极濒危植物中华水韭休宁居群的遗传结构   总被引:12,自引:0,他引:12  
采用超薄平板微型聚丙烯酰胺等电聚焦电泳方法对极濒危蕨类植物中华水韭(Isoetes sinensis)现存于安徽休宁的5个亚居群的等位酶多样性和遗传结构进行了研究。结果表明:中华水韭居群每位点平均等位基因数A=1.7,平均多态位点比率P=55.56%,平均预期杂合度He=0.201。居群中半数的多念位点表现为等位基因的“固定杂合”,5个亚居群的遗传多样性无显著差异,但都表现出严重偏离Hardy—Weinberg平衡的杂合子过昔;其遗传变异主要发生于亚居群内(94.27%),亚居群之间的遗传分化较小(Gsl=0.0573),亚居群间遗传一致度较高(I=0.960—0.999)。我们推断这可能是由于居群构建之初的奠基者效应或者原种群曾经历了较为严重的遗传瓶颈所导致;此外亚居群间便利的基因交流也可能起到了很大作用(Nm=4.5062)。取自休宁居群的75株样品由13个专一多位点基因型(18个位点等位酶基因型)组成,亚居群及个体间高度遗传均质。本文还就中华水韭休宁野生居群濒危的遗传因素进行了探讨,并提出了相应的保育策略。  相似文献

为全面了解国家一级保护植物中华水韭(Isoetessinensis)的濒危机制,作者对分布于浙江松阳的中华水韭居群的自然生境进行了野外观察,在1m×1m样方尺度上研究了中华水韭与群落中出现频率较高的27个物种之间的联结性。结果表明:群落中物种组成贫乏,其组成成分中温带性质明显,中华水韭在不同小群落中分布密度差异较大,在少数小群落中能形成优势种群。中华水韭与薄叶假耳草(Neanotishirsuta)、鳢肠(Ecliptaprostrata)、短尖苔草(Carexbrevicuspis)等7个种群的种间联结性极显著,与多数种群间联结性不显著;中华水韭与大多数种群的联结系数(AC)值在–0.4至0.4之间,即关联小,说明中华水韭在群落中的存在是相对独立的;仅少数小群落的优势种与中华水韭之间有种间竞争。中华水韭受生境和种间竞争影响较大,生态适应性差。此研究结果对制定该物种合适的就地保护措施和恢复计划具有参考价值。  相似文献

中华水韭(Isoetes sinensisPalmer)为水韭科(Isoetaceae)水韭属(Isoetes)多年生沼泽植物,由于人为干扰日益严重,现分布范围急剧缩小,种群数量稀少,处于濒危状态,已被列为国家一级重点保护野生植物。为科学合理的保护和抢救这一珍稀濒危植物资源提供理论依据,为此对中华水韭屯溪居群的生长环境、面积、株丛数量、水体的pH值、伴生植物种类和生存现状等的调查研究。中华水韭屯溪居群面积500m^2,株丛密度较大,水体pH值为6.55,伴生植物有40余种,其中10种有较高优势度,生存面临多重威胁,保护迫在眉睫。  相似文献

利用核基因LEAFY第二个内含子片段对中国现存中华水韭(Isoetes sinensis Palmer)的遗传多样性进行分析,探讨了中华水韭自然居群的遗传多样性结构及其形成机制。结果显示:现存中华水韭7个自然居群共105个样本中存在78个单倍型,单倍型多样性(Hd)为0.989,核苷酸多样性(π)为0.021,遗传差异主要存在于居群内(72%),且存在较高的基因流(Nm=0.59)。同时,居群遗传学分析结果发现中华水韭居群不存在明显的谱系地理格局(GST>NST);Mantel检验中Rxy值为-0.286,P(rxy-rand≥rxy-data)值为0.370,表明居群遗传距离和地理距离之间没有明显相关性;UPGMA聚类分析显示处于海拔较高位置的2个居群与其它5个居群遗传关系较远;中性检验(Taijima's D、Fu & Li's D*和F*)检测结果均为负值,基于稳定模型的失配分布检测显示为多峰。根据中华水韭居群的地理位置,推测中华水韭的遗传结构可能与水系、海拔分布及其杂交后代多倍化的物种形成过程相关。  相似文献

中华水韭(Isoetes sinensis Palmer)为国家一级保护野生植物,被评价为极度濒危级(CR),仅分布在长江中下游,对环境污染敏感。本文以其人工苗为材料,用石蜡切片法,连续观察了根和叶的发育特征:叶原基横切面呈端部渐圆的三角形,由表皮原、皮层原和中柱原三部分组成;维管束中央首先出现的一个原生管胞,与其近轴面两侧后出现的两个后生管胞共同组成倒“V”字型的外始式初生木质部;成熟叶分化出表皮与皮层,表皮细胞间有嵌合,皮层部分细胞裂解成彼此不相通的4个通气道。根横切面呈圆形,成熟区分化出表皮、皮层(外皮层、中皮层、内皮层)和维管束三部分,中皮层部分细胞裂解呈一个完整的筒状通气道并与外皮层无连接,由内皮层包围的维管束横切面呈三角形。讨论了根和叶的形态结构、功能及系统学意义。  相似文献

Habitat fragmentation can lead to a decline of genetic diversity, a potential risk for the survival of natural populations. Fragmented populations can become highly differentiated due to reduced gene flow and genetic drift. A decline in number of individuals can result in lower reproductive fitness due to inbreeding effects. We investigated genetic variation within and between 11 populations of the rare and endangered plant Silene chlorantha in northeastern Germany to support conservation strategies. Genetic diversity was evaluated using AFLP techniques and the results were correlated to fitness traits. Fitness evaluation in nature and in a common garden approach was conducted. Our analysis revealed population differentiation was high and within population genetic diversity was intermediate. A clear population structure was supported by a Bayesian approach, AMOVA and neighbour-joining analysis. No correlation between genetic and geographic distance was found. Our results indicate that patterns of population differentiation were mainly caused by temporal and/or spatial isolation and genetic drift. The fitness evaluation revealed that pollinator limitation and habitat quality seem, at present, to be more important to reproductive fitness than genetic diversity by itself. Populations of S. chlorantha with low genetic diversity have the potential to increase in individual number if habitat conditions improve. This was detected in a single large population in the investigation area, which was formerly affected by bottleneck effects.  相似文献

Primulina tabacum Hance, is a critically endangered perennial endemic to limestone area in South China. Genetic variability within and among four extant populations of this species was assessed using AFLP markers. We expected a low genetic diversity level of this narrowly distributed species, but our results revealed that a high level of genetic diversity remains, both at population level (55.5% of markers polymorphic, H E = 0.220, I S = 0.321), and at species level (P = 85.6% of markers polymorphic, H E = 0.339, I S = 0.495), probably resulting from its refugial history and/or breeding system. High levels of genetic differentiation among populations was apparent based on Nei’s genetic diversity analysis (G st=0.350). The restricted gene flow between populations is a potential reason for the high genetic differentiation. The population genetic diversity of P. tabacum revealed here has clear implications for conservation and management. To maintain present levels of genetic diversity, in situ conservation of all populations is necessary.  相似文献

Isoetes yunguiensis is an endangered and endemic fern in China.Field survey indicated that only one population and no more than 50 individuals occur in the wild.The genetic variation of 46 individuals from the population remaining at Pingha (Guizhou Province,China)was assessed by Random Amplified Polymorphic DNA (RAPD)fingerprinting.Twelve primers were screened from sixty ten-bp arbitrary primers,and a total of 95 DNA fragments were scored.Of these,62.1%were polymorphic loci,which indicated that high level genetic variation existed in the natural population.The accumulation of genetic variation in the history of the taxon and the apparent minimal reduction effect on genetic diversity following destruction of habitat might be responsible for the high level genetic diversity presently remaining in the I.yunguiensis population.However,with the continuing decrease of population size,the genetic diversity will gradually be lost.We suggest that the materials from the extant population should be used for re-establishment of the populations.  相似文献

珍稀濒危蕨类植物东方水韭的遗传多样性   总被引:9,自引:0,他引:9  
采用ISSR分子标记对珍稀濒危蕨类植物东方水韭和中华水韭4个自然居群共72个样品进行了DNA多态性分析。从65个随机引物中筛选出11个有效引物,共产生98条DNA片段,其中68条为多态性条带,多态位点百分率(PPL)为66.33%。东方水韭两个居群的多态位点百分比率(PPL)较低(8.16%和7.14%)。中华水韭两个居群的PPL也较低(9.18%和23.47%)。中华水韭物种水平上的PPL(51.02%)高于东方水韭(9.18%)。AMO-VA分析结果表明,东方水韭遗传变异中主要存在于居群内(94.74%),而中华水韭绝大部分的遗传变异却存在于居群间(86.71%)。东方水韭和中华水韭个体间UPGMA聚类结果表明:同一物种的个体完全能聚在一起,中华水韭的两个居群也能明显分开,而东方水韭两居群的个体并不能完全聚在一起。探讨了可能造成上述居群遗传结构模式的主要因素,同时提出今后工作中需重点解决的问题。  相似文献

All extant populations of Hebe speciosa (Plantaginaceae), a threatened endemic New Zealand shrub, were investigated using the amplified fragment length polymorphism technique (AFLP). Genetic diversity indices varied significantly among geographical regions and were positively correlated with population size. Among-population genetic differentiation was high (mean pairwise ΦST = 0.47), implying complex historical relationships between disjunct populations and negligible contemporary gene flow. Southern populations exhibited extremely low genetic diversity relative to those found in Northland, suggesting that these populations may be more recent in origin. Patterns of genetic relationship among some populations indicate pre-European Māori dispersal and cultivation. The three northernmost populations were found to contain the majority of the species' remaining genetic diversity and thus, should be a focus for future conservation management. 中华水韭(Isoetes sinensis Palmer)属水韭科(Isoetaceae)草本拟蕨类,是长江中下游特有的古老物种,对环境污染敏感,为国家一级重点保护的极度濒危的野生植物。本研究通过人工培养,按不同发育阶段,用常规石蜡切片法连续观察了雌配子体及胚胎发育的全过程。结果显示:大孢子培养15~20 d萌发,雌配子体为球状、属壁内发育型,每个雌配子体着生8~20个颈卵器。成熟的颈卵器发育有多种可能,但只有颈沟细胞和腹沟细胞都退化,卵才有受精的机会。合子首先进行横向分裂,培养约50 d发育成数十个细胞的球形胚胎,约80 d雌配子体的细胞萎缩,第一叶及其叶舌、第一根结构更明显,出现第二叶。文中还初步探讨了中华水韭濒危的生殖生物学机制。  相似文献

中华水韭遗传多样性的RAPD分析   总被引:18,自引:4,他引:18  
采用RAPD方法对珍稀濒危植物中华水韭(Isoetes sinensis)4个自然居群的48个样品进行了DNA多态性分析。从60个随机引物中筛选出14个有效引物,共产生124条DNA片段,其中72条为多态性条带,总的多态位点百分率(PPB)为58.06%。各居群间多态位点百分率差异显著(0.81%-12.90%)。AMOVA分析结果表明,4个居群间基因分化系数Фst=0.5894,即遗传变异中有相当一部分来源于群体间(58.94%)。日益缩小的种群规模而导致的居群内近交和遗传漂变的发生以及居群间有限的基因交流可能是中华水韭目前遗传结构的主要成因。鉴于目前中华水韭居群内个体数偏少、遗传多样性较低的现状,建议对其进行就地保护并保护尽可能多的生境,对不同自然居群内的个体进行植株相互移栽和育苗移栽,以提高不同居群间的基因交流,尽可能地保护中华水韭的遗传多样性。  相似文献

Correlates between genetic diversity at intra- and interpopulation levels and the species diversity in plant communities are rarely investigated. Such correlates may give insights into the effect of local selective forces across different communities on the genetic diversity of local plant populations. This study has employed amplified fragment length polymorphism to assess the genetic diversity within and between 10 populations of Ranunculus acris in relation to the species diversity (richness and evenness) of grassland communities of two different habitat types, 'seminatural' and 'agriculturally improved', located in central Germany. Within-population genetic diversity estimated by Nei's unbiased gene diversity (HE) was high (0.258-0.334), and was not correlated with species richness (Pearson's r = -0.17; P = 0.64) or species evenness (Pearson's r = 0.15; P = 0.68) of the plant communities. However, the genetic differentiation between R. acris populations was significantly correlated with the difference in species evenness (Mantel's r = 0.62, P = 0.02), but not with difference in species richness of plant communities (r = -0.17, P = 0.22). Moreover, we also found that populations of R. acris from the 'seminatural' habitat were genetically different (amova, P < 0.05) from those in 'agriculturally improved' habitats, suggesting that gene flow between these habitat types is limited. The results reported in this study may indicate that habitat characteristics influence the genetic diversity of plant species.  相似文献

We determined the genetic diversity and population structures ofCarex breviculmis (Cyperaceae) populations in Korea, using genetic variations at 23 allozyme loci.C. breviculmis is a long-lived herbaceous species that is widely distributed in eastern Asia. A high level of genetic variation was found in 15 populations. Twelve enzymes revealed 23 loci, of which 11 were polymorphic (47.8%). Genetic diversity at the speciesand population levels were 0.174 and 0.146, respectively. Total genetic diversity (HT = 0.363) and within-population genetic diversity (Hs = 0.346) were high, whereas the extent of the population divergence was relatively low (GST = 0.063). Deviation from random mating (Fis) within the 15 populations was 0.206. An indirect estimate of the number of migrants per generation(Nm = 3.69) indicated that gene flow was extensive among Korean populations of this species. Analysis of fixation indices revealed a substantial heterozygote deficiency in some populations and at some loci. Genetic identity between popu-lations was high, exceeding 0.956.  相似文献

Isoetes hypsophila Hand.-Mazz. (Isoetaceae) is an endangered alpine quillwort endemic to the Qinghai-Tibetan Plateau (QTP). A total of 247 individuals of I. hypsophila from 11 populations were analyzed using ten pairs simple sequence repeats (SSR) markers. Seventy-seven alleles were recorded, with an average of 7.7 alleles per locus (ranging from 3 to 12) and the expected heterozygosity (HE) ranged from 0.392 to 0.536 (average 0.471). Clear population structure among I. hypsophila populations was found and the proportion of genetic differentiation among populations accounted for about 20% of total genetic diversity (FST = 0.178; probability of allelic co-ancestory Fcoal = 0.238). This genetic structure was probably due to limited gene flow hindered by complex terrains, bottlenecks resulting from water fluctuation in habitats, and repeated population retreat and recolonization during the ice ages. In conservation, four management units corresponding to four geographic regions were suggested, and the populations from different management units with higher genetic variation should be the priority targets of both in situ and ex situ conservation.  相似文献

? Premise of the study: The first microsatellite primers were developed for Isoetes hypsophila, an endangered quillwort species endemic to the Qinghai-Tibetan Plateau in China, to further describe its genetic variability and population structure. We also examined their cross-amplification in a congeneric species, I. sinensis. ? Methods and Results: Using the Fast Isolation by AFLP of Sequences COntaining Repeats (FIASCO) protocol, nine microsatellite loci were isolated and characterized in 32 samples from four natural populations of I. hypsophila. The primers amplified di- and hexanucleotide repeats with three to 11 alleles per locus. Seven of nine primers were cross-amplified in I. sinensis with two to seven alleles per locus. ? Conclusion: The microsatellite loci primers will be useful for studies of genetic diversity and gene flow in natural populations of Isoetes species.  相似文献

以中国山东、安徽、陕西、河南、云南和新疆6个栽培石榴群体的85个品种类型为试材,利用荧光标记AFLP对中国石榴群体遗传多样性进行了研究。结果表明:8对引物组合在种级水平扩增的多态性位点数范围从135~185个不等,平均为158.25个,多态位点百分比范围为62.5%~86.11%,平均为73.26%,说明中国石榴品种遗传多样性较为丰富;石榴品种种级水平遗传多样性大于群体水平,6个群体的遗传多样性依次为河南群体〉新疆群体〉陕西群体〉安徽群体〉山东群体〉云南群体,并且具有显著性差异;群体间的遗传分化系数GST为0.2018,说明石榴遗传变异主要存在群体内,群体间的遗传变异占总变异的20.18%,根据基因分化系数,测得的基因流Nm为1.9027,说明中国石榴群体间存在适当的基因交流;UPGMA聚类分析结果表明,同一群体的大部分品种都聚在一起,但同时存在部分基因交流。所有遗传参数表明,中国石榴栽培品种遗传多样性较为丰富,其中河南群体遗传多样性显著高于其他群体,在中国石榴品种选育中具有更为广阔的应用前景。  相似文献

应用RAPD技术研究了祁连山中段中国沙棘(Hippophae rhamnoidoes ssp.sinensis)4个居群和1个对照居群的遗传多样性水平,探讨中国沙棘边缘居群的遗传变异以及天然屏障祁连山对中国沙棘居群遗传结构的影响。结果显示,祁连山地区中国沙棘居群水平的Nei’基因多样度(h)和Shannon多态信息指数(I)分别为0.2070和 0.297 4,基因分化系数(Gst)为0.319 3,均高于中国沙棘整个分布区的平均值,表明该地中国沙棘边缘居群遗传多样性水平及居群间的遗传分化都有增加的趋势;居群间每代迁移数(Nm=1.065 9)显著低于异交风媒植物(Nm= 5.380),表明祁连山的隔离对中国沙棘居群间的基因交流有限制作用,造成了该地中国沙棘边缘居群间较高的遗传分化。  相似文献

猪血木为山茶科特有的单型属珍稀濒危植物,目前仅在广东省阳春市八甲镇有分布,且种群数量不足200株,为给濒危植物猪血木的种群成功回归自然和种群复壮提供科学依据,采用AFLP分子技术,对7个种源人工繁殖的猪血木幼苗遗传多样性进行研究。结果表明:6对引物共检测到位点数为115个,其中多态位点数为68个,多态位点所占比率为59.13%,观察等位基因数(Na)为1.5826、有效等位基因数(Ne)为1.3813、Nei′s基因多样性(H)为0.2215、Shannon多样性指数(Ⅰ)为0.3280。UPGMA聚类将7个种源的幼苗划分为3支,不同种源间幼苗的遗传距离与种源地理距离存在显著相关性(P<0.05)。说明不同种源人工繁殖猪血木幼苗仍保持较高的遗传多样性。建议采集不同种源种子进行人工繁殖和人工种群构建,促进物种的种群复壮。  相似文献
