A cobalt-resistant strain of Neurospora crassa (cor) was obtained by repeated subculturing of the wild type on cobalt-containing agar medium. N. crassa cor is twentyfold more resistant to cobalt ions compared with the wild type. Resistance was stable on repeated subculturing of cor on cobalt-free media. N. crassa cor is also cross-resistant to nickel (fourfold), but not to zinc or copper. Higher concentrations of iron and magnesium ions are required to reverse growth inhibition due to cobalt toxicity in N. crassa cor, compared with the wild type. Germinating conidia and mycelia of the cor strain accumulated lower levels of cobalt ions compared with the parent N. crassa. The partial transport block for cobalt uptake is shown to be primarily due to decreased surface binding of cobalt to mycelia and cell walls. Efflux of mycelial cobalt was also observed in wild type and cobalt-resistant N. crassa. The characteristics of cor in comparison with wild type N. crassa are discussed in relation to the mechanisms of cobalt resistance.  相似文献

Summary Electropherograms of Neurospora crassa homogenates showed a polypeptide with a mobility slightly lower than that of a standard sample of clathrin (from bovine brain). Subcellular fractionation of the homogenate resulted in a 20-fold enrichment of the putative N. crassa clathrin in the microsomal fraction. Further fractionation of the microsomal fraction by glass bead permeation chromatography yielded a fraction enriched about 150-fold relative to the homogenate. Coated vesicles (42.5 ± 2.5 nm diameter) were found in this preparation by electron microscopy of negatively stained specimens. Ribosomes were virtually absent from this sample. N. crassa clathrin remained associated with the coated vesicles after repeated centrifugation and homogenization steps, even in the presence of 0.4 M-NaCl, but was released by treatment with Tris buffer pH 8.5. However the polypeptide was again sedimentable after dialysis against Mes buffer pH 6.5. Under the electron microscope this sediment resembled the empty coats of higher eukaryotes. The results taken together indicate that a clathrin-like protein occurs in wild type cells of N. crassa.  相似文献

Uptake of Co2+ by cobalt-resistant strain is dependent on Co2+ concentration in the medium and is linear with time. The uptake is unaffected by metabolic inhibitors and decreased at low pH values. The uptake is independent of temperature in the range 0–40‡ C. The transport system is a passive diffusion process, unlike in the parent wild type strain where it is energy-dependent. It is possible that Mg2+ transport system is not involved in Co2+ transport in this strain, since the Co2+ uptake is not suppressed by Mg2+ as in the parent strain.  相似文献

Three non-identical Zn-resistant strains of Neurospora crassa have been isolated. ZNR-1 and ZNR-2 strains were obtained after repeated subculturing of wild type N. crassa on Zn-containing agar media (8mM and 16mM), while ZNR-3 was isolated after mutagenesis with diethyl sulfate, followed by selection on Zn agar plates (16mM). All three ZNR strains showed two- to threefold resistance to Zn in liquid media when compared with the wild type. However, growth measured by hyphal elongation clearly distinguished between the resistant strains (ZNR-3>ZNR-2>ZNR-1wild). The ZNR-2 and ZNR-3 strains were also cross-resistant to Co, while ZNR-2 alone was cross-resistant to Cu. Both Mg and Fe reversed the growth inhibition caused by Zn; Mg by suppression of Zn uptake and Fe without affecting the same. Assay of catalase, iron-binding siderophores and glutathione in Zn toxicity revealed significant increases in catalase and glutathione levels in the ZNR-2 strain when compared with the wild type. Kinetics of Zn uptake by preformed mycelia showed a rapid initial phase of uptake followed by a slower phase. The rates of Zn uptake measured after leaching surface-bound metal with EDTA revealed that ZNR strains have significantly reduced Zn uptake rates when compared with the wild type. The overall data suggest a partial transport block for Zn uptake as the major mechanism for resistance in ZNR strains. Genetic analysis of ZNR strains showed that in the ZNR-3 strain the znr locus maps close to the mating type locus (mt) of N. crassa LG I, while that of ZNR-1 and ZNR-2 is linked to LG IV associated with chromosomal aberration.  相似文献

A cobalt-resistant wall-less mutant (slime) of Neurospora crassa was obtained by repeated sub-culturing of the sensitive wall-less mutant (W-sl) on agar medium containing toxic concentrations of cobalt. Resistance was stable on culturing Cor-sl on cobalt-free medium up to 15 weekly subcultures. Cor-sl is 10-fold more resistant to cobalt when compared to W-sl. It is also cross-resistant to Cu (10-fold) and Ni (3-fold). Cobalt accumulated by Cor-sl during growth and in short-term uptake experiments was lower when compared to W-sl. Cells previously loaded with cobalt was released into medium in both mutants, while in case of Cor-sl most of cobalt taken up (>80%), was released back into the medium when compared to W-sl. Metabolic inhibitor (Sodium azide) and magnesium ions inhibited cobalt uptake in both the mutants. Fractionation of cell-free extracts showed that most of the cobalt (70%) taken up by Cor-sl was bound to an inducible protein fraction which bound to DEAE-Cellulose, while in W-sl only 20% of cobalt was associated with this fraction. Subcellular localization of cobalt in W-sl indicated most of it to be cytoplasmic (70%) while nuclei and mitochondria had 10% and 5% respectively. In case of Cor-sl, mitochondrial cobalt accounted for only 2% while no significant differences were noted for other fractions. Our data implicate both transport block and intracellular sequestration of cobalt to play a major role in resistance.  相似文献

Energy metabolism and mitochondria have been discussed with respect to their role in the circadian rhythm mechanism for some time. Numerous examples of inhibitors that affect the mitochondria of plants and animals and microorganisms are known, which cause large phase shifts in the rhythms of these organisms. Analogous studies on the role of mitochondria in the Neurospora circadian rhythm mechanism have also been reported and summarized. This communication differs from previous studies on other organisms in that it will focus on two lines of evidence derived from studies on Neurospora strains carrying mutations affecting the mitochondria, (a) Strains whose growth rate is resistant to oligomycin (olit) owing to an altered protein in the F0 sector of the mitochondrial ATPase, showed no phase shifts when pulsed with oligomycin. Control strains (oli8) showed large phase shifts when pulsed with oligomycin. This indicates that the phase-shifting effect of oligomycin is due to the direct inhibition of the mitochondrial ATPase and not some side effect of this inhibitor, (b) In Neurospora, many different strains are known that carry mutations in the nuclear or mitochondrial genome that affect mitochondrially localized proteins. Some of these, such as oli', [MI-3], or cya-5, showed shorter (≥ 19-h) periods compared with the normal (21.5-h) period. Others showed little or no change in period. Those mutant strains exhibiting shorter periods also contained ≥60% more mitochondrial protein per gram total protein in extracts compared with the normal strains. Assays of the level of a mitochondrial-specific protein, acyl carrier protein, showed that the cellular content of this protein was approximately doubled. A parallel set of studies on the effects of antimycin or chloramphenicol on Neurospora demonstrated that these inhibitors also produced shorter periods as well as increased amounts of mitochondrial proteins. These two new lines of evidence may be interpreted to indicate that in Neurospora either some part of the oscillator is localized to the mitochondria and/or that mitochondria exert their effect on the clock mechanism through their effects on biosynthetic pathways or by their contribution in determining ion gradients.  相似文献

A gene (Ncphy) encoding a putative phytase in Neurospora crassa was cloned and expressed in Pichia pastoris, and the biochemical properties of the recombinant protein were examined in relation to the phytic acid hydrolysis in animal feed. The recombinant phytase (rNcPhy) hydrolyzed phytic acid with a specific activity of 125 U mg-1, Km of 228 micromol L-1, Vmax of 0.31 nmol (phosphate) s-1 mg-1, a temperature optimum of 60 degrees C and a pH optimum of 5.5 and a second pH optimum of 3.5. The enzyme displayed pH stability around pH 3.5-9.5 and showed satisfactory thermostability at 80 degrees C. The phytase from N. crassa has potential for improving animal feed processing at higher temperatures.  相似文献

Chitin synthetase activity in cell-free preparations from a wild-type strain and a 'slime' variant of Neurospora crassa was monitored over many days in samples stored at 0 degrees C. Total activity in whole-cell-free extracts and low-speed supernatants from both organisms was very unstable, losing more than 90% of the initial activity on storage at 0 degrees C for 96 h. Chitin synthetase detection was not masked by chitinase activity present in the preparations. Gel-filtration chromatography of these preparations increased the stability of the activity from the 'slime' variant, whereas removal of particulate structures by high-speed centrifugation stabilized the chitin synthetase activity in the supernatant, particularly in the wild type. These results suggest that factor(s) involved in the regulation of chitin synthetase may be differentially located or altered in 'slime' cells.  相似文献

【背景】粗糙脉孢菌LY03是从武平传统"红菌豆腐"中分离得到的主要发酵菌株。【目的】研究粗糙脉孢菌LY03菌株的基因组信息,揭示武平传统"红菌豆腐"发酵特性。【方法】采用形态学观察、ITS鉴定、重测序及框架图测序对所分离的LY03菌株进行鉴定和基因组信息解析。【结果】武平传统"红菌豆腐"中分离得到的主要发酵菌株LY03确定为粗糙脉孢菌,将其在中国普通微生物菌种保藏管理中心进行专利保藏,保藏号为CGMCC3.19233。LY03菌株比对到参考基因组上的总Read数目为95.85%,测序对应深度的位点占全基因组76.13%;各变异类型在内含子区域均无变异,主要变异存在于基因组的外显子区域,具体变异数量为:单核苷酸多态性位点(single nucleotide polymorphism,SNP)位点变异总数203128个、插入缺失(insertion/deletion,In Del)突变总和26859个、拷贝数变异(copy number variation,CNV)增加和减少的拷贝总数1 039个、结构变异(structure variation,SV)注释的变异总数777个;LY03菌株...  相似文献

Fungi are capable of potentially unlimited growth. We resolved nuclear types from multinuclear mycelium of a phenotypically normal wild isolate of the fungus Neurospora intermedia by plating its uninucleate microconidia and obtained a strain which, unlike the "parent" strain, exhibited clonal senescence in subcultures. The mutant gene, senescent, was introgressed into N. crassa and mapped four map units to the right of the his-1 locus on linkage group VR. senescent is the first nuclear gene mutant of Neurospora derived from nature that shows the death phenotype. Death of the sen mutant occurred faster at 34 degrees C than at 22 or 26 degrees C. Measurements of oxygen uptake of conidia using respiratory inhibitors and the spectrophotometric analyses of mitochondrial cytochromes showed that in sen cultures grown at 34 degrees C, cytochromes b and aa(3) were present but cytochrome c was absent. By contrast at 26 degrees C, cytochromes b and c were present but cytochrome aa(3) was diminished in the late subcultures. This suggested that the sen mutation does not affect the potential to produce functional cytochromes. The deficiency of the respiratory chain cytochromes may not be the cause of death of the sen mutant because the cytochrome c and aa(3) mutants of N. crassa are capable of sustained growth whereas sen is not. Possible explanations for the observations are discussed.  相似文献

Summary Slow-growing (inl +/-) spontaneous mutants have been isolated from an inositol requiring (inl) strain of Neurospora crassa that produces defective myo-inositol-1-phosphate synthase (MIPS), the enzyme responsible for the production of inositol-1-phosphate from glucose-6-phosphate. The defective enzyme has some residual activity. In the inl +/- strain the synthesis of the defective enzyme is enhanced, which enables the strain to grow slowly on minimal medium. The mutation (opi1) responsible for the partial inositol independence segregates independently from the inositol locus, and suppresses the inositolless character by overproduction of defective MIPS. opi1 acting upon the wild type (inl +) allele increases MIPS production and causes inositol excretion.  相似文献

A sharp decrease in oxygen uptake occurred in Neurospora crassa cells that were transferred from 30 degrees C to 45 degrees C, and the respiration that resumed later at 45 degrees C was cyanide-insensitive. Energization of mitochondria, measured in vivo with fluorescence microscopy and a carbocyanine dye, also declined sharply in cells at 45 degrees C. Electron microscopy showed no changes in mitochondrial complexity; however, the cytoplasm of heat-shocked cells was deficient in glycogen granules.  相似文献

粗糙脉孢菌是天然纤维素降解真菌,具有产纤维素酶能力,国内外对其纤维素降解机理和发酵产酶有一定的研究,但对其产酶的条件优化研究得不多,其产酶潜力需要进一步挖掘。以粗糙脉孢菌基因组测序菌株FGSC 2489为对象,采用响应面分析法对Neurospora crassa摇瓶发酵产纤维素酶进行培养基优化。采用Plackett-Burman(PB)实验设计考察发酵培养基中关键参数对产酶条件的影响,进而采用最陡爬坡实验逼近最大响应区域,并结合中心组合实验(central composite design,CCD)和响应面分析法对两个显著因素进行分析。PB实验结果显示:Peptone、Yeast extract对产纤维素酶有显著影响。通过响应面分析得到一元二次方程,对方程求解得到Peptone 7.27g/L、Yeast extract 5.51g/L。采用该优化培养基,最大纤维素酶活可达1.27FPU/ml,较优化前提高了2.03倍;CMC酶活14.15IU/ml,比优化前提高1.88倍;木聚糖酶活24.13IU/ml,比优化前提高1.86倍;葡萄糖苷酶酶活1.22IU/ml比优化前提高2.08倍。  相似文献

1. Increasing concentrations of cobalt in the medium result in increased production of an iron-binding compound and a corresponding fall in catalase activity of Neurospora crassa. 2. Cobalt rapidly depletes the medium of iron by enhancing the rate of iron uptake by the mycelium. 3. With toxic amounts of cobalt there is a fall in bound (59)Fe and haem (59)Fe as well as a decreased incorporation of [2-(14)C]glycine into the mycelial haem fraction. The production of the iron-binding compound precedes the fall in the iron-dependent systems mentioned. 4. The (59)Fe bound to the iron-binding compound acts as a better iron source for haem synthesis in cell-free extracts as compared with (59)FeSO(4). 5. Cobalt inhibits iron incorporation into protoporphyrin in cell-free extracts but is not itself incorporated to an appreciable extent.  相似文献

以粗糙脉胞霉CICIM F00021染色体DNA为模板,通过PCR扩增得到粗糙脉胞霉酸性蛋白酶结构基因。对其序列进行了测定和分析,表明扩增得到的片段为酸性蛋白酶基因。将扩增出的酸性蛋白酶基因克隆入酵母表达载体中获得重组质粒pPIC9K-ap。将其转化入毕赤氏酵母获得重组菌NA3。重组酸性蛋白酶在pH4.0和45℃下表现出最高活性。  相似文献

We have enriched a 47-kDa polypeptide (p47) from Neurospora crassa on the basis of its affinity to calmodulin. The p47 was purified to homogeneity by chromatography on a Mono S cation exchange column and evidence is presented that the polypeptide co-sediments specifically with F-actin. The intracellular distribution of p47 and actin was also examined using indirect double immunofluorescence staining of cells at different stages of development. Our results suggest that by altering the conformation binding site of actin to p47, calmodulin could play a regulatory role in the polarized hyphal growth of N. crassa.  相似文献

Abstract Highly specific antibodies against calmodulin from Neurospora crassa were produced in rabbits. These antibodies were immunopurified by chromatography on Neurospora calmodulin-Sepharose, and had a titer for purified calmodulin from Neurospora crassa and bovine testis of 80 ng/ml and 2.5 μg/ml respectively. By immunoblot, as little as 8 ng of pure Neurospora calmodulin could be detected, and the antibodies revealed calmodulin in crude homogenates from Neurospora crassa and Aspergillus nidulans while in homogenates from Allomyces arbuscula, Saccharomyces cerevisiae, Dictyostelium discoideum and bovine testis, calmodulin remained undetected.  相似文献

Abstract The effects in vitro of chloramphenicol on the mitochondrial electron transport of Neurospora crassa were investigated under phosphorylating, non-phosphorylating and uncoupled conditions. The antibiotic acted at more than one site of the respiratory chain: exogenous NADH dehydrogenase, Complex I, the rotenone-resistant by-pass and the alternative oxidase, although an unspecific effect could not be totally excluded.  相似文献

Cyclic nucleotide phosphodiesterase has been partially purified by calmodulin-Sepharose affinity chromatography from a soluble extract of Neurospora crassa. The phosphodiesterase activity remained bound to the affinity column even in the presence of 6 M urea and could only be eluted by calcium chelation. The enzyme exhibits cAMP and cGMP phosphodiesterase activities. Both activities can be enhanced by calmodulin in a Ca2+-dependent manner. Stimulation of cyclic nucleotide phosphodiesterase by calmodulin can be inhibited by calmodulin antagonists such as pimozide, trifluoperazine and chlorpromazine.  相似文献
