Establishing the phylogeny of fungi and protists often has proved difficult owing to the simple morphologies and convergent characters in these organisms. We used DNA sequences of nuclear small-subunit ribosomal RNA genes to determine phylogenetic relationships among three major classes of organisms considered to be fungi--Basidiomycetes, Ascomycetes and Chytridiomycetes--and to assess the taxonomic position of Neocallimastix, an economically important anaerobic rumen microorganism whose classification is controversial. The Basidiomycetes and Ascomycetes, two classes of nonflagellated fungi, are the most closely related taxa. Chytridiomycetes, though bearing flagella, group with these higher fungi rather than with the protists. Neocallimastix, a eukaryote lacking mitochondria and variously classified as a protist or as a fungus, shows closest molecular affinities with the Chytridiomycete fungi in the order Spizellomycetales.  相似文献

The nature of the ancestral red alga: inferences from a cladistic analysis   总被引:1,自引:0,他引:1  
A cladistic analysis of the orders of Rhodophyta is presented. Sixteen taxa and 34 characters comprise the data matrix. Included in the analysis are biochemical and ultrastructural features of pigments, cell walls, cell organelles, mitosis and pit connections as well as vegetative and reproductive characters. The traditional recognition of two classes or subclasses, Bangiophycidae and Florideophycidae, is not supported regardless of whether Porphyridiales, Rhodochaetales or Bangiales is designated the outgroup. Florideophycidae, however, appears to be monophyletic with Bangiales as its sister group. Relationships among taxa with one or two plug cap layers, i.e. Acrochaetiales, Palmariales, Corallinales, Nemaliales, Batrachospermales, Gelidiales and Hildenbrandiales are unresolved. Rhodochaetales, Bangiales and possibly Erythropeltidales are monophyletic, but Porphyridiales is polyphyletic. The class Cyanidiophyceae is not recognized and the included genera are considered to be unicellular red algae belonging to Porphyridiales. Taxa that have been proposed as sister groups for red algae, including Cyanobacteria, Cryptophyta, Glaucophyta and Chlorophyta, and Ascomycetes and Basidiomycetes are discussed in relation to the proposed phylogeny of Rhodophyta.  相似文献

Summary The molecular weights of the 18s and 25s ribosomal RNA components of fungi from all major classes were determined by electrophoresis in polyacrylamide gels. The molecular weight of the 18s RNA was found to be very similar for all fungi (range 0.71–0.75 million) and about 4–5% larger than the 18s RNA of HeLa cells and soybean. The molecular weight of the 25s RNA ranged between 1.45 million in the Myxomycetes and 1.30–1.31 million in the Ascomycetes and Basidiomycetes. The differences in the 25s RNA molecular weights between various classes of fungi were interpreted as being in agreement with a monophyletic origin of the Chytridiomycetes, Zygomycetes, Ascomycetes and Basidiomycetes, and independent origins for the Myxomycetes and the Oomycetes. The Hyphochytridiomycete examined could not be placed unequivocally in any group on the basis of its 25s RNA. Fungal RNA extracted with a p-aminosalicylate-triisopropylnaphthalene sulfonate-phenol mixture at 40–60°C contained a high molecular weight aggregate of the 18s and 25s ribosomal RNA; this suggested significant base sequence homology between the two ribosomal RNA species in fungi.  相似文献

The fungal community associated with the terrestrial photosynthetic orchid Gymnadenia conopsea was characterized through PCR-amplification directly from root extracted DNA and cloning of the PCR products. Six populations in two geographically distinct regions in Germany were investigated. New ITS-primers amplifying a wide taxonomic range including Basidiomycetes and Ascomycetes revealed a high taxonomic and ecological diversity of fungal associates, including typical orchid mycorrhizas of the Tulasnellaceae and Ceratobasidiaceae as well as several ectomycorrhizal taxa of the Pezizales. The wide spectrum of potential mycorrhizal partners may contribute to this orchid's ability to colonize different habitat types with their characteristic microbial communities. The fungal community of G. conopsea showed a clear spatial structure. With 43 % shared taxa the species composition of the two regions showed only little overlap. Regardless of regions, populations were highly variable concerning taxon richness, varying between 5 and 14 taxa per population. The spatial structure and the continuous presence of mycorrhizal taxa on the one hand and the low specificity towards certain fungal taxa on the other hand suggest that the fungal community associated with G. conopsea is determined by multiple factors. In this context, germination as well as pronounced morphological and genetic differentiation within G. conopsea deserve attention as potential factors affecting the composition of the fungal community.  相似文献

The nuclear DNa of 28 species (30 strains investigated) of yeasts classified currently or previously in the genus Trichosporon. was analysed for its molar percentage of guanine + cytosine (mol% G+C). This criterion, together with biochemical characteristics, suggested the separation of the organisms studied into two groups. The first group, which appears related to the Ascomycetes, includes thirteen species with a G+C content lower than 50 mol% (34.7–48.8), and lacks urease (except T. margaritiferum). The second group appears related to the Basidiomycetes and includes fifteen species with a G+C content higher than 50 mol% (57–64) and has the ability to hydrolyse urea.A DNA homology experiment with T. beigelii and twelve other species of the second group showed very low values of complementarity with T. beigelii-labeled DNA. All these species must be considered as taxa other than T. beigelii.  相似文献

Air sampling was conducted in Waterloo, Canada throughout 1992. Functional relationships between aeromycota and meteorological factors were analysed. The meteorological factors were, in descending order of importance: mean temperature, minimum temperature, maximum temperature, mean wind speed, relative humidity (RH), rain, maximum wind speed and snow. The most important airborne fungal propagules in descending order were: total fungal spores, unidentified Ascomycetes,Cladosporium, Coprinus, unidentified Basidiomycetes,Alternaria and unidentified fungi. Most airborne fungal taxa had highly significant relationship with temperature, butAspergillus/Penicillium, hyphal fragments andEpicoccum did not.Epicoccum and hyphal fragments were positively associated with wind speed. In comparison with other airborne fungal taxa,Leptosphaeria and unidentified Ascomycetes were more closely correlated with rain and RH during the growing season.  相似文献

Fluorescent probes have been applied to analyze the molecular architecture of fungal cell surfaces. Binding patterns of aniline blue and FITC-labeled wheat germ agglutinin (FITC-WGA) elucidated class-specific properties. Aniline-blue-induced fluorescence was distributed over the entire cell walls from Ascomycetes, but was confined to sporangial walls of Zygomocetes, hyphal tips and a few other sites in Basidiomycetes, while no fluorescence was found with sporangia and rhizoids of Chytridiales. FITC-WGA in Zygomycetes and in Ascomycetes was restricted to few sites (e.g. apex of hyphae), in Basidiomycetes and Chytridiales label was evenly associated with the entire surface of hyphal walls, or sporangia and rhizoids. For Oomycetes, Zygomycetes, Ascomycetes, and Basidiomycetes differences in the molecular architecture between apex and hyphal side walls were discerned, although the chemical nature of these differences is distinct for each class. Species specific differences, due to differences in binding patterns of several lectins are not apparent at fungal cell surfaces. The degree of intraspecies variation was found to be larger than interspecies diversification, suggesting changeableness of the molecular architecture of fungal cell walls. This is in contrast to assertions which we made by working on algae. There species-specific lectin binding patterns have been described.  相似文献

阐明植被恢复过程中土壤真菌群落的变化及其生态功能,对于制定科学有效的退化生态系统管理措施有重要参考价值。利用扩增子高通量测序技术和生物信息学分析解析了华北退化荒地自然恢复(对照组,CK)和建植豆科植物和禾本植物人工草地(分别为LG和GG处理)过程中土壤真菌群落结构和功能群特征差异。结果表明:(1)退化荒地土壤表层样品中共获取6315个真菌OTU,隶属于17门60纲145目347科896属,优势菌门为Ascomycetes、Mortierellomycota和Basidiomycetes,LG处理相较于CK的Basidiomycetes相对丰度明显升高,GG处理相较于CK的Mortierellomycota相对丰度明显提高。(2)土壤真菌功能群类型以腐生真菌为主,共生真菌次之,病原真菌占比最少。建植人工草地对腐生真菌和共生真菌相较于病原真菌的功能群组成影响更明显,并导致腐生真菌相对丰度升高,共生真菌相对丰度降低。(3)土壤真菌群落结构受植物物种丰富度、根系生物量等植被参数变化的显著(P<0.05)影响,且与土壤有机碳、总氮、速效氮、总磷等土壤养分水平显著(P<0.05)相关。本研究的结果有助于深入理解建植人工草地对土壤真菌群落结构和功能的影响,并为华北退化荒地植被恢复策略提供理论依据。  相似文献

植物根系与真菌形成菌根, 在自然生态系统的物质能量循环中具有重要的生态功能。作者在四川省的中国杜鹃园选取银叶杜鹃(Rhododendron argyrophyllum)和繁花杜鹃(R. floribundum), 通过直接扩增杜鹃花根部真菌rDNA-ITS区片段, 来揭示该地区杜鹃花属植物根部真菌的多样性。ITS序列分析结果表明: 从两种杜鹃的根部共检测到41个真菌分类单元, 分别属于子囊菌纲的柔膜菌目(Helotiales)、散囊菌目(Eurotiales)、盘菌目(Pezizales)、假球壳目(Pleosporales)和担子菌纲的蜡壳耳目(Sebacinales)、伞菌目(Agaricales)、Erythrobasidiales、线黑粉菌目(Filobasidiales)。银叶杜鹃和繁花杜鹃根部真菌种类丰富, 包括了杜鹃花类菌根真菌、外生菌根真菌和其他类型真菌, 其中担子菌纲的蜡壳耳目和子囊菌纲的柔膜菌目占有较大比例。  相似文献

This paper may be regarded as an addendum to previous work along the same lines, more particularly the work which dealt with some 700 of the larger Basidiomycetes. Both Ascomycetes and Basidiomycetes were tested for their antibiotic properties by means of sporophore extract. Of the ninety-nine fungi tested, twenty-eight showed a positive and seventy-one a negative reaction against the test bacteria, Bacterium coli and Staphylococcus aureus.  相似文献

Illus. 1962.—Electron microscopy of 13 carpomycete species (5 Ascomycetes, 5 Basidiomycetes, and 3 Deuteromycetes) presents evidence that suggests there is an Ascomycete- and a Basidiomycete-type septum. The observed Ascomycete septa taper slightly or not at all toward a simple, clear-channel, central pore, while the observed Basidiomycete septa flare sharply and broadly at the center to produce what is termed a dolipore septum. On each side of this septum is a double membrane structure that is crescent-shaped in section. This is given the name parenthesome. These dolipore-parenthesome septa were found to be characteristic of basidiocarpic, dikaryotic, hyphae. Interpretative drawings of both types of septa are presented. From these and the micrographs, it is not difficult to see how materials, including nuclei, could pass through the Ascomycete-type septum, but it would appear that the modifications of the Basidiomycete-type septum while maintaining cytoplasmic continuity would prohibit nuclear migration in dikaryotic hyphae. It is postulated that as the plant and animal phylogenetic lines have developed actual diploidism, the terminal group of the fungi may have achieved functional diploidism. Of the 3 species of Deuteromycetes examined, 2, Aspergillus variecolor and Stilbum zacalloxanthum, have typical Ascomycete-type septa; the third, QM 7739, has Basidiomycete-type septa. This fungus is a species of Sclerotium, but the present confusion and lack of definitive information regarding the taxonomy, genetics, and perfect stage of this genus prohibit the enlarging at present on the possible significance of this observation.  相似文献

Two fungal-like organisms (class Oomycetes) and 263 species of fungi from 111 genera, 42 families, 23 orders, and 7 classes have been found on the territory of Aksu-Zhabagly Nature Reserve. Twenty-two species are found for the first time. The vast majority of species are parasitic (202 species), 56 species belong to saprotrophs, and 7 to symbiotrophs. Classes Taphrinomycetes, Urediniomycetes, and Ustilaginomycetes are represented exclusively by parasites. There have been found 21 saprotrophic, 27 parasitic, and 4 symbiotrophic species in the class Ascomycetes. The most numerous group in the class Basidiomycetes is the group of humus saprotrophs; it contains 15 species. Groups of litter saprotrophs and parasites contain four species each, symbiotrophic species have three species, coprotrophs and xylotrophs have two species each, and litter and humus saprotrophs have one species each. Anamorphic fungi are represented by 91 parasitic and 11 saprotrophic species. In comparison with closely located Sairam-Ugam National Park and the Karatau Nature Reserve, the mycobiota of Aksu-Zhabagly is characterized by a significant number of species in the classes Basidiomycetes, Urediniomycetes, Hyphomycetes, and Coelomycetes. Thirty-one species are common to these protected areas. The most numerous group is parasites.  相似文献

Different types of molecular markers are available for use in evolutionary and population studies of microscopic fungi. These approaches have proved their merits and have been successfully applied to a wide range of fungal species belonging in the Ascomycetes and Basidiomycetes. Species in the class Zygomycetes have been rather neglected from this aspect. Ectomycorrhizal (ECM) fungal communities of Quercus liaotungensis of different ages (seedlings, young trees and mature trees) in the growing seasons (June and September) between 2007 and 2009 were studied in a temperate forest of northern China. A total of 66 ECM fungal taxa were identified based on ECM morphotyping, PCR-RFLP, and DNA sequence data. Of these fungal taxa, 51 were Basidiomycetes (77.3%) and 15 were Ascomycetes (22.7%). Cenococcum geophilum was the dominant species. Thelephoraceae (16 taxa), Sebacinaceae (12 taxa) and Russulaceae (seven taxa) were the most species-rich and abundant ECM fungi, accounting for 19.5%, 17.6% and 8.3% of the total ECM root tips, respectively. Results of multiple response permutation procedure (MRPP) analysis indicated that there were marginally significant effects of tree ages (A?=?0.01801, P?=?0.054) and growing seasons (A?=?0.01908, P?=?0.064) on the ECM fungal species composition of Q. liaotungensis in a temperate forest.  相似文献

Fungi are important actors in ecological processes and trophic webs in mangroves. Although saprophytic fungi occurring in the intertidal part of mangrove have been well studied, little is known about the diversity and structure of the fungal communities in this ecosystem or about the importance of functional groups like pathogens and mutualists. Using tag-encoded 454 pyrosequencing of the ITS1, ITS2, nu-ssu-V5 and nu-ssu-V7 regions, we studied and compared the fungal communities found on the marine and aerial parts of Avicennia marina and Rhizophora stylosa trees in a mangrove in New Caledonia. A total of 209,544 reads were analysed, corresponding to several thousand molecular operational taxonomic units (OTU). There is a marked zonation in the species distribution, with most of the OTU being found specifically in one of the microhabitat studied. Ascomycetes are the dominant phylum (82%), Basidiomycetes are very rare (3%), and 15% of the sequences correspond to unknown taxa. Our results indicate that host specificity is a key factor in the distribution of the highly diverse fungal communities, in both the aerial and intertidal parts of the trees. This study also validates the usefulness of multiple markers in tag-encoded pyrosequencing to consolidate and refine the assessment of the taxonomic diversity.  相似文献

蕈菌的定义是:具有明显的、肉眼可辨的子实体的大型真菌。其菌肉为肉质、木质或革质。大多数的种属担子菌,部份种属子囊菌,有些种可形成菌根。蕈菌有重要的经济价值,不少种类可供食用和药用。过去三年内,世界食用蕈菌年产量每年平均增加24.5%,总增加率为72.5%。1989—1990年度产量为3763000吨,年产值约为74.85亿美元。今后预计产量还会增加。一些药用菌已作为医药制剂进行工业生产并在临床上应用,有的已开发成为保健饮料和化妆品的重要原料。  相似文献
