTL in another recombinant inbred population, and is possibly related to malate excretion. A complete pair-wise search revealed 11 sets of epistatic interacting loci pairs, which accounted for the transgressive segregation among the RI population. Several epistatic interactions shared the same chromosomal region, indicating the possible involvement of regulatory proteins in Al tolerance in Arabidopsis.  相似文献

Complexes of aluminium (Al) with organic ligands are believed to represent an important detoxification mechanism in acid soils. However, relatively little is known about the particular ligands produced by decomposing vegetation or about their effects on plant growth in the presence or absence of toxic Al. This paper reports an experiment on the effects of decomposition products of Eucalyptus camaldulensis leaves on the root elongation of maize (Zea mays) cv. DK687 in the presence or absence of Al. The static solution culture experiment used fulvic acid (FA) and humic acid (HA), extracted from E. camaldulensis leaves, at three nominal concentrations, viz. 40, 120 and 360 mg C L-1, replicated 4 times in the presence and absence of 30 µM Al. In the absence of Al, root elongation was increased by 30% by HA at 40 mg C L-1 and by 36% by FA at 120 mg C L-1. In the presence of 30 µM Al, the effects of toxic Al on root elongation were negated by FA and HA at all concentrations. Aluminium was totally complexed in all treatments except FA at 40 mg C L-1 in which treatment only 2.7 µM Al was present in the monomeric form. The E. camaldulensis FA and HA at concentrations of 40 and 120 mg C L-1, either in the presence or absence of Al, stimulated maize root elongation. Aluminium was strongly complexed by the E. camaldulensis FA and HA. The present results, in which FA and HA alleviated Al toxicity limitations on root elongation of maize, are relevant to the protection afforded to plant growth in acid soils amended with organic materials. They highlight the need to focus more on the role of FA and HA.  相似文献

In order to develop a new positron emission tomography (PET) probe to study hepatobiliary transport mediated by the multi-drug and toxin extrusion transporter 1 (MATE1), 11C-labelled metformin was synthesized and then evaluated as a PET probe. [11C]Metformin ([11C]4) was synthesized in three steps, from [11C]methyl iodide. Evaluation by small animal PET of [11C]4 showed that there was increased concentrations of [11C]4 in the livers of mice pre-treated with pyrimethamine, a potential inhibitor of MATEs, inhibiting the hepatobiliary excretion of metformin. Radiometabolite analysis showed that [11C]4 was not degraded in vivo during the PET scan. Biodistribution studies were undertaken and the organ distributions were extrapolated into a standard human model. In conclusion, [11C]4 may be useful as a PET probe to non-invasively study the in vivo function of hepatobiliary transport and drug–drug interactions, mediated by MATE1 in future clinical investigations.  相似文献

Inhibition of root elongation and modification of membrane properties are sensitive responses of plants to aluminium. The present paper reports on the effect of AI on lipid peroxidation and activities of enzymes related to production of activated oxygen species. Soybean seedlings (Glycine max L. cv. Sito) were precultured in solution culture for 3–5 days and then treated for 1–72 h with Al (AICI3) concentrations ranging from 10 to 75 μM at a constant pH of 4.1. In response to Al supply, lipid peroxidation in the root tips (< 2 cm) was enhanced only after longer durations of treatment. Aluminium-dependent increase in lipid peroxidation was intensified by Fe2+ (FeSO4). A close relationship existed between lipid peroxidation and inhibition of root-elongation rate induced by Al and/or Fe toxicity and/or Ca deficiency. Besides enhancement of lipid peroxidation in the crude extracts of root tips due to Al, the activities of superoxide dismutase (EC 1.15.1.1) and peroxidase (EC 1.11.1.7) increased, whereas catalase (EC 1.11.1.6) activity decreased. This indicates a greater generation of oxygen free radicals and related tissue damage. The results suggest that lipid peroxidation is part of the overall expression of Al toxicity in roots and that enhanced lipid peroxidation by oxygen free radicals is a consequence of primary effects of Al on membrane structure.  相似文献
