Insect additives have been shown to improve the value of artificial media for Trichogramma species, but at the same time maintain dependence on parallel cultures of host insects. In the present study, Trichogramma dendrolimi Matsumura was reared in vitro from egg to adult on artificial media with different contents of pupal hemolymph of Chinese oak silkmoth Antheraea pernyi (Guérin‐Méneville) and with supplements of distilled water, or of trehalose dissolved in water or in Grace's insect medium. The results indicated insect hemolymph was the key component of artificial medium. Developmental parameters, including rates of parasitism, pupation, adult emergence and normal adults, and numbers of produced adults, were increased on media supplemented with trehalose even when the proportion of pupal hemolymph was reduced. Two artificial media, the first containing 30% hemolymph and 10% trehalose in water with 98.9% parasitism rate, 77.7.0% pupation rate, 77.2% emergence rate, 80.0% normal adult rate and 333 produced adults, and the second containing 25% hemolymph and 15% trehalose in Grace's insect medium with 97.8% parasitism rate, 91.0% pupation rate, 85.2% emergence rate, 76.1% normal adult rate and 757 produced adults, were believed to hold potential to mass produce T. dendrolimi. The use of trehalose to partially replace pupal hemolymph in artificial medium of this and other Trichogramma species may contribute to a significant reduction in their production cost and may as such help to evade problems related to short supplies of lepidopteran eggs, which currently constitute the main factitious host for the mass rearing of the parasitoids.  相似文献

A variety of semi-defined artificial diets were developed and tested for their ability to support the in vitro development of Edovum puttleri. In the most effective diet, 2.6% of E. puttleri pupated. This diet contained high levels of hen egg yolk combined with Manduca sexta larval hemolymph, or with a mixture of M. sexta egg homogenate and larval hemolymph. Egg homogenate alone (without the addition of hemolymph) was not capable of supporting the parasitoid's development. Thus, hemolymph appears to contain unidentified factor(s) important for inducing pupation of the wasp. Addition of M. sexta pupal fat body tissue extract (in place of hemolymph) also promoted pupation of E. puttleri. Gypsy moth (Lymantria dispar) larval hemolymph could not replace M. sexta larval hemolymph. Fractionation irreversibly reduced the growth-promoting effects of M. sexta larval hemolymph. However, the most effective fraction contained components whose molecular weights were 1000 kd. In diets that were devoid of insect materials, the best results were achieved when hen egg yolk, FreAmine, yeast extract, lactalbumin, trehalose, fetal bovine serum and bovine milk were included. This is the first report of an artificial diet for in vitro rearing an eulophid parasitoid from the egg through the pupal stage.  相似文献

Laboratory studies were conducted to investigate the effect of selected temperatures on the development, mortality, sex ratio, and emergence rate of Trichogramma dendrolimi Matsumura reared from a factitious host, oak silkworm, Antheraea pernyi (Guérin-Méneville) eggs. The comparison tests were conducted to investigate the fecundity on Dendrolimus spectabilis (Butler) eggs of T. dendrolimi reared from natural and factitious hosts, and artificial hosts. Developmental periods from egg to adult of T. dendrolimi reared at 26, 28, 30, and 32°C were 10.9, 9.6, 9.0, 8.6 days, respectively. Emergence rates of T. dendrolimi decreased 78.0, 88.4, 60.7, and 50.1% as temperature increased. The progeny sex ratios (i.e., females per male) were 7.3, 8.4, 8.2 and 6.9 at the respective temperatures. When adult T. dendrolimi emerged from A. pernyi eggs were kept in dark conditions for storage at different temperatures for 34 days, their mortality increased as temperature increased: 0% at 0°C; 10% at 4°C; 40% at 6°C; 50% at 18°C and 100% at ≥26°C. The fecundity of T. dendrolimi reared from three different hosts was investigated on D. spectabilis eggs. During the first day, the adult T. dendrolimi reared on A. pernyi eggs laid most eggs (99.0±10.7), followed by those reared on artificial hosts (76.6±24.5) and D. spectabilis eggs (63.4±35.9). Over the entire lifespan, T. dendrolimi reared on D. spectabilis eggs produced the highest number of eggs (218.0±27.9), followed by A. pernyi eggs (104.0±44.7) and artificial host (98.2±37.1). These results suggest that the temperature and three hosts factors had an effect on rearing of the T. dendrolimi on the factitious host, A. pernyi eggs.  相似文献

Mythimna separata (Walker) is a serious cosmopolitan pest. Trichogramma species have been used worldwide as biological control agents of lepidopteran pests. However, no data on the control efficiency of Trichogramma species on this pest have been reported. We evaluated the suitability of M. separata eggs for three Trichogramma species, Trichogramma dendrolimi Matsumura, T. ostriniae Pang et Chen and T. japonicum Ashmead, and compared the suitability with that of Corcyra cephalonica (Stainton) eggs. The parasitism rate, development time, emergence rate, female proportion, body size and egg load of female offspring indicated that the M. separata egg is a suitable host for the three Trichogramma species. On the other hand, the non‐emergence rate and the relationship between egg load and body size reflected that M. separata eggs are less suitable for the three Trichogramma species than C. cephalonica eggs. Egg resorption occurred in T. japonicum 4 days after emergence.  相似文献

The silkworm Bombyx mori is a lepidopteran insect with four developmental stages: egg, larva (caterpillar), pupa, and adult. The hemolymph of the silkworm is in an open system that circulates among all organs, and functions in nutrient and hormone transport, injury, and immunity. To understand the intricate developmental mechanisms of metamorphosis, silkworm hemolymph from different developmental stages, including the 3rd day of fifth instar, the 6th day of fifth instar, the 3rd day of pupation, the 8th day of pupal stage and the first day of the moth stage, was investigated by two-dimensional electrophoresis and mass spectrometry.  相似文献

Ooencyrtus kuvanae is a key egg parasitoid of Lymantria dispar (L.), an important defoliating pest in China and North America. We have developed mass rearing techniques for O. kuvanae and in this study compared the reproductive capacity of O. kuvanae when it was previously reared on the natural host, L. dispar or the factitious host, Antheraea pernyi Guerin‐Meneville. There was no significant difference in the oviposition period or total number of eggs laid between L. dispar‐reared and A. pernyi‐reared females. However, the mean number of offspring successfully emerging from those eggs was significantly larger from the A. pernyi‐reared females compared with L. dispar‐reared females. From this, we can conclude that, with respect to reproductive capability, eggs of A. pernyi are suitable factitious hosts for mass rearing of O. kuvanae.  相似文献

Anastatus disparis (Ruschka) (Hymenoptera: Eupelmidae) is an egg parasitoid and considered a potential biological control agent of the gypsy moth, Lymantria dispar (L.) (Lepidoptera: Lymantriidae). Only male offspring of A. disparis emerge from single eggs of L. dispar in the laboratory, and A. disparis exhibits low parasitism on L. dispar in the field. We therefore selected several lepidopteran species with various body sizes to evaluate the optimal egg size for hosting A. disparis. In addition, we explored whether the nutritional content of a single L. dispar egg influences the sex of A. disparis offspring and why female offspring can be reared from L. dispar eggs in the field. The results indicated that host egg size decisively influenced the body size and sex ratio of the parasitoid offspring. Therefore, larger hosts, especially the largest eggs of Antheraea pernyi Guérin‐Méneville (Saturniidae), might increase the fitness of A. disparis females. Lymantria dispar eggs concealed in the larger egg shell of A. pernyi produced female A. disparis, suggesting that adult A. disparis should prefer hosts with larger bodies and that the nutritional content of L. dispar eggs did not play a decisive role in the sex allocation of A. disparis. The results also indicated that the egg mass and the fur cover of L. dispar egg masses might be the key factors inducing female A. disparis to lay female offspring in L. dispar eggs.  相似文献

Bradysia cellarum Frey and Bradysia impatiens Johansen are major pests of vegetable crops, as well as edible mushrooms and ornamental plants, and damage to hosts resulting in economic losses. Temperatures above the optimum levels for these pests have been predicted to regulate their population growth during summer. The aim of the present study was to examine the effects of both heat stress and exposure time on the growth and development of eggs, larvae and pupae for two Bradysia species. The egg stage, egg hatching rate, 4th instar larval stage, pupation rate, pupal stage and adult emergence rate were observed after exposing at high temperatures of 34°C, 37°C and 40°C for 1, 2, 4 and 6 hr. The results showed that 34°C, 37°C and 40°C for 1-, 2-, 4- and 6-hr exposure treatments prolonged the developmental stage of egg, 4th instar larva and pupa, while decreasing the egg hatching rate, pupation rate and adult emergence rate. This suggests that increasing temperature or prolonging exposure time to the heat stress could significantly affect insect survival, growth and development. Our study could provide an ecological basis for pests’ management strategy by using short-term heat stress.  相似文献

This study has been proved that statistical experimental designs offer efficient and feasible approaches for determination of culture medium and fermentation conditions of Streptomyces lavendulae Xjy. Nutrition and cultural conditions have high influence on the antibiotic production of S. lavendulae Xjy. Soya bean meal, NaCl and (NH4)2SO4 were identified as the best ingredients for high antifungal activity of Xjy based on single variable experiments. Using response surface methodology, the best concentrations of medium ingredients were 1.46% soya bean meal, 2.25% NaCl and 1.26% (NH4)2SO4. The potential components were reduced from seven to three, and the antibiotic activity was increased by 10%. The optimized conditions for Xjy in a 250‐ml Erlenmeyer flask were initial pH 5, medium volume 25 ml/250 ml and inoculation volume 10% at 30°C with 200 rpm by response surface methodology. Validation experiments carried out to check the accuracy of the models indicated that the predicted values agreed with the experimental values. The extension of necrosis lesion of Botryosphaeria dothidea was impeded by the application of Xjy in apple fruit. In tomato plants, Xjy showed the 43.7–51.7% curative and protective effects against Fulvia fulva. The optimal medium not only gave a 20% increase in antifungal activity, but it provided a simpler formulation. The results will set a basis for further study on large‐scale fermentation volumes using S. lavendulae Xjy and be useful for the development of more advanced control strategies on plant diseases.  相似文献

Exorista larvarum (L.) (Diptera: Tachinidae), a larval parasitoid of Lepidoptera, can be reared from egg to fecund adult on artificial media composed of crude components. The standard in vitro culture is performed in 24‐well plastic rearing plates. Exorista larvarum eggs, removed from superparasitized larvae of Galleria mellonella (L.) (Lepidoptera: Pyralidae), are individually placed in the wells, each containing a cotton swab soaked in liquid medium. The plates are then sealed until parasitoid puparium formation. To avoid contamination by microorganisms, the artificial medium is routinely supplemented with 0.01% solution of gentamicin. Experiments were carried out to assess whether this broad‐spectrum antibiotic may be replaced with hydrolate of Monarda fistulosa L. (Lamiaceae), which was selected due to its high in vitro activity against pathogenic microorganisms for humans and plants. The hydrolate was either supplemented to the artificial medium (0.5% wt/wt) (first experiment) or placed in an empty well (200 μl) of the rearing plate, to be supplied as saturated air due to evaporation (second experiment). In both experiments, a standard medium with gentamicin and an antimicrobial‐free medium were maintained as positive and negative controls, respectively. In the first experiment, in the hydrolate‐supplemented medium fewer E. larvarum completed egg‐to‐adult development than in the standard medium, but significantly more parasitoids developed from egg to adult compared to the antimicrobial‐free medium. No significant difference was found between the numbers of eggs laid by the females obtained from the standard medium vs. those from the hydrolate‐supplemented medium. In the second experiment, the hydrolate‐saturated air significantly decreased E. larvarum egg hatching, puparium formation, and female fecundity compared to the standard medium. In perspective, M. fistulosa hydrolate supplemented to the artificial media for E. larvarum may be considered as a promising candidate to replace the gentamicin solution, as suggested also by the microbiological analyses of the media, performed at various growth stages of the parasitoid in a separate trial. Conversely, the hydrolate‐saturated air treatment was deemed unsuitable.  相似文献

Antheraea pernyi is a semi‐domesticated lepidopteran insect species valuable to the silk industry, human health, and ecological tourism. Owing to its economic influence and developmental properties, it serves as an ideal model for investigating divergence of the Bombycoidea super family. However, studies on the karyotype evolution and functional genomics of A. pernyi are limited by scarce genomic resource. Here, we applied PacBio sequencing and chromosome structure capture technique to assemble the first high‐quality A. pernyi genome from a single male individual. The genome is 720.67 Mb long with 49 chromosomes and a 13.77‐Mb scaffold N50. Approximately 441.75 Mb, accounting for 60.74% of the genome, was identified as repeats. The genome comprises 21,431 protein‐coding genes, 85.22% of which were functionally annotated. Comparative genomics analysis suggested that A. pernyi diverged from its common ancestor with A. yamamai ~30.3 million years ago, and that chromosome fission contributed to the increased chromosome number. The genome assembled in this work will not only facilitate future research on A. pernyi and related species but also help to progress comparative genomics analyses in Lepidoptera.  相似文献

In a previous study it has been reported that dsp28 is induced during desiccation in Tenebrio larvae. During that study it was observed that in non-stressed larvae the concentration of dsp28 in hemolymph drops dramatically just prior to pupation. These results suggested that control of dsp28 synthesis is subject to environmental as well as hormonal cues. This study identifies a site of synthesis as fat body, as dsp28 was secreted into the medium during in vitro incubation of larval fat bodies. Using immunoelectrophoresis to determine protein concentration a developmental profile showing changes in levels of dsp28 in hemolymph during larval, pupal and adult stages of Tenebrio molitor was established. The concentration of dsp28 in larval hemolymph dropped from 4 to 5 mg/ml to 1.5 mg/ml just prior to pupation. This lower level was maintained until adults emerged when the concentration of dsp28 rose to prepupal levels again. Hormonal regulation is suggested since application of methoprene to newly-emerged pupae resulted in an increased incorporation of radiolabeled cysteine into dsp28.  相似文献

The trace fossil Teisseirei barattinia, found in Cenozoic formations of Uruguay and Argentina, is an elongated chamber recognizable by its depressed cross‐section, antechamber, and its multi‐layered lining with an inner surface texture composed of densely packed sub‐rectangular to sub‐triangular pits. Our recent behavioural observations on larval and pupal stages of Sphingidae (Lepidoptera), particularly on Manduca rustica, suggest that Teisseirei barattinia is the pupation chamber of a sphinx moth. Last instar larvae of Manduca rustica, Eumorpha anchemolus and E. labruscae were placed in terraria to observe their burrowing behaviour and to recover pupation chambers. Chambers show depressed or plane convex cross‐sections as T. barattinia. The internal surface texture of walls is also similar to that of T. barattinia. The same pattern could be obtained experimentally by pressing the true legs of Manduca rustica larva against plasticine. The multi‐layered wall structure, shown by T. barattinia, is a new type of lining for insect trace fossils in palaeosols that result from soil packing combined with discharges of abundant liquid excretion by soft‐bodied larvae, as in the case of Manduca rustica. T. barattinia is the first trace fossil documented in palaeosols attributed to sphinx moths and supported by macro and micromorphological comparisons with extant pupation chambers. The shallow emplacement of moth pupation chambers in soils suggests that T. barattinia would be a good indicator of palaeosol upper horizons.  相似文献

Parasitism of fifth instar Manduca sexta larvae by the gregarious parasitoid Cotesia congregata prevented normal storage of tyrosine in the hemolymph, whereas total tyrosine levels increased over eight times in the hemolymph of unparasitized larvae by day 4. Tyrosine glucoside, the hemolymph storage form of tyrosine and the precursor for pupal cuticle sclerotizing agents, was found only in trace amounts in parasitized larvae at the time of parasitoid emergence, but had increased to over 6 mM in hemolymph of unparasitized larvae. Concentrations of dopamine and N-β-alanyldopamine (NBAD), precursors for melanization and sclerotization of cuticle, respectively, had approximately doubled in the hemolymph of parasitized larvae by the day of parasitoid emergence, but not in unparasitized larvae. Catecholamine biosynthesis may be transiently stimulated for wound-healing, as black melanic pigmentation appeared around the wasp emergence holes in the host integument. C. congregata larvae accumulate tyrosine, dopamine, and NBAD by the time of emergence and cocoon spinning, either by direct uptake or by synthesis from precursors obtained from the host. NBAD increased in parasitoid larvae close to pupation, suggesting it functions as the main precursor for pupal cuticle tanning. Both dopamine and NBAD increased dramatically in pharate adult wasps just before eclosion and N-acetyldopamine (NADA) appeared for the first time. Dopamine was highest in concentration and total amount, and it can serve both as a precursor for black melanic pigmentation of adult wasp cuticle and for synthesis of NADA and NBAD, the precursors for cuticle sclerotization. In vitro rearing of the egg parasitoidTrissolcus basalis (WOLL.) from eggs collected on the natural hostNezara viridula (L.) was initiated. Several oligidic diets containing insect material (Manduca sexta hemolymph or host egg content) were tested. Our initial medium with 50% hemolymph induced a high egg mortality, but by decreasing the hemolymph concentration, increasing the hen egg yolk concentration and adding 15% of free amino acids mixture, a hatching rate of 85% of the parasitoid eggs was obtained with 39% reaching the second instar and 33% the third instar. In a medium without hemolymph, but with 18% liquid from parasitized host eggs we obtained 90% to 100% hatching, 25 to 27% reaching the second instar and 8% the third instar. We did not obtain pupation from eggsin vitro, but did get pupae and adults from larvae rearedin vivo to second instar and transfered to anin vitro system.   相似文献

The acceptance of 40 different strains of 24 Trichogramma (Hymenoptera: Trichogrammatidae) species for oak-silkworm host eggs, Antheraea pernyi Guerin-Meneville (Lepidoptera: Anthelidae) was tested in laboratory experiments. The oak-silkworm, which is commercially used in China to mass produce Trichogramma dendrolimi Matsumura on a large scale, was accepted for egg laying by 10 out of the 24 species tested but only 3 species (four strains of T. dendrolimi, three strains of T. chilonis Ishii and one strain of T. cacoeciae Marchal) successfully completed development to adult emergence. The number of adults emerged per host egg averaged 83.2, 37.0, 42.3, 53.0 for four different strains of T. dendrolimi; 42.5, 10.0 for two strains of T. chilonis; 24.5 and 0 for two strains of T. cacoeciae.

Seven other Trichogramma species develoedp in A. pernyi eggs, but no adult emergence occurred and no emergence holes on the chorion were found. The number of Trichogramma larvae, pupae, and adults together per host egg averaged 81.7 and 67.4 for two strains of T. embryophagum Hartig; 39.0 for T. japonicum Ashmead; 35.0, 16.7, 19.0, 0 for four strains of T. evanescens Westwood; 18.7, 0, 0, 0 for four strains of T. brassicae Bezdenko; 11.5 for T. piceum Dyurich; 76.4 and 23.0 for two unidentified strains collected in apple and vine orchards in Germany, respectively.

The following 14 Trichogramma species did not parasitize any A. pernyi eggs: T. atopovirilia Oatman & Platner, T. bourarachae Pintureau & Babault, T. buesi Voegél, T. funiculatum Carver, T. ivelae Pang & Chen, T. meyeri Sorokina, T. minutum Riley, T. nerudai Pintureau, T. nubilale Ertle & Davis, T. ostriniae Pang & Chen, T. principium Sugonjaev & Sorokina, T. pretiosum Riley and two further unidentified strains that originated from France and Switzerland.

The results confirmed that A. pernyi is a suitable host for rearing T. dendrolimi and T. chilonis and that the two species T. cacoeciae and T. embryophagum, under optimal conditions, might be possible candidates for rearing.  相似文献

Artificial diets have been developed to sustain the mass rearing of a wide range of arthropod natural enemies, with varying success. In some cases, such diets can be optimized using insect‐derived materials, such as haemolymph. In this study, we examined the effect of supplementing haemolymph of the black soldier fly, Hermetia illucens, to a basic artificial diet for the phytoseiid mite Amblyseius swirskii. The survival, development and reproduction of the predatory mite were assessed when fed on artificial diets composed of honey, sucrose, tryptone, yeast extract and egg yolk, supplemented with 5%, 10%, or 20% of H. illucens pre‐pupal haemolymph. Developmental time from larva to adult was shorter for males and females offered artificial diets supplemented with 20% haemolymph vs. the basic diet. The oviposition rate and total fecundity of females reared on the basic diet were substantially lower than those of females supplied with the enriched diets. The intrinsic rate of increase was highest on the diet containing 20% haemolymph, followed by those containing 10% and 5% haemolymph. In a subsequent diet‐switching experiment, mites fed on the basic diet in their juvenile stages were switched upon adulthood to diet enriched with different concentrations of H. illucens haemolymph. The females that were fed with the enriched diets from the adult stage on had higher oviposition rates and fecundities than those maintained on the basic diet, but their reproductive parameters were not significantly affected by the concentration of the haemolymph in the artificial diet. In conclusion, supplementing artificial diets with black soldier fly haemolymph significantly improved their nutritional value for A. swirskii. Our findings indicate the potential of using H. illucens as a cheap source for haemolymph in artificial diets, as the fly can be cost‐effectively produced at a large scale on organic waste materials.  相似文献

Trichogramma dendrolimi can be successfully reproduced in fresh eggs dissected from ovaries of the Chinese tussah silkworm (Antheraea pernyi) and is widely used in biological control of lepidopteran agricultural and forest pests in China. Diapause induction of T. dendrolimi in A. pernyi eggs was investigated through exposing the parasitoid to six constant temperatures (16, 13, 10, 7, 4, and 1 °C) for 19 exposure periods between 10 and 46 days. The sensitive age of T. dendrolimi for diapause induction was explored through a separate experiment to examine the parasitoids that had developed for 2, 3, 4, 5, 6, 7, and 8 days at 26 °C after parasitization, under the six constant temperatures, respectively. Diapause was induced at 10 or 7 °C, and the induction period was 4–6 weeks. The sensitive age of T. dendrolimi to react at the induction temperature was 2–3 days (at 26 °C). At 7 and 10 °C, the diapause rate increased with increasing exposure period and decreased with increased T. dendrolimi age at exposure. The optimum method to induce diapause in T. dendrolimi consisted of exposing hosts for parasitization at 26 °C for 8 h, and then keeping them at 26 °C for 40 h, finally, moving them into 10 °C for 4 weeks.  相似文献

The effects of sublethal concentrations of indoxacarb on Plutella xylostella (L.) (Lepidoptera: Yponomeutidae) were examined. In comparison with the control, sublethal concentrations led to a significant decrease in adult emergence, the percentage of pupation and pupal weight of the parent generation, fecundity, and adult longevity. They also significantly decreased the net reproductive rate (R 0), intrinsic rate of increase (r m), finite rate of increase (λ), and gross reproduction rate (GRR), and had significantly increased egg developmental time and doubling time (Dt) of P. xylostella. However, sublethal concentrations of indoxacarb had no significant affect on adult emergence, percentage of pupation, larva, prepupa and pupa developmental periods, hatchability, and sex ratio of offspring. The results of this study indicated that sublethal concentrations can decrease the biological parameters of P. xylostella.  相似文献

The goal of this research was to develop an artificial diet for rearingTrichogramma galloiandTrichogramma pretiosum,which are important egg parasitoids of several pests in Brazil. The diet was based on different proportions of hemolymph ofHelicoverpa zealarvae, chicken egg yolk, powdered milk solutions, bovine fetal serum, and an extract fromHeliothis virescenseggs. The ideal volume of diet for rearingT. galloiwas also evaluated. The parasitoids showed different nutritional requirements for their development. The best proportion of diet components forT. galloiwas 70% hemolymph, 20% egg yolk, 10% fetal serum, and 0.2% streptomycin, and forT. pretiosumit was 70% hemolymph, 20% egg yolk, 5% fetal serum, 5% egg extract, and 0.2% streptomycin. Egg yolk was found to be essential for the development of both parasitoids, and the powdered milk solution was not an important component in the diet. Development was not obtained in diets with hemolymph concentrations lower than 40%. A 3- to 4-μl amount of diet was the ideal volume on which to rearT. galloiwith the techniques reported.  相似文献
