Pars intermedia (PI) tissue from fetal, perinatal, neonatal and juvenile rabbits has been maintained in organ culture for up to nine weeks after explantation. Autoradiography showed that DNA synthesis took place for at least 22 days of culturing. PI-glandular cells and interstitial cells remain identifiable throughout this period but ACT-type cells were recognised only up to six weeks. Material from fetal and perinatal animals had a higher proportion of surviving cells than that from adult animals. The degree of differentiation achieved by PI-glandular cells in vitro appears to depend on three factors: i) the stage of development reached before explantation; ii) the original topographic position in the PI tissue before explantation; and iii) the position in the explant in relation to the gas-liquid interphase.  相似文献

Dynamics of ultrastructural alterations of Paneth cells (PC) of the rat jejunum have been investigated after bilateral subdiaphragmal trunkal vagotomy (7, 14, 30 and 60 days). This operation results in a profound ultrastructural rearrangement in PC organization, which is especially manifested on the 7th and 14th days. The cisterns and canaliculi of the endoplasmic reticulum dilate, the Golgi complex structure is disturbed, the matrix of mitochondria is cleared, their crists are fragmented, large vacuoles with electron transparent contents appear, total amount of the secretory granules and relative number of their immature forms increase, the secreory granules form and topography of their core is changed, small structures resembling microtubules appear in them. Beginning from the second week, morphological alterations of compensatory-adaptive character progressively increase: the number of free ribosomes grows large and large mitochondria appear.  相似文献

Cell-mediated immune responses in mice infected with fonsecaea pedrosoi   总被引:4,自引:1,他引:3  
Time course of cellular and humoral immune responses in mice infected with Fonsecaea pedrosoi was investigated by using an antigen prepared from culture filtrate of this fungus. Mice were infected by intravenous injection with yeast-like cells of the fungus. Viable fungus was recovered from the brain of the infected mice until the 36th day after inoculation, and from the other organs examined until 14th to 16th day. Inflammatory lesions were observed in the brain, lung, heart, liver, spleen, kidney and intestine during the first 30 days after inoculation. Macrophage migration inhibition factor response in these mice was insignificant until 8 days after inoculation. A significant response was developed at day 10 and persisted until day 63. This response returned negative by 95 days after inoculation. Lymphocyte transformation response of these mice was negative until 4 days after inoculation. At day 6 blastogenic index increased to 1.5, and at day 10, 14 and 16 the indices were 1.8, 2.4 and 1.7 respectively. Precipitin response to this fungus could not be detected in these mice until 16 days after inoculation. Positive results were obtained at day 21 and lasted until 51 days after inoculation. The precipitin titers, however, did not exceed one fold in any of these mice.  相似文献

Lactating Friesian dairy cows (2nd-4th parity) which calved in spring (N = 7) or autumn (N = 15) were used. Their ovaries were examined by ultrasound scanning and blood samples were obtained daily for progesterone and oestradiol concentrations from the 5th day after calving until the first post-partum ovulation occurred. Five autumn-calving cows selected at random were bled every 15 min over a 6-h period on 1 day each week for 4 weeks after calving to assess the patterns of LH secretion. Follicular development during the post-partum anoestrous period was characterized by the growth and regression of small (less than or equal to 4 mm) and medium-sized (5-9 mm) follicles, until a dominant follicle (greater than 10 mm) was detected. The first detected dominant follicle ovulated in 14 cows, became cystic in 4 cows (all in autumn), and failed to ovulate in 1 cow. It was not possible to detect a dominant follicle in 3 cows due to scanning difficulties. The post-partum interval to detection of the first dominant follicle (mean +/- s.d.) was shorter (P less than 0.05) in autumn (6.8 +/- 1.8 days) than in spring (20 +/- 10.1 days). However, there was no significant difference between the respective intervals to first ovulation (autumn 27.4 +/- 25.9 and spring 27.3 +/- 18.9 days). Autumn-calved cows which had cysts had longer (P less than 0.001) intervals to first ovulation (58.2 +/- 23.5 days) than did normal cows (12.0 +/- 2.5 days). All cows with cysts had twin ovulations at their first post-partum ovulation. A pulsatile pattern of LH secretion was detected in the first week post-partum and LH pulse frequency was 2-3 per 6-h period in Weeks 1 and 2 post partum and increased to 5-7 pulses per 6-h period in the presence of a dominant or cystic follicle. Concentrations of progesterone in plasma during post-partum anoestrus were usually low (less than 0.2 ng/ml); oestradiol concentrations were also low (less than 5 pg/ml), but higher values (5-110 pg/ml) were observed in cows that had a dominant or a cystic follicle.  相似文献

Mouse urothelium is disrupted just before birth, followed by a postnatal restoration process which includes cell proliferation, death and differentiation. We assessed urothelial proliferation by the expression of proliferating cell nuclear antigen (PCNA), desquamation by electron microscopy, and apoptosis by TUNEL staining and urothelial differentiation by the expression of uroplakins and cytokeratin 20 (CK20) as well as the apical plasma membrane maturation. Our results indicated that urothelial proliferation was high from birth until about the 14th postnatal day. A majority of basal cells and even occasional superficial cells were PCNA positive during the first 5 postnatal days. Cell death occurred during the first 9 postnatal days. Between birth and day 5, single cells underwent apoptosis, whereas between days 6 and 9 cells mainly desquamated. CK20 and uroplakins were expressed in all superficial cells in postnatal urothelium. Their subcellular distribution characteristically changed in accordance with the progressive differentiation of superficial cells. During the urothelial postnatal development, proliferation activity slowly decreases to the proliferatively quiescent urothelium of the adult animal. Apoptosis is present in the first 9 postnatal days and within a few days of this period it appears simultaneously with desquamation. Superficial urothelial cells gradually differentiate, which is reflected in the changeable morphology of the apical plasma membrane.  相似文献

At 5 days post conceptionem (p.c.) shortly after implantation, giant cell transformation starts at the abembryonic pole of the blastocyst, spreading over the mural trophoblast; 1 day later, the first ectoplacental giant cells appear at the base of the fast growing ectoplacental cone (derived from the polar trophoblast). Giant cell transformation expands over it periphery. Thus, by the 8th day p.c., the conceptus is separated from the maternal tissue by a continuous layer of giant cells, variable in thickness. Giant cells reach their greatest size by 10 days p.c. in the mural tophoblast and by 12 days p.c. in the chorioallantoic placenta. They are probably no longer formed after that stage. Around the 8th day p.c., the allantois reaches contact with the ectoplacental cone, which develops into the chorioallantoic (definitive) placenta. At 9 days p.c., its four zones can already be discriminated: chorionic plate, labyrinth, junctional zone (trophospongium), and zone of giant cells, respectively. Within the next day, the chorioallantoic placental circulation is established. The yolk sac placental circulation is established by the 9th day p.c. The villi of the proximal layer of the yolk sac increase in size and number, and their capillary network becomes more dense until the 12th to 14th day p.c. This provides evidence that the yolk sac placenta exerts its function--to a certain extent--beyond the establishment of the definitive placenta. Around the 14th day p.c., the placental labyrinth reaches its definitive features. Fetal capillaries in the labyrinth, branching from unbilical blood vessels within the septa of connective tissue are surrounded by trophoblast cells. They form a dense vascular network bathing in maternal blood. The structures of the placental zones remain almost the same during further development, the borders becoming sometimes little blurred. Adjacent to the chorionic plate, subchorionic clefts appear at the 14th day p.c. These clefts become confluent to form the intraplacental space, regularly communicating with the yolk sac cavity. At the end of gestation (19th day p.c.) there is a considerable amount of eosinophilic material ('fibrinoid') between the zone of giant cells and the decidua, probably produced by the giant cells.  相似文献

Parturitions were induced in five cows, 2 weeks before term using prostaglandin (PG) F(2alpha). Two i.m. injections were performed with an interval of 24 h. All cows calved within 5 days (average 2.7 days) after the first injection of PGF(2alpha). Out of five cows, four had retained fetal membranes (RFM). Each animal was sampled for bacteriological examination using uterine biopsies twice a week during 42 days postpartum (PP). Jugular vein blood samples were withdrawn for PGF(2alpha)-metabolite and progesterone analyses five times per day during the first week PP and eight times per 24 h during the 2nd and 3rd weeks PP. From the 4th week, the sampling interval was reduced back to five times per day. From the 5th week PP, the sampling was reduced to two times per day and sampling was terminated after day 46 PP. Only morning samples were used for progesterone analyses. From day 10 PP, ultrasonography (US) was performed every 3rd day until day 39 PP for detection of ovarian activity and follicular dynamics. The highest incidence of bacteriological species was found during the first 3 weeks PP. After the 5th week of collection, all animals were free from bacteria. The species of bacteria found were Arcanobacterium (Actinomyces) pyogenes, Escherichia coli, alpha-hemolytic streptococcae and Pasteurella multocida. Immediately after parturition, very high levels of the PG-metabolite were seen in all animals, with a sharp decrease to line of significance around days 9-12 PP. Small increases above the line of significance were detected up to day 27 PP in cows with RFM, and after that time the levels were considered to be at baseline. Low levels of progesterone were seen in four animals during the whole experimental time. In one animal, an increase was seen on day 43 PP, which was maintained until the end of the experimental period on day 46 PP. Based on US, follicular waves were detected in all animals during the experimental period. In three animals, three non-ovulatory follicular waves were detected and in two animals, four non-ovulatory follicular waves were detected during 39 days of ultrasound sessions. Based on progesterone levels, only one animal was considered to have ovulated around day 40 PP. Results from the present study indicate that reproductive performance of cows after PG-induced parturitions differs from those of spontaneous cases of RFM. Differences regarding the resumption of ovarian activity were also observed between previous studies of dexamethasone-induced parturitions and the present study.  相似文献

Changes in lectin binding of developing fetal mouse testes and ovaries were examined by light and electron microscopy, with much attention paid particularly to those in carbohydrates of germ cells. Characteristic binding patterns were observed with three lectins (BPA, GS-I, and GS-II) in the germ cells and the somatic cells during the process of testicular and ovarian development. GS-I and BPA, which showed similar binding patterns, preferentially bound to the plasma membrane and small dense bodies (SDB) of germ cells in both testes and ovaries during the 12th to 14th day post coitum (p.c.). In the fetal testes on day 16 p.c., the reaction with both GS-I and BPA completely disappeared. While, in the ovaries, a weak reaction with these lectins was retained as it was in germ cells until the 16th day p.c. The reaction with GS-II was restricted to Sertoli cells in the fetal testes during the 12th to 14th day p.c., and thereafter disappeared on day 16 p.c. The distribution of GS-II binding sites was in agreement with that of the glycogen granules. No positive staining with GS-II was seen in the ovaries throughout their development. These results indicate that certain glycoconjugates containing D-galactose and N-acetyl-D-galactosamine residues are expressed on the cell surface and in the SDB of germ cells during the period of the 12th to 14th day p.c., and that striking changes in function as well as in structure may take place in both germ cells and somatic cells during the 14th to 16th day p.c. in association with testicular and ovarian development.  相似文献

The distribution of the West Nile virus (WNV) in the organs and tissues of the mosquito Culex pipiens pallens, a potential vector of WNV in China, was investigated up to 14 days after oral infection. The WNV antigen was detected in paraffin‐embedded mosquitoes using immunocytochemistry and viral titers of post‐infected mosquitoes determined by plaque assay. Viral titers sharply decreased 24 h post‐infection, were undetectable for the first few days, then rose over the course of infection. The first midgut infection appeared after one day, and the overall infection rate (based on midgut infection) was 43.9%. Other tissues, including hindgut, foregut, ovarian follicles, Malpighian tubules, and ommatidia, showed weak WNV antigens as early as three days post‐infection. Staining in the salivary glands first appeared after seven days, and the salivary gland infection rate on the 14th day was 37.5%. Specimens with no detectable WNV antigens in any tissues, and with positive results confined to the midgut, anterior midgut, and hindgut, were observed on the 14th day. The route of viral dissemination from the midgut, and the relative importance of amplifying tissues in mosquitoes' susceptibility to infection, were evaluated. The results indicate that Cx. p. pallens has the ability to harbor WNV throughout its alimentary system and that midgut epithelial cells may be the initial site of the replication of this virus in this species.  相似文献

Serum copper and zinc estimations in humans were made to find their diagnostic and prognostic value in cases of myocardial infarction. Following infarction, there was an increase in serum copper levels from the first 24 h up to the 7th day, with gradual decline that did not reach the normal value up to the 14th day. The serum zinc levels declined in the first 24 h until the 4th day and increased to the normal value on the 14th day. It is concluded that, for diagnosis of myocardial infarction, serum zinc levels are more useful during the first week and copper levels in the second week after the onset of infarction.  相似文献

The morphology of metacercariae of Massaliatrema misgurni Ohyama et al. (Ohyama et al., Parasitol Int 2001; 50; 267–71) was described, and their infectivity, egg output, growth and development in mice until day 35 post infection (PI) were studied. Metacercarial cysts from loaches imported from China to Japan were 199–349 μm in diameter and consisted of a very thick translucent outer layer and a refractile inner layer. Excysted metacercariae basically had the shape of miniature adults, and a pair of pre-developed testes but no other genital organs were recognized. The worm recovery rate from mice was 36.7–51.7% during days 3–7 PI, and decreased remarkably to 2.5 and 1.7% at days 28 and 35 PI. The prepatent period was 3–4 days, and the egg output quickly increased and sustained high levels at days 5–7 PI, then decreased suddenly at day 8 PI, and continued at a low level until day 28 PI. The size of the body and inner organs such as the oral sucker, pharynx, acetabulum, testes, ovary and seminal receptacle quickly increased until day 3 PI, and sustained at a plateau level until day 21 PI except testes which gradually decreased until 21 PI. The number of the uterine eggs increased with a short time lag compared to other genital organs and sustained a plateau level until day 21 PI. Compared with other Heterophyidae species, M. misgurni was characterized by the remarkably fast growth and development.  相似文献

将正常兔IgG与完全弗氏佐剂颈免疫大鼠,8天后注射亚致肾炎剂量的兔抗大鼠肾小球基底膜(GBM)血清,结果大鼠尿蛋白量在注射该血清后第1天即明显升高,第3天达高峰;在第21天时,血清肌酐显著升高。肾脏组织学检查:免疫荧光见第1天即有大鼠IgG、C3和兔IgG沿GBM呈典型线状沉积;光镜见第1天肾小球内细胞数目明显增加,内皮细胞肿胀,肾小球内见中性粒细胞浸润,第7、14.21天时肾小球毛细血管丛系膜区增宽,系膜细胞增生,间质可见炎症细胞浸润,PASM染色可见GBM增厚,电镜见GBM内有电子致密物线状沉积,上皮足突融合内皮窗孔消失,微血栓形成阻塞管腔,间质有细胞浸润和纤维化。  相似文献

目的:以鼠嗜铬神经瘤细胞(PC12)为模型,筛选锰对神经细胞增殖抑制作用的时间及剂量,观察锰作用下PC12细胞的氧化应激反应与细胞形态学、生化指标改变和丝裂原活化蛋白激酶pp38(p38MAPKs)的活化表达。方法:用200,400,600,800μmol/LMnCl2的培养液,分别作用对数生长期PC12细胞1,2,3,4d后,用MTT筛选锰的细胞毒性剂量;测定200-600μmol/L MnCl2作用4d后,PC12细胞还原型谷胱甘肽和丙二醛含量;透射电镜观察细胞形态学变化;琼脂糖凝胶电泳检测MnCl2对PC12细胞基因组DNA的影响。western-blot法检测p-p38。结果:MTT实验显示200~800μmol/LMnCl2作用1,2,3,4d对PC12有显著的抑制作用,呈剂量和时间依赖趋势,600μmol/LMnCl2作用4d对PC12的抑制率可达50%以上。200-600μmol/LMnCl2作用于细胞4d后,随着浓度的升高,还原型GSH逐渐降低,MDA的含量逐渐升高;600μmol/LMnCl2作用4d电镜可见细胞凋亡,同样条件下细胞DNA碎片化。Western-blot实验显示600μmol/LMnCl2作用1,2,3,4dp-p38逐渐升高,3d时较对照组增加6.6倍(n=3,p〈0.05),200,400,600μmol/L MnCl2作用4d时,磷酸化蛋白38(p-p38)也逐渐升高,400μmol/L MnCl2作用4d时较对照组升高了4.7倍(n=3,p〈0.05)。结论:PC12细胞在锰作用下发生氧化应激反应,上调p-p38,诱导细胞凋亡,细胞增殖抑制。  相似文献

Fasting and refeeding effects on gastrointestinal morphology and digestive enzyme activities of Atlantic salmon, held in tanks of seawater at 9°C and 31‰ salinity, were addressed in two trials. Trial 1: Fish (mean body mass 1190 g) were fasted for 40 days and intestines sampled at day 0, 2, 4, 11, 19 and 40. Trial 2: Fish (1334 g), fasted for 50 days, were refed and sampled at day 0, 3 and 7. Mass, length, protein, and maltase, lactase, and leucine aminopeptidase (LAP) activities were analyzed for stomach (ST), pyloric caeca (PC), proximal (PI), mid (MI), and distal intestine (DI). PC contributed 50% of gastrointestinal mass and 75% of enzyme capacity. Fasting decreased mass and enzyme capacities by 20–50% within two days, and 40–75% after 40 days. In PC, specific brush border membrane (BBM) maltase activity decreased whereas BBM LAP increased during fasting. Upon refeeding, enzyme capacities were mostly regenerated after one week. The results suggest that refeeding should start slowly with about 25% of estimated feed requirement during the first 3 days, but may then be stepped up rapidly. Investigations of digestive processes of fed fish should only be performed when intestines are feed-filled to avoid bias due to effects of fasting.  相似文献

In this study we examined the effect of various initial sensitizing doses of infective Toxocara canis eggs and the effect of murine host genotype on the level of trapping of larvae in the liver after larval challenge. In the initial experiments, C57BL/6J mice were infected with a sensitization dose of 5, 25, 75, 125, or 250 infective T. canis eggs on day 0 postinfection (PI). On day 28 PI all mice were challenged with 500 infective eggs. On days 7, 14, and 21 postchallenge (PC) larval numbers within individual livers were determined. Trapping of larvae was observed in mice receiving a sensitization dose of 25 or more eggs. At 7 and 14 days PC the level of trapping increased with sensitization egg dose up to a dose of 125 eggs. At 21 days PC the level of trapping reached a plateau at a sensitization dose of 75 eggs. The peak level of larval trapping was observed on day 7 and day 14 PC following sensitization doses of 125 and 250 eggs, respectively. In the subsequent experiments, mice of various strains and H-2 haplotypes were inoculated with an initial sensitization dose of 125 eggs and a challenge dose of 500 eggs on day 0 and day 28 PI, respectively. Larval trapping within the liver was determined on day 14 PC. C57BL/6J mice trapped significantly more larvae than DBA/2J mice (P less than 0.01); all other strains trapped larvae at a lower, but statistically similar, level to the C57BL6/J mice.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The ontogeny of the caudal neurosecretory cells (Dahlgren cells) in the caudal spinal cord of the chum salmon, Oncorhynchus keta, was examined by conventional electron microscopy and with immunohistochemistry for urotensins (U) and neuropeptide Y (NPY). The precursors of the Dahlgren cells first appeared as agranular ovoid cells in the caudal region of the neural tube of 40-day-old embryos about one week before hatching. The occurrence of cytoplasmic granules in the immature Dahlgren cells became evident by the 14th day after hatching. At this moment, the U-positive reaction was merely demonstrated in some of the granules. Close association of NPY-positive fibers with the caudal neurosecretory structures was recognizable in 1-month-old larvae. Thus, it is apparent that the salmon Dahlgren cells start their secretory activity (production of the secretory granules) in early larval stages and that, thereafter, NPYergic afferent innervation of the caudal neurosecretory system becomes evident.  相似文献

At the 14th day of gestation, embryonic thymocytes+ are large, functionally incompetent cells with H-2K+ Thy-1+ B14- Ly-2- L3T4- phenotype, some of which express TL antigen. Differentiation of these cells in organ culture is characterized by: 1) appearance of cells expressing Ly-2 and L3T4 molecules, first among the population of large cells, after 2 days of culture; 2) appearance of small H-2K- Thy-1+TL+B14+Ly-2+L3T4+ and H-2K-Thy-1+TL+B14-Ly-2+ L3T4+ cells between days 2 and 4; 3) accumulation of small H-2K- Thy-1+ TL+ B14- Ly-2+ L3T4+ (but not H-2K- Thy-1+ TL+ B14+ Ly-2+ L3T4+) cells until day 5 of culture, and their subsequent gradual disappearance which is paralleled by an increase of the proportion of medium-sized H-2K+ Thy-1+ TL- B14- cells with various Ly-2 L3T4 phenotypes; 4) appearance and subsequent accumulation of cytolytic and IL 2-producing cells between days 4 and 6. Comparison of these results with the data from similar in vivo studies shows that differentiation of organ-cultured thymocytes rather closely follows the in vivo development only during the first week of culture and shows significant deviations thereafter. Precursors of cytolytic cells and cytolytic effector cells as well as IL 2-producing cells are found among both Ly-2+ and Ly-2- populations of thymocytes, indicating that there is no clear association between Ly-2 phenotype and the ability to kill or to secrete IL 2.  相似文献

The growth and development of Gymnophalloides seoi were studied in C3H/HeN mice and effects of immunosuppression of the host on the worm development were observed. Two hundred metacercariae of G. seoi were orally administered to each mouse, and worms were recovered on days 1, 3, 5, 7, 14 and 21 post-infection (PI). The worm recovery rate was significantly higher in immunosuppressed (ImSP) mice than in immunocompetent (ImCT) mice except on days 1 and 3 PI. The worms attained sexual maturity by day 3 PI with eggs in the uterus, and worm dimensions and the number of uterine eggs continuously increased until day 14 PI in ImSP mice. Worms recovered from ImSP mice were significantly larger in size than those from ImCT mice on days 1 and 3 PI, and the number of uterine eggs was significantly larger in ImSP mice on days 5 and 7 PI. Genital organs such as the ovary, testes, and vitellaria, that were already developed in the metacercarial stage, grew a little in size until day 14 PI. The results show that the C3H/HeN mouse is, though not excellent, a suitable laboratory host for G. seoi.  相似文献
