Prephenate dehydratase has been purified 10,000-fold from the crude extracts of Bacillus subtilis. The procedure takes advantage of the dissociation of the enzyme to a 55,000-dalton form in the presence of the negative effector, phenylalanine, and its association to a 210,000-dalton form in the presence of the positive effector, methionine. These two forms of the enzyme were separated from the bulk of the other proteins present in the crude extracts by gel filtration alternately in the presence of the two effectors. Sodium dodecyl sulfate electrophoretic analysis showed the enzyme is composed of apparently identical 28,000-dalton polypeptides.  相似文献

The ecological and biogeochemical relevance of hydrolytic enzymes associated with the fungal cell wall has been poorly studied in ectomycorrhizal (ECM) fungi. We used a modified sequential extraction procedure to investigate the activity of various hydrolytic enzymes (β-glucosidase, acid-phosphatase, leucine-aminopeptidase, chitinase, xylanase and glucuronidase) and their association with the cell wall of three ECM fungi (Rhizopogon roseolus, Paxillus involutus and Piloderma croceum). Fungi were grown on C-rich solid medium under three different P concentrations (3.7, 0.37 and 0.037 mM). The sequential extraction procedure classifies enzymes as: (a) cytosolic, (b) loosely bound, (c) hydrophobically bound, (d) ionically bound and (e) covalently bound. Results showed that for the same fungus absolute enzymatic activity was affected by P concentration, whilst enzymatic compartmentalization among the cytosol and the cell wall fractions was not. The association of enzymes with the cell wall was fungus- and enzyme-specific. Our data indicate also that enzymes best known for being either extracellular or cytosolic or both, do act in muro as well. The ecological implications of cell wall-bound enzymes and the potential applications and limitations of sequential extractions are further discussed.  相似文献

A simple and effective cryogenic procedure for the extraction of glycolytic intermediates from whole Drosophila has been developed. This procedure gives consistent results when a measure (µM/liter/OD260) is adopted which corrects for differences in extraction efficiency. Using this measure and a homozygous strain of D. mercatorum, there are no significant differences among extracts for the levels of any of the 15 glycolytic intermediate or energy molecules considered. The profile of means is consistent across experimental designs and instrument types. Coefficients of variation are well below 50% for most variables. The methodology presented has the statistical power to detect a mean change of 10 to 50% using an experimental design which requires as few as 32 observations. The estimated energy charge for resting Drosophila from these studies is the expected value of 0.86.  相似文献

Definitions are proposed for the independent and joint contributions that the chemical groups A and B make to the free energy of association of the ligand A?B with a receptor. The definitions are independent of the choice of the standard state and are consistent with the basic thermodynamic cycle relating the association of the ligands A?B, A?Y and X?B to the receptor Rappaport 1976. The basic idea is the use of the excess free energy of association of the ligand A?Y over the free energy of association of the reference ligand X?Y as the measure of the “independent” contribution of the group A to the binding. This definition allows the free energy of association of the ligand A?B to be written as the sum of the independent contributions of the groups A and B, their joint contribution, and an invariant free energy of association of the reference ligand with any receptor. With the appropriate definition of the receptor-reference ligand complex, water can be chosen as the reference ligand. Using ΔG(A?OH)?AG(HOH), ΔG(H?B?H)?ΔG(HOH) and ΔG(HO?C)?ΔG(HOH) as the definitions of the “independent” contributions of the chemical groups A, B and C to the binding of the ligand A?B?C, the joint contribution of the groups A and C to the binding is ΔG(A?B?C) ? A rapid and quantitative procedure is described for the re-N-acetylation of amino sugar methyl glycosides prior to their analysis by gas-liquid chromatography. Two equivalents of pyridine are added to acidic methanolysates containing amino sugars, serving both to neutralize the acid and to act as a catalyst for the subsequent N-acetylation reaction with acetic anhydride. The N-acetylation is quantitative and complete within 10 min at ambient temperature. Excess acetic anhydride is destroyed by solvolysis with the methanolic solvent. The procedure has been used effectively for methanolysates containing 0.01–2.0 mg/ml glucosamine. The procedures utilizing ion-exchange columns and insoluble salts are thus circumvented and all reaction byproducts are volatile. The procedure is therefore ideally suited for the simultaneous workup of numerous samples for analytical procedures such as gas-liquid chromatography.  相似文献

Due to its high nutritional value, chickpea is one of the most important and cost-effective legumes for human diet. Nutrient elements, such as Cu, P, K have numerous essential functions for the human metabolism. In this study, association mapping of loci linked to the seed Cu, P and K concentrations were performed on a population consisting of 107 Cicer reticulatum and 73 Cicer arietinum individuals in four environments (two locations x two years). A total of 121,840 SNPs were genotyped across 180 individuals by GBS analysis. The association mapping between the SNP markers and the seed Cu, P, K concentrations were identified and eight SNPs were found to be significantly associated with variations in three nutrient elements in more than two environments This research suggests that association mapping is a useful methodology for the identification of loci controlling the Cu, P and K uptake in chickpea seeds for further association mapping, molecular breeding, and marker-assisted selection and plant breeding studies and provides a broader understanding of the relationship between the investigated Cicer species and the effects of environmental conditions.  相似文献

Photosynthesis in the Azolla-Anabaena association was characterized with respect to photorespiration, early products of photosynthesis, and action spectra. Photorespiration as evidenced by an O2 inhibition of photosynthesis and an O2-dependent CO2 compensation concentration was found to occur in the association, and endophyte-free fronds, but not in the endophytic Anabaena. Analysis of the early products of photosynthesis indicated that both the fern and cyanobacterium fix CO2 via the Calvin cycle. The isolated endophytic Anabaena did not release significant amounts of amino acids synthesized from recently fixed carbon. The action spectra for photosynthesis in the Azolla-Anabaena association indicated that the maximum quantum yield is between 650 and 670 nanometers, while in the endophyte the maximum is between 580 and 640 nanometers. Although the endophytic cyanobacterium is photosynthetically competent, any contribution it makes to photosynthesis in the intact association was not apparent in the action spectrum.  相似文献

A one-step procedure using a mixture of glutaraldehyde and osmium tetroxide was devised to fix in situ large unilamellar liposomes of phosphatidylserine for transmission electron microscopy (TEM), since the conventional fixation method was found to be inadequate in this respect. The new fixation procedure enabled us to visualize the sequence of events in the interaction of liposomes with protoplasts from Vinca rosea suspension cultures in the presence of polyethylene glycol. Liposomes were thus found adhering to the surface of protoplasts, in association with invaginating plasmalemma, and within intracellular vesicles. These observations showed that liposomes enter plant protoplasts via endocytosis. Ultrastructural profiles indicating fusion of liposomes with protoplasts were not observed.  相似文献

The floral biology ofNuphar lutea subsp.macrophylla, native to Central Texas, has been investigated. The flowers are protogynous with anthesis occurring over a period of several days. Flowers are visited by a number of insects but are effectively pollinated by the beetleDonacia piscatrix Lac., which spends its entire life history in association withNuphar. Evidence is assembled which suggests that the overall floral structure together with the sequence of floral development and the timing of sepal movements (i.e., the opening and closure of the flower) are primary adaptations to assure beetle pollination.  相似文献

In vitro measurements show that the X86 repressor, which has an increased affinity for the lac operator as compared to wild-type repressor, also has an increased affinity for non-operator sites on Escherichia coli DNA. The rate constant of association of repressor and operator is decreased by E. coli DNA fivefold more for X86 repressor than for wild-type repressor. Low inducer concentrations increase the rate of association of X86 repressor and operator in the presence of E. coli DNA. In a partial equilibrium situation where part of the X86 repressor is bound to the operator, and part to either non-operator sites on E. coli DNA or to an Oc operator, the formation of complexes between X86 repressor and wild-type operator is favored by low inducer concentrations. Repression of the lac enzymes increases drastically in the X86 mutant in the absence of DNA synthesis in vivo. A new explanation for the in vivo characteristics of the X86 mutant is suggested.  相似文献

Procedures for the determination of equilibrium constants of linear and helical associations based on molecular weight data have been derived. Theoretical construction of curves for M1/Mwapp vs. C reveals two clearly identifiable patterns for linear and helical association. The change in molecular conformation between linear and helical association is evidenced by a recurring pattern of irregular curves which obviously differ from those of an indefinite stacking model.  相似文献

The influence of magnesium ions, hydrogen ions, and oxygen on the monomer (17 S)-tetramer (39 S) equilibrium of the hemocyanin from Callianassa californiensis has been investigated. Data have been interpreted in terms of a theory integrating the allosteric equilibria and association equilibrium. Binding of oxygen and divalent cations by the hemocyanin has also been studied in terms of the theory, and the parameters in the model have been determined. The 17 S monomer, which contains six polypeptide chains, is found to be the allosteric unit which behaves as a self-contained co-operative system with allosteric properties. Both magnesium and hydrogen ions are shown to affect the association directly, whereas the effect of oxygen binding can be explained in terms of a difference in the allosteric equilibrium constant L′ in the different associated states of the protein. It is shown that whereas the effect of oxygen on the monomer-tetramer equilibrium is easily observable, the converse effect of the association equilibrium on the oxygen binding curve is at the limit of detectability.  相似文献

The influence of the two operator-like regions lying within or near the lac regulatory region on the binding of lac repressor to lac operator has been investigated. λdlac phages deleted either for the “second operator” in the beginning of the Z gene or deleted for the “third operator” at the end of the I gene were constructed. In in vitro binding experiments it could be shown that the deletion of secondary repressor binding sites from the lac regulatory region does not significantly alter the stability of the repressor—operator complex. Measuring the rate constant of association of repressor with operator in the presence of a 150-fold excess of unspecific DNA, we observed a concentration-dependent effect of the unspecific DNA, although the ratio of operator to non-operator DNA was kept constant. A small effect of the secondary binding sites is seen on the rate of association of repressor with operator, indicating that the secondary binding sites might play a role in facilitating association of repressor with operator under in vivo conditions.  相似文献

Spinal ganglia from 4- to 7-day [Stage 23–30; Hamburger and Hamilton (1951) J. Morphol.88, 49–92] chicken embryos were cultured in vitro to investigate the effect of various environmental conditions on cell differentiation. Culture morphology (i.e., degree of dispersion of the explanted ganglia, survival of neurons, and outgrowth of axons) was observed to depend upon several factors including: (1) the age of the explanted ganglia, (2) the presence or absence of nerve growth factor (NGF), and (3) the nature of the substratum on which the cultured tissue resides. These observations enabled us to disturb the association of neurons with the other cells in ganglion cultures and thereby modulate the differentiation of adventitious melanocytes. Thus, in medium permissive for melanogenesis, melanocytes appear when the association between neurons and small stellate nonneuronal cells in the ganglion is disrupted. This disruption is most extensive (1) when young (Stage 26–27, 5-day) ganglia are explanted on plastic substrata, in the initial absence of NGF, and (2) when cells from enzyme-dissociated ganglia are cultured on plastic substrata. In comparable media, pigment cell differentiation is not observed when the association between neurons and small stellate cells is preserved. Such associations tend to endure (1) in developmentally older (Stage 30+, 7- to 8-day) ganglia or (2) when ganglia are cultured on agar or fibroblast substrata. We conclude that loss of association between neurons and the nonneuronal cells in young ganglia is necessary for the latter to undergo melanogenesis in vitro.  相似文献

A greatly simplified procedure is proposed which employs C= f(r) as determined from sedimentation equilibrium measurements in graphical analysis of self-associating protein systems and in the enumeration of interacting species in the ultracentrifuge. Basic equations given here are applicable to any self-associating system. A simple and rapid procedure based on the gel filtration principle is described together with its applicability to the study of protein-protein interactions including subunit-subunit and enzyme-enzyme interactions. Using this procedure, it is shown that phosphoglycerate kinase (PGK) and glyceraldehyde-3-phosphate dehydrogenase (GPDH) interact with a stoichiometry of one PGK molecule combining with one monomeric subunit of GPDH. This interaction has been observed with both enzymes being from the same, as well as from different, species. The Kd values for rabbit muscle PGK and porcine muscle GPDH complex and that for the rabbit muscle PGK and yeast GPDH complex are found to be (4.5 ± 2.0) × 10−7 M and (6.5 ± 1.7) × 10−7 M, respectively. The specificity of bienzyme association is stronger when enzymes are from the same species than when they are from different species.  相似文献

A standardized survey of respiratory and cardiovascular findings was conducted among male telephone workers in Baltimore, Maryland who were over 40 years of age. Sputum produced within 1 hour after arising was requested as part of the examination. All but 1 of the 246 eligible employees returned the specimen bottles, but only 103 were able to produce sputum in time for examination. All suspected fungal growth was isolated and insofar as possible identified as to genus. Some fungus was isolated from 83 of the 103 specimens.Penicillium was found in about 40 per cent of the specimens,Hormodendrum in about 30 per cent, andAlternaria in about 10 per cent. The association of chronic wheezing and subsequent respiratory illness with the presence ofHormodendrum orAlternaria in the sputum was sufficiently marked to suggest further investigations of this association for possible etiologic relationships.  相似文献

The genus Multifurca is recorded for the first time from Australia. Multifurca stenophylla (Berk.) T.Lebel, C.W.Dunk & T.W.May comb. nov. is described and illustrated, and a lectotype and epitype designated. The species is characterized by the association with Nothofagus and Eucalyptus, the pale yellow, concentrically zoned pileus, abundant acrid white latex which becomes pale yellow then eventually greenish, and the small basidiospores.  相似文献

Adults and deutonymphs of a new water mite species of the genus Hygrobates Koch, 1837 (Acari, Hydrachnidia, Hygrobatidae) were found to parasitize the Hong Kong Newt, Paramesotriton hongkongensis Myers and Leviton, 1962 (Amphibia, Caudata, Salamandridae), endemic to Hong Kong and coastal areas of Guangdong Province: Hygrobates aloisii sp. nov. Males and females, as well as deutonymphs and larvae are described. A new subgenus - Lurchibates subgen. nov. - is defined to accommodate the species of the Hygrobates salamandrarum-group. The parasite-host association, infestation rate, attachment sites and the impact of mite infestation on the newts behaviour and body condition are investigated. These ecological aspects of the parasite-host association between freshwater mites and newts are examined for the first time in detail in their natural habitat.  相似文献
