Erythrina caffra seeds were shown to be true anaerobic germinators.They exhibit a Pasteur effect, high alcohol dehydrogenase activityand produce high levels of ethanol under anoxia, in which situationgermination starts to be suppressed by as little as 0.1% externallyapplied ethanol. Toxic levels of ethanol production appear tobe prevented by a decrease in the rate of ethanol accumulation.Carbon monoxide does not inhibit germination. Cyanide, SHAM,iodoacetate, pyrazole, and 4-methylpyrazole are more inhibitoryto anoxic than aerobic germination whereas azide, arsenate,and fluoride inhibit both. Azide, pyrazole, 4-methylpyrazoleand a low concentration of cyanide and SHAM tend to stimulateethanol production in air. At 10 mol m–3, 4-methylpyrazolestimulates anaerobic ethanol production. At higher concentrationsthis compound and all other inhibitors used suppress anaerobicethanol production initially. Inhibition of ethanol productionby 10 mol m–3 cyanide is paralleled by an accumulationof acetaldehyde. Azide and cyanide appear to exert their inhibitoryeffect at different loci. The effects of CO2 on dormancy and germination were examinedusing seeds of cocklebur (Xanthium pennsylvanicum Wallr.) andgiant foxtail (Setaria faberi Herrm.). The rate of germinationof the giant foxtail seeds as well as cocklebur was promotedby exogenously applied CO2 at a concentration of 30 mmol mol-1regardless of the sowing conditions. However, seeds which failedto germinate in the presence of CO2, entered a secondary phaseof dormancy under unfavourable germination conditions. If CO2was applied to seeds under conditions such as water stress imposedwith a 200 mol m-3 mannitol solution, a hypoxic atmosphere of100 mmol mol-1 O2 or a treatment of 0·1 mol m-3 ABA,development of secondary dormancy was accelerated. These contrastedeffects of CO2 were observed in ecological studies. Under naturalfield conditions germination of buried giant foxtail seeds respondedpositively to CO2 during a period of release from primary dormancyfrom Feb. to May, but CO2 accelerated secondary dormancy commencingin early Jun. In other words, in the presence of CO2, both theenvironmental conditions and the germination states of the seedsclearly showed secondary dormancy-inducing effects. Seed conditioning and germination in witchweed (Striga asiatica(L.) Kuntze) were temperature-dependent. With higher conditioningtemperatures, shorter conditioning time was required for germinationwith terminal dl-strigol (strigol) treatment at 30 °C. Maximumgermination (80–100%) was obtained by conditioning inwater at 20, 25, 30 and 35 °C for 14, 7, 5 and 3 d, respectively,and terminally treating with 10–6 M strigol at 30 °C.Seeds conditioned in 10–8 M strigol instead of water germinatedmuch less with the same terminal strigol treatment. Generally,conditioning was slower when seeds were conditioned in strigolrather than water. The reduction in germination rate by pretreatmentin strigol or pretreatment at low temperatures could be overcomeby increasing the terminal strigol concentration in the germinationtest. Conditioned seeds did not germinate at 10 and 15 °Cwith a terminal 10–6 M strigol treatment but yielded closeto maximum germination at 25, 30 and 35 °C with the sameterminal strigol treatment. To obtain maximum germination, boththe minimum conditioning temperature and the minimum germinationtemperature for conditioned seeds were 20 °C. Intact wild-type tomato (Lycopersicon esculentum cv. Moneymaker)seeds do not complete germination to the same percentage orat the same speed as intact ABA-deficient sitiens (sitw) mutantseeds when seeds of both genotypes are imbibed on polyethyleneglycol (PEG) solutions of –0.3 to –1.5 MPa osmoticpotential. However, if the thicker testas of wild-type seedsare removed (stripped) from the micropyle without damaging theendosperm, both the percentage and speed of germination at lowexternal water potential are similar to that of sitw mutantseeds. Removing the micropylar end of the testa from sitw seedsdid not enhance either the speed or percentage of germinationon PEG solution. Despite similar germination percentage and speed between strippedwild-type seeds and either stripped or intact sitw seeds underosmotic stress, some differences in seed metabolism are evidentbetween genotypes. The activity of endo-ß-mannanasewas greater in the endosperm of sitw mutant seeds compared tothe endosperm of wild-type seeds when seeds were exposed toosmotic stress. Although  相似文献

The effects on the germination of positively photoblastic uppercocklebur (X anthium pennsylvanicum Wallr.) seeds by pretreatingwith anoxia, chilling, cyanide or azide, which stimulates theirdark germination, were examined in relation to light actions.Prior to experiments, seeds were pre-soaked at 23 °C inthe dark for 1 or 2 weeks to remove the pre-existing Pfr. Whenthe prctreatment conditions were suboptimal for germinationinduction, the stimulating effects of the pretreatments on germinationduring a subsequent dark period at 23 °C were manifest onlywhen seeds were irradiated with red light before or after thepretreatment Red light promotion was reversed by blue or far-redlight treatment. However, both prc-chilling for 6 d at 8 °Cand prctreatment with 1· 5 mol m – 3 NaN3 for 2d could induce full germination without red light exposure.On the other hand, both pre-exposure to anoxia for 8 d and pretreatmentwith 30 mol m–3 KCN could induce the dark germinationonly when germination occurred at 33 °C which is known toaugment the ratio of an alternative respiration flux to a cytochromeone. Moreover, the dark germination in response to these inductionswere strongly inhibited by the inhibitors of alternative respiration,propyl gallate and benzohydroxamic acid, applied during a subsequentdark period. It was thus suggested that Pfr has some relationto the operation of two respiration systems of cocklebur seeds,but it is not indispensable to germination of this positivelyphotoblastic seed. Various photon doses (net number of photons per unit area perday), provided by varying both photon flux density and photoperiod,were applied to imbibing seeds of seven lots of four speciesof Compositae in various germination test regimes. In all fourspecies germination was dependent upon photon dose, the productof photon flux density and daily duration of exposure. The responsewas quantified by linear relations between the probit of percentagegermination and the logarithm of photon dose. In general, photonflux density and photoperiod only influenced the stimulationof germination by the low energy reaction indirectly (as factorsof daily photon dose), whereas there was a tendency for photoperiodto have a direct influence on the inhibition of germinationby the high irradiance reaction. Reducing the germination testtemperature from 25?C to 20?C and 15?C not only increased thedark germination of L. sativa L., but also broadened the photondose range at which full germination occurred by reducing theminimum value necessary for the germination of the most dormantseeds, and increasing the maximum value which failed to inhibitthe germination of any seeds. Differences between L. sativaand L. serriola L. in the response of germination to white lightwere only quantitative, rather than qualitative. The singlemost promotory dose for all four species was 3 ? 10–3mol m–2 d–1, although the inhibitory action of dosesup to 10– mol m– d– was generally only slight. The effects of kinetin and gibberellin were examined under anaerobicconditions (0% oxygen) and aerobic conditions (20% oxygen) onthe germination of dehusked seeds of indica and japonica ricecultivars that had been harvested at different times duringthe formation of seeds. Surjamkhi was used as a representativeof deep dormant indica cultivars and Assam IV as a less dormantindica cultivar. Sasanishiki was used as the japonica rice cultivar.Both phytohormones were applied at a concentration of 10-3Mwhichproved to have the greatest stimulatory effect in preliminarywork at concentrations of 10-3–10-5M. Under aerobic conditions,inhibition of germination by dehusking of Sasanishiki seedsthat had been harvested either 30 or 60 d after anthesis wasovercome by kinetin and all seeds germinated. Complete germinationinduced by kinetin under aerobic conditions was also achievedwith the dehusked seeds of the indica rice cultivar Assam IVthat had been harvested on two occasions and of Surjamkhi thathad been harvested 28 d after anthesis. In contrast, germinationof dehusked japonica seeds stimulated by anaerobiosis was inhibitedby kinetin. The stimulatory effects of gibberellin on the germinationof indica and japonica rice seeds were observed under aerobicand anaerobic conditions. Under anaerobic conditions, the responsesof dehusked indica and japonica rice seeds to kinetin and gibberellindiffered, being negative with kinetin and positive with gibberellin.Under aerobic conditions, the stimulatory effects of kinetinon germination of dehusked seeds were greater than those ofgibberellin. Thus, treatment with kinetin appears to be usefulfor breaking the considerable dormancy commonly observed inthe dehusked seeds of indica rice. Mechanisms are proposed toexplain the stimulatory effects of these phytohormones on thegermination of dehusked seeds of indica and japonica rice underaerobic and anaerobic conditions. Using X-ray photography and flow cytometry, the internal morphologyand DNA replication activity of wild type (wt), GA- (gib-1 )and ABA-deficient (sitw ) tomato (Lycopersicon esculentum Mill.cv. Moneymaker) mutant seeds were studied. During seed formation,from 30 to 45 d after pollination (DAP) the endosperm becomessolid and the seed starts to gain desiccation tolerance. Atthis time significant changes occur in the amounts of DNA inradicle tip cells. At 30 DAP, radicle tip cells of the threegenotypes manifest about 60% of 2C, 30% of 4C and 10% of 8Camounts of DNA. Upon maturation (45 DAP onwards), most cellsin the seeds of the three genotypes arrest in the G1phase ofthe cell-cycle with 2C amounts of DNA. However, a relativelyhigh proportion of cells with 4C amounts of DNA was detectedin the radicle tip cells ofsitw compared with wild type andgib-1. At the well-matured stage (60 DAP), there were about 2% ofseeds with free space in wild type andgib-1 , and about 13%insitw . At the over-matured stage (75 DAP), even more seedswith free space were found insitw , whereas no increase in theproportion of the seeds with free space was detected in theother two genotypes. In -1.0 MPa PEG-6000 with or without 10µM GA4+7, no germination occurred in well-matured wildtype andgib-1 seeds, whether or not they were dried after harvest.However,sitw seeds were able to germinate both in over-maturefruit and in -1.0 MPa PEG-6000. Priming of dried seeds in -1.0MPa PEG induced a large amount of free space in almost all seedsof the three genotypes, and nuclear DNA synthesis in the radicletip cells of wild type andsitw seeds. However, PEG priming offresh (non-dried) seeds had no effect on the amount of freespace and 2C/4C DNA ratios in wild type orgib-1 seeds, but didinduce free space in about 20–25% ofsitw seeds and provoked4C signals insitw seeds. Removal of the endosperm and testaopposite the radicle tip of seeds resulted in root protrusion,the induction of free space and an increase of 4C DNA signalsin the three genotypes. It is concluded that ABA is crucialfor the efficient arrest of tomato embryo radicle tip cellsin G1phase upon maturation, whereas GAs play an important rolein re-initiating 4C DNA levels upon germination. Studies have been made on the time to germination and earlyseedling growth of the tomato Lycopersicon esculentum var. Potentateand Lycopersicon pimpinellifolium var. Red Currant, as wellas on derivative generations from their hybridization. Althoughtime to germination showed a genetic component, the relationshipsbetween different genotypes was much influenced by environmentalfactors. A marked maternal effect on time to germination dueto the varying seed sizes was noted while variation betweendifferent genotypes of similar seeds size was ascribed to anendospermic effect. The F1 hybrid embryo with L. Pimpinellifoliumas maternal parent contained more cells than that of the parentbut the hybrid cmbryo with L. esculentum as maternal parentcontained a similar number of cells to that of the parent itself.It is suggested that the results for embryo size both supportAshby‘s assertion that embryo size may be important indetermining heterosis, and also Hatcher’s findings in1940, that heterosis for hypocotyl extension is found in hybridsfrom parcnts of different sized seeds provided the small seededvariety is the maternal parent.  相似文献

Ethylene (10 µ1–1) caused about one-third of highlydark-dormant seeds of common purslane (Portulaca oleracea L.)to germinate in the dark. Attempts were made to increase germinationin the dark with nitrate and ethylene combinations. When applieddirectly to the seeds, KNO3 did not stimulate germination andKNO3 plus ethylene did not increase germination above that ofethylene alone. Pre-incubation of seeds in KNO3 for 4 to 7 dbefore the ethylene applications significantly increased germination.The effects of the KNO3 pre-incubation were additive at eachof four ethylene concentrations (0.1–100 µ11–1).Potassium nitrate was effective only when ethylene followedthe KNO3 pre-incubation period. Potassium nitrite stimulatedabout 25 per cent of the seeds to germinate without a pre-incubationperiod and without ethylene. Also, ethylene plus KNO2 enhancedgermination above that achieved by either stimulus alone. Silvernitrate did not block the ethylene promotion of germination,but reversed the typical ethylene inhibition of seedling growthfollowing germination. The results support the views that nitrateexerted its effect via conversion to nitrite within the seedand that the rate of nitrate conversion may be a limiting factorin the dark germination of common purslane seeds. The intact dormant seeds of Dioscorea tokoro germinate slowlyif at all between 11-23°C; for full and rapid germinationthey require prior chilling treatment [Okagami and Kawai (1982)Bot. Mag. Tokyo 95: 155]. The germination abilities of zygoticembryos detached from dormant seeds of this species were studiedunder various nutritional and temperature regimes. For germinationof embryos, the minimum nutritional components in Murashigeand Skoog's (1962) medium that were required were sucrose andNO3 or SO2–4. As the source of carbohydrate forgermination of detached embryos, sucrose, mannose and maltosewere effective; glucose and fructose were less effective; andrhamnose was entirely unable to support germination. Embryos detached from dormant seeds, incubated with the sucroseplus KNO3, germinated more rapidly with increasing temperatureup to 35°C. However, application of sucrose and KNO3 didnot induce germination of intact seeds above 26°C. Therefore,it is very possible that the endosperm exerts an inhibitoryfunction on germination at such high temperatures. When seeds were incubated after a cut was made over a smallpart of the edge of the endosperm in which the radicle of theembryo is encased, germination occurred rapidly but the increasein germination percentage was slight. This result suggests thatthe endosperm suppots part of the germination inhibition bymeans of a mechanical barrier or its impermeability to wateror gases. Physiological features of the endosperm alone or interactionsbetween the embryo and endosperm may contribute significantlyto the characteristics of dormancy of intact seeds of this species. The response of the germination of seeds of Barbarea vema (Mill.)Aschers, Brassica chinensis L., Brassica juncea (L.) Czern.& Coss., Brassica oleracea L. var. gongylodes L., Camelinasaliva (L.) Crantz, Eruca saliva Mill., Lepidium sativum L.,Nasturtium officinale R. Br., and Rorippa palustris (L.) Besserto white fluorescent light of different photon flux densitiesapplied for different daily durations in a diurnal alternatingtemperature regime of 20 °C/30 °C (16 h/8 h) was quantifiedby linear relations between probit percentage germination andthe logarithm of photon dose, the product of photon flux densityand duration. The low energy reaction, in which increasing dosepromotes germination, was detected in all the seed populationsbut in Barbarea vema and Brassica Juncea the lowest photon doseapplied (10–5–2 and 10–5 7 mol m–2 d–1,respectively) was sufficient to saturate the response. Comparisons,where possible, between photoperiods demonstrated reciprocity,i.e. germination was proportional to photon dose irrespectiveof photoperiod, for the low energy reaction in Brassica oleracea(1 min d–1 to 1 h d–1), Camelina saliva (1 min d–1to 8 h d–1), Eruca saliva (1 min d–1 to 24 h d–1),Lepidium sativum (I min d–1 to 8 h d–1) and Rorippapalustris (1 min d–1 to 8 h d–1), but not in Brassicachinensis and Nasturtium officinale. The high irradiance reaction,in which increasing dose inhibits germination, was detectedin Barbarea vema, Brassica chinensis, Brassica juncea, Brassicaoleracea, and Camelina saliva. The minimum dose at which inhibitionwas detected was lO–0–3 mol m–2 d–1.These results are discussed in the context of devising optimallight regimes for laboratory tests intended to maximize germination The response of germination to photon dose was also quantifiedwith 3 x 10–4 M GA2, co-applied (Brassica chinensis, Camelinasaliva, and Lepidium sativum) and with 2 x 10–2 M potassiumnitrate co-applied (Brassica chinensis). In the latter casepotassium nitrate had no effect in the dark and inhibited germinationin the light, but GA2, promoted germination substantially inall three species. Variation amongst seeds in the minimum photondose required to stimulate germination was not affected by co-applicationof GA2, in Brassica chinensis and Camelina saliva, whereas seedsof Lepidium salivum showed a narrower distribution of sensitivitiesto the low energy reaction in the presence of GA2 Barbarea vema (Mill.) During in vitro growth and maturation of soybean seeds, cessationof embryo growth and dry weight accumulation occurred in thepresence of abundant C and N nutrients. Axis followed by cotyledontissues changed from green to yellow, and post-harvest germinationpotential declined if cultured after yellowing of axis tissues.A tissue specific accumulat;on of methanol occurred during thein vitro culture of immature seeds (i.e. initially 50 to 70mg fresh weight) to maturity in liquid medium. Methanol accumulatedto 3.0 g m–3 or 50 µg seed–1 in the medium,while methanol decreased from 37 to about 3.0 µg g–1fresh weight in cotyledons. By contrast, axis tissues increased20-fold in methanol concentration to 90 µg g–1 during20 d in culture. Ethanol was present only in trace amounts inaxis tissues and medium. Addition of exogenous methanol vapourto in situ grown seeds during precocious maturation decreasedsubsequent seedling vigour and germination with increasing levelsof exposure. Methanol accumulation in axis tissues during thegermination phase was not correlated with high temperature andtissue water content treatments which simulated pre-harvestdeterioration of seeds. However, the accumulation of methanolduring in vitro seed development and maturation in liquid culturemay contribute to reduced post-harvest germination performance. Damage and degradation of cellular proteins is observed duringage-induced seed deterioration. L-Isoaspartyl protein methyltransferase(EC 2.1.1.77 [EC] ) is an enzyme hypothesized to play a role in limitingand repairing age-induced damage to proteins. Tomato (Lycopersiconesculentum Mill. ‘New Yorker’) seeds were assayedfor changes in L-isoaspartyl methyl-transferase activity duringaccelerated ageing and after osmotic priming. Accelerated ageingof seeds for 1–4 d at 45C and 100% relative humidityreduced germination from 94% to 71%, increased the mean timeof germination (MTG) from 2.4 to 5.8 d, and was accompaniedby a correlative decrease in L-isoaspartyl methyltransferaseactivity (r2=0.90). Aged and untreated seeds were primed for7 d at 20C in darkness using aerated solutions of 3% KNO3 orpolyethylene glycol 8000 (PEG) with equivalent osmotic potential(–1.25 MPa). Priming with KNO3 decreased the MTG, butdid not improve germination percentage for untreated seeds.Priming did not affect L-isoaspartyl methyltransferase activityin untreated seeds, but restored activity in aged seeds primedin KNO3 to levels near that of untreated seeds. Priming withPEG did not effectively improve the MTG or increase L-isoaspartylmethyltransferase activity. During germination, L-isoaspartylmethyltransferase activity remained constant for 48 h post-imbibitionand then declined, suggesting that the enzyme was developmentallyregulated and inactivated or degraded as radicle emergence occurred. Dormant E. crus-galli seeds can be stimulated to germinate bycontact with solutions of several anaesthetic-like substances.The order of activity was n-pentanol > benzyl alcohol >n-butanol > n-propanol > ethanol. 2-propanol is nearlyinactive. The relative activity of the substances as germinationstimulants is closely related to their membrane-buffer partitioncoefficients in a manner similar to their effectiveness as anaestheticsin animals. Thus, n-pentanol is a more active stimulant of germinationthan ethanol and its increased effectiveness is correlated witha higher membrane-buffer partition coefficient Similarly, ananaesthetic that stimulated germination (n-propanol) causedgreater leakage of electrolytes, U.V. absorbing substances andamino acids from E. crus-galli seeds than a substance that wasnot active as a germination stimulant (2-propanol). In animals,action of anaesthetics can be prevented by application of externalpressure, an argument for membrane action. Action of n-propanolin stimulating germination can be similarly prevented by pressurein E. crus-galli seeds. The several lines of evidence supporta membrane action for anaesthetic-like substances in seeds and,accordingly, a membrane site as the locus of seed dormancy. The viability of seeds is associated with ageing and storageconditions. A loss of viability is accompanied by slow germination,reduced growth, and a decline in protein and poly(A)+RNA synthesis.This paper reports on the activity of poly(A) polymerase indry and germinating embryos of Triticum durum Desf. cv. Cappellicaryopses of different ages and viability. The enzyme was presentas a single form during ageing and germination. The poly(A)polymerase was active at decreasing levels in all aged dry embryos,in parallel with loss of viability. Its activity strongly increasedduring the germination only in viable embryos. The observedincrease was due to de novo synthesis of the enzyme. Poly(A)polymerase synthesis was low during germination of less viableembryos and absent in older ones. Reduced poly(A) polymeraseactivity in dry or germinated wheat embryos may cause a shorteningof poly(A) chains in vitro and a decline in poly(A)+RNA synthesis.Copyright1995, 1999 Academic Press Triticum durum Desf. cv. In contrast, seed germination (radicle protrusion)can occur when seeds as young as 35 DAF (68–79% moisture)are excised, but this germination docs not lead to comparableseedling growth frequencies unless seeds are first given a moistureloss treatment to artificially reduce their moisture below 60%.A moisture loss treatment applied at 35 DAF thus enables seedto undergo the transition from germination (cell expansion)to seedling growth (cell division and expansion) to the extentthat treated immature seed have a vigour index comparable toseeds matured on the plant (100%). The pattern of protein synthesisin vivo was examined in 35 DAF seed using [35S]-methionine incorporation.When moisture loss treatment was applied for 24 h to 35 DAFseeds, seeds synthesized several new polypeptides when comparedwith untreated seeds at the same developmental stage. The sameseed samples showed 0% seedling growth in the absence of moistureloss treatment and 80% seedling growth when the treatment hadbeen applied. Moisture loss from soybean seeds appears to bea prerequisite for the synthesis of new proteins which may bepart of the metabolic process or processes that allow the soybeanseed to undergo the transition from seed germination to seedlinggrowth. The relationship between coleoptile elongation and survivalvs. alcoholic fermentation of rice under anoxia is examinedusing eight cultivars differing in submergence tolerance. Anoxiawas imposed on either 1 or 4 d aerated seeds either by N2 flushingsubmerged tissues or by incubating tissues in stagnant deoxygenatedagar at 0·1% w/v; the latter simulated the stagnant conditionsof waterlogged soil. Two cultivars that were most tolerant tosubmergence also had the greatest tolerance to anoxia, whilea submergence intolerant cultivar was also intolerant to anoxia. Coleoptile growth under anoxia was related to rates of ethanolsynthesis (RE), however differences between growth during anoxiaand survival after anoxia indicated that post-anoxic injurymay also be important in rice seeds exposed to anoxia. The correlationbetween coleoptile growth and RE measured on a tissue basisusing intact seeds was r2 = 0·67 among six varietiesover 0-3 d anoxia. This correlation improved to about r2 = 0·85using RE of (embryos plus coleoptiles) over 0-3 d, or coleoptilesat 3 d after anoxia. Coleoptile growth of individual seeds wasusually poorly correlated to RE in these cultivars at 2-3 dafter anoxia. When coleoptiles of similar lengths were obtainedfrom different cultivars using 4 d aerated seeds, there weredifferences in RE and coleoptile growth which were related tocoleoptile growth during 3 or 5 d anoxia, either on a tissue(r2 = 0·85) or a fresh weight basis (r2 = 0·70-0·97respectively). A linear relationship between constant temperatures in the sub-optimaltemperature range and germination rate is shown in both Quercusrobur L and Castanea sativa Mill germinated under nominal darkconditions. The mean base temperature was interpolated for Qrobur as 0 8 ? or 2-4 ?, depending on seed lot provenance, andfor C. sativa as 1 -4? The optimum temperature for germinationin Q. robur was about 20? compared with around 28 ? in C. sativaOver the sub-optimal temperature range the distribution of thermaltimes was log-normal for each population studied their spreadvarying both between Q robur seed lots and between species However,in C. sativa germinated close to the mean base temperature,the distribution in thermal times was reduced Thermal timesto germination were decreased in Q. robur and C sativa by approximately0 3 and 0-5 log-units, respectively, when the pericarp was removed,i.e in the seeds, but the sensitivity of the response remainedrelatively unaltered In both species the germination rate was the same when nominaldark or safe green light conditions were employed during thegermination test. However, at 21 ? Q robur exhibited the highirradiance reaction (HIR) at photon doses above 30mmol m–2d–1. HIR first affected the germination rate by an inhibitionof radicle extension The sensitivity of the response to thermaltime was reduced as photon dose increased. This photo-inhibitionwas exacerbated at supra-optimal temperatures.
