We attempted to improve the bioavailability of green tea catechins by using food ingredients. The catechin bioavailability of a green tea extract administered to mice was significantly (p<0.05) increased by supplementing with steamed rice. This enhanced bioavailability was due to the increased concentration of plasma non-gallated catechins, especially epigallocatechin (EGC).  相似文献

Green tea, prepared from the steamed and dried leaves of the shrub Camellia sinensis, is known for its antioxidant and anti-carcinogenic effects. However, its effects on male gonadal functions have not been explored adequately and the present investigation has been undertaken to evaluate the effect of green tea extract on gonads of adult male albino rats. Results of in vivo studies showed that green tea extract (GTE) at mild (1.25 g%, identical to 5 cups of tea/day), moderate (2.5 g%, identical to 10 cups of tea/day) and high (5.0 g%, identical to 20 cups of tea/day) doses, for a period of 26 days, altered morphology and histology of testis and accessory sex organs. A significant dose-dependent decrease in the sperm counts, inhibited activities of testicular delta(5)3beta-and 17beta-hydroxysteroid dehydrogenase (delta5-3beta3-HSD and 17beta3-HSD respectively) and decreased serum testosterone level were noticed. Significant increase in serum LH level was observed after moderate and high doses; serum FSH level also increased but not significantly. Histopathological examination showed inhibition of spermatogenesis evidenced by preferential loss of matured and elongated spermatids. Results of this study showed that GTE at relatively high dose may cause impairment of both the morphological and normal functional status of testis in rodents and thus its consumption at relatively high doses raises concern on male reproductive function in spite of its other beneficial effects.  相似文献

To examine the possible benefits of tea flavonols, we compared anti-atherogenic effects between common and flavonol-rich tea cultivars. The tea infusion made from a flavonol-rich cultivar, but not a common cultivar, significantly decreased the plasma oxidized low-density lipoprotein level in mice fed a high-cholesterol diet. The result suggests that tea flavonols have the potential to protect against cardiovascular diseases.  相似文献

The antitranspirant effect of a number of substances on tea was studied and the effects of the most effective substance investigated in detail. The film type of antitranspirants reduced transpiration only at a considerably higher concentration than the stomatal type of antitranspirants. Of the stomatal type of antitranspirants phenyl mercuric acetate (PMA) was the most effective substance. Under actively transpiring conditions a 250 ppm solution of PMA reduced transpiration by about 30 per cent. The effect of PMA was localised and there was no translocation to untreated parts of the leaf blade. A spray of PMA reduced transpiration with gradually diminishing intensity for about 20 days, reduced vegetative growth of young plants grown both under non-water stress and water stressed conditions and reduced the yield of crop of mature plants during a drought. re]19760806  相似文献

The objective of this study was to investigate the antitumor activity of suspension cultures of tea callus cells grown in the presence of different concentrations of the growth regulator 2,4-dichlorophenoxy acetic acid (2,4-D) with or without light irradiation. The methanol and ethanol extracts of precipitated cells (MEP, EEP) exhibited stronger inhibitory effects on the growth of tumor cell lines than the water extract of precipitated cells (WEP) or the supernatant. Compared to culture under dark conditions, exposure to light irradiation led to significantly higher antitumor activity. The MEP from light irradiated cells at 250 μg/mL with 2.0 mg/L 2,4-D displayed more than 64% growth inhibition of HEP-2 cells, whereas normal cells showed less than 25% growth inhibition. The some fractions of MEP obtained from Diaion HP-20 column chromatography displayed the majority of inhibitory activity against the HEP-2 cell line. These results show that 2,4-D, and light stimulated the synthesis of antitumor compounds.  相似文献

Tamoxifen citrate (TAM), is widely used for treatment of breast cancer. It showed a degree of hepatic carcinogenesis. The purpose of this study was to elucidate the antioxidant capacity of green tea (Camellia sinensis) extract (GTE) against TAM-induced liver injury. A model of liver injury in female rats was done by intraperitoneal injection of TAM in a dose of 45mg Kg(-1) day(-1), i.p. for 7 successive days. GTE in the concentration of 1.5 %, was orally administered 4 days prior and 14 days after TAM-intoxication as a sole source of drinking water. The antioxidant flavonoid; epicatechin (a component of green tea) was not detectable in liver and blood of rats in either normal control or TAM-intoxicated group, however, TAM intoxication resulted in a significant decrease of its level in liver homogenate of tamoxifenintoxicated rats. The model of TAM-intoxication elicited significant declines in the antioxidant enzymes (glutathione-S-transferase,glutathione peroxidase, superoxide dismutase and catalase) and reduced glutathione concomitant with significant elevations in TBARS (thiobarbituric acid reactive substance) and liver transaminases; sGPT (serum glutamate pyruvate transaminase) and sGOT (serum glutamate oxaloacetate transaminase) levels. The oral administration of 1.5 % GTE to TAM-intoxicated rats, produced significant increments in the antioxidant enzymes and reduced glutathione concomitant with significant decrements in TBARS and liver transaminases levels. The data obtained from this study speculated that 1.5 % GTE has the capacity to scavenge free radical and can protect against oxidative stress induced by TAM intoxication. Supplementation of GTE could be useful in alleviating tamoxifen-induced liver injury in rats.  相似文献

Theanine, a unique non‐proteinogenic amino acid, is an important component of tea, as it confers the umami taste and relaxation effect of tea as a beverage. Theanine is primarily synthesized in tea roots and is subsequently transported to young shoots, which are harvested for tea production. Currently, the mechanism for theanine transport in the tea plant remains unknown. Here, by screening a yeast mutant library, followed by functional analyses, we identified the glutamine permease, GNP1 as a specific transporter for theanine in yeast. Although there is no GNP1 homolog in the tea plant, we assessed the theanine transport ability of nine tea plant amino acid permease (AAP) family members, with six exhibiting transport activity. We further determined that CsAAP1, CsAAP2, CsAAP4, CsAAP5, CsAAP6, and CsAAP8 exhibited moderate theanine affinities and transport was H+‐dependent. The tissue‐specific expression of these six CsAAPs in leaves, vascular tissues, and the root suggested their broad roles in theanine loading and unloading from the vascular system, and in targeting to sink tissues. Furthermore, expression of these CsAAPs was shown to be seasonally regulated, coincident with theanine transport within the tea plant. Finally, CsAAP1 expression in the root was highly correlated with root‐to‐bud transport of theanine, in seven tea plant cultivars. Taken together, these findings support the hypothesis that members of the CsAAP family transport theanine and participate in its root‐to‐shoot delivery in the tea plant.  相似文献

An experiment was conducted to determine the association of tea catechins to water stress in tea, with the objective of determining their suitability as indicators for predicting drought tolerance in tea (Camellia sinensis). The study consisted of six tea clones (BBK 35, TRFK 6/8, TRFK 76/1, TRFK 395/2, TRFK 31/30, and TRFK 311/287) and four levels of soil water content (38, 30, 22, and 14% v/v), which were arranged in a complete randomized design and replicated 3 times. The treatments were maintained for a period of 12 weeks. Tea shoots were sampled for catechin analysis during the 6th week of water treatment, in which fresh shoots with two leaves and a bud were plucked and steamed for 2 min, and dried at 70 degrees C to constant weight. Subsequently, the samples were ground and analyzed for catechins using an HPLC system. The total catechins showed significant correlation with shoot growth (r=0.65, P=0.006), soil water content (r=0.54, P=0.0066), and water stress index (r=0.67, P=0.0004). The epicatechin (EC) correlated with shoot growth (r=0.58, P=0.0032), soil water content (r=0.62, P=0.0014), and water stress index (r=0.63, P=0.0010). Similarly, epigallocatechin (EGC) correlated with shoot growth (r=0.65, P=0.0006), soil water content (r=0.50, P=0.0133), and water stress index (r=0.60, P=0.0021). However, epigallocatechin gallate (EGCg) and epicatechin gallate (ECG) showed no significant response to changes in soil water content. The shoot contents of EC and EGC in the six clones showed varied responses, with a distinct pattern in the water-stress tolerant clones (TRFK 6/8 and TRFK 31/30). The results suggest a potential use for EC and EGC as indicators in predicting drought tolerance in tea.  相似文献

Green tea (C. sinensis) extract (GTE) dose dependently produced reversal of despair in normal, reserpinised and diabetic mice, thereby demonstrating an antidepressant effect. Although the exact mechanism is yet to be explored, the possible inhibition of catechol-o-methyl transferase and monoamine oxidase enzymes may be responsible for antidepressant activity of GTE.  相似文献

Tea polyphenols have strong in vitro antioxidant activity. Due to their limited bioavailability, however, their contribution to in vivo antioxidant activity may depend on the form of administration. A human intervention study was performed to evaluate the bioavailability and antioxidant capacity of (-)-epigallocatechin-3-gallate (EGCG) administered as a single large dose in the form of either purified EGCG or as green tea extract (Polyphenon E). Plasma concentrations of tea polyphenols were determined by high-performance liquid chromatography (HPLC) analysis combined with coulometric array electrochemical detection (ECD). We found no differences in plasma EGCG concentrations and trolox equivalents determined by the trolox equivalent antioxidant capacity assay after administration of either form of EGCG. However, we found that the plasma antioxidant activity was significantly affected by changes in the plasma urate concentration, which may have interfered with the effect of tea polyphenols on the antioxidant activity. In addition, lymphocyte 8-hydroxydeoxyguanosine to deoxyguanosine (8-OHdG/10(6)dG) ratios were determined by HPLC with ECD. The 8-OHdG/10(6)dG ratios did not change significantly during the 24 h following both EGCG interventions but correlated significantly within individuals determined during the two interventions separated by 1 week. In summary, changes in plasma uric acid due to dietary intake were significantly correlated to the plasma antioxidant activity and exerted a stronger influence on the plasma antioxidant activity compared with the EGCG intervention. In future studies of dietary effects on the plasma antioxidant capacity, changes in plasma uric acid will need to be closely monitored.  相似文献

1. Methods for the separation and determination of the polyphenolic components of the tea plant by thin-layer chromatography and colorimetric reactions have been devised. 2. High concentrations of catechins, flavonols and depsides were found to be restricted to the young vegetative and floral shoots, whereas leucoanthocyanins or flavylogens were characteristic of the more bulky axial tissues of the plant. 3. In the young shoots cell growth was correlated with an increasing degree of flavonoid B-ring hydroxylation. 4. Maximal flavylogen concentrations occurred in the outer protective layers of stem and of seed coat. 5. Mature leaves were shown to contain derivatives of the flavones apigenin and luteolin. 6. Developing seedlings showed a steady increase in polyphenol complexity; flavylogens were concentrated at shoot and root apices and accumulated at the stem base. 7. It is postulated that the flavanols (leucoanthocyanins and catechins), because they can co-polymerize, are of use to the plant for protection of wood and bark against infection and decay.  相似文献

对广东省5个大型茶场中不同土壤背景值的8个茶园进行了土壤及茶(CamelliasinensisL.)树各部位的铜含量分析调查。地处我国南方的广东省茶园,土壤中铜含量在2.39~53.05mg kg之间。有效态铜含量相对较低,仅在0.45~3.17mg kg之间。茶树中各部位铜含量大小依次为枝条>幼叶>根>成熟叶。茶树枝条和幼叶铜含量较高,平均分别达14.5和14.3mg kg,远高于成熟叶铜含量水平(9.7mg kg)。而同时期相应茶场的茶叶制成品铜含量则较幼叶(一芽两叶)铜的含量高。土壤中的总钾、有效磷和总铜等都对茶树各部位铜的累积产生较大影响,但只有土壤有效磷含量对铜在茶树体内向枝条部分的转移和累积影响达显著程度,枝条铜含量与土壤有效磷之间的相关系数r=-0.74,呈显著负相关(P<0.05)。  相似文献

Plant polyphenols have gained prominence in quality of plant products and in human health. An experiment was conducted to determine the association of tea polyphenols with water stress and their suitability as indicators for drought tolerance. The experiment was conducted in a 'rain-out' shelter, and consisted of six tea clones (BBK 35, TRFK 6/8, TRFK 76/1, TRFK 395/2, TRFK 31/30, and TRFK 311/287) and four levels of soil water contents (38, 30, 22, and 14% v/v), which were maintained for a period of 12 weeks. The treatments were arranged in a completely randomized design and replicated three times. Plant growth was monitored over 6 weeks, and a water stress index was calculated to determine water-stress tolerant clones. Total polyphenols in tea shoots was analyzed and a regression analysis done. The results indicate that declining soil water content (SWC) reduced both growth and content of polyphenols in tea. Tolerant clones maintained a high polyphenol content at low SWC, and also showed less fluctuation in phenolics when subjected to changes in SWC. There was significant (P<0.001) correlation of total polyphenol content with shoot growth and WSI of tea, and a linear relationship (r2=0.97) between SWC for tea and both, water stress index and shoot polyphenol content. We report that there is a potential to use polyphenols as indicators for selection of drought-tolerant tea cultivars.  相似文献

Boron (B) re-translocation is an important factor determining tolerance to B deficiency in plants. In this work growth, B content of leaves with different ages, B partitioning between soluble and cell wall (CW) fractions, and B re-translocation were investigated in tea (Camellia sinensis (L.) O. Kuntze) plants grown hydroponically without (<2.5 μM) and with adequate (46 μM) B supply. Under B deficiency, the proportion of CW bound B increased in the old leaves but decreased in roots. Contrastingly, the proportion of CW bound B was not influenced by B supply in the young leaves. A continuous reduction of B content was observed in all fully expanded leaves as well as in roots of low B plants. Taken together, these results revealed considerable re-translocation of B from mature to growing leaves. Leaf extract and phloem exudate samples were analyzed and sucrose, glucose, and fructose were detected while xylitol, sorbitol, mannitol, maltose, galactose, cellobiose or rafinose were not found in these samples. In the leaf extracts, concentration of sucrose increased under B deficiency conditions, concentration of glucose decreased, while that of fructose remained unchanged. Our results provide circumstantial evidence for a considerable re-translocation of B in tea plants despite lacking polyol compounds.  相似文献

1. Flavonoid synthesis was able to proceed in darkness in young shoots and seedlings of the tea plant, but was increased by light. 2. The initial effect of darkness was to inhibit synthesis of the A ring or its linkage to the phenylpropane moiety of the flavonoid, but later the hydroxylation state of the flavanols was affected, leading to smaller proportions of gallocatechins and of complex leucoanthocyanins. 3. The esterification of catechins with gallic acid was less affected, so that the ratio of catechin gallates to simple catechins also increased. 4. The flavylogen content of darkened stems, especially in seedlings, was much less decreased than that of leaves; however, a short subsequent light-treatment caused an increase in polymerization.  相似文献

To identify the chemical forms of aluminum (Al) transported from roots to shoots of tea plants (C. sinensis L.), 27Al-nuclear magnetic resonance and 19F NMR spectroscopy were used to analyze xylem sap.The concentration of Al in collected xylem sap was 0.29 mM, twice as high as that of F. Catechins were not detected in xylem sap. The concentration of malic acid in xylem sap was higher than that of citric acid, whereas the concentration of oxalic acid was negligible.There were two signals in the 27Al NMR spectra of xylem sap, a larger signal at 11 ppm and a smaller one at −1.5 ppm. The former signal was consistent with the peak for an Al-citrate model solution, suggesting that an Al-citrate complex was present in xylem sap. Although the latter signal at −1.5 ppm was thought to indicate the presence of an Al-F complex (at 1.7 ppm) in xylem sap, there was only one signal at −122 ppm in the 19F NMR spectrum of xylem sap, indicating that the main F complex in xylem sap was F.These results indicate that Al might be translocated as a complex with citrate, while Al-malate, Al-oxalate and Al-F complexes are not major Al complexes in xylem sap of tea plants.  相似文献

Metal-binding sites in epigallocatechin gallate (egcg) were identified by measuring (1)H nuclear spin relaxation time T(1) under the coexistence of paramagnetic Mn(2+). The T(1) of the proton of gallate ring (D-ring) was shortened by the paramagnetic ion to a greater extent than the proton in gallocatechin ring (B-ring); the D-ring OH groups occupy the first coordination sphere around a metal ion to form a diolate chelate ring, while B-ring OH groups have a weak interaction with the metal ion. Comparison of changes in T(1) of egcg and epigallocatechin (egc) indicates that the gallate ring has a higher coordination capability than that of the gallocatechin ring so that the former plays a predominant role in the complex formation of egcg.  相似文献
